The dermal covering of most batoid fish is constituted by dermal denticles and by different series of tubercles or thorns. The repartition and the morphological variations of these structures can provide complementary information about the taxonomy of skates and rays. The variations in these dermal structures within Pristiforms, Rajiforms and Myliobatiforms have been studied, taking into consideration the number of tubercles, their location and their arrangement in different series. Following Hubbs and Ishiyama [30], two new terms and 15 new series are indicated. The characteristics of the arrangement and of the morphology of these structures can separate the Rajiforms, having spiny tubercles or thorns, from the Myliobatiformes, bearing lanceolate or heart-shaped tubercles. The main taxinomic characters found are: guitar fish characterized by two scapular series, one well-developed rostral series and tubercles with an anterolateral ornamentation (relief). Within this group, Rhinidae and Rhynchobatidae are set apart by the morphology of their tubercles (devoid of any anterolateral ornamentation), by the absence of a middorsal caudal series and by the presence of an outer supraspiracular series. Platyrhina and Platyrhinoidis are distinguishable by the absence of anterolateral relief and by the presence of anterolateral, lateral and parallel series. Rajoids are characterized by thorns, only one scapular series and sometimes a nucho-scapular triangle, malar and alar thorns in adults, and well-developed parallel and lateral series. Myliobatiforms are devoid of rostral, orbito-spiracular, malar, alar, anterolateral, parallel and lateral series but a caudal sting is present in most species. Sawfish are almost entirely devoid of tubercules, except for rostral ‘teeth’. The morphology and arrangement of the rostral teeth can differenciate the two genera within this family.  相似文献

We have previously shown that the costal and crural parts of the diaphragm have different actions on the rib cage (RC) and that the tension developed in one part is not transmitted perfectly to the other. Thus the diaphragm can be modeled pneumatically or electrically as two generators or pumps in series between the lung and abdomen. As such, the force developed by diaphragmatic contraction is the sum of the forces developed in each part, whereas the volume displaced is the same for each part and equal to the total volume displaced. The costal part of the diaphragm is in series with the intercostal and accessory (IA) muscles between the lung and RC, whereas the crural part is in parallel. The volume displaced by simultaneous contraction of the crural part and IA is the sum of volumes displaced by each part. The action of pleural and abdominal pressure [acting through the area of apposition (Aap) of the diaphragm to RC] can be modelled as a summing junction between IA and RC. With hyperinflation the costal part acts more and more in parallel with both IA and the crural part, whereas Aap diminishes, so that the ability to develop large forces decreases independently of the muscles' force-length relationships. The model also predicts that the factors determining the length of the costal and crural parts are different. Finally, the parallel and serial arrangement of the inspiratory musculature allows for increases in maximum power, maximum force, and maximum velocity by appropriate recruitment of the various muscle groups.  相似文献

The influence of the Bohr-Haldane effect (BH) on steady-state gas exchange has previously been described by its effect of gas transfer from the blood when arterial and venous blood gas tensions were held constant. This report quantifies by computer analysis the effects of BH when either or both arterial and venous blood gas tensions are subject to change. When mixed venous blood gas composition is held constant, elimination of BH from a single lung unit typically reduces CO2 output by 6.5% and O2 uptake by 0.5%. Similar effects occur in a two-compartment lung model whether alveolar ventilation-perfusion (VA/Q) mismatch occurs in a parallel or series ventilatory arrangement. When arterial blood gas composition is held constant, elimination of BH increases systemic venous CO2 partial pressure, but O2 partial pressure is hardly affected in the absence of metabolic acidosis. When both mixed venous and arterial blood gas tensions vary and gas exchange is stressed by VA/Q inequality, altitude, anemia, or exercise, elimination of BH predominantly affects mixed venous rather than arterial blood gas tensions. it is concluded that BH may act primarily to reduce tissue acidosis.  相似文献

