The life cycle of Gnathostoma nipponicum was examined by field survey and by experimental infection of animals with the larvae. Naturally infected larval G. nipponicum were found in loaches, catfish, and snakes. Experimentally, loaches, killifishes, frogs, salamanders, mice, and rats were successfully infected with the early third-stage larvae of G. nipponicum obtained from copepods (the first intermediate host), whereas snakes, quails, and weasels were not. Frogs, snakes, quails, and rats were experimentally infected with the advanced third-stage larvae (AdL3) from loaches. These results reveal that some species of fishes, amphibians and mammals can act as the second intermediate host and that some species of reptiles, birds and mammals can act as a paratenic host. The life cycle was completed in weasels, the definitive host, which were infected with AdL3 from loaches and started to evacuate eggs of G. nipponicun in faeces on days 65-90 postinfection.  相似文献

Five confirmed human cases of gnathostomiasis nipponica exhibiting creeping eruption and itching were found sporadically from the autumn of 1991 to the winter of 1992 in the northern region of the mainland of Japan. In all cases, a causative gnathostome with 3 transverse rows of hooklets on the head bulb was detected in biopsied skin. The morphological characteristics agreed with the advanced third-stage larvae of Gnathostoma nipponicum. Within a few weeks before development of symptoms, all patients had histories of eating raw freshwater fishes, kokanee (Salmo nerka nerka), carp (Cyprinus carpio), crucian carp (Carassius gibelio langsdorfi), or common ice-fish (Salangichthys microdon). However, they had never eaten raw loach, which is known as a source of human infections with G. nipponicum.  相似文献

The goal of our study was to investigate the molecular characteristics of mitochondrial DNA (mtDNA) and phylogenetic construction of the weather loach (Misgurnus anguillicaudatus) in Poyang Lake. The complete mitochondrial genome was 16,634?bp, and the gene order was identical to that of teleost fishes. Compared with the previous reported weather loach in China, there were numerous nucleotide substitutions and length polymorphisms on the structural genes of mitochondrial DNA in the loach from the Poyang Lake. The Phylogenetic tree indicated that the loach had its own molecular characteristics and was somewhat different from those in other regions of China. Fourteen unique haplotypes of the cytochrome b (cyt b) gene were obtained from 300 weather loaches. The Phylogenetic tree based on the cyt b gene showed that the loaches were substructured into two different populations in The Poyang Lake. Results indicated that the loaches in Poyang Lake not only showed the same phylogeny as the loaches in other areas of China, but also generated its own unique phylogenetic relationships.  相似文献

Gnathostomiasis by Gnathostoma hispidum has occurred mainly in southern and western areas of Japan since 1980. In all cases, patients had eaten live loaches, freshwater fish imported from southeast Asia. Chief symptoms were creeping linear skin eruptions and eosinophilia, both of which disappeared in most patients within three months after the onset, and never recurred thereafter. In order to study how gnathostome larvae may survive and behave in humans, rats were infected with gnathostome larvae. Loaches were purchased in city markets in Fukuoka. Gnathostome larvae (G. hispidum) were recovered from the fish and fed to rats. They were killed at intervals of one and six months and one, two and three years postinoculation. Encysted larvae were found in the muscles of all rats and recovered larvae were all viable. A migrating larva was observed in muscle tissue from one rat that was killed three years postinoculation. In histological sections, walls around encysted larva were much thinner in a rat killed three years postinoculation than in a rat killed one month postinoculation.  相似文献

The surface ultrastructure of advanced third-stage larvae (AL3) of Gnathostoma nipponicum was studied using scanning electron microscopy. The larvae were recovered from the grass snake Rhabdophis tigrina in the Republic of Korea. Parasites had a globular head bulb with a pair of lips at the anterior end and 2 labial papillae and an amphid on each lip. The head bulb was characteristically armed with 3 transverse rows of hooklets, averaging 36, 38, and 43 in number, increasing posteriorly. A total of 213-232 minute unidentate cuticular spines were present along the entire length of the larvae, forming the transverse striations. Two pairs of cervical papillae were located between the 8th and 12th transverse striations, and a pair of body papillae was seen laterally on the posterior third of the body. A pair of caudal phasmids was recognized near the posterior extremity. The surface ultrastructure of AL3 of G. nipponicum is unique compared with that of other species.  相似文献

