Manifestation of pleiotropic effects in the isogenic variants of luminescent bacteria Photobacterium leiognathi 54 was investigated. The decrease or increase of the expression level of bioluminescence was caused by changes in lux operon regulation. The dynamics of the bioluminescence of dark and dim variants did not differ from the dynamics of the initial luminescent variant, but dependence of the level of luminescence intensity on the exogenous autoinductor of the lux operon was revealed. The investigated variants of P. leiognathi 54 inherited fairly stable morphological characteristics, colony architectonics, level of luminescence, and activity of some enzymes; variants with reduced bioluminescence formed colonies of the S type. Stable bright variants with S- and R-type colonies appeared both in the initial strain population and in the dark variant population, but with smaller frequency. Populations of the bright variant with R-type colonies were most heterogeneous; this can be determined by the lack of glucose repression of the bioluminescence in contrast to other investigated variants of P. leiognathi.  相似文献

Direct calorimetric determinations of the rate of heat production along with simultaneous determinations of the rate of photon emission and the number of viable cells have provided insight into the growth of Beneckea harveyi and Photobacterium leiognathi. These experiments were performed with a Tronac isothermal microcalorimeter modified with a fiber optic light guide to allow in situ detection of light. Escherichia coli and a dark variant of P. leiognathi were also examined to provide points of reference. It is demonstrated that B. harveyi seems to pause in the rate of metabolic heat production at the same point in time that the enzyme luciferase begins to be synthesized. This effect is not removed if B. harveyi is grown in conditioned medium. The thermograms for all species are correlated with cell generation time. The heat production per cell indicates that uncrowded cultures produce more heat than older, more crowded cultures, supporting the original observation of Bayne-Jones and Rhees (1929). These observations reopen for examination the suggestion that living systems tend toward a state of minimum metabolism per unit mass.  相似文献

The ability of marine luminescent bacteria to synthesize polyesters of hydroxycarboxylic acids (polyhydroxyalkanoates, PHA) as reserve macromolecules was studied. Twenty strains from the collection of the luminescent bacteria CCIBSO (WDCM839) of the Institute of Biophysics, Siberian Branch, Russian Academy of Sciences, assigned to different taxa (Photobacterium leiognathi, Ph. phosphoreum, Vibrio harveyi, and V. fischeri) were analyzed. The most productive strains were identified, and the conditions ensuring high polymer yields in batch culture (40–70% of the cell dry mass weight) were determined. The capacity for synthesizing two-and three-component polymers containing hydroxybutyric acid as the main monomer and hydroxyvaleric and hydroxyhexanoic acids was revealed in Ph. leiognathi and V. harveyi strains. The results allow luminescent microorganisms to be regarded as new producers of multicomponent polyhydroxyalkanoates.  相似文献

