This paper describes a method for quantitating relative potencies of mixed lymphocyte interactions (MLI) in the rat by parallel line analysis of responder cell titrations. The MLI titration assay is based upon, and has certain advantages over, the popliteal lymph node graft-versus-host assay for investigations of allogeneic interactions determined by the major histocompatibility complex. Log-log plots of dose of responder cells versus cpm of [3H]thymidine incorporated were linear in the range 1 × 104 to 2 × 105 responder cells per microtiter well. Dilution experiments indicated that the overriding influence on the potency of any allogeneic interaction was the initial frequency of reactive cells. We have defined the limits within which the MLI titration assay behaved ideally and suggested explanations for the nonideal behavior seen when reactions of widely differing potency were compared.  相似文献

Short-term cultures of human tonsilar lymphocytes (HTL), 5 × 106 cells/culture, in medium RPMI 1640 supplemented with human group AB serum were studied for the production of plaque-forming cells (PFC) against sheep (SRBC) and bovine (BRBC) red blood cells following in vitro stimulation by various allogeneic lymphoid cells. Of 55 HTL specimens examined, 48 produced a significant number (50–300/culture) of PFC against SRBC and/or BRBC following the in vitro stimulation. The optimal doses of the stimulator HTL and peripheral blood lymphocytes (PBL) were 107 and 5 × 106/culture, respectively. After the stimulation, PFC appeared in significant numbers on the third day, reached the peak number on the sixth day, and decreased sharply in number thereafter. Removal of E-rosetting cells from both stimulator and responder populations abolished the PFC formation. PFC formation against SRBC was inhibited by solubilized Forssman antigen, while PFC formation against BRBC was inhibited strongly by Hanganutziu-Deicher antigen, hardly by Paul-Bunnell antigen and not at all by Forssman antigen. Supernatants of mixed lymphocyte culture of PBL were shown to enhance PFC formation of HTL cultures stimulated by allogeneic lymphocytes. The results of this study indicated that in vivo primed B cells of the HTL were triggered in vitro by allogeneic stimulation for the heterophile antibody formation. Since these antibodies are apparently directed against Forssman and Hanganutziu-Deicher antigens, the “allo” nature of these antigens as well as their relationship to the previously described heterophile transplantation antigens have to be clarified.  相似文献

Moloney leukemia virus-specific cytotoxic T lymphocytes (CTL), generated by secondary in vitro stimulation of spleen cells with syngeneic virus-infected cells, frequently lysed not only syngeneic virus-infected cells, but also noninfected allogeneic target cells. This phenomenon was studied with B6(H-2 b ) responder cells and a series of H-2K b -mutant responder cells. Thus, B6 Moloney-specific CTL lysed noninfected K b -mutant cells, but not B6 cells, whereas K b -mutant Moloney-specific CTL lysed noninfected B6 cells and not noninfected cells of the same mutant. Cold-target-inhibition studies showed that the CTL reactions against different allogeneic cells were mediated by different subpopulations of virus-specific CTL: lysis of allogeneic target cells was fully inhibited only by the same allogeneic and by syngeneic virus-infected cells, but not by another allogeneic cell, also lysed by the same effector-cell population. Lysis of syngeneic virus-infected cells could not be inhibited by allogeneic target cells. These data imply that a minority of virus-specific CTL shows cross-reactivity with a given allogeneic target cell. It is concluded that limited amino acid substitutions in the Kb molecule alter the repertoire of Moloney virus-specific CTL, as reflected in alloreactive CTL populations, even though the virus-specific CTL response. of B6 and all K b mutants is mainly Db-restricted. Thus, the development of tolerance to self class-I major histocompatibility complex (MHC) molecules affects the repertoire of self-restricted cytotoxic T cells.  相似文献

When spleen cells primed in vivo against allogeneic lymphoid cells were used as responder cells in secondary mixed lymphocyte cultures, a high degree of cytotoxicity was generated even in the absence of splenic adherent cells. However, removal of adherent cells from such primed responder spleen cells reduced the cytotoxicity to some extent. On the other hand, when these responder cells were transferred into the peritoneal cavity of irradiated syngeneic mice together with antigenic cells, unseparated responder cells generated a lower degree of cytotoxicity than did adherent cell-depleted responder cells. In an in vitro system, peritoneal adherent cells also suppressed the generation of cytotoxic T lymphocytes by unseparated responders; however they augmented the cytotoxic T lymphocyte generation by adherent cell-depleted responders. These adherent cell populations with augmenting activity became inhibitory when they coexisted. The mechanism of this inhibitory action remains unclear.  相似文献

