Summary The paper deals with the question, whether the Herring bodies and the granular structures formed by the so-called Gomori-substance in the neurohypophysis are artefacts or not. It is shown that the Gomori-substance and also its globular and granular structures are present in paraffin sections of the gland prepared according to the freezing-drying technique of Altmann-Gersh. All Gomori-substance is washed away, however, if the sections are treated with toluene and alcohol. If, on the contrary, the sections are treated with Bouin before the paraffin is removed, the Gomori-substance is precipitated and stands the treatment with alcohol. After staining with Gomori's haematoxylin and phloxine such preparations give the same picture of the Gomori-substance as common, wet-fixed Bouin preparations, provided the fixation has been carefully made. This strongly suggests that the classical opinion of the Gomori-substance as a granular matter which occasionally forms larger globulets (Herring bodies) truly reflects the conditions in the living tissue. Gersh and Tarr, who used the common technique when making their freezing-drying preparations, apparently lost the Gomori-substance when they treated the sections with alcohol. Their statement that the Herring bodies and other granular matter in the neurohypophyseal interstitia are artefacts is therefore to be abandoned.  相似文献

Summary Autoradiography of sections from neurohypophyses treated with tritiated clonidine has shown specific binding to 2-adrenoceptor sites in the neurohypophysis but not in the intermediate lobe. Other studies have shown that 2 agonists such as clonidine can cause a fall in circulating antidiuretic hormone; it is therefore possible to speculate that this action could be a direct one on the neurohypophysis since the appropriate binding sites have been shown to exist.  相似文献

AIM: To investigate the metabolic effects of recombinant human growth hormone (rhGH) in an Alstr?m syndrome patient with growth hormone deficiency. METHODS: A 15-year-old Alstr?m syndrome boy with growth hormone deficiency received rhGH therapy for 1 year. Biochemical parameters, including hepatic enzyme levels, lipid profiles, and insulin sensitivity, were measured. Body composition analysis and computed tomography scans of the liver were performed. RESULTS: After 1 year of rhGH treatment, body fat mass, fat infiltration in the liver, and serum lipid profiles had all decreased. Insulin sensitivity and acanthosis nigricans improved. CONCLUSION: rhGH therapy might have beneficial effects on body composition, liver fat content, lipid profiles, and insulin resistance in Alstr?m syndrome patients, with improvement of the glucose homeostasis.  相似文献

The activity of acid phosphatases was followed for one year in Lake Gårdsjön as well as in the inlet and the outlet of the lake. A budget of the phosphatases was calculated, including an estimation of the production of phosphatases. The phosphatase activity was also measured in two basins upstream of L. Gårdsjön: the north basin and the south basin of L. Stora Hästevatten.The acid phosphatase activity was very high compared with reported alkaline phosphatase activities in other lakes. About 95% of the phosphatases in L. Gårdsjön was produced in the lake, and the production was highest in early summer.Small Chrysophyceae (< 10 µm) probably produced the majority of the acid phosphatases in the investigated lakes, and accordingly could be favoured in environments with low phosphorus supply due to their ability to produce large amounts of phosphatases.  相似文献

Summary A strong positive immunoreaction with an -endorphin antiserum occurs in two distinct sites of the goldfish and carp neurohypophysis. Fluorescent nerve terminals are found in the laminar nerve processes located in the rostral pars distalis, but the immunocytological reaction is mainly localised on the nerve processes of the posterior neurohypophysis lying between the intermediate lobe cells. Almost all the digitations of the neurohypophysis are strongly fluorescent. The immunoreactive fibres probably originate from the hypothalamus, where perikarya displaying the same immunoreaction have been found in the pars lateralis of the nucleus lateralis tuberis and in some minor centres. The possibility that the immunoreactive substances revealed on the neurohypophyseal processes may originate in the intermediate lobe cells is also discussed. It has now to be established if this hypothalamo-hypophyseal system contains a substance with endorphic properties or only some immunologically related substance devoid of the corresponding physiological activities.  相似文献

