  • 1.1. Influence of some neurotransmitters and neuromodulators on the PMA-stimulated phosphorylation in vitro of calcium pump-like protein from rat cerebellum synaptosomal membranes was examined.
  • 2.2. The prolonged time (up to 6 min) of synaptosomal membranes preincubation with 1 and 10 μM serotonin results in the increase of phosphorylation. The decrease of phosphorylation up to 80% of control value was observed for 100 μM serotonin.
  • 3.3. The most stimulating effect on 130kDa protein phosphorylation was observed with 1μM of histamine (160% of control value).
  • 4.4. 1 and 0.1 μM somatostatin triggered a short-time transient increase of 130 kDa phosphorylation (up to 135% of control value).

  • 1.1. In the absence of sodium, the reabsorption rate of amino acid α-aminoisobutyric acid (AIB) by the nephridia of Sabella pavonina is reduced to 20% and the AIB accumulation in the cells is reduced to 10%. These results suggest the presence of sodium-dependant processes.
  • 2.2. The observed processes are reversible when control conditions are re-established.
  • 3.3. A minimum of 17mEq/l Na+ is required to restore the normal reabsorption rate.
  • 4.4. The addition ofamiloride (10−35 M) decreases the reabsorption rate, but to a lesser extent than the absence of sodium.

  • 1.1. Uptake and elimination of lindane, 3,4-dichloroaniline, phenol and 4-nitrophenol by the zebrafish Brachydanio rerio were investigated in tap water and in water of the river Rhine.
  • 2.2. The differences in bioconcentration of chemicals between the two water types did not exceed a factor of 2.5.
  • 3.3. Elimination kinetics were comparable in tap and river water.
  • 4.4. It can be concluded that water of the river Rhine does not influence the kinetics of the investigated xenobiotics.

  • 1.1. Two morphotypes of Myliobatis from the demersal fishery off the Rio Grande (Brazil) were studied.
  • 2.2. Thirty-two alleles were detected and resolved by 27 loci.
  • 3.3. Nei's measure of genetic identity was 0.8306 and Thorpe's similarity was 0.6990. Mean heterozygosities observed were 0.1327 for the “DE” morphotype and 0.0409 for the “DL” morphotype.
  • 4.4. Seven loci were fixed differently in the two taxa studied. This indicates the existence of a barrier to gene-flow between them, showing that both morphotypes belong to different species.
  • 5.5. Jaccard's measure of similarity was calculated and a phenogram with the two morphotypes and M. freminvillii was constructed using isoelectric focusing of total soluble proteins. This showed a higher similarity between the two morphotypes of Myliobatis than M. freminvillii.

  • 1.1. The activities of three lysosomal enzymes (acid phosphatase, β-galactosidase, catepsin D) was observed during metamorphosis in the fat body and midgut cells of two insects (Mamestra brassicae and Pieris brassicae).
  • 2.2. The activities increased slightly during the feeding period and showed a sharp rise at the beginning of the wandering period.
  • 3.3. Subsequently, a decrease was observed during the pre-pupal stage and pupation.
  • 4.4. The activities increased again 2 days after the larval-pupal moult.
  • 5.5. We suggest that an inhibitory mechanism works in the studied cells before pupation to protect the stored proteins from the degradation until the beginning of differentiation of imaginai cells in the pupal stage.

