Abstract The fatty acid composition of Pimelobacter sp. is of the complex type. When pyridine was used as a sole inhibitory substrate, the fatty acid composition of Pimelobacter sp. was quite different from that within other readily available substrates. When compared with the fatty acid composition in a complex medium, the proportion of isopalmitic acid ( iso -C16:0) drastically decreased from 68.4% to 7.7%, while the proportion of anteisoheptadecanoic acid ( anteiso -C17:0) remained almost constant at ca. 7%. Concomitantly, this decrease of the branched-chain fatty acid was accompanied by the increase of the straight-chain, especially long-chain, fatty acid such as heptadecanoic (C17:0), octadecenoic (Cig:i), 10-methylheptadecanoic (10-me-18) and 10-methyloctadecanoic (10-me-19) acids. Consequently, in response to membrane active organic solvents, Pimelobacter sp. was found to regulate its membrane fatty acid composition in a fashion different from Gram-negative bacteria.  相似文献

采用气相色谱仪和氨基酸分析仪测定了半滑舌鳎(Cynoglossus semilaevis)受精卵、卵黄囊仔鱼和开口仔鱼的氨基酸与脂肪酸组成的变化。结果表明:总氨基酸组成在受精卵和卵黄囊仔鱼之间变化明显,但是在卵黄囊仔鱼和开口仔鱼之间只有细微的变化。开口仔鱼与其摄食的轮虫的总必需氨基酸组成相关。受精卵、卵黄囊仔鱼、开口仔鱼的游离氨基酸含量分别为139 mg/g、3.6 mg/g和2.5 mg/g,占总氨基酸含量的22.3%、3.6%和2.5%。饱和脂肪酸的总量从受精卵到卵黄囊仔鱼明显下降,但是发育到开口仔鱼含量无显著变化。单不饱和脂肪酸和多不饱和脂肪酸的总量在不同发育阶段无显著变化,而EPA和DHA的含量从卵黄囊仔鱼到开口仔鱼有明显下降。这表明在早期发育阶段半滑舌鳎主要利用饱和脂肪酸作为能量代谢的基质,对饱和脂肪酸的利用程度大于单不饱和脂肪酸和多不饱和脂肪酸。半滑舌鳎似乎需要长链的多不饱和脂肪酸如EPA、DHA和ARA。  相似文献

分析了13种微藻(包括7种绿藻,5种杂色藻和1种红藻)的总脂含量和脂肪酸组成,结果表明,不同门类微藻的脂肪酸组成差异较大:绿藻的脂肪酸组成以C16和C18为主;杂色藻类的脂肪酸组成相近,金藻门含有14:0、16:0、18:1、18:4等特征脂肪酸,三角褐指藻主要的脂肪酸为14:0、16:0、16:1、16:3和20:5,而粉核油球藻的脂肪酸以14:0、16:0、20:5为主;紫球藻的脂肪酸组成以16:0、20:4和20:5为主。在测试的13种微藻中,杜氏盐藻的亚麻酸含量最高,占总脂肪酸的60.9%;等鞭金藻的十八碳四烯酸含量最高,占总脂肪酸的19.6%;紫球藻和粉核油球藻中花生四烯酸与二十碳五烯酸(EPA)含量分别占总脂肪酸的17.1%和20.9%。  相似文献

