Tannins exist widely in plants, but because they precipitate proteins, scientists frequently ignore them in search of bioactive components. Catechu, a traditional astringent, is rich in tannins. In this study, we found that condensed tannins from catechu potently inhibited animal fatty acid synthase (FAS). Among them, trimeric condensed tannin showed the most potent inhibition with IC50 of 0.47 μg/ml and it also exhibited strong time-dependent inhibition. Its inhibitory kinetics and reacting sites on FAS were obviously different from the known inhibitors of FAS. Furthermore, condensed tannins were found to suppress the growth of MCF-7 breast cancer cells, and the effect was related to their activity of FAS inhibition. The inhibition of both FAS activity and MCF-7 growth was exhibited by low concentrations of condensed tannins without FAS being precipitated. These results suggest tannins would be a valuable resource of bioactive substances.  相似文献

The ability of foliar tannins to increase plant resistance to herbivores is potentially determined by the composition of the tannins; hydrolyzable tannins are much more active as prooxidants in the guts of caterpillars than are condensed tannins. By manipulating the tannin compositions of two contrasting tree species, this work examined: (1) whether increased levels of hydrolyzable tannins increase the resistance of red oak (Quercus rubra L.), a tree with low resistance that produces mainly condensed tannins, and (2) whether increased levels of condensed tannins decrease the resistance of sugar maple (Acer saccharum Marsh.), a tree with relatively high resistance that produces high levels of hydrolyzable tannins. As expected, when Lymantria dispar L. caterpillars ingested oak leaves coated with hydrolyzable tannins, levels of hydrolyzable tannin oxidation increased in their midgut contents. However, increased tannin oxidation had no significant impact on oxidative stress in the surrounding midgut tissues. Although growth efficiencies were decreased by hydrolyzable tannins, growth rates remained unchanged, suggesting that additional hydrolyzable tannins are not sufficient to increase the resistance of oak. In larvae on condensed tannin-coated maple, no antioxidant effects were observed in the midgut, and levels of tannin oxidation remained high. Consequently, neither oxidative stress in midgut tissues nor larval performance were significantly affected by high levels of condensed tannins. Post hoc comparisons of physiological mechanisms related to tree resistance revealed that maple produced not only higher levels of oxidative stress in the midgut lumen and midgut tissues of L. dispar, but also decreased protein utilization efficiency compared with oak. Our results suggest that high levels of hydrolyzable tannins are important for producing oxidative stress, but increased tree resistance to caterpillars may require additional factors, such as those that produce nutritional stress.  相似文献

Fourteen chemically defined hydrolyzable tannins and six impure mixtures of either condensed or hydrolyzable tannins were found to inhibit the gibberellin-induced growth of light-grown dwarf pea seedlings. The highest ratio of tannins to gibberellic acid tested (1000: 1 by weight) inhibited from 80 to 95% of the induced growth for all tannins tested except for two monogalloyl glucose tannins which inhibited only 50% of the induced growth. The lowest ratio tested (10: 1) inhibited the induced growth by less than 25% except for the case of terchebin where 50% inhibition was found. The inhibition of gibberellin-induced growth was found to be completely reversed by increasing the amount of gibberellin in three cases tested. Tannins alone did not inhibit endogenous growth of either dwarf or nondwarf pea seedlings. Eight compounds related to tannins, including coumarin, trans-cinnamic acid, and a number of phenolic compounds were also tested as gibberellin antagonists. Most of these compounds showed some inhibition of gibberellin-induced growth, but less than that of the tannins. At the highest ratio (1000: 1) the greatest inhibition was 55%; at the lowest ratio (10: 1) no more than 17% was observed. These compounds did not inhibit endogenous growth, and the inhibition of gibberellin-induced growth could be reversed by increasing the amount of gibberellin in two cases tested.  相似文献

