Leaf gas exchange, chlorophyll fluorescence and activities of antioxidant enzymes were studied in two pepper (Capsicum annuum L.) cultivars grown under drought (D) and heat (H), as well as under drought and heat in combination (HD). The drought-tolerant cv. Zhengjiao 13 exhibited greater net photosynthetic rate (PN) and cytochrome respiratory pathway activity (RSHAM), and lower contents of superoxide radical and hydrogen peroxide, as compared to the drought-sensitive cv. Longkouzaojiao. The PN and RSHAM decreased and ROS production increased under D and HD in both cultivars. As compared to the Longkouzaojiao, Zhengjiao 13 retained higher non-photochemical quenching (NPQ), photorespiration rate (RL), and alternative respiratory pathways (RKCN) under D and HD. Drought increased the superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities in the cytosol, chloroplasts and mitochondria in the two cultivars. Although SOD and APX activities decreased in Longkouzaojiao under HD, SOD activity increased in Zhengjiao 13. There was no H-induced reactive oxygen species production due to increase of RL, NPQ, RSHAM, RKCN and activities of SOD and APX. However, H slightly decreased the PN. The results indicated that HD was more detrimental than either stress alone.  相似文献

In this work, the injuries caused by clethodim herbicide application as well as the use of exogenous salicylic acid (SA) as a protective agent against clethodim in Zea mays leaves were examined. Although the target for clethodim is the inhibition of acetyl coenzyme A carboxylase (ACCase) which is the key enzyme for fatty acid biosynthesis, it can indirectly affect the photosynthetic machinery, gaseous exchange and some biochemical parameters. Clethodim application caused chlorosis and yellowing of leaf-tip parts. Higher doses caused browning or reddening of leaves and sometimes dead parts of the leaf margins were observed. The rate of photosynthesis was significantly lowered and the pigments content was highly reduced as a response to clethodim spraying. Moreover, other gas-exchange properties were altered. Furthermore, accumulation of high amounts of carbohydrates, proteins and proline were detected. SA spraying three days prior clethodim application caused partially or totally disappearance of clethodim injuries and kept the leaves similar to those of control. Improved photosynthesis and enhanced pigments content were observed in leaves treated with SA. Other analyzed parameters showed values similar to those of the corresponding control. From the experimental work, an evidenced role of SA working against clethodim effects was suggested and discussed in this paper.  相似文献

Trigonella foenum graecum seed powder (TSP) and sodium orthovanadate (SOV) have been reported to have antidiabetic effects. However, SOV exerts hypoglycemic effects at relatively high doses with several toxic effects. We used low doses of vanadate in combination with TSP and evaluated their antidiabetic effects on antioxidant enzymes and membrane-linked functions in diabetic rat brains. In rats, diabetes was induced by alloxan monohydrate (15 mg/100 g body wt.) and they were treated with 2 IU insulin, 0.6 mg/ml SOV, 5% TSP and a combination of 0.2 mg/ml SOV with 5% TSP for 21 days. Blood glucose levels, activity of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), Na+/K+ ATPase, membrane lipid peroxidation and fluidity were determined in different fractions of whole brain after 21 days of treatment. Diabetic rats showed high blood glucose (P < 0.001), decreased activities of SOD, catalase and Na+/K+ ATPase (P < 0.01,P < 0.001 andP < 0.01), increased levels of GPx and MDA (P < 0.01 andP < 0.001) and decreased membrane fluidity (P < 0.01). Treatment with different antidiabetic compounds restored the above-altered parameters. Combined dose ofTrigonella and vanadate was found to be the most effective treatment in normalizing these alterations. Lower doses of vanadate could be used in combination with TSP to effectively counter diabetic alterations without any toxic effects.  相似文献

