Abstract. In the Calliphora blastoderm, cytokinesis is preceded, during the final cleavage mitosis, by a radical surface remodelling which leads to the initiation of cytokinetic furrows. The egg is initially covered with oval surface bulges, each of which contains a mitotic nucleus. The shallow furrows between these bulges are then retracted and replaced by smooth membrane areas. Concomitantly, the remnants of the bulges become covered with large numbers of microprojections, and each bulge splits into two new bulges. The new bulges then increase in size, and cytokinetic furrows appear between them. At this point, the nuclei have also divided and reached interphase. During the first 60 min of cytokinesis, the plasma-membrane area of the egg is increased by the growth of surface microprojections; however, the furrows grow very slowly. During the final 30 min of cytokinesis, the surface becomes almost perfectly smooth, and the furrows grow very rapidly, As a result, cytokinesis is almost complete, and a columnar blastodermal epithelium is formed. Thus, surface microprojections play an essential role in cytokinesis. Plasma membrane utilized for furrow extension is apparently provided by the unfolding of these microprojections. In addition, filamentous microprojections may play an active part in the remodelling of the surface.  相似文献

Globodera rostochiensis population densities and potato root growth were measured in field plots of one susceptible and two resistant potato cultivars. Root growth and nematode densities were estimated from soil samples taken at three depths between plants within the rows, three depths 22.5 cm from the rows, and at two depths midway between rows (furrows). Four weeks after plant emergence (AE), nematode densities in the rows had declined 68% in plots of the susceptible cultivar and up to 75% in plots of both resistant cultivars. Significant decline in nematode densities in the furrows 4 weeks AE occurred only in plots of the susceptible cultivar. Total decline in nematode density in fallow soil was 50%, whereas in plots of the resistant cultivars, decline was more than 70% in the rows and more than 50% in the furrows. Nematode densities increased in the rows of the susceptible cultivar but declined in the furrows. We conclude that G. rostochiensis decline or increase is correlated with host resistance and the amount of roots present at any particular site.  相似文献

The peculiar secondary growth in Doxantha unguis-cati provides several developmental problems concerning cambial activity. One of the most interesting of these problems is the presence of both unidirectional and bidirectional arcs of cambium within the same stem. This investigation reports the ontogenetic development of these two kinds of cambial arcs. The first cambial divisions are observed in the fascicular regions of the 11th to 16th internodes from the shoot tip. This event is initiated after internode elongation is completed. In the initial stages, secondary tissues have a cylindrical configuration, but subsequently four grooves become apparent. These grooves signify the first evidence of unidirectional cambial activity. The four unidirectional arcs occur near the four major vascular strands to which all of the leaf traces connect. As secondary growth continues, the bidirectional and unidirectional arcs of cambium become separated and radial fissues can be seen between the furrows of phloem and the lobes of secondary xylem. Additional furrows originate either as sets of four between the original set of four or as single furrows to either or both sides of an existing furrow. All furrows are bordered by multiseriate rays. The initials of the bidirectional and unidirectional cambial arcs are non-stratified and are similar in size and appearance. The phloem produced within the furrow differs in several respects from that produced by the bidirectional arcs. The two types of cambial activity and the precise locations of the unidirectional cambial arcs in the stem (i.e. near the four major strands) suggests that transported products from the leaves are involved in the control of unidirectional cambial activity.  相似文献

北方旱作区降水稀少,水分不足是限制作物生长的主要因素,沟垄二元覆盖技术可进一步提高降水的利用效率和作物生产水平,在国内外已备受关注.为深刻理解中国北方旱作区沟垄二元覆盖技术模式及研究进展,本文就沟垄二元覆盖技术的概念、研究现状、配套农机具、技术模式及其土壤生态与作物效应进行了综述;在总结现有研究的基础上,归纳了典型旱作区沟垄二元覆盖技术模式的类型、作业操作及应用效果;科学评价不同沟垄二元覆盖技术下土壤与作物的生态效应;指出了目前沟垄二元覆盖技术研究中所存在的问题,并提出相应的建议.综合国内外沟垄集雨栽培技术的研究进展,提出沟垄二元覆盖模式及技术体系今后的重点研究方向:1)建立适合北方不同旱作类型区及不同作物的垄沟比;2)重视土壤水分与温度、肥力等其他因子耦合性研究;3)探索最佳环保型覆盖材料;4)加强沟垄二元覆盖模式及体系的技术评价、技术推广与配套机具设计等方面的研究.  相似文献

