Infectivity of second‐stage juvenile (J2) populations of Heterodera schachtii was assayed with radish.The numbers of J2 in three‐day‐old seedlings were proportional to the numbers of J2 in two differently textured soils.In a microplot trial with a known H.schachtii‐supprcssivc soil, half of the plots contained untreated suppressive soil, the other half contained the same soil, but methyl iodide‐fumigated and therefore conducive.Both soils were infested with cysts introducing the equivalents of 0, 30, 60 or 120 H.schachtii eggs g‐1 soil, kept moist for 2 months, and then planted to Swiss chard.The numbers of J2 in radish roots were proportional to the numbers of H.schachtii eggs introduced into the microplots, at a low level of detection in suppressive soil and at a high level in conducive soil.Growth of Swiss chard was not different at increasing infestation levels in suppressive soil, but growth was reduced in conducive soil proportionally to increasing nematode infestation level.  相似文献

Soils collected from five districts of Hawaii county were infested with Rhtzoctonia solani in small inoculum particles and successfully planted with radish to induce suppression, Suppressiveness was induced in some, but not all, replicates of all. soils. When fresh inoculum was added, suppressiveness was demonstrated in some, but not all, replicates of two soils, but not in the other three soils. Acidity of soil was not important in successful induction of suppression. Characteristics of induced suppression in soil from one site (S. Kohala) were further investigated. Reduction of microbial population by heat treatment of suppressive soil completely nullified its inhibitory effect. The populations of actinomycetes, fungi in general and Trichoderma spp. in suppressive and conducive soil were not significantly different. However, the population of bacteria in suppressive soil was almost four times higher than that in conducive soil. The survival time of R. solani in suppressive soil was shorter than that in conducive soil. Hyphae of R. solani also lysed faster in suppressive soil than in conducive soil. It is suggested that suppressiveness of the South Kohala soil created by monoculture is due to enhanced competitive pressure generated by an increased bacterial population, which in turn causes the rapid autolysis of R. solani hyphae.  相似文献

Seaweed concentrate (SWC), prepared fromEcklonia maxima, when applied as a soil drench to tomato seedlings, significantly increased plant growth and reduced infestation byMeloidogyne incognita. Foliar applied SWC had little effect on plant growth and increased nematode galling. Ashing SWC reduced the suppressive effect on nematode infestation. In anin vitro experiment, SWC lessened infestation of root-knot nematodes on excised roots of a susceptible cultivar of tomato. Application of the same concentrations of SWC to a nematode-resistant cultivar increased the number of egg masses.  相似文献

Summary Certain volatile organic acids stimulated the growth of Stereum sanguinolentum when added through the gas phase or to the liquid nutrient medium. Some of the acids were identified in extract of heat-treated wood of pine (Pinus silvestris), which had been shown (Suolahti, 1951) to have a growth-promoting effect on Stereum sanguinolentum and other wood-rotting fungi.The effect of volatile materials from a heat-treated piece of pine-wood and of caproic acid was tested on 13 other wood-rotting fungi. The growth of 11 of them was stimulated by volatiles from pine-wood, only 2 by caproic acid.The morphology of Coniophora cerebella was altered when volatiles from a neutral fraction of pine-wood extract was added through the gas phase.  相似文献

Phytophthora cinnamomi-suppressivc soils were found to be widely distributed in nature. About 40 % of soil samples collected from locations with different vegetation, soil type or elevation throughout the island of Hawaii were suppressive to chlamydospore germination of Phytophthora cinnamomi. Soil samples collected from the same general areas varied greatly in degree of suppressive-ness to P. cinnamomi, ranging from conducive to strongly suppressive. Among the 155 soil samples tested, those with pH close to 4 or 8 tend to be more suppressive to P. cinnamomi than those with pH close to 6.  相似文献

