Because the major processes involved in muscle contraction require rapid utilization of ATP, measurement of ATP utilization can provide important insights into the mechanisms of contraction. It is necessary, however, to differentiate between the contribution made by cross-bridges and that of the sarcoplasmic reticulum (SR) Ca2+ pumps. Specific and potent SR Ca2+ pump blockers have been used in skinned fibers to permit direct measurement of cross-bridge ATP utilization. Up to now, there was no analogous cross-bridge blocker. Recently, N-benzyl-p-toluene sulfonamide (BTS) was found to suppress force generation at micromolar concentrations. We tested whether BTS could be used to block cross-bridge ATP utilization, thereby permitting direct measurement of SR Ca2+ pump ATP utilization in saponin-skinned fibers. At 25 µM, BTS virtually eliminates force and cross-bridge ATP utilization (both <4% of control value). By taking advantage of the toadfish swimbladder muscle's unique right shift in its force-Ca2+ concentration ([Ca2+]) relationship, we measured SR Ca2+ pump ATP utilization in the presence and absence of BTS. At 25 µM, BTS had no effect on SR pump ATP utilization. Hence, we used BTS to make some of the first direct measurements of ATP utilization of intact SR over a physiological range of [Ca2+]at 15°C. Curve fits to SR Ca2+ pump ATP utilization vs. pCa indicate that they have much lower Hill coefficients (1.49) than that describing cross-bridge force generation vs. pCa (5). Inorganic phosphate(Pi) accumulates in the fibers of actively working musclewhere it acts at various sites to modulate contraction. To characterizethe role of Pi as a regulator of the sarcoplasmic reticulum(SR) calcium (Ca2+) release channel, we examined the actionof Pi on purified SR Ca2+ release channels,isolated SR vesicles, and skinned skeletal muscle fibers. In singlechannel studies, addition of Pi to the cis chamberincreased single channel open probability (Po;0.079 ± 0.020 in 0 Pi, 0.157 ± 0.034 in 20 mMPi) by decreasing mean channel closed time; mean channelopen times were unaffected. In contrast, the ATP analog,,-methyleneadenosine 5'-triphosphate (AMP-PCP), enhancedPo by increasing single channel open time anddecreasing channel closed time. Pi stimulation of[3H]ryanodine binding by SR vesicles wassimilar at all concentrations of AMP-PCP, suggesting Pi andadenine nucleotides act via independent sites. In skinned musclefibers, 40 mM Pi enhanced Ca2+-inducedCa2+ release, suggesting an in situ stimulation ofthe release channel by high concentrations of Pi. Ourresults support the hypothesis that Pi may be an importantendogenous modulator of the skeletal muscle SR Ca2+ releasechannel under fatiguing conditions in vivo, acting via a mechanismdistinct from adenine nucleotides.

Recent studies on the role of nitric oxide (NO) ingastrointestinal smooth muscle have raised the possibility thatNO-stimulated cGMP could, in the absence of cGMP-dependent proteinkinase (PKG) activity, act as aCa2+-mobilizing messenger[K. S. Murthy, K.-M. Zhang, J.-G. Jin, J. T. Grider, and G. M. Makhlouf. Am. J. Physiol. 265 (Gastrointest. Liver Physiol. 28):G660-G671, 1993]. This notion was examined indispersed gastric smooth muscle cells with 8-bromo-cGMP (8-BrcGMP) andwith NO and vasoactive intestinal peptide (VIP), which stimulate endogenous cGMP. In muscle cells treated with cAMP-dependent protein kinase (PKA) and PKG inhibitors (H-89 and KT-5823), 8-BrcGMP (10 µM),NO (1 µM), and VIP (1 µM) stimulated45Ca2+release (21 ± 3 to 30 ± 1% decrease in45Ca2+cell content); Ca2+ releasestimulated by 8-BrcGMP was concentration dependent with anEC50 of 0.4 ± 0.1 µM and athreshold of 10 nM. 8-BrcGMP and NO increased cytosolic freeCa2+ concentration([Ca2+]i)and induced contraction; both responses were abolished after Ca2+ stores were depleted withthapsigargin. With VIP, which normally increases[Ca2+]iby stimulating Ca2+ influx,treatment with PKA and PKG inhibitors caused a further increase in[Ca2+]ithat reverted to control levels in cells pretreated with thapsigargin. Neither Ca2+ release norcontraction induced by cGMP and NO in permeabilized muscle cells wasaffected by heparin or ruthenium red.Ca2+ release induced by maximallyeffective concentrations of cGMP and inositol 1,4,5-trisphosphate(IP3) was additive, independent of which agent was applied first. We conclude that, in the absence ofPKA and PKG activity, cGMP stimulatesCa2+ release from anIP3-insensitive store and that itseffect is additive to that of IP3.

