Microbial fuel cell (MFC) is an emerging technology for sustainable energy generation and waste treatment. This paper reviews the potential of a gaseous substrate when it is combined with a mediator in an MFC to generate electricity and to treat toxic gaseous pollutants. Most MFCs for waste water treatment often cannot use mediator to enhance the electron transfer from the microbe to the anode because of the difficulty in recovering the expensive and potentially toxic compound. Combining gas feeds with mediators is possible since the soluble mediator would remain in the anode chamber as the gas passes through the reactor. In addition, this type of MFC is possible to be integrated into an anaerobic biofiltration system (BF-MFC), where the biofilter removes the gaseous contaminant and produces the reduced mediator and the MFC produces the electricity and recycles the reoxidised mediator. This paper also talks about the past research on gaseous feed MFCs, and reviews the mechanism and strategies of electron transfer in MFC using redox mediator. The advantages, process parameters and challenges of BF-MFC are discussed. This knowledge is very much required in the design and scale up of BF-MFC. This paper will be useful for those who work in the area of gaseous pollutant treatment and electricity generation.

New exoelectrogen Citrobacter sp. SX-1 isolated from a microbial fuel cell
Aims: Isolation, identification and characterization of a new exoelectrogenic bacterium from a microbial fuel cell (MFC). Methods and Results: Exoelectrogenic bacterial strain SX‐1 was isolated from a mediator‐less MFC by conventional plating techniques with ferric citrate as electron acceptor under anaerobic condition. Phylogenetic analysis of the 16S rDNA sequence revealed that it was related to the members of Citrobacter genus with Citrobacter sp. sdy‐48 being the most closely related species. The bacterial strain SX‐1 produced electricity from citrate, acetate, glucose, sucrose, glycerol and lactose in MFCs with the highest current density of 205 mA m?2 generated from citrate. Cyclic voltammetry analysis indicated that membrane‐associated proteins may play an important role in facilitating the electrons transferring from bacteria to electrode. Conclusions: This is the first study that demonstrates that Citrobacter species can transfer electrons to extracellular electron acceptors. Citrobacter strain SX‐1 is capable of generating electricity from a wide range of substrates in MFCs. Significance and Impact of the Study: This finding increases the known diversity of power generating exoelectrogens and provided a new strain to explore the mechanisms of extracellular electron transfer from bacteria to electrode. The wide range of substrate utilization by SX‐1 increases the application potential of MFCs in renewable energy generation and waste treatment.

Endogenously produced, diffusible redox mediators can act as electron shuttles for bacterial respiration. Accordingly, the mediators also serve a critical role in microbial fuel cells (MFCs), as they assist extracellular electron transfer from the bacteria to the anode serving as the intermediate electron sink. Electrochemical impedance spectroscopy (EIS) may be a valuable tool for evaluating the role of mediators in an operating MFC. EIS offers distinct advantages over some conventional analytical methods for the investigation of MFC systems because EIS can elucidate the electrochemical properties of various charge transfer processes in the bio‐energetic pathway. Preliminary investigations of Shewanella oneidensis DSP10‐based MFCs revealved that even low quantities of extracellular mediators significantly influence the impedance behavior of MFCs. EIS results also suggested that for the model MFC studied, electron transfer from the mediator to the anode may be up to 15 times faster than the electron transfer from bacteria to the mediator. When a simple carbonate membrane separated the anode and cathode chambers, the extracellular mediators were also detected at the cathode, indicating diffusion from the anode under open circuit conditions. The findings demonstrated that EIS can be used as a tool to indicate presence of extracellular redox mediators produced by microorganisms and their participation in extracellular electron shuttling. In microbial fuel cells and electrolysis cells (MXCs), anode-respiring bacteria (ARB) oxidize organic substrates to produce electrical current. In order to develop an electrical current, ARB must transfer electrons to a solid anode through extracellular electron transfer (EET). ARB use various EET mechanisms to transfer electrons to the anode, including direct contact through outer-membrane proteins, diffusion of soluble electron shuttles, and electron transport through solid components of the extracellular biofilm matrix. In this review, we perform a novel kinetic analysis of each EET mechanism by analyzing the results available in the literature. Our goal is to evaluate how well each EET mechanism can produce a high current density (>10 A m−2) without a large anode potential loss (less than a few hundred millivolts), which are feasibility goals of MXCs. Direct contact of ARB to the anode cannot achieve high current densities due to the limited number of cells that can come in direct contact with the anode. Slow diffusive flux of electron shuttles at commonly observed concentrations limits current generation and results in high potential losses, as has been observed experimentally. Only electron transport through a solid conductive matrix can explain observations of high current densities and low anode potential losses. Thus, a study of the biological components that create a solid conductive matrix is of critical importance for understanding the function of ARB.

Microbial fuel cells: novel biotechnology for energy generation
Microbial fuel cells (MFCs) provide new opportunities for the sustainable production of energy from biodegradable, reduced compounds. MFCs function on different carbohydrates but also on complex substrates present in wastewaters. As yet there is limited information available about the energy metabolism and nature of the bacteria using the anode as electron acceptor; few electron transfer mechanisms have been established unequivocally. To optimize and develop energy production by MFCs fully this knowledge is essential. Depending on the operational parameters of the MFC, different metabolic pathways are used by the bacteria. This determines the selection and performance of specific organisms. Here we discuss how bacteria use an anode as an electron acceptor and to what extent they generate electrical output. The MFC technology is evaluated relative to current alternatives for energy generation.

