Rats trained preoperatively to discriminate between acetic acid and caproic acid and between acetic acid and propionic acid were tested for their memory of these tasks and ability to discriminate between these odorants and between the enantiomers of carvone after receiving large bilateral bulbar lesions that included most of the fatty acid responsive areas identified in prior physiological studies. Concentrations of acid odorants were varied to insure that discrimination was based on the qualitative difference between acids. Experimental rats performed somewhat poorer than controls on the memory test but had no significant deficits in performing the acid discrimination tasks or discriminating between the enantiomers of carvone. These results demonstrate that removal of most bulbar sites identified as responsive to fatty acids and the consequent disruption of patterned input to the bulb is largely without effect on discriminating odor qualities of structurally similar acids.  相似文献

In this review, we use data obtained primarily from humans to argue that sniffs are not merely a stimulus carrier but are rather a central component of the olfactory percept. We argue that sniffs 1) are necessary for the olfactory percept, 2) affect odorant intensity perception and identity perception, 3) drive activity in olfactory cortex, 4) are rapidly modulated in an odorant-dependent fashion by a dedicated olfactomotor system, and 5) are sufficient to generate an olfactory percept of some sort even in the absence of odor.  相似文献

We show in a model of spiking neurons that synaptic plasticity in the mushroom bodies in combination with the general fan-in, fan-out properties of the early processing layers of the olfactory system might be sufficient to account for its efficient recognition of odors. For a large variety of initial conditions the model system consistently finds a working solution without any fine-tuning, and is, therefore, inherently robust. We demonstrate that gain control through the known feedforward inhibition of lateral horn interneurons increases the capacity of the system but is not essential for its general function. We also predict an upper limit for the number of odor classes Drosophila can discriminate based on the number and connectivity of its olfactory neurons. Contrast enhancement via lateral inhibitory circuits is a common mechanism in sensory systems. We here employ a computational model to show that, in addition to shaping experimentally observed molecular receptive fields in the olfactory bulb, functionally lateral inhibitory circuits can also mediate the elemental and configurational properties of odor mixture perception. To the extent that odor perception can be predicted by slow-timescale neural activation patterns in the olfactory bulb, and to the extent that interglomerular inhibitory projections map onto a space of odorant similarity, the model shows that these inhibitory processes in the olfactory bulb suffice to generate the behaviorally observed inverse relationship between two odorants' perceptual similarities and the perceptual similarities between either of these same odorants and their binary mixture.  相似文献

Sensory information is represented in a spatio-temporal code in the antennal lobe, the first processing stage of the olfactory system of insects. We propose a novel mechanism for decoding this information in the next processing stage, the mushroom body. The Kenyon cells in the mushroom body of insects exhibit lateral excitatory connections at their axons. We demonstrate that slow lateral excitation between Kenyon cells allows one to decode sequences of activity in the antennal lobe. We are thus able to clarify the role of the existing connections as well as to demonstrate a novel mechanism for decoding temporal information in neuronal systems. This mechanism complements the variety of existing temporal decoding schemes. It seems that neuronal systems not only have a rich variety of code types but also quite a diversity of algorithms for transforming different codes into each other.  相似文献

