Background  

Natural disease resistance is a cost-effective and environmentally friendly way of controlling plant disease. Breeding programmes need to make sure that the resistance deployed is effective and durable. Grapevine downy mildew, caused by the Oomycete Plasmopara viticola, affects viticulture and it is controlled with pesticides. Downy mildew resistant grapevine varieties are a promising strategy to control the disease, but their use is currently restricted to very limited acreages. The arising of resistance-breaking isolates under such restricted deployment of resistant varieties would provide valuable information to design breeding strategies for the deployment of resistance genes over large acreages whilst reducing the risks of the resistance being defeated. The observation of heavy downy mildew symptoms on a plant of the resistant variety Bianca, whose resistance is conferred by a major gene, provided us with a putative example of emergence of a resistance-breaking isolate in the interaction between grapevine and P. viticola.  相似文献

Downy mildew (Peronospora parasitica (Pers. ex Fr.) Fr.) is a serious disease of brassicas in several countries. Seedlings are very susceptible to this pathogen and crops require frequent fungicide treatments to reach a good marketable yield. The use of resistant cultivars can be the most economical, reliable and environmental friendly method for managing this disease. In this work 32 Sicilian landraces and 16 commercial cultivars of cauliflower and broccoli (B. oleracea) were screened for downy mildew resistance at the cotyledon stage using one P. parasitica strain from Portugal and one from Sicily (Italy). Seven-day old seedlings were inoculated by deposing a droplet of a spore suspension on the cotyledons, incubated under controlled environment and scored 7 days later using a seven-class scale of interaction phenotype (IP), which took into consideration host response and pathogen sporulation. There were no differences in virulence between the two P. parasitica isolates. Accessions ranged from very susceptible to highly resistant to downy mildew showing a variable number of resistant individuals per accession. Forty accessions were very susceptible to downy mildew and are of no interest as sources of resistance, since most of the seedlings were scored in the most susceptible IP classes. Seven accessions had intermediate resistance and included individuals that expressed some degree of resistance. Accession Cv 90 (Cavolfiore Torino) and Br 63 (Sparaceddu) showed the majority of seedlings in the resistant IP classes and may constitute valuable sources of resistance to downy mildew to be used in breeding programs.  相似文献

The effects of seed dressing, sowing date and cultivar on incidence and severity of downy mildew of pearl millet induced by Sclerospora graminicola and yield were studied in a two-year field trial conducted at the Research farm of University of Maiduguri. The millet cultivars, Ex-Borno, SOSAT-C88, GB 8735 and Gwagwa were each dressed with metalaxyl at 0.75 and 1.50 g a.i./kg seed; and a batch of undressed seeds of each cultivar served as control. Both dressed and undressed seeds were used for dry-planting and wet-planting in early and late seasons. The results showed that seed dressing with the fungicides significantly (p ≤ 0.05) reduced the incidence and severity of downy mildew and increased grain yield. Dry-planting also significantly (p ≤ 0.01) increased grain yield irrespective of disease incidence. Delay in sowing led to a significant reduction in incidence and severity of downy mildew. Differences between the cultivars in relation to incidence and severity of downy mildew and grain yield were significant. SOSAT-C88 developed low or no downy mildew in both seasons. Sowing of dressed SOSAT-C88 as soon as rainfall established appeared most beneficial in the control of downy mildew. Dry- or wet-planting Ex-Borno dressed with any of the metalaxyl formulations proved to be effective for downy mildew management and for high yield.  相似文献

52 entries including landraces, old cultivars and wild accessions of B. oleracea and closely related Brassica species were screened for resistance against downy mildew and clubroot. Several accessions resistant to downy mildew and a few to clubroot were found. Genetic inheritance of the resistance in downy mildew was investigated by screening F1 and BC1F1 offspring from three resistant landrace accessions crossed with both a resistant and a susceptible father. The seedling resistance against downy mildew was found to be inherited recessively. This is a bit surprising as earlier papers mostly report of inheritance controlled by a single dominant gene. Previous screenings of B. oleracea resistance against downy mildew at the cotyledon stage have been done with P. parasitica isolated from B. oleracea as the original host plant. The recessive nature of the cotyledon resistance found in this screening might be due to the fact that the P. parasitica isolate was collected from B. napus fields. The clubroot seedling resistance was found to be controlled by recessive inheritance after screening the F1 offspring, this in agreement with earlier results/reports.  相似文献

