Nine distinct anaerobic benzene-degrading cultures were enriched from sediment samples from four different sites. These cultures used nitrate, sulphate or CO2 as electron acceptors. The shortest doubling times were observed in nitrate-reducing cultures, although cell yield was lowest in these cultures. The highest substrate concentration utilized and maximum absolute rates of benzene degraded (in micro M day-1) were observed in methanogenic cultures. The microbial compositions of a methanogenic and nitrate-reducing culture were determined from a clone library of 16S rRNA genes. Five Bacterial 16S rRNA sequences, one of which resembled a clone previously found in a sulphate-reducing, benzene-degrading culture and four Archaeal 16S rRNA sequences were identified in a methanogenic culture. Four Bacterial and no Archaeal 16S rRNA sequences were identified in a nitrate-reducing culture. The relative abundance of the four nitrate-reducing putative species was determined by slot blot hybridization. Two green sulphur bacteria together formed 52% of the clone library, but were found to be less than 4% of the culture by slot blot analysis. One of the cloned 16S rRNA gene sequences comprised 70% of the culture and was phylogenetically 93% similar to both Azoarcus and Dechloromonas species, which have been shown to degrade aromatic compounds, including benzene, under nitrate-reducing conditions.  相似文献

A modified dynamic method is introduced to determine the oxygen transfer coefficient, KL a, in aerobic fermentation systems which are not mechanically agitated. The dissolved oxygen concentration is measured continuously following a step down or a step up in aeration rate. The response curve is analyzed to obtain the value of KLa Experiments were carried out at several different air flow rates using mixed culture in concurrent tower fermentors with motionless mixers. The effect of sieve trays and Koch motionless mixers on oxygen transfer was investigated using a 3 in. diameter column. The values of KL aobtained at the bottom of each column were found to be higher than those obtained at the top. Comparison of the results showed that the values ofKL a were higher when the Koch mixers were used than when the sieve trays were employed. The oxygen uptake rate by the organisms rX, is also calculated by using the KL a values obtained. They compare favorably withthe experimentally measured values.  相似文献

The interaction between epithelium (dog kidney epithelium MDCJ/clone 20) and fibroblasts (diploid human fibroblasts M19 and AG-1523) was studied in mixed heterotypic cell cultures. The mode of cell interaction depends on the manner of their collision. At collision of the epithelium lamella and the lateral side of fibroblast, the lamella was seen to creep under the lateral side to force back the fibroblast. At the frontal collision of epithelium and fibroblast lamellae, the mode of interaction depends on the local situation. With the presence of a free substratum around, the fibroblast formed a new lamella and moved aside from the place of collision. In the case, when the neighboring cells prevented fibroblast from moving, it migrated under the epithelium. In this work, we have first demonstrated the formation of specialized intercellular adhesions between epithelium and fibroblasts. The cultures were studied by phase contrast, interference reflection or video tape recording, using an image processing system (Hamamatsu). For studying adhesion, immuno-fluorescent methods were performed.  相似文献

Natural biofilms rarely exist as monocultures. Usually they are formed from various microorganism species that interact with each other, have shared metabolites, strengthen the attachment of each other to the support substrate, provide expression of "foreign" genes etc. Material on factors and mechanisms that determine the formation of mixed (polymicrobial) biofilms is analyzed in the review. The significance of interspecies interaction between bacteria based on QS system signal autoinductors is underlined. Examples of humoral and contact communications between bacteria and eukaryotes including host cells are provided. Study of polymicrobial processes and their interaction with innate and adaptive immune response seems important for further development of medical microbiology (especially regarding chronic infectious diseases).  相似文献

Abstract The degradation of the following amino acids was investigated in mixed cultures obtained from a waste water purification plant: aspartate, glutamate, serine, alanine, valine and leucine. Inhibition of sulfate-reducing bacteria in these mixed cultures by molybdate was found to inhibit amino acid degradation. The degradation of serine, alanine, valine and leucine was accelerated considerably by active sulfate reduction. The fermentation of aspartate and glutamate was not stimulated by the presence of sulfate-reducing bacteria. The existence of species which are able to ferment valine and leucine by coupling their oxidation to the reduction of exogenous acetate to butyrate was demonstrated.  相似文献

A method to characterise a dynamic microbial consortium is described. By exploiting differences in surface properties between different cells and between cells of different physiological status, it was possible to develop a partition pattern for a mixed culture under different conditions. The separation method used was partition in aqueous two-phase systems and when using a counter current extraction process one could clearly differentiate the partition profile between resting, active and overloaded biomethanation cultures.  相似文献

