We conducted an experiment to assess the predictive capability of a leaf optical meter for determining leaf pigment status of Acer mono Maxim., A. ginnala Maxim., Quercus mongolica Fisch., and Cornus alba displaying a range of visually different leaf colors during senescence. Concentrations of chlorophyll (Chl) a, Chl b, and total Chl [i.e., Chl (a+b)] decreased while the concentration of carotenoids (Car) remained relatively static for all species as leaf development continued from maturity to senescence. C. alba exhibited the lowest average concentration of Chl (a+b), Chl a, and Car, but the highest relative anthocyanin concentration, while Q. mongolica exhibited the highest Chl (a+b), Chl b, and the lowest relative anthocyanin concentration. A. mono exhibited the highest Chl a and Car concentrations. The relationships between leaf pigments and the values measured by the optical meter generally followed an exponential function. The strongest relationships between leaf pigments and optical measurements were for A. mono, A. ginnala, and Q. mongolica (R 2 ranged from 0.64 to 0.95), and the weakest relationships were for C. alba (R 2 ranged from 0.13 to 0.67). Moreover, optical measurements were more strongly related to Chl a than to Chl b or Chl (a+b). Optical measurements were not related to Car or relative anthocyanin concentrations. We predicted that weak relationships between leaf pigments and optical measurements would occur under very low Chl concentrations or under very high anthocyanin concentrations; however, these factors could not explain the weak relationship between Chl and optical measurements observed in C. alba. Overall, our results indicated that an optical meter can accurately estimate leaf pigment concentrations during leaf senescence — a time when pigment concentrations are dynamically changing — but that the accuracy of the estimate varies across species. Future research should investigate how species-specific leaf traits may influence the accuracy of pigment estimates derived from optical meters.  相似文献

New spectral absorption photometry methods are introduced to estimate chlorophyll (Chl) content of corn leaves by smart phones. The first method acquires light passing through a leaf by smartphone camera, compensating for differences in illumination conditions. In order to improve performance of the method, spectral absorption photometry (SAP) with background illumination has been considered as well. Data were acquired by smartphone camera in Iowa State University maize fields. Various indices were extracted and their correlation with Chl content were examined by Minolta SPAD-502. Hue index in SAP reached R 2 value of 0.59. However, with light-aided SAP (LASAP), R 2 of 0.97 was obtained. Among traits, the vegetation index gave the most accurate indication. We can conclude that the high performance of LASAP method for estimating Chl content, leads to new opportunities offered by smart phones at much lower cost. This is a highly accurate alternative to SPAD meters for estimating Chl content nondestructively.  相似文献

At present, chlorophyll meters are widely used for a quick and nondestructive estimate of chlorophyll (Chl) contents in plant leaves. Chl meters allow to estimate the Chl content in relative units - the Chl index (CI). However, using such meters, one can face a problem of converting CI into absolute values of the pigment content and comparing data acquired with different devices and for different plant species. Many Chl meters (SPAD-502, CL-01, CCM-200) demonstrated a high degree of correlation between the CI and the absolute pigment content. A number of formulas have been deduced for different plant species to convert the CI into the absolute value of the photosynthetic pigment content. However, such data have not been yet acquired for the atLEAF+ Chl meter. The purpose of the present study was to assess the applicability of the atLEAF+ Chl meter for estimating the Chl content. A significant species-specific exponential relationships between the atLEAF value (corresponding to CI) and extractable Chl a, Chl b, Chl (a+b) for Calamus dioicus and Cleistanthus sp. were shown. The correlations between the atLEAF values and the content of Chl a, Chl b, and Chl (a+b) per unit of leaf area was stronger than that per unit of dry leaf mass. The atLEAF value- Chl b correlation was weaker than that of atLEAF value-Chl a and atLEAF value-Chl (a+b) correlations. The influence of light conditions (Chl a/b ratio) on the atLEAF value has been also shown. The obtained results indicated that the atLEAF+ Chl meter is a cheap and convenient tool for a quick nondestructive estimate of the Chl content, if properly calibrated, and can be used for this purpose along with other Chl meters.  相似文献