The mechanism underlying H2O2-inducedactivation of frog skeletal muscle ryanodine receptors was studiedusing skinned fibers and by measuring single Ca2+-releasechannel current. Exposure of skinned fibers to 3-10 mM H2O2 elicited spontaneous contractures.H2O2 at 1 mM potentiated caffeine contracture.When the Ca2+-release channels were incorporated into lipidbilayers, open probability (Po) and open timeconstants were increased on intraluminal addition ofH2O2 in the presence of cis catalase,but unitary conductance and reversal potential were not affected.Exposure to cis H2O2 at 1.5 mM failedto activate the channel in the presence of trans catalase.Application of 1.5 mM H2O2 to the transside of a channel that had been oxidized by cisp-chloromercuriphenylsulfonic acid (pCMPS; 50 µM) still led to anincrease in Po, comparable to that elicited bytrans 1.5 mM H2O2 without pCMPS.Addition of cis pCMPS to channels that had been treated with orwithout trans H2O2 rapidly resulted inhigh Po followed by closure of the channel. Theseresults suggest that oxidation of luminal sulfhydryls in theCa2+-release channel may contribute toH2O2-induced channel activation and musclecontracture.

Fibroblasts form fibers when grown inculture medium containing native type 1 collagen. The contractileforces generated can be precisely quantified and used to analyze thesignal transduction pathways regulating fibroblast contraction. Calfserum (30%) induces a sustained contraction that is accompanied by atransient increase in intracellular calcium([Ca2+]i). W-7, a calmodulin inhibitor,KN-62, an inhibitor of calcium/calmodulin-dependent protein kinase, andML-7, a myosin light-chain kinase inhibitor, had no effects on eitherthe contraction or the [Ca2+]i responses.Neither genistein, a tyrosine kinase inhibitor, nor calphostin C, aprotein kinase C inhibitor, had major effects on force or[Ca2+]i. In contrast, the Rho kinaseinhibitors(R)-(+)-trans-N-(4-pyridyl)-4-(1-aminoethyl)-cyclohexanecarboxamide (Y-27632) and HA1077 depressed the contraction in a dose-dependent manner without affecting the [Ca2+]iresponse. Stress fiber formation was also suppressed by Y-27632. Surprisingly, calf serum, Y-27632, and calf serum plus Y-27632 did notalter mono- or diphosphorylation of the myosin regulatory light chain(MRLC) compared with control untreated fibers. These results suggestthat the sustained contraction of NIH 3T3 fibroblast fibers induced bycalf serum is mediated by Rho kinase but is independent of a sustainedincrease in [Ca2+]i, calcium/calmodulin- orprotein kinase C-dependent pathways, or increases in MRLC phosphorylation.

The regulationof intracellular Ca2+ signals in smooth muscle cells andarterial diameter by intravascular pressure was investigated in ratcerebral arteries (~150 µm) using a laser scanning confocal microscope and the fluorescent Ca2+ indicator fluo 3. Elevation of pressure from 10 to 60 mmHg increased Ca2+spark frequency 2.6-fold, Ca2+ wave frequency 1.9-fold, andglobal intracellular Ca2+ concentration([Ca2+]i) 1.4-fold in smooth muscle cells,and constricted arteries. Ryanodine (10 µM), an inhibitor ofryanodine-sensitive Ca2+ release channels, or thapsigargin(100 nM), an inhibitor of the sarcoplasmic reticulumCa2+-ATPase, abolished sparks and waves, elevated global[Ca2+]i, and constricted pressurized (60 mmHg) arteries. Diltiazem (25 µM), a voltage-dependentCa2+ channel (VDCC) blocker, significantly reduced sparks,waves, and global [Ca2+]i, and dilatedpressurized (60 mmHg) arteries. Steady membrane depolarization elevatedCa2+ signaling similar to pressure and increased transientCa2+-sensitive K+ channel current frequencye-fold for ~7 mV, and these effects were prevented by VDCCblockers. Data are consistent with the hypothesis that pressure inducesa steady membrane depolarization that activates VDCCs, leading to anelevation of spark frequency, wave frequency, and global[Ca2+]i. In addition, pressure inducescontraction via an elevation of global[Ca2+]i, whereas the net effect of sparks andwaves, which do not significantly contribute to global[Ca2+]i in arteries pressurized to between 10 and 60 mmHg, is to oppose contraction.

