从甘蓝型油菜与白菜型油菜的种间杂交获得的甘蓝型油菜(Brassica napus)中发现了雄性不育单株,兄妹交株系和不育株与甘蓝型油菜常规杂交F1和F2株系的育性分离分析表明,该不育材料属于双隐性雄性核不育类型.利用育性分离株系的可育株自交和可育株与不育株间兄妹交等方法筛选出7个纯合可育株系,等位测验表明这7个纯合可育株系(B1~B7)中存在两种基因型:Ms1Ms1ms2ms2和ms1ms1Ms2MS2.该材料对油菜核不育基因定位和杂种优势利用研究有重要意义.  相似文献

用Olympus BH2型光学显微镜对甘蓝型油菜单显性核不育系(GMS)及其等位可育系小孢子发育过程进行解剖学观察,发现在正常小孢子发育过程中,绒毡层在小孢子发育的四分体前后开始解体,为小孢子继续发育提供营养,而不育系小孢子的败育在减数分裂前就已经发生,并且不能形成四分体,小孢子逐渐解体,且小孢子解体在绒毡层解体之前发生,最后花药成为干瘪的空壳,导致不育。  相似文献

以5种不同类型甘蓝型油菜雄性不育系为材料,以保持系作对照,对甘蓝型油菜雄性不育系的花蕾及雌雄蕊干物质变化动态进行了研究。结果表明,干物质重量雄性不育系花蕾和雄蕊显著小于保持系,而雌蕊则明显大于保持系。大多数不育系雌雄蕊的发育曲线存在拐点,它们出现拐点的时期相同,但方向完全相反。暗示可能雄蕊败育后降解的生物大分子物质被部分运到了雌蕊,才引起了雌蕊干物重的异常增加。  相似文献

Male and female sterility was induced in canola or rapeseed(Brassica napus L. cv. Westar) flowers grown under high temperatureconditions (32°C/26°C; day/night). At maturity, themajority of flower buds remained closed but had protruding stigmas.The stamens were reduced in size and the anthers showed abnormalmicrosporogenesis. The gynoecia, although normal in appearance,did not set seed and ovule development was aberrant. Flowersof the ogu CMS line of B. napus also showed similar ovular abnormalitiesunder high temperatures and male fertility was not restoredin them. 在显性细胞核雄性不育系Rs1046A和双低油菜品种Samourai构建的回交分离群体中,运用AFLP和SSR两种标记技术构建了一个甘蓝型油菜(Brassica napus L.)的分子标记遗传连锁图谱。该图谱共包含138个AFL.P标记、83个SSR标记和1个形态标记,分布于18个主要连锁群、2个三联体和1个连锁对上,图谱总长度为2646cM,偏分离标记的比例为11.7%。显性细胞核雄性不育基因Ms被定位到第10连锁群(LG10)上。同时,偏分离标记聚集于第8连锁群(LG8)和第16连锁群(LGl6)的末端,形成了十分明显的偏分离标记密集区域。研究结果对于油菜核不育两型系的分子标记辅助选择育种具有重要意义,同时也为克隆和分离核不育基因以及研究核不育的分子机理打下了良好的基础。  相似文献

甘蓝型油菜细胞质雄性不育材料1575A的分子鉴定   总被引:1,自引:0,他引:1  
根据已报道的油菜育性相关基因orf224、orf138和orf222设计引物对1575A、陕3A、Ogura和nap等4份甘蓝型油菜的mtDNA进行PCR扩增.结果显示:引物对Syworf1382/ Xyworf1382在1575A和Ogura的mtDNA中均有1000 bp左右的扩增片段;测序结果表明,在1575A中的该扩增片段包含已报道基因orf138(登录号为AB055435)的全部编码序列;该引物对在陕3A和nap中都没有相应扩增片段.引物对Syworf222/Xyworf222在除Ogura外的其他3个材料的mtDNA中都有196 bp左右的扩增片段;测序结果显示,扩增产物与已报道基因orf222(登录号为DQ872162)同源性为100%.初步推测1575A属于Ogu CMS的改良类型.  相似文献

采用石蜡切片方法,对甘蓝型油菜隐性上位互作核不育材料1665的可育株与不育株花药进行细胞学观察.结果显示:(1)不育株花药在花粉母细胞减数分裂时期出现异常,部分花粉母细胞细胞分裂相不均等分裂或分裂异常.导致部分四分体形状异常.(2)不育株绒毡层细胞在四分体时期开始生长膨大,单核花粉时期出现液泡化和巨型化,侵占药室,使得小孢子不能正常释放或无法继续发育;部分释放出的小孢子未及时形成花粉壁,阻碍花粉继续发育.不能发育形成二核期和三核期花粉,导致花药败育.  相似文献