Leaf senescence can be induced by numerous factors. In order to explore the relationship between root respiration and leaf senescence, we utilized different types of phloem girdling to control the root respiration of Alhagi sparsifolia and its physiological response. Our results showed that both girdling and inhibition of root respiration led to a decline of stomatal conductance, photosynthesis, transpiration rate, chlorophyll (Chl) a, Chl b, carotenoid (Car) content, Chl a/b, Chl/Car, water potential, and Chl a fluorescence, as well as to an increase of abscisic acid (ABA), proline, and malondialdehyde content in leaves and to upregulation of senescence-associated gene expression. Our present work implied that both inhibition of root respiration and girdling can induce leaf senescence. In comparison with phloem girdling, the leaf senescence caused by inhibition of root respiration was less significant. The reason for girdling-induced senescence was ABA and carbohydrate accumulation. Senescence induced by inhibition of root respiration occurred due to leaf water stress resulting from inhibition of water absorption.  相似文献

The present study revealed that Ginkgo biloba and Platanus occidentalis, the most abundant roadside trees in Seoul, grown under polluted environmental conditions, displayed lower contents of total chlorophyll (Chl), carotenoids (Car), and ascorbic acid (AsA) compared to the trees grown under clean conditions. The reduction in Chl, Car, and AsA contents was 59, 53, and 50%, respectively, in G. biloba, contrary to 26, 23, and 24%, respectively, in P. occidentalis. Furthermore, relative ion leakage and leaf temperature was higher in the trees grown under polluted conditions than in those grown under clean conditions. The increase in relative ion leakage and leaf temperature was 58 and 3% for G. biloba and 17 and 4% for P. occidentalis, respectively. Our results, therefore, highlighted the negative impact of urban environmental pollution on the physiological and biochemical parameters in roadside trees.  相似文献

The objective of this study was to investigate a response to low-light environments in hybrids and commercial cultivars of Boehmeria nivea L. Two hybrids (Chuanzhu 11 and Chuanzhu 8) and two commercial cultivars (Chuanzhu 12 and Chuanzhu 6) of ramie were subjected to a shade treatment for 6, 12, and 18 days. The shade treatment led to a significant decrease in some plant traits and fiber yield in four ramie cultivars, whereas their leaf area and plant height increased. In addition, net photosynthesis and stomatal conductance significantly declined in response to shade, while transpiration rate and intercellular CO2 did not significantly change. Moreover, chlorophyll (Chl) and carotenoid (Car) concentration, Chl/Car, and Chl (a+b) per leaf dry mass significantly increased in the response to shade, while the Chl a/b ratio decreased. Furthermore, Chuanzhu 6 and Chuanzhu 11 were more tolerant to shade than Chuanzhu 12 and Chuanzhu 8, thus, they could be potentially used for management practices and breeding programs.  相似文献

The finding of unique Chl d- and Chl f-containing cyanobacteria in the last decade was a discovery in the area of biology of oxygenic photosynthetic organisms. Chl b, Chl c, and Chl f are considered to be accessory pigments found in antennae systems of photosynthetic organisms. They absorb energy and transfer it to the photosynthetic reaction center (RC), but do not participate in electron transport by the photosynthetic electron transport chain. However, Chl d as well as Chl a can operate not only in the light-harvesting complex, but also in the photosynthetic RC. The long-wavelength (Qy) Chl d and Chl f absorption band is shifted to longer wavelength (to 750 nm) compared to Chl a, which suggests the possibility for oxygenic photosynthesis in this spectral range. Such expansion of the photosynthetically active light range is important for the survival of cyanobacteria when the intensity of light not exceeding 700 nm is attenuated due to absorption by Chl a and other pigments. At the same time, energy storage efficiency in photosystem 2 for cyanobacteria containing Chl d and Chl f is not lower than that of cyanobacteria containing Chl a. Despite great interest in these unique chlorophylls, many questions related to functioning of such pigments in primary photosynthetic processes are still not elucidated. This review describes the latest advances in the field of Chl d and Chl f research and their role in primary photosynthetic processes of cyanobacteria.  相似文献