以短枝木麻黄(Casuarina equisetifolia)小枝中提取的单宁处理其幼苗,探讨其化感作用及其对幼苗单宁含量的影响.结果表明,单宁对木麻黄幼苗的生长有显著抑制作用,且随着处理浓度的升高,抑制作用逐渐增强,处理15 d后,单宁的化感作用对芽长的抑制程度显著高于根长.单宁处理后,幼苗体内单宁含量也发生显著变化,随着处理浓度的升高而逐渐降低,尤其是总酚和可溶性缩合单宁的含量,单宁溶液的浓度与被处理幼苗体内单宁含量间有显著的线性负相关.因此,短枝木麻黄体内的单宁会对同种的幼苗产生化感作用,这种化感作用不仅影响幼苗的生长和发育,也会通过影响幼苗体内次生代谢物质的形成而影响幼苗对食草动物和其他不利因素的抵抗能力.  相似文献

Tannins are water-soluble polyphenolic compounds having wide prevalence in plants. Hydrolysable and condensed tannins are the two major classes of tannins. These compounds have a range of effects on various organisms – from toxic effects on animals to growth inhibition of microorganisms. Some microbes are, however, resistant to tannins, and have developed various mechanisms and pathways for tannin degradation in their natural milieu. The microbial degradation of condensed tannins is, however, less than hydrolysable tannins in both aerobic and anaerobic environments. A number of microbes have also been isolated from the gastrointestinal tract of animals, which have the ability to break tannin-protein complexes and degrade tannins, especially hydrolysable tannins. Tannase, a key enzyme in the degradation of hydrolysable tannins, is present in a diverse group of microorganisms, including rumen bacteria. This enzyme is being increasingly used in a number of processes. Presently, there is a need for increased understanding of the biodegradation of condensed tannins, particularly in ruminants.  相似文献

Strategies are sought to reduce intestinal colonisation of food-producing animals by Campylobacter jejuni, a leading bacterial cause of human foodborne illness worldwide. Presently, we tested the antimicrobial activity of hydrolysable-rich blackberry, cranberry and chestnut tannin extracts and condensed tannin-rich mimosa, quebracho and sorghum tannins (each at 100 mg/mL) against C. jejuni via disc diffusion assay in the presence of supplemental casamino acids. We found that when compared to non-tannin-treated controls, all tested tannins inhibited the growth of C. jejuni and that inhibition by the condensed tannin-rich mimosa and quebracho extracts was mitigated in nutrient-limited medium supplemented with casamino acids. When tested in broth culture, both chestnut and mimosa extracts inhibited growth of C. jejuni and this inhibition was much greater in nutrient-limited than in full-strength medium. Consistent with observations from the disc diffusion assay, the inhibitory activity of the condensed tannin-rich mimosa extracts but not the hydrolysable tannin-rich chestnut extracts was mitigated by casamino acid supplementation to the nutrient-limited medium, likely because the added amino acids saturated the binding potential of the condensed tannins. These results demonstrate the antimicrobial activity of various hydrolysable and condensed tannin-rich extracts against C. jejuni and reveal that condensed tannins may be less efficient than hydrolysable tannins in controlling C. jejuni in gut environments containing high concentrations of amino acids and soluble proteins.  相似文献

