The stem succulent perennial halophyteHaloxylon recurvum Bungeex. Boiss. grows and produces seed under highly saline conditionsand seeds are deposited in saline soils. Experiments were conductedto determine the effect of salinity and temperature on the germinationof seeds. Results indicate that seeds can germinate at veryhigh salt concentrations (500 mM). However, highest germinationpercentages were obtained in distilled water. Cooler thermoperiodspromoted germination, while high temperatures significantlyinhibited the germination of seeds at all NaCl concentrationstested. Rate of germination decreased with increases in salinity.At higher thermoperiods the rate of germination was significantlylower in comparison to lower thermoperiods. Seeds recover afterbeing transferred to distilled water and recovery was higherfrom higher salinity concentrations and lower thermoperiods.Final recovery germination percentages in high salt treatmentswere significantly lower than non-saline controls, indicatingthat exposure to high concentration of NaCl permanently inhibitedgermination. A rapid and economical method is reported for the preparation of an immobilized enzyme reactor (IMER) using silica-encapsulated equine butyrylcholinesterase (BuChE) as a model system. Peptide-mediated silica formation was used to encapsulate BuChE, directly immobilizing the enzyme within a commercial pre-packed column. The silica/enzyme nanocomposites form and attach simultaneously to the metal affinity column via a histidine-tag on the silica-precipitating peptide. BuChE-IMER columns were integrated to a liquid chromatography system and used as a rapid and reproducible screening method for determining the potency of cholinesterase inhibitors. The IMER preparation method reported herein produces an inert silica-encapsulation matrix with advantages over alternative systems, including ease of preparation, high immobilization efficiency (70-100%) and complete retention of activity during continuous use.  相似文献

Several marine fungi collected from the waters of Prince Edward Island, Canada, were screened for the presence of natural products exhibiting antibacterial activity. Both broths and mycelia of these fungi were studied using the bioassay-guided chromatographic separation. The 4 fractions from the extract of mycelia of Corollospora lacera exhibited weak antibacterial activity and were analyzed further. From these fractions, 2 sterols (5 alpha,8 alpha-epidioxyergosterol and 22E,24R-ergosta-7,22-diene-3beta,5 alpha,6 beta-triol) and a 3:1 mixture of linoleic and oleic acids were isolated. The presence of ergosterol was confirmed in dichloromethane extracts of mycelia of every fungus in this study and this sterol was isolated from the extract of mycelium of Corollospora lacera. Two other known compounds (5-hydroxymethylfuran-2-carbaldehyde and bis(2-ethylhexyl) phthalate), were isolated from the dichloromethane extract of mycelium of Monodictys pelagica. The phthalate was reported in the literature as a metabolite isolated from the fungi, but in our study it was proven to be an artifact of the culturing and (or) extraction procedures rather than a true fungal metabolite.  相似文献

An examination of the Australian sponge Phakellia aruensis led to the isolation and identification of sterols with six different nuclei. Eight new sterols were isolated which included three delta 15-A-nor sterols, three delta 7-A-nor sterols, and two saturated A-nor sterols. Their structures were established through mass spectrometry and 1H-NMR spectral studies.  相似文献

Several polysaccharides have been isolated from the cell walls of Phytophthora parasitica, a phytopathogenic fungus of carnation. The crude polysaccharides were fractionated by successive chromatographies on DEAE-cellulose, Sephadex G-25, concanavalin-A-Sepharose and Sephadex G-200 columns. The neutral polysaccharides consist of a mixture of beta(1----3, 1----6)-D-glucans whose relative molecular masses varied from 9000 to about 200 000. All these polysaccharides have a main chain of beta(1----3)-linked D-glucose residues. They differ by the presence of 1----6 branched chains consisting of D-glucose and D-Glc-(1----3)-D-Glc, for the lowest molecular mass polysaccharides or D-Glc-(1----3)-D-Glc-(1---3)-D-Glc for the highest molecular mass polysaccharides.  相似文献

A total of 27 natural products were isolated from Saussurea cauloptera, including two new eudesmane-type sesquiterpenoids, the gerin derivatives 2 and 3, and one new ent-labdane diterpenoid, compound 9. The known compounds included six sesquiterpenoids, eleven triterpenoids, six sterols, and one lignan. Their structures were elucidated on the basis of extensive spectroscopic and mass-spectrometric analyses, as well as by X-ray crystallography in the case of gerin (1). The structurally related compounds 1-4 were found to exhibit strong inhibitory activities against human gastric carcinoma (SGC-7901) cells.  相似文献