To assess the relationship between chlorophyll (Chl) fluorescence (CF) and photosynthetic pigments, soybean was grown under varying phosphorus (P) nutrition at ambient and elevated CO2 (EC). The EC stimulated, but P deficiency decreased plant height, node numbers, and leaf area concomitantly with the rates of stem elongation, node addition, and leaf area expansion. Under P deficiency, CF parameters and pigments declined except that carotenoids (Car) were relatively stable indicating its role in photoprotection. The CF parameters were strongly related with Chl concentration but not with Chl a/b or Car. However, total Chl/Car showed the strongest association with CF parameters such as quantum efficiency and yield of photosystem II. This relationship was not affected by CO2 treatment. The high correlation between CF and total Chl/Car underscores the significance of the quantification of both, Chl and Car concentrations, to understand the photochemistry and underlying processes of photoprotection and mechanisms of excess energy dissipation in a given environment.  相似文献

The harderian gland is located within the orbit of the eye of most terrestrial vertebrates. It is especially noticeable in rodents, in which it synthesises lipids, porphyrins, and indoles. Various functions have been ascribed to the harderian gland, such as lubrication of the eyes, a site of immune response, and a source of growth factors. Carbonic anhydrases (CAs) are zinc-containing metalloenzymes that catalyse the reaction \( {\text{CO}}_{2} + {\text{H}}_{2} {\text{O}} \Leftrightarrow {\text{H}}^{ + } + {\text{HCO}}_{3}^{ - } \). They are involved in the adjustment of pH in the secretions of different glands. Thirteen enzymatically active isozymes have been described in the mammalian α-CA family. Here, we first investigated the mRNA expression of all 13 active CAs in the mouse harderian gland by quantitative real-time PCR. Nine CA mRNAs were detectable in the gland. Car5b and Car13 showed the highest signals. Car4, Car6, and Car12 showed moderate expression levels, whereas Car2, Car3, Car7, and Car15 mRNAs were barely within the detection limits. Immunohistochemical staining was performed to study the expression of Car2, Car4, Car5b, Car12, and Car13 at the protein level. The epithelial cells were intensively stained for CAVB, whereas only weak signal was detected for CAXIII. Positive signals for CAIV and CAXII were observed in the capillary endothelial cells and the basolateral plasma membrane of the epithelial cells, respectively. This study provides an expression profile of all CAs in the mouse harderian gland. These results should improve our understanding of the distribution of CA isozymes and their potential roles in the function of harderian gland. The high expression of mitochondrial CAVB at both mRNA and protein levels suggests a role in lipid synthesis, a key physiological process of the harderian gland.  相似文献

The source-sink relationship is one of major determinants of plant performance. The influence of reproductive sink demand on light-saturated photosynthesis (Pmax), dark respiration (RD), stomatal conductance (gs), intrinsic water-use efficiency (WUEi), contents of soluble sugar (SSC), nitrogen, carbon, and photosynthetic pigments was examined in blueberry (Vaccinium corymbosum L. cv. ‘Brigitta’) during the final stage of rapid fruit growth. Measurements were performed three times per day on developed, sun-exposed leaves of girdled shoots with 0.1, 1, and 10 fruit per leaf (0.1F:L, 1F:L, and 10F:L, respectively) and nongirdled shoots bearing one fruit per leaf (NG). Girdling and lower fruit amount induced lower Pmax, gs, N, and total chlorophyll (Chl) and higher WUEi, SSC, RD, Chl a/b ratio and carotenoids-to-chlorophylls ratio (Car/Chl) for the 1F:L and 0.1F:L treatments. The impact of girdling was counterbalanced by 10F:L, with NG and 10F:L having similar values. Variables other than Pmax, RD, gs, WUEi, and SSC were unaffected throughout the course of the day. Pmax and gs decreased during the course of the day, but gs decreased more than Pmax in the afternoon, while WUEi was increasing in almost all treatments. SSC increased from the morning until afternoon, whereas RD peaked at noon regardless of the treatment. Generally, Pmax was closely and negatively correlated to SSC, indicating that sugar-sensing mechanisms played an important role in regulation of blueberry leaf photosynthesis. With respect to treatments, Pmax and N content were positively related, while RD was not associated to substrate availability. The enhanced Car/Chl ratio showed a higher photoprotection under the lower sink demand. Changes in the source-sink relationship in ‘Brigitta’ blueberry led to a rearrangement of physiological and structural leaf traits which allowed adjusting the daily balance between carbon assimilation and absorbed light energy.  相似文献

