The objectives of this experiment were to compare estrous synchronization responses and AI pregnancy rates of beef heifers using protocols that included either CIDR or MGA as the progestin source. The hypotheses tested were that: (1) estrous synchronization responses after (a) progestin removal, and (b) PGF(2alpha); and, (2) AI pregnancy rates, do not differ between heifers synchronized with either progestin source. At the start of the experiment (Day 0) in both years, heifers were assigned randomly to receive, MGA supplement for 14 days (MGA-treated; n=79) or CIDR for 14 days (CIDR-treated; n=77). On Day 14 progestin was removed and heifers were observed for estrus up to and after PGF(2alpha) on Days 31 and 33 for CIDR-treated and MGA-treated heifers, respectively. Heifers that exhibited estrus within 60h after PGF(2alpha) were inseminated by AI 12h later; the remaining heifers were inseminated at 72h after PGF(2alpha) and given GnRH (100mug). More (P<0.05) CIDR-treated heifers exhibited estrus within 120h after progestin removal than MGA-treated heifers. Intervals to estrus after progestin removal were shorter (P<0.05) for CIDR-treated heifers than MGA-treated heifers. More (P<0.05) CIDR-treated heifers exhibited estrus and were inseminated within 60h after PGF(2alpha) than MGA-treated heifers. Pregnancy rates did not differ (P>0.10) between MGA-treated (66%) and CIDR-treated (62%) heifers. In conclusion, the use of CIDR as a progestin source in a 14-day progestin, PGF(2alpha), and timed AI and GnRH estrous synchronization protocol was as effective as the use of MGA to synchronize estrus and generate AI pregnancies in beef heifers.  相似文献

Two experiments were conducted to study effects of cloprostenol sodium (cloprostenol) and clenbuterol HCl (clenbuterol) during postpartum anestrus on subsequent reproductive performance in cows. In Experiment I, 96 cows received either 0.5 mg cloprostenol (PGF, n = 25), 364 mg clenbuterol (CLEN, n = 24), 0.5 mg cloprostenol and 364 mg clenbuterol (CLEN+PGF, n = 21) or no treatment (Control, n = 26) on Day 20 post partum. Treatments failed to influence postpartum interval, pregnancy rate or the incidence of short estrous cycles preceding the first normal estrous cycle. In Experiment II, anestrous cows were administered cloprostenol (0.5 mg) on either Day 20 (PGF20, n = 27) or Day 35 post partum (PGF35, n = 25), or served as untreated controls (Control, n = 26). Neither postpartum interval nor pregnancy rate were affected by cloprostenol treatment. In conclusion, treatment of postpartum cows with PGF did not alter the resumption of normal estrous cycles following parturition.  相似文献

Four experiments were conducted in postpartum beef cows to evaluate the influence of reducing the interval from GnRH to PGF(2alpha) from 7 to 5d in a Select-Synch + CIDR or CO-Synch + CIDR estrous synchronization program. In Expt 1, cows (n=156) were treated with either a 7 or 5d Select-Synch + CIDR program. A second PGF(2alpha) treatment was given to all cows in all experiments at 12h after the initial PGF(2alpha) (to ensure that luteolysis occurred with the 5d program). Estrous response, interval to estrus, conception rate, and first service AI pregnancy rates were similar between treatments. In Expt 2, cows (n=223) were treated with either a 7 or 5d CO-Synch + CIDR program, with timed-AI concomitant with GnRH at 60 h after PGF(2alpha). Timed-AI pregnancy rates were similar between treatments. In Expt 3 (n=223) and 4 (n=400) cows were treated with either a 7 or 5d CO-Synch + CIDR program with timed-AI concurrent with GnRH at either 60 h (7d) or 72 h (5d) after CIDR withdrawal. Timed-AI pregnancy rates were 13.3% (P<0.05; Expt 3) and 9.1% (P<0.05; Expt 4) greater for the 5 than 7d program. In conclusion, timed-AI pregnancy rates were improved with a 5d CO-Synch + CIDR program with timed-AI at 72 h after CIDR withdrawal, compared to a 7d CO-Synch + CIDR program with timed-AI at 60 h after CIDR withdrawal.  相似文献