The present article describes the three-dimensional arrangement of collagen fibrils in dermal plates of different species of Ostraciidae. These dermal plates or 'scutes' are transformed scales, which have a polygonal shape and form a rigid tiling. They are natural composites, associating a fibrous network with a mineral deposit lying at two different levels of the scute, the 'ceiling' and the 'floor', plus a set of similarly mineralized walls joining the two levels. The three-dimensional structure of the collagen network can be compared to that of 'plywood': fibrils align parallel within superposed layers of uniform thickness, and their direction changes from layer to layer. In the dermal plate, two types of plywood have been evidenced: (1) one lying between the two mineralized plates, where the orientation of fibrils rotates continuously, and (2) one under the lower plate, with thick layers of fibrils, each showing a constant orientation, but abrupt angular changes are observed at the transition from one layer to the following one. In oblique sections, both types of plywood reveal large series of arced patterns, testifying to a twisted arrangement of collagen fibrils, analogous to the arrangement of molecules or polymers in cholesteric liquid crystals. The network is reinforced by some collagen fibrils running unidirectionally and almost normally to the lamellate structure. Moreover in the overall organization of the scute, these plywood systems form a set of nested boxes. This original architecture is compared to the arrangement of the collagenous network previously described in most fish scales and in other extracellular matrices.  相似文献

Most restriction endonucleases, including FokI, interact with two copies of their recognition sequence before cutting DNA. On DNA with two sites they act in cis looping out the intervening DNA. While many restriction enzymes operate symmetrically at palindromic sites, FokI acts asymmetrically at a non-palindromic site. The directionality of its sequence means that two FokI sites can be bridged in either parallel or anti-parallel alignments. Here we show by biochemical and single-molecule biophysical methods that FokI aligns two recognition sites on separate DNA molecules in parallel and that the parallel arrangement holds for sites in the same DNA regardless of whether they are in inverted or repeated orientations. The parallel arrangement dictates the topology of the loop trapped between sites in cis: the loop from inverted sites has a simple 180° bend, while that with repeated sites has a convoluted 360° turn. The ability of FokI to act at asymmetric sites thus enabled us to identify the synapse geometry for sites in trans and in cis, which in turn revealed the relationship between synapse geometry and loop topology.  相似文献

The egg capsule of the dogfish is a composite material containing collagenous fibrils and 2 mum spherical hydrophobic protein granules. The latter appear to owe much of their hydrophobicity to an exceptionally high tyrosine content (approximately 20% of total amino acid residues). The hydrophobic component appears to form as an emulsion in the secretory granules of the D and E zone gland cells of the nidamental gland. Droplets of the hydrophobic material appear to become coated with remarkably regular layers of radially-arranged collagen molecules which form a series of concentric, evenly spaced layers around each hydrophobic granule. Numerous disclinations were seen where the layers around adjacent granules interfered with one another. The layers are thought to represent a lamellar liquid crystalline phase previously described for this collagen (Knight et al., 1993). The fine structural appearance of the concentric layers and evidence for radial arrangement of collagen molecules within them is compatible with the suggestion that the layers are built from a dumbbell-shaped unit approximately 35 nm long with hydrophobic groups concentrated at the ends. This unit may represent a dumbbell-shaped molecule or an oligomer of two or more molecules lying parallel with one another in a head-to-tail arrangement. Such a unit can be readily incorporated into models for the micellar, hexagonal columnar and final fibrillar phases previously described for this collagen (Knight et al., 1993). Evidence from the TEM study of stretched egg capsule wall suggests that there is a mechanical interaction between the hydrophobic granules and the collagen fibrils in the fully formed material. We suggest that the radial, concentric layered arrangement of collagen molecules is established by hydrophobic interactions within the liquid crystalline material and locked into place by oxidative covalent cross-linking to give a 3-dimensional cross-linked meshwork of collagen fibrils and hydrophobic granules. The latter arrangement helps to account for the high tensilestrength and toughness of this material.  相似文献

Three-way DNA junctions can adopt several different conformers, which differ in the coaxial stacking of the arms. These structural variants are often dominated by one conformer, which is determined by the DNA sequence. In this study we have compared several three-way DNA junctions in order to assess how the arrangement of bases around the branch point affects the conformer distribution. The results show that rearranging the different arms, while retaining their base sequences, can affect the conformer distribution. In some instances this generates a structure that appears to contain parallel coaxially stacked helices rather than the usual anti-parallel arrangement. Although the conformer equilibrium can be affected by the order of purines and pyrimidines around the branch point, this is not sufficient to predict the conformer distribution. We find that the folding of three-way junctions can be separated into two groups of dinucleotide steps. These two groups show distinctive stacking properties in B-DNA, suggesting there is a correlation between B-DNA stacking and coaxial stacking in DNA junctions.  相似文献