The floral ontogeny and anatomy ofKoelreuteria paniculata have been investigated to understand the developmental basis for the occurring monosymmetry and the origin of the septal cavities. Petals arise sequentially and one petal is missing between sepals 3 and 5, or rarely between sepals 2 and 5. The eight stamens arise sequentially before petal initiation is completed. The last formed petal and one stamen arise on a common primordium. Two stamen positions are empty (opposite the petal between the sepals 2 and 5, and the petal between sepal 1 and 3); consequently two antesepalous stamens have become displaced. The derivation of octandry from a diplostemonous ancestry, and reduction of the petal are discussed. The triangular gynoecium has a strong impact in obliquely reorganizing the symmetry of the flower, loss of organs, and shifts of stamens. The so-called septal slits occurring within the style are a deepreaching non-nectariferous extension of the stigma. Alternating locular furrows are present which could play a role as pollen transmitting tissue and in the loculicid dehiscence of the capsule.  相似文献

The postembryonic development of the turtle carapace was studied in the aquatic Еmys orbicularis and the terrestrial Тestudo graeca. Differences in the structure of the bony shell in aquatic and terrestrial turtles were shown to be associated with varying degrees of development of epidermal derivatives, namely, the thickness of the scutes and the depth of horny furrows. Sinking of the horny furrows into the dermis causes local changes in the structure of the collagen matrix, which might precondition the acceleration of the ossification. Aquatic turtles possess a relatively thin horny cover, whose derivatives are either weakly developed or altogether absent and thus make no noticeable impact on the growth dynamics of bony plates. Carapace plates of these turtles outgrow more or less evenly around the periphery, which results in uniform costals, relatively narrow and partly reduced neurals, and broad peripherals extending beyond the marginal scutes. In terrestrial turtles (Testudinidae), horny structures are much more developed and exert a considerable impact on the growth of bony elements. As a result, bony plates outgrow unevenly in the dermis, expanding fast in the zones under the horny furrows and slowly outside these zones. This determines the basic features of the testudinid carapace: alternately cuneate shape of costals, an alternation of broad octagonal and narrow tetragonal neurals, and the limitation of the growth of peripherals by pleuro-marginal furrows. The evolutionary significance of morphogenetic and constructional differences in the turtle carapace, and the association of these differences with the turtle habitats are discussed.  相似文献

The effects of nerve growth factor (NGF) on the intracellular content of acetylcholine (ACh) in cultured septal neurons from developing rats have been examined. The content of ACh could be measured by using HPLC and electrochemical detection (HPLC-ECD), coupled with an immobilized enzyme column. This method of determination is very simple and rapid, and is highly sensitive. The content of ACh and the activity of choline acetyltransferase (ChAT) in cultured postnatal day 1 (P1) septal neurons grown on an astroglial "feeder" layer was increased during the period of cultivation by the addition of NGF. The activities of ChAT and the content of ACh increased in a dose-dependent manner in direct relationship to the different amounts of NGF employed. These effects of NGF, i.e., elevating the intracellular content of ACh, accompanied by an increase in activity of ChAT, also were confirmed in the P1 septal organotypic cultures. Additionally, embryonic day 17 (E17) septal neurons in a serum-free medium displayed a similar responsiveness to NGF with respect to the elevation in the content of ACh and the increase in activity of ChAT. These results suggest that intracellular levels of ACh are likely to be regulated by NGF in a fashion similar to that of the activity levels of the biosynthetic enzyme.  相似文献

We have previously reported that injection of purified basal bodies or sperm into unfertilized eggs of Xenopus laevis induced the formation of asters and irregular cleavage furrows. Fully grown oocytes were found to be unable to form asters or cleavage furrows. In this paper we show that the oocyte acquires the ability to form asters upon basal body injection at the time of germinal vesicle breakdown during in vitro maturation. Our evidence indicates that aster formation requires progesterone-stimulated changes in the oocyte and mixing of cytoplasm and germinal vesicle plasm. The ability of the oocyte to form cleavage furrows arises six to eight hours after germinal vesicle breakdown. We infer that some maturational change in the cell cortex occurs to enable the egg surface to furrow. Experiments on the relationship of aster formation to furrow initiation indicates that asters stimulate furrow formation. However, some furrowing could be induced without aster formation in mature oocytes and unfertilized eggs by an activation stimulus, showing that asters are not essential for cleavage initiation. The significance of these observations are discussed in the light of our current understanding of meiotic maturation, cell cleavage and aster growth.  相似文献