This study evaluated the effect of exogenous PGF(2)alpha on circulating LH concentrations in postpartum multiparous (n = 32) and primiparous (n = 46) Brahman cows. The cows were randomly allotted within parity and calving date to receive 0, 1, 2 or 3 mg im PGF(2)alpha (alfaprostol)/100 kg body weight (BW), with or without GnRH on Day 30 after calving. Blood samples were collected at weekly intervals from calving through treatment. Serum progesterone concentrations were determined using RIA procedures to assure that only anestrous cows were treated. Sterile marker bulls were maintained with cows on Coastal bermudagrass pastures until the first estrus was detected. Multiparous cows had a shorter (P < 0.05) interval from calving to estrus than did primiparous cows. Serum LH was affected by time (P < 0.0001), PGF(2)alpha dose (P < 0.0002), GnRH (P < 0.0001), parity by PGF(2)alpha dose (P < 0.0003), PGF(2)alpha dose by GnRH (P < 0.0009), parity by GnRH (P < 0.0008), and by parity by PGF(2)alpha dose by GnRH (P < 0.0005). Multiparous cows not receiving GnRH had higher mean serum LH (P < 0.02), LH peak pulse height (P < 0.03), and area under the LH release curve (P < 0.03) compared with primiparous cows. The number of LH pulses/6 h was greater (P < 0.06) in multiparous than primiparous cows, and was greater (P < 0.02) in multiparous cows receiving 3 mg/100 kg BW than in cows receiving 2 mg/100 kg BW, but not in the controls or in cows receiving 1 mg/100 kg BW. Exogenous GnRH resulted in increased (P < 0.0001) serum LH concentrations in all cows, and LH was enhanced (P < 0.0009) by simultaneous treatment with PGF(2)alpha. Primiparous cows had a greater response (P < 0.0005) to PGF(2)alpha and GnRH compared with multiparous cows. Pituitary release of LH in response to GnRH was enhanced by simultaneous exposure to PGF(2)alpha in Day 30 postpartum cows.  相似文献

The objective of this study was to evaluate the effectiveness of superovulatory protocols by synchronizing the emergence of the follicular wave using estradiol benzoate (EB) or GnRH in CIDR-treated, Korean cows. Sixty-six cows were used in the study and these were divided into three groups. The standard group comprised cows that were between days 8 and 12 of their estrous cycle (n=22). The remaining 44 cows, at all other stages of the estrous cycle, received CIDR and were assigned to two treatment groups that received either 2mg EB (EB-CIDR group, n=22) or 100 microg GnRH (GnRH-CIDR group, n=22) 1 day after CIDR insertion. Gonadotropin treatment began between the 8th and 12th days of the estrous cycle in the standard group, 5 days after EB injection in the EB-CIDR group, and 3 days after GnRH injection in the GnRH-CIDR group. All cows were superovulated with porcine FSH (pFSH) twice daily, with the dose (total 28 mg) decreasing gradually over 4 days. On the 5th and 6th injections of pFSH, 25 and 15 mg doses of PGF(2alpha) were administered. CIDR was withdrawn at the 7th pFSH injection and the cows received 200 microg GnRH at 24h after CIDR withdrawal. Cows were artificially inseminated twice at 36 and 48 h post-CIDR withdrawal and embryos were recovered 7 days after the 1st insemination. The numbers of preovulatory follicles (22.9-28.2), ovulated preovulatory follicles (17.6-21.7) and CL (15.9-17.9) detected by ultrasonography did not differ among groups (P>0.05). Similarly, the numbers of total ova (6.7-10.0), transferable embryos (4.0-6.0), degenerate embryos (1.1-1.8) and unfertilized ova (1.3-4.3) did not differ among groups (P>0.05). Progesterone and estradiol concentrations during superovulation treatments and at embryo recovery were also the same in all groups (P>0.05). We conclude that in CIDR-treated Korean native cows, superovulatory treatments that follow administration of either EB or GnRH (at any stage of the estrous cycle) result in both a superovulatory response and embryo yield comparable to conventional superovulation protocols.  相似文献

Synthesis and secretion of prostaglandin F2alpha (PGF2alpha) is elevated following parturition and exerts divergent effects on the re-establishment of fertile estrous cycles in cows. The objective of these experiments was to determine if oil seed supplements differing in fatty acid composition differentially influence serum concentrations of the specific PGF2alpha metabolite, PGFM. Safflower seed supplements were formulated to provide 5% of dry-matter intake as fat. In Trial 1, 24 multiparous beef cows were individually fed control (beet pulp-soybean meal) or cracked high-linoleate safflower seed (78% 18:2n-6) supplements for 80 d postpartum. Linoleate supplemented cows had greater (P < 0.001) serum concentrations of PGFM than control cows. In Trial 2, primiparous beef cows (n = 36) were individually fed control (cracked corn-soybean meal), cracked high-linoleate (76% 18:2n-6) or -oleate (72% 18:1n-9) safflower seed supplements for 92 d postpartum. As in Trial 1, serum concentrations of PGFM were greater (P < or = 0.04) in linoleate than control or oleate supplemented cows. Serum concentrations of PGFM, however, did not differ (P = 0.40) among oleate and control supplemented cows. Although potential impacts on reproductive performance remain to be proven, dietary oil supplements high in linoleate, but not oleate, increased serum concentrations of PGFM compared to control supplements.  相似文献

