Summary The immunogold technique, employing antisera with clear-cut specificities, was used to localise different processing stages of pro-opiomelanocortin (POMC) in rabbit melanotropic cells. While the antiserum against 3-MSH labelled all the secretory granules including intrasaccular condensations in the Golgi apparatus, antisera against -MSH only labelled extra-Golgi secretory vesicles (SV). All extra-Golgi SV were likewise labelled with the three antisera against -MSH used, despite their different specificities for the desacetylated, N-acetylated or C-amidated forms of the peptide. The antibody against -endorphin also labelled the extra-Golgi SV, while only some SV were labelled with the antibody against -endorphin. These results correlate with biochemical data in favour of mainly — if not exclusively — intragranular processing of POMC. Except for 3-MSH, the cleavage of which could coincide with Golgi packaging of secretory material, other post-translational modifications of the precursor seem to occur when SV are discharged outside the Golgi area. The cleavage of -endorphin appears to be a later step in POMC processing, occurring in some mature SV.  相似文献

Summary The dorsal tegument of the mature cercaria of Notocotylus attenuatus is a syncytial, cytoplasmic layer, containing two types of secretory granule which are identifiable ultrastructurally. The type 1 secretory bodies are electron lucid, whereas most type 2 granules have a banded appearance. The ventral tegument contains granules which are secreted from the type 3 cells; the type 3 granules are membrane bound, electron dense, and consist of both an amorphous and a finely striated zone. The type 4 cells mainly contain cigar-shaped granules consisting of an amorphous core surrounded by concentric striations. The granules exhibit structural variability in shape and content. The type 4 cells undergo a cellular migration to the tegument during encystment. The structure of the posterior-lateral glands and mode of secretion of the granules are described. Possible functions of microtubules are discussed for each cell type. Details of some secretory processes involved in the formation of the hemispherical cyst wall are described. The layers of the cyst wall may be related to the granular contents of the various parenchymal cells of the cercaria. The tegument of the metacercaria originates primarily from the cytoplasm of the type 1, type 2, type 3 and type 4 cells.  相似文献

Summary Isoelectric focusing of extracts from the endosperm of mature grains of hexaploid wheat and related species was used to study the genetic control of Iodine binding factor (IBF). Ten IBF bands were present in Chinese Spring (CS) and analysis of the nullisomictetrasomic and ditelosomic lines of CS showed nine of them to be controlled by genes on the long arms of the homoeologous group 5 chromosomes. Five alleles were detected at Ibf-A1 locus, four at Ibf-B1 and four at Ibf-D1 among a sample of 46 wheat genotypes. Homoeoloci were found on chromosome 5R of Secale cereale, 5E of Agropyron elongatum, 5U of Aegilops umbellulata, 5Agi of Agropyron intermedium, 5S1 and 4S1 of Aegilops sharonensis and 4H of Hordeum vulgare.  相似文献

Water uptake and transport were studied in eight liana species in a seasonally dry tropical forest on Barro Colorado Island, Panama. Stable hydrogen isotope composition (D) of xylem and soil water, soil volumetric water content (v), and basal sap flow were measured during the 1997 and 1998 dry seasons. Sap flow of several neighboring trees was measured to assess differences between lianas and trees in magnitudes and patterns of daily sap flow. Little seasonal change in v was observed at 90–120 cm depth in both years. Mean soil water D during the dry season was –19 at 0–30 cm, –34 at 30–60 cm, and –50 at 90–120 cm. Average values of xylem D among the liana species ranged from –28 to –44 during the middle of the dry season, suggesting that water uptake was restricted to intermediate soil layers (30–60 cm). By the end of the dry season, all species exhibited more negative xylem D values (–41 to –62), suggesting that they shifted to deeper water sources. Maximum sap flux density in co-occurring lianas and trees were comparable at similar stem diameter (DBH). Furthermore, lianas and trees conformed to the same linear relationship between daily sap flow and DBH. Our observations that lianas tap shallow sources of soil water at the beginning of the dry season and that sap flow is similar in lianas and trees of equivalent stem diameter do not support the common assumptions that lianas rely primarily on deep soil water and that they have higher rates of sap flow than co-occurring trees of similar stem size.  相似文献

