Male Drosophila fruit flies acquire and defend territories in order to attract females for reproduction. Both, male-directed agonistic behavior and female-directed courtship consist of series of recurrent stereotypical components. Various studies demonstrated the importance of species-specific sound patterns generated by wing vibration as being critical for male courtship success. In this study we analyzed the patterns and importance of sound signals generated during agonistic interactions of male Drosophila melanogaster. In contrast to acoustic courtship signals that consist of sine and pulse patterns and are generated by one extended wing, agonistic signals lack sine-like components and are generally produced by simultaneous movements of both wings. Though intra-pulse oscillation frequencies (carrier frequency) are identical, inter-pulse intervals are twice as long and more variable in aggression signals than in courtship songs, where their precise temporal pattern serves species recognition. Acoustic signals accompany male agonistic interactions over their entire course but occur particularly often after tapping behavior which is a major way to identify the gender of the interaction partner. Since similar wing movements may either be silent or generate sound and wing movements with sound have a greater impact on the subsequent behavior of a receiver, sound producing wing movements seem to be generated intentionally to serve as a specific signal during fruit fly agonistic encounters.  相似文献

The innate sexual behaviors of Drosophila melanogaster males are an attractive system for elucidating how complex behavior patterns are generated. The potential for male sexual behavior in D. melanogaster is specified by the fruitless (fru) and doublesex (dsx) sex regulatory genes. We used the temperature-sensitive activator dTRPA1 to probe the roles of fru(M)- and dsx-expressing neurons in male courtship behaviors. Almost all steps of courtship, from courtship song to ejaculation, can be induced at very high levels through activation of either all fru(M) or all dsx neurons in solitary males. Detailed characterizations reveal different roles for fru(M) and dsx in male courtship. Surprisingly, the system for mate discrimination still works well when all dsx neurons are activated, but is impaired when all fru(M) neurons are activated. Most strikingly, we provide evidence for a fru(M)-independent courtship pathway that is primarily vision dependent.  相似文献

Male mating behavior is an important component of fitness in Drosophila and displays segregating variation in natural populations. However, we know very little about the genes affecting naturally occurring variation in mating behavior, their effects, or their interactions. Here, we have mapped quantitative trait loci (QTL) affecting courtship occurrence, courtship latency, copulation occurrence, and copulation latency that segregate between a D. melanogaster strain selected for reduced male mating propensity (2b) and a standard wild-type strain (Oregon-R). Mating behavior was assessed in a population of 98 recombinant inbred lines derived from these two strains and QTL affecting mating behavior were mapped using composite interval mapping. We found four QTL affecting male mating behavior at cytological locations 1A;3E, 57C;57F, 72A;85F, and 96F;99A. We used deficiency complementation mapping to map the autosomal QTL with much higher resolution to five QTL at 56F5;56F8, 56F9;57A3, 70E1;71F4, 78C5;79A1, and 96F1;97B1. Quantitative complementation tests performed for 45 positional candidate genes within these intervals revealed 7 genes that failed to complement the QTL: eagle, 18 wheeler, Enhancer of split, Polycomb, spermatocyte arrest, l(2)05510, and l(2)k02206. None of these genes have been previously implicated in mating behavior, demonstrating that quantitative analysis of subtle variants can reveal novel pleiotropic effects of key developmental loci on behavior.  相似文献

Corrolations between female rejection behaviors and male wing display were calculated for both Drosophila simulans and Drosophila melanogaster intraspicific pair-matings. No significant correlations were found for D. melanogaster, but in D. simulans flicking by the female appeared to be associated with a shift in male wing display pattern resulting in higher levels of vibration. Flicking did not appear to discourage courtship by males in either species.  相似文献

The Y chromosome in Drosophila melanogaster is composed of highly repetitive sequences and is essential only in the male germ line. We employed P-element insertional mutagenesis to induce male sterile mutations in the Y chromosome. By using a combination of two modifiers of position effect variegation, adding an extra Y chromosome and increasing temperature, we isolated 61 P(ry+) elements in the Y chromosome. Six of these Y-linked insertions (approximately 10%) induced male sterile mutations that are mapped to two genes on the long and one on the short arms of the Y chromosome. These mutations are revertible to the wild type in a cell-autonomous and germ-line-dependent manner, consistent with previously defined Y-linked gene functions. Phenotypes associated with these P-induced mutations are similar to those resulting from deletions of the Y chromosome regions corresponding to the male fertility genes. Three alleles of the kl-3 gene on the Y long arm result in loss of the axonemal outer dynein arms in the spermatid tail, while three ks-2 alleles on the Y short arm induce defects at early postmeiotic stages. The recovery of the ms(Y) mutations induced by single P-element insertions will facilitate our effort to understand the structural and functional properties of the Y chromosome.  相似文献