通过田间测坑试验研究了长期处于不同土壤水分状况下花生和早稻叶片气体交换的一些特点.结果表明,花生分枝期轻度和中度水分胁迫使气孔导度(Gs)和蒸腾速率(Tr)略有下降,净光合速率(Pn)和叶片水分利用效率(WUE)减小,轻度水分胁迫Gs/Tr略有上升而中度胁迫Gs/Tr变小.花生结荚期轻度和中度水分胁迫都使Gs、Tr、Gs/Tr和Pn显著降低,WUE大幅度上升.花生结荚期明显受土壤水分胁迫影响.早稻灌浆期轻度和中度水分胁迫Gs、Tr和Gs/Tr变化不显著,Pn和WUE增加,并且轻度水分胁迫下籽粒产量增加.Gs和Gs/Tr变化情况相结合可以作为作物水分胁迫程度的一个参考指标,即如果Gs和Gs/Tr同时下降则作物已经受到水分胁迫影响.  相似文献

Thermopsis turcica is distributed naturally in saline soils. Interestingly, how T. turcica can live in harsh salt conditions is unknown. To study its defense responses under salinity, T. turcica was grown in a medium containing 100 and 200 mM NaCl for 7 and 14 days. Physiological parameters, ion contents, reactive oxygen species accumulation, activities of antioxidant enzymes/isozymes, NADPH oxidase enzyme/isozyme, lipid peroxidation (TBARS) and osmolyte contents were investigated. Stress caused a rapid decline in relative growth rate, relative water content and chlorophyll fluorescence (F v/F m) under both NaCl treatments. These traits were more suppressed at 200 mM NaCl. The decline in osmotic potential (Ψ Π) with salinity increased the gradient for water flux into the cell and assisted in turgor maintenance. The increased membrane permeability under stress caused the entrance of excess Na+ and K+ into the cell. Stress decreased superoxide dismutase, catalase and peroxidase after 14 days of growth in 200 mM NaCl, whereas glutathione reductase (GR) increased throughout the experiment. While ascorbate peroxidase (APX) increased by 44 % at 7 days, it decreased after 14 days exposure to 200 mM NaCl. 200 mM NaCl caused the highest increase in TBARS at 14 days, indicating a decrease in OH· scavenging activity. Increasing concentrations of salinity caused an increase in glycine betaine (GB) and choline (Cho), though an increase in proline was only observed at 200 mM NaCl for 14 days. Briefly, H2O2 was more efficiently eliminated in 100 mM-treated plants by the ascorbate–glutathione cycle in which APX acts a strong catalyst together with GR. Gross photosynthetic capacity (P G ) of greenhouse-grown tomato plants (Lycopersicon esculentum Mill.) decreased as the leaf aged. The P G of the 10th, 15th and 18th leaves from the top was only 76, 37, and 18 % of P G of the 5th leaf, respectively. Quantum yield (Y Q ) and dark respiration rate (R D ) were also lower in older leaves than in the younger ones. Net photosynthetic rate (P G ) was apparent in young fruits (about 10 g FM) or young petioles but no P N was found in large fruits (40 g or more FM) and stems because of high R D . Both P G and R D were lower in older fruits and petioles or in lower parts of the stem compared to the younger ones or upper parts of stem. A sharp decrease in chlorophyll (Chl) content was only measured in the senescing 18th leaf. The Chl content in petioles, stems and fruits was proportional to P G . Decreases in P G of older leaves were attributed to decreases in content rather than activity of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) since soluble protein content was lower in older leaves than in the younger ones but the specific activity (activity per unit of protein) of RuBPCO was not so. The estimated values of P N of the 10th, 15th and 18th leaves inside the canopy were only 50, 21, and 7 % of that in the 5th leaf. Therefore, leaves below the 18th can be removed in order to ensure a good air circulation and prevent diseases. The significance of photosynthesis in fruit, stem and petioles is not negligible because photosynthesis re-fixes the respired CO2. The antioxidant status of birch and ginkgo leaves during autumnal senescence was characterized by the activities of catalase (CAT), peroxidase (POD), ascorbate peroxidase (APX) and superoxide dismutase (SOD). The contents of leaf H2O2 and ascorbate were used as indicators of oxidative stress. Degradation of chlorophyll (chl) during natural senescence was not accompanied either by an increase of H2O2 or by a decrease of reduced ascorbate. A transient decrease of reduced ascorbate in ginkgo and birch leaves in early senescence was accompanied by CAT inactivation. The activity of ionically-bound PODs was stimulated in late senescence in both species, when more than 30% of chl was degraded. Induction of MnSOD in both species and new isoforms of CuZnSOD in birch in late senescence was accompanied by the disappearance of other CuZnSOD isoforms in birch and FeSOD in ginkgo. The role of antioxidative enzymes in keeping ascorbate reduced and endogenous H2O2 at low levels in senescent leaves of deciduous trees was discussed.  相似文献