To determine theeffect of plasma growth hormone (GH) on skeletal muscle function, wemeasured the free Ca2+concentration-tension relationship of slow-twitch (soleus) and fast-twitch (peroneus longus) muscles isolated from rats undergoing acromegaly in response to implanted, GH-secreting tumors. Muscles fromadult (9 mo) and aged rats (24 mo) were studied after the tumor-bearingrats weighed over 50% more than their age-matched controls.Ca2+-activated isometric tensionwas recorded from skinned muscle fibers. For soleus muscles, the freeCa2+ concentration producing 50%of maximal tension([Ca2+]50)was 2.0 µM for rats with tumors and 3.4-3.6 µM for controls. For peroneus longus fibers,[Ca2+]50shifted from 6.1-6.7 µM in controls to 3.5 µM after tumors were introduced into either adult or aged rats. Soleus muscle fibersfrom neonatal rats (14 days) were less sensitive toCa2+ than those isolated fromadult rats, having a[Ca2+]50of 7.3 µM. The Ca2+ sensitivityof peroneus longus fibers did not change with age. We conclude thatsignificant increases in myofibrillarCa2+ sensitivity occur in skeletalmuscles undergoing rapid growth induced by GH-secreting tumors.

To find whether cytoplasmic streaming in Acetabularia is controlledby Ca2+, a tonoplast-permeabilized cell model was prepared usinga vacuolar perfusion technique. The cytoplasmic streaming remainedalmost normal after perfusion with EGTA medium (10 mM EGTA,40 mM PIPES, 5mM MgCl2 and 800 mM sorbitol, pH 6.9), but stoppedwithin 10 min when saponin medium (EGTA medium plus 50 µg/mlsaponin, 50 µg/ml hexokinase and 5 mM glucose) was perfused.This model system was reactivated with a solution containing0.5 mM ATP and different concentrations of Ca2+ (reactivationmedium). With the reactivation medium at pCa 6–5, theresumed streaming lasted for about 10 min before the cytoplasmaggregated. At pCa 4–3, the streaming was observed onlyfor a few minutes because the cytoplasm aggregated quickly.At pCa 7, no reactivated movement was observed. Reactivationwas not induced in an ATP- or Mg2+-deficient medium even inthe presence of an adequate concentration of Ca2+, and was inhibitedby 50 µg/ml cytochalasin B or 1 mM N-ethylmaleimide. We concluded from these observations that the cytoplasmic streamingin Acetabularia is very likely to be driven by the actomyosinsystem in the presence of Mg-ATP and Ca2+ at pCa 6–5. We investigatedthe role of intracellular calcium concentration([Ca2+]i) in endothelin-1 (ET-1) production,the effects of potential vasospastic agents on[Ca2+]i, and the presence of L-typevoltage-dependent Ca2+ channels in cerebral microvascularendothelial cells. Primary cultures of endothelial cells isolated frompiglet cerebral microvessels were used. Confluent cells were exposed toeither the thromboxane receptor agonist U-46619 (1 µM),5-hydroxytryptamine (5-HT; 0.1 mM), or lysophosphatidic acid (LPA; 1 µM) alone or after pretreatment with the Ca2+-chelatingagent EDTA (100 mM), the L-type Ca2+ channel blockerverapamil (10 µM), or the antagonist of receptor-operated Ca2+ channel SKF-96365 HCl (10 µM) for 15 min. ET-1production increased from 1.2 (control) to 8.2 (U-46619), 4.9 (5-HT),or 3.9 (LPA) fmol/µg protein, respectively. Such elevated ET-1biosynthesis was attenuated by verapamil, EDTA, or SKF-96365 HCl. Toinvestigate the presence of L-type voltage-dependent Ca2+channels in endothelial cells, the [Ca2+]isignal was determined fluorometrically by using fura 2-AM. Superfusionof confluent endothelial cells with U-46619, 5-HT, or LPA significantlyincreased [Ca2+]i. Pretreatment ofendothelial cells with high K+ (60 mM) or nifedipine (4 µM) diminished increases in [Ca2+]i inducedby the vasoactive agents. These results indicate that 1)elevated [Ca2+]i signals are involved in ET-1biosynthesis induced by specific spasmogenic agents, 2) theincreases in [Ca2+]i induced by thevasoactive agents tested involve receptor as well as L-typevoltage-dependent Ca2+ channels, and 3) primarycultures of cerebral microvascular endothelial cells express L-typevoltage-dependent Ca2+ channels.