Odor presentation generates both fast oscillations and slow patterning in the spiking activity of the projection neurons (PNs) in the antennal lobe (AL) of locusts, moths and bees. Experimental results indicate that the oscillations are the result of the interaction between the PNs and the inhibitory local neurons (LNs) in the AL; e.g., blocking inhibition by application of GABA-receptor antagonists abolishes these oscillations. The slow patterning, on the other hand, was shown to be somewhat resistant to such blockage. In a H-H model, we reproduce both the oscillations and the slow patterning. As previously suggested, the oscillations are the result of the interaction between the PNs and LNs. We suggest that calcium and calcium-dependent potassium channels (found in PNs of bees and moths) are sufficient to account for the slow patterning resistant to the application of GABA-receptor antagonists. The intrinsic bursting property of the PNs, resulting from these additional modeled currents, give rise to another network feature that was seen experimentally in locusts: A relatively small increase in the number of additional generated PN action potentials when LN input is blocked. Consequently, the major effect of network inhibition is to redistribute the action potentials of the PNs from bursting to one action potential per cycle of the oscillations. The coding of odor intensity by an olfactory receptor neuron model was studied under steady-state stimulation. Our model neuron is an elongated cylinder consisting of the following three components: a sensory dendritic region bearing odorant receptors, a passive region consisting of proximal dendrite and cell body, and an axon. First, analytical solutions are given for the three main physiological responses: (1) odorant-dependent conductance change at the sensory dendrite based on the Michaelis-Menten model, (2) generation and spreading of the receptor potential based on a new solution of the cable equation, and (3) firing frequency based on a Lapicque model. Second, the magnitudes of these responses are analyzed as a function of odorant concentration. Their dependence on chemical, electrical, and geometrical parameters is examined. The only evident gain in magnitude results from the activation-to-conductance conversion. An optimal encoder neuron is presented that suggests that increasing the length of the sensory dendrite beyond about 0.3 space constant does not increase the magnitude of the receptor potential. Third, the sensivities of the responses are examined as functions of (1) the concentration at half-maximum response, (2) the lower and upper concentrations actually discriminated, and (3) the width of the dynamic range. The overall gain in sensitivity results entirely from the conductance-to-voltage conversion. The maximum conductance at the sensory dendrite appears to be the main tuning constant of the neuron because it determines the shift toward low concentrations and the increase in dynamic range. The dynamic range of the model cannot exceed 5.7 log units, for a sensitivity increase at low odor concentration is compensated by a sensitivity decrease at high odor concentration.  相似文献

The ability of individuals to learn about chemosensory stimuli in the prenatal, or immediate postnatal, period may be advantageous in acquiring information about "safe" foods after weaning. In this study, we examined the influence of perinatal exposure to aniseed via the mother's diet on a two-choice food test in the domestic dog. Pups were tested at 10 weeks of age following "prenatal" exposure to aniseed (the last 20 days of gestation), "postnatal" exposure to aniseed (the first 20 days after birth), "perinatal" exposure to aniseed (pre- and postnatal exposure combined), or no exposure to aniseed prenatally or postnatally (control). Perinatal exposure resulted in a significantly greater preference for the aniseed food than the other types of exposure. At 10 weeks, there was no evidence for the retention of any prenatal learning of the aniseed. It is suggested that exposure to a chemosensory stimulus across the perinatal period results in a greater effect than simply the sum of pre- and postnatal exposure due to priming of the chemosensory system via prenatal chemosensory experience. Such a system may confer survival advantages by promoting the acquisition of information about safe foods.  相似文献

Olfactory sensory stimulation induces a fast-phase arrest response (FPA-R) of the blowfly heart activity that has been described as a sensitive tool for testing insect reactivity to odor perception. We analyzed FPA-R occurrence to repeated olfactory stimulation with low and high 1-hexanol concentrations that are behaviorally attractant and repellent, respectively, in the blowfly. FPA-R occurrence diminished and ceased with repeated presentations of low and medium odor concentrations, according to dynamics inversely related to odor doses. On the other hand, repeated stimulation with higher odor concentrations induced persistent FPA-Rs. Sensory input amplitude to repeated presentations of singly tested odor concentrations did not change throughout stimulation sessions. A spontaneous restoration of FPA-R to olfactory stimulation was recorded 30 min after cessation of FPA-R to a previous olfactory stimulation session. However, a prompt restoration of FPA-R to olfactory stimulation after cessation of FPA-R was obtained following mechano-taste stimulation of labellar sensilla. Our findings show that the FPA-R habituates to olfactory sensory stimulation with low and medium odor concentrations according to dynamics inversely related to odor intensities. On the other hand, the FPA-R does not habituate to higher odor concentrations. Therefore, flies learn to disregard nonaversive odor information, but they cannot ignore iterative detection of a repellent volatile.  相似文献