Abstract

The sequence of the pig ob cDNA, which codes for the protein leptin, has been determined by screening a pig adipose cDNA library with an RT‐PCR amplified cDNA fragment of this gene. The 501 bp ob cDNA has 89% identity to the human ob cDNA, 92% identity to the bovine ob cDNA, 84% identity to the mouse ob cDNA and 84% identity to the rat ob cDNA. At the amino acid level, pig leptin which codes for a protein with a predicted molecular weight of 18,661‐dalton, has 86% identity to human leptin, 93% identity to bovine leptin, 84% identity to rat leptin and 84% identity to mouse leptin. RT‐PCR screening of RNA isolated from pig adipose, skeletal muscle, cardiac muscle, pancreas, stomach, kidney, spleen and jejunum detected ob mRNA only in adipose tissue; Northern blots with an ob cDNA probe identified a 4.0 kb species in adipose tissue. The conservation of sequence and expression pattern of leptin in the pig reported here indicates that as in other species, this protein likely plays an important role in controlling food intake and fat deposition in the pig.  相似文献

Colonies of Montastrea annularis from Carysfort Reef, Florida, that remained bleached seven months after the 1987 Caribbean bleaching event were studied to determine the long term effects of bleaching on coral physiology. Two types of bleached colonies were found: colonies with low numbers of zooxanthellae with normal pigment content, and a colony with high densities of lowpigment zooxanthellae. In both types, the zooxanthellae had an abnormal distribution within polyp tissues: highest densities were observed in basal endoderm and in mesenteries where zooxanthellae are not normally found. Bleached corals had 30% less tissue carbon and 44% less tissue nitrogen biomass per skeletal surface area, but the same tissue C:N ratio as other colonies that either did not bleach (normal) or that bleached and regained their zooxanthellae (recovered). Bleached corals were not able to complete gametogenesis during the reproductive season following the bleaching, while recovered corals were able to follow a normal gametogenic cycle. It appears that bleached corals were able to survive the prolonged period without nutritional contribution from their zooxanthellae by consuming their own structural materials for maintenance, but then, did not have the resources necessary for reproduction. The recovered corals, on the other hand, must have regained their zooxanthellae soon after the bleaching event since neither their tissue biomass nor their ability to reproduce were impaired.  相似文献

We investigated mechanisms whereby peroxisome proliferator-activated receptor γ (PPARγ) agonism redistributes lipid from visceral (VF) toward subcutaneous fat (SF) by studying the impact of PPARγ activation on VF and SF glucose uptake and metabolism, lipogenesis, and enzymes involved in triacylglycerol (TAG) synthesis. VF (retroperitoneal) and SF (inguinal) of rats treated or not for 7 days with rosiglitazone (15 mg/kg/day) were evaluated in vivo for glucose uptake and lipogenesis and in vitro for glucose metabolism, gene expression, and activities of glycerolphosphate acyltransferase (GPAT), phosphatidate phosphatase-1 (or lipin-1), and diacylglycerol acyltransferase. Rosiglitazone increased SF glucose uptake, GLUT4 mRNA, and insulin-stimulated glucose oxidation, conversion to lactate, glycogen, and the glycerol and fatty acid components of TAG. In VF, only glucose incorporation into TAG-glycerol was stimulated by rosiglitazone and less so than in SF (1.5- vs. 3-fold). mRNA levels of proteins involved in glycolysis, Krebs cycle, glycogen synthesis, and lipogenesis were markedly upregulated by rosiglitazone in SF and again less so in VF. Rosiglitazone activated TAG-glycerol synthesis in vivo (2.8- vs. 1.9-fold) and lipin activity (4.6- vs. 1.5-fold) more strongly in SF than VF, whereas GPAT activity was increased similarly in both depots. The preferential increase in glucose uptake and intracellular metabolism in SF contributes to the PPARγ-mediated redistribution of TAG from VF to SF, which in turn favors global insulin sensitization.  相似文献

The effect of temperature on the flow of diluted blood [Hct = 0.21], through 5 μm Nuclepore filters, is described by the Arrhenius equation with an energy of activation of 27.7 kJ/mol. Plasma, diluted with PBS, is almost three times less sensitive to temperature, with an energy of activation of 9.8 kJ/mol, while red cells are of intermediate sensitivity, with an energy of activation of 14.7 kJ/mol. The most sensitive elements to changes in temperature are leukocytes, with energies of activation of 31 kJ/mol and 35 kJ/mol for fast-flowing leukocytes (granulocytes and lymphocytes) and slow-flowing leukocytes (monocytes) respectively. Hence, the major determinants of the decline in filterability of blood through micropore filters are the leukocytes. This effect is compounded when blood is kept for 10 minutes or more at 10° C due to activation of granulocytes, which leads to permanent pore blocking when the affected blood is filtered at room temperature. The combination of increased passage time of leukocytes through peripheral areas at abnormally low temperatures and subsequent activation might influence the flow of blood in non-affected tissues. The effect of temperature on the filterability of red blood cells through 3 μm filters is not described by the Arrhenius equation and the deviations are seen as a gradual change of slope rather than a sharp break between two straight lines. The data are consistent with a gradual shift in rate limiting step away from the entry event into pores, which dominates at low temperature but becomes progressively less important at elevated temperatures. The changing parameter is probably the volume of the red cell, which is less important when flow is measured through 5 μm pores.  相似文献