Soleus musclefibers from the rat display a reduction in peak power andCa2+ sensitivity after hindlimbsuspension. To examine human responses to non-weight bearing, weobtained soleus biopsies from eight adult men before and immediatelyafter 17 days of bed rest (BR). Single chemically skinned fibers weremounted between a force transducer and a servo-controlled positionmotor and activated with maximal (isotonic properties) and/orsubmaximal (Ca2+ sensitivity)levels of free Ca2+. Gelelectrophoresis indicated that all pre- and post-BR fibers expressedtype I myosin heavy chain. Post-BR fibers obtained from one subjectdisplayed increases in peak power andCa2+ sensitivity. In contrast,post-BR fibers obtained from the seven remaining subjects showed anaverage 11% reduction in peak power (P < 0.05), with each individualdisplaying a 7-27% reduction in this variable. Post-BR fibersfrom these subjects were smaller in diameter and produced 21% lessforce at the shortening velocity associated with peak power. However,the shortening velocity at peak power output was elevated 13% in thepost-BR fibers, which partially compensated for their lower force.Post-BR fibers from these same seven subjects also displayed a reducedsensitivity to free Ca2+(P < 0.05). These results indicatethat the reduced functional capacity of human lower limb extensormuscles after BR may be in part caused by alterations in thecross-bridge mechanisms of contraction.

We investigatedthe relationship between voltage-operatedCa2+ channel current and thecorresponding intracellular Ca2+concentration([Ca2+]i)change (Ca2+ transient) in guineapig gastric myocytes. Fluorescence microspectroscopy was combined withconventional whole cell patch-clamp technique, and fura 2 (80 µM) wasadded to CsCl-rich pipette solution. Step depolarization to 0 mVinduced inward Ca2+ current(ICa) andconcomitantly raised[Ca2+]i.Both responses were suppressed by nicardipine, an L-typeCa2+ channel blocker, and thevoltage dependence of Ca2+transient was similar to the current-voltage relation ofICa. When pulseduration was increased by up to 900 ms, peakCa2+ transient increased andreached a steady state when stimulation was for longer. The calculatedfast Ca2+ buffering capacity(B value), determined as the ratio ofthe time integral ofICa divided bythe amplitude of Ca2+ transient,was not significantly increased after depletion of Ca2+ stores by the cyclicapplication of caffeine (10 mM) in the presence of ryanodine (4 µM).The addition of cyclopiazonic acid (CPA, 10 µM), a sarco(endo)plasmicreticulum Ca2+-ATPase inhibitor,decreased B value by ~20% in areversible manner. When KCl pipette solution was used,Ca2+-activatedK+ current[IK(Ca)]was also recorded during step depolarization. CPA sensitivelysuppressed the initial peak and oscillations of IK(Ca) withirregular effects on Ca2+transients. The above results suggest that, in guinea pig gastric myocyte, Ca2+ transient is tightlycoupled to ICaduring depolarization, and global[Ca2+]iis not significantly affected byCa2+-inducedCa2+ release from sarcoplasmicreticulum during depolarization.