Landraces in situ Conservation: A Case Study in High-Mountain Home Gardens in Vall Fosca, Catalan Pyrenees, Iberian Peninsula. Interest in landrace conservation has grown over the last few decades with much research focusing on the maintenance of on-farm crop genetic diversity in the tropics. Research on landraces is less abundant in temperate climates. In this paper we assess landrace conservation status in home gardens in Vall Fosca (Catalan Pyrenees, Iberian Peninsula). We estimate the individual socio-demographic attributes associated with in situ conservation of landraces and explore the reasons for their conservation. Fieldwork was conducted March–September 2008, during which time we surveyed 60 home gardens, owned by 53 tenders from 16 villages. We recorded occurrence, abundance, uses, and management of plants cultivated in home gardens. We also inquired about the informants’ reasons for conserving landraces. We found 148 different species. We identified 39 landraces corresponding to 31 species. Women, people over 65 years of age, experienced gardeners, and people who grow their home garden organically were more likely to conserve landraces than people without those characteristics. Although the informants express a strong preference for landraces, they mainly grow commercial varieties. Landraces seem to be displaced by less labor-intensive commercial varieties.  相似文献

Background  

Due to the variation and mutation of the races of Pseudoperonospora cubensis, downy mildew has in recent years become the most devastating leaf disease of cucumber worldwide. Novel resistance to downy mildew has been identified in the wild Cucumis species, C. hystrix Chakr. After the successful hybridization between C. hystrix and cultivated cucumber (C. sativus L.), an introgression line (IL5211S) was identified as highly resistant to downy mildew. Nucleotide-binding site and leucine-rich repeat (NBS-LRR) genes are the largest class of disease resistance genes cloned from plant with highly conserved domains, which can be used to facilitate the isolation of candidate genes associated with downy mildew resistance in IL5211S.  相似文献

The maize weevil Sitophilus zeamais Motsch. is an important pest of maize that attacks the grain both in the field and during storage. The damage caused by the maize weevil S. zeamais on maize landraces, Amarelo Antigo, Asteca, Caiano, Carioca, and Ferrinho, was evaluated by no-choice tests under laboratory conditions. The commercial varieties Sol da Manh?, BR 106, BR 451, and the synthetics PC 0203 and PC 9903 were evaluated for comparisons with the maize landraces. The parameters evaluated were susceptibility index, number of weevil progeny, development time, weevil progeny dry weight, and grain dry weight loss. The landraces were more susceptible to the maize weevil as compared to the commercial varieties. Based on the cluster analysis, two groups of susceptibility to the maize weevil were observed: one of more susceptible populations formed by local landraces and BR 451, and another less susceptible, with commercial varieties, synthetics, and the landrace Amarelo.  相似文献

Plasmopara halstedii, the causal agent of downy mildew of cultivated sunflower (Helianthus annuus), was documented in Germany for the first time in commercial fields. The pathogen was first observed in the Württemberg area, where races 1 and 4 were identified using a set of differential lines. Later, commercial fields near Baden were found to be infected by race 5, which is the first occurrence of that race outside of North America. With the discovery of race 5, there are now eight races of the sunflower downy mildew fungus that have been found in Europe. The sunflower cultivars most frequently grown in Germany were investigated for resistance to race 1, 4 and 5; while all were resistant to race 1, none were resistant to either race 4 or 5.  相似文献

Among four fungicides, viz. metalaxyl (two formulations), fosetyl-Al, pro-pamocarb and cyomaxanil tested in vitro against sporangial germination inhibition of Sclerospora graminicola, cyomaxanil was found to be most inhibitory. In an artificially contaminated plot, when used as seed treatment or foliar spray for the control of downy mildew of pearl millet, only metalaxyl was effective. Metalaxyl 25 (Ridomil) and metalaxyl 35 (Apron) seed treatments protected the pearl millet plants from downy mildew up to 30 days. As a foliar spray, metalaxyl 25 used once at 20 days or twice after 20 and 38 days of plant growth gave less disease at harvest time. Seed treatment (metalaxyl 25 or 35) followed by one metalaxyl 25 spray was found to be effective in controlling the downy mildew. These treatments improved the growth of plants and yield significantly.  相似文献