Atriplex gmelini plants were regenerated via organogensis from hypocotyl explants. Callus lines were induced from the hypocotyl explants on Linsmaier and Skoog (LS) medium supplemented with 1 M benzyladenine and 5 M -naphthaleneacetic acid in the dark. Shoots were regenerated from the callus lines on LS medium supplemented with 20 M thidiazuron and 0.1 M -naphthaleneacetic acid under a high-intensity light condition (450 mol m–2 s–1). The regenerated shoots were rooted on LS medium without growth regulators to obtain fully developed plants. We succeeded in transforming Atriplex gmelini from callus lines using Agrobacterium tumefaciens.  相似文献

Using isoenzyme-specific antisera, we have studied the distribution of protein kinase C isoforms in fetal mouse organs at the developmental age of 17 days. Two different sets of antibodies, produced by different manufacturers, were employed in this study. The specificity of the antisera was tested by immunoblotting experiments using whole fetal mouse extracts. Immunohistochemistry was carried out by means of an alkaline phosphatase-conjugated secondary antibody. Analysis of fetal mouse longitudinal cryostat sections stained with the antibodies demonstrated a distinct distribution of protein kinase C isoforms in the tissues. Protein kinase C- and C-I were present in all tissues examined, whereas the C-II isoform was absent in the lung and the liver. Protein kinase C- was identified in brain, spinal ganglia, and adrenal gland. The C- isoenzyme was abundantly expressed in spinal ganglia and in the smooth muscle cells of the bronchial wall. Antisera to C- and C- isoforms heavily stained liver, kidney, and spinal ganglia, whereas the C- isozyme was mainly detected in brain, stomach and kidney. Thus, protein kinase C-, C-I, C-II, C-, C- and C- were the isoforms present in many of the organs investigated. The two sets of antibodies gave slightly different results that might be ascribed to the different epitopes recognized by the antisera. One set of antisera was employed to investigate the distribution of the isoforms in selected organs from an earlier developmental age (15 days) and from adult animals. Occupational exposure to beryllium (Be) and Be compounds occurs in a wide range of industrial processes. A large number of workers are potentially exposed to this metal during manufacturing and processing, so there is a concern regarding the potential carcinogenic hazard of Be. Studies were performed to determine the carcinogenic potential of beryllium sulfate (BeSO4) in cultured mammalian cells. BALB/c3T3 cells were treated with varying concentrations of BeSO4 for 72 h and the transformation frequency was determined after 4 weeks of culturing. Concentrations from 50–200 g BeSO4/ml, caused a concentrationdependent increase (9–41 fold) in transformation frequency. Nontransformed BALB/c3T3 cells and cells from transformed foci induced by BeSO4 were injected into both axillary regions of nude mice. All ten Beinduced transformed cell lines injected into nude mice produced fibrosarcomas within 50 days after cell injection. No tumors were found in nude mice receiving nontransformed BALB/c3T3 cells 90 days postinjection. Gene amplification was investigated in Kras, cmyc, cfos, cjun, csis, erbB2 and p53 using differential PCR while random amplified polymorphic DNA fingerprinting was employed to detect genomic instability. Gene amplification was found in Kras and cjun, however no change in gene expression or protein level was observed in any of the genes by Western blotting. Five of the 10 transformed cell lines showed genetic instability using different random primers. In conclusion, these results indicate that BeSO4 is capable of inducing morphological cell transformation in mammalian cells and that transformed cells induced by BeSO4 are potentially tumorigenic. Also, cell transformation induced by BeSO4 may be attributed, in part, to the gene amplification of Kras and cjun and some BeSO4induced transformed cells possess neoplastic potential resulting from genomic instability.  相似文献

