The flight take-off frequency of adult Colorado potato beetles, Leptinotarsa decemlineata (Say), from potato plants, Solanum tuberosum L. 'Red Pontiac' at the bloom stage of development was 2.2-2.5-fold that of Colorado potato beetle from plants at the vegetative stage. Tests were conducted in a flight chamber over a period of 3 h. Prefeeding Colorado potato beetles for 48 h on potato plants at the bloom or at the vegetative stage before placing them into the flight chamber resulted in the same significantly higher flight take-off frequency from potato plants at the bloom stage than from plants at the vegetative stage. These results demonstrate that the factor in potato plants in bloom that stimulates the flight take-off of the Colorado potato beetle is independent of the feeding history of the beetles and begins acting only when the beetles are in the presence of the plant. According to these results, the dispersal of adult Colorado potato beetles from potato fields in bloom to younger potato fields with plants at the vegetative stage, previously reported in the literature, is at least partly explained by the effect of plant phenology on the frequency of flight take-off. Results confirm the value of planting potato fields of similar phenology over as wide an area as possible to reduce Colorado potato beetle dispersal between fields. Results also imply that staggering the planting dates of conventional potato refuge areas near Colorado potato beetle transgenic or conventionally resistant potato fields is a sound management practice, because it promotes the movement of wild beetles over to the adjacent younger resistant crops.  相似文献

Survival and fecundity of Colorado potato beetle adults, Leptinotarsa decemlineata (Say), that had or had not fed previously on non-transgenic potato before exposure to transgenic potato containing the Bacillus thuringiensis subsp. tenebrionis Cry3A toxin (Bt) was investigated. In the laboratory, < 5% of first-generation adults survived after two weeks when restricted to Bt foliage since eclosion, but over 85% of adults that had fed initially on non-Bt potato survived exposure to Bt potato for two weeks. In field experiments, less than 0.5% of adults that were exclusively provided Bt potato plants survived overwinter, whereas 44% to 57% survived overwinter when fed non-Bt potato plants for two weeks before being provided Bt potato as a final pre-overwintering host. Survival through the winter increased as the duration of initial feeding on non-Bt potato increased and was similar for beetles provided either tubers or Bt potato plants as a final pre-overwintering host. Only overwintered beetles that fed initially on non-Bt potato before encountering either tubers or Bt potato as a final pre-overwintering host laid eggs the following spring. Survival and reproduction of potato beetle adults after colonizing Bt potato fields should not be adversely affected as long as they have had sufficient time to feed initially on non-Bt potato. Implications for how potato production practices in the Mid-Atlantic US may affect the utility of general resistance management plans for Bt potato are discussed.  相似文献

A field experiment in which main‐crop potatoes were grown every other year was conducted on a sandy soil from 1994 to 1999. The aim of the experiment was to control soil‐borne pathogens of potato with ecologically sound methods. Potato grown as a trap crop from the end of April to the end of June (8 wk) was used to control potato cyst nematodes (PCN) (Globodera pallida), and its effects on other important soil pathogens and on the growth of a subsequent potato crop were also assessed. Additional experimental treatments were a potato crop from which the haulm was removed and a green manure crop. Three potato cultivars with different degrees of resistance to PCN were grown as the main crop. Duplicate sets of the experiment were run concurrently. The PCN were effectively controlled by the potato trap crop. When a highly resistant potato cultivar was grown as a main crop after the trap crop, the post‐harvest soil infestation was very low. When a moderately resistant cultivar was grown after the trap crop the soil infestation also remained low. When the trap crop was alternated with a susceptible potato cultivar as a main crop, soil infestation increased slightly, but the degree of control when compared with no trap crop averaged 96%. Soil infestation with root‐knot nematodes (mainly Meloidogyne hapla) increased when potato was grown as a trap crop, but soil infestation with the root‐lesion nematode Pratylenchus crenatus was not affected. Stem canker caused by Rhizoctonia solani was not affected by the trap crop but black scurf (sclerotia of R. solani) on tubers was reduced. Soil infestation with Verticillium dahliae declined in one of the duplicate sets of the experiment but not in the other. However, stem infections by V. dahliae were significantly decreased in both sets, although the effect depended on the PCN‐resistance level of the potato cultivar. When a highly resistant potato cultivar was grown Verticillium stem infections were not significantly affected, they were decreased with a moderately resistant cultivar but the decrease was most pronounced with a PCN‐susceptible cultivar. Senescence of a following potato crop was not influenced by the trap crop when a highly PCN‐resistant cultivar was grown, but it was delayed in the case of a moderately resistant or a susceptible cultivar, resulting in higher tuber yields for those cultivars. The experiment proved that a trap crop can be an alternative to chemical soil disinfection but, for several reasons, the potato itself is not an ideal crop for this purpose; a trap crop other than potato must be developed.  相似文献