Our objectives were to compare: (1) conception rates (in early postpartum Japanese Black beef cows) to timed-artificial insemination (timed-AI) among Ovsynch and Ovsynch plus CIDR protocols, and a protocol that used estradiol benzoate (EB) in lieu of the first GnRH of the Ovsynch plus CIDR; and (2) the effects of these protocols on blood concentrations of ovarian steroids. Cows in the control group (Ovsynch; n=35) underwent a standard Ovsynch protocol (GnRH analogue on Day 0, PGF(2 alpha) analogue on Day 7 and GnRH analogue on Day 9), with timed-AI on Day 10, approximately 20 h after the second GnRH treatment. Cows in the Ovsynch+CIDR group (n=31) received a standard Ovsynch protocol plus a CIDR for 7 days (starting on Day 0). Cows in the third treatment group (EB+CIDR+GnRH; n=41) received 2mg of EB on Day 0 in lieu of the first GnRH treatment, followed by the same treatment as in the Ovsynch+CIDR protocol. The conception rate tended to be greater in the Ovsynch+CIDR group (67.7%, P<0.15) and was greater in the EB+CIDR+GnRH (73.2%, P<0.05) and CIDR-combined (both CIDR-treated groups were combined) groups (70.8%, P<0.05) than in the Ovsynch group (48.6%). Plasma progesterone concentrations were higher on Day 7 (P<0.01) and lower on Days 14, 17 and 21 (P<0.001) in the CIDR-combined group than in the Ovsynch group. Plasma estradiol-17beta concentrations were higher on Day 7 in the Ovsynch group of non-pregnant cows than in the CIDR-combined group of non-pregnant cows and in an all-combined group (all treatment groups combined) of pregnant cows (P<0.01). Furthermore, estradiol-17beta concentrations were lower on Day 9 in the Ovsynch and CIDR-combined groups of non-pregnant cows than in the all-combined group of pregnant cows (P<0.05). In conclusion, both protocols using CIDR improved conception rates following timed-AI in early postpartum suckled Japanese Black beef cows relative to the Ovsynch protocol. Treatment with a CIDR may prevent early maturation of follicles observed in non-pregnant cows treated with the Ovsynch protocol, by maintaining elevated blood progesterone concentrations until PGF(2 alpha) treatment.  相似文献

This experiment was conducted to compare pregnancy rates in postpartum beef cows resulting from fixed-time AI (FTAI) after treatment with controlled internal drug release (CIDR)-based protocols to synchronize estrus. Cows assigned to the Show-Me-Synch (n=167) protocol received a CIDR from d 0 to 14, and prostaglandin F(2α) (PGF(2α)) on d 30. Cows assigned to 7-d CO-Synch+CIDR (n=177) received a CIDR and gonadotropin releasing hormone (GnRH) on d 23. On d 30, CIDRs were removed and PGF(2α) was administered. Blood sampling occurred on d -10 and 0 of treatment to determine estrous cyclicity status (progesterone ≥0.5 ng/mL estrous cycling). Treatments were balanced on age, DPP and BCS. Estrous detection was performed using HeatWatch from PGF(2α) to FTAI. Artificial insemination was performed at predetermined fixed times (72 h, Show-Me-Synch; 66h, 7-d CO-Synch+CIDR) and all cows were administered GnRH at FTAI. This experiment was conducted over a two year period; no differences were found between years so the data were pooled for further analysis. Pregnancy rate resulting from FTAI did not differ (P>0.10) between technicians or AI sires. Pregnancy rate resulting from FTAI was similar between treatments (P=0.20); however, cows that exhibited estrus prior to FTAI had a higher pregnancy rate (P<0.01) than for those that did not. Pregnancy rate at the end of the breeding period was similar between treatments (P=0.28). In summary, FTAI pregnancy rates were similar among postpartum beef cows following treatment with either a short- or long-term CIDR-based estrous synchronization protocol.  相似文献