Summary A diverse collection of mutants of Arabidopsis with altered seed lipid compositions was isolated by determining the fatty acid composition of samples of seed from 3,000 mutagenized lines. A series of mutations was identified that caused deficiencies in the elongation of 181 to 201, desaturation of 181 to 182, and desaturation of 182 to 183. In each of these cases the wild type exhibited incomplete dominance over the mutant allele. These results, along with results from earlier studies, point to a major influence of gene dosage in determining the fatty acid composition of seed lipids. A mutation was also isolated that resulted in increased accumulation of 183. On the basis of the effects on fatty acid composition, the nature of the biochemical lesion in three of the mutants could be tentatively attributed to deficiencies in activities of specific enzymes. The other mutant classes had relatively less pronounced changes in fatty acid composition. These mutants may represent alterations in genes that regulate lipid metabolism or seed development. The fatty acid composition of milk fat as influenced by feeding oilseeds   总被引:5,自引:0,他引:5  
The fatty acid composition of bovine milk fat can be substantially altered by feeding lipid sources which alter the fatty acid profile of lipid entering the intestine from the rumen. As long-chain fatty acids of dietary origin can be incorporated directly into milk fat the opportunity exists to alter the ratio of short and long-chain fatty acids as well as the degree of saturation of milk fat. In practice our ability to alter the fatty acid profile of milk fat is limited not by the synthetic capacity of the mammary gland, but rather by the challenge of achieving effective protection of unsaturated dietary fatty acids from biohydrogenation in the rumen, as well as keeping the level of polyunsaturated fatty acids within the range where the organoleptic quality and shelf-life of milk and dairy products are not compromised. The fatty acid composition of oilseeds such as canola are considered desirable from a human health perspective and thus their inclusion in the diet of dairy cattle as a means of achieving a more desirable fatty acid profile in milk fat may enhance the nutritive quality of milk.  相似文献

Two yellow and two red pigmented strains of Thermus were monitored for changes in fatty acid content and composition with reference to growth phase at the optimum temperature. Fatty acid content per mg of dry weight increased as the cultures aged. In addition the quantities of iso C 15:0, iso C 17:0 and iso C 16:0 increased in yellow pigmented strains, but in red pigmented strains, an increase was seen in iso C 15:0, but C 16:0 and iso C 16:0 levels decreased. Thus the fatty acid composition of these organisms varies with growth phase, and shows also strain specific variability.  相似文献

The fatty acid composition of five embryo stages following the induction of embryogenesis in cultures of Daucus carota have been studied. Quantitative rather than qualitative changes in fatty acid content were observed, although globular embryos do possess some fatty acids of long (C 20 and above) chain length not observed in earlier or later stages in development. Abstract The relationship between freezing resistance and cellular long-chain fatty acid composition of 18 selected yeast strains was studied. All strains produced a series of saturated and unsaturated evennumbered fatty acids ranging from 14 to 20 carbons in length. The majority of the freeze-resistant yeasts were found among fermentative species with a content of oleic acid above 40%.  相似文献

The haptophyte microalga Tisochrysis lutea was heterotrophically grown in F2 medium with different combinations of pH and salinity. Growth, oil content and fatty acids (FAs) profile were determined under each set of conditions. The salinity was adjusted using NaCl at concentrations of 0.4, 0.6, 0.8, or 1.0 M, while pH was adjusted at 7, 8, or 9, and heterotrophic growth was performed using organic carbon in the form of sugar cane industry waste (CM). Fatty acid methyl esters (FAMEs) were identified by gas chromatography. The results showed that pH of 8.0 was the optimal for dry weight and oil production, regardless of the salinity level. At pH 8.0, growth at a salinity of 0.4 M NaCl was optimal for biomass accumulation (1.185 g L-1). Under these conditions, the maximum growth rate was 0.055 g L-1 d-1, with a doubling time of 17.5 h and a degree of multiplication of 2.198. Oil content was maximal (34.87%) when the salinity was 0.4 M and the pH was 9.0. The ratio of saturated to unsaturated FAs was affected by the pH value and salinity, in that unsaturated FAs increased to 58.09% of the total FAs, considerably greater than the value of 40.59% obtained for the control (0.4 M NaCl and pH 8.0).  相似文献