Proper management of water and fertilizer placement in irrigated corn (Zea mays L.) has the potential to reduce nitrate leaching into the groundwater. Potential management practices tested in a two year field experiment included row or furrow fertilizer placement combined with every or alternate furrow irrigation. To understand how fertilizer availability to plants could be affected by these management practices, root growth and distribution in a Ulm clay loam soil were examined. Spring rains were greater than normal in both years providing adequate moisture for early root growth in both irrigated and non-irrigated furrows. As the non-irrigated furrow began to dry, root biomass increased as much as 126% compared with the irrigated furrow. The greatest increase was at lower depths, however, where moisture was still plentiful. When early season moisture was available, roots proliferated throughout the soil profile and quickly became available to take up fertilizer N in both irrigated and non-irrigated furrows. Root growth responded positively to fertilizer placement in the furrow in 1996 but not in 1995. Excessive N leaching in 1995 may have limited the response to fertilizer N.  相似文献

The dwarf globular cactus Aztekium ritteri shows a high degree of surface differentiation. The stem consists of 8–15 ribs, these in turn being sculptured by transverse furrows: compared with a sphere of equal size, the surface/volume ratio is increased by more than 10-fold. At the same time, only 20% of the stem surface is exposed to direct sunlight, 80% lying in the shade of its own body, in a system of furrows and crypts. The exposed areas have few stomata and the epidermal cell walls are notably thickened and cutinised. In addition, these form an exoskeleton by means of a hypodermis containing calcium oxalate crystals while the unexposed cell surface regions, hidden in the furrows, lack xeromorphic characters and calcium oxalate crystals. By far the greatest number of stomata are found in the furrows mostly in deep crypts. When dehydration occurs as a consequence of longer periods of drought, the furrows and crypts close together, thereby reducing transpiration losses. The complicated interlocking nature of the surface architecture of Aztekium is discussed in terms of the functional aspects as an adaptation to an extreme habitat (vertical shale cliffs in Nuevo León, Mexico).  相似文献

Calcium signaling is known to be associated with cytokinesis; however, the detailed spatio-temporal pattern of calcium dynamics has remained unclear. We have studied changes of intracellular free calcium in cleavage-stage Xenopus embryos using fluorescent calcium indicator dyes, mainly Calcium Green-1. Cleavage formation was followed by calcium transients that localized to cleavage furrows and propagated along the furrows as calcium waves. The calcium transients at the cleavage furrows were observed at each cleavage furrow at least until blastula stage. The velocity of the calcium waves at the first cleavage furrow was approximately 3 microns/s, which was much slower than that associated with fertilization/egg activation. These calcium waves traveled only along the cleavage furrows and not in the direction orthogonal to the furrows. These observations imply that there exists an intracellular calcium-releasing activity specifically associated with cleavage furrows. The calcium waves occurred in the absence of extracellular calcium and were inhibited in embryos injected with heparin an inositol 1,4,5-trisphosphate (InsP3) receptor antagonist. These results suggest that InsP3 receptor-mediated calcium mobilization plays an essential role in calcium wave formation at the cleavage furrows.  相似文献

The septal growth of Bacillus subtilis 168/s has been studied by making a number of observations from thin sections of cells from exponentially growing cultures. The process was initiated by the formation of a new cross wall under a preexisting layer of cylindrical wall. An annular notch appeared to cut through the overlying wall and presumably allowed the cross wall to split into two layers of peripheral wall. During this initial notching process, two raised bands of wall material were produced which resembled those previously observed in morphological studies of Streptococcus faecalis. Through an improved fixation technique, it was possible to preserve the bands seen in B. subtilis to the extent that they were used as markers to study the subsequent stages of septal growth. These stages included (i) the continued displacement of the two bands from the cross wall (as the two nascent polar surfaces enlarged and as the diameter of the cross wall decreased), (ii) the closure of the cross wall, and (iii) the final severance of the common cross wall connection between two completed poles. To study this process in a more quantitative manner, three-dimensional reconstructions of the envelope observed between pairs of the raised bands were made from axial thin sections of cells. The process of reconstruction was based on a technique by which x, y coordinates were taken from thin sections and were rotated around the cell's central axis. These reconstructions were used to estimate the surface area or volume of the reconstructed zones or their parts. A round of septal growth was then simulated by arranging 118 reconstructions in order of increasing surface area or volume. The topology of the process was studied by noting how various measurements of septal thickness, length, surface area, and volume varied as a function of increasing septal zone size. This analysis was based on several assumptions, of which three of the most important are: (i) the bands produced by the initial notching process are markers which separate septal from cylindrical wall growth; (ii) a septal zone observed between pairs of bands is made up of two nascent poles and a single cross wall; and (iii) as septal zones develop in terms of relative age they increase in size (volume or surface area) or amount of wall. The data suggested that the S. faecalis model of surface growth (in which polar growth occurs through a regulated constrictive separation and expansion of a cross wall) also seems applicable to the pattern of septal growth observed here for B. subtilis. This was indicated from measurements which showed that increases in the size of nascent polar surfaces were correlated with decreases in cross wall diameter. An explanation of these observations may be that decreases in cross wall diameter were due to a progressive splitting of the cross wall that removed surface from the outer circumference of the cross wall and converted it into new polar surface. Calculations further suggested that if the poles of B. subtilis were made by this model a sizeable and variable increase in surface area of the cross wall would also be required to convert these separating cross wall layers into two curved polar structures. Measurements of wall thickness taken from various locations within septal zones indicated that while the thickness of the polar wall of B. subtilis was constant over its surface, the width of the cross wall varied considerably during a round of synthesis. Again, one of the simplest explanations compatible with these observations and those previously made in S. faecalis is that the B. subtilis cross wall is brought to a constant thickness (possibly by remodeling or precursor addition) before or during separation. Although most observations made from the reconstruction of the septal zones of B. subtilis may fit the S. faecalis model of surface growth, differences in the pattern of septal growth were seen when the two organisms were compared. These have been discussed in terms of differences in the regulation of their respective septal growth sites and basic mechanisms of wall assembly and modification.  相似文献