The purpose of the present paper was to study the influence of bacteria harbouring the luciferase‐encoding Vibrio harveyi luxAB genes upon the spectral emission during growth in batch‐culture conditions. In vivo bioluminescence spectra were compared from several bioluminescent strains, either naturally luminescent (Vibrio fischeri and Vibrio harveyi) or in recombinant strains (two Gram‐negative Escherichia coli::luxAB strains and a Gram‐positive Bacillus subtilis::luxAB strain). Spectral emission was recorded from 400 nm to 750 nm using a highly sensitive spectrometer initially devoted to Raman scattering. Two peaks were clearly identified, one at 491–500 nm (± 5 nm) and a second peak at 585–595 (± 5 nm) with the Raman CCD. The former peak was the only one detected with traditional spectrometers with a photomultiplier detector commonly used for spectral emission measurement, due to their lack of sensitivity and low resolution in the 550–650 nm window. When spectra were compared between all the studied bacteria, no difference was observed between natural or recombinant cells, between Gram‐positive and Gram‐negative strains, and growth conditions and growth medium were not found to modify the spectrum of light emission. Photobacteria were used as test objects for rapid monitoring of ecotoxicants. Specific inhibitory effects of Hg2+ on bioluminescence and cell growth as well as the lux gene expression of Photobacterium leiognathi were studied. The 3‐(4,5‐Dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide reduction assay was used to evaluate cellular proliferation and mortality. The luminescent inhibition effect on P. leiognathi cells was found to increase as cellular mortality increased; y = 0.744x ‐ 4.8916, R² = 0.9794. However, this trend was not observed in cell growth processes. Quantitation of lux mRNAs by semi‐quantitative RT‐qPCR indicated that increases and decreases in luciferase mRNA integral level coincided with changes in luminescence intensity (R2 = 0.93). Addition of Hg2+ changed luminescence but without concomitant changes in extractable luciferase activity. Nevertheless, the presence of Hg2+ changed lux gene expression. Summary The incidence of H2-oxidizing chemolithotrophic bacteria associated with rice grown under continuous wetland, upland, and rainfed wetland conditions was studied by14C-autoradiographic technique in a neutral soil at IRRI (Maahas) and an acid rainfed wetland soil (Luisiana).In Maahas soil, H2-oxidizing chemolithotrophic bacteria were not detected in the endorhizosphere, rhizosphere, and nonrhizosphere soil of rice grown under dryland conditions. Under continuously flooded conditions a very large population of these bacteria were found in the endorhizosphere but not in the oxidized and reduced soil.A very low population of these bacteria were found in the endorhizosphere and basal culm of rice grown under rainfed wetland conditions at Luisiana. Bacteria isolated from Maahas wetland rice and inoculated to rice seedling planted in Luisiana soil failed to establish.Both Maahas and Luisiana soils consumed externally supplied H2 and produced H2 and CH4 almost at the same rate when they were amended with rice straw or sucrose. This paper discusses possible causes of variation in the number of these bacteria and their distribution in rice grown under different cultural and soil conditions.  相似文献

A carbon mass balance methodology employing minimal measurements was applied to heterotrophic and photoheterotrophic marine bacteria grown under constant dilution and exposed to 12-h intervals of light or darkness. Carbon mass balance calculations using measurements taken every 3 h closed to within 93–103% using dissolved organic carbon, biomass carbon and CO2 production data only, indicating that background interference from dissolved inorganic carbon variations in the amended seawater medium was not significant. Neither strain was observed to sustain a net CO2 fixation using paramagnetic measurement of oxygen uptake rates (OUR), indicating a need for more sensitive on-line measurement techniques for OUR. Photoheterotrophic growth demonstrated lower carbon-mole biomass yields (0.41±0.026 vs. 0.64±0.013 mol mol–1) despite higher specific glucose uptake rates (0.025 vs. 0.02 mol mol–1 h–1), suggesting that bioreactor-based study of marine bacteria can present growth modes that are different from those encountered in the marine environment.  相似文献

【目的】对菌株L1和XH1的混合发酵条件进行优化,为混合菌发酵生物破乳剂的实际生产和应用提供理论依据。【方法】利用响应面实验(RSM)的中心组合旋转设计方法(CCRD)针对混合菌的发酵条件进行优化,通过对模型乳状液进行破乳实验,以排油率作为发酵液破乳效能的评价标准。【结果】经模型的分析与验证,确定最佳发酵条件为:种子液比例(L1:XH1)为3:2,葡萄糖投加时间为第4天,投加葡萄糖后再培养21 h,液体石蜡含量3.6% (体积比)。【结论】与破乳菌XH1和L1单独培养相比,经混合培养后获得复合生物破乳剂具有投加量少、破乳接触时间短的优势。同时双株破乳菌复配培养有效地提高了培养基中主要营养物质的利用率,减少了对底物的浪费。  相似文献