Inskeletal muscle fibers, the intracellular loop between domains II andIII of the 1-subunit of the dihydropyridine receptor (DHPR) may directly activate the adjacent Ca2+ releasechannel in the sarcoplasmic reticulum. We examined the effects ofsynthetic peptide segments of this loop on Ca2+ release inmechanically skinned skeletal muscle fibers with functional excitation-contraction coupling. In rat fibers at physiological Mg2+ concentration ([Mg2+]; 1 mM), a20-residue skeletal muscle DHPR peptide[AS(20);Thr671-Leu690; 30 µM], shown previously toinduce Ca2+ release in a triad preparation, caused onlysmall spontaneous force responses in ~40% of fibers, although itpotentiated responses to depolarization and caffeine in all fibers. TheCOOH-terminal half of AS(20)[AS(10)] induced much larger spontaneousresponses but also caused substantial inhibition of Ca2+release to both depolarization and caffeine. Both peptides induced orpotentiated Ca2+ release even when the voltage sensors wereinactivated, indicating direct action on the Ca2+ releasechannels. The corresponding 20-residue cardiac DHPR peptide [AC(20);Thr793-Ala812] was ineffective, but itsCOOH-terminal half [AC(10)] had effects similar to AS(20). In the presence of lower[Mg2+] (0.2 mM), exposure to eitherAS(20) or AC(10) (30 µM) induced substantial Ca2+ release. PeptideCS (100 µM), a loop segment reported to inhibit Ca2+ release in triads, caused partial inhibition ofdepolarization-induced Ca2+ release. In toad fibers, eachof the A peptides had effects similar to or greater than those in ratfibers. These findings suggest that the A and C regions of the skeletalDHPR II-III loop may have important roles in vivo.

We used a reconstituted fiber formed when 3T3fibroblasts are grown in collagen to characterize nonmusclecontractility and Ca2+ signaling. Calf serum (CS) andthrombin elicited reversible contractures repeatable for >8 h. CSelicited dose-dependent increases in isometric force; 30% produced thelargest forces of 106 ± 12 µN (n = 30), whichis estimated to be 0.5 mN/mm2 cell cross-sectionalarea. Half times for contraction and relaxation were 4.7 ± 0.3 and 3.1 ± 0.3 min at 37°C. With imposition of constant shortening velocities, force declined with time, yieldingtime-dependent force-velocity relations. Forces at 5 s fit thehyperbolic Hill equation; maximum velocity(Vmax) was 0.035 ± 0.002 Lo/s.Compliance averaged 0.0076 ± 0.0006 Lo/Fo. Disruption of microtubules with nocodazole in a CS-contracted fiber had no net effects on force, Vmax, or stiffness; force increased in 8, butdecreased in 13, fibers. Nocodazole did not affect baselineintracellular Ca2+ concentration([Ca2+]i) but reduced (~30%) the[Ca2+]i response to CS. The force afternocodazole treatment was the primary determinant of stiffness andVmax, suggesting that microtubules were not amajor component of fiber internal mechanical resistance. Cytochalasin Dhad major inhibitory effects on all contractile parameters measured butlittle effect on [Ca2+]i.

J. Appl.Physiol. 83(2): 444-450, 1997.This investigationtested the notion that fatiguing stimulation induces intrinsic changes in the contractile apparatus and sarcoplasmic reticulum (SR) and thatthese changes are initiated by elevated intracellularCa2+ concentration([Ca2+]i).Immediately after stimulation of frog semitendinosus muscle, contractile apparatus and SR function were measured. Despite a largedecline in tetanic force (Po),maximal Ca2+-activated force(Fmax) of the contractileapparatus was not significantly altered. However,Ca2+ sensitivity was increased. Inconjunction, the rate constant ofCa2+ uptake by the SR wasdiminished, and the caffeine sensitivity ofCa2+ release was decreased. Duringrecovery, Po, contractileapparatus, and SR function each returned to near-initial levels.Exposure of skinned fibers to 0.5 µM freeCa2+ for 5 min depressed bothFmax andCa2+ sensitivity of thecontractile apparatus. In addition, caffeine sensitivity ofCa2+ release was diminished.Results suggest that fatigue induces intrinsic alterations incontractile apparatus and SR function. Changes in contractile apparatusfunction do not appear to be mediated by increased[Ca2+]i.However, a portion of the change in SRCa2+ release seems to be due toelevated[Ca2+]i.