The changes in cell morphology of Bacillus subtilis rodB during a temperature shift from 20 to 42 degrees C, in the absence of added anions, are described. At 20 degrees C the organisms grow as rods but gradually become spherical in shape when placed at 42 degrees C. The shape change is initiated by an increase in diameter at the cell equator, resulting in a bulged morphology, which is further modified to the morphology of a coccus. This change may involve a modification of the pattern of normal cylindrical extension such that incorporation of newly synthesized wall leads only to increase in diameter, perhaps from a growth zone of limited extent. The pattern of surface growth was followed by reconstructing the sequence of cross wall formation and pole construction in rods grown at 20 degrees C and in organisms incubated at 42 degrees C for 75 and 150 min. In thin section, wall forming the septum and nascent poles can be distinguished from the surface distal to the division site by the presence of raised tears, perhaps analogous to the wall bands of streptococci. By using an analog rotation technique involving the three-dimensional reconstruction of cells by mathematical rotation of axial thin sections about their longitudinal axis, it is shown that the proportion of septal wall increases during the shape change. In the coccal forms, all surface growth may arise from septal growth sites.  相似文献

Members of the genus Chorda represent the simplest form of sporophyte in the order Laminariales. The present study deals with reproduction in Chorda tomentosa, involving the initiation, growth, and structure of the sporangium and the process of zoosporogenesis. The simple tube-like sporophyte of Chorda tomentosa grows in diameter by means of repeated anticlinal divisions in a superficial meristematic layer known as the meristoderm. The onset of reproduction is marked by the conversion of the meristoderm from contributing cells to the vegetative plant body to producing 2 new cell types: paraphyses and sporangial mother cells. At the time of initiation, sporangial mother cells are crescent shaped and possess a densely staining cytoplasm. Sporangial mother cells increase in size, become ellipsoid, decrease in staining density, and undergo meiosis. After meiosis, sporangia increase in size while their nuclei undergo successive cycles of synchronous mitotic divisions. Sporangia increase to a maximum length of 120 μ;m at which time they possess the characteristic “cap” found in all members of the order studied thus far. At this stage the protoplast of the sporangium is organized into a peripheral layer of nucleus-chloroplast pairs and a central region of vacuoles. Cleavage furrows begin to form at the cell membrane and are met by furrows developing in the interior of the cytoplasm resulting in the division of the entire protoplast into separate units. Each unit is an individual zoospore. The biflagellate zoospores contain a nucleus, one chloroplast with eyespot, perinuclear Golgi, and several bodies of presumed storage carbohydrate. The occurrence of a small population of early developing sporangia is described. In essential details, the origin, development, and structure of sporangia in Chorda tomentosa are identical to all earlier observations in the Laminariales.  相似文献

Summary Microplasmodia ofPhysarum polycephalum Schw differentiate into numerous hard walled spherules when deprived of nutrients. Cleavage occurs by invagination of the plasmalemma and growth of furrows centripetally. Cleavage furrows consistently contain ruthenium red positive fibrillar material. As the spherules become more rounded, a compact layer of ruthenium red positive material surrounds their membranes. A spherule wall which does not bind ruthenium red is deposited inside of the stained layer. Clusters of spherules are enclosed by ruthenium red positive material similar to the microplasmodial slime.Partially supported by a grant from Research Corporation.  相似文献