A microbial fuel cell (MFC) is a relatively new type of fixed film bioreactor for wastewater treatment, and the most effective methods for inoculation are not well understood. Various techniques to enrich electrochemically active bacteria on an electrode were therefore studied using anaerobic sewage sludge in a two-chambered MFC. With a porous carbon paper anode electrode, 8 mW/m2 of power was generated within 50 h with a Coulombic efficiency (CE) of 40%. When an iron oxide-coated electrode was used, the power and the CE reached 30 mW/m2 and 80%, respectively. A methanogen inhibitor (2-bromoethanesulfonate) increased the CE to 70%. Bacteria in sludge were enriched by serial transfer using a ferric iron medium, but when this enrichment was used in a MFC the power was lower (2 mW/m2) than that obtained with the original inoculum. By applying biofilm scraped from the anode of a working MFC to a new anode electrode, the maximum power was increased to 40 mW/m2. When a second anode was introduced into an operating MFC the acclimation time was not reduced and the total power did not increase. These results suggest that these active inoculating techniques could increase the effectiveness of enrichment, and that start up is most successful when the biofilm is harvested from the anode of an existing MFC and applied to the new anode.

The microbial communities associated with electrodes in closed and open circuit microbial fuel cells (MFCs) fed with glucose were analyzed by 16S rRNA approach and compared. The comparison revealed that bacteria affiliated with the Aeromonas sp. within the Gammaproteobacteria constituted the major population in the closed circuit MFC (harvesting electricity) and considered to play important roles in current generation. We, therefore, attempted to isolate the dominant bacteria from the anode biofilm, successfully isolated a Fe (III)‐reducing bacterium phylogenetically related to Aeromonas sp. and designated as strain ISO2‐3. The isolated strain ISO2‐3 could grow and concomitantly produce current (max. 0.24 A/m2) via oxidation of glucose or hydrogen with an electrode serving as the sole electron acceptor. The strain could ferment glucose, but generate less electrical current. Cyclic voltammetry supported the strain ISO2‐3 was electrically active and likely to transfer electrons to the electrode though membrane‐associated compounds (most likely c‐type cytochrome). This mechanism requires intimate contact with the anode surface. Scanning electron microscopy revealed that the strain ISO2‐3 developed multiplayer biofilms on the anode surface and also produced anchor‐like filamentous appendages (most likely pili) that may promote long‐range electron transport across the thick biofilm. Microbially catalyzed treatment of wastewater was evaluated in both the anode and cathode chambers in dual chambered microbial fuel cell (MFC) under varying biocathode microenvironment. MFC operation with aerobic biocathode showed significant increment in both TDS (cathode, 90.2±1%; anode, 39.7±0.5%) and substrate (cathode, 98.07±0.06%; anode, 96.2±0.3%) removal compared to anaerobic biocathode and abiotic cathode operations (COD, 80.25±0.3%; TDS, 30.5±1.2%). Microbially catalyzed reduction of protons and electrons at cathode will be higher during aerobic biocathode operation which leads to gradual substrate removal resulting in stable bio-potential for longer periods facilitating salts removal. Bio-electro catalytic behavior showed higher exchange current density during aerobic biocathode operation resulting in induced electrochemical oxidation which supports the enhanced treatment. Anaerobic biocathode operation depicted relatively less TDS removal (anode, 16.35%; cathode, 16.04%) in both the chambers in spite of good substrate degradation (anode, 84%; cathode, 87.39%). Both the chambers during anaerobic biocathode operation competed as electron donors resulting in negligible bio-potential development.

A mediator-less three-stage two-chamber microbial fuel cell (MFC) system was developed and operated continuously for more than 1.5 years to evaluate continuous power generation while treating artificial wastewater containing glucose (10 mM) concurrently. A stable power density of 28 W/m3 was attained with an anode hydraulic retention time of 4.5 h and phosphate buffer as the cathode electrolyte. An overall dissolved organic carbon removal ratio was about 85%, and coulombic efficiency was about 46% in this MFC system. We also analyzed the microbial community structure of anode biofilms in each MFC. Since the environment in each MFC was different due to passing on the products to the next MFC in series, the microbial community structure was different accordingly. The anode biofilm in the first MFC consisted mainly of bacteria belonging to the Gammaproteobacteria, identified as Aeromonas sp., while the Firmicutes dominated the anode biofilms in the second and third MFCs that were mainly fed with acetate. Cyclic voltammetric results supported the presence of a redox compound(s) associated with the anode biofilm matrix, rather than mobile (dissolved) forms, which could be responsible for the electron transfer to the anode. Scanning electron microscopy revealed that the anode biofilms were comprised of morphologically different cells that were firmly attached on the anode surface and interconnected each other with anchor-like filamentous appendages, which might support the results of cyclic voltammetry. The end-product profile of the glucose fermentation by Propionibacterium freudenreichii ET-3 changed on an electrochemical treatment, in which the culture vessel was filled with a carbon felt anode. Acetate and propionate were produced as final end products in a molar ratio of 2:3 without any electrochemical treatments at the point of the consumption of lactate as an intermediate of the glucose fermentation. The ratio was changed to 1:1 at the point of the lactate consumption by the electrochemical incubation at an electrode potential of 0.4 V versus Ag|AgCl for 100 h. During further electrochemical incubation, propionate was oxidized to acetate as a final end-product in the microbe-containing anode chamber. 1,4-Dihydroxy-2-naphthoic acid produced by P. freudenreichii ET-3 itself would receive electrons from the metabolic pathway and serve as an electron transfer mediator from the microbial cells to the electrode.