A bacterium which can utilize potato starch granules as sole carbon source was isolated and identified as Bacillus circulans from its physiological and biochemical properties. Scanning electron microscopic observation of potato starch granules recovered from the culture broth revealed that granules were degraded gradually from their surface resulting in elongated granules with layered structures on their surface. This bacterium produced extracellular amylase which can digest potato starch granules in vitro. The amylase has a unique property in that it produces only maltohexaose from gelatinized starch in the early stage of the reaction. For the production of this amylase potato starch was found to be most effective while soluble sugars including gelatinized starch and maltose had little effect.  相似文献

Insecticide resistance problems have increased interest in trap crops as a cultural control strategy for overwintered Colorado potato beetle adults, Leptinotarsa decemlineata Say (Coleoptera: Chrysomelidae). Growers in the north central United States have noticed and treated concentrations of adults at the edge of some of their potato fields each spring. Based on sampling in commercial potato fields over a 2-year period, early planted fields that are adjacent to the previous year's potato crop are most likely to have concentrations of adults at the field edge. Frequency of fields with significantly more adults at the edges than in the center sections as well as adult population density in the center sections of fields declined with both distance from the previous potato field and later planting date. The effects of both physical and chemical barriers to movement into potato fields from the field edges were studied in small plot trials and at the edges of commercial potato fields. In small plot trials, physical barriers had a greater impact than chemical barriers on adult beetle movement from a potato trap crop to the protected potatoes beyond the barrier. Barrier treatments reduced beetle numbers in and just beyond the barrier in commercial fields, but the effects were localized and no significant reduction of beetles was observed further into the field. Beetle flight was hypothesized to be responsible for the localized effects of barrier treatments and the lack of edge concentrations in later planted and more distantly rotated fields. In field studies, larger potato plants attracted more colonizing potato beetles than smaller plants. Attracting Colorado potato beetles to trap crops containing potato plants that were larger than those in the remainder of the field, however, provided no significant reduction of beetles in the remainder of the field. We found little opportunity to reduce beetle populations with trap crops at the edges of potato fields without controlling the adults in the trap crop itself. Growers can exploit naturally occurring concentrations of adults at the edges of early and adjacent potato plantings if they are prepared to monitor and regularly treat the field edges.  相似文献

Landrace potato cultivars are native to two areas in South America: the high Andes from eastern Venezuela to northern Argentina and the lowlands of south-central Chile. Potato first appeared outside of South America in Europe in 1567 and rapidly diffused worldwide. Two competing hypotheses suggested the origin of the "European" potato from the Andes or from lowland Chile, but the Andean origin has been widely accepted over the last 60 years. All modern potato cultivars predominantly have Chilean germplasm, explained as originating from breeding with Chilean landraces subsequent to the late blight epidemics beginning in 1845 in the UK. The Andean origin has been questioned recently through examination of landraces in India and the Canary Islands, but this evidence is inferential. Through a plastid DNA deletion marker from historical herbarium specimens, we report that the Andean potato predominated in the 1700s, but the Chilean potato was introduced into Europe as early as 1811 and became predominant long before the late blight epidemics in the UK. Our results provide the first direct evidence of these events and change the history of introduction of the European potato. They shed new light on the value of past breeding efforts to recreate the European potato from Andean forms and highlight the value of herbarium specimens in investigating origins of crop plants.  相似文献

Chinese farmers frequently use a wheat–potato cropping system. The land area planted to transgenic potatoes is increasing because transgenic potatoes have greater resistance to pests and diseases. However, little is known about the bio-compatibility of transgenic potatoes with wheat straw. The objective of this tissue culture study was to determine the allelopathic effects of wheat straw on transgenic potato seedlings. Seedlings were cultured on normal MS medium (normal treatment) and nutrient-deficient MS medium (acclimated treatment) and then transferred to MS medium, which contained wheat straw powder. Wheat straw powder inhibited potato seedling growth in both treatments. Among the parameters analyzed in this study, inhibition was greatest for plant fresh weight and least for plant height. The inhibitive effects of wheat straw were greater for seedling roots compared to shoots. Resistance to allelopathic pressure from wheat straw was greater in acclimated seedlings compared to normal seedlings. This suggested that previous pressure may have induced tolerance in the transgenic potato seedlings. Furthermore, growth inhibition of potato seedlings from the normal treatment increased as the amount of wheat straw powder in the culture medium increased. Calculations indicated that the presence of wheat straw would lead to a 55% reduction in the total biomass of normal potato seedlings compared to a 39% reduction for acclimated seedlings. Parameters such as net photosynthesis rate (Pn) and quantum yield (Y(II)) changed as the nutrient content of the culture medium increased or decreased, but the changes in the parameters were smaller for acclimated seedlings compared to normal seedlings. This suggests that nutrient status during the culture period could help transgenic potato seedlings adapt and compensate for energy loss from seedlings in defending against allelopathic pressure. In summary, the results show that previous exposure to pressures such as nutrient deficiency may increase the allelopathic pressure resistance of transgenic potato seedlings.  相似文献

