Proteomics of Arabidopsis seeds revealed the differential accumulation during germination of two housekeeping enzymes. The first corresponded to methionine synthase that catalyses the last step in the plant methionine biosynthetic pathway. This protein was present at low level in dry mature seeds, and its level was increased strongly at 1-day imbibition, prior to radicle emergence. Its level was not increased further at 2-day imbibition, coincident with radicle emergence. However, its level in 1-day imbibed seeds strongly decreased upon subsequent drying of the imbibed seeds back to the original water content of the dry mature seeds. The second enzyme corresponded to S -adenosylmethionine synthetase that catalyses the synthesis of S -adenosylmethionine from methionine and ATP. In this case, this enzyme was detected in the form of two isozymes with different p I and M r. Both proteins were absent in dry mature seeds and in 1-day imbibed seeds, but specifically accumulated at the moment of radicle protrusion. Arabidopsis seed germination was strongly delayed in the presence of dl -propargylglycine, a specific inhibitor of methionine synthesis. Furthermore, this compound totally inhibited seedling growth. These phenotypic effects were largely alleviated upon methionine supplementation in the germination medium. The results indicated that methionine synthase and S -adenosylmethionine synthetase are fundamental components controlling metabolism in the transition from a quiescent to a highly active state during seed germination. Moreover, the observed temporal patterns of accumulation of these proteins are consistent with an essential role of endogenous ethylene in Arabidopsis only after radicle protrusion.  相似文献

Abstract

In spite of the simplicity of its molecules, the complex effects of benzoic acids on the regulation of plant growth are an increasingly attractive field of research to chemists and biologists. Halide substituted benzoic acids offer an excellent opportunity to explore the effect of electron withdrawing substituents (fluoro-, chloro-, bromo- and iodo-) on the response of plant growth stage. Under normal physiological conditions, benzoic acids are ionized molecules that exhibit low solubility in water. Monoethanolamine, a natural alkanolamine, was used to generate salts of monoethanolamine of halogenated para-substituted benzoic acids, new compounds with biological activity. This study reports on the biological effects of these substances at different concentrations on Arabidopsis thaliana seed germination and early seedling growth. Seed germination at 22°C, in a vertical position, under a photoperiod of 16 h light and 8 h darkness, was variable depending on the concentration of the compounds applied. Final germination percentages were similar for all treatments and control at 0.05 mM and 0.1 mM (exception p-Br BA and p-I MEASPBA). No germination occurred when seeds were treated with more than 0.5 mM. The results also revealed that the primary root length and the number of secondary roots are reduced in a concentration-dependent manner and also in relation to increasing atomic size of the substituents (F < Cl < Br < I). It is concluded that uptake rates of benzoic acid anions by roots decrease with a decrease in hydrophilic character of the anion and with an increase in molecular size.  相似文献

Fatty acid desaturases play important role in plant responses to abiotic stresses. However, their exact function in plant resistance to salt stress is unknown. In this work, we provide the evidence that FAD2, an endoplasmic reticulum localized ω-6 desaturase, is required for salt tolerance in Arabidopsis. Using vacuolar and plasma membrane vesicles prepared from the leaves of wild-type (Col-0) and the loss-of-function Arabidopsis mutant, fad2, which lacks the functional FAD2, we examined the fatty acid composition and Na+-dependent H+ movements of the isolated vesicles. We observed that, when compared to Col-0, the level of vacuolar and plasma membrane polyunsaturation was lower, and the Na+/H+ exchange activity was reduced in vacuolar and plasma membrane vesicles isolated from fad2 mutant. Consistent with the reduced Na+/H+ exchange activity, fad2 accumulated more Na+ in the cytoplasm of root cells, and was more sensitive to salt stress during seed germination and early seedling growth, as indicated by CoroNa-Green staining, net Na+ efflux and salt tolerance analyses. Our results suggest that FAD2 mediated high-level vacuolar and plasma membrane fatty acid desaturation is essential for the proper function of membrane attached Na+/H+ exchangers, and thereby to maintain a low cytosolic Na+ concentration for salt tolerance during seed germination and early seedling growth in Arabidopsis.  相似文献

