A survey of the pectic content of nonlignified monocot cell walls   总被引:3,自引:1,他引:2       下载免费PDF全文
The primary cell walls of graminaceous monocots were known to have a low content of pectin compared to those of dicots, but it was uncertain how widespread this feature was within the monocots as a whole. Nonlignified cell walls were therefore prepared from 33 monocot species for determination of their pectin content. It was not possible to solubilize intact pectins quantitatively from the cell walls, and the pectin content was assessed from three criteria: the total uronic acid content; the content of α-(1,4′)-D-galacturonan isolated by partial hydrolysis and characterized by electrophoresis and degradation by purified polygalacturonase; and the proportion of neutral residues in a representative pectic fraction solubilized by sequential β-elimination and N,N,NN′-cyclohexanediaminetetraacetic acid extraction. Low galacturonan contents were restricted to species from the Gramineae, Cyperaceae, Juncaceae, and Restionaceae. Other species related to these had intermediate galacturonan contents, and the remainder of the monocots examined had high galacturonan contents comparable with those of dicots. Cell walls extracted from cv. Rutgers, 7711 (ripening inhibited), and nor (non-ripening) tomato ( Lycopersicon eseulentum Mill.) pericarp tissue at various stages of post-maturation development have been separated into four distinct fractions and their carbohydrate composition characterized. The amount of ionically-associated, chelator-soluble (CDTA, cyclohexanediaminetetraacetic acid) uronic acid in 'Rutgers' fruit cell walls remained constant during ripening, whereas the amount of residual pectin, which was extracted with cold alkali (Na2CO3) and was apparently covalently bound, decreased. These changes did not occur in rin and nor mutant fruit at a similar chronological age. The galactose content in pectic polysaccharide preparations extracted from tomato cell walls with CDTA and Na2,CO3, decreased by 65% during ripening. A similar but diminished decrease also occurred in rin and nor fruit. A non-cellulosic polysaccharide(s) was present in walls which resisted extraction with Na-acetate/CDTA, Na2CO3, and 4 M KOH. In 'Rutgers' fruit, the content of galactose in this polysaccharide(s) decreased 44% during ripening, whereas little or no significant change was observed in rin or nor mutant fruit.  相似文献

Following a solid phase extraction, GC-MS and GC-FID procedures, the production of three kaurane derivatives (grandiflorenic, kaurenoic and monoginoic acids) was detected in callus and cell suspension batch cultures of Montanoa tomentosa. From different hormonal combinations, the addition of 0.5 mg 2,4-dichlorophenoxyacetic acid l–1 + 2 mg kinetin l–1 increased the accumulation of total kaurenoids in 6 months old calluses to 2.1 mg g–1 dry weight and in cell suspensions cultures up to 0.76 mg g–1 dry weight. Monoginoic acid, which has not been detected before in leaves of wild plants, accumulated in both in vitro systems.  相似文献

Galacturonan, the main constituent of the backbone (core) of the comaruman macromolecule, a pectin from the marsh cinquefoil Comarum palustre L., was obtained on partial acid hydrolysis of the pectin. Using atomic force microscopy and methylation analysis of the galacturonan, the backbone of the comaruman macromolecule was shown to contain branches as side chains consisting of α-1,4-linked residues of D-galactopyranosyl uronic acid attached to the 2-and 3-positions of the galacturonic acid residues of the core, in addition to linear regions of α-1,4-D-galacturonan. Jasmonic acid altered the accumulation of major anthocyanins in Vitis vinifera cell culture. Peonidin 3-glucoside content at day three was increased from 0.3 to 1.7 mg g–1 dry cell wt while other major anthocyanins were increased by smaller increments. By day 14, the content of methylated and acylated anthocyanins (peonidin 3-p-coumaroylglucoside and malvidin 3-p-coumaroylglucoside) was 6.3 mg g–1 DCW, in response to treatment with jasmonic acid, and comprising 45% (w/w) of total anthocyanins. In comparison, the untreated control culture contained 1.2 mg g–1 DCW which made up 32% (w/w) of total anthocyanins. Light further enhanced anthocyanin accumulation induced by jasmonic acid elicitation. The content of peonidin 3-glucoside at day 3 was 6.6 mg g–1 DCW, 22-fold higher than control cultures while the content in response to light irradiation alone was 0.6 mg g–1 DCW. When a highly pigmented cell line was elicited with jasmonic acid total anthocyanins increased from 9.2 to 20.7 mg g–1 DCW, but there was no change in the anthocyanin composition.  相似文献

