Cytokinins from leaf laminae, buds, petioles, stems, roots,and root exudate of mature vegetative plants of Xanthium strumariumL. were extracted, fractionated, and partially characterizedby means of column chromatography with Sephadex LH20. Two peaksof cytokinin activity with elution volumes corresponding tozeatin and zeatin riboside were detected, in varying concentrations,in all plant parts. A third cytokinin, detected only in petiolesand in expanding and mature leaves, eluted off the Sephadexcolumn before zeatin riboside. This cytokinin (peak ‘a’)was converted to zeatin or to a zeatin-like cytokinin followingboth acid hydrolysis and treatment with ß-glucosidase.Peak ‘a’ was not detected in buds or in the youngestdeveloping leaves but was the predominant cytokinin presentin half-expanded and more mature leaves. By contrast, the zeatinriboside-like peak (peak ‘b’) constituted the majorcytokinin in root exudate, apical buds, and the youngest developingleaves, while not greatly contributing to the cytokinin contentof mature leaves. The detopped root system was shown to be capable of cytokininproduction. The distribution of cyrtokinins in the plant isdiscussed in relation to their probable origin in the root system.  相似文献

Vegetative plants of Sinapis alba L. grown in short days were induced to flower by expsoure to one or continuous long days. In both inductive conditions, the first flowers were initiated about 60 h after the start of the treatment. Soluble protein extracts were prepared from apical buds and just-expanded leaves of both vegetative and induced plants. Rabbit antisera were prepared using extracts from vegetative and reproductive buds. Immunodiffusion tests were performed. Analysis of the precipitin bands indicated that: (1) one antigenic protein was present in the vegetative buds and disappeared from the buds of induced plants between 96 and 240 h after the start of the inductive treatment; (2) the concentration of a another antigenic protein increased in buds of induced plants 30 h after the start of the inductive treatment; (3) the concentration of a third antigenic proteín increased in buds of induced plants at 96 h.  相似文献

Extracellular extracts from leaves were applied to moderately induced cuttings of potato. The involvement of nitrogenous substances in the transition to flowering was investigated in Sinapis alba and Arabidopsis thaliana (Columbia). Both species grown in short days (SD) are induced to flower by one long day (LD). In S. alba, the phloem sap (leaf and apical exudates) and the xylem sap (root exudate) were analysed in LD versus SD. In A. thaliana, only the leaf exudate could be analysed but an alternative system for inducing flowering without day‐length extension was used: the displaced SD (DSD). Significant results are: (i) in both species, the leaf exudate was enriched in Gln during the inductive LD, at a time compatible with export of the floral stimulus; (ii) in S. alba, the root export of amino acids decreased in LD, whereas the nitrate remained unchanged – thus the extra‐Gln found in the leaf exudate should originate from the leaves; (iii) extra‐Gln was also found very early in the apical exudate of S. alba in LD, together with more Glu; (iv) in A. thaliana induced by one DSD, the leaf export of Asn increased sharply, instead of Gln in LD. This agrees with Asn prevalence in C‐limited plants. The putative role of amino acids in the transition to flowering is discussed.  相似文献

Summary Variation in nitrate reductase activity associated with vegetative development and morphological differences inLolium perenne L. cv S24 was examined using anin vivo technique of enzyme assay. Little difference in enzyme activity per unit weight of leaf laminar tissue was found between tillers, between and within leaf laminae but activity in leaf laminae, was much higher than that of leaf sheaths; the activity of shoot material was greater than the apparent root activity. Senescence of leaf material was not accompanied by marked changes invivo nitrate reductase activity.  相似文献

The influence of extracts from oak bark, St. John's-wort leaves and pine buds on natural immunity characteristics of mice has been studied. The injection of these extracts into mice has been found to enhance their resistance to infection with Staphylococcus aureus and Bordetella pertussis virulent cultures, to decrease the enzymatic activity of 5'-nucleotidase in the peritoneal exudate macrophages of mice and to increase the level of lysozyme in their blood. The action of these extracts has proved to depend on their dosage and the time of observation.  相似文献