This paper describes an assay for the determination of glyphosate (GLYP), glyphosate metabolites [(aminomethyl) phosphonic acid] (AMPA), and glufosinate (GLUF) in human serum. After protein precipitation using acetonitrile and solid-phase extraction, serum samples were derivatized and analyzed by gas chromatography-mass spectrometry (GC-MS). The assay was linear over a concentration range of 3-100.0 microg/ml for GLYP, AMPA, and GLUF. The overall recoveries for the three compounds were >73%. The intra- and inter-day variations were <15%. Precision and accuracy were 6.4-10.6% and 88.2-103.7%, respectively. The validated method was applied to quantify the GLYP and AMPA content in the serum of a GLYP-poisoned patient. In conclusion, the method was successfully applied for the determination of GLYP and its metabolite AMPA in serum obtained from patient of GLYP-poisoning.  相似文献

We investigated the effects of brushing on the chilling tolerance and metabolism of nonstructural carbohydrates (soluble sugars and starch) in tomato leaves before, during and after a chilling stress. Tomato plants ( Lycopersicon esculentum Mill. cv. Caruso) were cultivated either without mechanical stress application (control plants) or with daily brushing treatments for 15 days (brushed plants), prior to a 7-day chilling treatment (8/5°C day/night). Brushing resulted in shorter plants with a 34% reduction in leaf dry weight per area and a 59% reduction of soluble sugars and starch, on a dry weight basis. The sugar to starch ratio was not affected by brushing. A greater chilling tolerance in the brushed plants was demonstrated by the maintenance of a significantly higher PSII efficiency in brushed plants (42%) compared to that of the control plants (30%) after 7 days of chilling treatment, less visible damage to the leaf tissue, and a more rapid resumption of growth during 3 days of recovery as compared to control plants. During the chilling treatment levels of soluble sugars per leaf dry weight increased 15-fold in the brushed plants and 5-fold in control plants. In the present study we have demonstrated that brushing can increase chilling tolerance in tomato plants. The observed differences in chilling tolerance and concentration of soluble sugars in the leaves may indicate an involvement of soluble sugar levels in acclimation to chilling.  相似文献

The relationship between loss of ribulose-1,5-bisphosphate carboxylase (RuBPCase) and the decline in photosynthesis during the senescence of barley primary leaves was assessed. Loss of RuBPCase accounted for about 85% of the decrease in soluble protein. RuBPCase was highly correlated with in vitro RuBPCase activity (r = 0.95) and gross photosynthesis (r = 0.96). However, the rate of photosynthesis per milligram RuBPCase increased during the early stages of leaf senescence. The concentration of nonreducing sugars was negatively correlated (1% level) with photosynthesis. Free α-amino N, in contrast to nonreducing sugars, declined markedly during senescence. A decrease in chlorophyll and an increase in in vitro protease activity was observed, but these changes did not appear to be closely related to the decline in photosynthesis and RuBPCase. Mesophyll resistance increased at the same rate that photosynthesis and RuBPCase declined. Stomatal resistance increased more rapidly than mesophyll resistance and accounted for about 24% of the total increase in resistance to CO2 diffusion. The concentration of CO2 in the intercellular air spaces decreased during the last stage of senescence. Although loss of RuBPCase probably is the primary event responsible for the decline in photosynthesis during leaf senescence, other factors such as in vivo regulation and stomatal aperture must also be considered.  相似文献

