Homogenates of Chironomus cells synthesize chitin as effectively as intact cells. Chitin is produced in a dose-dependent manner, when GlcN, GlcNAc, or UDP-GlcNAc is used as precursor. Due to the lability of UDP-GlcNAc incorporation of this substrate is underestimated. No allosteric effect is observed when GlcN or GlcNAc is used as a substrate. Chitin synthesis is stimulated by Mg2+ and inhibited by uridine monophosphate (UMP), uridine diphosphate (UDP), and uridine triphosphate (UTP). The apparent temperature optimum is 30°C, the apparent pH optimum is 5.5–6. Addition of the chitinase inhibitor allosamidin does not enhance chitin synthesis significantly. The time course of chitin formation reveals a lag period of about 12 h, which can be overcome by trypsin treatment. Addition of protease inhibitors prevents chitin synthesis.  相似文献

In three separate experiments, the upper leaf surface of the fifth formed leaf of wheat cv. Highbury, the fourth and fifth leaves of barley cv. Julia and the third and fourth leaves of oat cv. Mostyn were inoculated in a spore settling tower with wheat brown rust (Puccinia recondita f. sp. tritici), barley brown rust (P. hordei) or oat crown rust (P. coronata f. sp. avenae), respectively. Fewer pustules developed on distal portions of leaves of plants infected with barley yellow dwarf virus (BYDV) than on similar portions of leaves from virus-free plants. There were no significant differences in the number of pustules on proximal leaf portions. In barley and oats, the number of pustules on distal leaf portions was negatively correlated with the amount of yellowing of the leaf areas scored. In wheat, symptoms of BYDV were mild and leaves were little affected by yellowing. The latent period of rust on wheat and oats was not affected by BYDV. In barley, BYDV reduced the latent period of rust on leaf 5, but not on leaf 4, and reduced it on proximal, but not distal, leaf portions. In other experiments, BYDV reduced the yield of wheat and oats by 44% and 66%, respectively, while BYDV-infected barley was almost sterile. The appropriate rust reduced the yield of wheat, barley and oats by 33%, 13% and 86%, respectively. When infected with both BYDV and rust, yield of wheat and oats was reduced by 63% and 91%, respectively. Neither BYDV nor rust affected the percentage crude protein content of wheat grain, nor did rust affect that of barley. In oats, BYDV and rust each significantly increased crude protein of grain, but rust infection of BYDV-infected plants tended to reduce it.  相似文献

The shoot development ofAucuba japonica was studied morphologically. The shoot shows dichasial branching in connection with the formation of a terminal inflorescence and shows a decussate phyllotaxis even in the reproductive phase. The sequence of initiation of successive foliar appendages is very precise, hence the foliage leaf, scale leaf and bract can be compared with each other even at their stages of initiation. In the stage of proximal foliage leaf formation the shoot apex is flat, while in the stage of formation of distal foliage leaves, bud scales and proximal bracts, it becomes concave. In the stage of formation of distal bracts the apex becomes domed. Plastochron durations are relatively long in the vegetative phase in comparison with other plants, and the duration from initiation of the first pair of appendages to that of the second is about one and a half months. Both male and female inflorescences exhibit basically a thyrsoid type of monotelic synflorescence.  相似文献

  • One benefit of clonal integration is that resource translocation between connected ramets enhances the growth of the ramets grown under stressful conditions, but whether such resource translocation reduces the performance of the ramets grown under favourable conditions has not produced consistent results. In this study, we tested the hypothesis that resource translocation to recipient ramets may reduce the performance of donor ramets when resources are limiting but not when resources are abundant.
  • We grew Mikania micrantha stolon fragments (each consisting of two ramets, either connected or not connected) under spatially heterogeneous competition conditions such that the developmentally younger, distal ramets were grown in competition with a plant community and the developmentally older, proximal ramets were grown without competition. For half of the stolon fragments, slow‐release fertiliser pellets were applied to both the distal and proximal ramets.
  • Under both the low and increased soil nutrient conditions, the biomass, leaf number and stolon length of the distal ramets were higher, and those of the proximal ramets were lower when the stolon internode was intact than when it was severed. For the whole clone, the biomass, leaf number and stolon length did not differ between the two connection treatments. Connection did not change the biomass of the plant communities competing with distal ramets of M. micrantha.
  • Although clonal integration may promote the invasion of M. micrantha into plant communities, resource translocation to recipient ramets of M. micrantha will induce a cost to the donor ramets, even when resources are relatively abundant.