To study the interaction of voltage-sensitive Na+-channels with membrane lipids, the phospholipid and fatty acid composition of highly purified membrane fragments from the remarkably differentiated plasma membrane of Electrophorus electricus has been analyzed. After density gradient fractionation and carrier free electrophoresis, fractions with up to 30 pmol tetrodotoxin binding/mg protein can be obtained, which may correspond to a 50% pure preparation of the extrasynaptic part of the excitable face. Phospholipid classes and cholesterol are separated by one-dimensional thin-layer chromatography in acidic and alkaline solvent systems. The following mean molar contents are found: 40% phosphatidylcholine, 23% phosphatidylserine, 30% phosphatidylethanolamine and 7% sphingomyelin. In a series of 11 animals, significant deviations from these mean values have been observed. The fatty acid composition of the phospholipids has been determined by gas chromatography. Phosphatidylcholine contains more than 50% 16:0, and about 20% unsaturated fatty acids in the C-18 group. Compared to other plasma membrane fractions, this phospholipid is the least differentiated. By contrast, phosphatidylethanolamine and phosphatidylserine show many characteristics in different membrane fractions, especially in their unsaturated components representing more than 50%. 22:6, as the major constituent in these fractions, accounts for a quarter to a third of all fatty acids in these fractions. 18:0 is the main saturated component in these two phospholipids with abundances of typically a quarter or less of all fatty acids. Knowledge of the lipid composition of these excitable membranes may help to conserve binding and structural properties when analyzing lipid-sensitive Na+-channels in vitro. This study investigated the effect of single and mixed algal diets on growth, fatty acid composition and ingestion rates for Dendraster excentricus larvae. Larvae were assigned to three single algal diet treatments Isochrysis galbana, Dunaliella tertiolecta or Rhodomonas sp. and four mixed algal diet treatments D. tertiolecta and Rhodomonas, I. galbana and D. tertiolecta, I. galbana and Rhodomonas, D. tertiolecta, Isochrysis galbana and Rhodomonas sp. Small amounts (0.36-0.6%) of stearic acid (18:0) were seen in the three algae used but a relatively large percentage (7-25%) of this SAFA was found in Dendraster larvae. The alga D. tertiolecta had the highest percentage (51.7%) of the short chain polyunsaturated fatty acid (PUFA) linolenic acid 18:3(n-3) and trace amounts (0.02-0.14%) of the long chain PUFAs eicosapentanoic (EPA, 20:5(n-3) and docosahexanoic acids (DHA, 22:6(n-3)). However, sand dollar larvae demonstrated the ability to elongate and desaturate shorter chain (18 carbon) polyunsaturated fatty acids (PUFA) to longer chain (20 carbon) n-3 PUFA. Thus high levels of 18:3(n-3) in D. tertiolecta led to high levels of EPA and low levels of 18:3(n-3) in Dendraster larvae fed this diet. Rhodomonas sp. had the highest percentage of stearidonic acid (18:4(n-3), 38.14%) and EPA (10.6%). Despite high levels of 18:4(n-3) in Rhodomonas sp. this acid was absent or found at very low levels in larvae fed this alga, or any combination of this alga. I. galbana had the highest percentage of DHA(14.3%) but was almost devoid of EPA (0.43%). Although Rhodomonas sp. and I. galbana had high levels of EPA and DHA sand dollar larvae did not incorporate higher levels of these long chain PUFAs into their lipids compared to those fed the alga D. tertiolecta. Dendraster larvae synthesized a number of 20 and 22 carbon non-methylene interrupted dienes (NMID), with levels increasing with larval stage. Higher ingestion rates were observed for Dendraster larvae fed single algal diets (Rhodomonas sp. or D. tertiolecta) and lower ingestion rates for those fed mixed algal diets. The highest ingestion rates were for 8-arm Dendraster larvae fed the large alga Rhodomonas sp. presented as a single algal diet. When fed a combination of three algae, selection of particles varied slightly depending on stage with 8-arm larvae ingesting slightly more of the larger algal cell in the mixture than 6-arm larvae. The present study suggests that regardless of the ratios given larvae might have an optimum ratio of different sized particles at which they can feed. The mixed algal diet of I. galbana and D. tertiolecta was the best algal diet leading to significantly larger larvae with high survival and development to metamorphosis. The single algal diets of Rhodomonas sp. or Dunaliella tertiolecta were the second and third best algal diets based on growth and survival to metamorphosis.  相似文献