The sweet potato sporamin promoter was used to control the expression in transgenic potato of the E. coli appA gene, which encodes a bifunctional enzyme exhibiting both acid phosphatase and phytase activities. The sporamin promoter was highly active in leaves, stems and different size tubers of transgenic potato, with levels of phytase expression ranging from 3.8 to 7.4% of total soluble proteins. Phytase expression levels in transgenic potato tubers were stable over several cycles of propagation. Field tests showed that tuber size, number and yield increased in transgenic potato. Improved phosphorus (P) acquisition when phytate was provided as a sole P source and enhanced microtuber formation in cultured transgenic potato seedlings when phytate was provided as an additional P source were observed, which may account for the increase in leaf chloroplast accumulation (important for photosynthesis) and tuber yield of field-grown transgenic potato supplemented with organic fertilizers. Animal feeding tests indicated that the potato-produced phytase supplement was as effective as a commercially available microbial phytase in increasing the availability of phytate-P to weanling pigs. This study demonstrates that the sporamin promoter can effectively direct high-level recombinant protein expression in potato tubers. Moreover, overexpression of phytase in transgenic potato not only offers an ideal feed additive for improving phytate-P digestibility in monogastric animals but also improves tuber yield, enhances P acquisition from organic fertilizers, and has a potential for phytoremediation.  相似文献

A chimeric gene containing the patatin promoter and the transit-peptide region of the small-subunit carboxylase gene was utilized to direct expression of Escherichia coli glycogen synthase (glgA) to potato (Solanum tuberosum) tuber amyloplasts. Expression of the glgA gene product in tuber amyloplasts was between 0.007 and 0.028% of total protein in independent potato lines as determined by immunoblot analysis. Tubers from four transgenic potato lines were found to have a lowered specific gravity, a 30 to 50% reduction in the percentage of starch, and a decreased amylose/amylopectin ratio. Total soluble sugar content in these selected lines was increased by approximately 80%. Analysis of the starch from these potato lines also indicated a reduced phosphorous content. A very high degree of branching of the amylopectin fraction was detected by comparison of high and low molecular weight carbohydrate chains after debranching with isoamylase and corresponding high-performance liquid chromatography analysis of the products. Brabender viscoamylograph analysis and differential scanning calorimetry of the starches obtained from these transgenic potato lines also indicate a composition and structure much different from typical potato starch. Brabender analysis yielded very low stable paste viscosity values (about 30% of control values), whereas differential scanning calorimetry values indicated reduced enthalpy and gelatinization properties. The above parameters indicate a novel potato starch based on expression of the glgA E. coli gene product in transgenic potato.  相似文献

We characterized the uncoupling activity of the plant uncoupling protein from Solanum tuberosum (StUCP) using mitochondria from intact potato tubers or from yeast (Saccharomyces cerevisiae) expressing the StUCP gene. Compared with mitochondria from transfected yeast, StUCP is present at very low levels in intact potato mitochondrial membranes (at least thirty times lower) as shown by immunodetection with anti-UCP1 antibodies. Under conditions that ruled out undesirable effects of nucleotides and free fatty acids on uncoupling activity measurement in plant mitochondria, the linoleic acid-induced depolarization in potato mitochondria was insensitive to the nucleotides ATP, GTP, or GDP. In addition, sensitivity to linoleic acid was similar in potato and in control yeast mitochondria, suggesting that uncoupling occurring in potato mitochondria was because of a UCP-independent proton diffusion process. By contrast, yeast mitochondria expressing StUCP exhibited a higher sensitivity to free fatty acids than those from the control yeast and especially a marked proton conductance in the presence of low amounts of linoleic acid. However, this fatty acid-induced uncoupling was also insensitive to nucleotides. Altogether, these results suggest that uncoupling of oxidative phosphorylation and heat production cannot be the dominant feature of StUCP expressed in native potato tissues. However, it could play a role in preventing reactive oxygen species production as proposed for mammalian UCP2 and UCP3.  相似文献

