Saussurea involucrata (Asteraceae) is a medicinal and second-degree national priority endangered plant that is mainly distributed in the high latitude region of the western Tianshan Mountains. The population is fragmented and isolated, and extensive human impact merits a suitable and specific conservation strategy, which can be compiled based on the genetic diversity, population structure, and demographic history. Phylogeographic studies were conducted on a total of five natural populations and 150 individuals were sampled. Data from three cpDNA intergenic spacer regions (trnL-F, matK, and ndhF-rpl32) and nrDNA ITS sequences showed that twelve haplotypes in cpDNA and five haplotypes in nrDNA indicated high genetic diversity among populations sampled (H T?=?0.820 and 0.756) and within populations sampled (H S?=?0.792 and 0.721). Additionally, the high genetic diversity did not mirror genetic structure in either cpDNA (F ST?=?0.03153, G ST?>?N ST, p?<?0.05) or nrDNA (F ST?=?0.03666, meaningless G ST and N ST). Two groups (north and south) were determined for a SAMOVA analysis. Based on this analysis, the demographic history was conducted with a Bayesian Skyline Plot and Isolation with Migration analysis, which showed sustainable and stable extension without a marked bottleneck. Divergence time was indicated at c. 6.25 Mya (90%HPD: 15.30–0.22 Mya) in the Miocene, which is consistent with the formation of the Kelasu section of Tianshan. The southern populations in the Bayanbulak and Gonglu regions require additional attention and transplanting would be an effective way to restore rare cpDNA haplotypes, increase effective population size, and migration rate. Our results suggested that in situ conservation of S. involucrata in western Tianshan should be the main strategy for protection and recovery of the species.  相似文献

Understanding genetic connectivity is fundamental for ecosystem-based management of marine resources. Here we investigate the metapopulation structure of the edible sea cucumber Holothuria edulis Lesson, 1830 across Okinawa Island, Japan. This species is of economic and ecological importance and is distributed from the Red Sea to Hawai‘i. We examined sequence variation in fragments of mitochondrial cytochrome oxidase subunit I (COI) and 16S ribosomal RNA (16S), and nuclear histone (H3) at six locations across Okinawa Island. We found higher haplotype diversity for mtDNA (COI: Hd = 0.69 and 16S: Hd = 0.67) and higher heterozygosity of nDNA (H3: H E = 0.39) in populations from the west coast of Okinawa compared to individuals from populations on the east coast (COI: Hd = 0.40; 16S: Hd = 0.21; H3: H E = 0.14). Overall population structure was significant (AMOVA results for COI: Φ ST = 0.49, P < 0.0001; 16S: Φ ST = 0.34, P < 0.0001; H3: Φ ST = 0.12, P < 0.0001). One population in the east, Uruma, showed elevated pairwise Φ ST values in comparisons with all other sites and a marked reduction of genetic diversity (COI: Hd = 0.25 and 16S: Hd = 0.24), possibly as a consequence of a shift to a more dominant asexual reproduction mode. Recent reports have indicated that coastal development in this area influences many marine organisms, and ecosystem degradation in this location could cause the observed decrease of genetic diversity and isolation of H. edulis in Uruma. Our study should provide valuable data to help with the urgently needed management of sea cucumber populations in Okinawa, and indicates particular attention needs to be paid to vulnerable locations.  相似文献

Pelagic larval dispersal habits influence the population genetic structure of marine mollusk organisms via gene flow. The genetic information of the clam Gomphina aequilatera (short larval stage, 10 days) which is ecologically and economically important in the China coast is unknown. To determine the influence of planktonic larval duration on the genetic structure of G. aequilatera. Mitochondrial markers, cytochrome oxidase subunit i (COI) and 12S ribosomal RNA (12S rRNA), were used to investigate the population structure of wild G. aequilatera specimens from four China Sea coastal locations (Zhoushan, Nanji Island, Zhangpu and Beihai). Partial COI (685 bp) and 12S rRNA (350 bp) sequences were determined. High level and significant FST values were obtained among the different localities, based on either COI (FST?=?0.100–0.444, P?<?0.05) or 12S rRNA (FST?=?0.193–0.742, P?<?0.05), indicating a high degree of genetic differentiation among the populations. The pairwise Nm between Beihai and Zhoushan for COI was 0.626 and the other four pairwise Nm values were >?1, indicating extensive gene flow among them. The 12S rRNA showed the same pattern. AMOVA test results for COI and 12S rRNA indicated major genetic variation within the populations: 77.96% within and 22.04% among the populations for COI, 55.73% within and 44.27% among the populations for 12S rRNA. A median-joining network suggested obvious genetic differentiation between the Zhoushan and Beihai populations. This study revealed the extant population genetic structure of G. aequilatera and showed a strong population structure in a species with a short planktonic larval stage.  相似文献