The objective of this study was to identify single nucleotide polymorphisms (SNPs) in the thioesterase (TE) domain of the bovine fatty acid synthase (FASN) gene and to evaluate the extent to which they were associated with beef fatty acid composition. The four exons in FASN that encode for the TE domain were sequenced, and three SNPs, AF285607:g.17924A>G, g.18663T>C and g.18727C>T, were identified. Purebred Angus bulls (n = 331) were classified into three genotype groups, g.17924AA (n = 121), g.17924AG (n = 168) and g.17924GG (n = 42). The g.17924A>G genotype was significantly associated with fatty acid composition of longissimus dorsi muscle of Angus bulls. Cattle with the g.17924GG genotype had lower myristic acid (C14:0; P < 0.0001), palmitic acid (C16:0, P < 0.05) and total saturated fatty acid contents (P < 0.01), greater health index (P < 0.001), oleic acid content (C18:1; P < 0.001) and total monounsaturated fatty acid concentration (P < 0.01) in the total lipids and triacylglycerols fraction than did those with the g.17924AA genotype. Because of the linkage disequilibrium between SNPs g.17924A>G and g.18663T>C, similar significant associations of fatty acid contents with the g.18663T>C genotypes were observed. In conclusion, the SNPs g.17924A>G and g.18663T>C may be used as DNA markers to select breeding stock that have a healthier fatty acid composition.  相似文献

本研究测定了瘤背石磺产卵前后肌肉、肝胰腺、两性腺、卵蛋白腺、雄性附性腺和黏液腺指数、水分含量、总脂含量、脂类和脂肪酸组成.结果表明:(1) 瘤背石磺产卵后的肝胰腺指数(HIS)、两性腺指数(DGSI)和卵蛋白腺指数(EAGI)都显著下降(P<0.05), 其水分含量显著上升, 而脂肪含量显著下降; (2) 瘤背石磺各种组织总脂主要有磷脂(PL)、胆固醇(Cho)、游离脂肪酸(FFA)和甘油三酰(TG), 仅在肝胰腺中检测出少量甘油一酰(MG), 产卵后肝胰腺中的FFA含量和两性腺中的PL含量大幅度下降, 分别下降了41.36%和80.53%, 其它组织中的脂类成分变化幅度不大;(3) 产卵后瘤背石磺肝胰腺中的C16:1n7、C20:5n3(EPA)和C22:6n3(DHA)显著下降, 而饱和脂肪酸(∑SFA)含量显著上升.产卵后两性腺中的C20:2n6、C22:2n6和C22:5n3(DPA)显著下降, 而C20:4n6(AA)含量显著上升.产卵后雄性附性腺和黏液腺中的AA和EPA含量均有所下降, 而C16:0和C18:1n9却有所上升.结果表明:卵蛋白腺和黏液腺对于瘤背石磺产卵和胚胎的形成具有重要的作用, PL和多不饱和脂肪酸(PUFA)对瘤背石磺的两性腺发育具有十分重要的作用, 肝胰腺中的脂肪可能是瘤背石磺产卵过程中主要的能源物质之一, 性腺发育过程中肝胰腺中的PUFA可能被转运到两性腺中.  相似文献

The correlation between the fluidity of phospholipids and their fatty acid composition was studied by spin label technique and gas-liquid chromatography for three major phospholipid species in Tetrahymena pyriformis during temperature acclimation. The fluidity of 2-aminoethylphosphonolipid increased within the first 10 h of the cold-acclimation when the content of γ-linolenic acid in 2-aminoethylphosphonolipid was highest, and it then decreased up to 24 h. On the other hand, the fluidities of phosphatidylethanolamine and phosphatidylcholine showed a gradual decrease up to 24 h after the temperature shift, although γ-linolenic acid contents were highest at 10 h after the temperature shift. Thus the fluidity changes of these two phospholipids were interpreted as resulting from the altered content of other fatty acids in addition to γ-linolenic acid, since the γ-linolenic acid content was smaller than that of 2-aminoethylphosphonolipid. The results suggest that the content of γ-linolenic acid in 2-aminoethylphosphonolipid plays a role in regulating the thermal adaptation process.  相似文献