对新选育的甘蓝型油菜PL CMS不育系和Pol CMS及Ogu CMS不育系材料花器形态进行比较,并对3种不育系10份材料mtDNA进行RAPD分析.结果表明,新选育的PL CMS不育系与Pol CMS、Ogu CMS在花瓣长和宽、花冠直径、雄蕊大小等形态特征方面有极显著差异;mtDNA分析结果表明,新发现的细胞质雄性不育材料L04-02A、L04-05A、L04-01A为同一类型的细胞质雄性不育系,它们与Pol CMS及Ogu CMS不育材料有很大差异,由此推知PL CMS不育系是一种新型甘蓝型油菜不育系.  相似文献

提取细胞质雄性不育系301A、212A和'陕3A'及其共同保持系'中双2号'线粒体DNA,根据已报道的Polima油菜细胞质雄性不育相关基因orf224序列设计引物,对3种不同来源的线粒体DNA PCR产物进行分析.结果显示,这3种甘蓝型油菜细胞质雄性不育材料所获得片段序列完全一致,且与已报道的Polima油菜线粒体中细胞质雄性不育相关基因orf224序列完全相同.研究表明,不育材料301A从分子角度讲属于Polima系统,不育系301A、212A和'陕3A'线粒体中与细胞质雄性不育相关线粒体基因片段orf224具有高度同源性.  相似文献

以陕西省杂交油菜研究中心选育的单显性核不育油菜分离群体为材料,利用集群分离法(BSA)对该油菜单显性核不育基因进行了RAPD分析。在随机选取的300个10碱基随机引物中,引物S243(5′CTATGCCGAC3′)在可育集团与不育集团间扩增出特异而可重复的1.5kb的多态性片段OPU-031500,而在细胞质雄性不育和其它核不育类型油菜中均未扩增出上述特异性片段,从而确证此RAPD标记OPU-031500。片段是与甘蓝型油菜单显性核不育基因连锁的。将该多态性片段克隆并测序,发现其序列与拟南芥的一段DNA序列高度同源。根据同源序列及测序结果设计两对特异引物(P1/P2和P3/P4),引物P3/P4在可育系中可扩增到约1.5kb的单一特异片断,而在不育系中无带,从而将RAPD标记转化为稳定可靠的SCAR标记。  相似文献

通过对NK58s和NK58F这一对光敏核不育水稻等位突变系的AFLP分析,比较了AFLP,RAPD及RFLP检测DINA多态性的相对效率。结果表明,这三种分子标记的DNA多态性检出效率依次为AFLP>RAPD>RFLP;找出了水稻AFLP分析的最适反应条件;通过AFLP和集群混合分析(Bulked segregating analysis,BSA),筛选出了一批与水稻光敏核不育(PGMS)基因连锁的多态性AFLP产物,已完成了对4个多态性AFLP产物的克隆,Southem杂交证明其中2个为单拷贝顺序,另外2个为低拷贝顺序。对上述三种分子标记各自的优缺点及它们在DNA多态性检测中的适用之处进行了分析探讨。  相似文献

采用双向凝胶电泳对现蕾初期苜蓿雄性不育植株(Ms-4)及其可育植株(MF)花蕾蛋白质进行了分离,获得了分辨率和重复性较好的双向电泳图谱。通过ImageMaster 2D软件对Ms-4和MF银染图谱分析发现,两者在等电点5~7、分子量20~60 kD范围内蛋白质斑点分布最多,可识别的总蛋白质点数均在6 000个左右,其中差异表达的蛋白质点数为98个;进一步通过质谱分析成功鉴定了22个差异蛋白点。利用Blast2GO程序对 22个蛋白点进行功能注释和代谢途径分析发现,核酮糖羧化酶小亚基、尿苷三磷酸-葡萄糖-1-磷酸尿苷酰基转移酶等蛋白在光合作用、碳水化合物代谢、多细胞生物有机体的发育等过程中起着重要的作用,同时参与了细胞质、细胞壁等组成,并具有绑定、催化、结合和水解等功能。研究结果初步推断,在苜蓿花药发育过程中,蛋白的缺失及表达量的变化可能会使与花粉发育有关的能量缺失,物质合成发生改变,导致雄性不育。  相似文献

A dominant genetic male sterility trait obtained through transformation in rapeseed (Brassica napus) was studied in the progenies of 11 transformed plants. The gene conferring the male sterility consists of a ribonuclease gene under the control of a tapetum-specific promoter. Two ribonuclease genes, RNase T1 and barnase, were used. The chimaeric ribonuclease gene was linked to the bialophos-resistance gene, which confers resistance to the herbicide phosphinotricine (PPT). The resistance to the herbicide was used as a dominant marker for the male sterility trait. The study presented here concerns three aspects of this engineered male sterility: genetics correlated with the segregation of the T-DNA in the progenies; expression of the male sterility in relation to the morphology and cytology of the androecium; and stability of the engineered male sterility under different culture conditions. Correct segregation, 50% male-sterile, PPT-resistant plants, and 50% male-fertile, susceptible plants were observed in the progeny of seven transformants. The most prominent morphological change in the male-sterile flowers was a noticeable reduction in the length of the stamen filament. The first disturbances of microsporogenesis were observed from the free microspore stage and were followed by a simultaneous degeneration of microspore and tapetal cell content. At anthesis, the sterile anthers contained only empty exines. In some cases, reversion to fertility of male-sterile plants has been observed. Both ribonuclease genes are susceptible to instability. Instability of the RNase T1-male sterility trait increased at temperatures higher than 25[deg] C. Our results do not allow us to confirm this observation for the barnase male-sterile plants. However, the male-sterile plants of the progeny of two independent RNase T1 transformants were stably male sterile under all conditions studied.  相似文献