The estimates of genetic diversity in populations of chum salmon Oncorhynchus keta from different regions of Sakhalin Island, Iturup Island, and the Anadyr’ River were obtained on the basis of analysis of allozyme variability. These estimates together with our published earlier data on chum salmon from the Amur River basin and the rivers of the northern coast of the Sea of Okhotsk demonstrate pronounced regional genetic differentiation in the Asian part of the fish range. The intraregional level of interpopulation genetic diversity was maximum on Sakhalin Island (G ST = 6.6%) and was small on Iturup Island (G ST = 0.9%) and the northern coast of the Sea of Okhotsk (G ST = 0.6%). Interpopulation genetic diversity of Sakhalin chum salmon was almost commensurable to the diversity of the whole pool of studied Asian populations (G ST = 7.6%) and would be presented more completely in baselines assigned for genetic identification of mixed stocks. It was demonstrated that the character and degree of genetic differentiation between populations of chum salmon from the main hatcheries situated in different regions of the Sakhalin oblast and connected to one another by numerous transplantations of fertilized eggs did not change significantly during an approximately 20-year period of our observation, and this fact suggests low efficiency of such transplantations.  相似文献

Knowledge of the influence of both spatial and temporal environmental gradients on life history traits and population demographics is a critical requirement in the management of exploited fish populations. This study examined variation in the demographics of Arripis trutta, an economically-important pelagic fish species with a broad latitudinal distribution in the waters of coastal south-eastern (SE) Australia, a region dominated by the influence of the East Australian Current (EAC). A validated ageing protocol was first developed using sectioned sagittal otoliths, which in turn permitted examination of latitudinal variation in A. trutta growth, size compositions and age compositions. The von Bertalanffy growth function parameters for A. trutta in SE Australia were estimated to be L ?=?63.20?±?0.37 cm fork length (FL), k?=?0.26?±?0.01 yr?1 and t o?=??0.14?±?0.03 yr, with a maximum estimated age of 12.7 years. Growth was shown to be faster with decreasing latitude likely due to a simple relationship with the latitudinal gradient in water temperature; fastest growth occurring in northern NSW and slowest growth occurring in Tasmania. Latitudinal patterns in growth were remarkably similar to those previously reported for this species, despite age being estimated using scale readings some 40 years ago. This consistency in latitudinal growth patterns of a temperate fish can be attributed to the life history-related movements undertaken by A. trutta in this region. This temporally-consistent movement pattern is supported by the spatial gradient in the size and age composition of A. trutta sampled from different latitudes in both the current and historical research, whereby numbers of large and old fish increase progressively from Tasmania to northern NSW. These results highlight the need to consider the potential for spatial size- or age-structuring in the development of sampling designs and interpretation of results for any study examining spatial or temporal variation in demographic parameters of exploited fish populations.  相似文献

Understanding patterns of genetic diversity of plants is important in guiding conservation programs. The aim of our study was to characterize genetic diversity in Afzelia quanzensis, an economically important African tree species. We genotyped 192 individuals at 10 nuclear microsatellite loci. Samples were collected from nine sites in Zimbabwe, five in the north and four in the south, separated by a mountain range, the Kalahari-Zimbabwe axis. Overall, genetic diversity was relatively low across all sites (expected heterozygosity (H E)?=?0.452, mean number of alleles (A)?=?4.367, allelic richness (A R)?=?2.917, effective number of alleles (A E)?=?2.208, and private allelic richness (PAR)?=?0.197). Genetic diversity estimates, H E, A, A R, and PAR, were not significantly different between northern and southern sites. Allelic richness was significantly higher in southern sites. Significant population differentiation was observed among all sites (F ST ?=?0.0936, G′ ST ?=?0.1982, G ST ?=?0.1001, D JOST?=?0.0598). STRUCTURE analysis and principal components analysis identified two gene pools, one predominantly made up of southern individuals, and the other of northern individuals. A Monmonier’s function detected a genetic barrier that coincided with the Kalahari-Zimbabwe axis. The relatively low level of genetic diversity in A. quanzensis may reduce adaptability and limit future evolutionary responses. All sites should be monitored for deleterious effects of low genetic diversity, and genetic resource management should take into consideration the existence of the distinct gene pools to capture the entire extant genetic variation.  相似文献