A field experiment in which main‐crop potatoes were grown every other year was conducted on a sandy soil from 1994 to 1999. The aim of the experiment was to control soil‐borne pathogens of potato with ecologically sound methods. Potato grown as a trap crop from the end of April to the end of June (8 wk) was used to control potato cyst nematodes (PCN) (Globodera pallida), and its effects on other important soil pathogens and on the growth of a subsequent potato crop were also assessed. Additional experimental treatments were a potato crop from which the haulm was removed and a green manure crop. Three potato cultivars with different degrees of resistance to PCN were grown as the main crop. Duplicate sets of the experiment were run concurrently. The PCN were effectively controlled by the potato trap crop. When a highly resistant potato cultivar was grown as a main crop after the trap crop, the post‐harvest soil infestation was very low. When a moderately resistant cultivar was grown after the trap crop the soil infestation also remained low. When the trap crop was alternated with a susceptible potato cultivar as a main crop, soil infestation increased slightly, but the degree of control when compared with no trap crop averaged 96%. Soil infestation with root‐knot nematodes (mainly Meloidogyne hapla) increased when potato was grown as a trap crop, but soil infestation with the root‐lesion nematode Pratylenchus crenatus was not affected. Stem canker caused by Rhizoctonia solani was not affected by the trap crop but black scurf (sclerotia of R. solani) on tubers was reduced. Soil infestation with Verticillium dahliae declined in one of the duplicate sets of the experiment but not in the other. However, stem infections by V. dahliae were significantly decreased in both sets, although the effect depended on the PCN‐resistance level of the potato cultivar. When a highly resistant potato cultivar was grown Verticillium stem infections were not significantly affected, they were decreased with a moderately resistant cultivar but the decrease was most pronounced with a PCN‐susceptible cultivar. Senescence of a following potato crop was not influenced by the trap crop when a highly PCN‐resistant cultivar was grown, but it was delayed in the case of a moderately resistant or a susceptible cultivar, resulting in higher tuber yields for those cultivars. The experiment proved that a trap crop can be an alternative to chemical soil disinfection but, for several reasons, the potato itself is not an ideal crop for this purpose; a trap crop other than potato must be developed.  相似文献

Culture supernatants of splenic T cells from susceptible CBA mice chronically infected with Trypanosoma cruzi contain a suppressive substance which can inhibit the induction of delayed-type hypersensitivity (DTH) to a wide range of antigens. The suppressive substance is distinct from T. cruzi antigen inasmuch as the supernatant depleted of any residual T. cruzi antigen by an affinity column still retains the suppressive activity, whereas addition of T. cruzi antigens to control supernatant did not confer suppressive function. The suppressive supernatant does not contain detectable levels of IL-1, IL-2, IL-3, or IFN-gamma but a modest level of IL-1 and IL-2 inhibitory activities. However, both these inhibitory activities elute at a different position from the DTH suppressive activity on gel filtration. The DTH suppressive activity is heat labile (1 h, 56 degrees C), cryostable, but destroyed by trypsin treatment. It binds to ricin but not to lentil lectin. Sepharose 4B gel filtration and HPLC analysis in mild chaotropic agents (urea, ethylene glycol) demonstrate that the suppressive substance has an apparent Mr of 30 to 60 kDa, but full DTH-suppressive activity is retained only in an aggregated form.  相似文献

Sensitivity of conidia of Cochliobolus victoriae to fungistasis decreased markedly following incubation on moist sand for at least 1 h. Germination was greater on Conover loam or on sand being leached with water than on an alkaline clay loam soil known to produce a volatile fungistatic substance. Evolution of 14CO2 began within 3 min after [14C]glucose was applied to the soils; the rate of 14CO2 evolution was faster with Conover loam. Germination of Thielaviopsis basicola conidia per unit of glucose remaining in agar discs initially containing 0-1% glucose, was lower for discs incubated on the clay loam soil than on Conover loam, and was greatest on a bed of sand undergoing aqueous leaching. Germination of ascospores of Neurospora tetrasperma and conidia of C. victoriae was suppressed on discs of washed, Purified Agar or polyacrylamide gel incubated on or over the clay loam soil, but no suppression resulted when discs were incubated on Conover loam. Extensive aeration of either soil did not remove its fungistatic effect. Fungistasis in Conover loam appears to be caused primarily by nutrient deprivation, whereas volatile fungistatic substances may play a major role in the clay loam soil.  相似文献