In this study, we investigated the role of DR antigens in human mixed lymphocyte reactions (MLR) at the responder cell level. Upon stimulation by allogeneic lymphocytes or leukemic cell lines, a large proportion of T cells underwent blastogenesis and began expressing DR antigens. Analysis by a fluorescence-activated cell sorter revealed that both subpopulations of large activated T cell blasts and of small T lymphocytes became DR+ by synthesis and/or uptake. Depletion of DR+ responder cells from 6-day-old MLRs by treatment with anti-DR monoclonal antibodies (mAbs) plus complement (C) reduced but did not completely abrogate the natural killer (NK)-like activity of the responder lymphocytes, suggesting that the MLR-induced cytotoxic cells include both DR+ and Dr- populations. The expression of NK-like activity by the responder cells was also greatly reduced upon addition of anti-DR mAbs (without C) at the start of the mixed cultures. This effect was observed regardless of the presence of DR antigens on the stimulator cells, indicating that the anti-DR mAbs can interact with the antigens present on both the stimulator and responder populations. These data show that during an MLR, the continued presence of DR antigens on the responding population is essential for the expression and maintenance of the proliferative and cytotoxic capabilities of these cells.  相似文献

The capacity of spleen cell populations enriched for T and B lymphocytes by a physical adherence column method to respond in vitro to phytomitogens and allogeneic lymphocytes was determined. Column filtrate cells (T lymphocytes) responded well to phytohaemagglutinin- and mitomycin-C-treated allogeneic spleen cells, but poorly to pokeweed mitogen. Adherent cell populations from the column (B and some T lymphocytes) responded well to pokeweed mitogen, but poorly to phytohaemagglutinin- and mitomycin-C-treated allogeneic cells.Purified peripheral T lymphocytes prepared from normal mouse spleen by the column method reconstituted the depleted in vitro antibody response to the thymic-dependent SRBC antigen of all B lymphocyte sources tested, namely, spleen cells from congenitally athymic mice, neonatally thymectomized mice, and adult thymectomized mice which had been reconstituted with bone marrow, and a lymphocyte population prepared by incubating spleen cells with anti-θ serum and complement. When transferred with sheep erythrocytes to congenitally athymic mice, purified peripheral T cells restored the in vivo IgM and IgG responses of these animals. These results confirm that the column filtrate is a thymus derived subpopulation of cells capable of cell-mediated immunity and cooperation with B lymphocytes in humoral immunity both in vitro and in vivo.  相似文献

The effect of cell-free liver extract (LE) on the proliferation of spleen cells in vitro was examined using [3H]thymidine incorporation. LE inhibited the blastogenic response of murine lymphocytes stimulated with plant mitogens, phytohemagglutinin, and concanavalin A and in the mixed lymphocyte reaction (MLR). Suppression of cell proliferation occurred whether the LE was syngeneic or allogeneic to the responding cells. This effect was observed only when LE was present in cultures, as preincubation of cells with LE did not impair their capacity to respond to stimulation. Profound suppression of proliferation was achieved with the addition of LE to the culture up to 48 hr after the onset of stimulation. However, the inhibitory effect was readily reversible upon removal of LE from the culture. Furthermore, although LE was capable of suppressing the generation of cytotoxic lymphocytes, LE did not interfere with their capacity for cytolysis. These findings indicate the presence of a potent inhibitor of lymphocyte proliferation in a cell-free extract of murine liver.  相似文献

THE mixed skin cell-leukocyte reaction (MSLR) is an in vitro reaction between dissociated skin cells and allogeneic peripheral blood leukocytes. It is similar in its immunological implications to the reaction between allogeneic leukocytes, commonly known as the mixed leukocyte reaction (MLR) or the mixed lymphocyte interaction (MLI). In the latter, immunological recognition between lymphocytes from genetically disparate donors can be detected and measured either by blastoid transformation, enhanced mitotic rate or induction of DNA synthesis1–4. This system has been useful in the study of the transfer of transplantation immunity in vivo by lymphocytes sensitized in vitro5 and also in the identification of tissue incompatibility between donor and recipient6, 7. In studies with inbred strains of rats the proliferative phase of the MLR has been shown to possess characteristics required for an immunological response; that is, selectivity and specificity8, 9. Furthermore, mouse peripheral lymphocytes sensitized in mixed culture with allogeneic lymphocytes have been shown to engender immunospecific destruction of target cells10, an event which is directly associated with allograft rejection.  相似文献