In Ascaris suum chitin is formed in the zygote immediately after oocyte fertilization, and its synthesis is completed in the eggs from the distal half of the uterus. Incorporation of radiocarbon [14C] glucose into chitin of the eggshell was 40-fold higher than incorporation of [14C] glucosamine. The same rank order also holds for the incorporation of label from these isotopes into the glycogen of the ovaries. A large part of the radiolabel was incorporated first into oocyte glycogen and only after fertilization was it incorporated into eggshell chitin. Actinomycin D inhibited chitin synthesis in the eggs from the distal half of the uterus and it significantly reduced incorporation of radiocarbon from glucose into chitin.  相似文献

Ethanol-soluble carbohydrates in healthy and blast-infected leaves and also cultures of Pyricularia oryzae were analyzed using gas chromatography. Arabitol, mannitol, and trehalose occurred in infected leaf tissue, but not in healthy controls. Cultures of P. oryzae contained mannitol and trehalose, but not arabitol and sucrose. The presence of arabitol only in blast-infected rice leaves suggests that arabitol synthesis may be induced in infected leaf tissue as a result of the rice-blast fungus interaction, but may not be within blast fungus itself. The amounts of glucose, fructose, and sucrose in infected leaves were slightly increased until 5 days, and greatly enhanced at 7 days after inoculation, There were no differences in amounts of these sugars between the cultivars Nakdong and Dobong. At 7 days after inoculation, increases in amounts of arabitol, mannitol, and trehalose were pronounced in the susceptible cultivar Nakdong than in the moderately susceptible cultivar Dobong. The increased amounts of glucose and fructose in infected plants of the two cultivars were closely correlated with the presence of a very active invertase.  相似文献

The area covered by visible mycelium of E. cichoracearum on the upper surface of leaves 4, 8, 12 and 16 of tobacco plants in field plots in Rhodesia was expressed as percentages of the proximal and distal halves at weekly intervals. Free amino nitrogen and carbohydrate in discs from proximal and distal halves of the same leaves were analysed when each leaf was expanding rapidly and was not infected, and several weeks later, when the rate of expansion had slowed down and there was slight infection. On two other occasions, similar leaf discs were inoculated with conidia, to measure the percentage germination and hyphal length from individual conidia after incubation for 2–3 days at constant temperature and humidity; duplicate discs were chemically analysed. Leaves were not susceptible until at least 6 weeks after they had emerged from the bud. Soluble carbohydrate increased and free amino nitrogen decreased during the change from resistance to susceptibility. Proximal parts of leaves were usually infected first; they initially contained less amino nitrogen and soluble carbohydrate than distal parts. All parts of the leaf seemed to be equally susceptible later, when there were no differences in their amino nitrogen or soluble carbohydrate. Upper leaves of intact plants had more natural infection than those from corresponding leaves from topped plants. More conidia germinated on discs from them and produced longer hyphae. The discs from intact plants contained less free amino nitrogen and more soluble carbohydrate than those from topped plants. The accuracy of visual assessments of susceptibility was, generally, confirmed by measurements of percentage germination and length of hyphae from individual conidia on leaf discs. Regressions of hyphal length on leaf composition showed that susceptibility was apparently related inversely to free amino nitrogen and water content and directly to insoluble carbohydrate per unit dry matter.  相似文献

The aim of the present study was to test whether rapid accumulation of solutes in response to salinity in leaf tissues of barley (Hordeum vulgare L.) contributes to recovery and maintenance of residual elongation growth. Addition of 100 mM NaCl to the root medium caused an immediate reduction close to zero in elongation velocity of the growing leaf 3. After 20–30 min, elongation velocity recovered suddenly, to 40–50% of the pre-stress level. Bulk osmolality increased first, after 60 min, significantly in the proximal half of the elongation zone. Over the following 3 days, osmolality increases became significant in the distal half of the elongation zone, the adjacent, enclosed non-elongation zone and finally in the emerged portion of the blade. The developmental gradient and time course in osmolality increase along the growing leaf was reflected in the pattern of solute (Cl, Na and K) accumulation in bulk tissue and epidermal cells. The partitioning of newly accumulated solutes between epidermis and bulk tissue changed with time. Even though solute accumulation does not contribute to the sudden and partial growth recovery 20–30 min after exposure to salt, it does facilitate residual growth from 1 h onwards. The levels of adenosine, the adenine nucleotides AMP, ADP, ATP, as well as NAD, NADP, protein and chlorophyll were determined in young barley plants of which the primary leaves were infected by Erysiphe graminis DC. f. sp. hordei MARCHAL. The largest changes of these metabolite levels, compared to the non-infected control, occurred in the infected leaf and to a lesser degree also in the roots and in healthy younger leaves. The increase in the levels of most metabolites in the primary leaf revealed the sink property of this infected tissue and possible stress or defence reactions of the host, whereas the reductions in the roots showed the impaired supply of this natural sink organ due to the infection. Changes in the healthy leaves were most pronounced in the tertiary leaf and may reflect metabolic stimulation in that healthy organ. The changes of the adenosine pool, a precursor of the adenine nucleotides, were discussed in terms of translocation and its possible role as a precursor for fungal purine nucleotide synthesis.  相似文献