Phytochemical coevolution theory posits that specialist herbivores will be less sensitive than generalists to the defensive compounds of their host. On the other hand, both types of herbivores should allegedly be similarly sensitive to ‘quantitative’ defences, such as tannin compounds. In this paper, we critically examine the biological effects of two types of tannins: vescalagin (a quantitatively dominant hydrolysable tannin of Quercus robur), and a mix of condensed tannins. In a phylogenetically controlled design, we compare the response of two specialist moth species (Dichonia aprilina and Catocala sponsa) and two generalist species (Acronicta psi and Amphipyra pyramidea) to four artificial diets: a control diet, a diet with 50 mg g?1 vescalagin, a diet with 15 mg/g condensed tannins, and a diet with both 50 mg g?1 vescalagin and 15 mg g?1 condensed tannins. Overall, we find drastic effects of vescalagin and pronounced differences in the responses of generalist and specialist herbivores, but no detectable effects of condensed tannins, and no interaction between the two types of compounds. More specifically, vescalagin reduced the growth of generalist species to one‐half of control levels over 72 h. The compound served as a strong feeding deterrent to generalists, reducing ingestion rates by two‐thirds. Vescalagin also reduced the metabolic and growth efficiency of generalist species to between 16% and 56% of control levels – effects which were lacking or even reversed in specialist species. These patterns suggest that vescalagin forms an important part of the oak's defence against herbivores, and that specialist species have adapted to deal with such substances. In terms of biological effects, condensed tannins seem much less important. Given a quantitative dominance of hydrolysable tannins over condensed tannins in oak leaves, and a seasonal decline in overall tannin levels, these findings contradict the previous notion that widespread spring feeding among oak herbivores could be attributed to tannins.  相似文献

Crude extracts of water and solvent extractable tannin fractions from pine needles were found to contain tannin concentrations of 10.15% and 13.15% tannic acid equivalents respectively. Thin Layer Chromatography revealed the presence of four distinct phenolic compounds, amongst which two were tannic acid like compounds. Both the extracts were found to be inhibitory to several microbes of agricultural importance. Amongst the bacterial strains studied, Azotobacter sp (VL-A2) was able to tolerate upto 1000 ppm of crude tannin concentration without any growth inhibition. While growth of Rhizobium (VL-R1) and Bacillus halodurans (MTCC 7181) was inhibited by crude tannin concentrations of 50 and 100 ppm respectively of both water and solvent extracted tannins. Among the fungal genera, Pleurotus djamor was found to tolerate up to 10000 ppm of crude tannins, while Trichoderma virescens (MTCC 6321) and T. reesii could tolerate up to 3000 ppm of both water extractable and acetone extractable crude tannins without any growth inhibition.  相似文献

The following tannins, Chinese gallotannin, 1,2,3,4,6-pentagalloyl glucose, chebulinic acid, procyanidin dimers, and procyanidin trimers were tested and found to be antagonists of seven gibberellins (GAs). Each tannin inhibited the growth induced by any of the gibberellins GA(1), GA(3), GA(4), GA(7), GA(9), GA(13), and GA(14) in the dwarf pea assay. Endogenous growth was not affected. The highest ratio of tannin to gibberellin tested (1000:1 by weight) inhibited from 60 to 95% of the induced growth for all tannins and all gibberellins tested. The tannins were particularly inhibitory against GA(4) and GA(14) where a ratio of 10:1 (tannins: GA by weight) resulted in up to 85% growth reduction. Inhibition could be completely reversed by increasing the amount of gibberellin in all combinations studied. The procyanidin dimers and trimers were the first purified components of condensed tannins to be tested in this system and were potent inhibitors particularly against growth induced by GA(4) and GA(14). Inhibition by these compounds along with similar inhibition by previously tested hydrolyzable tannins demonstrates that the effect is general to tannins of all classes.  相似文献

Tannins are plant-derived polyphenols with antimicrobial effects. The mechanism of tannin toxicity towards Escherichia coli was determined by using an extract from Acacia mearnsii (Black wattle) as a source of condensed tannins (proanthocyanidins). E. coli growth was inhibited by tannins only when tannins were exposed to oxygen. Tannins auto-oxidize, and substantial hydrogen peroxide was generated when they were added to aerobic media. The addition of exogenous catalase permitted growth in tannin medium. E. coli mutants that lacked HPI, the major catalase, were especially sensitive to tannins, while oxyR mutants that constitutively overexpress antioxidant enzymes were resistant. A tannin-resistant mutant was isolated in which a promoter-region point mutation increased the level of HPI by 10-fold. Our results indicate that wattle condensed tannins are toxic to E. coli in aerobic medium primarily because they generate H2O2. The oxidative stress response helps E. coli strains to overcome their inhibitory effect.  相似文献

