Pineal glands of male hamsters 8 weeks after removal of both eyes or both superior cervical ganglia and those of untreated animals were studied by electron microscopy. In the blinded hamsters the reproductive organs were remarkably involuted, whereas the pinealocytes enlarged and were characterized by a tremendous hypertrophy of the smooth-surfaced endoplasmic reticulum, in the mesh of which some dense cored vesicles were distributed. In constrast the pinealocytes of ganglionectomized hamsters atrophied and were noted by a large number of lysosomes and sparsity of the agranular reticulum, the testes being significantly larger than the controls. The findings were interpreted to be compatible with the view that the pineal capability of producing antigonadal substance is augmented by blinding and is suppressed by cervical ganglionectomy due to the impairment of normal functioning of the pineal by denervation. It is assumed that the degree of development of the smooth endoplasmic reticulum in the pinealocyte is parallel to, and reflects that of the pineal activity. Probably the membranous organelle is involved in the elaboration of the pineal antigonadal substance.  相似文献

Summary Semiquantitative immunocytochemistry by immuno-gold techniques revealed differences in the spatial distribution of glutamate, glutamine, and taurine within the pineal gland, with greatest labeling over pinealocytes, glia, and endothelia, respectively. At the subcellular level, glutamate labeling tended to be highest over pinealocyte synaptic ribbons and mitochondria, and lowest over lipid inclusions. Pineal levels of glutamate, glutamine and taurine, as measured by high performance liquid chromatography, did not vary over a light: dark cycle. Superior cervical sympathetic denervation, which abolishes pineal melatonin synthesis, resulted in a nearly 50% reduction in pineal glutamate levels, but had no effect on levels of glutamine and taurine. Other amino acids (alanine, arginine, aspartate, serine) were reduced by 23%–33% following sympathectomy. These data suggest an important role for glutamate in pinealocyte function(s) possibly related to the noradrenergic innervation of the gland.  相似文献

Female rats were subjected to superior cervical ganglionectomy (Gx), blinding and anosmia (BAs) or combined procedures (BAsGx). Onset and growth of dimethylbenz(a)anthracene (DBMA)-induced mammary tumors was studied in these animals and compared to tumorigenesis in intact control rats. Carcinostatic effects were present in all surgically altered animals, as evidenced by a trend toward reduced tumor incidence, reduced final tumor mass, and a significant reduction in mean number of tumors in Gx and BAsGx rats, and increased regression of tumors in BAs rats compared to intact group. Reduced tumorigenesis was paralleled by a trend toward either an increase (BAs) or a decrease (Gx and BAsGx) in the activity of pineal hydroxyindole-O-methyltransferase (HIOMT) compared to intact group. In addition, BAs and BAsGx animals showed a significant reduction in body weight. These results suggest that Gx reduces mammary tumorigenesis in both sighted and BAs rats. They further confirm the findings of others on reduced mammary tumorigenesis in BAs rats. Possible involvement of multiple carcinostatic mechanisms in different animal models is discussed.  相似文献

In male rats under anaesthesia, dialysis of the venous blood from sella turcica region was carried out. Vasopressin and oxytocin content was determined in the dialysates by radioimmunoassay. The obtained results indicate that: 1. Electrical stimulation of the superior cervical ganglion causes an increase in vasopressin and oxytocin release. 2. 20 days after superior cervical ganglionectomy the vasopressin and oxytocin release increased. 3. Superior cervical ganglionectomy immediately before the dialysis evoked a several times increase in vasopressin and oxytocin release. 4. Application of alpha1-blocker, prazosin, as well beta-blocker, propranolol, has partially prevented the increase in vasopressin release which was found immediately after superior cervical ganglionectomy. 5. Contrary to vasopressin, the increase in oxytocin release after superior cervical ganglionectomy is completely prevented by the beta-blocker, propranolol, and only partially by the alpha1-blocker, prazosin.  相似文献