Following a 2-week treatment with glyphosate [N-(phosphonomethyl)glycine] changes in peroxidase (EC 1.11.1.7) and polyphenol oxidase (PPO; EC 1.14.18.1) activities of yellow nutsedge ( Cyperus esculentus L.) plants, were determined. Glyphosate caused significant increases of both activities. Isoelectric focusing gave 3 species (F1, F2 and F3) of peroxidase activity, at pl 3.8, 4.4 and 4.8, and 4 species (Fa, Fb, Fc and Fd) of PPO activity at pl 7.0, 7.5, 7.8 and 9.5. The activity of the 4 active forms of PPO increased with increasing glyphosate dose up to 10−2 M . The effect of the herbicide on the 3 fractions with peroxidase activity was to change their relative activities. Highest F1 activity was found in control plants whereas the F2 fraction was the predominant form in the plants treated with glyphosate at 10−2 M and the highest F3 activity occurred in plants treated with 5 × 10−3 M glyphosate. The increased PPO activity could produce phytotoxic o -quinones, and variations in peroxidase isoenzymes activity could enhance isoperoxidases with lignin biosynthetic activity.  相似文献

Diet profoundly influences the behavior of animals across many phyla. Despite this, most laboratories using model organisms, such as Drosophila, use multiple, different, commercial or custom-made media for rearing their animals. In addition to measuring growth, fecundity and longevity, we used several behavioral and physiological assays to determine if and how altering food media influence wild-type (Canton S) Drosophila melanogaster, at larval, pupal, and adult stages. Comparing 2 commonly used commercial food media we observed several key developmental and morphological differences. Third-instar larvae and pupae developmental timing, body weight and size, and even lifespan significantly differed between the 2 diets, and some of these differences persisted into adulthood. Diet was also found to produce significantly different thermal preference, locomotory capacity for geotaxis, feeding rates, and lower muscle response to hormonal stimulation. There were no differences, however, in adult thermal preferences, in the number or viability of eggs laid, or in olfactory learning and memory between the diets. We characterized the composition of the 2 diets and found particularly significant differences in cholesterol and (phospho)lipids between them. Notably, diacylglycerol (DAG) concentrations vary substantially between the 2 diets, and may contribute to key phenotypic differences, including lifespan. Overall, the data confirm that 2 different diets can profoundly influence the behavior, physiology, morphology and development of wild-type Drosophila, with greater behavioral and physiologic differences occurring during the larval stages.  相似文献

BACKGROUND: Rheumatoid arthritis (RA) is a prevalent and debilitating disease that affects the joints. Infiltration of blood-derived cells in the affected joints upon activation generate reactive oxygen/nitrogen species, resulting in an oxidative stress. One approach to counteract this oxidative stress is the use of antioxidants as therapeutic agents. OBJECTIVES: Kalpaamruthaa (KA), a modified indigenous Siddha preparation constituting Semecarpus anacardium nut milk extract (SA), Emblica officinalis (EO) and honey was evaluated for its synergistic antioxidant potential in adjuvant induced arthritic rats than sole SA treatment. MATERIALS AND METHODS: Levels/activities of reactive oxygen species (ROS)/reactive nitrogen species (RNS), myeloperoxidase, lipid peroxide and enzymic and non-enzymic antioxidants were determined in control, arthritis induced, SA and KA treated (150 mg/kg b.wt.) animals. RESULTS AND CONCLUSION: The levels/activities of ROS/RNS, myeloperoxidase and lipid peroxide were increased significantly (p<0.05) and the activities of enzymic and non-enzymic antioxidants were in turn decreased in arthritic rats, whereas these changes were reverted to near normal levels upon SA and KA treatment. KA showed an enhanced antioxidant potential than sole treatment of SA in adjuvant induced arthritic rats. KA via enhancing the antioxidant status in adjuvant induced arthritic rats than sole SA treatment proves to be an important therapeutic modality in the management of RA and thereby instituting the role of oxidative stress in the clinical manifestation of the disease RA. The profound antioxidant efficacy of KA than SA alone might be due to the synergistic action of the polyphenols such as flavonoids, tannins and other compounds such as vitamin C and hydroxycinnamates present in KA.  相似文献