用化肥减量和分期施肥、增施有机肥来替代化肥是提高半干旱区全膜覆盖垄沟种植马铃薯水、肥利用效率的有效途径.在4年大田定位试验基础上,设置传统施肥(F)、化肥减量25%花期追施(DF)、化肥减量50%花期追施并增施有机肥(OF)3种养分管理模式,通过测定马铃薯不同生育期的土壤含水量和产量,计算阶段耗水量和水分利用效率,研究施肥方式对半干旱区马铃薯耗水过程的调控及其对产量和水分利用效率的影响.结果表明: 马铃薯花期的土壤贮水量DF最高,但处理间差异不显著;花后DF和OF的耗水深度较F有明显增加趋势.与F相比,2011—2014年DF花前耗水量显著下降,花后耗水量分别增加了36.2%、23.0%、24.8%和19.0%;OF未显著降低马铃薯花前耗水,但2011、2012年花后耗水量增加了20.7%和16.3%.DF的马铃薯块茎产量在2012—2014年较F平均增加2595.1 kg·hm-2,水分利用效率(WUE)在2013、2014年分别显著增加14.4%和6.3%,达到显著差异;OF在2011—2014年平均马铃薯块茎产量较F增加了2945 kg·hm-2,且WUE在2012—2014年显著高于F.DF和OF均能显著调节马铃薯花前花后耗水量,使马铃薯块茎产量、水分利用效率增加,但OF的增加幅度更大.  相似文献

In 1998, Verticillium sp. (CE98Vt1 and CE98Vt2) were isolated from discolored vascular structures of potato tubers sold at a market in Chiba Prefecture. These isolates were identified as Verticillium tricorpus on the basis of cultural and morphological characteristics and PCR diagnosis. This observed vascular discoloration of the potato tuber was demonstrated in three cultivars (Touya, Toyoshiro, and Waseshiro) among eight cultivars by inoculation to seedlings. External and internal symptoms of these isolates were not distinct in potato plants. The virulence of these isolates to potato was very low as compared with Verticillium dahliae. These two isolates were not pathogenic to Chinese cabbage, eggplant, green pepper, larkspur, parsley, snapdragon, soybean, tobacco, and tomato. This is the first report of V. tricorpus from potato in Japan.  相似文献

Belonolaimus longicaudatus has long been recognized as a pathogen of potato (Solanum tuberosum). However, a damage function relating expected yield of potato to population densities of B. longicaudatus at planting has not been derived, and the economic threshold for nematicide application is unknown. The objectives of this study were to derive the damage function of B. longicaudatus on potato and to calculate the economic threshold population density. The damage function data for B. longicaudatus on potato were obtained from an ongoing field study to evaluate cropping systems and nematode management practices. Soil samples were collected from experimental field plots, and nematodes were extracted from a 130-cm³ subsample with a centrifugal-flotation method. A damage function was derived by linear regression of potato yield on nematode population density at planting. Based on this derived damage function and published potato prices, the economic threshold for nematicide application was calculated at 2 to 3 B. longicaudatus/130 cm³ of soil, which was near the detection threshold based on methodology used in this study.  相似文献

Starch branching enzyme was purified from potato (Solanum tuberosum L.) tubers as a single species of 79 kilodaltons and specific antibodies were prepared against both the native enzyme and against the gel-purified, denatured enzyme. The activity of potato branching enzyme could only be neutralized by antinative potato branching enzyme, whereas both types of antibodies reacted with denatured potato branching enzyme. Starch branching enzymes were also isolated from maize (Zea mays L.) kernels. All of the denatured forms of the maize enzyme reacted with antidenatured potato branching enzyme, whereas recognition by antinative potato branching enzyme was limited to maize branching enzymes I and IIb. Antibodies directed against the denatured potato enzyme were unable to neutralize the activity of any of the maize branching enzymes. Antinative potato branching enzyme fully inhibited the activity of maize branching enzyme I; the neutralized maize enzyme was identified as a 82 kilodalton protein. It is concluded that potato branching enzyme (Mr = 79,000) shares a high degree of similarity with maize branching enzyme I (Mr = 82,000), in the native as well as the denatured form. Cross-reactivity between potato branching enzyme and the other forms of maize branching enzyme was observed only after denaturation, which suggests mutual sequence similarities between these species.  相似文献