Lewis rats were rendered hematopoietic and lymphoid cell chimeras by injection of (LBN)F1 hybrid cells at birth or following treatment with cyclophosphamide in adult life. The establishment of transplantation tolerance was indicated by acceptance of (LBN)F1 skin grafts and specific unresponsiveness in graft vs. host reaction (GvHR) and mixed lymphocyte interaction (MLI) in vitro. Tolerance was abolished by adoptively transferred Lewis lymphocytes, and the loss of chimerism and recovery of specific reactivity by blood lymphocytes were monitored independently by mixed lymphocyte cultures. Recovery of competence to initiate GvHR by splenic and lymph node cells was monitored by the local renal graft vs. host technique. Both techniques measure essentially the proliferative response of certain lymphocytes to foreign cellular AgB antigens, and both detected a prolonged, but gradually weakening, state of partial tolerance to the AgB factors to which tolerance had originally been induced. During this phase of partial tolerance the former chimera rejects skin and lymph node cell grafts from (LBN)F1 donors with alacrity, but in some cases accepts (LBN)F1 kidney grafts. Cytotoxic antibodies appear in the serum soon after allogeneic chimerism is terminated. These results are interpreted to indicate that a state of partial tolerance exists among the cells which proliferate in response to certain AgB antigens in GvHR and MLI in the formerly tolerant chimera, and that a state of transplantation immunity (possibly to other determinants) coexists with this partial tolerance.  相似文献

To determine the potential role of macrophages as regulators of the immune response, the effect of mouse peritoneal macrophages on transforming mouse spleen lymphocytes was investigated. Mitogen and antigen stimulated lymphocyte transformation, as measured by DNA synthesis, was enhanced by all concentrations of normal macrophages tested, but only by low concentrations of activated macrophages. High concentrations of activated macrophages markedly inhibited lymphocyte transformation. This inhibition occurred whether lymphocyte DNA synthesis was measured by incorporation of [3H]TdR or of 32P. Activated macrophages cultured with lymphocytes within 4 hr of being removed from the peritoneal cavity inhibited lymphocyte transformation. When activated macrophages were cultured alone for 24 or more hours before addition of lymphocytes, enhancement of transformation was noted. Once lymphocytes were exposed to activated macrophages, they could not be induced to undergo transformation in the presence of Con A. Whereas heat-killed activated macrophages, which appeared intact morphologically, lost their capacity to inhibit lymphocyte transformation, macrophages treated with mitomycin C to inhibit DNA synthesis retained this capacity. Syngeneic and allogeneic macrophages had similar inhibitory ability. Supernatants from cultures of many cell types (including normal or activated macrophages, lymphocytes, lymphocytes plus macrophages, and L cells) inhibited [3H]TdR incorporation by both mitogen stimulated lymphocytes and tumor cells. These studies demonstrate the capacity of macrophages to regulate lymphocyte transformation in vitro and suggest a role for these cells as regulators of cell-mediated immunity in vivo.  相似文献

Strong, adult (Type II) autologous mixed lymphocyte reactions (AMLR) were observed in cultures of lymphoid cells from both A.TH and A.TL mice. These were suppressed by more than 90% in the continuous presence of 7.5 × 10?8M hydrocortisone-21-sodium succinate. This concentration of hormone had minimal effects on the allogeneic mixed lymphocyte response (MLR) and the mitogenic response to concanavalin A (Con A). Higher concentrations suppressed all three responses. Treatment of autologous cell mixtures for the first 30 hr with 7.5 × 10?8M hydrocortisone resulted in a 78% suppression of the AMLR. This was not associated with a detectable decrease in the quantity of Ia antigens on the stimulator-cell surface, as evaluated by the susceptibility of treated cells to antibody dependent, complement-mediated lysis, using [A.TH × B.10M]F1 anti-A.TL antiserum. Hence, this suppression did not appear to result from an alteration of the antigens putatively associated with stimulation of the AMLR. Separate pretreatment of stimulator and responder cells with 7.5 × 10?8M hydrocortisone followed by culturing with appropriate companion cells had no major effect on the AMLR. Therefore, low-dose hydrocortisone did not appear to selectively eliminate or permanently inactivate subpopulations of responder or stimulator cells. Rather, it appeared to regulate active cellular processes that are initiated by the coculturing of these cells and are required for the early stages of autologous lymphocyte activation.  相似文献