Copy-DNA clones have been obtained that distinguish eight messenger mRNAs, moderately abundant in the axes of the germinating soybean (Glycine max (L.) Merr.) seedling. These clones have been used to characterize the size of the mRNAs and to anlyze the accumulation of the mRNAs at different time points and in different parts of the axis during germination and early seedling growth. Three of the mRNAs accumulate to a substantial level by 9 h, a time point before either the beginning of growth or the accumulation of polyribosomes. Four other mRNAs reach a substantial level only at 24 h, a period when rapid seedling growth is occurring. Those mRNAs whose accumulation begins at 24 h were found only in the top (hypocotyl) half of the 24-h seedlings, while the remaining mRNAs were present also in the bottom half of the seedlings in different amounts. By 44 h, the bottom 0.5 cm of the seedlings, i.e., the region of meristematic growth, had little or none of the mRNAs, with the exception of one mRNA. These temporal and spatial observations indicate that many of the mRNAs are not involved simply in the general maintenance of ongoing cell proliferation, but that they may be related to differentiation during early seedling formation. Summary Butylester of morphactin (n-butyl-9-hydroxy-fluorence-(9)-carboxylate) greatly inhibited seed germination and seedling growth of two strains of lettuce, Attraktion and Hohlblättriger Butter. The inhibitory effect of morphactin on seed germination was completely overcome by kinetin but the latter chemical was ineffective in reversing the morphactin induced inhibition of seedling growth. Thus it appears probable that the two substances effect seedling growth independently.  相似文献

Morphactin-butylester (a flourene-9-carboxylic acid derivative) inhibited seed germination of two strains of lettuce. Morphactin induced inhibition of germination could be partially or wholly reversed by simultaneous addition of gibberellic acid. However, gibberellic acid played very little part in reversing the inhibitory effect of morphactin on seedling growth. It is concluded that gibberellin can not reverse all the growth effects induced by morphactin.  相似文献

在不同光照梯度的人工遮荫和森林生境中,研究了西双版纳季节雨林标志树种望天树的种子萌发和幼苗早期生长特征.结果表明,裸地上的强光照和深度遮荫均不利于望天树种子的萌发,中等程度的遮荫有利于种子萌发.望天树种子萌发率在林窗中央最大,而且萌发迅速,林窗边缘和林下生境不利于种子萌发.幼苗株高、基径和单株叶面积等生长指标均在部分遮荫处理条件下最大;幼苗根冠比在裸地上最高,且随遮荫程度的增加而降低;幼苗比叶面积在一定光照强度范围内随遮荫程度的增加而增大,在3层遮荫最大.除幼苗根冠比以外的其它生长参数均在林窗中央最大.讨论了环境因子(主要是光照强度和光质)对望天树种子萌发和幼苗生长的影响.  相似文献

Monoterpenes, the chemical constituents of essential oils found in plants, are known biologically active compounds. The present study was conducted to investigate the inhibitory effects of 30 monoterpenes including monoterpene hydrocarbons and oxygenated monoterpenes on seed germination and seedling growth of Amaranthus retroflexus, Chenopodium album and Rumex crispus under laboratory conditions. The monoterpenes were applied at contents of 10 and 20 microl for liquid compounds and 10 and 20 microg for solid compounds. The results show that most of the monoterpenes significantly inhibited seed germination and seedling growth of the tested plants. Oxygenated monoterpenes including beta-citronellol, nerol and terpinen-4-ol completely inhibited seed germination and seedling growth of all tested plants. Their inhibitory effects were also stronger than that of the herbicide 2,4-D. In general, monoterpenes were less effective against seed germination and seedling growth of C. album as compared with R. crispus and A. retroflexus. Phytotoxic effects of monoterpene hydrocarbons were found to be lower than those of oxygenated monoterpenes. The alcohol derivatives of oxygenated monoterpenes were also found to be more phytotoxic as compared with their acetate derivatives. Based on the present results, it can be concluded that the oxygenated monoterpenes can be used as potential bio-herbicides.  相似文献

为探讨醉马草(Achnatherum inebrians)种群繁殖和扩散机理,并为其防控和开发利用提供理论基础,研究了不同沙埋深度(0、1、2、3、4和5cm)对醉马草种子萌发和幼苗生长的影响。结果表明:埋深对醉马草幼苗出土率、首次出苗时间、幼苗生长高度以及生物量的分配均有极显著影响(P<0.001);种子萌发和幼苗出土率在2cm沙埋下达到最高(92%),在5cm沙埋下最低(58.7%);幼苗最大高度(10.8cm)出现在3cm的沙埋,最小高度(6.3和7.1cm)出现在0和5cm的埋深;埋深为2cm时,地上和地下生物量最大,埋深5cm时地上和地下生物量均最小;最大根长(约5cm)和最小根长(1cm)分别出现在2~3cm和5cm的埋深。2~3cm的埋深是醉马草种子萌发和幼苗生长的最佳深度。  相似文献

