Alpha-amylases are ubiquitous proteins which play an important role in the carbohydrate metabolism of microorganisms, animals and plants. Living organisms use protein inhibitors as a major tool to regulate the glycolytic activity of alpha-amylases. Most of the inhibitors for which three-dimensional (3-D) structures are available are directed against mammalian and insect alpha-amylases, interacting with the active sites in a substrate-like manner. In this review, we discuss the detailed inhibitory mechanism of these enzymes in light of the recent determination of the 3-D structures of pig pancreatic, human pancreatic, and yellow mealworm alpha-amylases in complex with plant protein inhibitors. In most cases, the mechanism of inhibition occurs through the direct blockage of the active center at several subsites of the enzyme. Inhibitors exhibiting "dual" activity against mammalian and insect alpha-amylases establish contacts of the same type in alternative ways.  相似文献

The baculovirus vector systems has been extensively used for the expression of foreign gene products in insect and mammalian cells. New advances increase the possibilities and applications of the baculovirus expression system, which has the capability to express multiple genes simultaneously within a single infected insect cells and to use recombinant virus with mammalian cell-active expression cassettes to permit expression of recombinant proteins in mammalian cells in vitro and in vivo. Future investigations of the baculovirus expression system designed for specific target cells, can open wide variety of applications. This review summarizes the recent achievements in applications the baculovirus vector systems and optimization recombinant protein expression in both insect and mammalian cell lines.  相似文献

Permethrin-impregnated clothing and three topical repellent formulations of deet (diethyltoluamide) were field tested against natural populations of tsetse flies, mostly Glossina morsitans centralis Machado, in central Zambia. Volunteers wore different combinations of clothing impregnated with permethrin 0.125 mg ai/cm2 and repellents while riding in a vehicle that was driven slowly (4-6 km/h), with the windows and rear door open, through fly-infested areas. The mean rate of tsetse bites was about twenty per 75 min for unprotected people. The treatment combination of permethrin-impregnated clothing (blue cotton coveralls) and either of two controlled-release deet formulations on exposed skin of face and arms provided 91% mean protection, but this was not significantly better (P greater than 0.05) than wearing deet repellent alone (76-87% protection). No significant differences of protection were observed between the three repellent treatments, although the two controlled-release formulations (intended to be more persistent) were applied at approximately half the dosage of the standard 75% deet. Wearing permethrin-impregnated coveralls alone provided relatively poor protection (34%) for the untreated and exposed skin of head and hands. However, olive drab mesh jackets treated with permethrin reduced the tsetse biting rate by 75%.  相似文献

Penetration of the nervous systems of suckling mice by prototype strains of the three mammalian reovirus serotypes was studied after footpad inoculation of a dose (10(7) PFU) representing 3.5 x 10(3) 50% lethal doses (LD50) for reovirus type 3 Dearing and less than 1 LD50 for reoviruses type 1 Lang and type 2 Jones. Type 3 Dearing entered both motor and sensory neurons; infected neurons were clearly detectable by immunohistochemical staining 19 h after inoculation. By day 2, a second cycle of infection had occurred, and by day 4, several hundred motor and sensory neurons and interneurons were infected. By this time, infection also involved large areas of the brain stem and brain. There was evidence of both retrograde and anterograde movement of viral antigen within axons and dendrites. Unexpectedly, reovirus type 1 Lang followed neuronal pathways as well as being disseminated in the bloodstream. Reovirus type 2 Jones also entered neurons. While the number of motor neurons and interneurons infected with type 1 Lang or type 2 Jones remained limited within the first 4 days after inoculation, infection of sensory neurons increased with time and reached a level by day 4 comparable to that observed after infection with type 3 Dearing. Viral antigen was also found in the brain stem and brain, but this infection was limited. These three strains multiplied in nonneuronal tissues. Connective tissue in the footpad was massively infected by all three strains 19 h after inoculation. By this time, foci of infection were also present in muscle and skin. Viral antigen was repeatedly observed in the endothelium of blood vessels and in the meninges after infection with type 1 Lang. The titer of type 1 Lang increased in the blood with time, which was not observed after infection with strains of the other two serotypes. In this study, we found that prototype strains of the three reovirus serotypes exhibited different degrees of neurotropism, all being capable of entering neurons. Transmission of the infection occurred through synapses rather than from cell body to cell body. Thus reovirus, like herpesvirus and rabies virus, is a good marker for the identification of neuronal pathways, although its capacity to grow in neurons, unlike that of herpesvirus and rabies virus, is restricted to newborn animals.  相似文献