Objectives were to evaluate progesterone concentrations after cows had initiated estrous cycles following calving and induction of estrous cycles in postpartum anovular high-producing Holstein dairy cows treated with controlled internal drug releasing (CIDR). In experiment 1 (EXP1), 62 cows that had initiated estrous cycles received a new CIDR (NCIDR) containing 1.38 g of progesterone or a 7-d used autoclaved CIDR (UCIDR) 48h after luteolysis for 7 d. Ovaries were examined by ultrasonography, and plasma analyzed for concentrations of progesterone. In experiment 2 (EXP2), 515 cows diagnosed as anestrus were randomly assigned to untreated control, NCIDR or UCIDR for 6d. Plasma was analyzed for concentration of progesterone 12 d after CIDR removal to determine ovulation. In EXP1, milk yield and body condition did not influence progesterone concentrations. Concentration of progesterone tended to increase faster (P=0.10) in cows receiving UCIDR than NCIDR, but both treatments reached a plateau at 90min. Cows receiving the NCIDR had greater (P=0.04) concentrations of progesterone during the 7-d treatment, but they were mostly subluteal (<1.0 ng/mL) after d 2. After removal, concentrations of progesterone were greater for NCIDR than UCIDR for the first 45 min, and were similar thereafter. Multiparous cows had lesser (P=0.004) concentrations than primiparous cows throughout the study. The pattern of ovarian follicular development was not affected by treatment. In EXP2, induction of onset of estrous cycles increased (P<0.01) with progesterone treatments, but was similar between NCIDR and UCIDR. Proportion of cows experiencing shorter than typical length estrous cycles after first AI tended to be greater (P=0.09) for control cows than those receiving the CIDR, and for cows remaining anestrous than those in which onset of estrous cycles was induced. Pregnancy per AI and pregnancy loss were similar among treatments. Cows that resumed estrous cyclicity prior to first AI had greater (P=0.01) pregnancy per AI. Treatment of high-producing Holstein cows that had previously initiated onset of estrous cycles with CIDR resulted in subluteal concentrations of progesterone, but in anestrous high-producing cows increased induction of estrous cycles with no effect on fertility at first insemination.  相似文献

The reproductive efficiency of Friesian dairy cows was investigated in a three (oestrous synchronisation technique) x two (seasons of the year) factorial design. The 90 primiparous and multiparous cows (winter, n=42; summer, n=48) were allocated at random to three synchronisation treatments (n=30 cows per treatment). In treatment 1 (GPG), the cows were administered 15 mg PGF(2alpha) i.m. at 30 +/- 3 days postpartum, 100 microg GnRH i.m. at 51 +/- 3 days and 15 mg PGF(2alpha) 7 days later. A second 100 microg dose of GnRH was given after, further 2 days and fixed time AI occurred 16-20 h later. In treatment 2 (PG-PG), 15 mg PGF(2alpha) was administered i.m. to each cow on three occasions at successive 14 days interval starting at 30 +/- 3 days postpartum and the cows were inseminated at observed oestrus following the third dose of PGF(2alpha). Cows in treatment 3 (PG) had a single administration of 15 mg PGF(2alpha) i.m. at 57+/-3days postpartum and were inseminated as in treatment 2. Mean daily ambient temperature was 10.9 degrees C in winter (November-March) and 20.2 degrees C in summer (June-October). The cows were confined in an open-fronted shed and had ad libitum access to a complete diet with a 37:63 forage to concentrate ratio. Body condition score was assessed at 57 +/- 3 days postpartum. Cow rectal temperature at insemination, milk yield, reproductive data and climatic variables were recorded. Blood samples were collected for progesterone assay on days 4, 11, 18, 25, 32, 39 and 46 post-AI from 54 of the cows (19 GPG; 17 PG-PG; 18 PG). Pregnancy rate to first AI was 36.7% (11/30) for GPG and 16.7% (5/30) for both PG-PG and PG treatments. The difference was not significant. The cumulative pregnancy rate after third AI were GPG 83.3% (25/30), PG-PG 60.0% (18/30) and PG 60.0% (18/30; P<0.057). The cumulative pregnancy rate for cows inseminated in the winter (81.0%; 34/42) was higher (P<0.01) than for those inseminated in the summer (56.3%; 27/48). The interval from calving to first service was shorter (P<0.05) in treatment PG-PG (65.4+/-1.3 days) than in PG (69.2+/-1.3 days). Mean plasma progesterone concentrations post-AI of pregnant cows were higher (P<0.001) for GPG cows than those for PG-PG and PG cows. Plasma progesterone levels of pregnant cows tended to be higher (P=0.087) in winter than in summer. In conclusion, although the cumulative pregnancy rate was higher for GPG cows, it may be appropriate to correct the nutrition and management of the herd before resorting to synchronisation techniques to improve animal reproductive performances.  相似文献