Changes in the genetic structure of the Katsunuma natural population of Drosophila melanogaster have been examined during the past 35 years. The frequency of recessive lethal genes on the second chromosome once increased from 15% to 30% in the early 1970s, then decreased to about 24% in the late 1970s, and thereafter showed no significant changes. Sterility genes, the frequency of which is always less than the lethals, showed a similar tendency. The SD (segregation distorter) mutant gene disappeared but some others such as rbl (reduced bristle) and bw (brown) persisted in the population. The frequency of inversion-carrying chromosomes gradually decreased in the period, such that the standard chromosome frequency in the second and third chromosomes increased from about 40% to more than 80%. Coincident with these frequency changes is the invasion of a transposable element P into the Katsunuma population. The P element should have invaded into Katsunuma in the late 1960s. It spread over the population apparently inducing deleterious mutations, causing the decrease in the allelism rate, and hence increasing the effective population size. Soon, however, most flies became resistant to the P element-mediated transposition as they began to harbor defective P elements. During the course of spreading, the P element must also have induced deleterious mutations on the polymorphic inversions, breaking up the heterotic gene complexes along the chromosomes, which probably caused the reduction in the frequency of inversion chromosomes. Temporal invasion of D. simulans, a sibling species of D. melanogaster, into Katsunuma occurred several times after 1978, and the species seems to have been settled since 1990. This, however, did not have any effect on the genetic structure of D. melanogaster population.  相似文献

Spontaneous meiotic recombination events do not normally occur in the male germ line of Drosophila melanogaster. However, such events are induced in males when a P transposable element or a source of P element encoded transposase protein is present in its genome. This report concerns a molecular analysis of the meiotic exchanges that were induced in the male Drosophila by P elements within a genetically marked region of the third chromosome. The marked region also harbors a single P-element called P(lArB). Fifty-six percent of the P(lArB) region crossovers indicated some alterations in the P element 5' fragment. Such alterations appear to be related to asymmetric or unequal genetic exchanges. Finally, P(lArB) excision was found to be independent of P(lArB) region crossover events.  相似文献

A total of 1020 B s Yy +chromosomes was screened for the induction of male sterile mutations by X irradiation. The 29 recovered mutations were analyzed by genetic complementation and the metaphase chromosomes stained with Hoechst 33258 and observed with fluorescence microscopy. The cytological and genetic maps derived from this analysis were compared to similar maps of the Y chromosome mutations isolated in an earlier study (Brosseau, 1960). Unlike the previous work we have identified only 6 male fertility loci (2 on the short arm, 4 on the long arm) on the Y chromosome. These loci are distributed along the length of the long arm and are likely to reside at two separate sites on the short arm. There is no apparent clustering of these fertility factors in this heterochromatic chromosome. The deletions obtained in this study were observed to be unstable and the nature of this instability was investigated. The original Y chromosome was marked at both telomeres with normally X-linked genes. The loss of one or the other of these markers was accompanied in many cases by the concomitant loss of large segments of Y chromosome material. The analysis of inter-strain heterochromatin polymorphism in mitotic chromosomes of Drosophila melanogaster was extended to some stocks characterized by chromosomal mutations. In particular, the present investigation aims to compare, in the same cell, the quinacrine banding of two different Y chromosomes of male hybrids derived from crosses using special stocks. A direct comparison of homologous heteromorphic chromosomes in F1 hybrids provided additional evidence of differences in the fluorescence pattern of the Y chromosome, as well as in the length of the heterochromatin segment of the X chromosome.  相似文献

Male meiotic sterile mutations were selected among X-linked male—steriles by detection of micronuclei in early spermatids. Despite severe defects in the 1st or 2nd meiotic spindles in all mutants, no effect on mitosis was observed. Various features of spindle structure, chromosome segregation, and centriole movements were compared in seven meiotic steriles and in XO males. Chromosome behaviour and centriole movement were always affected concomitantly, and were both shown to be genetically independent of centre formation in the meiotic spindles. Precocious and delayed centromere separation was observed in the various mutants in both divisions, and similarly attributed to basic spindle lesions rather than chromosome defects. Attachment of the centriole body to the membrane of the spermatid nucleus was normal only in mutants where second division nuclei were formed. The role of the centriole body was shown to be independent of membrane attachment.—The phenomena observed in this study were discussed mainly with regard to genetic interdependence of morphogenetic processes during male meiosis. A common base for the pleiotropic defects of meiotic steriles and XO males is suggested, and the genetic control of meiosis is re-evaluated in the light of comparison with fertile meiotic mutants.  相似文献