Animal cells are cleaved by the formation and contraction of an extremely thin actomyosin band. In most cases this contractile band seems to form synchronously around the whole equator of the cleaving cell; however in giant cells it first forms near the mitotic apparatus and then slowly grows outwards over the cell. We studied the relationship of calcium to such contractile band growth using aequorin injected medaka fish eggs: we see two successive waves of faint luminescence moving along each of the first three cleavage furrows at approximately 0.5 micron/s. The first, narrower waves accompany furrow extension, while the second, broader ones, accompany the subsequent apposition or slow zipping together of the separating cells. If the first waves travel within the assembling contractile band, they would indicate local increases of free calcium to concentrations of about five to eight micromolar. This is the first report to visualize high free calcium within cleavage furrows. Moreover, this is also the first report to visualize slow (0.3-1.0 micron/s) as opposed to fast (10-100 microns/s) calcium waves. We suggest that these first waves are needed for furrow growth; that in part they further furrow growth by speeding actomyosin filament shortening, while such shortening in turn acts to mechanically release calcium and thus propagates these waves as well as furrow growth. We also suggest that the second waves act to induce the exocytosis which provides new furrow membrane.  相似文献

Citron kinase is a Rho-effector protein kinase that is related to Rho-associated kinases of ROCK/ROK/Rho-kinase family. Both ROCK and citron kinase are suggested to play a role in cytokinesis. However, no substrates are known for citron kinase. We found that citron kinase phosphorylated regulatory light chain (MLC) of myosin II at both Ser-19 and Thr-18 in vitro. Unlike ROCK, however, citron kinase did not phosphorylate the myosin binding subunit of myosin phosphatase, indicating that it does not inhibit myosin phosphatase. We found that the expression of the kinase domain of citron kinase resulted in an increase in MLC di-phosphorylation. Furthermore, the kinase domain was able to increase di-phosphorylation and restore stress fiber assembly even when ROCK was inhibited with a specific inhibitor, Y-27632. The expression of full-length citron kinase also increased di-phosphorylation during cytokinesis. These observations suggest that citron kinase phosphorylates MLC to generate di-phosphorylated MLC in vivo. Although both mono- and di-phosphorylated MLC were found in cleavage furrows, di-phosphorylated MLC showed more constrained localization than did mono-phosphorylated MLC. Because citron kinase is localized in cleavage furrows, citron kinase may be involved in regulating di-phosphorylation of MLC during cytokinesis.  相似文献

We have isolated a new female sterile mutant from Drosophila melanogaster, which arrests the embryonic development during the transition from syncytial to cellular blastoderm. Cytological analysis of the mutant embryos indicates that pseudocleavage furrows in the syncytial blastoderm are abnormal but not completely disrupted. However, cleavage furrows during cellularization are totally disorganized, and no embryos can develop beyond this stage. Consistent with this observation, the expression of this gene peaks around the cellular blastoderm and not in any later developmental stages. Based on immunofluorescence experiments, the protein product of this gene is localized in both pseudocleavage furrows at the syncytial blastoderm and in the cleavage furrows during the cellularization stage. Sequence homology analysis demonstrates a modest, but statistically significant, similarity of this protein with the carboxyl-terminal domains of dystrophin and a family of proteins collectively known as apodystrophins. It is possible that this protein may play an essential role in organizing and maintaining a specialized cytoskeletal structure, a function also suggested for dystrophin and apodystrophins.  相似文献

The 95F myosin, a class VI unconventional myosin, associates with particles in the cytoplasm of the Drosophila syncytial blastoderm and is required for the ATP- and F-actin-dependent translocation of these particles. The particles undergo a cell cycle-dependent redistribution from domains that surround each nucleus in interphase to transient membrane invaginations that provide a barrier between adjacent spindles during mitosis. When 95F myosin function is inhibited by antibody injection, profound defects in syncytial blastoderm organization occur. This disorganization is seen as aberrant nuclear morphology and position and is suggestive of failures in cytoskeletal function. Nuclear defects correlate with gross defects in the actin cytoskeleton, including indistinct actin caps and furrows, missing actin structures, abnormal spacing of caps, and abnormally spaced furrows. Three- dimensional examination of embryos injected with anti-95F myosin antibody reveals that actin furrows do not invaginate as deeply into the embryo as do normal furrows. These furrows do not separate adjacent mitoses, since microtubules cross over them. These inappropriate microtubule interactions lead to aberrant nuclear divisions and to the nuclear defects observed. We propose that 95F myosin function is required to generate normal actin-based transient membrane furrows. The motor activity of 95F myosin itself and/or components within the particles transported to the furrows by 95F myosin may be required for normal furrows to form.  相似文献