The objective of this study was to characterize endometrial secretion (in vitro) of prostaglandin F (PGF), 15-keto-13,14-dihydro-prostaglandin F2 alpha (PGFM), prostaglandin E2 (PGE2), and 6-keto-prostaglandin F1 alpha (6-keto-PGF1 alpha) on Day 5 following the first postpartum estrus of cows anticipated to have a short compared to a normal estrous cycle. Twenty-seven beef cows were randomly assigned into four groups. The Short Cycle (n = 6; control) and Short Cycle/Explant (n = 8; endometrial explants) groups had their calves weaned at 30-32 days postpartum. The Normal Cycle (n = 5, control) and Normal Cycle/Explant (n = 8; endometrial explants) groups received norgestomet (progestin) implants for 9 days beginning 21-23 days postpartum, and calves were weaned at implant insertion. Estrous cycle length (mean +/- SE; p less than 0.01) for the Short Cycle group was 11.5 +/- 1.9 days compared to 18.8 +/- 0.6 days for the Normal Cycle group. On Day 5 following the first postpartum estrus, cows in the Short Cycle/Explant and Normal Cycle/Explant groups were hysterectomized, and endometrial explants were incubated in Earle's Balanced Salt solution/Medium 199 for 90 min with or without arachidonic acid (AA) in the presence of three levels of oxytocin. Mean concentrations of PGF and PGFM were combined to obtain a value for total PGF. Luteolysis in the cow depends upon an interaction between prostaglandin F(2alpha) (PGF(2alpha)) and oxytocin. The objectives of our study were 1) to determine oxytocin concentrations in postpartum dairy cows and 2) to identify the temporal relationship between oxytocin and PGF(2alpha) release patterns during luteolysis in normal and abbreviated estrous cycles in the postpartum period. Serum oxytocin and PGF(2alpha) metabolite (PGFM) concentrations from nine cows which had short estrous cycles (< 17 d) were compared with those of six cows which had normal estrous cycles. Serum basal oxytocin concentrations in short estrous cycle cows (23.7 to 31.1 pg/ml) were higher (P<0.05) than those of normal estrous cycle cows (14.6 to 19.8 pg/ml). Oxytocin concentrations increased to peak values in both short and normal cycle cows, during luteolysis. Basal PGFM concentrations (112.2 to 137.4 pg/ml) were higher in cows with short cycle (P<0.05) than in cows with normal cycles (62.9 to 87.5 pg/ml). The increase in PGFM concentrations during luteolysis was significant in both normal cycle and short cycle cows (P<0.05). Increases in serum PGFM concentrations were always associated with increases in serum oxytocin concentrations in normal cycle and short cycle cows and the levels decreased simultaneously before the subsequent estrus. Results support the idea of a positive relationship between PGF(2alpha) and oxytocin concentration during the estrous cycle as well as a possible synergistic action of these hormones in the induction of luteolysis in dairy cattle.  相似文献

In a previous study we showed that estrus synchronization with 2 treatments of PGF2 alpha 13 d apart reduced conception rate at the synchronized estrus and that this reduction occurred mainly in cows in the early luteal phase at the second PGF2 alpha treatment. The objective of the present study was to determine the efficacy of a synchronization regimen in which PGF2 alpha was administered during the mid- to late-luteal phase to cows that had previously been synchronized with progesterone. Spring-calving cows from 6 dairy herds were used in this study. On Day -32 (Day 1 = the start of the breeding season), cows that had calved 2 or more weeks ago were randomly assigned to a synchronization (S, n = 732) or control (C, n = 731) group. Cows in Group S were treated with an intravaginal progesterone device (CIDR) for 12 d from Day -32 to Day -20, while those in Group C were left untreated. Similar percentages of cows in Group S (80.6%) and C (82.9%) had cycled by Day -7. The CIDR treatment synchronized the onset of estrus, resulting in 92.9% of cows in estrus being detected within 7 d after CIDR removal. Cows in Group S that had cycled by Day -7 were treated with PGF2 alpha (25 mg, i.m., Lutalyse) on Day -2. Cows in both groups that were anestrous on Day -7 were treated with a combination of progesterone and estradiol benzoate (EB) to induce estrus and ovulation (CIDR and a 10 mg EB capsule on Day -7, CIDR removal on Day -2, and injection of 1 mg EB 48 h after CIDR removal). The PGF2 alpha treatment synchronized the onset of estrus in 87.5% of the cows. Group S and C cows had similar conception rates to first (61.0 vs 58.3%) and second (58.4 vs 60.9%) AI; similar pregnancy rates over the AI period (82.8 vs 79.2%) and over the whole breeding season (91.9 vs 90.6%); and required a similar number of services per pregnancy to AI (1.7 vs 1.8). The interval from the start of the breeding season to conception for cows conceiving to AI or to combined AI and natural mating was shorter (P < 0.001) by 5.7 and 6.2 d, respectively, for the Group S cows. It is concluded that the treatment regimen tested in the present study achieved satisfactory estrus synchronization, had no detrimental effect on fertility at the synchronized estrus, and shortened the interval from start of the breeding season to conception.  相似文献