The genome of the model cyanobacterium, Synechococcus sp. PCC 7002, encodes two paralogs of CruA-type lycopene cyclases, SynPCC7002_A2153 and SynPCC7002_A0043, which are denoted cruA and cruP, respectively. Unlike the wild-type strain, a cruA deletion mutant is light-sensitive, grows slowly, and accumulates lycopene, γ-carotene, and 1-OH-lycopene; however, this strain still produces β-carotene and other carotenoids derived from it. Expression of cruA from Synechocystis sp. PCC 6803 (cruA 6803) in Escherichia coli strains that synthesize either lycopene or γ-carotene did not lead to the synthesis of either γ-carotene or β-carotene, respectively. However, expression of this orthologous cruA 6803 gene (sll0147) in the Synechococcus sp. PCC 7002 cruA deletion mutant produced strains with phenotypic properties identical to the wild type. CruA6803 was purified from Synechococcus sp. PCC 7002 by affinity chromatography, and the purified protein was pale yellow-green due to the presence of bound chlorophyll (Chl) a and β-carotene. Native polyacrylamide gel electrophoresis of the partly purified protein in the presence of lithium dodecylsulfate at 4 °C confirmed that the protein was yellow-green in color. When purified CruA6803 was assayed in vitro with either lycopene or γ-carotene as substrate, β-carotene was synthesized. These data establish that CruA6803 is a lycopene cyclase and that it requires a bound Chl a molecule for activity. Possible binding sites for Chl a and the potential regulatory role of the Chl a in coordination of Chl and carotenoid biosynthesis are discussed.  相似文献

Photochemically induced dynamic nuclear polarization (photo-CIDNP) has been observed in the homodimeric, type-1 photochemical reaction centers (RCs) of the acidobacterium, Chloracidobacterium (Cab.) thermophilum, by 15N magic-angle spinning (MAS) solid-state NMR under continuous white-light illumination. Three light-induced emissive (negative) signals are detected. In the RCs of Cab. thermophilum, three types of (bacterio)chlorophylls have previously been identified: bacteriochlorophyll a (BChl a), chlorophyll a (Chl a), and Zn-bacteriochlorophyll a′ (Zn-BChl a′) (Tsukatani et al. in J Biol Chem 287:5720–5732, 2012). Based upon experimental and quantum chemical 15N NMR data, we assign the observed signals to a Chl a cofactor. We exclude Zn-BChl because of its measured spectroscopic properties. We conclude that Chl a is the primary electron acceptor, which implies that the primary donor is most likely Zn-BChl a′. Chl a and 81-OH Chl a have been shown to be the primary electron acceptors in green sulfur bacteria and heliobacteria, respectively, and thus a Chl a molecule serves this role in all known homodimeric type-1 RCs.  相似文献