  • 1.1. The malate dehydrogenase (MHD) activity from the ribbed mussel gill is polymorphic with two distinct mitochondrial forms (M1 and M2) and five forms that could be resolved from cytosolic extracts (C1 to C5) by DEAE-cellulose chromatography and starch gel electrophoresis.
  • 2.2. Two of the cytosolic forms (C3 and C4) may represent interchangeable conformational states.
  • 3.3. With kinetic analysis there appear to be three distinct cytosolic forms (C1, C2 and C3–C4), with C2 possibly behaving as a heterodimer.
  • 4.4. The identity of C5 is uncertain.
  • 5.5. The forms isolated from the mitochondria (M1 and M2) exhibited lower apparent Kms for oxaloacetate (OAA) than the cytosolic forms.
  • 6.6. For all isozymic forms, the apparent Kms for OAA increased as the pH increased between pH 6 and 9
  • 7.7. Increasing the salt concentration raised the Km for OAA for all forms.
  • 8.8. The mMDHs were more sensitive to inhibition by NaCl than the cMDHs.
  • 9.9. Representative cMDH (C1) and mMDH (M2) isozymes exhibited substrate inhibition by high concentrations of OAA with the mMDH possessing lower Kis for substrate inhibition than the cMDH at each pH tested.
  • 10.10. Differences and similarities in Km app. for OAA at the different pHs and salt concentrations indicated that C1, C2 and C3–C4 and C5 were distinct forms, that M1 and M2 were distinct but very similar to each other, and that C1, C2, C3–C4 and C5 were distinct from M1 and M2.

  • 1.1. Embryonic and posthatch turkey skeletal muscle development was compared in in vitro studies using clonal-derived embryonic myoblasts and satellite cells.
  • 2.2. Although population doubling times were similar between the two lines (25.4 hr for satellite cells and 26.4 hr for embryonic myoblasts), embryonic myoblasts consistently began log phase growth 24 hr earlier than satellite cells.
  • 3.3. Differentiation (fusion) of embryonic myoblasts was maximized by 36 hr in Dulbecco's Modified Eagle's Medium containing 1% horse serum compared with 72 hr for satellite cells.
  • 4.4. When administered a serum-free medium which supports proliferation of turkey satellite cells, embryonic myoblasts differentiated to form myotubes.

  • 1.1. The planktonic diatom Ditylum brightwellii, grown in light-limited resp. nitrogen-limited continuous culture, has been exposed to Cu levels, comparable with those in the Scheldt estuary.
  • 2.2. At increasing levels D. brightwellii initially detoxified Cu, producing metal-binding ligands (amino acids), and increasing its cell volume.
  • 3.3. In light-limited D. brightwellii, photosynthesis could be adjusted to increasing Cu stress, division rates remained constant, and cells proved to be adaptable to 200 nM dissolved Cu.
  • 4.4. Nitrogen-limited D. brightwellii detoxified Cu inadequately: it stored large amounts of Cu (30–60 μM) that inhibited cell division.

  • 1.1. Biliverdin reductase from the liver of eel, Anguilla japonica was characterized and purified with a novel enzymatic staining method on polyacrylamide electrophoretic gel.
  • 2.2. This enzyme could use both NADPH and NADH as coenzyme. The Km of NADPH was 5.2 μM, while that of NADH was 5.50 μM.
  • 3.3. The optimum reaction pH for using HADPH as coenzyme was 5.3. That for NADH was 6.1. The optimum reaction temperature is 37°C.
  • 4.4. When NADPH was used as coenzyme, the Km of biliverdin was 0.6 μM. When NADH was used as coenzyme, the Km of biliverdin was 7.0 μM.
  • 5.5. The activity of the enzyme was inhibited by the concentration of biliverdin. Also, the potency of the enzyme was much less than that of the analogous enzyme isolated from mammals.
  • 6.6. This is a fairly stable enzyme with a mol. wt around 67,000. Its estimated pI was pH 3.5–4.0.
  • 7.7. This is the first time biliverdin reductase has been isolated and characterized from a vertebrate other than mammals. The property of it is quite different from that of mammals.