Monoclonal antibodies (mAbs) were produced against the proteins of advanced third-stage larvae (AdL3) of Gnathostoma nipponicum. Six mAbs (Gn2C3, Gn2H3, Gn4C3, Gn4E9, GnSH1, and Gn10B7) were obtained as determined by enzyme-linked immunosorbent assay (ELISA). Gn4E9 and GnSH1 seemed to be genus-specific, as they did not cross-react with Anisakis sp., Dirofilaria immitis, Gongylonema pulchrum, Toxocara canis, Trichinella sp., Trichuris vulpis, Metagonimus sp., or Spirometra erinaceieuropaei by ELISA. Immunohistochemistry showed that Gn2C3, Gn4E9, and Gn5H1 reacted strongly with the central esophagus; Gn2H3 reacted with cuticle,muscle, intestine, and the cervical sac; and Gn4C3 and Gn10B7 reacted with cuticle, muscle, esophagus, intestine, and the cervical sac of AdL3. In Western blotting analysis, Gn2C3, Gn4E9, and Gn5H1 reacted to 60-, 53-, 46-, and 41-kDa proteins; Gn4C3 reacted to the AdL3 protein of G. nipponicum (>42 kDa). Moreover, proteins purified using a mAb Gn4E9 immunoprecipitation method (sizes 60-, 53-, 46-, and 41-kDa) were used as antigens in ELISAs. A significant difference (P < 0.01) was shown between mouse sera infected with G. nipponicum and sera infected with Trichnella sp. or not infected. These results provide a rationale for evaluating esophageal proteins for the development of diagnostic methods for detecting G. nipponicum or Gnathostoma sp. infections.  相似文献

In Memanbetsu town, Hokkaido island, Japan, a high frequency of natural triploid loaches Misgurnus anguillicaudatus (7.4% on average) was detected by flow cytometry for relative DNA content. Among sympatric diploid females (n=6) from a single population, we found two unique females that laid unreduced diploid eggs. They gave normal diploid progeny even after induction of gynogenesis with genetically inert UV-irradiated sperm. When fertilized with normal loach sperm, some unreduced eggs developed into triploids, but the rest into diploids. Hybridization using goldfish Carassius auratus sperm gave both normal diploid loaches and inviable allotriploid hybrids possessing the diploid loach genome and the haploid goldfish genome. Microsatellite genotyping and DNA fingerprinting demonstrated that the diploid progeny developing from the unreduced eggs were genetically identical to the mother, while the triploids had some of the paternal DNA. These results indicate that the diploid eggs reproduced unisexually as a diploid clone and in other cases developed into triploids after accidental incorporation of sperm nucleus. The presence of at least one clonal line in this area was shown by the identical DNA fingerprint detected in five out of 17 diploid loaches examined.  相似文献

用从20个随机引物中筛选出的7个引物对泥鳅(Misgurnus anguillicaudatus)和大鳞副泥鳅(Paramisgurnus dabryanus)及其自交与杂交子代进行了RAPD遗传稳定性分析。结果表明,RAPD谱带主要呈孟德尔式遗传,该类带纹明亮稳定,在泥鳅子代中占99.11%;在大鳞副泥鳅子代中占100%;在杂交子代中占99.36%。也存在非孟德尔式遗传谱带,该类带纹较暗、稳定性差,在泥鳅子代中为0.89%;在杂交子代中为0.64%;在大鳞副泥鳅中未见此类带纹。实验结果说明RAPD谱带具有很高的遗传稳定性。  相似文献

Phosphoglucomutase of two groups of the loach Cobitis biwae , a fresh-water teleost, was examined by starch gel electrophoresis. Some of the fishes had six-banded electrophoretic patterns not observed in other loach species (e.g. Misgurnus anguillicaudatus and Cobitis delicata ). The complexity of the pattern is explainable by an assumption that the loaches are tetraploid fishes. The result suggests that a diploid-tetraploid relationship exists within the species Cobitis biwae .  相似文献

A human case of creeping eruption due to Gnathostoma nipponicum was confirmed indirectly by finding infective advanced third-stage larvae in leftover largemouth bass meat. Hitherto unknown metacercariae were found encysted in loaches (Misgurnus anguillicaudatus) from China. They were experimentally fed to golden hamsters, and gravid adults were recovered 1 week post-infection from their small intestines. A new species, Massaliatrema misgurni n. sp. (Heterophyidae), is described from the adults. This new species is different from M. gyrinicola Dollfus and Timon-David, 1960, in having a smaller acetabulum/oral sucker ratio, less branched vitellaria widely entering the intercecal anteroacetabular area and an almost median seminal receptacle; and from M. yamashitai Kamiya and Ohbayashi, 1975, in having a larger acetabulum/oral sucker ratio, with the seminal vesicle situated in the uterine loop and the vitellaria entering the intercecal anteroacetabular area. This report is the first record of M. anguillicaudatus as a second intermediate host of the genus Massaliatrema Dollfus and Timon-David, 1960.  相似文献