Leaf composts were studied for their suppressive effects on Pythium ultimum sporangium germination, cottonseed colonization, and the severity of Pythium damping-off of cotton. A focus of the work was to assess the role of fatty-acid-metabolizing microbial communities in disease suppression. Suppressiveness was expressed within the first few hours of seed germination as revealed by reduced P. ultimum sporangium germination, reduced seed colonization, and reduced damping-off in transplant experiments. These reductions were not observed when cottonseeds were sown in a conducive leaf compost. Microbial consortia recovered from the surface of cottonseeds during the first few hours of germination in suppressive compost (suppressive consortia) induced significant levels of damping-off suppression, whereas no suppression was induced by microbial consortia recovered from cottonseeds germinated in conducive compost (conducive consortia). Suppressive consortia rapidly metabolized linoleic acid, whereas conducive consortia did not. Furthermore, populations of fatty-acid-metabolizing bacteria and actinobacteria were higher in suppressive consortia than in conducive consortia. Individual bacterial isolates varied in their ability to metabolize linoleic acid and protect seedlings from damping-off. Results indicate that communities of compost-inhabiting microorganisms colonizing cottonseeds within the first few hours after sowing in a Pythium-suppressive compost play a major role in the suppression of P. ultimum sporangium germination, seed colonization, and damping-off. Results further indicate that fatty acid metabolism by these seed-colonizing bacterial consortia can explain the Pythium suppression observed.  相似文献

A field soil, artificially infested with pathogenic isolates of Fusarium oxysporum f. sp. melonis was continuously used for screening resistant varieties of melon to Fusarium wilt. After 9–10 years of continuous cropping with resistant varieties, the soil had developed induced suppressiveness. Seven to 9 experimental replantings of the induced suppressive soil with the susceptible cultivar of melon, ‘Ein-Dor', nullified its suppressiveness. This was expressed by 90 % disease incidence. Only 2 replantings were required to obtain the same disease incidence in an adjacent field of a conducive soil. Nonpathogenic isolates of F. oxysporum, isolated from the rhizospheres of melon seedlings, induced various degrees of soil suppressiveness when added to soil at various ratios to the pathogenic isolate.  相似文献

This study assessed the potential impact of various Fusarium strains on the population development of sugarbeet cyst nematodes. Fungi were isolated from cysts or eggs of Heterodera schachtii Schmidt that were obtained from a field suppressive to that nematode. Twenty-six strains of Fusarium spp. were subjected to a phylogenic analysis of their rRNA-ITS nucleotide sequences. Seven genetically distinct Fusarium strains were evaluated for their ability to influence population development of H. schachtii and crop performance in greenhouse trials. Swiss chard (Beta vulgaris) seedlings were transplanted into fumigated field soil amended with a single fungal strain at 1,000 propagules/g soil. One week later, the soil was infested with 250 H. schachtii J2/100 cm3 soil. Parasitized eggs were present in all seven Fusarium treatments at 1,180 degree-days after fungal infestation. The percentage of parasitism ranged from 17 to 34%. Although the most efficacious F. oxysporum strain 471 produced as many parasitized eggs as occurred in the original suppressive soil, none of the Fusarium strains reduced the population density of H. schachtii compared to the conducive check. This supports prior results that Fusarium spp. were not the primary cause of the population suppression of sugarbeet cyst nematodes at this location.  相似文献

Strains of Streptomyces were tested for their ability to reduce population densities of the root-lesion nematode (RLN), Pratylenchus penetrans, in roots of alfalfa (Medicago sativa) in growth chamber assays. Previously, these strains were shown to suppress potato scab disease, caused by Streptomyces scabies, in field experiments and to inhibit in vitrogrowth of a wide range of plant-pathogenic fungi and bacteria. Inoculation with Streptomyces at planting significantly reduced RLN population densities in roots of both susceptible and resistant alfalfa varieties grown in either heat-treated or untreated soil. Reductions in RLN population densities were observed 6 weeks after nematode inoculation. Shoot dry matter was not affected by any treatment; root dry weight was reduced in Streptomycesplus nematode treatments compared to the nematode inoculation alone in some experiments but was not affected by Streptomyces when RLN was absent. Mutant strains not producing antibiotics in vitro also reduced RLN population densities in alfalfa roots and all strains maintained high population densities after inoculation into heat-treated soil and on alfalfa roots. These strains may be useful in multi-crop, multi-pathogen management programs to augment genetic resistance to plant diseases.  相似文献