Knowing how microevolutionary processes, such as genetic drift and natural selection, shape variation in adaptive traits is strategic for conservation measures. One way to estimate local adaptation is to compare divergences in quantitative traits (QST) and neutral loci (FST). Therefore, we have assessed the pattern of phenotypic and molecular genetic divergence among natural subpopulations of the fruit tree Eugenia dysenterica DC. A provenance and progeny test was performed to assess the quantitative traits of the subpopulations collected in a wide distribution area of the species in the Brazilian Cerrado. The sampled environments are in a biodiversity hotspot with heterogeneous soil and climate conditions. By associating quantitative trait variation in initial seedling development with neutral microsatellite marker variation, we tested the local adaptation of the traits by the QSTFST contrast. Genetic drift was prevalent in the phenotypic differentiation among the subpopulations, although the traits seedling emergence time and root green mass, which are relevant for adaptation to the Cerrado climate, showed signs of uniform selection. Our results suggest that E. dysenterica has a spatial genetic structure divided into two large groups, separated by a line that divides the Cerrado biome in a southwestern to northeastern direction. This structure must be taken into account for managing E. dysenterica genetic resources both for conservation and breeding purposes.  相似文献

Although it is well established that different coral species have different susceptibilities to thermal stress, the reasons behind this variation are still unclear. In this study, 384 samples across five dominant coral species were collected seasonally between September 2013 and August 2014 at Luhuitou fringing reef in Sanya, Hainan Island, northern South China Sea, and their algal symbiont density and effective photochemical efficiency (Φ PSII) were measured. The results indicated that both the Symbiodinium density and Φ PSII of corals were subject to significant interspecies and seasonal variations. Stress-tolerant coral species, including massive Porites lutea and plating Pavona decussata, had higher symbiont densities but lower Φ PSII compared to the vulnerable branching species of Acropora over the course of all four seasons. Seasonally, coral symbiont densities were the lowest during winter, while during the same period, Φ PSII of corals was at the highest point. Further analysis suggested that dissolved inorganic nutrients and upwelling in the reef area were probably responsible for the observed seasonal variations in symbiont density. The fact that Porites lutea has the lowest Φ PSII during all four seasons is likely related to their symbionts’ lower capacity to provide required photosynthates for calcification. These results suggest that a coral’s thermal tolerance is primarily and positively dependent on its symbiont density and is less related to its effective photochemical efficiency.  相似文献

Haplotype frequencies and allele distributions at 11 STR loci of the Y chromosome were evaluated in 180 unrelated individuals from Russian population of Southern Federal district of the Russian Federation (Rostov oblast, Krasnodar krai, and Stavropol krai). Among 153 Y-chromosomal haplotypes discovered, 62 were unique. In the sample of Russian population, the most frequent haplotype (frequency of 5.56%) was 16-11,14-13-30-25-11-11-13-14-11-10 (for the loci DYS19, DYS385a,b, DYS389I, DYS389II, DYS390, DYS391, DYS392, DYS393, DYS437, DYS438, and DYS439, respectively). Despite the high diversity of Y-chromosomal haplotypes in the Russian populations from the south of Russia (the h value was 0.997, 0.995, and 0.994 in Rostov, Krasnodar, and Stavropol samples, respectively), analysis of molecular variance (AMOVA) showed the absence of differentiation between the populations (ΦST = 0.1%, P = 0.36). Comparative differentiation analysis performed for 13 Russian populations from the European part of Russia pointed to low among-population differentiation in Y-chromosomal lineages (ΦST = 0.52%, P = 0.03).  相似文献