研究了在可控生态条件下。一株分离自厦门西海域沉积物的海洋细菌S10在不同pH和盐度条件下对赤潮原因种塔玛亚历山大藻(Alexandrium tamarense)生长和产毒的影响.结果表明。实验用藻株适宜生长pH为6~8,适宜盐度为20~34;该藻株在不同pH及不同盐度条件下,藻细胞毒力差异显著,且随着pH升高而下降。随着盐度增加而加大,到盐度为30时达到最高值。然后逐渐下降;菌株Sl0(1.02×10^l0cells·ml^-1)在pH7~9和盐度15~34下均能有效抑藻生长和产毒。且在pH7、盐度34时其抑藻生长作用最强;在pH7时抑藻产毒效果较好,且其抑藻产毒作用强度不随盐度变化而异.  相似文献

研究了ATP生物荧光检测法与国标法《GB4789.2-2010食品卫生微生物学检验菌落总数测定》检测鸡蛋壳表面细菌总数的相关性。采用ATP生物荧光检测法和国标法对40个混合样品表面细菌总数进行检测,以log CFU/个蛋壳为横坐标(x),以logRLU/个蛋壳为纵坐标(y),分别进行线性、对数、乘幂、指数拟合。结果表明,ATP荧光检测法与国标法检测结果 Pearson相关系数为0.912,线性模型y=0.7306x-1.0041(R2=0.8322)拟合度较高。该试验结果为ATP荧光检测法在鸡蛋壳表面细菌总数快速检测中应用的可行性提供了依据。  相似文献

The toxicities of solutions of 10 mercury compounds to luminescent bacteria were measured using the Microtox Toxicity Bioassay. The aim of this study was to assess the influence that the counter-ions have on the aquatic toxicity of mercury salts. The toxicities of these mercury compounds were very similar, except for mercurous tannate and mercuric salicylate. This can be attributed to differences in the ionization and speciation patterns of these compounds relative to the other compounds tested. In general, the toxicity of the solutions at pH 5 was not significantly different from the toxicity of these solutions at pH 6, but a clear reduction in toxicity was observed when the pH of the solution was adjusted to pH 9. Significant differences were found between the toxicity of Hg(I) and Hg(II) salts of the same anion at pH 9. When cysteine was added to a mercuric nitrate solution (at pH 6), a reduction in the toxicity was observed. This can be explained in terms of the strong binding of mercury to cysteine, thus reducing the concentration of mercury species available to cause an observable toxic effect to the bioluminescent bacteria.  相似文献

Simplified anaerobic media and a convenient method for the cultivation of Rhodospirillaceae, Chlorobiaceae, Chloroflexaceae and Chromatiaceae are described. The modified conditions assure almost complete anaerobiosis for media, growth and maintenance.Strains representing several species of Rhodospirillaceae, Chlorobiaceae and Chromatiaceae were successfully grown within relatively short times with full pigmentation, indicating that the new media and cultivation conditions were most suited for photoautotrophic growth.  相似文献

目的研究宁波市宁海县贝类养殖滩涂中异养细菌、反硝化细菌、氨化细菌和硫酸还原菌等几类细菌的动态分布及异养细菌的菌群组成。方法异养细菌的计数采用平板菌落计数法;反硝化细菌、氨化细菌和硫酸还原菌的计数采用MPN法;并参照Oliver提供的海洋细菌鉴定检索图、《海洋调查规范》和《一般细菌常用鉴定方法》提供的图式将分离到的细菌鉴定至属。结果滩涂沉积物环境中异养细菌的数量波动在8.00×103~7.60×104CFU/g(湿重),反硝化细菌的数量变动在5.00×102~5.00×104个/g(湿重),氨化细菌的数量变动在3.00×105~5.00×107个/g(湿重),硫酸还原菌的数量分布在9.00×104~9.00×106个/g(湿重),温度的变化对这几类细菌的数量分布没有直接的影响。所分离的细菌经过鉴定,可归于15个属与肠杆菌科的部分属,其中肠杆菌科(Enterobacteriaceae)的部分属、梭状芽胞杆菌属(Clostridium)、芽胞杆菌属(Bacillus)、棒状杆菌属(Corynebacterium)、发光杆菌属(Photobacterium)、假单胞菌属(Pseudomonas)等为优势菌属。沉积物中氨化细菌、反硝化细菌和硫酸还原菌的检出率均为100%,且含量很高。结论经过多年不间歇的养殖后,宁海贝类养殖滩涂已经不同程度地受到了污染,应尽快调整养殖容量并对养殖环境进行重新布局。  相似文献