To verify current thresholds for Bollgard II® cotton in Australia, the impact of Helicoverpa spp. (Lepidoptera: Noctuidae) larvae on yield, development, and quality under various infestation intensities and durations, and stages of growth, was tested using small plot field experiments over two seasons. Infestation with up to 80 eggs m?1 of Helicoverpa armigera (Hübner) and Helicoverpa punctigera Wallengren showed that species, infestation level, and stage of growth had no significant effect on yields of seed‐cotton or lint and on maturity and fibre quality. The duration of infestation of white flowers with H. punctigera neonates (maximum of every day for up to 4 weeks) had no impact on the yield of seed‐cotton or lint, maturity, and fibre quality, but when 100% of flowers were infested (compared with 0 or 50%), seed‐cotton and lint yields were significantly reduced and maturity was delayed. Infestation with up to 18 medium H. armigera larvae m?1 at several plant stages did not significantly affect yields of seed‐cotton and lint, maturity, and fibre quality. A heliocide spray applied on a commercial farm at the current threshold resulted in a significantly higher lint yield, compared with a farm where no spray was applied. In conclusion, Bollgard II® cotton is highly resistant to Helicoverpa spp. infestation.  相似文献

The effects of coffee extracts on mutagenicity were studied using the Salmonella typhimurium system. Coffee extracts showed inhibitory effects on the mutagenicity of such mutagens as 3-amino-l-methyl-5H-pyrido[4,3–b]indole (Trp-p-2), 2-acetylaminofluorene (AAF), and benzo(a)pyrene (B(a)P), whose mutagenicity require metabolic activation by rat liver microsomal fraction, S-9 mix. The inhibition of mutagenicity increased in proportion to the level of roasting or to the darkness in color of the coffee extracts. When the coffee extracts were applied to a Sephadex G-50 column, two color pigment peaks were observed, the second peak showing the inhibitory activity towards mutagenicity. The inhibitory substance towards the mutagenicity was formed only in a heat-treated mixture of sucrose with either chlorogenic or caffeic acid, among various heat-treated combinations of components in raw coffee beans. The decrease of mutagenicity by coffee extracts was due to inhibiting the metabolic activation by S-9 mix.  相似文献

The mechanisms that permit maternal tolerance of the conceptus allograft during early pregnancy in the mare have not been investigated. Embryos and endometria were collected from mares 14 days after ovulation and cultured for 20.5 h. The effect of addition of culture supernatant on incorporation of [3H]thymidine by equine peripheral blood lymphocytes was studied. Culture supernatant from endometrium of nonpregnant mares did not affect lymphocyte blastogenesis, but supernatant from both embryos and endometrium of pregnant mares reduced concanavalin A (Con A)- and phytohemagglutinin-induced blastogenesis. Five of six cultures performed in the present of indomethacin did not contain immunosuppressive factors. The suppressive effect on Con A-induced blastogenesis was eliminated by charcoal treatment of the supernatants and reduced by treatment with trypsin or heat. Blastogenesis of bovine lymphocytes was inhibited by culture supernatant of endometrium from pregnant mares, but not by embryo supernatant. Preincubation of blood lymphocytes with supernatants from endometrium of pregnant mares enhanced subsequent incorporation of [3H]thymidine by lymphocytes. A 24-h delay in addition of embryo culture supernatants significantly reduced the degree of immunosuppression. These results suggest that probably more than one substance interacts with the lymphocyte cultures and the observed blastogenesis reflects the end result of the interaction between suppressive and stimulating factors. The lymphocyte inhibitory effect evident in supernatants from embryos and endometrium from pregnant mares may be important in local immunosuppression and maternal acceptance of pregnancy.  相似文献