采用cDNA-AFLP技术从梭梭中获得了一个干旱诱导的TDF序列,编码RAN基因的cDNA片段,利用5'-RACE和3'-RACE技术克隆到一个1 083 bp的全长基因,含有一个编码221个氨基酸的开放阅读框,具有结合GTP/GDP的4个保守结构域,属于RAN1亚家族,命名为HaRAN1.Blast分析表明,HaRAN1与单、双子叶植物(毛果杨、拟南芥、小麦、水稻)的一致性高达95%~96%以上;与哺乳动物(人类)和真菌(烟曲霉)的一致性高达88%以上;半定量RT-PCR分析显示,干旱、PEG和NaCl胁迫明显诱导HaRAN1基因的表达.结果表明,HaRAN1基因编码蛋白在物种间具有高度保守性,可能在梭梭适应极端干旱的沙漠生境中起重要作用.  相似文献

The phenolic compounds methyl 2-O-feruloyl-1a-O-vanillactate (1), caffeic anhydride (2), and trans 4-hydroxycyclohexyl-2-O-p-coumaroyl beta-D-glucopyranoside (3) have been isolated from the AcOEt-soluble fraction of Ehretia obtusifolia, along with methyl rosmarinate (4) and rosmarinic acid (5), which are reported for the first time from this species. Their structures were determined by means of 1D- and 2D-NMR techniques. Compounds 1-5 inhibited lipoxygenase in a concentration-dependent manner, with Ki values ranging from 0.85-57.6 microM. Compounds 1, 2, 4, and 5 showed noncompetitive inhibition, whereas 3 was found to be an uncompetitive inhibitor of lipoxygenase.  相似文献

By gas-liquid chromatography, it was demonstrated that the content of beta-sitosterol made 0.3--0.4% of its cholesterol complex in summer milk fat. Emergence and accumulation of beta-sitosterol in milk were not associated with the content of oil-cake and meal but depended on the content of green mass in the cow's diet.  相似文献

Four lanostane triterpenes, 3,7,11-trioxo-5α-lanosta-8,24(E)-dien-26-oic acid, methyl 11α-3,7-dioxo-5α-lanosta-8,24(E)-dien-26-oate, methyl 3,7,11,12,15,23-hexaoxo-5α-lanost-8-en-26-oate, and ethyl 3,7,11,12,15,23-hexaoxo-5α-lanost-8-en-26-oate, two sterols, (14α,22E)-14-hydroxyergosta-7,22-diene-3,6-dione and a steroid named as camphosterol A were isolated from a mixture of fruiting bodies and mycelia of solid cultures of Antrodia camphorata. The 1H and 13C NMR spectra of all compounds were fully assigned using a combination of 2D NMR experiments, including COSY, HMQC, HMBC and NOESY sequences. Six compounds were evaluated for cytotoxicity against several human tumor cell lines, all of which has moderate activity.  相似文献

为探讨多花地宝兰(Geodorum recurvum)胚胎发育的系统分类学意义,采用石蜡制片法对多花地宝兰胚囊和胚的发育进行解剖学观察。结果表明,在开花前,多花地宝兰胚珠原基发育缓慢,开花授粉后胚珠原基快速发育成"树状二杈分枝结构",随后在"分枝结构"末端形成孢原细胞,开始胚囊发育。多花地宝兰的胚囊发育属于单孢蓼型胚囊,胚珠具有双层珠被。孢原细胞形成后,经过细胞膨大延长发育形成胚囊母细胞,胚囊母细胞经过减数分裂形成线性四分体,在珠孔端形成1个功能大孢子,功能大孢子经过3次有丝分裂形成8核胚囊。多花地宝兰的胚发育具有藜型和紫苑型两种方式。双受精完成后,多花地宝兰合子进行一次橫裂后形成基细胞和顶细胞;基细胞经过多次分裂形成细胞团,细胞团中的细胞向不同方向膨大延长形成多个胚柄细胞;顶细胞有两种分裂方式,一种是横裂形成藜型胚,一种是纵裂形成紫苑型胚。因此,推测多花地宝兰在兰科植物系统分类学上属于较为原始种。  相似文献