Baculovirus vector systems are extensively used for the expression of foreign gene products in insect and mammalian cells. New advances increase the possibilities and applications of the baculovirus expression system, which makes it possible to express multiple genes simultaneously within a single infected insect cell and to obtain multimeric proteins functionally similar to their natural analogs. Recombinant viruses with expression cassettes active in mammalian cells are used to deliver and express genes in mammalian cells in vitro and in vivo. Further improvement of the baculovirus expression system and its adaptation to specific target cells can open up a wide variety of applications. The review considers recent achievements in the use of modified baculoviruses to express recombinant proteins in eukaryotic cells, advantages and drawbacks of the baculovirus expression system, and ways to optimize the expression of recombinant proteins in both insect and mammalian cell lines.  相似文献

Using acetylthiocholine as substrate, microscopically localizable cholinesterase (ChE) activity is demonstrated in neural and glial elements of central and peripheral nervous systems of the lobsters, Panulirus argus and Homarus americanus. Moderate to very intense ChE activity occurs in all synaptic regions of the central ganglia and stomatogastric ganglion, in glial sheaths around neuron somata and peripheral nerve axons, and in cytoplasm of a few nerve cell bodies. Axons, identified as motor, contain extremely little ChE. The principal reaction in peripheral nerves occurs in sheath elements of sensory fibres; in most cases, much of the reaction is lost as the nerves lose the sheaths at the point of entry into brain.  相似文献

Methods to measure resistance to inhibition by organophosphorus toxicants (OP) for mutants of butyrylcholinesterase (EC 3.1.1.8; BChE) and acetylcholinesterase (EC 3.1.1.7; AChE) enzymes were devised. Wild-type cholinesterases were completely inhibited by 0.1 mM echothiophate or 0.001 mM diisopropylfluorophosphate, but human BChE mutants G117H, G117D, L286H, and W231H and snake AChE mutant HFQT retained activity. Tissues containing a mixture of cholinesterases could be assayed for amount of G117H BChE. For example, the serum of transgenic mice expressing human G117H BChE contained 0.5 microg/ml human G117H BChE, 2 microg/ml wild-type mouse BChE, and 0.06 microg/ml wild-type mouse AChE. The oligomeric structure of G117H BChE in the serum of transgenic mice was determined by nondenaturing gel electrophoresis followed by staining for butyrylthiocholine hydrolysis activity in the presence of 0.1 mM echothiophate. Greater than 95% of the human G117H BChE in transgenic mouse serum was a tetramer. To visualize the distribution of G117H BChE in tissues of transgenic mice, sections of small intestine were treated with echothiophate and then stained for BChE activity. Both wild-type and G117H BChE were in the epithelial cells of the villi. These assays can be used to identify OP-resistant cholinesterases in culture medium and in animal tissues.  相似文献

The prevention and treatment of drug-resistant malaria is becoming increasingly difficult. On the Thai–Myanmar border multi-drug resistant strains of falciparum malaria are increasing and, because the malaria vector Anopheles bite outdoors during early evening, insecticide house-spraying or impregnated bednets provide only limited protection. Therefore, the protective efficacy of repellent formulations containing di-methyl benzamide (deet) and permethrin against local vectors was estimated, when applied to the skin, and their acceptability amongst pregnant Karen women who are at relatively high risk from malaria was assessed. Human landing catches of mosquitoes showed that almost complete protection was achieved using different formulations of 20% deet and 0.5% permethrin for up to 6 h. All-night collections from human subjects indicated that this repellent combination reduced exposure to malaria parasites by at least 65 and 85% for those transmitted by Anopheles minimus and An. maculatus , respectively, the two principal vectors in this area. Pregnant women in the camps preferred repellents which were mixed with 'thanaka', a root paste made from pulp of the wood apple tree, Limonia acidissima , used locally as a cosmetic. Apart from a temporary warming sensation where repellent thanaka was applied to the skin, the repellents were well tolerated. An intervention trial is currently in progress to determine whether deet mixed with thanaka can protect pregnant women against malaria in this part of the world. Bioassays using a laboratory strain of Aedes aegypti demonstrated that thanaka is itself slightly repellent at high dosages and the mixture with deet provides protection for over 10 h. This treatment would therefore also provide some personal protection against dengue, which is increasing locally, transmitted by Ae. aegypti and Ae. albopictus biting during the daytime.  相似文献

Nitric oxide (NO) is a membrane permeant signalling molecule which activates soluble guanylyl cyclase and leads to the formation of cyclic GMP (cGMP) in target cells. In the nervous system, NO/cGMP signalling is thought to play essential roles in synaptic plasticity during development and also in the mature animal. This review summarizes neurochemical, cell biological, and physiological investigations of NO/cGMP signalling in the nervous system of insects. The anatomical localization of donor and target cells suggests functions in olfaction, vision, and mechanosensation. Behavioural assays have uncovered contributions of NO signalling in oxygen sensing, habituation to chemosensory stimuli, and associative memory formation. During development, NO regulates cell proliferation, axonal outgrowth, and synaptic maturation. The cellular distribution of NO-responsive cells suggests that NO can serve as a retrograde synaptic messenger, as an intracellular messenger, and as a lateral diffusible messenger irrespective of conventional synaptic connectivity.  相似文献