Adult gnathostomes were discovered in the stomach of the Jeju weasel, Mustela sibilica quelpartis, road-killed in Jeju-do (Province). Their morphological characters were examined to identify the species. Total 50 gnathostome adults were collected from 6 out of 10 weasels examined. In infected weasels, 4-6 worms were grouped and embedded in each granulomatous gastric tumor, except 1 weasel. Male worms were 25.0×1.4 mm in average size, and had a tail with pedunculate papillae, a spicule, and minute tegumental spines. Females were 40.0×2.5 mm in average size, and had a tail without tegumental spines. Pointed and posteriorly curved hooklets were arranged in 8-10 rows on the head bulb. Tegumental spines were distributed from behind the head bulb to the middle portion of the body. The spines were different in size and shape by the distribution level of the body surface. Fertilized eggs were 65.5×38.9 μm in average size, and had a mucoid plug at 1 pole. These gnathostomes from Jeju weasels were identified as Gnathostoma nipponicum Yamaguti, 1941. By the present study, it was confirmed for the first time that G. nipponicum is distributed in Jeju-do, the Republic of Korea, and the Jeju weasel, M. sibilica quelpartis, plays a crucial role for its definitive host.  相似文献

本文报告,应用不同来源肠道菌株,进行了仔鳗肠道微生态重建试验。菌株包括肠微球菌,贝内克菌分离自成鳗体内。非O-I群弧菌(NCV)分离自黑斑泥鰍(Misgurnus anguillicaudatus)肠道。两歧双歧杆菌分离自婴儿粪便。结果表明,两试验组仔鳗肠道微生态重建后,仔鳗的嗜食性比对照组均有显著性提高(t=8.282,p<0.001;t=5.458,p<0.005)。这一结果,可清楚地看出肠道微生态重建,在宿主生理功能上的重要作用。  相似文献

用含低浓度Hg2 的水饲养泥鳅(Misgurnus anguillicaudatus),研究Hg2 污染对视网膜中S100免疫反应阳性结构数量及分布的影响。用免疫组织化学ABC法标记S100免疫阳性(S100-IR)细胞,显微镜下观察S100免疫反应阳性细胞并计数。结果显示,处理组S100首先在神经节细胞层(GCL)、神经纤维层(NFL)表达,然后逐渐在其他各层表达。与对照组相比,处理组S100免疫反应阳性细胞数显著增多,且该变化与环境中无机汞处理时间成正相关;阳性细胞胞体明显膨胀,突起稠密粗大,S100阳性反应强。研究提示,Hg2 的神经毒害作用导致泥鳅视网膜中S100-IR细胞数的增多。推测Hg2 污染可能对视网膜有伤害性影响,而神经胶质细胞在视网膜修复过程中起了重要作用。  相似文献

Triploid loaches Misgurnus anguillicaudatus are derived from unreduced diploid gametes produced by an asexual clonal lineage that normally undergoes gynogenetic reproduction. Here, we have investigated the reproductive system of two types of triploids: the first type carried maternally inherited clonal diploid genomes and a paternally inherited haploid genome from the same population; the second type had the same clonal diploid genomes but a haploid genome from another, genetically divergent population. The germinal vesicles of oocytes from triploid females (3n=75) contained only 25 bivalents, that is, 50 chromosomes. Flow cytometry revealed that the majority of the progeny resulting from fertilization of eggs from triploid females with normal haploid sperm were diploid. This indicates that triploid females mainly produced haploid eggs. Microsatellite analyses of the diploid progeny of triploid females showed that one allele of the clonal genotype was not transmitted to haploid eggs. Moreover, the identity of the eliminated allele differed between the two types of triploids. Our results demonstrate that there is preferential pairing of homologous chromosomes as well as the elimination of unmatched chromosomes in the course of haploid egg formation, that is, meiotic hybridogenesis. Two distinct genomes in the clone suggest its hybrid origin.  相似文献

由G蛋白β2亚基类似物1基因(GNB2L1)编码的蛋白激酶C受体(RACK1)是一个高度保守的锚定蛋白,属于WD40结构域蛋白家族成员,在细胞信号转导等生命过程中发挥着重要作用。本文采用RACE技术和基因克隆技术分别对大鳞副泥鳅(Paramisgurnus dabryanus)和泥鳅(Misgurnus anguillicaudatus)精巢组织的GNB2L1基因c DNA序列进行了克隆。序列分析表明,大鳞副泥鳅GNB2L1基因c DNA序列全长1 115 bp,开放阅读框(ORF)长965 bp,编码317个氨基酸;泥鳅GNB2L1基因c DNA序列的开放阅读框长965 bp,编码317个氨基酸;两种泥鳅GNB2L1基因编码的蛋白与其他鱼类的RACK1蛋白的同源性为94%~97%,且不同进化地位物种的GNB2L1基因均由8个外显子和7个内含子组成。以GNB2L1基因为标记基因,构建的鱼类系统发育树显示,大鳞副泥鳅和泥鳅在进化上的亲缘关系最近。RT-PCR结果显示,GNB2L1基因在大鳞副泥鳅成体各组织中均有表达,且在脑组织的表达量高于其他组织。以上结果表明,GNB2L1基因为一个进化保守基因,可能在大鳞副泥鳅的细胞活动中发挥着重要作用。  相似文献