Coculture of normal human T lymphocytes with autologous cells enriched in B lymphocytes results in increased DNA synthesis. In our system, the responder cells are mononuclear cells stimulated by irradiated autologous non-T cells in the presence of 20% inactivated autologous plasma. The results indicate that the proliferative response is age and sex related; significantly higher levels of thymidine incorporation are observed with female peripheral blood lymphocytes and, moreover, it appears that this strong stimulation is more pronounced in older female subjects. In contrast, the proliferative responses against allogeneic cells are not modified irrespective of the age and sex group considered. The involvement of autorosette-forming cells (A-RFC) in such autologous mixed lymphocyte reaction (A-MLR) is demonstrated by the marked impairment of the A-MLR after removal of A-RFC from responding cells. These data suggest that older healthy subjects, particularly females, exhibit a higher incidence of in vitro autologous reactions.  相似文献

We have studied the induction of cytotoxic activity in human peripheral blood lymphocytes by heated allogeneic cells. By separating T and B cells from the responder and stimulator cell populations we found that cytotoxic cells are generated in responder T cell populations by both T and the B stimulator cells. Rabbit antisera to a membrane glycoprotein complex (33,000 and 27,000 m.w. by SDS-gel electrophoresis) isolated from a human B cell line were utilized to explore further the nature of the effector cells in this type of cytotoxicity. This antiserum, present during the 6-day-culture period, blocked generation of cytotoxic effector cells. Depletion of cells bearing the B cell antigen from the responder cell population by anti-B cell serum and complement (C) eliminated cytotoxicity. Furthermore, heated cell-induced cytotoxicity was blocked by simply pretreating the responder or the stimulator cell populations with anti-B cell serum in the absence of C. Apparently the human lymphocyte that functions as the effector cell in heated cell-induced cytotoxicity bears the Ia-like antigen that might be important in triggering this type of cytotoxicity.  相似文献

Fc receptor (FcR)-carrying blast cells were separated from nonFcR blast cells after priming in primary mixed lymphocyte culture (MLC) by erythrocyte-antibody rosetting and by 1-g velocity sedimentation. Both types of blast cells were cytotoxic to relevant allogeneic target cells in vitro. The FcR-positive and FcR-negative blasts were then plated on a feeder layer syngeneic to the primary MLC responder cells. After feeder layer culture both types of cells reverted into secondary small lymphocytes. When restimulated with the original stimulator cells, both types of secondary lymphocytes produced the relevant secondary cell-mediated lysis responses. Thus no functionally dissimilar subclasses of secondary T lymphocytes can be distinguished in the MLC-stimulated T-cell population on the basis of the FcR.  相似文献

Spleen cells from DBA/2 mice bearing the DBA/2 P815X mastocytoma for approximately 2 weeks can be stimulated in vitro by mastocytoma cells to generate cytotoxicity measured as 51Cr release from mastocytoma cells in a 4-hr assay. These cytotoxic cells will not kill allogeneic cell lines but will kill a series of first transplant generation syngeneic tumors. T cells are involved in that treatment of the responding or the cytotoxic cell populations with either anti-T or anti-theta antibody + complement will abrogate all cytotoxicity. Anti-Ly 2.1 antibody + complement treatment of either responder cells (prior to the in vitro culture with irradiated tumor cells) or effector cells after culture markedly decreases cytotoxicity whereas treatment with anti-Ly 1.1 was more effective prior to culture compared to its effect on cytotoxic cells per se. These T cells are in the small lymphocyte class and occur either singly or in aggregates. Suppression of antisyngeneic tumor cytotoxicity by antibody inhibits preferentially the expression of cytotoxicity in the aggregate fractions.  相似文献

Peripheral blood mononuclear cells from 48 healthy subjects of ages varying from 20 to 94 years were evaluated for the ability to generate suppressor cell activity following in vitro incubation with concanavalin A. The suppression of proliferative responses by autologous and young, allogeneic lymphocytes to phytohemagglutinin was assessed using suppressor/ responder cell ratios (SR) of 2:1 and 1:1 and by using a summation index. Inducible suppressor cell activity for autologous responder cells was comparable between 24 aged (76.0 ± 10.9 years) and 20 young (26.8 ± 4.6 years) subjects. However, aged subjects exhibited a significant decrease in suppressor cell activity (SR = 1) when allogeneic responder cells were utilized. Our results indicate that autologous inducible suppressor cell activity is preserved in the aged population, whereas allogeneic activity is impaired.  相似文献