Vanadium compounds are potent in controlling elevated blood glucose levels in experimentally induced diabetes. However the toxicity associated with vanadium limits its role as therapeutic agent for diabetic treatment. A vanadium compound sodium orthovanadate (SOV) was given to alloxan-induced diabetic Wistar rats in lower doses in combination withTrigonella foenum graecum, a well-known hypoglycemic agent used in traditional Indian medicines. The effect of this combination was studied on lens morphology and glucose metabolism in diabetic rats. Lens, an insulin-independent tissue, was found severely affected in diabetes showing visual signs of cataract. Alterations in the activities of glucose metabolizing enzymes (hexokinase, aldose reductase, sorbitol dehydrogenase, glucose-6-phosphate dehydrogenase) and antioxidant enzymes (glutathione peroxidase, glutathione reductase) besides the levels of related metabolites, [sorbitol, fructose, glucose, thiobarbituric acid reactive species (TBARS) and reduced glutathione (GSH)]were observed in the lenses from diabetic rats and diabetic rats treated with insulin (2 IU/day), SOV (0.6 mg/ml),T. f. graecum seed powder (TSP, 5%) and TSP (5%) in combination with lowered dose of vanadium SOV (0.2 mg/ml), for a period of 3 weeks. The activity of the enzymes, hexokinase, aldose reductase and sorbitol dehydrogenase was significantly increased whereas the activity of glucose-6-phosphate dehydrogenase, glutathione peroxidase and glutathione reductase decreased significantly in lenses from 3 week diabetic rats. Significant increase in accumulation of metabolites, sorbitol, fructose, glucose was found in diabetic lenses. TBARS measure of peroxidation increased whereas the levels of antioxidant GSH decreased significantly in diabetic condition. Insulin restored the levels of altered enzyme activities and metabolites almost to control levels. Sodium orthovanadate (0.6 mg/ml) andTrigonella administered separately to diabetic animals could partially reverse the diabetic changes, metabolic and morphological, while vanadate in lowered dose in combination withTrigonella was found to be the most effective in restoring the altered lens metabolism and morphological appearance in diabetes. It may be concluded that vanadate at lowered doses administered in combination withTrigonella was the most effective in controlling the altered glucose metabolism and antioxidant status in diabetic lenses, these being significant factors involved in the development of diabetic complications, that reflects in the reduced lens opacity  相似文献

Photosynthesis and nitrogen relationships in leaves of C3 plants   总被引:53,自引:0,他引:53  
Summary The photosynthetic capacity of leaves is related to the nitrogen content primarily bacause the proteins of the Calvin cycle and thylakoids represent the majority of leaf nitrogen. To a first approximation, thylakoid nitrogen is proportional to the chlorophyll content (50 mol thylakoid N mol-1 Chl). Within species there are strong linear relationships between nitrogen and both RuBP carboxylase and chlorophyll. With increasing nitrogen per unit leaf area, the proportion of total leaf nitrogen in the thylakoids remains the same while the proportion in soluble protein increases. In many species, growth under lower irradiance greatly increases the partitioning of nitrogen into chlorophyll and the thylakoids, while the electron transport capacity per unit of chlorophyll declines. If growth irradiance influences the relationship between photosynthetic capacity and nitrogen content, predicting nitrogen distribution between leaves in a canopy becomes more complicated. When both photosynthetic capacity and leaf nitrogen content are expressed on the basis of leaf area, considerable variation in the photosynthetic capacity for a given leaf nitrogen content is found between species. The variation reflects different strategies of nitrogen partitioning, the electron transport capacity per unit of chlorophyll and the specific activity of RuBP carboxylase. Survival in certain environments clearly does not require maximising photosynthetic capacity for a given leaf nitrogen content. Species that flourish in the shade partition relatively more nitrogen into the thylakoids, although this is associated with lower photosynthetic capacity per unit of nitrogen.  相似文献

