Presented in this paper is a neural network model that can be used to investigate the possible self-organizing mechanisms occurring during the early ontogeny of spinal neural circuits in the vertebrate motor system. The neural circuit is composed of multiple types of neurons which correspond to motorneurons, Renshaw cells and a hypothetical class of interneurons. While the connectivity of this circuit is genetically predetermined, the efficacies of these connections – the synaptic s trengths – evolve in accordance with activity-dependent mechanisms which are initiated by the intrinsic, autonomous activity present in the developing spinal cord. Using Oja's rule, a modified Hebbian learning scheme for adjusting the values of the connections, the network stably self-organizes developing, in the process, reciprocally activated motorneuron pools analogous to those which exist in vivo. Rhythmic limb movements are controlled by pattern-generating neurons within the ventral spinal cord, but little is known about how these locomotor circuits are assembled during development. At early stages of embryogenesis, motor neurons are spontaneously active, releasing acetylcholine that triggers the depolarization of adjacent cells in the spinal cord. To investigate whether acetylcholine-driven activity is required for assembly of the central pattern-generating (CPG) circuit, we studied mice lacking the choline acetyltransferase (ChAT) enzyme. Our studies show that a rhythmically active spinal circuit forms in ChAT mutants, but the duration of each cycle period is elongated, and right-left and flexor-extensor coordination are abnormal. In contrast, blocking acetylcholine receptors after the locomotor network is wired does not affect right-left or flexor-extensor coordination. These findings suggest that the cholinergic neurotransmitter pathway is involved in configuring the CPG during a transient period of development.  相似文献

Metamorphosis in the anuran frog, Xenopus laevis, involves profound structural and functional transformations in most of the organism's physiological systems as it encounters a complete alteration in body plan, habitat, mode of respiration and diet. The metamorphic process also involves a transition in locomotory strategy from axial-based undulatory swimming using alternating contractions of left and right trunk muscles, to bilaterally-synchronous kicking of the newly developed hindlimbs in the young adult. At critical stages during this behavioural switch, functional larval and adult locomotor systems co-exist in the same animal, implying a progressive and dynamic reconfiguration of underlying spinal circuitry and neuronal properties as limbs are added and the tail regresses. To elucidate the neurobiological basis of this developmental process, we use electrophysiological, pharmacological and neuroanatomical approaches to study isolated in vitro brain stem/spinal cord preparations at different metamorphic stages. Our data show that the emergence of secondary limb motor circuitry, as it supersedes the primary larval network, spans a developmental period when limb circuitry is present but not functional, functional but co-opted into the axial network, functionally separable from the axial network, and ultimately alone after axial circuitry disappears with tail resorption. Furthermore, recent experiments on spontaneously active in vitro preparations from intermediate metamorphic stage animals have revealed that the biogenic amines serotonin (5-HT) and noradrenaline (NA) exert short-term adaptive control over circuit activity and inter-network coordination: whereas bath-applied 5-HT couples axial and appendicular rhythms into a single unified pattern, NA has an opposite decoupling effect. Moreover, the progressive and region-specific appearance of spinal cord neurons that contain another neuromodulator, nitric oxide (NO), suggests it plays a role in the maturation of limb locomotor circuitry. In summary, during Xenopus metamorphosis the network responsible for limb movements is progressively segregated from an axial precursor, and supra- and intra-spinal modulatory inputs are likely to play crucial roles in both its functional flexibility and maturation.  相似文献