The hypolipidemic fibrates have been identified as agonists of the peroxisome proliferator-activated receptor alpha (PPARalpha), which plays a critical role in the regulation of cardiac fatty acid metabolism. Despite the widespread clinical use of fibrates, their role in myocardial oxidative stress and fatty acid composition is less known. In this study, male Sprague-Dawley rats were treated with either vehicle (olive oil, 1 ml/kg) or clofibrate (300 mg/kgday i.p.) for 1-14 days. Lipid peroxidation in heart homogenate was determined by thiobarbituric acid reactive substance (TBARS) assay. Results show that hearts from clofibrate-treated rats are more susceptible to FeSO(4)-induced TBARS production. The antioxidants including catalase and glutathione-related enzymes were marginally affected. We demonstrated that myocardial fatty acid composition was dramatically altered by clofibrate treatment. In hearts from clofibrate-treated rats, the principal n-6 polyunsaturated fatty acids (PUFAs), linoleic acid (C18:2 n-6) and arachidonic acid (C20:4 n-6), was significantly reduced, while the content of the principal n-3 PUFA, docosahexaenoic acid (C22:6 n-3), was markedly increased. The overall effect was to reduce n-6/n-3 ratio and increase the unsaturation extent of myocardial fatty acids. Functional study showed that hearts from clofibrate-treated rats had an improved recovery of post-ischemic contractile function and reduced ischemia/reperfusion (I/R)-induced infarct size. The data shows that clofibrate has a profound impact on cardiac fatty acid composition, which may contribute to its cardioprotective effect.  相似文献

The fatty acid composition during the transformation of Trypanosoma cruzi epimastigotes into metacyclic trypomastigotes (metacyclogenesis) was analysed by gas-liquid chromatography and mass spectrometry. Significant qualitative and quantitative changes in the fatty acid composition occurred during incubation of epimastigotes derived from LIT medium in the triatomine artificial urine (TAU). Metacyclogenesis was also followed by alterations in the fatty acid pattern but these were considerably less pronounced when compared to the pattern obtained for TAU-incubated epimastigotes. These results suggest that changes in the lipid composition precede the morphological transformation of epimastigotes into metacyclic trypomastigotes.  相似文献

为探讨内生菌和植物的生态关系,以生物量和脂肪酸组分作为主要指标,研究了乌桕韧皮部分离获得的5种内生真菌(丝核菌、小菌核菌、小单头孢、毛壳菌、拟盘多毛孢)在不同环境因子下的生长.与合成培养基相比,在液体马铃薯培养基上发酵,生物量较高,脂肪酸不饱和指数较低;其脂肪酸主要为棕榈酸、油酸、亚油酸.与未添加乌桕汁的合成培养基相比,添加乌桕浸汁对小菌核菌生长有促进作用,对其余4种菌有抑制作用;脂肪酸不饱和指数均进一步增加.在合成培养基中添加NaCl培养小菌核菌,生物量均无显著差异;在0~0.5mol·L-1 NaCl时,脂肪酸不饱和指数无显著差异;在0.6~1.0mol·L-1 NaCl时随着盐浓度增加,脂肪酸不饱和指数却下降;表明该菌有较强的耐盐能力.添加植物油对小菌核菌菌丝生长有促进作用,其中在添加1.5%的植物油时,生物量最大;其脂肪酸随添加植物油而改变.以上特点均和二者的共生关系有关.  相似文献

Fatty acid composition of lipids in various organs of the bonito (Euthynnus pelamis), a typical species within the tuna species, caught in the sea off the Pacific coast of Japan, was determined by gas-liquid chromatography. The major fatty acids of all organs examined were palmitic acid (16:0), palmitoleic acid (16:1), oleic acid (18:1), icosapentaenoic acid (20:5), and docosahexaenoic acid (22:6). 22:6 (DHA) was the dominant fatty acid accounting for 25% or more of the total fatty acids in all organs of the bonito, caught in the sea off the Japanese coast.  相似文献

Microbial fatty acids are an attractive source of precursors for a variety of renewable commodity chemicals such as alkanes, alcohols, and biofuels. Rerouting lipid biosynthesis into free fatty acid production can be toxic, however, due to alterations of membrane lipid composition. Here we find that membrane lipid composition can be altered by the direct incorporation of medium-chain fatty acids into lipids via the Aas pathway in cells expressing the medium-chain thioesterase from Umbellularia californica (BTE). We find that deletion of the aas gene and sequestering exported fatty acids reduces medium-chain fatty acid toxicity, partially restores normal lipid composition, and improves medium-chain fatty acid yields.  相似文献