The osmometric behavior of Drosophila melanogaster embryos in permeabilized eggs was studied in a microscope diffusion chamber designed to impose a rapid change in osmotic environment at various temperatures. A numerical model of NaCl diffusion in the chamber predicted that radial variations in concentration arising from the presence of a thin film of solution at the top of the chamber were negligible. On the basis of transient electrical conductance measurements in the chamber, characteristic time constants for the change in concentration averaged over the chamber depth occupied by the eggs were 0.99, 0.77, and 0.60 min at 0, 10, and 20 degrees C, respectively. The chamber response was sufficiently rapid that the characteristic response of the embryo was not masked. Equilibrium volumetric behavior of the embryos indicated that they behaved as nearly ideal osmometers over the range of 0.256 to 2.000 osm, and followed the relation FVeq = 0.123C-1 + 0.541, where FVeq is equilibrium fractional volume and C is osmolality. Nonlinear regression of volumetric data during osmotic contraction yielded an average Lp of 0.722 micron/(min.atm) at 20 degrees C and an apparent activation energy delta E of 8.11 kcal/mol. The coefficients of variation in the Lp estimates among individual embryos were 38, 18, and 47% at 0, 10, and 20 degrees C, respectively. With the use of probability rules and a model for volumetric behavior during freezing, it was determined that the observed variability in Lp (assuming delta E is fixed) considerably broadens the transition range of cooling rates over which the predicted probability of intracellular ice formation goes from 0 to 1. However, experimental observations (21) show the actual transition range is even wider, indicating that there exist other important sources of variability which determine the event of ice formation in D. melanogaster embryos.  相似文献

Male courtship ritual is among the main behavioral characteristics of Drosophila. This is a complex, genetically determined process consisting of four general stages: orientation, vibration, licking, and attempts at copulation (or successful copulation). Several genes are known that control some stages of this behavior. Most of them have pleiotropic effects and are involved in other biological processes. Earlier, we have shown that a mutation in locus flamenco (20A1-3), which controls transposition and infectivity of retrotransposon gypsy (MDG4), is involved in the genetic control of behavior. In strains mutant for this locus, the male mating activity is decreased and the structure of courtship ritual is changed. To understand the mechanisms of these changes, it is important to study all behavioral stages in genetically identical strains. For this purpose, the normal allele of gene flamenco from the X chromosome of the wild-type strain (stock) Canton S was introduced into strain SS carrying flamMS. This offers new opportunities in studying the role of gene flamenco in the control of mating behavior in Drosophila.  相似文献

In many species, males have the capacity to directly influence (either positively or negatively) the fitness of their mates and offspring, not only via parental care contributions and/or precopulatory resource provisioning, but also via the post‐copulatory activity of those substances passed on to their mates in their ejaculates. Here, we examine how an individual male's identity may be related to phenotypic variation in short‐term female fecundity in the model species, Drosophila melanogaster. The effect of male identity on short‐term fecundity stimulation of females was repeatable across time and accounted for over a fifth of the total observed phenotypic variation in fecundity in two independent populations. The functional explanations for these results and the implications for our understanding of the factors that contribute to the adaptive significance of mating preferences and/or sexual conflict are discussed.  相似文献

The use of Drosophila as an organism in which to study aging has been limited by the fact that few biomarkers of aging exist in the adult. In this paper we examine behavior loss relative to longevity in wild-type populations maintained at 22°C and 29°C to determine whether behavior loss—that is, loss of ability to perform certain innate behavioral responses within a defined test interval—can be used as biomarkers of aging. We find that under controlled conditions behavior loss can be used as a landmark of aging in populations maintained at either 22°C or 29°C. The ability to perform normal geotactic and phototactic responses is lost during the reproductive phase of the adult populations, whereas motor activity is not lost until well into the death phase. We feel that the use of behavior loss, together with other parameters of longevity in Drosophila, will allow comparisons to be made between different strains or between different environmental conditions to test their effect on aging. In the companion paper we demonstrate the use of behavior loss to identify a mutation which may accelerate the aging process.  相似文献