Although the mammalian locomotor CPG has been localized to the lumbar spinal cord, the functional-anatomical organization of flexor and extensor interneurons has not been characterized. Here, we tested the hypothesis that flexor and extensor interneuronal networks for walking are physically segregated in the lumbar spinal cord. For this purpose, we performed optical recordings and lesion experiments from a horizontally sectioned lumbar spinal cord isolated from neonate rats. This ventral hemi spinal cord preparation produces well-organized fictive locomotion when superfused with 5-HT/NMDA. The dorsal surface of the preparation was visualized using the Ca(2+) indicator fluo-4 AM, while simultaneously monitoring motor output at ventral roots L2 and L5. Using calcium imaging, we provided a general mapping view of the interneurons that maintained a stable phase relationship with motor output. We showed that the dorsal surface of L1 segment contains a higher density of locomotor rhythmic cells than the other segments. Moreover, L1 segment lesioning induced the most important changes in the locomotor activity in comparison with lesions at the T13 or L2 segments. However, no lesions led to selective disruption of either flexor or extensor output. In addition, this study found no evidence of functional parcellation of locomotor interneurons into flexor and extensor pools at the dorsal-ventral midline of the lumbar spinal cord of the rat.  相似文献

Sections of the medulla known as the locomotor strip were investigated during cat experiments using the horseradish peroxidase retrograde transport technique. The enzyme was administered under functional control (stimulating the injection site evoked locomotion). Results showed no evidence of a hypothetical column of cells either along the locomotor strip or medially to the strip. This would cast doubts on the existence of such a strip as a separate structural formation. It was shown by comparing findings from morphological research and experiments involving electrical stimulation that the bulbar locomotor strip may in fact consist of the spinal trigeminal tract and its nucleus. It is postulated that non-specific afferent activation of the brainstem reticular formation plays a crucial role in initiating locomotion.A. A. Summary We have tested the hypothesis that the circadian oscillators in the eyes ofAplysia are coequal driver oscillators for the circadian locomotor rhythm. Three predictions based on this hypothesis were tested. Prediction 1: at a time when the phase difference between the eye rhythms is small, the amplitude of the locomotor rhythm in two eyed animals will be as great or greater than the amplitude in one eyed animals. Prediction 2: the amplitude of the locomotor rhythm of two eyed animals will decline under conditions in which the two eye rhythms become out of phase with each other. Prediction 3: the form of the locomotor rhythm will broaden or become biphasic in two eyed animals when the two eye rhythms become out of phase with each other.None of the predictions was confirmed. One eyedAplysia had higher amplitude locomotor rhythms than two eyedAplysia, even under conditions in which the two eye rhythms were probably not far out of phase with each other. There was no tendency for the amplitude of the locomotor rhythm of two eyed animals to decline under circumstances in which the phase difference between the two eye rhythms changes from less than 4 h to as much as 11.5 h. There was no tendency in two eyed animals for the locomotor rhythm to broaden or become biphasic as the eye rhythms became more out of phase with each other.The results led us to reject the hypothesis that the eyes are co-equal drivers for the locomotor rhythm. The ocular influence on locomotion is more likely to be mediated via mechanisms in the central nervous system that do not faithfully conserve the phase of the eye rhythms. A statistical analysis of unit activity in spinal locomotor centers was undertaken on immobilized thalamic cats at rest and during generation of efferent discharges. Activation of the spinal locomotor generator was accompanied by shortening of interspike intervals in the spike sequences of neurons and a decrease in their fluctuations. Histograms of interspike intervals became more symmetrical under these circumstances and there was a considerable increase in the number of neurons whose activity showed regular fluctuations on autocorrelation histograms. Spike trains at rest were characterized by dependence of successive intervals, which increased during efferent discharge generation. The possible mechanisms of modification of the time structure of unit activity in spinal locomotor centers during their activation are discussed.A. A. Locomotion in mammals is a complex motor act that involves the activation of a large number of muscles in a well-coordinated pattern. Understanding the network organization of the intrinsic spinal networks that control the locomotion, the central pattern generators, has been a challenge to neuroscientists. However, experiments using the isolated rodent spinal cord and combining electrophysiology and molecular genetics to dissect the locomotor network have started to shed new light on the network structure. In the present review, we will discuss findings that have revealed the role of designated populations of neurons for the key network functions including coordinating muscle activity and generating rhythmic activity. These findings are summarized in proposed organizational principles for the mammalian segmental CPG.  相似文献