The course of regeneration of aminergic nerves in rat cerebral arteries was studied by means of histochemical methods, after uni- or bilateral cervical sympathectomy. Degeneration of aminergic nerves started on day 1 and was complete between days 3 and 7 after surgery. Between weeks 4 and 6, regenerating nerves started to appear from the proximal internal carotid artery. Regenerated aminergic nerve fibres were generally unbeaded and intensity of fluorescence was weak. The circular nerves appeared earlier than the longitudinal ones. The number of regenerating nerves reached the maximum, between months 9 and 12, at about half the normal level. AChE activity of the cerebral arteries showed no significant changes at any stage.  相似文献

The distributions of noradrenaline and dopamine in subcellular fractions of bovine superior cervical ganglia were measured fluorimetrically and were compared with that of acetylcholine. Results indicate that the crude synaptosomal pellet (P2), which contained the bulk of the bound acetylcholine, was not seriously contaminated with catecholamines. The microsomal fraction showed the highest concentration of noradrenaline relative to protein content, while dopamine was richest in P2, possibly due to formation of synaptosomes from nerve endings of the dopaminergic interneurones which have been described in this tissue.  相似文献

Adult male Long Evans and Sprague-Dawley rats and Mongolian gerbils were sacrificed at 4 hour intervals over a 24 hour period. Pineal noradrenaline (NA) levels in these animals were determined by a radioisotopic procedure. Statistically significant circadian rhythms were observed in pineal NA content in both strains of rats but not in the gerbils. These data show that daily rhythms in pineal NA are a characteristic phenomenon observed in rats either with or without eye pigment. The absence of a daily rhythm in NA content in the gerbil pineal may explain the relatively low amplitude rhythm in N-acetyl transferase activity observed in this species compared to the rat.  相似文献

The endogenous γ-aminobutyric acid (GABA) content of the rat pineal gland and superior cervical ganglion (SCG) was measured by high pressure liquid chromatography. It was found that GABA levels in both tissues increased after decapitation of the animals. The GABA content of tissues frozen within 20 seconds after decapitation was the same as that of tissues removed from animals killed by microwave irradiation. Amino-oxyacetic acid, a GABA-transaminase inhibitor, increased the endogenous GABA content of both of these tissues. Dimethylphenylpiperizinium or isoniazid administration did not alter GABA levels in these tissues. Isoproterenol increased the GABA content of the SCG but did not change the pineal gland GABA levels. The ability of the pineal gland to take up and accumulate 3H-GABA was significantly reduced in rats that had been ganglionectomized. A fluctuation in endogenous GABA levels in the pineal gland was seen to occur when measures were taken at different times of the day. These results tend to suggest that GABA may have some functional role in the pineal gland and the superior cervical ganglion.  相似文献

Summary From studies conducted with the pineal organ of the mouse, it was ascertained that for the in vitro investigation of secretory processes (synthesis and release) of proteic/peptidic compound(s), a culture time of 5 to 14 days is optimal. A 5-day organ culture was therefore chosen to study the effects of noradrenaline on these secretory processes.Addition of noradrenaline to the culture medium provokes, in pineal explants of the normal mouse and the eyeless mouse, an inhibition of the secretory process, characterized by the formation of granular vesicles. In the hamster and rat, however, opposite results were obtained. Moreover, it appears that noradrenaline, at least in the rat, may also be involved in the regulation of the ependymal-like secretory process.The present results indicate clearly that noradrenaline (thus, the sympathetic innervation) is implicated in the regulation of the production of proteic/peptidic hormonal agents, but that the effect of this neurotransmitter is species-specific. The effect of sympathetic denervation (bilateral superior cervical ganglionectomy) on several aspects of pituitary adrenal function was studied in 250-300g rats. Group I=Sham-op, Group II=superior cervical ganglionectomy. (1) There was no significant difference between these groups in the plasma corticosterone (B) response to ether or immobilization stress or to ACTH administration. (2) Circadian periodicity of plasma B was determined by sequential (q.4h) tail vein sampling under LD (lights on 0800) off 2000) and under DL (lights on 2000, off 0800). Both groups showed similar peak and trough concentrations. Phase shifting occurred in both groups at day 11. (3) Compensatory adrenal hypertrophy of similar magnitude was present in both groups 10 days following unilateral adrenalectomy. These studies indicate that sympathetic input from the superior cervical ganglia is not involved in stress induced, circadian or some feedback aspects of pituitary adrenal function.  相似文献