Tissue antioxidant status may be compromised under conditions of dietary restriction, either as the result of a deficiency in a specific cofactor required by a particular antioxidant enzyme or of more complex alterations of a generalized nature triggered by metabolic responses to starvation. Many similarities exist between insulin-reversible abnormalities in tissue antioxidant enzyme activities seen in experimental diabetes and in animals subjected to food deprivation-induced weight loss which is associated with hypoinsulinemia. The complex alterations in tissue antioxidant enzyme activities resulting from nutritional deficiency states, disease or drug administration may have important clinical consequences. Free radical-related processes have been implicated in the pathology of certain conditions in which weight loss is frequently recommended (e.g., diabetes and atherosclerosis). It will be important to investigate the possible adverse effects of this intervention on the underlying disease process involved. Glutathione-dependent hepatic detoxification processes are impaired under conditions of nutritional deficiency. This finding not only has important clinical implications but the standard practice of fasting small laboratory animals overnight to ensure reliable drug absorption can markedly influence the results of pharmacological/toxicological experiments. Further studies of the influence of nutritional status on free radical-related processes are likely to yield valuable information which may be applicable to a variety of research and clinical problems.  相似文献

Background:

The aim of this study was to evaluate the possible protective effect of sodium selenite on serum, liver, and kidney antioxidant enzymes activities in alloxan-induced type 1 diabetic rats.

Methods:

Forty Sprague-Dawley male rats were randomly divided into four groups; Group one as control, Group two as sham-treated with sodium selenite by 1 mg/kg intraperitoneal (i.p.) injections daily, Group three as diabetic untreated, and Group four as diabetic treated with sodium selenite by 1 mg/kg i.p. injections daily .Diabetes was induced in the third and fourth groups by subcutaneous alloxan injections. After eight weeks the animals were euthanized and livers and kidneys were immediately removed and used fresh or kept frozen until analysis. Before the rats were killed blood samples were also collected to measure glutathione peroxidase (GPX) and catalase (CAT) activities in sera.

Results:

Glutathione peroxidase and CAT activities serum, liver, and kidney were all significantly less in the diabetic rats than in the controls. Sodium selenite treatment of the diabetic rats resulted in significant increases in GPX activity in the kidneys and livers, and CAT activity in the sera and livers.

Conclusions:

Our results indicate that sodium selenite might be a potent antioxidant that exerts beneficial effects on both GPX and CAT activities in alloxan-induced type 1 diabetic rats. Key Words: Diabetes, Rat, Sodium selenite, Antioxidant enzymes activity  相似文献

We hypothesized that salinity and light interactively affect mangroves, such that net photosynthesis, growth, and survivorship rates increase more with increase in light availability at low than high salinity. Using greenhouse and field experiments, we determined that net photosynthesis, growth rates, and size increased more with light at low than high salinity. At high salinity, the ratio of leaf respiration to assimilation increased fourfold, suggesting that salinity may have contributed to declines in net photosynthesis. Stomatal conductance, leaf‐level transpiration, and internal CO2 concentrations were lower at high salinity. Ratios of root mass to leaf mass were higher at high salinity. Stomatal limitations and increased respiratory costs may explain why at high salinity, the seedlings did not respond to increased light availability with increased net photosynthesis. Increased root mass relative to leaf mass suggests that at high salinity, either water or nutrient limitations may have prevented the seedlings from increasing growth with increasing light availability. At both low‐ and high‐salinity zones in the field, seedling survivorship increased with light availability, and the effect of light was stronger at low salinity. However, at low light, survivorship was higher at high than low salinity, indicating that there may be a trade‐off between survivorship and growth. The interactive effects observed in the greenhouse were robust in the field, despite the presence of other factors in the field such as inundation and nutrient gradients and herbivory. This study provides a robust test of the hypothesis that salinity and light interactively effect mangrove seedling performance.  相似文献