Correlation analysis of unit activity in spinal locomotor centers was carried out in immobilized thalamic cats. Within a short time interval (the time shift of one spike train relative to the other during plotting of the cross-correlation histogram did not exceed 54 msec) correlation between the spike flows of these cells was absent, irrespective of the distance between them, both at rest and during efferent discharge generation. Spike flows of neurons could correlate only in the case of a long time interval (maximal time shift of one spike train relative to the other not less than 4–8 sec during plotting of the cross-correlation histogram). Weak correlation with a long time interval (4–8 sec) was found between changes in the momentary frequency of a neuron and the intensity of the discharge in the motor nerve, but no correlation was found between changes in momentary frequency of the neuron and intensity of discharge. The possible causes of the absence of correlation with a short time interval and its presence with a long time interval are discussed.A. A. The adult brain is a plastic place. To ensure that the mature nervous system's control of behaviour is flexible in the face of a varying environment, morphological and physiological changes are possible at many levels, including that of the entire cell. In two areas of the adult brain - the olfactory bulb and the dentate gyrus - new neurons are generated throughout life and form an integral part of the normal functional circuitry. This process is not fixed, but highly modulated, revealing a plastic mechanism by which the brain's performance can be optimized for a given environment. The functional benefits of this whole-cell plasticity, however, remain a matter for debate.  相似文献

This paper reports the development of a large-scale model of some spinal reflex circuitry, useful for studying the dynamic interactions among neuronal populations during simple behaviors. The included populations and properties of the neurons and terminals were derived from the literature, mainly on cat spinal cord. The model was conceived as a symmetrical controller of a pair of antagonistic muscles, within the behavioral domain of the stretch and Golgi-tendon-organ reflexes, and was scaled to include realistic numbers of motoneurons. Inputs to the model were fiber populations providing random synaptic drive to some of the populations and sensory stimuli appropriate for the reflexes. The resulting model contained roughly 2300 neurons in six pairs of populations. The total number of connections in the model was about 600 000, and individual postsynaptic potentials were small (0.1–0.6 mV). Model responses were calibrated by examination of their ability to reproduce known aspects of the reflexes. Published algorithms were used to construct the environment, which is easily expandable, in terms of membrane channels, neuronal geometry, and synaptic properties. The system was built to combine a system-level perspective of spinal circuitry with the single-unit perspective common in electrophysiological investigation. It provides a computational tool for system-level investigations of spinal cord similar to the tools available at the level of membrane currents. As compared with sham operated animals, an increase in night and daytime locomotion and a shift of the acrophase of the circadian locomotor rhythm to earlier night hours were observed in pinealectomized rats. A reorganization of the rhythm spectral characteristics took place with increase in the share of ultradian waves (14-20 h) and decrease in the number of short periodic (2-5 h) fluctuations.  相似文献

Hepatic encephalopathy (HE) arises from acute or chronic liver diseases and leads to several problems, including motor impairment. Animal models of chronic liver disease have extensively investigated the mechanisms of this disease. Impairment of locomotor activity has been described in different rat models. However, these studies are controversial and the majority has primarily analyzed activity parameters. Therefore, the aim of the present study was to evaluate locomotor and exploratory behavior in bile duct-ligated (BDL) rats to explore the spatial and temporal structure of behavior. Adult female Wistar rats underwent common bile duct ligation (BDL rats) or the manipulation of common bile duct without ligation (control rats). Six weeks after surgery, control and BDL rats underwent open-field, plus-maze and foot-fault behavioral tasks. The BDL rats developed chronic liver failure and exhibited a decrease in total distance traveled, increased total immobility time, smaller number of rearings, longer periods in the home base area and decreased percentage of time in the center zone of the arena, when compared to the control rats. Moreover, the performance of the BDL rats was not different from the control rats for the elevated plus-maze and foot-fault tasks. Therefore, the BDL rats demonstrated disturbed spontaneous locomotor and exploratory activities as a consequence of altered spatio-temporal organization of behavior.  相似文献