Excised root cultures of Gloriosa superba reached 7.5 g dry wt l–1 and accumulated 240±40 g colchicine g–1 cell dry wt after 4 weeks growth. While all precursors (except trans-cinnamic acid) enhanced colchicine content of root cultures without adversely affecting root growth, treatment with p-coumaric acid + tyramine (each at 20 mg l–1) increased colchicine content to 1.9 mg g–1 cell dry wt.  相似文献

Increasing sucrose from 20 to 50 g l−1 in Uncaria tomentosa cell suspension cultures enhanced ursolic acid and oleanolic acid production from 129 ± 61 to 553 ± 193 μg g−1 cell dry wt. The maximal concentration of both triterpenes (1680 ± 39 μg g−1 cell dry wt) was 8 days after elicitation by jasmonic acid, while yeast extract or citrus pectin treatments produced 1189 ± 20 or 1120 ± 26 μg g−1 cell dry wt, respectively. The ratio of ursolic acid:oleanolic acid was constant at 70:30.  相似文献

Summary Aiming to elucidate the possible involvement of pectins in auxin-mediated elongation growth the distribution of pectins in cell walls of maize coleoptiles was investigated. Antibodies against defined epitopes of pectin were used: JIM 5 recognizing pectin with a low degree of esterification, JIM 7 recognizing highly esterified pectin and 2F4 recognizing a pectin epitope induced by Ca2+. JIM 5 weakly labeled the outer third of the outer epidermal wall and the center of filled cell corners in the parenchyma. A similar labeling pattern was obtained with 2F4. In contrast, JIM 7 densely labeled the whole outer epidermal wall except the innermost layer, the middle lamellae, and the inner edges of open cell corners in the parenchyma. Enzymatic de-esterification with pectin methylesterase increased the labeling by JIM 5 and 2F4 substantially. A further increase of the labeling density by JIM 5 and 2F4 and an extension of the labeling over the whole outer epidermal wall could be observed after chemical de-esterification with alkali. This indicates that both methyl- and other esters exist in maize outer epidermal walls. Thus, in the growth-controlling outer epidermal wall a clear zonation of pectin fractions was observed: the outermost layer (about one third to one half of wall thickness) contains unesterified pectin epitopes, presumably cross-linked by Ca2+ extract. Tracer experiments with3H-myo-inositol showed rapid accumulation of tracer in all extractable pectin fractions and in a fraction tightly bound to the cell wall. A stimulatory effect of IAA on tracer incorporation could not be detected in any fraction. When, on the 15th day of growth, an elicitor from Fusarium solani was added at 40 mg l–1 to Cistanche deserticola cell suspension cultures, the contents of echinacoside, acteoside and total phenylethanoid glycosides (PeGs) in cultured cells all increased over the next 27 d by over 100% to 15 mg g–1 dry wt, 9 mg g–1 dry wt and 57 mg g–1 dry wt, respectively. The final biomass (1.3 mg dry wt ml–1) was not affected.  相似文献

Atmospheric N deposition can enhance biological P limitation in terrestrial ecosystems and increase the importance of organic P to plants and microorganisms. We used NaOH–EDTA extraction and solution 31P NMR spectroscopy to determine the P composition of soils in the Upper Teesdale National Nature Reserve, northern England, an upland region influenced by such deposition for at least 150 years. Three characteristic soil types were sampled on three occasions during an annual cycle: blanket peat (318 mg g–1 total C, 607 g g–1 total P, pH 3.9); acid organic soil under grassland (354 mg g–1 total C, 1190 g g–1 total P, pH 3.7); calcareous soil under grassland (140 mg g–1 total C, 649 g g–1 total P, pH 7.3). Between 58 and 99% of the total P in soil and litter layers was extracted by 0.25 M NaOH + 0.05 M EDTA. Extracts of all soils were dominated by organic P, mainly in the form of orthophosphate monoesters (43–69% extracted P). The two acidic soils also contained large proportions of orthophosphate diesters (6–19% extracted P) and phosphonates (7–16% extracted P), suggesting that these compounds become stabilised at low pH. However, a seasonal trend of increasing orthophosphate monoester-to-diester ratios, most evident in the calcareous grassland soil, indicated the preferential degradation of orthophosphate diesters during the growing season. Orthophosphate was the major inorganic P compound (17–34% extracted P), and all soils contained pyrophosphate (1–5% extracted P). However, orthophosphate determined in the NaOH–EDTA extracts by solution 31P NMR spectroscopy was substantially greater than that determined by molybdate colourimetry, suggesting that orthophosphate occurred in complexes with humic compounds that were not detected by conventional procedures. Our results suggest that organisms able to use recalcitrant soil organic P may have a competitive advantage in environments under enhanced atmospheric N deposition.  相似文献