The purpose of the study reported here was to determine conditions for electrocardiographic monitoring in the G?ttingen minipig in view of its use as a second non-rodent species in toxicology studies. Electrocardiograms were recorded from conscious minipigs (6/sex) maintained in a sling. The three standard bipolar limb leads (I, II, III), the three augmented unipolar limb leads (aVR, aVL, aVF), the triangular Nehb-Sp?ri leads (dorsal, axial, ventral) and their corresponding unipolar leads were recorded, and automated analysis of amplitudes and intervals was made. Major QRS patterns were not observed for any of the bipolar and unipolar leads. For triangular leads, the amplitude of waves was higher than that for limb leads, and the rS pattern dominated for dorsal, axial ventral and aV(F)-Ventral leads. The qR pattern dominated in the aV(R)-dorsal lead, whereas consistency and dominant patterns were not observed for the aV(L)-axial lead. For limb leads, the position of the electrode affected the ECG. Electrodes placed on the cubital and stifle joints were the preferred positions since the P- and R-waves were clearly identifiable with amplitudes > 0.2 mV. Also, the T-wave amplitude was (positive or negative) > 0.2 mV in at least two leads, making the determination of the QT-interval accurate. For the triangular leads, the position of the electrode had less influence on the amplitude of deflections. However, if the axial lead is to be used for calculation of intervals and amplitudes, the xyphoid process is the preferred position. In conclusion, the triangular lead system is recommended for recording ECGs in minipigs. Limb leads could be used in connection. The cubital and stifle joints for standard limb leads and the neck, sacrum, and xyphoid process for triangular leads are the preferred positions for electrodes.  相似文献

Until the 1940s research traditions were often imported from Germany to Sweden, and young scientists went to German universities to learn new techniques and get in touch with the latest ideas. In developmental biology, the comparative, phylogenetic embryology advocated most forcefully by Ernst Haeckel co-existed with the “Entwickelungsmechanik” tradition developed by Wilhelm His, Wilheln Roux and others partly as a reaction to Haeckel’s ideas. I use the zoology department at Uppsala University as a microcosmos to reflect the tensions between these traditions: Gösta Jägersten (1903–1993) and Sven Hörstadius (1898–1996) are used as exemples. Jägersten was a marine biologist who worked on the morphology and evolution of invertebrates, especiallly their larval forms. He developed a comprehensive theory describing the evolution of the life cycle in early metazoans. Recapitulation was an important ingredient, and Jägersten explicitly based his reasoning on Ernst Haeckel’s “biogenetic law”. Jägersten developed Haeckel’s “Gastraea” theory into another hypothetical animal—Bilaterogastraea—that came into being when the holopelagic Blastaea settled on the ground as an adult and kept a pelagic, planktonic larval form. This was the birth of the pelago-benthic life cycle, which plays such an important role in Jägersten’s speculations on the deep phylogeny of metazoans. Sven Hörstadius was a leading experimental embryologist in the mid-twentieth century. His most important work was on the determination and differentiation of the sea urchin embryo. Early work inspired by his teacher John Runnström’s double gradient theory showed that gradients of animalness (ectodermal determination) and vegetalness (endodermal determination) existed in the 16- and 32- cell embryos. Hörstadius became famous for his elegant extirpation and transplantation experiments using glass needles, and for his microsugical skills. He also made important contributions to the study of cranial neural crest development in the Mexican axolotl, in collaboration with his student Sven Sellman. Hörstadius was the great experimentalist, but did not develop speculative hypotheses the way Jägersten did. The very different styles of scientific research might have played a role also in the development of the personal difficulties that existed for a long time between the two professors.  相似文献