The effect of norepinephrine and acetylcholine on the 32P incorporation into phospholipids of normal and sympathetically denervated rabbit iris muscle was investigated. (1) In the absence of exogenously added neurotransmitters sympathetic denervation exerted little effect on the incorporation of 32P into the phospholipids of the excised iris muscle. In vivo thr iris muscle incorporated 32P into phosphatidylinositol, phosphatidic acid, phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine and sphingomyelin in that order of activity while in vitro phosphatidylinositol was followed by phosphatidylcholine. (2) Tension responses of iris dilator muscle from denervated irises exhibited supersensitivity to norepinephrine. Furthermore, norepinephrine at concentrations of 3 μM and 30 μM produced 1.6 times and 3 times stimulation of the phosphatidic acid of the denervated muscle respectively. In contrast at 30 μM it stimulated this phospholipid by 1.6 times in the normal muscle. This stimulation was completely blocked by phentolamine. (3) While in the normal muscle acetylcholine stimulated the labelling of phosphatidic acid and phosphatidylinositol by more than 2 times, in the denervated muscle it only stimulated 1.4 to 1.7 times. (4) Similarly when 32Pi was administered intracamerally, the labelling found in the various phospholipids of the denervated iris was significantly lower than that of the normal. (5) It was concluded that denervation decreases the 32P labelling in the presence of acetylcholine. (6) The norepinephrine-stimulated 32P incorporation into phosphatidic acid appears to be post-synaptic.  相似文献

Summary Extirpation of the superior cervical ganglion was performed in a series of Mongolian gerbils. One or two weeks after the ganglionectomy the animals were injected with a monoamine oxidase inhibitor. Subsequently perfusion fixation was performed using the glyoxylic acid-paraformal-dehydemagnesium method (Lorén et al., 1976) for fluorescence histochemical investigation of the monoamines of the pineal complex. In the ganglionectomized animals all of the blue-fluorescent sympathetic fibers in the pineal complex (superficial pineal gland, deep pineal gland and the pineal stalk) completely disappeared. The yellow indolamine fluorescence of the cells in the superficial pineal and the deep pineal, as well as in the pineal stalk, was markedly reduced after ganglionectomy. No change in the morphology or number of sympathetic fibers in the medial habenular nucleus was observed. These results indicate that the presence of sympathetic nerve fibers with perikarya in the superior cervical ganglion is necessary for maintaining a high indolamine content in all three parts of the pineal complex. In addition, the results also indicate that the deep pineal gland is a functional part of the pineal complex. In order to investigate a possible functional relationship between the submandibular salivary gland (SSG) and the central nervous system (CNS), we have extirpated the salivary organs from thirty male rats. Twenty days after ablation both the pineal glands and the cervical superior ganglions (CSG) were dissected, homogenized and frozen until AMPc and TOH were assayed respectively. We observed a significant decrease in pineal AMPc (53.9 +/- 6.2 vs 76.1 +/- 7.6% of maximum value; p less than 0.02) which seems to be linked with a significant drop in TOH activity measured at CSG level (1.5 +/- 0.6 vs 3.7 +/- 0.9 nmoles of DOPA/h/pair GCS; p less than 0.03). Our results suggest that both findings might be due to the lack of NGF normally reaching the CSG from SSG. This data reinforces the idea of a functional link between SSG and CNS via the pineal gland.  相似文献