The native alpine plant Saussurea superba is widely distributed in Qinghai–Tibetan Plateau regions. The leaves of S. superba grow in whorled rosettes, and are horizontally oriented to maximize sunlight exposure. Experiments were conducted in an alpine Kobresia humilis meadow near Haibei Alpine Meadow Ecosystem Research Station (37°29′–37°45′N, 101°12′–101°33′E; alt. 3200 m). Leaf growth, photosynthetic pigments and chlorophyll fluorescence parameters were measured in expanding leaves of S. superba. The results indicate that leaf area increased progressively from inner younger leaves to outside fully expanded ones, and then slightly decreased in nearly senescent leaves, due to early unfavorable environmental conditions, deviating from the ordinary growth pattern. The specific leaf area decreased before leaves were fully expanded, and the leaf thickness was largest in mature leaves. There were no significant changes in the content of chlorophylls (Chl) and carotenoids (Car), but the ratios of Chl a/b and Car/Chl declined after full expansion of the leaves. The variation of Chl a/b coincided well with changes in photochemical quenching (q P) and the fraction of open PSII reaction centers (q L). The maximum quantum efficiency of PSII photochemistry after 5 min dark relaxation (F (v)/F (m)) continuously increased from younger leaves to fully mature leaves, suggesting that mature leaves could recover more quickly from photoinhibition than younger leaves. The light-harvesting capacity was relatively steady during leaf expansion, as indicated by the maximum quantum efficiency of open PSII centers (\(F_{\text{v}}^{{\prime }}\)/\(F_{\text{m}}^{{\prime }}\)). UV-absorbing compounds could effectively screen harmful solar radiation, and are a main protection way on the photosynthetic apparatus. The decline of q P and q L during maturation, together with limitation of quantum efficiency of PSII reaction centers (L (PFD)), shows a decrease of oxidation state of QA in PSII reaction centers under natural sunlight. Furthermore, light-induced (Φ NPQ) and non-light-induced quenching (Φ NO) were consistent with variation of L (PFD). It is concluded that the leaves of S. superba could be classified into four functional groups: young, fully expanded, mature, and senescent. Quick recovery from photoinhibition was correlated with protection by screening pigments, and high level of light energy trapping was correlated with preservation of photosynthetic pigments. Increasing of Φ NPQ and Φ NO during leaves maturation indicates that both thermal dissipation of excessive excitation energy in safety and potential threat to photosynthetic apparatus were strengthened due to the declination of q P and q L, and enhancement of L (PFD).  相似文献

Mesophyll structure and content of photosynthetic pigments in the leaves of three species of steppe plants, Centaurea scabiosa L., Euphorbia virgata Waldst. et Kit., Helichrysum arenarium (L.) Moench, were investigated in four geographical sites of the Volga region and the Urals located in the forest-steppe and steppe zones. Variations of the studied parameters between geographical points depended both on the species and on the structural organization of the leaf. The highest level of variation was observed for leaf area and pigment content per unit leaf area, the size and the number of chloroplasts in the cell changed to a lesser extent. The leaf thickness, leaf area and mesophyll cell sizes mostly depended on the plant species. C. scabiosa had large leaves (40–50 cm2) with large thickness (280–290 μm) and large mesophyll cells (up to 15000 μm3). The leaves of H. arenarium and E. virgata were ten times smaller and characterized by 1.5 times smaller thickness and 2?3 times smaller cell size. Geographical location and climate of the region affected leaf density, proportion of partial tissue volume, and the ratio of the photosynthetic pigments. In the southern point of Volga region with the highest climate aridity, all studied species were characterized by maximum values of volumetric leaf density (LD), due to the high proportion of sclerenchyma and vascular bundles, and specificity of the mesophyll structure. With the decline in latitude, chlorophyll (Chl) and carotenoid (Car) contents in leaf area were reduced, the ratio Chl/Car was increased, and the ratio Chl a/b was declined. The reduction of the pigment content in the leaf in all species was associated with a reduction in the amount of Chl per chloroplast, and for C. scabiosa and H. arenarium it was associated also with the reduction of chloroplast amount in the leaf area. In turn, chloroplast number per leaf area and the total cell area (Ames/A) depended on the ratio of the number and size of mesophyll cells inherent to this plant species. At the same time, we found a similar mechanism of spatial organization of leaf restructuring for all studied species—decrease in Ames/A was accompanied by increasing in the proportion of intercellular air spaces in the leaf. It is concluded that variations in structural and functional parameters of the photosynthetic apparatus of steppe plants were associated with plant adaptation to climate features. General direction of the changes of leaf parameters of the studied species with aridity was the increase of LD and the decrease of pigment content per leaf area however the cellular mechanisms of changes in the pigment content and integral parameters of mesophyll were determined by the plant species properties.  相似文献