Tagged Colorado potato beetles (CPB), Leptinotarsa decemlineata (Say), were released on potato plants, Solanum tuberosum L., and tracked using a portable harmonic radar system to determine the impact of host plant spatial distribution on the tendency of the pest to remain on the colonized host plant or patch. Results confirmed the long residency time on the host plant and showed that close connection of the plant to neighboring plants hastened dispersal between plants. Tracking walking CPB for over 6 h in small potato plots revealed that all types of mixed borders tested (potato/bare ground, potato/timothy and potato/woodland) acted as a strong barrier and retained beetles within the patch. In another experiment in potato patches surrounded by bare ground borders, tracked walking CPB displayed similar behaviour for up to four days. The distribution of turning angles in the CPB walking paths was not uniform and corresponded to beetles following the edge rows of potato patches in response to the crop border barrier or reversing their direction as they reached the end of a row and therefore a border. Patch size had no or little effect on beetle retention in the patch. The relative distribution of counts of tagged beetles detected among small (16 m2), medium (64 m2) and large size (256 m2) patches of potato four days after initial release remained similar to that of numbers released. Even though mixed crop borders were a strong barrier to walking CPB emigrating from potato patches, the departure rate of beetles over time was high. Results suggest that the effect of mixed borders is largely limited to dispersal by walking and does not apply to beetles leaving host patches by flight. The manipulation of crop borders and patch size seem to have limited potential for the management of CPB emigrating from potato fields.  相似文献

Some properties of sucrose-P synthetases obtained from various plant tissues, including sweet potato roots, potato tubers and leaves of barley, rape and ladino clover were studied. The specific enzyme activity of the sucrose-P synthetase from sweet potato roots was much lower than that of the sucrose synthetase of the other tissues. The enzyme activity decreased gradually as the roots developed. The optimum pH did not differ between enzyme preparations from sweet potato roots and barley leaves. Manganese chloride exhibited a marked stimulative effect on the sucrose-P synthetase from sweet potato roots and potato tubers, whereas it was inhibited the barley leaf enzyme.

Kinetic studies of sucrose-P synthetase showed that the behavior of the enzyme to the substrates did not differ in the enzyme sources examined. The substrate saturation curve of the enzyme with respect to fructose-6-P was sigmodal in shape, giving a straight line with a slope of 1.35~1.5 (n value) in a plot of the data using the empirical Hill equation. On the other hand, enzymes from all the various tissues exhibited a hyperbolic substrate saturation curve for UDP-glucose, obeying the ordinary Michaelis-Menten type reaction. Manganese chloride had no effect on the Km for UDP-glucose, the S0.5 for fructose-6-P and the n value of the enzyme from potato tuber tissues.  相似文献

The mitochondrial sn-glycerol-3-phosphate and 1-acyl-sn-glycerol-3-phosphate O-acyltransferases from potato tubers and pea leaves were investigated with respect to their intraorganelle localization, their positional and substrate specificities, and their fatty acid selectivities. In mitochondria from potato tubers both enzymes were found to be located in the outer membrane. The 1-acyl-sn-glycerol-3-phosphate O-acyltransferase of pea mitochondria showed the same intraorganelle localization whereas the sn-glycerol-3-phosphate O-acyltransferase behaved like a soluble protein of the intermembrane space. The sn-glycerol-3-phosphate O-acyltransferase of both potato and pea mitochondria used sn-glycerol-3-phosphate but not dihydroxyacetone phosphate as acyl acceptor and exclusively catalyzed the formation of 1-acyl-sn-glycerol-3-phosphate which subsequently served as substrate for the second acylation reaction at its C-2 position. Both acyltransferases of potato as well as pea mitochondria showed higher activities with acyl-CoA than with the corresponding acyl-(acyl carrier protein) thioesters. When different acyl-CoA thioesters were offered separately, the sn-glycerol-3-phosphate O-acyltransferase of potato mitochondria displayed no fatty acid specificity whereas the enzyme of pea mitochondria revealed one for saturated acyl groups. On the other hand, the mitochondrial 1-acyl-sn-glycerol-3-phosphate O-acyltransferases from both potato tubers and pea leaves were more active on unsaturated than on saturated acyl-CoA thioesters. Furthermore, these enzymes preferentially used oleoyl- and linoleoyl-CoA when they were offered in a mixture with saturated ones, although the fatty acid selectivity of the pea enzyme was less pronounced than that of the potato enzyme. The sn-glycerol-3-phosphate O-acyltransferase of potato mitochondria displayed a slight preference for saturated acyl groups.  相似文献