Abstract

Downy mildew (Sclerospora graminicola [Sacc.] Schroet.) is a serious agricultural problem for pearl millet (Pennisetum glaucum [L.] R. Br.) grain production under field conditions. Six medicinally important plant species Azadirachta indica, Argemone mexicana, Commiphora caudata, Mentha piperita, Emblica officinalis and Viscum album were evaluated for their efficacy against pearl millet downy mildew. Seeds of pearl millet were treated with different concentrations of aqueous extract of the plants to examine their efficacy in controlling downy mildew. Among the plant extracts tested, V. album treatment was found to be more effective in enhancing seed quality parameters and also in inducing resistance against downy mildew disease. Germination and seedling vigor was improved in seeds treated with V. album extracts over control. Seeds treated with 10% concentration of V. album showed maximum protection against downy mildew disease under greenhouse and field conditions. The downy mildew disease protection varied from 44–70% with different concentrations. Leaf extract of V. album did not inhibit sporulation and zoospore release from sporangia of Sclerospora graminicola, indicating that the disease-controlling effect was attributed to induced resistance. Seed treatment with V. album extract increased pearl millet grain yield considerably. In V. album, treated pearl millet seedlings increased activities of peroxidase, and phenylalanine ammonia-lyase enzyme was detected. FTIR analysis of V. album extracts showed the presence of amides and other aromatic compounds which are antimicrobial compounds involved in plant defense.  相似文献

Downy mildew is a fungal disease of sunflower that can lead to severe yield losses. The damage caused by the pathogen can be controlled by growing resistant sunflower varieties. Gene Pl arg was introgressed into cultivated sunflower from the wild species Helianthus argophyllus and provides resistance against all known downy mildew races. In this study, we used a mapping population from the cross-RHA 419/RHA-N-49. We identified a new co-segregating simple sequence repeat marker ORS675 and confirmed the co-segregation of markers ORS716 and ORS662 with Pl arg gene. The markers were validated on two registered resistant inbred lines RHA 443 and RHA 464, as well as on twenty inbred lines RH 1–20 obtained through methods of classical breeding. Molecular marker ORS716 was assessed for usefulness in selecting resistant progeny in 12 BC populations. Markers were found to be valuable for molecular breeding in diverse genetic backgrounds and enabled transfer of the resistance gene in different sunflower genotypes.  相似文献

Downy mildew, caused by the Oomycete pathogen Pseudoperonospora cubensis, is one of the most destructive diseases of cucumber (Cucumis sativus L.) and muskmelon (C. melo L.). Although the process of pathogenesis is well understood, there are few disease control options available. The development and deployment of resistant cultivars is generally considered to be the best approach to control downy mildew. The recently completed sequencing of the cucumber genome provides a great opportunity for reliable and thorough study of the sequence and function of resistance genes in the Cucurbitaceae, which will help us to understand the resistance mechanisms and metabolic pathways activated by these genes. It can be anticipated that, in the near future, we will have more information about the genetic bases of resistance to downy mildew in Cucumis, which will facilitate efforts to breed for resistance to this pathogen.  相似文献

Pearl millet downy mildew (DM) incidence, severity and yield losses of two pearl millet varieties (local and improved) due to the disease were determined in the field. Significant differences in the disease incidence and severity were recorded in the plots sown with metalaxyl-treated seeds and those sown with non-treated seeds, indicating the efficacy of the fungicide on the fungus. Yield losses due to non-treatment of seeds with metalaxyl was 40.88 and 45.39% in a local variety and 43.00 and 18.60% in an improved variety in the 2000 and 2001 cropping seasons respectively. Significant differences between plots sown with metalaxyl-treated and those sown with non-treated seeds were obtained for other yield components such as 1000-grains weight, panicle length and weight.  相似文献