The kinetics of batch anaerobic hydrogen production by mixed anaerobic cultures was systemically investigated in this study. Unstructured models were used to describe the substrate utilization, biomass growth and product formation in the hydrogen production process. The relationship between the substrate, biomass and products were also evaluated. Experimental results show that the Michaelis-Menten equation, Logistic model and modified Gompertz equation all could be adopted to respectively describe the kinetics of substrate utilization, biomass growth and product formation. Furthermore, the relationship between the acidogenic products and biomass was simulated by Luedeking-Piret model very well. The experimental results suggest that the formation of hydrogen and the main aqueous products, i.e., butyrate and acetate, was all growth-associated.  相似文献

A comprehensive study on the effects of different carbon sources during the bacterial enrichment on the removal performances of benzene, toluene, ethylbenzene, and xylenes (BTEX) compounds when present as a mixture was conducted. Batch BTEX removal kinetic experiments were performed using cultures enriched with individual BTEX compounds or BTEX as a mixture or benzoate alone or benzoate–BTEX mixture. An integrated Monod-type non-linear model was developed and a ratio between maximum growth rate (μ max) and half saturation constant (Ks) was used to fit the non-linear model. A higher μ max/Ks indicates a higher affinity to degrade BTEX compounds. Complete removal of BTEX mixture was observed by all the enriched cultures; however, the removal rates for individual compounds varied. Degradation rate and the type of removal kinetics were found to be dependent on the type of carbon source during the enrichment. Cultures enriched on toluene and those enriched on BTEX mixture were found to have the greatest μ max/Ks and cultures enriched on benzoate had the least μ max/Ks. Removal performances of the cultures enriched on all different carbon sources, including the ones enriched on benzoate or benzoate–BTEX mixture were also improved during a second exposure to BTEX. A molecular analysis showed that after each exposure to the BTEX mixture, the cultures enriched on benzoate and those enriched on benzoate–BTEX mixture had increased similarities to the culture enriched on BTEX mixture.  相似文献

Co-immobilized Aspergillus awamori and Zymomonas mobilis cultures were investigated in a stirred tank reactor on synthetic medium with starch as substrate at various dissolved oxygen concentrations. In a gaslift loop reactor, freely suspended and immobilized A. awamori were cultivated on synthetic medium and soluble potato starch. In the same reactor, the growth and ethanol production of freely suspended and immobilized Z. mobilis cultures were studied on synthetic medium and glucose. Co-immobilized A. awamori and Z. mobilis were cultivated in batch and continuous operations in the gaslift loop reactor on synthetic medium with starch substrate at different dissolved oxygen concentrations. This study attempted to determine the optimal temperature and initial cultivation pH by conducting a series of batch tests in stirred-tank bioreactor using fructose-producing wastewater as an organic substrate. The bioreactor temperature was controlled at 35–55°C with an initial pH of 4–8. Hydrogen production efficiency was assessed using specific hydrogen production potential (SHPP) and the maximum specific hydrogen production rate (SHPRm). Experimental results indicated that temperature and initial pH markedly affected SHPP and SHPRm, volatile fatty acids distribution as well as the ratio of butyrate/acetate (BHu/HAc). Two-fold higher SHPP and SHPRm were obtained at thermophilic condition (55°C) than those at mesophilic condition (35°C). The optimal initial pH was 6 for hydrogen production with peak values of SHPP of 166.8 ml-H2/g-COD and SHPRm of 26.7 ml-H2/g-VSS-h for fructose-processing wastewater. Molasses-processing wastewater had a higher SHPP (187.0 ml-H2/g-COD) and SHPRm (42.7 ml-H2/gVSS-h) than fructose-processing wastewater at pH 6. The DGGE profiles indicated that molasses-processing wastewater is a better substrate than fructose-processing wastewater for growth of hydrogen-producing bacteria due to the high staining intensity of bands.  相似文献

Summary Erythromycin-resistant mitochondria from species 1, 5 and 7 of P. aurelia were injected into erythromycin-sensitive paramecia of each of the same three species. Mitochondria from species 1 and 5 were successfully transferred to all three species, but species 7 mitochondria failed to develop in species 1 and 5. Minor differences were indicated in the frequency of successful transfers of species 1 mitochondria into species 1 and 5 cells. From studies on the transferability of mitochondria from hybrid cells, containing mitochondria from one species and nuclei from another, it was concluded that mitochondrial compatibility was mainly under control of the nuclear genome, with a possible minor control also by the mitochondrial genome.Dedicated to T.M. GET是一种国外新研发的在田间利用秸秆进行甲烷生产的技术。为了推动该技术在我国的实际应用,本研究以稻草与牛粪为发酵原料,水稻田土壤作为基底,采用正交试验法,测试了温度、发酵基质配比(稻草∶牛粪)和秸秆腐熟剂浓度对混合厌氧发酵产气量的影响。结果表明:各处理因子的产气动态呈“S”型增长模式,但在快速增长时间和增长幅度上有一定区别。30 ℃和40 ℃处理的累积产气量显著高于10 ℃和20 ℃处理的累积产气量;发酵基质配比以1∶1处理的累积产气量最大,分别比1∶0(对照)、1∶2和1∶3配比高34.12%、20.39%和21.78%;秸秆腐熟剂浓度的产气效果在实验范围内随着浓度的增加而增加。厌氧发酵过程中各实验因子下挥发性脂肪酸(VFAs)含量和pH在20 d左右分别有一个高峰和低谷值且都有较大差异,各因子主要是通过在这个时期对厌氧发酵进程的影响而对整个产气量产生影响。综合分析显示稻草厌氧发酵产生物甲烷的最优组合为:温度30 ℃、发酵基质配比1∶1、秸秆腐熟剂浓度8%。  相似文献