The polypeptide chain initiation factor 2 (eIF-2) binds phospholipid (PL) and becomes a potent inhibitor of translation in hemin-supplemented reticulocyte lysates [De Haro et al. (1986) Proc. Natl. Acad. Sci. USA 83, 6711–6715]. This binding is independent of calcium ions and seems to be specific for phosphatidylinositol or phosphatidylserine; phosphatidic and arachidonic acids are inactive. Like -subunit-phosphorylated eIF-2, eIF-2·PL traps GEF in a non-dissociable eIF-2·PL·GEF complex whereby GEF is no longer able to recycle. Initiation is inhibited when no free GEF is available. Translational inhibition by eIF-2·PL is rescued by equimolar amounts of eIF-2·GEF. On the basis of this stoichiometry, we have estimated that reticulocyte lysates contain about 60 pmol of GEF/ml (60 nM) eIF-2·PL also inhibits translation in cell-free mouse liver extracts and this inhibition is prevented by reticulocyte eIF-2·GEF suggesting that GEF also functions in liver. However, the eIF-2·PL complex does not affect translation in such non-mammalian eukaryotic systems as wheat germ and Drosophila embryos.  相似文献

Purification of factors for DNA replication in an amount sufficient for detailed biochemical characterization is essential to elucidating its mechanisms. Insect cell expression systems are commonly used for purification of the factors proven to be difficult to deal with in bacteria. We describe first the detailed protocols for purification of mammalian Mcm complexes including the Mcm2/3/4/5/6/7 heterohexamer expressed in insect cells. We then describe a convenient and economical system in which large-sized proteins and multi-factor complexes can be transiently overexpressed in human 293T cells and be rapidly purified in a large quantity. We describe various expression vectors and detailed methods for transfection and purification of various replication factors which have been difficult to obtain in a sufficient amount in other systems. Availability of efficient methods to overproduce and purify the proteins that have been challenging would facilitate the enzymatic analyses of the processes of DNA replication.  相似文献

Summary Acetylcholinesterase was demonstrated at ultrastructural level in the motor nerve cells of rat's spinal cord using the Karnovsky-Roots modification of Koelle's thiocholine method. Selective inhibitors were employed to check the validity of the reaction.Prolonged formaldehyde fixation improved the poor penetration of the reactive agents and diminished the relatively large crystal size of the end product, which were the two main difficulties of the method. The preservation of ultrastructure was highly improved, when thin sections were made without freezing using a tissue chopper.Acetylcholinesterase was localized in the nuclear envelope, in the rough-surfaced endoplasmic reticulum, in medium-sized vesicles of the Golgi apparatus, and around synaptic terminals. Kinetics and mechanism of in vitro hydrolyses of acetylcholine and acetylthiocholine by carbamates were studied in a batch reactor at 25 degrees C, pH 8, and ionic strength of 0.11 M. Every hydrolysis was monitored by 3-4 independent methods. All studied hydrolyses can be described by the model of competitive inhibition with an irreversible step (k3). A table of obtained average values of rate constants and discussion of the resultes are given.  相似文献

Several lines of evidence indicate that inhibition of butyrylcholinesterase (BuChE) is important in the treatment of certain dementias. Further testing of this concept requires inhibitors that are both BuChE-selective and robust. N-alkyl derivatives (2, 3, 4) of phenothiazine (1) have previously been found to inhibit only BuChE in a mechanism involving pi-pi interaction between the phenothiazine tricyclic ring system and aromatic residues in the active site gorge. To explore features of phenothiazines that affect the selectivity and potency of BuChE inhibition, a series of N-carbonyl derivatives (5-25) was synthesized and examined for the ability to inhibit cholinesterases. Some of the synthesized derivatives also inhibited AChE through a different mechanism involving carbonyl interaction within the active site gorge. Binding of these derivatives takes place within the gorge, since this inhibition disappears when the molecular volume of the derivative exceeds the estimated active site gorge volume of this enzyme. In contrast, BuChE, with a much larger active site gorge, exhibited inhibition that increased directly with the molecular volumes of the derivatives. This study describes two distinct mechanisms for binding phenothiazine amide derivatives to BuChE and AChE. Molecular volume was found to be an important parameter for BuChE-specific inhibition.  相似文献