霜霉病是危害大白菜的三大病害之一,该病的发生会严重影响大白菜的产量及品质,因而研究与霜霉病抗性QTL紧密连锁的分子标记对大白菜抗病新品种培育具有重要意义。该研究在前期工作的基础上,选用高感霜霉病株系91-112、高抗霜霉病株系T12-19以及由二者为双亲构建的DH群体为实验材料,针对大白菜霜霉病抗性主效QTL——BrDW所在的标记区间,利用已有的大白菜基因组信息发展与抗性QTL紧密连锁的分子标记,通过Blast和IMap分析,将与BrDW连锁的RAPD标记K14-1030定位于大白菜KBrB058M10上(位于Contig214上),根据KBrB058M10附近的BAC及BAC-end序列设计引物,结合限制性内切酶酶切及HRM分析方法,筛选得到5个与BrDW连锁的分子标记,包括1个Indel标记Brb062-Indel230,3个CAPS标记Brb094-DraⅠ787、Brb094-AatⅡ666和Brb043-BglⅡ715,1个SNP标记Brh019-SNP137;同时,通过筛选与目标区域具有同源性的Unigene序列得到了1个与BrDW紧密连锁的SSR标记bru1209。标记Brb062-Indel230、Brb094-DraⅠ787、Brb094-AatⅡ666、Brb043-BglⅡ715、Brh019-SNP137和bru1209与RAPD标记K14-1030之间的遗传距离分别为4.3 cM、1.7 cM、5.9 cM、5.9 cM、4.6 cM和0.8 cM,在对DH群体中的抗性株系选择上准确率分别为69.7%、70.9%、72.4%、72.4%、58.3%和74.2%,可应用于分子标记辅助选择,为霜霉病抗性分子育种奠定了良好基础。  相似文献

Summary The influence of different levels of nitrogen on the incidence of downy mildew disease of pearl millet on different varieties was studied under field conditions. Nitrogen nutrition to the host did not have much influence on the disease incidence. Among the three hybrid varieties of pearl millet tested J 1270 exhibited resistance to downy mildew incidence while J 934 and HB3 were found to be highly susceptible.  相似文献

Arachidonic acid (AA) induces hypersensitive response (HR) on coleoptile/root regions of two-day-old pearl millet seedlings. The response is comparable to the HR induced by the downy mildew pathogen, Sclerospora graminicola. A time gap in the appearance of cell necrosis among genotypes of pearl millet was related to the degree of resistance to downy mildew. Based on the time required for the development of necrotic spots induced by AA, the pearl millet genotypes were categorised as highly resistant/resistant (HR in 3–6 h), susceptible (HR in 7–12 h) and highly susceptible (HR in 13 h and above). The percentage disease incidence in each genotype was compared with the time required for the development of AA-induced HR. The appearance of hypersensitive cell necrosis was rapid in genotypes having high resistance to downy mildew and was slow in genotypes with high susceptibility. This simple method of screening various pearl millet genotypes in the absence of the pathogen aids in identifying the downy mildew resistant/susceptible host cultivars without the risk of introducing the virulent race of the pathogen.  相似文献

Six strains of Plant growth promoting Rhizobacteria (PGPR) were tested for their ability to promote growth and induce resistance in pearl millet against downy mildew disease. All the PGRP strains showed a significant (P < 0.01) increase in growth promotion in laboratory as well as greenhouse conditions. Only two strains of Pseudomonas spp., UOM ISR 17 and UOM ISR 23, were capable of protecting pearl millet against downy mildew significantly. Pseudomonas UOM ISR 17 and UOM ISR 23 were able to offer 56.3 and 47.5%, respectively against downy mildew disease. When tested for the time gap needed to offer maximum protection, it was found that both the strains needed four days to offer maximum protection of 73.3% and 59.7%, respectively. While both the Acetobacter strains UOM Ab9 and Ab11 and Azospirillum strain UOM Az3 were able to promote growth and offered disease protection of 39.2, 22.3 and 17.40% respectively, they were not as efficient as the two Pseudomonas strains in protecting pearl millet against downy mildew. Maximum growth promotion was recorded by Pseudomonas spp. UOM ISR 17 with 33.9 cm height which was 44, 45, 42 and 46.8% more in height, fresh weight, dry weight and leaf area over the control which recorded 27 cm height, 8.1 g fresh weight, 2.1 g dry weight and 29 cm2 leaf area, respectively.  相似文献
