As a crucial member of the Hedgehog (Hh) protein family, desert hedgehog (dhh) plays a vital role in multiple developmental processes, cell differentiation and tissue homeostasis. However, it is unclear how it regulates development in fish. In this study, we cloned and characterized the dhh gene from Pseudopleuronectes yokohamae. The full-length cDNA of Pydhh comprises 3194 bp, with a 1386 bp open reading frame (ORF) that encodes a polypeptide of 461 amino acids with a typical HH-signal domain, Hint-N and Hint-C domains. Multiple sequence alignment revealed that the putative PyDHH protein sequence was highly conserved across species, especially in the typical domains. Phylogenetic analysis showed that the PyDHH clustered within the Pleuronectiformes. Real-time quantitative PCR showed that Pydhh was detected in fourteen different tissues in adult-female and adult-male marbled flounder, and nine different tissues in juvenile fish. During early embryonic development stages, the expression of Pydhh was revealed high levels at hatching stage of embryo development. Moreover, the relative expression of Pydhh was significantly higher in the juvenile liver than adults’, and higher in the female skin than the male skin. To further investigate its location, the in situ hybridization (ISH) assay was performed, the results showed that the hybridization signal was obviously expressed in the immune organs of Pseudopleuronectes yokohamae, with weak signal expression in the other tissues. Our results suggested that Pydhh is highly conserved among species and plays a vital role in embryonic development and formation of immune related organs.  相似文献

为了解星斑川鲽(Starry Flounder,Platichthys stellatus)脑组织基因表达与卵巢发育调控的关系,发掘相关功能基因,研究提取卵巢成熟期和退化期星斑川鲽雌鱼脑垂体和下丘脑组织总RNA,运用HiSeq 2000高通量测序技术进行转录组测序分析。测序结果经拼接组装后共获得30640条Unigenes,Blast同源性比对显示,其中24128条Unigenes获得注释;经eggNog功能注释后29137条Unigenes序列分为26类,分别涉及信号转导、翻译机制等生理生化过程。KEGG pathway数据比对显示,卵巢成熟期涉及98种代谢途径,135条序列表达上调;卵巢退化期涉及192种代谢途径,648条序列表达上调。Unigenes表达量及表达差异分析表明在卵巢成熟期334条序列表达上调,卵巢退化期987条序列表达上调。获得功能注释的Unigene中,408条涉及生殖调控和内分泌调控,参与生殖调控的信号分子有1508条。试验采用实时定量PCR研究了涉及生殖调控和内分泌调控基因促性腺激素释放激素(GnRH)、神经激肽B(NKB)、促性腺激素(GtH)、促滤泡激素(FSH)、肿瘤转移抑制因子(Kiss)以及催乳素释放肽受体(PrRPR)在卵巢两个不同发育时期脑垂体和下丘脑的表达情况,结果表明除FSH外,其余均在卵巢退化期时期脑组织中表达升高,与转录组测序结果趋势一致。  相似文献

This paper aims to assess the diel feeding pattern and seasonal variation in the daily ration of immature and mature marbled flounder (Pseudopleuronectes yokohamae). A day‐night collection by bottom trawls was conducted in Sendai Bay in July 2014. Marbled flounder (131–493 mm total length; Number of collected individuals = 1830, Number of analyzed individuals = 497) fed mainly on polychaetes during the day. At night, stomach content weight decreased with time, but the weight and proportion of bivalve siphons were consistently higher at night than during the day, suggesting nocturnal feeding by the flounder on bivalve siphons. Daily ration was greater in females (<300 mm: 2.6%–3.2% body weight; ≥300 mm: 1.5%–2.5%) than in males (<300 mm: 1.7%–2.6%; ≥300 mm: 1.3%–1.9%). Seasonal surveys were also carried out, and the greater ration in females than males were consistent throughout the year, suggesting that greater growth in females than males attributes to the greater food intake of females. The ration was highest in June, especially for large individuals, although water temperature in June was lower than that in September. These results indicate that the amount of food intake is related to the annual life cycle of the marbled flounder.  相似文献

Although Streptococcus parauberis is the major bacterial pathogen affecting olive flounder, the translocation and dissemination of this pathogen in infected fish are not well understood. Therefore, we conducted real-time PCR and histopathologic examination to monitor the intensity of infection in multiple organs of the olive flounder after challenge with S. parauberis through subcutaneous injection. The bacterial burden in the fish kidney, when sampled at 0, 3, and 7 dpc, was 0, 6.2?±?4.5?×?105, and 6.7?±?5.5?×?106 CFU/100 mg of tissue, respectively, indicating that the infection progressed rapidly over time. Of the ten different tissues sampled, the heart and the brain were the major target organs of S. parauberis based on highest copy number as detected by our modified real-time PCR method. Histopathologic examination also showed that S. parauberis caused severe inflammation accompanied by leucocyte infiltration, connective tissue expansion, and a loss of cardiomyocytes in the brain and heart of fish sampled at dpc 7. However, the number of S. parauberis-positive fish at 3 dpc was much higher in the spleen (6/8 fish) than in the remaining organs, suggesting that the spleen is targeted in the early stages of infection relative to the heart (2/8 fish) or brain (3/8 fish). This study provides essential information for studies to find treatments for the effective elimination of S. parauberis in target organs (i.e., the brain and heart) of olive flounder.  相似文献

Olive flounder (Paralichthys olivaceus) is regarded as one of economical fish species in the world. Genome information of Olive flounder has been revealed by next generation sequencing. Endogenous retroviruses (ERVs) as a member of transposable elements have been involved in functional roles of various genes in variety organisms. In this study, we identified an Olive flounder ERV (OF-ERV5) using RepeatMasker program, and examined expression pattern of pol gene of OF-ERV5, which indicated the high expression in two kidney samples (head kidney and body kidney). In addition, the 5′LTR sequences of OF-ERV5 are cloned into pGL4.11 vectors to confirm promoter activity. Luciferase assay indicated that the OF-ERV5_LTR showed promoter activity in both HepG2 and HINAE cell lines. These data could be of great use for further study to understand biological function of transposable elements in Oliver flounder.  相似文献