Summary Seedlings of Inga oerstediana Benth. (Mimosaceae) growing in three different light environments (the understory, tree-fall gaps and full sun) were tested for differences in chemistry (nutrients and tannins), wound-induced increases in tannins, growth, and susceptibility to leaf-cutter ants, Atta cephalotes (L.) (Formicidae: Attini). I hypothesized that seedlings of I. oerstediana would contain higher concentrations of tannins when growing in high light conditions and, therefore, would be less susceptible to leaf-cutter ants.Foliar concentrations of condensed tannins were much higher in plants growing in full sun compared to those growing in the understory. The concentrations of condensed tannins did not increase following damage. Despite higher concentrations of condensed tannins in sun foliage, leaf-cutter ants found these leaves more acceptable. The preference for sun leaves was consistent with higher concentrations of foliar nutrients. I suggest that the magnitude of the increase in condensed tannins was not great enough to override the benefits of increased concentrations of foliar nutrients. Finally, based on these results and those of others, I suggest that foraging by leaf-cutter ants may be an important factor determining patterns of succession in early successional habitats.  相似文献

Tannins are secondary metabolites that are widely distributed in the plant kingdom. They act as growth inhibitors for many microorganisms: they are released upon microbial attack, helping to fight infection in plant tissues. Extraction of tannins from plants is an active industrial sector with several applications, including oenology, animal feeding, mining, the chemical industry, and, in particular, the tanning industry. However, tannins are also considered very recalcitrant pollutants in wastewater of diverse origin. The ability to grow on plant substrates rich in tannins and on industrial tannin preparations is usually considered typical of some species of fungi. These organisms are able to tolerate the toxicity of tannins thanks to the production of enzymes that transform or degrade these substrates, mainly through hydrolysis and oxidation. Filamentous fungi capable of degrading tannins could have a strong environmental impact as bioremediation agents, in particular in the treatment of tanning wastewaters.

Summary The effects of tannins on survival, growth, and digestion were compared in two polyphagous species of Lepidoptera (one, the southern armyworm, a forb-feeder; and the other, the promethea silkmoth, a tree-feeder). Two different types of tannins (hydrolyzable and condensed) were incorporated into artificial basal diets in order to determine whether or not differential survival and growth would result between the forb feeder, which normally does not encounter tannins in its natural diet, and the tree-feeder, whose host species include many tanniniferous plants from several different families.Neonate larvae of the forb-feeding armyworms exhibited significantly suppressed 10-day growth rates at all tannin concentrations tested (0.5, 1.0, 1.5, 2.0, 2.5, and 3.0% of wet weight) for both the hydrolyzable and the condensed tannin compared to the control diet, however no dose-effect was detectable. In contrast, there were no detectable differences in neonate survival or growth through the first 10 days for the tree-feeding promethea silkmoth larvae fed diets with either tannic acid or quebracho tree condensed tannin.In order to determine the physiological mechanisms of action of these tannins against armyworms, we conducted detailed physiological bioassays of biomass and nitrogen utilization by penultimate instar larvae. Standard gravimetric feeding studies with both tannic acid and the quebracho tree condensed tannin demonstrated that reduced relative growth rates (RGR's) of Spodoptera eridania Cram. were due to the suppressed relative consumption rates (RCR's) and decreased conversion efficiencies (ECD's) rather than due to digestibility-reduction (as reflected by approximate digestibility, AD). As with the neonate larval growth rate suppression, there were no detectable dose responses at the different concentrations of tannic acid (0.25, 0.50, 0.75, 1.00, 2.50, and 5.0 percent) and condensed tannins from quebracho (0.25, 0.50, 0.75, 1.0, and 2.5 percent) in our penultimate instar studies.  相似文献