Abundance, spatial distribution, population structure and growth of non-native Eurasian perch (Perca fluviatilis) were examined in the Lake Skadar, a large, unstratified lake in the Mediterranean region inhabited by several endemic, rare and threatened fish species. Fish were caught in the infra-littoral and littoral habitats of the northern and central parts of the lake using multi-mesh gill nets over three consecutive years. Eurasian perch was among the most dominant species in both habitats. The mean relative abundance is - NPUE (0.556 individuals m?2) and relative biomass is - WPUE (23.56 g m?2) (years and habitats combined). Its population consisted primarily of individuals in first, second and third year of life span (0+???2+ age classes). The youngest fish in first and second year showed low mortality rates, older fish in third, fourth, fifth and sixth year of life span (age classes 2+???5+) experienced great losses, with mortality rates ranging from 0.6–1.0. The von Bertalanffy growth curve formula, expressing the expected total length L t ?=?36.98 (1-e-0.23 (t?+?1.018)) with growth performance value Φ ?=?2.50, revealed fast growth-in-length. Length-to-weight relationship was Wt?=?6.2?×?10?3 L t 3.27, indicating positive growth-in-weight. It has been concluded that 35 years after the first finding in the Lake Skadar, Eurasian perch adapted well to local conditions in this Mediterranean environment, with the life span changed in favor of fast growth and early maturation.  相似文献

Knowledge of larval dispersal and connectivity in coral reef species is crucial for understanding population dynamics, resilience, and evolution of species. Here, we use ten microsatellites and one mitochondrial marker (cytochrome b) to investigate the genetic population structure, genetic diversity, and historical demography of the powder-blue tang Acanthurus leucosternon across more than 1000 km of the scarcely studied Eastern African region. The global AMOVA results based on microsatellites reveal a low but significant F ST value (F ST = 0.00252 P < 0.001; D EST = 0.025 P = 0.0018) for the 336 specimens sampled at ten sample sites, while no significant differentiation could be found in the mitochondrial cytochrome b dataset. On the other hand, pairwise F ST, PCOA, and hierarchical analysis failed to identify any genetic breaks among the Eastern African populations, supporting the hypothesis of genetic homogeneity. The observed genetic homogeneity among Eastern African sample sites can be explained by the lengthy post-larval stage of A. leucosternon, which can potentiate long-distance dispersal. Tests of neutrality and mismatch distribution signal a population expansion during the mid-Pleistocene period.  相似文献

The uplift of mountains and climatic oscillations are important for understanding of the demographic history and genetic structure of species. We investigated the biogeographic history of the boreal tree species Ulmus lamellosa (Ulmaceae) in China, by using a combined phylogeographic and paleodistribution modeling approach. In this study, 14 populations of endangered U. lamellosa were analyzed by using chloroplast DNA (cpDNA) sequences. A high level of genetic differentiation (Φ ST = 86.22%) among populations with a significant phylogeographic pattern (N ST > G ST, P < 0.05) was found in U. lamellosa. Ten haplotypes were detected by combining chloroplast DNA data, and haplotype 3 (H3) was found to be common and widespread. The intraspecific divergence of all U. lamellosa cpDNA haplotypes (9.27 Ma; 95% HPD 5.17–13.33 Ma) most probably began in the late Miocene. The pairwise difference among haplotypes and neutrality tests (Tajima’s D and Fu’s Fs statistic) indicated that populations of U. lamellosa, except group I, have not experienced recent sudden expansions. Multiple refuge areas were identified across the entire distribution ranges of U. lamellosa. The low level of gene flow (Nm = 0.14) among populations may have resulted from isolation resulting from distance and complex topography during climatic oscillations; this isolation was probably the major process that shaped the present distribution of haplotypes. These results support the hypothesis that U. lamellosa persisted in situ during glaciations and occupied multiple localized glacial refugia, contrary to the hypotheses of large-scale range contraction and long-distance southward migration.  相似文献