In the preceding paper it has been shown that human or mouse lymphocytes stimulated by a variety of agents, damaged allogeneic target cells while damage of xenogeneic target cells was weak or absent. In this study, the species specificity of the cytotoxicity of PHA activated lymphocytes has been studied in greater detail. Effector cells were purified lymphocytes either from human peripheral blood, or from spleen or lymph nodes of inbred mice. Target cells were 51Cr-labeled human Chang liver cells or mouse L cells.PHA stimulated human or mouse lymphocytes were significantly more cytotoxic to allogeneic than to xenogeneic target cells. At low PHA doses at which damage of allogeneic target cells was significant, damage of xenogeneic target cells was very weak or absent. At higher PHA doses, damage of xenogeneic target cells became also significant but always remained at a lower level than that of allogeneic target cells.Prestimulation of human lymphocytes with PHA for 3 days increased their cytotoxic efficiency. Furthermore, damage of human Chang cells by human lymphocytes had a dose-response relationship similar to that valid for stimulation of DNA synthesis. However, damage of mouse L cells by human lymphocytes increased at PHA-doses at which stimulation of DNA-synthesis declined. For mouse lymphocytes, these doseresponse relationships were less clear-cut, probably due to differences in origin and survival of the effector cells. This confirms previous observations that cytotoxicity and DNA-synthesis are different but probably interdependent expressions of lymphocyte activation.  相似文献

The age-dependent alteration in the proliferative response of C57B1/6J lymph node cells to stimulation by H-2- and M-locus alloantigens was examined in one-way mixed lymphocyte cultures (MLC). Balb/c (H-2d, Mlsb) and DBA (H-2d, Mlsa) spleen cells served as stimulating cells differing from C57B1/6J (H-2b, Mlsb) at the H-2 and H-2 plus Mls loci, respectively. The day of peak response and the ratio of responder to stimulator cells required for optimal stimulation were the same for all the age groups (3 to 29 months) tested, irrespective of the stimulator strain used. Results obtained in MLC under optimal conditions showed a maximal response to both Balb/c and DBA/2 stimulation at the age of 6 months, followed by a gradual decline in the response with age. In order to determine whether the decline with age in mixed lymphocyte reactivity can be attributed to a reduction in the proliferative capacity of the responding lymphocytes of aged mice, cell cycle analyses were performed. Auto-radiographic studies of MLC containing lymphocytes from CS7B1/6J mice aged 6 and 24 months showed no difference in generation time, S, G2, G1, and M phases of the cell cycle. In addition, lymphocytes of both age groups underwent two identical mitotic waves within the period of examination. Our results determine that the functional decline with age in proliferative activity in mixed lymphocyte cultures is attributable to a neither decrease in sensitivity to alloantigen nor to a decrease in generation time or the ability to undergo several mitotic divisions, and suggest that such a decline is caused by fewer cells capable of response in old mice.  相似文献

Incubation of mouse thymic lymphocytes with irradiated allogeneic spleen cells gave rise to suppressor cells. The suppressor activity was assayed by adding the incubated cell mixture to a mixed lymphocyte culture (MLC) in which the responder cells were syngeneic with the sensitized thymocytes and the stimulator cells were syngeneic with the sensitizing spleen cells. Such addition suppressed significantly thymidine incorporation in the mixed lymphocyte reaction (MLR). The suppressor cells were found to carry the θ antigen and to function allospecifically, as shown by cross-testing in three allogeneic combinations. Our data suggest that these cells may originate from immature cortisone-sensitive thymic lymphocytes and also provide some preliminary information concerning their mode of action.  相似文献

In unidirectional mixed lymphocyte cultures containing (as responders, stimulators, or regulators) spleen cells from mice infected with Trypanosoma cruzi, alloantigen responses were less than in cultures containing normal spleen cells only. Depletion of plastic adherent cells from infected spleen cells (stimulators or regulators) reversed their inhibitory effect on normal spleen cells (responders); removal of adherent responder cells and/or B lymphocytes did not alter the low alloantigen responses of normal spleen cells (stimulated by infected spleen cells) or infected spleen cells (stimulated by normal spleen cells). Infected spleen cells were effective in regulating mixed lymphocyte cultures only when added at the initiation of the culture. Serum from infected mice suppressed mixed lymphocyte cultures containing responder spleen cells syngeneic to the serum donor if added up to 24 hr after initiation of cultures, whereas the “suppressor serum” had to be present at the initiation of cultures when responder cells were allogeneic to the serum donor. Cultures of infected spleen cells (whole or macrophage enriched) produced a factor which was suppressive when added to mixed lymphocyte cultures containing syngeneic responder cells at initiation. It is proposed that the serum suppressor substance regulates cell-mediated immune responses directly by suppressing the response-potential of cells and indirectly by triggering the release of a factor from adherent splenic cells which induces a hyporesponsive state in T lymphocytes.  相似文献