In the mammalian olfactory system, oscillations related to odour representation have been described in field potential activities. Previous results showed that in olfactory bulb (OB) of awake rats engaged in an olfactory learning, odour presentation produced a decrease of oscillations in gamma frequency range (60-90 Hz) associated with a power increase in beta frequency range (15-40 Hz). This response pattern was strongly amplified in trained animals. The aim of this work was twofold: whether learning also induces similar changes in OB target structures and whether such OB response depends on its centrifugal inputs. Local field potentials (LFPs) were recorded through chronically implanted electrodes in the OB, piriform and enthorhinal cortices of freely moving rats performing an olfactory discrimination. Oscillatory activities characteristics (amplitude, frequency and time-course) were extracted in beta and gamma range by a wavelet analysis. First, we found that odour induced beta oscillatory activity was present not only in the OB, but also in the other olfactory structures. In each recording site, characteristics of the beta oscillatory responses were dependent of odour, structure and learning level. Unilateral section of the olfactory peduncle was made before training, and LFPs were symmetrically recorded in the two bulbs all along the acquisition of the learning task. Data showed that deprivation of centrifugal feedback led to an increase of spontaneous gamma activity. Moreover, under this condition olfactory learning was no longer associated with the typical large beta band. As a whole, learning modulation of the beta oscillatory response in olfactory structures may reflect activity of a distributed functional network involved in odour representation.  相似文献

To study the projection of a special type of sensory neuron called crypt cells in the olfactory system in crucian carp, Carassius carassius, we applied the neural tracer 1,1-dilinoleyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI) in the olfactory bulb (OB). Small crystals of DiI were applied in a small area at the synaptic region at the ventral part of the OB, where a population of secondary neurons specific for sex pheromones has been identified. In those samples (4 out of 24) where only axons in the lateral bundle of the medial olfactory tract were stained, the majority (50-66%) of olfactory sensory neurons stained were crypt cells situated in the peripheral layer of the olfactory epithelium. Because this bundle of the tract mediates reproductive behavior, it is conceivable that crypt cells express olfactory receptors for sex pheromones.  相似文献

Odors can have repulsive effects on rodents based on two complementary adaptive behaviors: the avoidance of predator odors (potentially dangerous) and the avoidance of trigeminal stimulants (potentially noxious). The present study aimed to compare the behavioral effects on mice of odors according to their trigeminal properties and ecological significance. We used three different odors: 2,4,5-trimethylthiazoline (TMT: a fox feces odor frequently used to elicit fear-induced behaviors), toluene (a strong stimulant of the trigeminal system) and phenyl ethyl alcohol (PEA: a selective stimulant of the olfactory system). First, we checked preference and avoidance behaviors in mice with and without anosmia towards these odors to ensure their olfactory/trigeminal properties. Secondly, we used a standard test (open-field and elevated plus-maze) to assess the behaviors of mice when exposed to these odors. The results show that the anosmic and control mice both avoided TMT and toluene odors. In the open-field and the elevated plus-maze, mice exhibited "anxious" behaviors when exposed to TMT. Conversely, exposure to PEA induced "anxiolytic" effects confirmed by low blood corticosterone levels resulting from completion of the elevated plus-maze. Compared with TMT exposure, toluene exposure induced moderate "anxious" effects.  相似文献

Rats trained to detect propyl acetate and valeric acid and to discriminate between propyl acetate and amyl acetate and between valeric acid and butyric acid were injected with a low dose of 3-methyl indole, a treatment that produces well-defined and selective deafferentation of the olfactory bulbs. Treatment completely deafferented most but not all bulbar loci for aliphatic acids and at least disrupted those for propyl and amyl acetate. In posttreatment tests, experimental rats performed somewhat but not significantly more poorly than controls and about as well on the acid detection and discrimination tasks as on the corresponding acetate tests.  相似文献

Animals were trained to discriminate two natural odors while another group was trained to discriminate between a patterned electrical stimulation distributed on the lateral olfactory tract (LOT), labelled olfaco-mimetic stimulation (OMS), used as an olfactory cue versus a natural odor. No statistically significant difference was observed in behavioral data between these two groups. The animals trained to learn the meaning of the OMS exhibited a gradual long-term potentiation (LTP) phenomenon in the piriform cortex. When a group of naive animals was pseudo-conditioned, giving the OMS for the same number of sessions but without any olfactory training, no LTP was recorded. These results indicate that the process of learning olfactory association gradually potentiates cortical synapses in a defined cortical terminal field, and may explain why LTP in the piriform cortex is not elicited by the patterned stimulation itself, but only in an associative context. As olfactory and hippocampus regions are connected via the lateral entorhinal cortex, the olfactomimetic model was used to study the dynamic of involvement of the dentate gyrus (DG) in learning and memory of this associative olfactory task. Polysynaptic field potentials, evoked by the LOT stimulation, were recorded in the molecular layer of the ipsilateral DG. An early and rapid (2nd session) potentiation was observed when a significant discrimination of the two cues began to be observed. The onset latency of the potentiated response was 30–40 ms. When a group of naive animals was pseudoconditioned, no change was observed. Taken together, these results support the hypothesis that early activation of the DG during the learning of olfactory cue allows the progressive storage of olfactory information in a defined set of potentiated cortical synapses. The onset latency of the polysynaptic potentiated responses suggests the existence of a reactivating hippocampal loops during the processing of olfactory information.  相似文献