Taxus chinensis var. mairei was grown in two-liquid-phase culture in a 2.5 l airlift external loop reactor for production of Taxol. A mixture of oleic acid and dibutyl phthalate (1:1, v/v) in a volume ratio of 8:92 to the culture medium gave higher production of Taxol (up to 12 mg l–1) with an aeration rate of 1 vvm. By combination of the in situ extraction, precursor feeding and additional sugar introduction, Taxol production (16.7 mg l–1) was 5 times higher than that in the case without any additives (3.4 mg l–1) and 1.4 times of that in the case with only thein situ extraction (12 mg l–1). Taxol was extracted from the organic solvents with water and methanol (1:1:1.4, by vol.) and lyophilized giving a total yield of ca. 90%.  相似文献

A newly established Forsythia × intermedia cell suspension culture was shown to accumulate (+)- and (–)-pinoresinol as well as matairesinol. The influence of the sucrose content of the culture medium and of the cultivation time on pinoresinol and matairesinol accumulation was evaluated. The highest pinoresinol yield was achieved from cells grown in medium containing 6% sucrose for 12 ± 2 days with levels of 0.6–0.8 mg g–1 dry weight and an average enantiomeric composition of 75 ± 5% (+)-pinoresinol. The highest matairesinol amount was reached in the same medium at the 14th ± 2 culture day with levels of 1.0–2.7 mg g–1 dry weight. To our knowledge, this is the first report on pinoresinol accumulation in Forsythia × intermedia plants or cell suspension cultures.  相似文献

Hairy roots of Rauvolfia micrantha were induced from hypocotyl explants of 2–3 weeks old aseptic seedlings using Agrobacterium rhizogenes ATCC 15834. Hairy roots grown in half-strength Murashige & Skoog (MS) medium with 0.2 mg indole 3-butyric acid l–1 and 0.1 mg -naphthaleneacetic acid l–1 produced more ajmaline (0.01 mg g–1 dry wt) and ajmalicine (0.006 mg g–1 dry wt) than roots grown in auxin-free medium. Ajmaline (0.003 mg g–1 dry wt) and ajmalicine (0.0007 mg g–1 dry wt) were also produced in normal root cultures. This is the first report of production of ajmaline and ajmalicine in hairy root cultures of Rauvolfia micrantha.  相似文献

The partitioning of carbon between reserve polysaccharide and alkaloid secondary products was investigated in batch cultures of transformed roots of Datura stramonium grown in media in which the carbon substrate concentration was held constant and the level of mineral nutrients was varied. The growth and accumulation of starch and hyoscyamine was examined in roots grown at temperatures of 20°C, 25°C or 30°C in media containing 5% sucrose and levels of mineral nutrients varying from 1/4 to twice the standard level of Gamborg's B5 salts. The dry matter content was highest (up to 15% w/w) in roots grown at either 20°C or 25°C in medium of the lowest ionic strenth (1/4 B5 salts) and decreased as the ionic strength was raised (down to 7% w/w with 2 B5 salts). Up to half of this decrease could be accounted for by loss of starch from the roots. At 20°C and 25°C, the starch content of the roots grown in medium of the lowest ionic strength (1/4 B5) was 40 mg g-1 and 22 mg g-1 fresh weight respectively but decreased to less than 1 mg g-1 weight at either temperature when the ionic strength of the medium was raised to 2 B5. At 30°C, starch accumulation was severely inhibited in all media. In contrast, varying either the temperature or the ionic strength of the medium had only a small effect on hyoscyamine accumulation which remained at between 0.4–0.6 mg g-1 fresh weight. Although increases in the level of mineral salts had little effect on the hyoscyamine content of the roots, total yields however, increased due to stimulation of growth. Time course experiments showed that cultures grown at either 20°C or 25°C continued to accumulate both starch and hyoscyamine into late stationary phase.  相似文献

In Taxus cuspidata callus, vanadyl sulfate (10 mg l–1) induced a high (146 g g–1 dry wt) production of 10-deacetylbaccatin III in comparison to 7 g g–1 dry wt of the control. The content of paclitaxel in this species increased from 16 g g–1 to 74 g g–1 dry wt when 20 mg phenylalanine l–1 was used. In T. media, p-aminobenzoic acid induced the highest content of 10-deacetylbaccatin III (481 g g–1 dry wt) versus 181 g g–1 in the control. Paclitaxel increased from 89 to 139 g g–1 dry wt after adding chitosan (20 mg l–1) to the cultures.  相似文献