The structure of shoots, in particular of winter buds, ofHydrangea macrophylla was examined. The non-flower-bearing shoot is usually composed of a lower and an upper part, between which a boundary is discernible by means of a distinctly short internode. This internode is the lowermost of the upper part, and it is usually shorter than the internodes immediately above and below, although the internodes tend to shorten successively from the proximal to the distal part of the shoot. Variations exist in the following characters among the terminal bud, the axillary bud on the lower part of the shoot and the axillary bud on the upper part: (1) length of bud; (2) character of the outermost pair of leaf primordia; (3) degree of development of secondary buds in the winter bud; and (4) the number of leaf primordia. Usually, the terminal bud contains several pairs of foliage leaf primordia with a primordial inflorescence at the terminal of the bud, but the axiallary bud contains only the primordia of foliage leaves in addition to a pair of bud scales.  相似文献

Pathogens may alter their hosts, which consequently increases transmission efficiency by vectors. We examined the effects of Raspberry leaf mottle virus [RLMV; Closterovirus (Closteroviridae)] and Raspberry latent virus [RpLV; Reovirus (Reoviridae)], alone and in a co‐infection in raspberry, Rubus idaeus L. (Rosaceae) cv. Meeker, on the behavior and performance of its vector, Amphorophora agathonica Hottes (Hemiptera: Aphididae). Longevity was increased in aphids feeding on all infected‐plant treatments compared with healthy plants, but aphid fecundity only increased in the co‐infection treatment. In a two‐way choice study between infected and healthy plants, aphids showed no difference in preference between plants after 30 min of exposure. After 24 h, aphids significantly preferred to settle on plants infected with RLMV over healthy, but healthy plants over plants infected with RpLV. There were no differences in settling preferences between healthy and co‐infected plants. An electrical penetration graph study showed no differences in aphid feeding behavior on plants infected with RLMV and RLMV+RpLV when compared with healthy controls. Our results are consistent with past findings that infected plant's impact vector performance and behavior, but also highlight the need to further investigate greater virus diversity and effects of mixed infections.  相似文献

The talc-based formulation of two Pseudomonas fluorescens strains (Pf1 and VPT10) and its mixture (with and without chitin) were tested against tomato leaf curl virus in tomato under greenhouse and field conditions. The mean percentage of tomato leaf curl virus infected plants were significantly lower (25%) with less symptom severity and delayed symptom expression up to nine additional days in Pseudomonas with chitin (VPT10 + chitin) treated tomato plants compared to non-bacterised control plants upon challenge inoculation with tomato leaf curl virus. Tomato leaf curl virus was partially purified and antiserum was developed. Using the antiserum the tomato leaf curl virus was detected in symptomatic leaves and in whitefly vector through direct antigen coating enzyme linked immunosorbent assay which revealed the low virus titre in Pseudomonas treated plants (VPT10 + chitin) and insect vector compared to untreated tomato plants. The results indicate the potentiality of plant growth promoting rhizobacteria strains and talc-powder formulations in the effective management of this tomato leaf curl virus in tomato under field conditions.  相似文献