Chemically skinned muscle fibers,prepared from the rat medial gastrocnemius and soleus, were subjectedto four sequential slack tests in Ca2+-activating solutionscontaining 0, 15, 30, and 0 mM added Pi. Pi (15 and 30 mM) had no effect on the unloaded shortening velocity (Vo) of fibers expressing type IIb myosin heavychain (MHC). For fibers expressing type I MHC, 15 mM Pi didnot alter Vo, whereas 30 mM Pireduced Vo to 81 ± 1% of the original 0 mM Pi value. This effect was readily reversible whenPi was lowered back to 0 mM. These results are notcompatible with current cross-bridge models, developed exclusively fromdata obtained from fast fibers, in which Vo isindependent of Pi. The response of the type I fibers at 30 mM Pi is most likely the result of increased internal drag opposing fiber shortening resulting from fiber type-specific effects ofPi on cross bridges, the thin filament, or therate-limiting step of the cross-bridge cycle.

The myoplasmic free Ca2+concentration([Ca2+]i)was measured in intact single fibers from mouse skeletal muscle withthe fluorescent Ca2+ indicatorindo 1. Some fibers were perfused in a solution in which theconcentration of Na+ was reducedfrom 145.4 to 0.4 mM (low-Na+solution) in an attempt to activate reverse-modeNa+/Ca2+exchange (Ca2+ entry in exchangefor Na+ leaving the cell). Undernormal resting conditions, application oflow-Na+ solution only increased[Ca2+]iby 5.8 ± 1.8 nM from a mean resting[Ca2+]iof 42 nM. In other fibers,[Ca2+]iwas elevated by stimulating sarcoplasmic reticulum (SR)Ca2+ release with caffeine (10 mM)and by inhibiting SR Ca2+ uptakewith2,5-di(tert-butyl)-1,4-benzohydroquinone(TBQ; 0.5 µM) in an attempt to activate forward-modeNa+/Ca2+exchange (Ca2+ removal from thecell in exchange for Na+ influx).These two agents caused a large increase in[Ca2+]i,which then declined to a plateau level approximately twice the baseline[Ca2+]iover 20 min. If the cell was allowed to recover between exposures tocaffeine and TBQ in a solution in whichCa2+ had been removed, theincrease in[Ca2+]iduring the second exposure was very low, suggesting thatCa2+ had left the cell during theinitial exposure. Application of caffeine and TBQ to a preparation inlow-Na+ solution produced a large,sustained increase in[Ca2+]iof ~1 µM. However, when cells were exposed to caffeine and TBQ in alow-Na+ solution in whichCa2+ had been removed, a sustainedincrease in[Ca2+]iwas not observed, although[Ca2+]iremained higher and declined slower than in normalNa+ solution. This suggests thatforward-modeNa+/Ca2+exchange contributed to the fall of[Ca2+]iin normal Na+ solution, but whenextracellular Na+ was low, aprolonged elevation of[Ca2+]icould activate reverse-modeNa+/Ca2+exchange. The results provide evidence that skeletal muscle fibers possess aNa+/Ca2+exchange mechanism that becomes active in its forward mode when [Ca2+]iis increased to levels similar to that obtained during contraction.

In cultured porcine aortic endothelial monolayers, theeffect of ATP on myosin light chain (MLC) phosphorylation, whichcontrols the endothelial contractile machinery, was studied. ATP (10 µM) reduced MLC phosphorylation but increased cytosolicCa2+ concentration ([Ca2+]i).Inhibition of the ATP-evoked [Ca2+]i rise byxestospongin C (10 µM), an inhibitor of the inositol trisphosphate-dependent Ca2+ release from endoplasmicreticulum, did not affect the ATP-induced dephosphorylation of MLC. MLCdephosphorylation was prevented in the presence of calyculin A (10 nM),an inhibitor of protein phosphatases PP-1 and PP-2A. Thus ATP activatesMLC dephosphorylation in a Ca2+-independent manner. In thepresence of calyculin A, MLC phosphorylation was incremented afteraddition of ATP, an effect that could be abolished when cellswere loaded with the Ca2+ chelator1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acidacetoxymethyl ester (10 µM). Thus ATP also activates aCa2+-dependent kinase acting on MLC. In summary, ATPsimultaneously stimulates a functional antagonism toward bothphosphorylation and dephosphorylation of MLC in which thedephosphorylation prevails. In endothelial cells, ATP is the firstphysiological mediator identified to activate MLC dephosphorylation bya Ca2+-independent mechanism.