Vendace (Coregonus albula (L.)) is a very plastic species of freshwater whitefish which is widespread in Europe. But in Latvia this species is included into the list of specially protected fish species with limited use. We examined cross-species amplification of 14 microsatellite loci (Cocl-Lav22, Cocl-Lav23, Cisco-59, Cisco-106, Cisco-90, Cisco-126, Cisco-157, Cisco-179, Cisco-181, Cisco-183, Cisco-200, BWF1, BWF2, and C4-157) which were successfully used for genetic studies, monitoring, protection and management of different Coregonus species. Five microsatellite markers (BWF1, Cisco90, Cisco126, Cisco157, Cisco 200) had a good representation in populations of vendace from seven Latvian lakes (Sventes, Rāznas, Nirzas, Dridzas, Stirnu, Aluksnes, E?ezers) and were used for genetic analysis of these populations. The mean number of alleles per locus (from 4.4 to 6.2), the observed (from 0.53 to 0.70) and expected (from 0.49 to 0.70) heterozygosity, population structure (F ST and R ST ) and gene flow (Nm) were also analysed in Latvian vendace populations.  相似文献

Rare species consisting of small populations are subject to random genetic drift, which reduces genetic diversity. Thus, determining the relationship between population size and genetic diversity would provide key information for planning a conservation strategy for rare species. We used six microsatellite markers to investigate seven extant populations of the rare conifer Pseudotsuga japonica, which is endemic to the Kii Peninsula and Shikoku Island regions that are geographically separated by the Kii Channel in southwest Japan. The population differentiation of P. japonica was relatively high (FST = 0.101) for a coniferous species, suggesting limited gene flow among populations. As expected, significant regional differentiation (AMOVA; p?<?0.05) indicated genetic divergence across the Kii Channel. A strong positive correlation between census population size and the number of rare alleles (r?=?0.862, p?<?0.05) was found, but correlations with major indices of genetic diversity were not significant (allelic richness: r?=?0.649, p?=?0.104, expected heterozygosity: r?=?0.361, p?=?0.426). The observed order of magnitude of correlation with three genetic diversity indices corresponded with the theoretically expected order of each index’ sensitivity (i.e., the rate of decline per generation) to the bottleneck event. Thus, features that exhibit a faster response, i.e., the number of rare alleles, would have been subject to deleterious effects of the recent decline in population size, which is presumably caused by the development of extensive artificial plantations of other tree species over the last several decades. Finally, we propose a conservation plan for P. japonica based on our findings.  相似文献

Major histocompatibility complex (MHC) genes are excellent markers for the study of adaptive genetic variation occurring over different geographical scales. The Chinese egret (Egretta eulophotes) is a vulnerable ardeid species with an estimated global population of 2600–3400 individuals. In this study, we sampled 172 individuals of this egret (approximately 6 % of the global population) from five natural populations that span the entire distribution range of this species in China. We examined their population genetic diversity and geographical differentiation at three MHC class II DAB genes by identifying eight exon 2 alleles at Egeu-DAB1, eight at Egeu-DAB2 and four at Egeu-DAB3. Allelic distributions at each of these three Egeu-DAB loci varied substantially within the five populations, while levels of genetic diversity varied slightly among the populations. Analysis of molecular variance showed low but significant genetic differentiation among five populations at all three Egeu-DAB loci (haplotype-based ?ST: 0.029, 0.020 and 0.042; and distance-based ?ST: 0.036, 0.027 and 0.043, respectively; all P < 0.01). The Mantel test suggested that this significant population genetic differentiation was likely due to an isolation-by-distance pattern of MHC evolution. However, the phylogenetic analyses and the Bayesian clustering analysis based on the three Egeu-DAB loci indicated that there was little geographical structuring of the genetic differentiation among five populations. These results provide fundamental population information for the conservation genetics of the vulnerable Chinese egret.  相似文献