Understanding the environmental factors that influence the rhizosphere and inner root colonization of the disease‐suppressive strains of fluorescent pseudomonads is an essential step towards improving the level and reliability of their biocontrol activity. Soil amendment with Zn at 0.8 or 1.6 mg/kg of soil alone or in combination with Pseudomonas aeruginosa IE‐6S+significantly reduced nematode penetration in tomato roots. Zn applied alone did not reduce root infection caused by Macrophomina phaseolina or Fusarium solani but did reduce when used in combination with IE‐6S+. Soil amendment with Zn at 0.8 or 1.6 mg/kg of soil alone or in conjunction with IE‐6S+ markedly suppressed Rhizoctonia solani infection. Plant height, fresh weight of shoot and protein contents of the leaves substantially improved when used with Zn, however, plants growing in the soil treated with 1.6 mg/kg of Zn in the absence of IE‐6S+ not only reduced plant growth but also showed necrotic symptoms on the leaves. Zn application in the soil decreased populations of IE‐6S+ both in the rhizosphere and root. A positive correlation between bacterial rhizosphere and inner root colonization was also observed. With an increase in nematode densities in the soil, nematode penetration and subsequent galling due to Meloidogyne javanica increased. Regardless of the nematode densities, Zn applied alone or in combination with IE‐6S+ caused marked suppression of M. javanica. At all the population densities of M. javanica, Zn enhanced the efficacy of IE‐6S+ to reduce nematode invasion and subsequent gall development. IE‐6S+ caused significant suppression of soil‐borne root‐infecting fungi both in Zn‐sufficient and Zn‐deficient soil although this suppressive effect accentuated in Zn‐sufficient soils. In the absence of IE‐6S+ and/or Zn, increased nematode densities in the soil significantly reduced plant height, fresh weight of shoot and protein contents of the shoots. With an increase in nematode densities, populations of IE‐6S+ in the rhizosphere and root increased regardless of the Zn application. However, Zn‐deficient soils supported larger populations of IE‐6S+ compared with those of Zn‐sufficient soils.  相似文献

The effect of the water-soluble UV-absorbing substance (UVAS) extracted from the marine red alga Porphyra yezoensis Ueda on UV-dependent thymine photodimer production was investigated. The T<>T pyrimidine-pyrimidone 6-4 dimer and the cyclobutane cis-syn T<>T 5-6 dimer produced by UV irradiation with a xenon lamp were analyzed by reverse-phase high-performance liquid chromatography. Although the dimer production was reduced when the irradiation was filtered through a UVAS solution, it decreased more when thymine was mixed with UVAS. Furthermore, UVAS inhibited the degradation of UV-irradiated thymine. The inhibitory effect of UVAS was significantly greater than that of exogenously added adenine or guanine, which forms complementary base pairs with thymine. These data suggest that in addition to its filtering effect against UV radiation, UVAS also protects thymine by a direct molecule-to-molecule energy transfer process. The protective function of UVAS against UV irradiation is advantageous for this alga under strong UV irradiation.  相似文献

Progesterone-treated pregnancy lymphocytes release an immunologic blocking factor. The mode of action of this substance was investigated. The supernatant of progesterone-treated pregnancy lymphocytes was highly suppressive of natural cytotoxicity toward human embryonic fibroblast target cells as well as of natural killer cell activity. The effect was not observed when progesterone induction was performed in the presence of RU 486, a progesterone receptor blocking agent. The factor was able to inhibit mixed lymphocyte reactions (MLRs), and transfer coculture experiments revealed that this effect was dependent on major histocompatibility complex nonspecific, nonrestricted suppressor T cells. The activation/expansion of suppressor inducer and suppressor effector T cells was further proved by fluorescence-activated cell sorter analysis of the populations from MLRs cultured in the presence of the inhibitory factor. These changes were not observed with MLRs performed in the presence of supernatants from progesterone + RU 486-treated peripheral blood lymphocytes. The inhibitory material, on the other hand, did not affect either production or function of IL-2. We conclude that in the presence of high local concentrations of progesterone, a suppressive pathway dependent on specific progesterone-CD8+ lymphocyte interaction might be established. This mechanism might play an important role in the maintenance of pregnancy.  相似文献

Crude oil from Eucalyptus globulus and E. citriodora was extracted and the rich components, cineole and limonene were fractionated. The vapours of these oils and fractions were adsorbed onto the soil in one set of germination trials while in the other set a vapour column of volatile oils was maintained above the oil-treated soil. In both sets seed germination, seedling growth, relative growth rate, water content, height and number of leaves of Phaseolus aureus var. ML-267 were compared to those of controls. All parameters were found to be significantly affected. The effect was more pronounced with a combination of eucalyptus oil onto soil and a vapour-rich air column. There was a strong correlation between the vapour concentration and its inhibitory effect.  相似文献