Three experiments were conducted, using multiparous crossbred beef cows, to test the ability of exogenous prostaglandin F(2alpha) (PGF) and/or naloxone to reduce the duration of the postpartum interval to estrus and to improve subsequent reproductive performance. In each experiment, postpartum cows were assigned to treatments by calving date. In Experiment 1, cows (n=44) were assigned to 1 of 4 treatment groups: 1) control, 2) PGF on Day 25 post partum, 3) 400 mg naloxone (3 doses) at 12-h intervals on Day 30 post partum, and 4) PGF on Day 25 followed by 3 400-mg doses naloxone at 12-h intervals on Day 30 post partum. In Experiment 2, cows (n=126) were assigned either to 1) control or 2) PGF on Day 30 post partum In Experiment 3, cows (n=67) were again assigned to 1 of 4 treatments 1) control, 2) PGF on Day 30 post partum, 3) PGF on Day 40 post partum, and 4) PGF on Day 30 and 40 post partum. Serum progesterone was used to determine the postpartum interval to estrus in Experiments 1 and 3. In all 3 experiments, serum progesterone was used to determine the proportion of cows that had reestablished estrous cycles at the start of breeding. Pregnancy rate and calving interval were analyzed for all trials. Naloxone had no effect (P > 0.20) on any reproductive variable measured. The postpartum interval to estrus was similar (P > 0.30) for PGF-treated and control cows in Experiments 1 and 3. The proportion of cows cycling at the start of breeding and the calving interval were not affected (P > 0.20) by PGF treatment in any of the experiments. Only the administration of PGF on Day 40 post partum in Experiment 3 improved (P=0.04) the subsequent pregnancy rate. Analysis of data pooled across experiments showed that the pregnancy rate was higher (P=0.03) for cows treated with PGF than for control cows (91.4 and 72.9%, respectively). A total of 226 out of 245 postpartum lactating dairy cows in a commercial dairy farm were allocated to two groups of oestrous synchronisation protocols in order to evaluate reproductive performance. One group was treated with oestradiol benzoate (ODB) and PGF2alpha on day 10 of the oestrous cycle with insemination at the detected oestrus, the second group underwent the Ovsynch (OVS) protocol (GnRH + PGF2alpha + GnRH) with timed AI. Pregnancy was diagnosed by ultrasonography on day 28 after AI and confirmed by rectal palpation on day 45. A higher (P < 0.001) proportion of cows in OVS (100%) were inseminated within (19.2 +/- 3.8 h) following the second GnRH injection than those of cows in EPE (ODB + PGF2alpha + ODB) (70.6%) inseminated at the detected oestrus within (35.6 +/- 5.2 h) following the second ODB injection. Pregnancy rates for the first AI at day 28 (64.0 +/- 4.6, 62.4 +/- 5.5%) and at day 45 post-insemination (40.4 +/- 4.7, 40.0 +/- 5.6%) for OVS and EPE cows respectively, did not differ between the two treatments, whereas, the overall pregnancy rates tended to be higher (P < 0.08) for the OVS (85.1 +/- 3.8%) cows than the EPE cows (74.1 +/- 4.5%). No differences were observed in pregnancy rates for first AI and overall up to fourth AI between primiparous (34.7 +/- 5.8 and 85.3 +/- 4.7%) and multiparous cows (43.5 +/- 4.5 and 77.4 +/- 3.6%). Days open for pregnant cows tended to be lower (P < 0.08) for OVS (76.2 +/- 3) than for EPE cows (84.7 +/- 4), while days open were higher (P < 0.05) in primiparous cows (85.3 +/- 4) than in multiparous cows (75.6 +/- 3). The results indicate that pregnancy rates for first AI were similar, but overall pregnancy rates up to the fourth AI tended to be higher for OVS than EPE cows, while days open was tended to be lower for OVS than EPE cows.  相似文献