Summary The seasonal progression of phenophases in 21 shrub species of the Chilean matorral was analyzed. Five modules or basic units that are responsible for the aboveground architecture of the plants were characterized. These modules appear to be organized in seven different spatial arrangements. In drought-deciduous species a module type with an absolute short shoot with limited apical growth, leafy or spiny, predominated. In evergreen species long shoot and temporal leafy short shoot module types were more frequent. The spatial arrangement of morphologically different modules and the temporal sequence of their formation allow a dynamic interpretation of the modular architecture of the plants.  相似文献

Axillary shoot induction and plant regeneration were obtained in Plantago ovata. The optimum medium for inducing axillary shoots was Murashige & Skoog (MS) medium [5] supplemented with 4.6 M kinetin and 0.05 M NAA. Rooting of shoots was best on half-strength MS medium containing 5.0 M IBA and 0.05 M kinetin. The regenerated plants were similar to the control plants in karyotypic and phenotypic details.  相似文献

Zusammenfassung Unter den Cornealinsen des Komplexauges von Stylops befindet sich ein Kristallkegel vom pseudoconen Typ, der von zahlreichen Pigmentzellen umhüllt wird. An seinem proximalen Ende liegen 6 meist pigmentfreie Zellen (Sempersche Zellen).Das Ommatidium besteht aus etwa 60 Retinulazellen. Ihre distal kranzartig miteinander verbundenen Mikrovillisäume bilden ein einziges offenes Rhabdom, das extrazelluläres (?) granuläres Material und die Basis der Semperschen Zellen umgibt. Stellenweise wird das Rhabdom samt granulärem Material von homogen erscheinenden distalen Ausläufern einzelner Retinulazellen überlagert. Proximad zerfällt das Rhabdom zunehmend in kleinere Rhabdomteile. Im zentralen Teil des Ommatidiums liegen 1–2 auffallend große Retinulazellen, die meist weniger elektronendicht erscheinen und kleinere Pigmentgrana haben.Die einzelnen Ommatidien werden von ungemein zahlreichen, sehr pigmentarmen Stützzellen umhüllt. Diese werden — wie die basalen Teile der Retinulazellen — teilweise durch Gliazellfortsätze isoliert.Bei Stylops, einem Vertreter der Strepsipteren, handelt es sich nicht um ocelläre Komplexaugen (Strohm, 1910), auch nicht um eucone Ommatidien (Kinzelbach, 1967), sondern um Ommatidien vom pseudoconen Typ. Zumindest der Bau des Rhabdoms ähnelt dem des Larvenauges (Stemma), dessen rezeptorischer Teil entgegen den Annahmen früherer Autoren in der Imago nicht reduziert wird.
On the fine structure of the compound eye of Stylops spec. (Insecta, Strepsiptera)
Summary In the compound eye of Stylops a crystalline cone of the pseudocone type is found beneath the corneal lens. It is enveloped by several pigment cells. At the proximal part of the cone there are 6 cells (Semper cells) mostly pigment-free.The ommatidium consists of approximately 60 retinula cells. Their rhabdomeres distally rim-like connected to another form a single open rhabdom which encircles extracellular granular material as well as the bases of the Semper cells. Here and there the rhabdom plus granular material is overlain with distal protrusions of single retinula cells which appear to be homogeneous. Towards the proximal part the rhabdom increasingly divides up into smaller rhabdomal segments. One or two conspicuous large retinula cells were found in the central part of the ommatidium, appearing to be less electron-dense and containing pigment granules of a smaller size. Each ommatidium is surrounded by numerous cells (Stützzellen) lacking in pigment. These cells are partially insulated from another—as well as the basal parts of retinula cells—by protrusions of glia cells.Our investigations show that the eyes of Stylops (as a representative of Strepsiptera) are not of the ocellar complex eye type. At least the structure of the rhabdom resembles to that of the larval eye (stemma), the receptor part of which is not reduced in the imago.