In the present study mixed cultures that could grew in the molasses media were isolated from textile dye effluent and its decolorization activity was studied in a batch system under anaerobic conditions, in order to determine the optimal conditions required for the highest decolorization activity. The optimum pH value for decolorization was determined as 8 for all the dyes tested. In the experiment with pH 8 dye decolorizations by mixed cultures were investigated at about 96.2–1031.3 mg l−1 initial dye concentrations. The highest dye removal rates of mixed cultures were 94.9% for Reactive Red RB, 91.0% for Reactive Black B and 63.6% for Remazol Blue at 953.2, 864.9 and 1031.3 mg l−1 initial dye concentrations respectively within 24 h incubation period. When the Reactive Red RB was used, approximately 82–98% total color removal was obtained at between 96.2 and 953.2 mg l−1 initial dye concentrations after 12 h of incubation at 35 °C. These results show that our enriched mixed cultures have the potential to serve as an excellent biomass for the use in reactive dye removal from wastewaters under anaerobic conditions.  相似文献

By using a yeast functional complementation assay, we have identified AtTDX, a new Arabidopsis thaliana gene, encoding a two-domain 42-kDa protein. The amino-terminal domain of AtTDX is closely related to the co-chaperone Hsp70-interacting protein HIP, whereas its carboxyl-terminal part contains a thioredoxin domain. Both in vivo and in vitro assays showed that AtTDX is a protein-disulfide reductase. We next found that the HIP domain of AtTDX is capable of interacting with the ATPase domain of Ssb2, a yeast heat-shock protein 70 chaperone. Strikingly, the AtTDX-Ssb2 interaction can be released under oxidative stress, a redox-dependent regulation involving the thioredoxin activity of AtTDX. A mutation inactivating the cysteine 20 of the ATPase domain of Ssb2 was found to stabilize the AtTDX-Ssb2 interaction that becomes redox-insensitive. As cysteine 20 is conserved in virtually all the Hsp70 chaperones, our results suggest that this residue might be more generally the target of redox regulations of chaperone binding activity.  相似文献

Tests were made to determine the effects of inorganic and organic sulfur sources on the degradation of cellulose to methane in a chemically defined medium with sulfur-poor inoculum prepared from sewage sludge. The results show that a sulfur source of about a 0.85 mM concentration is essential for the degradation of cellulose to CH4. However, the production of CH4 from CO2 and H2 provided in the headspace occurred with 0.1 mM sulfate or sulfide. At a 9 mM concentration, all inorganic sulfur compounds other than sulfate inhibited both cellulose degradation and methane formation, and this inhibition increased in the order thiosulfate less than sulfite less than sulfide less than H2S. It appears that the degradation of cellulose to CH4 in a sulfate-free medium by inoculum maintained in a low-sulfur medium is inhibited because of the lack of availability of sulfur for growth of bacteria and synthesis of cell materials and sulfur-containing cofactors involved in cellulose degradation and methanogenesis. The reduction of methanogenesis by higher levels of sulfate probably occurs as a result of stimulation of reactions converting acetate and H2 to end products other than CH4.  相似文献

A completely mixed anaerobic filter, in which the influent organic matter concentration is diluted with recirculated effluent, was found to effectively remove organic matter concentrations in high strength acidic wastewater, at a range of organic loadings and shock loads. Increases in organic loading had a substantial effect on the relative organic matter composition of the effluent and the magnitude of the fatty acid fraction showed a pattern inverse to that of the nitrogenous organics. A fixed film model was formulated which indicated that at high substrate concentrations the substrate removal rate is proportional to the square root of the substrate concentration and the specific area of the filter medium. A comparison of the biofilm model and the measured effluent concentrations tended to indicate that the substrate removal rate is primarily affected by substrate concentration, specific surface area, flow rate, and temperature of the unit.  相似文献