  • 1.1. Carp red cells were treated with drugs that affect the cell membranes. The water content of the cells and the accumulation of cAMP in the cells were measured in normoxia and in hypoxia using non-stimulated and adrenergically stimulated cells.
  • 2.2. WGA, DIDS + CCCP and A23187 increased the water content of nonstimulated normoxic cells.
  • 3.3. In hypoxia ouabain and DIDS + CCCP increased the water content but cytochalasin B, NPM, DIDS, CCCP and A23187 + CA2+ abolished the hypoxia-induced swelling.
  • 4.4. Any membrane perturbation induced some cAMP formation, Sophora and Anquilla lectins being most potent.
  • 5.5. Also in adrenergically stimulated cells, membrane perturbation generally increased cAMP formation.
  • 6.6. However, cAMP accumulation diminished in cells treated with cytochalasin B, CCCP and DIDS + CCCP.
  • 7.7. The adrenergic swelling of carp red cells was reduced in normoxia by DIDS. NPM and CCCP increased the adrenergic swelling in normoxia to hypoxic level.

  • 3.3. Kidney, heart and neuronal cells (monitored photographically) swelled less than expected for strictly semipermeable cells, but exhibited no regulatory volume decrease.
  • 4.4. Long-term viability of the cells was not compromised following acute hyposmotic stress.
  • 5.5. Quinidine, which blocks SA channels in Lymnaea, intensified stress-induced swelling most markedly in kidney cells.

  • 2.2. All the cells composing this gland show PhA activity, the distribution pattern differing according to the cell type.
  • 3.3. The digestive cells show the most widely distributed reaction product (brush border, phagolysosomes, multivesicular bodies and autophagic vacuoles).
  • 4.4. In the excretory cells this activity appears in large sacs, while in the calcium cells the reaction product is abundant in the calcium granules.
  • 5.5. Cellular digestion processes performed by each of these cell types is discussed together with their role in the detoxification of heavy elements derived from the environment.

  • 5.5. In situ monitoring of pH levels revealed a lower pH level in the intestinal proximal section of hybrid striped bass compared with the distal section.
  • 6.6. In contrast, higher pH levels were detected at the proximal compared with the distal sections of D. labrax intestine.

  • 1.1. The effect of adenosine separately or in combination with alpha-1 adrenergic antagonist prazosin and alpha-2 adrenergic antagonist yohimbine as well as adenosine antagonists 8-phenyltheophylline and xanthine amine conjugate on glucose-induced insulin secretion from isolated rat pancreatic islets was studied.
  • 2.2. Their in vivo effects on serum glucose and insulin levels were also investigated. Adenosine at 10 and 100 μM inhibited significantly, insulin secretion from the isolated islets whereas at 10 mM slightly increased the secretion of insulin.
  • 3.3. Prazosin used at 100 μM inhibited insulin secretion. When it combined with adenosine (10 μM) it augmented the inhibitory effect of adenosine.
  • 4.4. In vivo prazosin (21 mg/kg bodywt) caused a hyperglycaemia which was accompanied by hypoinsulinaemia.
  • 5.5. Concurrent administration of this drug with adenosine neither affect the hyperglycaemic nor the hypoinsulinaemic effects of adenosine.
  • 6.6. On the other hand, yohimbine (100 μM) has no effect neither separately nor in combination with adenosine (10 μM) in modulating the inhibitory effect of adenosine on insulin secretion.
  • 7.7. When Yohimbine administered at 19.5 mg/kg body wt it did not alter serum glucose but it markedly increased the serum insulin level. Its combined administration with adenosine reduced the hyperglycaemic effect of adenosine with a remarkable increase in serum insulin.
  • 8.8. Both adenosine-antagonists were ineffective in alteration of insulin secretion.
  • 9.9. However, combination of 8-phenyltheophylline with adenosine (10 μM) totally blocked the inhibitory effect of adenosine on insulin secretion while xanthine amine conjugate failed to prevent this effect of adenosine.
  • 10.10. These results indicate that the inhibitory effect of adenosine on insulin secretion is neither mediated via alpha-1 nor alpha-2 adrenoceptors. It might be via activation of specific adenosine receptors on rat islets which are sensitive to blockade by 8-phenyltheophylline.