Bovine adrenalzona fasciculata cells (AZF) express a noninactivatingK+ current(IAC) whoseinhibition by adrenocorticotropic hormone and ANG II may be coupled tomembrane depolarization andCa2+-dependentcortisol secretion. We studiedIACinhibition byCa2+ and theCa2+ionophore ionomycin in whole cell and single-channel patch-clamp recordings of AZF. In whole cell recordings with intracellular (pipette)Ca2+concentration([Ca2+]i)buffered to 0.02 µM,IAC reachedmaximum current density of 25.0 ± 5.1 pA/pF(n = 16); raising[Ca2+]ito 2.0 µM reduced it 76%. In inside-out patches, elevated[Ca2+]idramatically reducedIAC channelactivity. Ionomycin inhibited IAC by 88 ± 4% (n = 14) without altering rapidlyinactivating A-type K+ current.Inhibition of IACby ionomycin was unaltered by adding calmodulin inhibitory peptide tothe pipette or replacing ATP with its nonhydrolyzable analog5'-adenylylimidodiphosphate.IAC inhibition byionomycin was associated with membrane depolarization. When[Ca2+]iwas buffered to 0.02 µM with 2 and 11 mM1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid (BAPTA), ionomycin inhibitedIAC by 89.6 ± 3.5 and 25.6 ± 14.6% and depolarized the same AZF by 47 ± 8 and 8 ± 3 mV, respectively (n = 4). ANG II inhibitedIAC significantlymore effectively when pipette BAPTA was reduced from 11 to 2 mM. Raising[Ca2+]iinhibits IACthrough a mechanism not requiring calmodulin or protein kinases,suggesting direct interaction withIAC channels. ANGII may inhibitIAC anddepolarize AZF by activating parallel signaling pathways, one of whichuses Ca2+ asa mediator.

We testedthe hypothesis that strain is the primary mechanical signal in themechanosensitive modulation of intracellular Ca2+concentration ([Ca2+]i) in airway smoothmuscle. We found that [Ca2+]i wassignificantly correlated with muscle length during isotonic shorteningagainst 20% isometric force (Fiso). When the isotonic loadwas changed to 50% Fiso, data points from the 20 and 50% Fiso experiments overlapped in thelength-[Ca2+]i relationship. Similarly, datapoints from the 80% Fiso experiments clustered near thosefrom the 50% Fiso experiments. Therefore, despite 2.5- and4-fold differences in external load, [Ca2+]idid not deviate much from the length-[Ca2+]irelation that fitted the 20% Fiso data. Maximal inhibition of sarcoplasmic reticular (SR) Ca2+ uptake by 10 µMcyclopiazonic acid (CPA) did not significantly change[Ca2+]i in carbachol-induced isometriccontractions and isotonic shortening. CPA also did not significantlychange myosin light-chain phosphorylation or force redevelopment whencarbachol-activated muscle strips were quickly released from optimallength (Lo) to 0.5 Lo. These results are consistent with thehypothesis and suggest that SR Ca2+ uptake is not theunderlying mechanism.

To investigatethe Ca2+-dependent plasticity ofsarcoplasmic reticulum (SR) function in vascular smooth muscle,transient responses to agents releasing intracellularCa2+ by either ryanodine(caffeine) orD-myo-inositol1,4,5-trisphosphate [IP3;produced in response to norepinephrine (NE),5-hydroxytryptamine (5-HT), arginine vasopressin (AVP)] receptorsin rat tail arterial rings were evaluated after 4 days of organculture. Force transients induced by all agents were increased comparedwith those induced in fresh rings. Stimulation by 10% FCSduring culture further potentiated the force andCa2+ responses to caffeine (20 mM)but not to NE (10 µM), 5-HT (10 µM), or AVP (0.1 µM). The effectwas persistent, and SR capacity was not altered after reversibledepletion of stores with cyclopiazonic acid. The effects of serum couldbe mimicked by culture in depolarizing medium (30 mMK+) and blocked by the additionof verapamil (1 µM) or EGTA (1 mM) to the medium, loweringintracellular Ca2+ concentration([Ca2+]i)during culture. These results show that modulation of SR function canoccur in vitro by a mechanism dependent on long-term levels of basal[Ca2+]iand involving ryanodine- but notIP3 receptor-mediatedCa2+release.  相似文献