Gibberellin (GA)-like substances were analyzed in extracts from cotyledons and phloem exudate collected from cotyledons in photoinduced and vegetative seedlings of the short-day plant Pharbitis nil Chois. var. Violet, using high performance liquid chromatography (HPLC) and the dwarf rice bioassay, to see whether any specific GA-like substances were transported from the photoinduced cotyledons via phloem. Cotyledon extracts exhibited five peaks of free GA-like activity in HPLC, whereas only one or two active peaks were detected in phloem exudate extracts. The level of free GA-like activity was considerably lower in phloem exudate than in the cotyledons. In five out of six analyses of cotyledons and phloem exudate, there were substantially higher levels of free GA-like substances in photoinduced plants. Conjugated GA-like substances were present in much higher levels than free GA-like substances in the cotyledon extracts but the levels were not influenced by daylength. In phloem exudate extracts there was no conjugated GA-like substances. The free GA-like substances that are transported via phloem cochromatographed with GA5/20 and GA19 on HPLC. These were significantly higher in photoinduced plants and thus could have some influence on the photoperiodically-induced flowering in P. nil.  相似文献

Water extracts of fresh leaves, buds, and leaf litter of Populus balsamifera (balsam poplar) were tested at different dilutions for allelopathic effects on nodulation, nitrogenase activity and growth of nodulated green alder (Alnus crispa) seedlings, and on growth of unnodulated green alder seedlings. All extracts inhibited height growth, root elongation and dry weight increment of nodulated and unnodulated green alder seedlings to some degree during a 2-month experiment. Foliar nitrogen content of both nodulated and unnodulated seedlings was significantly lower in extract treated plants than in controls. Growth inhibition was about 25% less in nodulated than in unnodulated seedlings. The number of nodules per plant in seedlings treated with any balsam poplar extract was only 51% that of control plants. Acetylene reduction by seedlings treated with bud and leaf litter extracts indicated a decrease of 62% compared to controls. Growth inhibition was not mediated by pH or osmotic effects of the extracts. The possible ecological significance is discussed.  相似文献

The cytokinin content of the root exudate and leaves of fruiting white lupin plants (Lupinus albus L.) was investigated at 2 weekly intervals after anthesis of the lowest flower on the primary inflorescence. Up to 8 weeks after anthesis the level of cytokinins in the root exudate increased. However, at 10 weeks after anthesis insufficient sap was produced for analysis. Cytokinins co-eluting with zeatin and zeatin riboside were detected in the root exudate after fractionation on Sephadex LH-20. The cytokinin levels in the mature leaves steadily increased up to 8 weeks after anthesis and thereafter remained relatively constant. Three peaks of activity, co-eluting with zeatin, zeatin riboside and the glucoside cytokinins were recorded in the leaf extracts. The level of glucoside cytokinins in the leaves was high at 8 and 10 weeks after anthesis. Paper chromatography of extracts of fruits collected at 2 weeks after anthesis indicated that as fruit development proceeded there was a build up of cytokinin in this region of the plant. It is suggested that, in the white lupin, the cytokinins translocated to the shoot are accumulated in the leaves and in the fruits and that it is only later when there is a considerable decrease in sap (10 weeks after anthesis) production that a decreasing supply of cytokinins leads to shoot senescence.  相似文献

Sinapis alba is a long-day (LD) plant that can be induced to flower by a single LD. A number of changes normally occurring in the meristem of plants subjected to the LD can be produced in short day by a single application of a cytokinin to the apical bud. However, flower buds are not produced indicating that evocation by the cytokinin is only partial. In this work, the cytokinin content of root exudate, obtained under vacuum, and of leaf exudate, obtained by the EDTA-method, has been analyzed comparatively in vegetative and induced plants, using reversed-phase HPLC coupled to the Amaranthus bioassay. The results show that, as early as 16 hours after the start of the LD, there is an increase of cytokinin activity in both the root and leaf exudates of induced plants. These observations fit nicely with previous results obtained on Sinapis, and they indicate that cytokinins are part of the floral stimulus in this species.  相似文献

Summary The levels of gibberellin (GA)-like activity in the root exudate of two seedless varieties of Vitis vinifera were examined by the barley endosperm assay, and compared with levels determined for other parts of the plant. That activity was due to GA-like substances was confirmed with dwarf-5 corn.When acidic, ethyl acetate soluble GA-like substances from sap and leaf extracts were chromatogrammed on thin layers of silica gel in chloroform/ethyl acetate/formic acid (50:50:1), activity moved to the same Rf as GA3 and GA1 (Rf 0.05–0.25). However, substances inhibitory to the barley endosperm assay were detected in both sap and leaf extracts. In the above solvent system the inhibitor(s) co-chromatogrammed with a GA1/GA7 mixture, and with abscisin II. The GA-like activity co-chromatogrammed with GA3 on paper developed in isopropanol/ammonia/water (10:1:1).Calculations on the rate of gibberellin movement from the roots seemed to be compatible with levels of activity in the leaves, although these levels could also be a reflection of the general gibberellin level in the plant.The relevance of the findings is discussed.  相似文献

