The gradual change of enzymes of glycogen metabolism proceeds during the skeletal muscle differentiation in the loach. The portion of the muscle type phosphorylase in the skeletal muscles of the embryo at the stage of the beginning of movement amounts to 30% and that at the stage of hatching to slightly over 50%. At the stage of yolk resorption, the skeletal muscles contain the muscle type phosphorylase only. At the same time the value of KM(UDPG) for glycogen synthetase gradually increases from 0,1 X 10(-3) up to 0,57 X 10(-3) M. The activity of alpha-glycerophosphate dehydrogenase increases more than 70 times.  相似文献

At normal ambient temperatures, in absence of stress, in babies during first months of life (in contrast to less maturely born mammals), the respiratory activity of skeletal muscles is not realized, being observed only under stress conditions. In 1-1 1/2 months old puppies, after deafferentation of the hindlimb, the frequency of movements of a jerk type increases approximating the level observed in newborn animals. At normal ambient temperatures, these puppies exhibit respiratory activity of skeletal muscles which is not observed at this age in control animals.  相似文献

The intraperitoneal administration of corticotropin (ACTH) in the rate of 1 and 2 units per 100 g of body weight and that of hydrocortisone in the rate of 1 mg and 5 mg per 100 g body weight were studied for their effects on carbohydrate metabolism rate in musculus gastrocnemius as well as on the level of 11-oxycorticosteroids in blood plasma of rats. The glycogen level in muscles was found to rise 3 hours after ACTH and hydrocortisone administration and it correlated with the hydrocortisone level increase in blood plasma (r = 0.714 and 0.863, respectively); the activity of pyruvate kinase decreased. Simultaneously ACTH did not change while hydrocortisone lowered the phosphorylase activity and the content of both fructose-6-phosphate and lactate.  相似文献

Three skeletal muscles viz., gastrocnemius, pectoralis and diaphragm from rats acclimated to a low temperature (4 +/- 1 degrees C; 16 hr daily; maximum for 8 weeks) exhibit an increased myosin ATPase activity. An analysis of native myosin from these muscles under non-dissociating conditions reveals two myosin isozymes instead of a single isozyme expressed in control muscles. Isoelectric focusing (IEF) coupled with two dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis (2-D SDS-PAGE) confirms an increased phosphorylation of myosin light chain 2 (MLC2) in muscles from cold acclimated rats.  相似文献

In the bivalve mollusksCrenomytilus grayanus, Mizuhopecten yessoensis, Mercenaria stimpsoni, andPeronidia venulosa, the activity of hexokinase (HK) and pyruvate kinase (PK), the key enzymes of glycolysis, and of glucose-6-phosphate dehydrogenase (G6PhDH), the main enzyme of the pentose phosphate path, was determined in the presence of heightened cadmium concentrations (500 mg/l). Under the effect of cadmium, the enzyme activity either decreased immediately or underwent an initial increase and decreased later. Such a response is consistent with the general theory of stress and suggests a difference in the adaptive capacities of the mollusks studied.  相似文献

Some aminopeptidase activities, dipeptidase-, tripeptidase-, and carboxypeptidase activities were measured in two different types of skeletal muscle in rabbit soleus muscle as a slow oxidative, and gastrocnemius muscle as a fast glycolytic type after immobilization in full extension with a plaster cast for 1, 2, 4, 7, 14 or 28 days. In correlation to the higher protein turnover in red muscles, the activities except of leucine and alanine aminopeptidase were higher in the normal soleus muscle than in the gastrocnemius muscle. Much higher activities of the tested enzymes were obtained in the immobilized soleus muscle than in the normal one after 2 weeks of immobilization. In the gastrocnemius muscle the tested enzyme activities generally did not change or decrease. The results demonstrate that the peptidases play a role in the process of protein breakdown in normal and disused skeletal muscles.  相似文献

Activity changes of a number of enzymes involved in carbohydrate metabolism were determined in cell extracts of fractionated exponential-phase populations of Saccharomyces cerevisiae grown under excess glucose. Cell-size fractionation was achieved by an improved centrifugal elutriation procedure. Evidence that the yeast populations had been fractionated according to age in the cell cycle was obtained by examining the various cell fractions for their volume distribution and their microscopic appearance and by flow cytometric analysis of the distribution patterns of cellular DNA and protein contents. Trehalase, hexokinase, pyruvate kinase, phosphofructokinase 1, and fructose-1,6-diphosphatase showed changes in specific activities throughout the cell cycle, whereas the specific activities of alcohol dehydrogenase and glucose-6-phosphate dehydrogenase remained constant. The basal trehalase activity increased substantially (about 20-fold) with bud emergence and decreased again in binucleated cells. However, when the enzyme was activated by pretreatment of the cell extracts with cyclic AMP-dependent protein kinase, no significant fluctuations in activity were seen. These observations strongly favor posttranslational modification through phosphorylation-dephosphorylation as the mechanism underlying the periodic changes in trehalase activity during the cell cycle. As observed for trehalase, the specific activities of hexokinase and phosphofructokinase 1 rose from the beginning of bud formation onward, finally leading to more than eightfold higher values at the end of the S phase. Subsequently, the enzyme activities dropped markedly at later stages of the cycle. Pyruvate kinase activity was relatively low during the G1 phase and the S phase, but increased dramatically (more than 50-fold) during G2. In contrast to the three glycolytic enzymes investigated, the highest specific activity of the gluconeogenic enzyme fructose-1, 6-diphosphatase 1 was found in fractions enriched in either unbudded cells with a single nucleus or binucleated cells. The observed changes in enzyme activities most likely underlie pronounced alterations in carbohydrate metabolism during the cell cycle.  相似文献