Bromide uptake was measured in single maturing erythroblastic cells of rabbits by means of X-ray microanalysis. Increase in bromide uptake as the cells matured was observed. The order of cells from low to high bromide uptake was: early erythroblast less than late erythroblast less than marrow red cells less than peripheral red blood cells. The transition from low to high bromide uptake is correlated to the accumulation of iron which begins in the late erythroblast. A decrease in rubidium uptake also occurs as iron accumulates in the cell. These results indicate that the anion and cation transport changes during maturation are parallel in time course but opposite in direction. In addition, the increase in bromide uptake can be accounted for by the increase in surface-to-volume ratios of the cells. Surface-to-volume ratios were estimated by morphometric techniques.  相似文献

The single-pass, bolus-injection method was used to study the effect of serotonin (5-HT) concentration on the extraction of 5-HT by isolated perfused rabbit lungs. The extraction pattern suggested that an uptake model, which includes multiple parallel uptake processes, provided a better representation of the data than the simple Michaelis-Menten equation, which has commonly been used to fit the saturable uptake data in previous studies. In particular, the rabbit lung data could be fit with two such parallel pathways. Since the 5-HT uptake could virtually be completely blocked by imipramine, both pathways can be considered to be carrier-mediated processes. The high-affinity pathway was saturable within the range of concentrations studied, with a Km and Vmax of approximately 0.84 microM and 0.21 nmol.s-1.g wet lung wt-1. The Km for the low-affinity pathway was larger than concentrations for which accurate uptake measurements are practical in the perfused organ. Thus, for the low-affinity pathway, only Vmax/Km was identifiable. Vmax/Km values for the high- and low-affinity pathways were approximately 2.87 and 0.35 ml/s, respectively. The results suggest that it will be worthwhile to investigate the behavior of these uptake parameters in response to changes in lung physiology and endothelial function in future studies.  相似文献

The recently published microcrystal structures of amyloid fibrils from small peptides greatly enhanced our understanding of the atomic-level structure of the amyloid fibril. However, only a few amyloid fibrils can form microcrystals. The dansyl-tryptophan fluorescence resonance energy transfer (FRET) pair was shown to be able to detect the inter-peptide arrangement of the Transthyretin (105-115) amyloid fibril. In this study, we combined the known microcrystal structures with the corresponding FRET efficiencies to build a model for amyloid fibril structure classification. We found that fibrils with an antiparallel structural arrangement gave the largest FRET signal, those with a parallel arrangement gave the lowest FRET signal, and those with a mixed arrangement gave a moderate FRET signal. This confirms that the amyloid fibril structure patterns can be classified based on the FRET efficiency.  相似文献

Summary The theoretical treatment of diffusion of solutes to a number of parallel, competing roots is difficult, but an electrical analog has been constructed which allows solute uptake by such a system to be simulated easily and rapidly. The construction, theory and operation of the analog are described. Differences in diffusion coefficients, dimensions, root size and uptake properties can all be dealt with. Approximate methods are available for simulating mass flow with diffusion, slow release of nutrients in the soil, the presence of root hairs, and incomplete root-soil contact.  相似文献