玉米雄性不育材料是一种宝贵的种质资源,不育基因的遗传分析与定位研究对玉米分子育种和杂种优势利用具有重要价值。通过对从美国引进的玉米雄性不育突变体材料ms14进行雄花育性鉴定和花药I2-KI染色,表明该突变体是无花粉型雄性不育;通过不育突变体ms14与正常自交系郑58、昌7-2杂交获得F1,然后自交构建两个F2遗传分离群体(ms14×郑58和ms14×昌7-2),并进行雄花育性调查、数据统计和遗传分析,发现可育株数与不育株数的分离比是3∶1,表明该突变体由隐性单基因控制;通过SSR等分子标记与不育位点的连锁分析,将ms14基因定位在玉米第1染色体的SSR标记umc2025和umc1676之间,遗传距离分别是2.2cM和0.3cM。对玉米不育基因ms14的遗传分析和初步定位,为该基因的精细定位和克隆、不育机理的解析及其产业化应用奠定了基础。  相似文献

在甘蓝型油菜杂交种C022(其母本为由3对基因控制育性的隐性细胞核雄性不育系9012A)不育株开放受粉的后代中,发现一种稀有的黄白双色嵌合花瓣的甘蓝型油菜突变体991S。其具有3个形态特征:(1)4片花瓣每片中央均为条带状黄色色斑,而两侧为白色,为嵌合双色花瓣;(2)4片花萼也可发生中间条带状白化;(3)目前只在不同群体的雄性不育株中出现,与同源的黄色花不育株形态相似,植株纤细矮小,花器也较小,花瓣较为平整,雌蕊弯曲,雄蕊退化,花药干缩。通过对其材料来源及后代花色表型分析,初步认为黄白双色性状由可局部表达的隐性白化基因控制。  相似文献

在我们以前研究热激(40℃)诱导雄性不育高粱3197A呈现雄性可育态时的内部物质的变化,即热激蛋白的表现的基础上,进一步研究观察到,在逐渐升温过程中,雄性不育高粱幼苗出现了热激蛋白的先后合成与消失的现象,即有阶段性变化的3次协同调控现象。雄性不育高粱与雄性可育高粱比较,在常温(28℃)不育系的自显影区带33KD、35KD比可育高粱的对应带强,在逐渐升温过程中,两个高粱系的这些蛋白带的差别逐渐缩小,到40℃时,两系的标记带图型趋于一致,表现了化学物质的变化与热激使不育系转变为可育态时的温度是一致的。比较高粱的总蛋白与可溶性蛋白,探明了某些蛋白为结合蛋白。用基因表达抑制剂探明热激反应可能与细胞核基因转录和转译两级调控有关。而与细胞质关系不大。试验探明逐渐升温到46—49℃温度区间,不育系幼穗的蛋白电泳图谱中出现特异的80KD热激蛋白,可育的保持系没有,表明此蛋白质与雄性不育有关。  相似文献

Anther and pollen development in male-fertile and male-sterile green onions was studied. In the male-fertile line, both meiotic microspore mother ceils and tetrads have a callose wall. Mature pollen grains are 2-celled. The elongated generative cell with two bended ends displays a PAS positive cell wall. The tapetum has the character of both secretory and invasive types. From microspore stage onwards, many oil bodies or masses accumulate in the cytoplasm of the tapetal cells. The tapetum degenerates at middle 2-celled pollen stage. In male-sterile line, meiosis in microspore mother cells proceeds normally to form the tetrads. Pollen abortion occurs at microspore with vacuole stage. Two types of pollen abortion were observed. In type I, the protoplasts of the microspores contract and gradually disintegrate. At the same time the cytoplasm of microspores accumulates oil bodies which remain in the empty pollen. The tapetal cells behave normally up to the microspore stage and early stage of microspore abortion, but contain fewer oil bodies or masses than those in the male-fertilt line. At late stage of microspore abortion, three forms of the tapetal ceils can be observed: (1) the tapetal cells with degenerating protoplasts become flattened, (2) the tapetal cells enlarge but protoplasts retractor, (3) the cells break down and tile middle layer enlarges. In type Ⅱ, the cytoplasm degenerates earlier than the nucleus of the microspores and no protoplast is found in the anther locule. There are fibrous thickenings iii the endothecium of both types. It is difficult to verify whether the tapetum behavior and pollen abortion is the cause or the effect.  相似文献