To investigate differences in leaf structure, chlorophyll and nutrients on terminal branches of the understory tree Asimina triloba, the first (proximal) and the last (distal) leaves to develop in the spring were compared. Proximal leaf expansion was completed before the overstory canopy was fully closed but distal leaf expansion occurred during and after the development of the overstory canopy. Fully expanded proximal leaves were 76% smaller in area, were 18% thicker and had 36% more stomates per m of leaf area when compared to distal leaves. In addition, maximum stomatal conductance to water vapor was greater (150 vs. 120 mmol m−-2s−-1) and the minimum PPFD required for maximum conductance was higher (200 vs. 150 μmol m−-2s−-1) for the proximal leaves. Chlorophyll content was also greater for proximal leaves, but nitrogen and phosphorus contents were lower throughout the entire summer. Seasonal measurements indicated an increase in chlorophyll a content and reductions in nitrogen content throughout the summer growth period for leaves from both positions. The results suggest that distal and proximal leaves differed physiologically and that the measured differences were related to the changing irradiance environment during leaf development. The time of leaf expansion, as indicated by leaf position on the branch, may be an important consideration when examining the water and photosynthetic relations of understory trees.  相似文献

The gradients in photosynthetic and carbohydrate metabolism which persist within the fully expanded second leaf of barley ( Hordeum vulgare ) were examined. Although all regions of the leaf blade were green and photosynthetically active, the basal 5 cm, representing approximately 20% of the leaf area, retained some characteristics of sink tissue. The leaf blade distal from the leaf sheath exhibited characteristics typical of source tissue; the activities of sucrolytic enzymes (invertase and sucrose synthase) were relatively low, whilst that of sucrose phosphate synthase was high. These regions of the leaf accumulated sucrose throughout the photoperiod and starch only in the second half of the photoperiod whilst hexose sugars remained low. By contrast the leaf blade proximal to the leaf sheath retained relatively high activities of sucrolytic enzymes (especially soluble, acid invertase) whilst sucrose phosphate synthase activity was low. Glucose, as well as sucrose, accumulated throughout the photoperiod. Although starch accumulated in the second half of the photoperiod, a basal level of starch was present throughout the photoperiod, by contrast with the rest of the leaf. The 14CO2 feeding experiments indicated that a constant amount of photosynthate was partitioned towards starch in this region of the leaf irrespective of irradiance. These findings are interpreted as the base of the leaf blade acting as a localized sink for carbohydrate as a result of sucrose hydrolysis by acid invertase.  相似文献

Populations of Arisaema triphyllum commonly harbor a systemic fungal pathogen, Uromyces ari-triphylli, which reduces leaf area and leaf longevity. Infected plants produced 42% fewer asexual progeny (cormlets). Furthermore, all asexual progeny from infected parents were themselves infected with Uromyces. Mean seed production of infected plants was only 21% of that among healthy plants. A hand pollination experiment demonstrated that 1) infected male plants can produce viable pollen, 2) the low seed production of naturally pollinated infected females was not due to inadequate pollination, and 3) seed production among healthy female plants was strongly pollinator limited. Seeds from infected and healthy parents had similar viability, and all seeds germinating from both types of parents were uninfected. The contrast in disease transmission to sexual vs. asexual progeny suggests that pathogen attack may be one selective factor favoring sexual reproduction in A. triphyllum populations.  相似文献

Abstract

Regeneration of new plants from the leaves of Atrichum undulatum will take place after 6 weeks if they are placed with the abaxial surface in contact with damp filter-paper in a saturated atmosphere.

The new plants arise from the surface cells of the midrib area of the leaf where there is a discontinuity of the photosynthetic filaments. This occurs mainly in the distal third of the leaf.

As the leaf ages it loses it capacity for regeneration. There are two regeneration processes, one which occurs when the leaf is separated from the stem and the only wound is at the point of attachment to the stem, and the second when other wounds are inflicted on the leaf. Regeneration due to wounding is greatest in the distal half of the leaf, mutilations near the base of the leaf having less effect on the percentage of regenerated plants.

There seems to be a potential for regeneration (both normal and as a result of wounding) which is greatest in the distal third of the leaf and decreases towards the base of the leaf of A. undulatum. This is compared with the leaf of Physcomitrium turbinatum, and the greater complexity of the process in the former is considered to be the result of greater differentiation of the leaf.  相似文献