海洋环境中存在着大量未被培养和利用的微生物资源。本研究对一份南海沉积物样品采用不同培养温度、盐度、pH、样品稀释倍数和营养浓度条件进行可培养细菌的多样性研究。经过16S rRNA基因序列分析,获得825株菌分属于5个门,8个纲,17个目,26个科,57个属。分离到最多的属级类群为芽胞杆菌属(Bacillus)。通过不同培养条件的分离实验发现,厚壁菌类群在4 °C~60 °C、0%~15%盐度、pH 5~8及不同的样品稀释倍数和营养浓度实验条件中均为优势培养类群,具有广泛的环境适应性,但2~200的样品稀释倍数可以大大减少分离培养基中厚壁菌的数量。放线菌类群在4 °C低温和0%NaCl添加条件下的可培养多样性较高,同时pH 6和寡营养培养基有助于分离获得稀有放线菌类群。另外,本研究发现新物种资源的获取几率分别在寡营养培养基、5%~10%较高盐度和60°C高温培养有所增加。分离获得的5个主要细菌门类分别为厚壁菌门(Firmicutes,86%)、放线菌门(Actinobacteria,13%)、变形菌门(Proteobacteria,1%)、拟杆菌门(Bacteroidetes)和异常球菌-栖热菌门(Deinococcus-Thermus)。本研究共分离得到29株潜在新种,分别属于芽胞杆菌纲(Bacilli)、放线菌纲(Actinobacteria)、酸微菌纲(Acidimicrobiia)、嗜热油菌纲(Thermoleophilia)、红色杆菌纲(Rubrobacteria)和异常球菌纲(Deinococci)。海洋环境微生物新类群、海洋放线菌稀有类群等微生物新资源的选择性分离培养提供了有效的方法和方案,为后期的深入开展打下良好的基础。  相似文献

以新疆、福建和浙江等地采集的132份土壤样品为对象,利用平板计数和Biolog等方法研究了不同耕作与栽培制度下瓜类土壤细菌数量及瓜类枯萎病拮抗细菌的分布与主要类群.结果表明,瓜类耕作土壤中的可培养细菌种群数量平均为非耕作土壤的16.2倍,根际为根围的2.0倍,轮作为连作的3.5倍,未嫁接平均为嫁接的1.5倍;水旱轮作和嫁接处理的瓜类土壤易获得拮抗细菌,其中荧光假单胞菌是主要类群,占总数的80%以上.  相似文献

The biosynthesis of proteinases with various substrate specificities was studied inBacillus firmus 44b andBacillus oligonitrophilus 21p as influenced by the growth conditions and growth phases of the bacteria. The period of the maximum synthesis of plasmin-like enzymes was observed 6 h later than the period of the maximum growth rate ofB. firmus 44b, and 3 h later than the maximum growth rate period ofB. oligonitrophilus 21 p. The periods of the maximum accumulation of activating enzymes were delayed 9 and 12 h, respectively, as compared to the periods of rapid growth of these two bacteria. Catabolite repression of proteinase synthesis and stimulation of the latter with substrate proteins were insignificant. The production of both plasmin-like and plasminogen-activating enzymes was most sensitive to repression by nitrogen deficiency. The production of plasminogen-activating proteinases was less dependent on the carbon source than the production of plasmin-like enzymes.  相似文献