Understanding biochemical system dynamics is becoming increasingly important for insights into the functioning of organisms and for biotechnological manipulations, and additional techniques and methods are needed to facilitate investigations of dynamical properties of systems. Extensions to the method of Ingalls and Sauro, addressing time-dependent sensitivity analysis, provide a new tool for executing such investigations. We present here the results of sample analyses using time-dependent sensitivities for three model systems taken from the literature, namely an anaerobic fermentation pathway in yeast, a negative feedback oscillator modeling cell-cycle phenomena, and the Mitogen Activated Protein (MAP) kinase cascade. The power of time-dependent sensitivities is particularly evident in the case of the MAPK cascade. In this example it is possible to identify the emergence of a concentration of MAPKK that provides the best response with respect to rapid and efficient activation of the cascade, while over- and under-expression of MAPKK relative to this concentration have qualitatively different effects on the transient response of the cascade. Also of interest is the quite general observation that phase-plane representations of sensitivities in oscillating systems provide insights into the manner with which perturbations in the envelope of the oscillation result from small changes in initial concentrations of components of the oscillator. In addition to these applied analyses, we present an algorithm for the efficient computation of time-dependent sensitivities for Generalized Mass Action (GMA) systems, the most general of the canonical system representations of Biochemical Systems Theory (BST). The algorithm is shown to be comparable to, or better than, other methods of solution, as exemplified with three biochemical systems taken from the literature.  相似文献

Taurine (TAU) is a free amino acid that is particularly abundant in the olfactory bulb. In the frog, TAU is located in the terminations of the primary olfactory axons and in the granular cell layer. TAU action seems to be associated with gamma amino butyric acid (GABA), the main inhibitory neurotransmitter involved in the processing of the sensory signal. The present study was designed to assess the action of TAU in vivo during the olfactory network's stimulation by odors. It was performed by recording the single-unit activity of mitral cells, the main bulbar output neurons. TAU effects were tested on both their spontaneous and odor-induced firing activity. Interactions between TAU and GABA were examined by analyzing TAU effects under the selective blocking action of GABAA or GABAB antagonists. TAU was found to suppress the spontaneous firing of mitral cells, mainly without altering their odor response properties. By testing GABA antagonists, we further show that TAU action is associated with GABAergic inhibitory mechanisms mainly via GABAB receptors. Thus, TAU action clearly reduces background activity in favor of the emergence of the odor-induced activity in the same manner as GABA action does via GABAB receptors. As a conclusion, we propose that, in the frog olfactory bulb, the joint actions of TAU and GABA may favor the processing of the primary sensory information by increasing the signal to noise ratio.  相似文献

In natural conditions, pheromones released continuously by female moths are broken in discontinuous clumps and filaments. These discontinuities are perceived by flying male moths as periodic variations in the concentration of the stimulus, which have been shown to be essential for location of females. We study analytically and numerically the evolution in time of the activated pheromone-receptor (signaling) complex in response to periodic pulses of pheromone. The 13-reaction model considered takes into account the transport of pheromone molecules by pheromone binding proteins (PBP), their enzymatic deactivation in the perireceptor space and their interaction with receptors at the dendritic membrane of neurons in Antheraea polyphemus sensitive to the main pheromone component. The time-averaged and periodic properties of the temporal evolution of the signaling complex are presented, in both transient and steady states. The same time-averaged response is shown to result from many different pulse trains and to depend hyperbolically on the time-averaged pheromone concentration in air. The dependency of the amplitude of the oscillations of the signaling complex on pulse characteristics, especially frequency, suggests that the model can account for the ability of the studied type of neuron to resolve repetitive pulses up to 2 Hz, as experimentally observed. Modifications of the model for resolving pulses up to 10 Hz, as found in other neuron types sensitive to the minor pheromone components, are discussed.  相似文献