Calcium channel subunits have profound effects on how 1 subunits perform. In this article we summarize our present knowledge of the primary structures of subunits as deduced from cDNAs and illustrate their different properties. Upon co-expression with 1 subunits, the effects of subunits vary somewhat between L-type and non-L-type channels mostly because the two types of channels have different responses to voltage which are affected by subunits, such as long-lasting prepulse facilitation of 1C (absent in 1E) and inhibition by G protein dimer of 1E, absent in 1C. One subunit, a brain 2a splice variant that is palmitoylated, has several effects not seen with any of the others, and these are due to palmitoylation. We also illustrate the finding that functional expression of 1 in oocytes requires a subunit even if the final channel shows no evidence for its presence. We propose two structural models for Ca2+ channels to account for 1 alone channels seen in cells with limited subunit expression. In one model, dissociates from the mature 1 after proper folding and membrane insertion. Regulated channels seen upon co-expression of high levels of would then have subunit composition 1. In the other model, the chaperoning remains associated with the mature channel and 1 alone channels would in fact be 1 channels. 1,3-Galactosylation of radiolabelled bi-antennary acceptors Gal1-4GlcNAc1-3(Gal1-4GlcNAc1-6)Gal-R (R=1-OH, 1-4GlcNAc or 1-4Glc) with bovine thymus 1,3-galactosyltransferase was studied. At all stages of the reactions the three acceptors reacted faster at the 1 6 linked arm than at the 1 3 linked branch. Hence, in addition to the doubly 1,3-galactosylated products, practically pure Gal1-4GlcNAc1-3(Gal1-3Gal1-4GlcNAc1-6)Gal-R could be obtained from the three acceptors in reactions that had proceeded to near completion. The conformational properties of the oligosaccharide chain of GM1 ganglioside containingN-glycolyl-neuraminic acid, -Gal-(1-3)--GalNAc-(1-4)-[-Neu5Gc-(2-3)]--Gal-(1-4)--Glc-(1-1)-Cer, were studied through NMR nuclear Overhauser effect investigations on the monomeric ganglioside in dimethylsulfoxide, and on mixed micelles of ganglioside and dodecylphosphocholine in water. Several interresidual contacts for the trisaccharide core--GalNAc-(1-4)-[-Neu5Gc-(2-3)]--Gal-were found to fix the relative orientitation of the three saccharides, while the glycosidic linkage of the terminal -Gal-was found to be quite mobile as the -Gal-(1-3)--GalNAc-disaccharide exists in different conformations. The role of -tocopherol uptake system in human erythrocyte in the uptake of plasma -tocopherol has been suggested. However no information is available on -tocopherol uptake activity of human erythrocytes in the presence of high levels of D-glucose which is known to lead to pathological alterations in different cells including human erythrocytes. Therefore, in order to examine the effect of D-glucose on the binding of -tocopherol to human erythrocytes, the binding characteristics of -tocopherol to these cells were established first. Binding of [3H]-tocopherol to human erythrocytes was both saturable and specific. Scatchard analysis of -tocopherol binding to these cells showed the presence of two independent classes of binding sites with widely different affinities. The high affinity binding sites had a dissociation constant (Kd1) of 90 nM with a binding capacity (n1) of 900 sites per cell, whereas the low affinity binding sites had a dissociation constant (Kd2) of 5.2 M and a binding capacity (n2) of 105,400 sites per cell. Trypsin treatment abolished all the -tocopherol binding activity. Competition for the binding of -tocopherol to human erythrocytes was effective with other homologues of -tocopherol (-tocopherol, -tocopherol and -tocopherol) and their potency was almost equal to -tocopherol itself. The order of preference was -tocopherol > -tocopherol -tocopherol -tocopherol. Incubation of human erythrocytes with various concentrations of D-glucose did not affect -tocopherol uptake activity. Our data demonstrate the presence of an -tocopherol uptake system in human erythrocytes and that the -tocopherol uptake activity is not modulated by the presence of D-glucose.  相似文献