The pattern of synthesis of rapidly-labelled RNA of hen sciatic nerve was studied during Wallerian degeneration. At 2,4,8, 16 and 30 days of degeneration the proximal and distal stumps of the severed nerve as well as the intact contralateral sciatic nerve (functional control) were excised and incubated with either [5-3H]uridine or [2-14C]uridine for 0.5 h. The electrophoretic pattern of RNA from the normal adult sciatic nerve showed that most of the radioactivity was incorporated into RNA species migrating between the 18 S and 4 S components of the bulk RNA. The synthesis of RNA was sensitive to actinomycin-D, an indication that it was directed by a DNA template. The electrophoretic patterns of the rapidly-labelled RNA in the proximal and distal nerve stumps demonstrated a change following nerve section. After 2–4 days of Wallerian degeneration the degenerating distal nerves incorporated more radioactivity in the 4 S region than the corresponding controls, but at 8 and 16-days after degeneration relatively more label appeared in higher molecular weight RNA species. In the intact sciatic nerve of the operated hens progressively more radioactivity was detected in the 4 S region with increasing time after the contralateral nerve section. At each stage of Wallerian degeneration the specific radioactivities of RNA in the control nerves from experimental hens were higher than those of the normal adult sciatic nerve. These results indicated a change of RNA metabolism in increased functional activity and during Wallerian degeneration.  相似文献

We transplanted imaginal disks of Drosophila melanogaster from larvae of the second half of the third larval instar into prepupae. Disks from the youngest donors differentiated bristles of only the distal segments of the leg. These disks also produced unusually large areas of cuticle that had no bristles. Disks from older donors differentiated bristles of more proximal segments and the area of cuticle with no bristles was reduced. To account for the regional variation in these results, there must be regional differences among the prospective leg cells at some time during the period from the second half of the third larval instar to the end of adult bristle differentiation. We asked whether prospective distal cells were more advanced than prospective proximal cells during bristle differentiation. We estimated when bristle precursor cells undergo their final cell divisions by heavily irradiating prepupae and pupae. We assumed that cells that were insensitive to the radiation had completed their cell divisions. The distal segments were the first to have insensitive bristles. Most leg bristles became insensitive between 12 and 18 hr after pupariation. The tarsus had a larger proportion of its bristles insensitive than the femur at 15 hr after pupariation. We also investigated when bristle-forming cells begin elongating their bristle shafts. We used the length of bristle rudiments as an indicator of when elongation is initiated. At 35 hr after pupariation, bristle rudiments of distal segments were two to three times longer than bristle rudiments of proximal segments. We discuss how these intersegmental differences observed during bristle differentiation can account for the regional variation in response of discs transplanted into older hosts. However, we do not exclude the possibility that regional differences among cells of the leg tissue exist at stages earlier than the time of bristle differentiation.  相似文献

The influence of infection with Septoria nodorum of leaves belowthe flag leaf on the translocation of 14C-labelled assimilatesin wheat was followed. In the vegetative phase export of assimilatesfrom a single infected leaf was reduced, but export from a healthyleaf on a heavily infected plant was increased. During the reproductivephase export from leaves was not affected by disease. Heavyleaf infection had little effect on the patterns of distributionof export especially during reproductive growth when only changesin the proportion of assimilates in leaf sheaths and tillerstumps were found. Distribution of export from a healthy flagleaf on an otherwise heavily infected plant was unaltered. Duringvegetative growth changes in the distribution of assimilateswere more marked, the greatest changes occurring when a singleinfected leaf on a healthy plant was exposed to 14CO2.  相似文献

Bioassays on ether-soluble acid extracts from healthy and Verticillium -infected tomato plants, showed the presence of substances inhibiting growth of wheat coleoptiles in both healthy and infected leaves and stems, but the amounts were greater in the infected.
Assays of infected stems and leaves showed increases in growth-promoting activity expressed as indole-3-acetic acid equivalents (IAAe), up to 200% of those for healthy controls.
Similar assays of cultures of V. albo-atrum showed growth-promoting activity. No acid substance capable of inhibiting the growth of wheat tissue was detected in the culture filtrate. IAA was identified by colour test with Ehrlich's reagent on chromatograms from extracts of both infected stems and fungal culture filtrates.
The vertical distribution of IAAe was determined in healthy and infected plants at the eight-leaf stage by assaying individual leaves and four stem segments separately. In healthy plants the IAAe content was greatest in the young leaves (6–8) but no gradient was observed as between leaves 1–5. In infected leaves increases over the controls were found in leaves, 1, 3 and 6 and a decrease in leaf 8.
In healthy stems IAAe was highest in the distal segment and infected stems showed higher values at all four levels, with the relative increase greatest in the distal region.
It is suggested that the major part of the Verticillium syndrome including petiolar epinasty, tylosis, pith hyperplasia and the formation of adventitious roots is the result of an accumulation of growth substances in infected tissue.  相似文献