The photosynthesis was investigated 30 d after Pb treatment in Myrica rubra seedlings. The Pb treatment resulted in significantly increased Pb concentrations in shoots. Low Pb concentration exposure (≤2 mM) reduced the net photosynthetic rate (PN), transpiration rate (E), and stomatal conductance (gs) without affecting the intercellular CO2 concentration (Ci), chlorophyll (Chl) content, and Chl fluorescence parameters. At 10 d after severe Pb treatment (≥4 mM), PN was inhibited and accompanied by Chl damage, while at 30 d, the inhibition of PN was followed by an increase of Ci and a decrease of gs, E, Chl content, and Chl fluorescence parameters. M. rubra showed a promising prospect for use in the soil phytoremediation, when Pb concentration is low, but the remediation efficiency of M. rubra is limited if Pb exceeds 2 mM.  相似文献

Chlorophylls (Chl) are important pigments in plants that are used to absorb photons and release electrons. There are several types of Chls but terrestrial plants only possess two of these: Chls a and b. The two pigments form light-harvesting Chl a/b-binding protein complexes (LHC), which absorb most of the light. The peak wavelengths of the absorption spectra of Chls a and b differ by c. 20 nm, and the ratio between them (the a/b ratio) is an important determinant of the light absorption efficiency of photosynthesis (i.e., the antenna size). Here, we investigated why Chl b is used in LHCs rather than other light-absorbing pigments that can be used for photosynthesis by considering the solar radiation spectrum under field conditions. We found that direct and diffuse solar radiation (PARdir and PARdiff, respectively) have different spectral distributions, showing maximum spectral photon flux densities (SPFD) at c. 680 and 460 nm, respectively, during the daytime. The spectral absorbance spectra of Chls a and b functioned complementary to each other, and the absorbance peaks of Chl b were nested within those of Chl a. The absorption peak in the short wavelength region of Chl b in the proteinaceous environment occurred at c. 460 nm, making it suitable for absorbing the PARdiff, but not suitable for avoiding the high spectral irradiance (SIR) waveband of PARdir. In contrast, Chl a effectively avoided the high SPFD and/or high SIR waveband. The absorption spectra of photosynthetic complexes were negatively correlated with SPFD spectra, but LHCs with low a/b ratios were more positively correlated with SIR spectra. These findings indicate that the spectra of the photosynthetic pigments and constructed photosystems and antenna proteins significantly align with the terrestrial solar spectra to allow the safe and efficient use of solar radiation.  相似文献

Using chlorophyll (Chl) a fluorescence many aspects of the photosynthetic apparatus can be studied, both in vitro and, noninvasively, in vivo. Complementary techniques can help to interpret changes in the Chl a fluorescence kinetics. Kalaji et al. (Photosynth Res 122:121–158, 2014a) addressed several questions about instruments, methods and applications based on Chl a fluorescence. Here, additional Chl a fluorescence-related topics are discussed again in a question and answer format. Examples are the effect of connectivity on photochemical quenching, the correction of F V /F M values for PSI fluorescence, the energy partitioning concept, the interpretation of the complementary area, probing the donor side of PSII, the assignment of bands of 77 K fluorescence emission spectra to fluorescence emitters, the relationship between prompt and delayed fluorescence, potential problems when sampling tree canopies, the use of fluorescence parameters in QTL studies, the use of Chl a fluorescence in biosensor applications and the application of neural network approaches for the analysis of fluorescence measurements. Data on the chlorophyll a (Chl a) content in water of the Volga River reservoirs sampled at 64–73 sampling stations in the summer period of 2015 and 2016, analyzed using the standard spectrophotometric method, are presented. Chl a content varied from a minimum of 0.9–3.0 to a maximum of 7.6–32.1 μg/L in June 2016, from 2.1–15.5 to 9.4–86 μg/L in August 2015, and from 1.1–6.7 to 15.1–62.7 μg/L in August 2016. A local increase in Chl a occurred in the near-dam areas of the reservoirs, as well as downstream of the confluence with tributaries. The highest Chl a concentrations were constantly recorded in the highly eutrophic Shoshinskii Reach in the Ivankovo Reservoir and at the Oka River mouth in the Cheboksary Reservoir. The average values of Chl a concentrations make it possible to classify the Ivankovo, Uglich, Rybinsk, and Cheboksary reservoirs as eutrophic; the Gorky and Kuibyshev reservoirs as moderately eutrophic; and the Saratov and Volgograd reservoirs as mesotrophic.  相似文献