The greater amberjack (Seriola dumerili) is a commercially and recreationally important marine fish species in the southeastern United States, where it has been historically managed as two non-mixing stocks (Gulf of Mexico and Atlantic). Mark-recapture studies and analysis of mitochondrial DNA have suggested the two stocks are demographically independent; however, little is currently known about when and where spawning occurs in Gulf of Mexico amberjack, and whether stock mixture occurs on breeding grounds. The primary objective of this study was to quantify stock mixture among breeding populations of amberjack collected from the Atlantic and Gulf of Mexico. Genetic data based on 11 loci identified very low, though statistically significant differentiation among Gulf of Mexico samples (GST = 0.007, \(G_{{{\text{ST}}}}^{\prime }\) = 0.009; all P?=?0.001) and between reproductive adults collected from two spawning areas (GST = 0.007, \(G_{{{\text{ST}}}}^{\prime }\) = 0.014; all P?=?0.001). Naïve Bayesian mixture analysis supported a single genetic cluster [p(S|data)?=?0.734] whereas trained clustering (using Atlantic and Gulf spawning fish) gave the highest support to a two-cluster model (p(S|data)?=?1.0). Our results support the argument that the genetic structuring of greater amberjack is more complex than the previously assumed two, non-mixing stock model. Although our data provide evidence of limited population structure, we argue in favour of non-panmixia among reproductive fish collected from the Gulf of Mexico and Florida Keys.  相似文献

Identification of population units is crucial for management and monitoring programs, especially for endangered wild species. The roughskin sculpin (Trachidermus fasciatus Heckel) is a small catadromous fish and has been listed as a second class state protected aquatic animal since 1988 in China. To achieve sustainable conservation of this species, it is necessary to clarify the existing genetic structure both between and within populations. Here, population genetic structure among eight populations of T. fasciatus were analyzed by using 16 highly polymorphic microsatellites. High levels of genetic variation were observed in all populations. All pairwise F ST estimates were significant after false discovery rate correction (overall average F ST = 0.054). Furthermore, both STRUCTURE and discriminant analysis of principal components (DAPC) analysis showed that the eight populations were grouped into six clusters. BAYESASS analysis showed generally low recent and asymmetric migration among populations. All these results suggested significant genetic structure across populations. However, there was no isolation by distance relationship among populations, likely resulting from barriers to gene flow created by habitat fragmentation. Our results highlight the need for in situ conservation efforts for T. fasciatus across its entire distribution range, through maximizing habitat size and quality to preserve overall genetic diversity and evolutionary potential.  相似文献

Polar cod, Boreogadus saida, is a key species in Arctic marine ecosystems; however, its genetic population structure is largely undescribed. The population genetic structure of 472 B. saida specimens among nine locations in the north-east Atlantic was revealed using 12 microsatellite loci. Pairwise F ST comparisons showed significant population differentiation between B. saida sampled inside fjords in Svalbard and north-east Greenland, as compared to B. saida from the shelf. The observed genetic variation was not a function of isolation by distance, and it is speculated that B. saida populations inhabiting fjords may have become reproductively isolated from shelf-dwelling B. saida during the last post-glacial recolonization.  相似文献

Variability of microsatellite DNA loci Gmo3, Gmo34, and Gmo35 is studied in samples of Pacific cod Gadus macrocephalus and Atlantic cod G. morhua. The results show high values of identity of the samples within the North Pacific basin (0.9766–0.9924) and within the Northeast Atlantic basin (0.9580). Based on the pairwise assessment of genetic differentiation, the F ST values are significantly different in all variants between the samples of Pacific and Atlantic cod (F ST = 0.5235–0.6719, p < 0.001). Within the basins, the significant differences in the frequencies of main alleles are revealed in the loci Gmo3 and Gmo34 for the samples from the Pacific and Atlantic oceans, respectively.  相似文献

This study investigates changes in body proportions during ontogenesis of the bivalves Astarte arctica and A. borealis from the subtidal zone of northeastern Sakhalin. Marked differences in the shell form of bivalves were found. The shell of A. arctica is convex and compact, shell length and height are comparable, and shell width is more than 50% of shell height. A. borealis has a slightly elongate and flattened shell, its width is less than 50% of its height. Shell proportions can provide a reliable criterion for differentiating the species. Mollusks grow at nearly the same rate but attain different maximum sizes. Linear growth is described by Bertalanffy’s equation: Lt = 48.2[1 ? e ?0.4850(t ? 0.4396)] for A. arctica and Lt = 53.1[1 ? e?0.4106(t ? 0.4253)] for A. borealis. In northeastern Sakhalin, the life span of A. arctica is 7 years and of A. borealis, 8 years. By life span, the two bivalve species from Sakhalin slightly differ from their counterparts in other regions, particularly the arctic seas.  相似文献