Two forage legumes, birdsfoot trefoil (Lotus corniculatus L.) and sainfoin (Onobrychis viciifolia Scop.), containing condensed tannins in their leaves and stems were used as source material to study condensed tannins in tissue culture. More protoplasts were isolated from mesophyll tissue of a low tannin-containing strain of birdsfoot trefoil than from a high tannin-containing strain, but more tannin-filled protoplasts were observed in the latter. Growth rates of leaf explant-derived callus tissue were greater for the high-tannin than for the low-tannin strain. In sainfoin, callus cultures from leaf explants produced numerous tannin-filled cells by 21 days. Explants from sainfoin cotyledons and roots, tissues which normally do not contain tannins, also formed callus with tannin-filled cells in 21 days but in almost every case, a cytokinin was required for tannin formation to occur. The occurrence of tannin-filled cells in callus from root and cotyledon explants was variable and genotype specific. We examined the extent to which carbon investment into secondary compounds in loblolly pine (Pinus taeda L.) is changed by the interactive effect of elevated CO2 and N availability and whether differences among treatments are the result of size-dependent changes. Seedlings were grown for 138 days at two CO2 partial pressures (35 and 70 Pa CO2) and four N solution concentrations (0.5, 1.5, 3.5, and 6.5 mmol l−1 NO3NH4) and concentrations of total phenolics and condensed tannins were determined four times during plant development in primary and fascicular needles, stems and lateral and tap roots. Concentrations of total phenolics in lateral roots and condensed tannins in tap roots were relatively high regardless of treatment. In the smallest seedlings secondary compound concentrations were relatively high and decreased in the initial growth phase. Thereafter condensed tannins accumulated strongly during plant maturation in all plant parts except in lateral roots, where concentrations did not change. Concentrations of total phenolics continued to decrease in lateral roots while they remained constant in all other plant parts. At the final harvest plants grown at elevated CO2 or low N availability showed increased concentrations of condensed tannins in aboveground parts. The CO2 effect, however, disappeared when size differences were adjusted for, indicating that CO2 only indirectly affected concentrations of condensed tannins through accelerating growth. Concentrations of total phenolics increased directly in response to low N availability and elevated CO2 in primary and fascicular needles and in lateral roots, which is consistent with predictions of the carbon-nutrient balance (CNB) hypothesis. The CNB hypothesis is also supported by the strong positive correlations between soluble sugar and total phenolics and between starch and condensed tannins. The results suggest that predictions of the CNB hypothesis could be improved if developmentally induced changes of secondary compounds were included. Summary Mangrove-derived tannins negatively effected laboratory-reared nematode populations and natural communities of meiobenthos in tropical mangrove forests along the northeastern coast of Australia. In the low and mid intertidal zones of five mangrove estuaries, nearly all of the dominant meiofaunal taxa correlated negatively with concentrations of sediment tannins. Only nematodes correlated with low tannin concentrations in the high intertidal zones. The negative exponential equation y=be -mx represented the best-fit for most of the meiofauna-tannin relationships. The mangrove-dwelling nematode, Terschellingia longicaudata did not grow (r=0.001) in the laboratory on fresh, tannin-rich leaves of the red mangrove, Rhizophora stylosa. Population growth of the nematode was significantly greater on fresh, tannin-poor leaves of the grey mangrove, Avicennia marina (r=0.081) with best growth (r=0.112) attained on a diet of tannin-free, mixed cereal. These preliminary field and laboratory results suggest that hydrolyzable tannins leached from mangrove roots and leaf litter are an important factor regulating intertidal meiobenthic communities in tropical mangrove forests along the northeastern Australian coast.  相似文献