The objectives of the present study were to evaluate the induction of estrus and fertility in yak cows treated with Co-Synch regimens or progesterone (P(4)). In Experiment 1, postpartum suckled yaks were assigned to three treatments: (1) A (n=28), insertion of an intravaginal device containing P(4) (CIDR) on Day 0, PGF(2alpha) (i.m.) on Day 6 and PMSG (i.m.) at the time of CIDR removal on Day 7 (P(4)-PGF(2alpha)-PMSG); (2) B (n=21), PGF(2alpha) (i.m.) on Day 6 and PMSG on Day 7; (3) C (n=26), control group. Seven yak bulls were grazed with the cows for natural breeding. Rate of estrus within 96h of the end of treatment was greater (P<0.05) in A (100.0%) than in B (28.6%) or C (0.0%). First service conception rate (CR) determined by serum P(4) on Day 21 after breeding was greater (P<0.05) in A (78.6%) than in B (22.2%). Also, pregnancy rate (PR) during the breeding season was greater (P<0.05) in A (82.1%) than in B (19.0%) and C (7.7%). In Experiment 2, non-suckled yaks that calved in previous years but not in the current year were assigned to three treatments: (1) A (n=31), GnRH (i.m.) on Day 0, followed by PGF(2alpha) on Day 7 and timed artificial insemination (TAI) concurrently with GnRH treatment on Day 9 (Co-Synch regimen); (2) B (n=50), a CIDR device for 7 days plus PGF(2alpha) and PMSG at the time of CIDR withdrawal on Day 7 and TAI on Day 9 (P(4)-PGF(2alpha)-PMSG); (3) C (n=50), yak cows were artificially inseminated at spontaneous estrus. Frozen semen of Holstein and Jersey were used for insemination in Experiment 2. The CR assessed by rectal palpation 35 days after TAI was not different in A (22.6%), B (30.0%) and C (33.3%), but PR was greater in A and B than in C, when based on those cows presented for estrous synchronization programs. It is concluded that P(4)-PGF(2alpha)-PMSG protocol could efficiently induce estrus and result in an acceptable pregnancy rate in postpartum suckled yak cows. This technique and Co-Synch regimen can be applied successfully for TAI of non-suckled yak cows.  相似文献

One hundred and sixty-five suckled postpartum beef cows were utilized to evaluate the effectiveness of 2 estrus synchronization systems for the initiation and synchronization of estrus. The treatment groups consisted of 1) melengestrol acetate (MGA)-PGF2alpha (cows were given 0.5 mg MGA/head/day for 14 d with 25 mg PGF2alpha injected 17 d after the last day of MGA administration); 2) MGA-48-h calf removal (CR)-PGF2alpha (cows were given 0.5 mg MGA/head/day for 14 d with 48-h calf removal starting on the second day after completion of the MGA regimen plus 25 mg PGF2alpha administered 17 d after the last day of MGA); and 3) unsynchronized controls. Cows were assigned to treatments by the numbers of days post partum, body condition, age, and breed of sire. The cows were observed for estrus at 12-h intervals for 5 d after PGF2alpha administration and were artificially inseminated 12 to 18 h after the observed estrus. Both the MGA-PGF2alpha and MGA-CR-PGF2alpha treatments (64.8 and 61.8%) had greater (P < 0.05) 5-d estrus rates than the control treatments (34.5%). The synchronized pregnancy rate was greater (P < 0.05) for the MGA-CR-PGF2alpha than the control treatment.(52.7 vs 30.9%, respectively). The MGA-CR-PGF2alpha cows had a higher 25-d pregnancy rate than either the MGA-PGF2alpha (P < 0.05) or control cows (P < 0.08). Of the anestrous cows at the beginning of treatment, more MGA-CR-PGF2alpha (P = 0.1) and MGA-PGF2alpha cows were cyclic posttreatment than control cows (58.7 and 55.1 vs 44.7%, respectively), suggesting that treatment initiated estrous cycles in only a small number of the anestrous cows. Both MGA-PGF2alpha and MGA-CR-PGF2alpha treatments appear to be effective methods of synchronizing estrus in suckled postpartum beef cows. However, MGA-CR-PGF2alpha was more effective in establishing pregnancy earlier in the breeding season than MGA-PGF2alpha.  相似文献

The objective of this study was to compare the effectiveness of the Ovsynch and controlled internal drug releasing (CIDR) protocols under commercial conditions for the treatment of cystic ovarian disease in dairy cattle. A total of 401 lactating dairy cows with ovarian cysts were alternatively allocated to two treatment groups on the day of diagnosis. Cows in the Ovsynch group were treated with GnRH on Day 0, PGF2alpha on Day 7, GnRH on Day 9, with timed insemination 16-20 h later. Cows in the CIDR group were treated with a CIDR insert on Day 0 for 7 days; on Day 7, the CIDR was removed, and cows were treated with PGF2alpha. All cows in the CIDR group were observed for estrus and cows exhibiting estrus within 7 days following removal of the CIDR and PGF2alpha administration were inseminated. The outcomes of interest for this experiment were the likelihood to be inseminated, return to cyclicity (determined by a CL on Day 21), conception and pregnancy rates. Data for these variables were analyzed using logistic regression. The percentage of cows inseminated in the Ovsynch and CIDR groups were 82 and 44%, respectively. Cows in the Ovsynch group were 5.8 times more likely to be inseminated than cows in the CIDR group. Cows with a low BCS were 0.48 times less likely to be inseminated than cows with a high BCS. The percentage of cows with a CL on Day 21 for the Ovsynch and CIDR groups was 83 and 79%, respectively (P > 0.05). Cows with a low BCS were 0.49 times less likely to have CL on Day 21 than cows with a high BCS. Conception and pregnancy rates for cows in the Ovsynch group were 18.3 and 14.4%, respectively. Conception and pregnancy rates for cows in the CIDR group were 23.1 and 9.5%, respectively. There was no significant differences between conception or pregnancy rates in cows in both groups. Primiparous cows were 2.6 times more likely to conceive than multiparous cows. In conclusion, the results of this study suggested that fertility was not different between cows with ovarian cysts treated with either the Ovsynch or the CIDR protocols in this dairy herd. In addition, primiparous cows had an increased likelihood for conception compared to multiparous cows, and cows with a low BCS were less likely to be inseminated or have a CL on Day 21, regardless of treatment.  相似文献