  • Location and degree of protection of aerial buds are important functional traits in disturbance- or stress-prone environments since aerial buds ensure the development of new organs under favourable growing conditions. This study was carried out in a Brazilian Cerrado area under regeneration after long-term Pinus cultivation, where the trees were clear-cut in 2012 and the remaining material was burned in 2014.
  • After the fire treatment, several species resprouted from belowground organs and their aboveground organs were directly exposed to full sunlight. We collected 15 terminal branches with fully expanded leaves from three individuals of each of three Eugenia species to investigate if those with well-developed belowground organs invest in bark for aboveground bud protection. The samples were analysed using light and electron microscopy.
  • In addition to terminal and axillary buds, all species presented accessory buds, and the number varied according to the node analysed. None of the aerial buds were protected by bark, but all were well protected by cataphylls and densely pubescent leaf primordia. There were also inter- and intra-petiolar colleters that released a mucilaginous protein exudate. The distance between the shoot apical meristem and the outer surface was longer in the terminal bud than in axillary buds. The bud leaf primordia covering the shoot apical meristem had a thick cuticle, unicellular non-glandular trichomes that accumulate phenolic and lipophilic compounds, and secretory cavities.
  • Our study shows that all three Eugenia species studied here had highly protected aerial buds allocated from belowground organs. These morphological traits may improve the chances of the species' persistence in areas subjected to frost events, low relative humidity, high irradiance and harmful UV levels.

Stems and buds of Glossopteris skaarensis Pigg and buds of G. schopfii Pigg from the Permian Skaar Ridge locality in the central Transantarctic Mountains, Antarctica demonstrate the first anatomically preserved glossopterids known with stem/leaf attachment. Stems of G. skaarensis are 1–12 mm in diameter ( = 3.1 mm) with a broad pith, poorly defined primary xylem, and a zone of secondary xylem up to 6 mm thick. Pycnoxylic wood conforming to Araucarioxylon Kraus is composed of tracheids with uni- to biseriate oval to hexagonal bordered pits on radial walls, uniseriate rays one to a few cells high, and cupressoid to taxodioid cross-field pitting. Stems have a narrow zone of secondary phloem, aerenchymatous cortex with scattered sclereids, and sometimes a narrow periderm. Two wedge-shaped leaf traces each bifurcate to form four strands in the base of each petiole. Small axillary branches are vascularized by double branch traces that fuse at the margin of the main axis. Buds of G. skaarensis have leaves with narrow lateral laminae and a thickened midrib containing a wide lacuna, delicate vascular strands, and a prominent hypodermis. In contrast, buds of G. schopfii have uniformly thick leaves with prominent, circular vascular bundle sheaths. These anatomical details are used to reconstruct individual types of glossopterid plants, providing new information toward understanding the ecology and evolution of this important group of Permian seed plants.  相似文献