Plant intraspecific variability has been proposed as a key mechanism by which plants adapt to environmental change. In boreal forests where nitrogen availability is strongly limited, nitrogen addition happens indirectly through atmospheric N deposition and directly through industrial forest fertilization. These anthropogenic inputs of N have numerous environmental consequences, including shifts in plant species composition and reductions in plant species diversity. However, we know less about how genetic differences within plant populations determine how species respond to eutrophication in boreal forests. According to plant defense theories, nitrogen addition will cause plants to shift carbon allocation more towards growth and less to chemical defense, potentially enhancing vulnerability to antagonists. Aspens are keystone species in boreal forests that produce condensed tannins to serve as chemical defense. We conducted an experiment using ten Populus tremula genotypes from the Swedish Aspen Collection that express extreme levels of baseline investment into foliar condensed tannins. We investigated whether investment into growth and phenolic defense compounds in young plants varied in response to two nitrogen addition levels, corresponding to atmospheric N deposition and industrial forest fertilization. Nitrogen addition generally caused growth to increase, and tannin levels to decrease; however, individualistic responses among genotypes were found for height growth, biomass of specific tissues, root:shoot ratios, and tissue lignin and N concentrations. A genotype’s baseline ability to produce and store condensed tannins also influenced plant responses to N, although this effect was relatively minor. High-tannin genotypes tended to grow less biomass under low nitrogen levels and more at the highest fertilization level. Thus, the ability in aspen to produce foliar tannins is likely associated with a steeper reaction norm of growth responses, which suggests a higher plasticity to nitrogen addition, and potentially an advantage when adapting to higher concentrations of soil nitrogen.  相似文献

【目的】桉树枝瘿姬小蜂是我国近年来发现的一种主要危害桉属树种的外来有害生物。本研究旨在通过探究中国桉树枝瘿姬小蜂主要伴生细菌在桉树枝瘿姬小蜂成功定殖中的作用。【方法】测定不同抗性品系桉树的次生代谢物质黄酮和单宁的含量以及易感品系桉树在枝瘿姬小蜂危害前后的含量变化。通过体外抑菌和化学物质降解实验,探究桉树枝瘿姬小蜂主要伴生细菌对抗虫性物质黄酮和单宁的耐受性及降解能力。【结果】抗性品系桉树的黄酮和单宁的含量明显高于易感品系,易感品系在桉树枝瘿姬小蜂危害后黄酮和单宁的含量显著提高;高浓度的黄酮和单宁会抑制桉树枝瘿姬小蜂伴生细菌的生长,在中低浓度黄酮和单宁的条件下,主要伴生细菌能够适应,并继续繁殖;桉树枝瘿姬小蜂伴生细菌具有一定的降解黄酮和单宁的能力,其中细菌Staphylococcus cohnii降解黄酮的能力比Pseudomonas geniculate稍强,而Bacillus wiedannii、Serratia macescens对单宁具有较强的降解能力。【结论】桉树枝瘿姬小蜂侵染桉树后,可以诱导桉树产生抗性,桉树产生大量的次生代谢物质来抵御桉树枝瘿姬小蜂的危害,而桉树枝瘿姬小蜂部分伴生细菌可降解桉树次生代谢物质来帮助小蜂克服植物抗性完成定殖。  相似文献

Zusammenfassung Bestimmungen des Gerbstoffgehalts in vitro kultivierter Calli von Juniperus communis L. innerhalb einer Kulturperiode von 90 Tagen ergaben ein Maximum zu Beginn der Phase stärkeren Wachstums, etwa 30 Tage nach Versuchsbeginn.Durch Veränderungen der Glucosekonzentration des Substrates wird mit der Gerbstoffproduktion gleichzeitig auch das Wachstum der Calli beeinflußt. Deshalb wird eine deutliche Zunahme des Gerbstoffgehalts erst bei einer Steigerung der Kohlenhydratkonzentration von 1 auf 6% sichtbar.
Tannin production in the Callus cultures of Juniperus Communis L.
Summary Callus cultures of Juniperus communis L. were analysed for tannins at intervals during a period of 90 days. The tannin content (mg tannins x 100/mg dry weight) reached a maximum at the onset of rapid growth, 30 days after inoculation, and then decreased. Variation of the glucose concentration in the medium influences both the growth and the production of tannins. An increase of the tannin content can be observed only if the carbohydrate concentration is raised from 1 to 6%.