Estrous cycles of 10 postpartum cyclic Holstein cows were synchronized using prostaglandin f(2alpha) (PGF(2alpha)) given twice 12 d apart to study the relationship of the onset of estrus, body temperature, milk yield, luteinizing hormone (LH) and progesterone concentration to ovulation. Blood samples and body temperatures (vaginal and rectal) were taken every 4 h until ovulation, starting 4 h prior to the second PGF(2alpha) treatment. All cows were observed for estrus following the second administration of PGF(2alpha). Ultrasound scanning of the ovaries commenced at standing estrus and thereafter every 2 h until the disappearance of the fluid filled preovulatory follicle (ovulation). Two cows failed to ovulate and became cystic following the second PGF(2alpha) treatment. The remaining eight cows exhibited a decline in progesterone to <1.0 ng/ml within 28 h, standing estrus and a measurable rise (> 1.0 degrees C) in vaginal but not rectal temperature, and ovulated 90 +/- 10 h after the second PGF(2alpha) treatment. Onset of standing estrus, LH peak and vaginal temperature were highly correlated (P<0.05) with time of ovulation (0.82, 0.81 and 0.74, respectively). Intervals to ovulation tended to depend upon parity. Pluriparous (n = 4) and biparous (n = 4) cows ovulated within 24 and 30 +/- 3 h from the onset of standing estrus; 22 and 31 +/- 2 h from the LH peak; and 22 and 27 +/- 3 h from peak vaginal temperature (mean +/- standard error of the mean), respectively. The results indicated that the onset of standing estrus and rise in vaginal temperature are good practical parameters for predicting ovulation time in dairy cattle.  相似文献

Two experiments were conducted to investigate the effects of timing of prostaglandin F2(alpha) (PGF2(alpha)) administration, controlled internal drug release device (CIDR) removal and second gonodotropin releasing hormone (GnRH) administration on the pregnancy outcome in CIDR-based synchronization protocols. In Experiment 1, suckled Angus crossbred beef cows (n = 580) were given 100 microg of GnRH+a CIDR on Day 0. Cows in Group 1 (modified Ovsynch-P) received 25 mg of dinoprost (PGF2(alpha)) and CIDR device removal on Day 8 (AM), 100 microg of GnRH 36 h later on Day 9 (p.m.), and fixed-time AI (FTAI) 16 h later on Day 10 (47.5+/-1.1 h after PGF2(alpha)). Cows in Group 2 (Ovsynch-P) received 25mg of PGF2(alpha) and CIDR device removal on Day 7 (p.m.), 100 microg of GnRH 48 h later on Day 9 and FTAI 16 h later on Day 10 (66.6+/-1.2 h after PGF2(alpha)). Pregnancy rates were 56.5% (170/301) for Group 1 and 55.6% (155/279) for Group 2, respectively (P = 0.47). In Experiment 2, beef cows (n=734) were synchronized with 100 microg of GnRH+CIDR on Day 0, 25 mg of PGF2(alpha) and CIDR device removal on Day 7 and either 100 microg of GnRH 48 h later on Day 9 (Ovsynch-P) and FTAI 16 h later on Day 10 (64.9+/-3.3 h from PGF2(alpha)) or 100 microg of GnRH on Day 10 (CO-Synch-P) at the time of AI (63.2+/-4.2 h from PGF2(alpha)). Pregnancy rates were 48.8% (180/369) for Ovsynch-P and 44.7% (163/365) for CO-synch-P groups, respectively (P = 0.11). In both experiments, there was a locationxtreatment interaction (P<0.05); pregnancy rates between locations were different (P < 0.05) in the Ovsynch-P group. In conclusion, in a CIDR-based Ovsynch synchronization protocol, delaying administration of prostaglandin and CIDR removal by 12 h, or timing of the second GnRH by 16 h, did not affect pregnancy rates to FTAI. Therefore, there may be an opportunity to make changes in synchronization protocols with out adversely affecting FTAI pregnancy rates.  相似文献