应用RAPD技术,分析了采自于我国黄河、长江和珠江三大水系中游的大鳞副泥鳅和泥鳅不同群体间的遗传变异。结果表明:种内不同群体间带纹相似度在0.730-0.938之间;遗传距离为0.089-0.245;种间不同群体间的带纹相似度为0.392-0.505,遗传距离为0.620-0.800。两种泥鳞采自武汉的群体,其不同个体间的相似度较之其它群体为低,表明其群体内遗传变异程度较高。  相似文献

A survey was performed to find out the intermediate hosts of Gnathostoma nipponicum in Jeju-do (Province), the Republic of Korea. In August 2009 and 2010, a total of 82 tadpoles, 23 black-spotted pond frogs (Rana nigromaculata), 7 tiger keelback snakes (Rhabdophis tigrinus tigrinus), 6 red-tongue viper snakes (Agkistrodon ussuriensis), and 2 cat snakes (Elaphe dione) were collected in Jeju-do and examined by the pepsin-HCl digestion method. Total 5 gnathostome larvae were detected in 3 (50%) of 6 A. ussuriensis, 70 larvae in 3 of 7 (42.9%) R. tigrinus tigrinus, and 2 larvae in 2 of 82 (8.7%) frogs. No gnathostome larvae were detected in tadpoles and cat snakes. The larvae detected were a single species, and 2.17 × 0.22 mm in average size. They had characteristic head bulbs, muscular esophagus, and 4 cervical sacs. Three rows of hooklets were arranged in the head bulbs, and the number of hooklets in each row was 29, 33, and 36 posteriorly. All these characters were consistent with the advanced third-stage larvae of G. nipponicum. It has been first confirmed in Jeju-do that R. nigromaculata, A. ussuriensis, and R. tigrinus tigrinus play a role for intermediate and/or paratenic hosts for G. nipponicum.  相似文献

In the loach, or Oriental weatherfish Misgurnus anguillicaudatus (Teleostei: Cobitidae), diploid (2n = 50) and tetraploid individuals (4n = 100) are often sympatric in central China. The evolutionary mechanism of this tetraploidization was analyzed with the observation of meiotic behavior of chromosomes in both the germinal vesicles of mature oocytes and the primary spermatocytes in diploid and tetraploid loaches. Whereas diploid specimens usually showed 25 bivalents in meiotic cells, tetraploid loaches exhibited 0-6 quadrivalents and 38-50 bivalents in both sexes, with the modal number of quadrivalents as three in females and four in males. In the diploid specimens, the two largest metacentric chromosomes bearing nucleolar organizing regions (NORs) identified by chromomycin A(3) staining and fluorescence in situ hybridization with a 5.8S + 28S rDNA probe formed one bivalent with terminal association. In the tetraploids, four NOR-bearing chromosomes never formed a quadrivalent, but were organized into two terminally-associated bivalents. These findings suggest an autotetraploid origin of the natural tetraploid loach and subsequent rediploidization of whole genome. The latter process, however, seems still in progress as inferred from the concurrence of up-to several quadrivalents and the majority of bivalents.  相似文献

Male Chinese loaches (Misgurnus anguillicaudatus) were exposed to 17beta-estradiol (E2) in the ambient water at nominal concentrations of 0.5, 1, 5 and 10 mug/L continuously for 8 weeks using a semi-static system. The gonadosomatic index (GSI), total plasma protein, total plasma calcium, magnesium and zinc, and plasma vitellogenin (Vtg) were determined as test endpoints. Our results indicate no significant changes in the GSI between exposed and control groups (P<0.05). All exposure concentrations of E2 dramatically induced the production of Vtg within 7 days. The vitellogenic response of male loach was time- and dose-dependent. At higher concentrations of E2 (5 and 10 microg/L), total plasma protein, total plasma calcium, and total plasma magnesium presented a significant time- and dose-dependent increase, and the three parameters were significantly correlated with plasma Vtg. We conclude that Chinese loaches are sensitive to estrogenic compounds and may be chosen as potential sentinel species in field and laboratory studies. In addition, total plasma protein, total plasma calcium, and total plasma magnesium can be used as indirect indicators to predict Vtg levels in estrogenized Chinese loach.  相似文献