Removal of fruit from potted cuttings of Vitis vinifera L. increased the concentration of a cytokininglucoside in leaf tissue extracts and decreased the level of extractable gibberellin-like substances. The glucoside (of zeatin riboside) is not present in xylem exudate of V. vinifera L., and appears to be synthesized in the leaves. Berry extracts contain zeatin-riboside and smaller amounts of cytokinin-glucoside. Immunodiffusion tests were used for studying protein composition of apical buds ofRudbeckia bicolor andPerilla nankinensis during their transition from vegetative to reproductive state under inductive photoperiodic conditions or GA3 treatment. In both species the induced buds differ from the vegetative ones in the presence of specific proteins (P): P1, P2, P3 appear inRudbeckia apical buds 2, 8, 16 d after the start of inductive treatment; P4 appears inPerilla apical buds 6 d after inductive treatment. P1, P2, P4 are revealed in induced buds in the early period of apex development when morphogenetic changes are not yet present. The similarity between antigenic spectra of induced buds and of those treated by GA3 appears only inRudbeckia. These observations support the hypothesis of a change in gene expression at floral evocation. The phloem exudate prepared from the cotyledons of Pharbitisseedlings that had been exposed to a single dark period (oflonger than 10 h) induced flowering in cultured apices excisedfrom non-induced seedlings. The flower-inducing activity ofthe exudate increased as the seedlings were exposed to longerperiods of darkness. The highest activity was associated withthe exudate taken from cotyledons exposed to a single 16-h darkperiod. The activity of the exudate taken from cotyledons exposedto an inductive dark period was clearly reduced by interruptionof the dark period. The addition of exudate taken from threecotyledons to 10 ml of medium resulted in the highest flower-inducingactivity. About 50% of cultured apex explants formed floralbuds, even when the concentration of the exudate was reducedto 0.1 cotyledon equivalents per 10 ml of medium. Summary The effects of the kinin 6-benzylamino-purine and of 1-naphthaleneacetic acid (NAA) on the epiphyllous bud formation in Bryophyllum were studied under controlled environment.In B. daigremontianum which requires long days for epiphyllous budding, buds were formed under continuous short days after application of the kinin. Similarly, such a treatment caused budding in attached non-aging leaves of B. calycinum which normally form buds only after detachment from the plant. This stimulatory effect of the kinin was strictly bound to the treated leaves (or leaf parts), which also showed an increased growth compared with the opposite non-treated leaves. Root formation in the developing buds was inhibited by the kinin.In both species NAA inhibited the epiphyllous budding under inductive conditions. A similar inhibitory effect was exerted by terminal and axillary buds.The results are discussed in the light of other investigations in this and related fields. It is concluded that epiphyllous bud formation is under the control of a correlative inhibition similar to apical dominance. It is further concluded that even though day-length controls both flowering and epiphyllous budding in B. daigremontianum the two processes must be affected through different biochemical systems.  相似文献

The cytokinin activity of the root exudate, the leaves, and the apices of vegetative and flowering white lupin plants (Lupinus albus L.) was investigated. The level of cytokinin activity in the root exudate decreased over the 11-week experimental period. Four peaks of cytokinin activity were recorded in the root exudate of 8-week-old plants after fractionation on Sephadex LH-20. Two of these peaks co-eluted with zeatin and zeatin riboside. It is suggested that the remaining peaks represent nucleotide and glucoside cytokinins. The cytokinin levels in extracts of the mature leaves fluctuated slightly over the experimental period. Three peaks of activity co-eluting with zeatin, zeatin riboside and the glucoside cytokinins were recorded in extracts of mature leaves of 8-week-old plants. In the apices cytokinin activity could only be detected in the inflorescences of flowering plants. It would appear that cytokinins produced by the roots accumulate in the fully expanded mature leaves, but are utilized in the rapidly growing apical region and in young expanding leaves.  相似文献

A convenient assay method for estimation of barnacle, Balanus amphitrite, settlement inductive activity was developed. To avoid the inductive effect by comrades and laborious observation requirements, cyprid larvae were put individually into wells of a 96-well plate. The settlement ratio from the experiment without any inducers was quite low; therefore this assay allowed easy estimation of settlement inductive activities. Some known inductive agents, such as serotonin and barnacle extracts, clearly showed inductive activities. This assay method is proven to be suitable for estimation of barnacle settlement-inducing activities of both water-soluble and -insoluble compounds. Radioactivity translocation after [14C]-spermidine application over the third trifoliate leaf of soybean plants ( Glycine max . [L.] Merr, cv. Williams) was checked during the first 72 h of short day (SD) treatment to study the involvement of polyamines (PAs) in photoperiodic flowering induction. PAs and/or their metabolites were translocated from the supplied leaf to all parts of the plant. Radioactivity reached its highest concentration in the upper portion of the stem, i.e. the apical bud and the youngest leaf. After the beginning of the first inductive night, the detected radioactivity showed two peaks of maximal concentration. The first arose after the first inductive night, coinciding with the proper flowering induction process; the second one arose after the third inductive night, coinciding with the first morphological symptoms of the transition of vegetative meristems to the reproductive condition. Soluble free PAs showed a different balance in the apical bud of SD-induced plants compared with LD-non induced control plants. Soluble conjugated PAs were detected as traces. It is suggested that under flowering inductive conditions, PAs play a different role according to the stage of the flowering process. Thus, their translocation from the leaves to the axillary and apical buds might be related, in a first step, to the fact that they were part of the complex mechanism of the flowering signal, and in a second step, to the flower transition of vegetative buds.  相似文献