Summary Antisera against oLH1, oLH and hFSH were used to localize gonadotropic cells in the pars distalis of Cercopithecus aethiops and Papio hamadryas. Three separate cell types were observed for FSH and LH: 85% of immunohistochemically identified gonadotropic cells reacted to all the various antisera; 10% reacted with the anti-LH antibody only; and 5% with the anti-hFSH antibody only. Comparisons between adjacent serial sections treated with various antisera, other than anti-gonadotropic hormones, demonstrated that the gonadotropic cells of these monkeys did not respond to these antisera.
Résumé Des anticorps anti-LH ovine, anti-LH ovine et anti-FSH humaine ont été utilisés pour localiser les cellules gonadotropes dans la pars distalis de l'hypophyse des Singes Cercopithecus aethiops et Papio hamadryas. Trois catégories cellulaires distinctes, réagissant avec des anticorps anti-hormones gonadotropes, ont été observées. 85% des cellules immunoréactives identifiées en tant que cellules gonadotropes réagissent simultanément avec les différents anticorps mentionnés; 10% des cellules gonadotropes réagissent seulement avec l'anticorps anti-oLH et 5% de ces cellules seulement avec l'anticorps anti-hFSH. La comparaison avec des coupes adjacentes traitées par divers anticorps autres que les anticorps anti-gonadotropines prouve que les cellules gonadotropes de ces Singes ne réagissent jamais simultanément avec l'un ou l'autre de ces anticorps.

Zusammenfassung Es wird über Laboratoriums-Versuche mit 3 verschiedenen Typen von Bacillus thuringiensis-Präparaten an Honigbienen berichtet: (a) mit dem Sporen- Endotoxin-Komplex, (b) mit vegetativen Zellen und (c) mit von diesen produziertem wasserlöslichem thermostabilem Exotoxin. Während Sporen-Endotoxin-Präparate (geprüft an 5 Varietäten) in den für die Bekämpfung von Lepidopteren-Raupen benutzten Dosierungen ungefährlich sind, wirken hohe Dosen toxisch, wenn sie im Futtersaft adulten Bienen appliziert werden. Ein durch vegetative Zellen bedingter nachteiliger Einfluß auf Bienen wurde nicht beobachtet. Das Exotoxin welches von bestimmten Varietäten (z.B. var. thuringiensis) in die Kulturflüssigkeit abgegeben wird, wirkt auf Bienen giftig, wenn es im Futtersaft und als Konzentrat verfüttert wird.
Summary In laboratory trials the effects of three types of preparations of Bacillus thuringiensis were tested on adult honey bees (Apis mellifera L.): (a) the spore-endotoxin-complex, (b) vegetative cells, and (c) the exotoxin. Though spore-endotoxin-preparations (of 5 varieties) were harmless to bees in dosage used for biological control, higher dosages applied in sugar solution were toxic. By using a medium concentration of vegetative cells no disease symptoms could be observed on bees. But the water-soluble thermostable factor, the so-called exotoxin, which is produced by the vegetative cells of several varieties (for example var. thuringiensis), was toxic to bees after feeding. The consequences of these observations are discussed.