G?ttingen minipigs were trained on a set-shifting procedure involving discriminations, reversals, and extra-dimensional shifts. The discriminations used were black-white discriminations and right-left discriminations. The initial visual and spatial discrimination seemed equally difficult, and only for the visual modality was reversal found to be more difficult than the initial discrimination. Visual reversal was more difficult than spatial reversal, and a larger number of perseverative sessions were found for visual reversal compared to spatial reversal. The acquisition of the extra-dimensional shift from the visual to the spatial dimension was not inferior to the learning of spatial reversal. Neither was the acquisition of the extra-dimensional shift from the spatial to the visual dimension inferior to the learning of visual reversal. Thus, no evidence was found for attention to stimulus dimensions in discrimination learning of the pigs.  相似文献

Intracerebroventricular (icv) injection of purified recombinant human tumour necrosis factor (TNF , 4–8g) in conscious rats, produced increases in colonic temperature (1.0°C) and resting oxygen consumption (VO2, 14%) which were maximal after 80–90 minutes. Pretreatment with propranolol (10mg/kg s.c) significantly inhibited the rise in VO2, and prevented the increase in body temperature. Icv injection of an antagonist to corticotropin releasing factor (-helical CRF 9-41, 25 g), which prevents the pyrogenic and thermogenic actions of interleukin-1, did not influence the effects of TNF on temperature or VO2. Injection of a fragment of TNF (113–130 amino acid sequence) did not affect body temperature or VO2. TNF injection (icv) significantly increased brown adipose tissue (BAT)in vitro mitochondrial GDP binding, and this effect was slightly inhibited, but not prevented, by surgical denervation of the tissue, and was unaffected by pretreatment with -helical CRF 9-41. These data indicate that TNF can stimulate thermogenesis by a direct central action. The effects are largely, but not totally, dependent on the sympathetic nervous system but, unlike the thermogenic actions of interleukin they do not require release of CRF.  相似文献

There is much evidence that suggests that freshwater systems are more sensitive to introduced predators than are terrestrial or marine systems. We argue here that this dichotomy reflects widespread naiveté toward introduced predators among freshwater prey. Continental terrestrial animals are seldom naive toward novel predators owing to the homogenizing effects of historical biotic interchanges. Comparable biotic interchanges might have also precluded prey naiveté in most marine systems. By contrast, freshwater systems exhibit persistent large- and small-scale heterogeneity in predation regimes. This heterogeneity promotes naiveté at multiple spatial scales in freshwater prey, thereby producing a systemic vulnerability to introduced predators that is not seen in continental terrestrial or marine systems.  相似文献

The technique of horseradish peroxidase retrograde axonal transport and local electrical stimulation of the pre-Botzinger complex was used to study the connections between neurones of the bulbar respiratory nucleus and descending pathways from bulbar nuclei in the cat spinal cord. Twenty five years ago it was proposed that conserved components of constitutive heterochromatin assemble heterochromatin-like complexes in euchromatin and this could provide a general mechanism for regulating heritable (cell-to-cell) changes in gene expressibility. As a special case, differences in the assembly of heterochromatin-like complexes on homologous chromosomes might also regulate the parent-of-origin-dependent gene expression observed in placental mammals. Here, the progress made in the intervening period with emphasis on the role of heterochromatin and heterochromatin-like complexes in parent-of-origin effects in animals is reviewed.  相似文献

The life cycle of Empoasca vitis and its most important parasitoid Anagrus atomus in southern Switzerland can be divided into three phases. In phase I the adults of E. vitis move from overwintering sites, i.e. primarily conifers, over deciduous plants into the vineyards. The parasitoid A. atomus on the other hand, appears to overwinter in leafhopper eggs, mainly on roses and blackberries. It subsequently completes one generation in leafhopper eggs primarily on blackberries and hazelnuts, before it attacks E. vitis eggs in the vineyards. For phase II, a population model with time‐varying age structures and stochastic properties was constructed for E. vitis. For this purpose a time‐varying distributed delay model with attrition was constructed. Given the calibrated initial density of overwintering females, the model predicts an unacceptable number of E. vitis for the growing season. However, if egg parasitism of A. atomus and Stethynium triclavatum is introduced into the model as an external variable E. vitis densities are predicted which are economically irrelevant. Inphase III E. vitis adults leave the vineyards for the overwintering sites. The model shows the importance of the parasitoid A. atomus during phase II. A. atomus should be favoured by surrounding the vineyards with host plants carrying leafhoppers’ eggs. Consequently, habitat management measures for E. vitis control could be applied to the surroundings of the vineyards.  相似文献