Zanthoxylum piperitum (prickly ash) was grown as a suspension culture in Schenk and Hildebrandt medium supplemented with 50 g sucrose l–1 and 0.5 mg 2,4-dichlorophenoxyacetic acid l–1 for 21 days with elicitation by pectin added at day 15. Volatile compounds were extracted from the culture and 3-hydroxy-2-butanone was identified by GC-MS as the most abundant compound, followed by -butyrolactone and 2,3-butanedione. 2,3-Butanedione, ethyl 3-methylbutyrate, and ethyl 2-methylpropanoate were identified as the most intense aroma-active compounds and represented the characteristic aroma of the culture.  相似文献

Fourteen species of crop plants which differ in their reportedtissue boron requirements were grown in B-replete or B-deficientmedium. Leaf samples were collected and analysed for B and cellwall components. There was a significant positive correlationamong the species between B concentration in the leaf or thecell wall and uronic acid, rhamnose and galactose (indicativeof pectin) in the cell wall. The concentration of cell wallpectin was also positivety related with reported tissue-B requirementsand observed sensitivity to B deficiency. Boron deficiency didnot alter the amount of uronic acid present in cell walls, suggestingthat there was no effect of B deficiency on pectin metabolism.Under B-deficient conditions the amount of ‘soluble’B (i.e. B not associated with the cell wall) declined dramaticallywhile the proportion of cellular B that was ‘insoluble’(i.e. B associated with the cell wall) increased. The positiverelationship between pectin content, insoluble B and tissue-Brequirement of diverse species suggests that the amount of cellwall pectin may be significant in determining the relative tissue-Brequirements of the species. These results indicate that either(1) species with high cell wall pectin contents require greateramounts of B for the construction of the cell wall, or (2) pectinin cell walls forms an insoluble complex with B, thereby reducingits availability for other putative B-requiring metabolic functions.Thus, species with a high pectin content would have a highertissue-B requirement. Extracellular polysaccharides (EPS) of a copper-sensitive (Cus) and a copper-resistant (Cur) Pseudomonas aeruginosa strain were investigated in terms of their production, chemical nature and response towards copper exposure. The extent of EPS synthesis by the resistant strain (4.78 mg mg–1 cell dry wt.) was considerably higher over its sensitive counterpart (2.78 mg mg–1 dry wt.). FTIR-spectroscopy and gas chromatography revealed that both the polymers were acidic in nature, containing alginate as the major component along with various neutral- and amino-sugars. Acid content in the Cur EPS (480.54 mg g–1) was greater than that in the Cus EPS (442.0 mg g–1). Presence of Cu2+ in the growth medium caused a dramatic stimulation (approximately 4-fold) in EPS synthesis by the Cur strain, while in a similar condition, the Cus failed to exhibit such response. The polymer of the resistant strain showed elevated Cu2+ binding (320 mg g–1 EPS) compared to that of the sensitive type (270 mg g–1). The overall observations show the potential of the Cur EPS for its deployment in metal bioremediation.  相似文献

Three reactive dyes were rapidly adsorbed by the mycelium pellets of Penicillium oxalicum. Dye removal of Reactive Blue 19 was up to 60% in 10 min and 91% in 80 min. Dye adsorption isotherms fitted Langmuir model well and the maximum adsorption capacities at 20 °C were calculated to be 160 mg g–1 for Reactive Blue 19, 122 mg g–1 for Reactive Red 241 and 137 mg g–1 for Reactive Yellow 145, respectively. The pellets exhibited a high dye adsorption capacity (80–180 mg g–1) for all of the 3 dyes over a wide pH range (pH 2–10), and the maximum adsorption was obtained at pH 2. The adsorption capacity was mildly increased by increasing salinity.  相似文献

Casein hydrolysate at 2.0 g l–1 significantly enhanced forskolin content (2.3 mg g–1 cell dry wt) in a rhizogenic tumourous line, GCO-RCH-2 of Coleus forskohlii. In rooty teratoma line, RC-ST-2/4, forskolin content enhanced to 1.7 mg g–1 cell dry wt in presence of 2.5 g l–1 casein hydrolysate. Unlike untransformed calli and rhizogenic/root cultures, all the forskolin yielding transformed cultures of C. forskohlii have been maintained for over 5 years.  相似文献