Zusammenfassung Die Retinula im Ommatidium der Mehlmotte besteht aus einer wechselnden Anzahl (9–12, meist 11) langgestreckter, prismatischer Sinneszellen. Außerdem enthält jede Retinula nahe der Basalmembran im Zentrum zwischen diesen distalen Retinulazellen noch eine basale Retinulazelle. Die Längsachse der Retinula wird von der Achsenstruktur eingenommen, die aus Mikrovilli besteht. Ihr distaler Teil ist der Achsenfaden, der breitere, proximale Teil bildet das Rhabdom. Dieses erscheint im Querschnitt meist vierstrahlig gelappt, da seine Außenseite in Längsrichtung tief gekehlt ist. Der Rhabdomquerschnitt gliedert sich in mehrere Schöpfe parallel angeordneter Mikrovilli (Rhabdomsektoren); jeder Rhabdomsektor besteht aus 1 oder 2 Rhabdomeren. Die basale Retinulazelle entsendet einen kleinen Schopf von Mikrovilli in die proximale Spitze des Rhabdoms. Die distalen Retinulazellen setzen sich proximal in Neuriten fort, welche sich in Einkehlungen der basalen Retinulazelle bzw. der Tracheenendzelle einschmiegen. Jeweils eine Tracheole durchbricht zusammen mit dem Neuritenstrang einer Retinula die Basalmembran; sie verzweigt sich distal zu ca. 30 Tracheolen, die die Retinula umhüllen.Die Kristallkegelzellen grenzen distal an die Cornea; proximal laufen die Kristallkegelzellen eines Ommatidiums in einen gemeinsamen Fortsatz aus, der zwischen den Retinulazellen unmittelbar am Achsenfaden endet. — Nur das helladaptierte Auge wurde untersucht. Hierbei erscheint im distalen Teil der Retinula nur der Achsenfaden lichtdurchlässig, das Cytoplasma der Retinulazellen hingegen von Pigmentgrana durchsetzt und für Licht undurchlässig.
Fine structure of the eye of the meal moth, Ephestia kuehniella Zeller (Lepidoptera, Pyralididae)
Summary In each ommatidium of the meal moth a retinula is formed from a varying number (9–12, mostly 11) of elongated, prismatic sense cells. In addition, a basal retinular cell is situated near the basement membrane in the center of the other (distal) retinular cells. The axis of the retinula is occupied by many microvilli forming the axial structure, the distal section of which is the slender axial thread. Proximally, the axial structure widens (to 8.5 m instead of 1 m in diameter) and is now called rhabdom. Cross sections of the rhabdom mostly look like a petaloid with four petals; this figure is due to longitudinal infoldings along the length of the rhabdom surface. The rhabdom cross section is subdivided into several brushes of microvilli (rhabdom sectors), each one being characterized by an approximately parallel arrangement of its microvilli. One rhabdom sector may be composed of one or two rhabdomeres respectively.The basal retinular cell participates in rhabdom formation through a small brush of microvilli at the proximal end of the rhabdom. Proximally, the distal retinular cells taper into slender neurites which are embedded in grooves at the surface of the basal retinular cell and the tracheal end cell respectively. One tracheole piercing the basement membrane together with the neurites of one retinula branches into about 30 tracheoles surrounding the retinula.The crystalline cone cells touch the cornea; proximally, their cytoplasm forms a point which eventually terminates amongst the distal tips of the retinular cells, immediately at the axial thread.—Our work was restricted to light adapted eyes; in this condition, light transmission in the distal part of the retinula seems to be blocked by retinular cell pigment except inside the axial thread.

Summary The effect of a non-indole compound, phenylacetic acid (PAA), on the induction of somatic embryogenesis in tissue cultures of geranium (Pelargonium x hortorum Bailey cv. Scarlet Orbit Improved) was investigated. Hypocotyl explants derived from young, dark-grown seedlings were cultured on Murashige and Skoog (1962) medium (MS) supplemented with PAA or IAA (0.01–120 M) alone or in combination with BAP (8 M). Somatic embryogenesis was induced by both PAA and IAA at 0.01–20 M with 8 M BAP, however, the optima differed considerably for the two compounds. Maximal activity of IAA for somatic embryogenesis was found at 0.1–2.5 M, whereas PAA gave best results at 10 and 20 M under identical culture conditions. Summary Transforming growth factor- (TGF-) is a biologically active polypeptide present in normal tissues as well as transformed cells. Two structurally related forms of this peptide are TGF- 1 and TGF- 2. Using freshly isolated cardiomyocytes and non-myocyte heart cells, and a [32P]-labelled cDNA probe to human TGF- 1, we demonstrated that mRNA for TGF- 1 could be detected only in the nonmyocyte fraction of heart cells. In the present study, the distribution of TGF- 1 in the heart was determined by immunofluorescence staining by use of a polyclonal antibody to porcine TGF- 1 in cryostat sections of rat heart. Immunofluorescence staining was intense around the blood vessels and radially diffuse in the surrounding myocardium.  相似文献

Summary The seven possible primary trisomics of Petunia (2 n= 14) located in the progenies of triploid, hypertriploid and hypotriploid plants were distinguished from one another and from diploid on the basis of cytological and morphological criteria. They were provisionally named as Oval, Semi, Slender, Pseudonormal, Arrow, Narrow and Giant. In three of the trisomics, the extra chromosome was identified for the first time at pachytene stage. Postpachytene studies revealed no precise relationship between the length of extra chromosome and the frequency of multiple association.  相似文献