  1. Both the organization of behaviour and communicative interactions can be established by analyzing behavioural time series. By means of an analysis of this kind, conclusions about the control of behaviour and the principles of interindividual communication can be reached. An analysis may be based on simultaneous and successive behavioural events. In these multi-channel time series the temporal arrangement of patterns (“Strukturierung”), the organization and the conditions determining the occurrence of behavioural events can be specified.
  2. Data-analysis by digital computer permits rapid processing of a large quantity of material in regard to several respects. “Sets of data” can be stored, thus enabling the user to correct, alter, combine and finally analyze them with respect to various questions. For this purpose, a program system for the digital processing of behavioural time series (title “PROVED”) was developed.
  3. The course of behavioural events is composed of temporal patterns which are ordered in a definite hierarchy (Fig. 1). Several quantitative and qualitative characteristics (“Muster-Merkmale”) can be attributed to each pattern (Figs. 2,4). With the PROVED-System, the temporal arrangement of behavioural patterns is reestablished by the computer according to the control orders of the user. These orders must take into consideration the physiological relevance that is to be confirmed by an analysis of time serics (Fig. 3). The orders controlling the temporal arragement as well as the asignment of pattern characteristics to sets of data are available for further evaluation.
  4. The PROVED-System includes several procedures involving input, output, storage, administration, correction, assignment of characteristics, temporal arrangement and analysis of data. The examiner can select and combine these procedures at will using statements of a particular control language.
  5. During the first step of data analysis, a fixed number of parameters for each pattern is determined, the number of parameters varying with the complexity of the problem. Any measured value which is related to the pattern under investigation can be regarded as a parameter. During the second step, multidimensional frequency distributions of crossclassified parameters are established which can be examined by means of statistical tests (third step) (Figs. 5,6).
  6. Determination of the parameters by the examiner decisively influences the further course of the analysis. The control statements for one parameter consist of two parts: 1. quality of the parameter (type of pattern, temporal distance, etc.), 2. position of the parameter in the behavioural time series (pattern under investigation, immediately subsequent pattern, next pattern with the same characteristics, etc.). The latter is determined by a series of shifts of indices controlled by the user (Fig. 7,8).
  7. The PROVED-System can be applied to the analysis of various behavioural time series (succession of sounds, recordings of movements, observations of behavioural acts, etc.). Depending upon the aim of the analysis (temporal arrangement, organization, control of behaviour or communication), spontaneous behavioural events, input-output experiments (stimulus-reaction) and social interaction can be evaluated.

Summary The cell wall of root hairs ofLimnobium stoloniferum is composed of two fibrillar layers: an outer layer with a dispersed texture and an inner layer with a helicoidal texture. In stained oblique sections the helicoidal layer appears as a series of bow-shaped structures. In sections which were shadow-casted after the embedding medium was removed, the following properties of the helicoidal layer can be directly observed. (1) It is build up of superimposed lamellae. (2) Each lamella consists of parallel oriented microfibrils. (3) Going into the helicoidal layer, there is a counter-clockwise discontinuous rotation of the microfibril orientation in successive lamellae. (4) Between adjacent lamellae the average angular displacement of the microfibril orientation is about 23 degrees. The dispersed outer layer is also polylamellated, but with randomly arranged microfibrils in each lamella. Both layers are present in the lateral wall as well as in the apical wall of the root hairs. Observations indicate that in the cell wall of the tip the parallel oriented microfibrils of the outermost helicoidal lamellae become distorted towards a dispersed arrangement. The suggestion is made that the dispersed outer layer is derived from the helicoidal layer.  相似文献

The formation of fibril aggregates by long polyglutamine sequences is assumed to play a major role in neurodegenerative diseases such as Huntington. Here, we model peptides rich in glutamine, through a series of molecular dynamics simulations. Starting from a rigid nanotube-like conformation, we have obtained a new conformational template that shares structural features of a tubular helix and of a beta-helix conformational organization. Our new model can be described as a super-helical arrangement of flat beta-sheet segments linked by planar turns or bends. Interestingly, our comprehensive analysis of the Protein Data Bank reveals that this is a common motif in beta-helices (termed beta-bend), although it has not been identified so far. The motif is based on the alternation of beta-sheet and helical conformation as the protein sequence is followed from the N to the C termini (beta-alpha(R)-beta-polyPro-beta). We further identify this motif in the ssNMR structure of the protofibril of the amyloidogenic peptide Abeta(1-40). The recurrence of the beta-bend suggests a general mode of connecting long parallel beta-sheet segments that would allow the growth of partially ordered fibril structures. The design allows the peptide backbone to change direction with a minimal loss of main chain hydrogen bonds. The identification of a coherent organization beyond that of the beta-sheet segments in different folds rich in parallel beta-sheets suggests a higher degree of ordered structure in protein fibrils, in agreement with their low solubility and dense molecular packing.  相似文献