测定了Hg2+、Cd2+、Cu2+、Pb2+单一重金属胁迫对拟南芥种子发芽和幼苗生长的影响.结果表明,重金属对幼苗生长的毒性大于对种子发芽的毒性,以抑制种子发芽的IC50为指标,4种重金属的毒性顺序为Hg2+>Cd2+>Pb2+/Cu2+,以幼苗生长为指标,则毒性顺序为:Cu2+>Hg2+>Cd2+/Pb2+,并随着胁迫时间延长,种子萌发率下降.此外,不同重金属在不同发芽时段对种子的毒性也不尽相同,Cd2+的毒性在种子吸水后的0~12 h大于12~24 h,而Hg2+毒性在12~24 h大于0~12 h,其中,种皮对减轻重金属毒性起着十分重要的作用.通过非毒性离子(Ca2+、Mg2+、K+、Na+)与重金属离子(Hg2+、Cd2+、Cu2+、Pb2+)交互作用对拟南芥种子发芽及幼苗生长效应的研究发现, mmol·L-1的Ca2+、Mg2+、K+、Na+可以增强Hg2+对种子发芽的毒性,但对Cd2+的毒性却没有影响.对于幼苗来说,Ca2+、Mg2+、K+、Na+可以显著增强Hg2+的毒性,Ca2+可以缓解Cd2+的毒性,但却增加Cu2+的毒性,K+可以缓解Pb2+对幼苗的毒害作用.最后,本文对重金属的毒害机理进行了探讨.  相似文献

The tripeptide antioxidant glutathione (γ-l-glutamyl-l-cysteinyl-glycine; GSH) essentially contributes to thiol-disulphide conversions, which are involved in the control of seed development, germination, and seedling establishment. However, the relative contribution of GSH metabolism in different seed structures is not fully understood. We studied the GSH/glutathione disulphide (GSSG) redox couple and associated low-molecular-weight (LMW) thiols and disulphides related to GSH metabolism in bread wheat (Triticum aestivum L.) seeds, focussing on redox changes in the embryo and endosperm during germination. In dry seeds, GSH was the predominant LMW thiol and, 15?h after the onset of imbibition, embryos of non-germinated seeds contained 12 times more LMW thiols than the endosperm. In germinated seeds, the embryo contained 17 and 11 times more LMW thiols than the endosperm after 15 and 48?h, respectively. This resulted in the embryo having significantly more reducing half-cell reduction potentials of GSH/GSSG and cysteine (Cys)/cystine (CySS) redox couples (EGSSG/2GSH and ECySS/2Cys, respectively). Upon seed germination and early seedling growth, Cys and CySS concentrations significantly increased in both embryo and endosperm, progressively contributing to the cellular LMW thiol-disulphide redox environment (Ethiol-disulphide). The changes in ECySS/2Cys could be related to the mobilisation of storage proteins in the endosperm during early seedling growth. We suggest that EGSSG/2GSH and ECySS/2Cys can be used as markers of the physiological and developmental stage of embryo and endosperm. We also present a model of interaction between LMW thiols and disulphides with hydrogen peroxide (H2O2) in redox regulation of bread wheat germination and early seedling growth.  相似文献

利用NaCl和Na2SO4复盐溶液胁迫处理36个燕麦品种,采用培养皿纸上发芽法,进行种子萌发和幼苗的耐盐能力鉴定,通过分析盐胁迫对不同燕麦种子萌发及幼苗的影响,为燕麦品种耐盐性筛选、盐碱地燕麦栽培和耐盐育种提供理论参考.结果表明:盐分抑制种子的萌发和幼苗生长,低盐浓度(0.4%)适宜各类燕麦生长,对较耐盐的品种有促进萌发、生长的作用,盐浓度1.2%是鉴定燕麦耐盐性的适宜盐浓度;36个品种耐盐性差异较大,可划分为耐盐型、中度耐盐型、不耐盐型3类;燕麦不同品种及不同类型种子萌发的盐浓度范围达极显著差异,各耐盐指标对于燕麦耐盐性的体现存在一定的差异,发芽率、发芽指数、简易活力指数和单株干质量是主要的耐盐指标.  相似文献

In order to enhance sink strength, we expressed a heterologous plant cell wall invertase (CrCIN1) under the control of a root-specific promoter (ppyk10) in Arabidopsis thaliana. Slightly elevated apoplastic invertase activity resulted in apparent phenotypic changes. Transgenic plants developed more secondary roots and subsequently, possibly because of a higher capacity to acquire nutrients, a higher shoot and whole plant biomass. Furthermore, an early flowering phenotype was detected. The data presented here demonstrate that it is possible to modulate carbohydrate metabolism by ectopic expression of cell wall invertases and thereby influence sink organ size and whole plant development.  相似文献