In the lateral musculature of the axolotl tail a multipennate arrangement of muscle fibres is found in the 3rd postanal myomere and all following myomeres. This arrangement appears in larval stages as soon as the myomeres concerned are fully differentiated. Within a myomere a uniform degree of contraction of the muscle fibres may be achieved either by helical arrangement as seen in Teleosts, by diminishing fibre length in medial direction or by a combination of both phenomena. The relation between muscle fibre length and angle of insertion in the myomeres showing pennate arrangement is approximated by the Benninghoff and Roll-h?user equation but the values recorded deviated systematically from the values calculated. In the rather slender tail of the axolotl the transition from the helical or parallel fibre system into the pennate arrangement will be in favour of the development of relatively strong bending moments in the region concerned while a reasonable uniformity of the rate of contraction is maintained.  相似文献

The kinetics of substrate uptake and product formation in the process of citric acid accumulation by Aspergillus niger on sucrose as a sole carbon source are presented. The experiments are aimed at studying if glucose and fructose obtained from the hydrolysis of sucrose are equivalent carbon sources for A. niger and how the presence of the two different carbon substrates might influence the citric acid formation process. Beet sugar was used as a sole carbon source in the first series of experiments conducted in two types of bioreactors: stirred tank and air-lift. The fructose uptake rate was significantly lower than the glucose uptake rate in the late idiophase. A substrate utilisation breakpoint occurred when a large amount of citric acid was accumulated in the fermentation broth. A similar phenomenon was also detected in repeated fed-batch fermentation. This phenomenon was confirmed by the second series of parallel shake culture runs, in which fungal growth and citric acid accumulation by A. niger was simultaneously tested on the media containing the following carbon sources: sucrose, glucose and fructose, with and without addition of concentrated citric acid solution. Finally, it was shown that high concentration of citric acid strongly depleted fructose uptake rate.  相似文献

The surface area of various types of montmorillonites (MMT) with different values of layer charge plays a very important role in surface arrangement of methylene blue cations (MB). Photoluminescence measurements can be strongly or partially influenced by this surface arrangement of cations. For these reasons and on the basis of our previous results, molecular simulations were performed for various types of montmorillonites covered with methylene blue cations. Adsorption of methylene blue cations on Na-Wyoming MMT surface is different from Ca-Cheto MMT. In the case of Wyoming with a lower layer charge, MB cations lie parallel to the silicate layer for all investigated samples. On the other hand, Cheto surface is covered with a higher amount of MB cations. The results obtained from molecular modeling indicate that MB lies parallel to low loading case and become tilted with respect to layer for a higher loading. Moreover, a higher amount of MB cations covering the silicate layer are much less energy-stable. A higher loading of MB cations leads to aggregates but at low loading MB cations degrade to monomers.  相似文献

The double refraction of the chitinous hair of the crayfish is positive with respect to the axis of the hair, and is largely caused by the arrangement of submicroscopic, elongated chitin particles parallel to this axis (form birefringence). Using a series of relatively unreactive liquids and fluid mixtures which permeate the chitin framework, the type of curve relating double refraction and refractive index of the imbibed fluid is found to depend greatly on the chemical nature of the fluid. Either a positive or a negative residual birefringence may be found, depending on the choice of imbibing liquid. Separation of form and crystalline elements in double refraction by means of Ambronn''s imbibition technique is therefore unsafe in a system like chitin, where some type of oriented association of the imbibed molecules with the chitin framework is prevalent.  相似文献

The ontogeny of the uptake of glutamate, GABA and glycine into synaptic vesicles isolated from rat brain has been investigated. The vesicular uptake of the three amino acids increased with developmental age in parallel with synaptogenesis, indicating a functional role of uptake of the amino acids by synaptic vesicles in the nerve terminals. Uptake of the amino acids by plasma membrane particles (synaptosomes) in brain homogenate showed a somewhat different developmental profile. The uptake of glutamate increased markedly with developmental time, while the uptake of GABA showed only a slight increase. Uptake of glycine by plasma membrane particles was very low and therefore not registered. The observed developmental increase in uptake of glycine by synaptic vesicles isolated from brain, supports previous reports indicating that glycine can be taken up by vesicles from non-glycine terminals.Special issue dedicated to Dr. Morris H.
