Sucrose (Suc) plays a central role in plant growth and development. It is a major end product of photosynthesis and functions as a primary transport sugar and in some cases as a direct or indirect regulator of gene expression. Research during the last 2 decades has identified the pathways involved and which enzymes contribute to the control of flux. Availability of metabolites for Suc synthesis and ‘demand’ for products of sucrose degradation are important factors, but this review specifically focuses on the biosynthetic enzyme sucrose-phosphate synthase (SPS), and the degradative enzymes, sucrose synthase (SuSy), and the invertases. Recent progress has included the cloning of genes encoding these enzymes and the elucidation of posttranslational regulatory mechanisms. Protein phosphorylation is emerging as an important mechanism controlling SPS activity in response to various environmental and endogenous signals. In terms of Suc degradation, invertase-catalyzed hydrolysis generally has been associated with cell expansion, whereas SuSy-catalyzed metabolism has been linked with biosynthetic processes (e.g., cell wall or storage products). Recent results indicate that SuSy may be localized in multiple cellular compartments: (1) as a soluble enzyme in the cytosol (as traditionally assumed); (2) associated with the plasma membrane; and (3) associated with the actin cytoskel-eton. Phosphorylation of SuSy has been shown to occur and may be one of the factors controlling localization of the enzyme. The purpose of this review is to summarize some of the recent developments relating to regulation of activity and localization of key enzymes involved in sucrose metabolism in plants.  相似文献

Research on NO in plants has gained considerable attention in recent years mainly due to its function in plant growth and development and as a key signalling molecule in different intracellular processes in plants. The NO emission from plants is known since the 1970s, and now there is abundant information on the multiple effects of exogenously applied NO on different physiological and biochemical processes of plants. The physiological function of NO in plants mainly involves the induction of different processes, including the expression of defence-related genes against pathogens and apoptosis/programmed cell death (PCD), maturation and senescence, stomatal closure, seed germination, root development and the induction of ethylene emission. NO can be produced in plants by non-enzymatic and enzymatic systems. The NO-producing enzymes identified in plants are nitrate reductase, and several nitric oxide synthase-like activities, including one localized in peroxisomes which has been biochemically characterized. Recently, two genes of plant proteins with NOS activity have been isolated and characterized for the first time, and both proteins do not have sequence similarities to any mammalian NOS isoform. However, different evidence available indicate that there are other potential enzymatic sources of NO in plants, including xanthine oxidoreductase, peroxidase, cytochrome P450, and some hemeproteins. In plants, the enzymatic production of the signal molecule NO, either constitutive or induced by different biotic/abiotic stresses, may be a much more common event than was initially thought.  相似文献

Caffeine (1,3,7-trimethylxanthine) and theobromine (3,7-dimethylxanthine) are the major purine alkaloids in plants. To investigate the diversity of N-methyltransferases involved in purine alkaloid biosynthesis, we isolated the genes homologous for caffeine synthase from theobromine-accumulating plants. The predicted amino acid sequences of N-methyltransferases in theobromine-accumulating species in Camellia were more than 80% identical to caffeine synthase in C. sinensis. However, there was a little homology among the N-methyltransferases between Camellia and Theobroma. The recombinant enzymes derived from theobromine-accumulating plants had only 3-N-methyltransferase activity. The accumulation of purine alkaloids was, therefore, dependent on the substrate specificity of N-methyltransferase determined by one amino acid residue in the central part of the protein.  相似文献

Starch plays a major role as a transitory and long-term storage compound in higher plants, and therefore is of prime importance for plant growth and development. Additionally, starch serves as a widely used material for a variety of industrial uses. The formation of starch can arbitrarily be divided into three types of event: (I) those leading to the supply of glucose-1-phosphate in the plastids; (II) the conversion of glucose-1-phosphate to ADP-glucose catalysed by the enzyme ADP-glucose pyrophosphorylase; and (III) the enzymatic reactions converting ADP-glucose to long-chain glucans (amylopectin, amylose). In recent years, numerous cDNA and genomic sequences encoding enzymes involved in starch metabolism have been identified. Some of these have been used to down-regulate enzyme activities via the antisense RNA technique. Additionally, bacterial genes have been ectopically expressed in transgenic plants in order to increase corresponding enzyme activities. By modulating the activity of ADP-glucose pyrophosphorylase in plastids, it was possible to decrease and increase, respectively, the starch content in source and sink organs of transgenic plants. In addition, down-regulation of granule-bound starch synthase (isoform I) resulted in the production of starch that was almost completely free of amylose. Further experiments aimed to modulate starch structure are currently underway and will briefly be discussed.  相似文献

Changes in the sucrose metabolism of Cur-cuma longa L. plants were studied under treatment with different triazole compounds viz., triadimefon (TDM) and propiconazole (PCZ). Plants were treated with TDM at 15 mg/L and PCZ at 10 mg/L separately by soil drenching on 80, 110, and 140 days after planting (DAP). The plants were harvested randomly on 90, 120, and 150DAP to determine the effect of both the triazoles on sucrose metabolizing enzymes and phenol content. The sucrose metabolism was studied by analyzing sucrose metaboliz-ing enzymes like sucrose synthase and sucrose phosphate synthase. All the analyses were assayed in leaves and tubers of both control and treated plants. It was found that both of the triazole compounds had profound effects on these parameters.  相似文献

Mitochondria isolated from root tissue of maize plants grown on a modified Knop solution containing 10.9 mM nitrate ± 7.2 mM ammonium were purified on the discontinuous Percoll density gradient with polyvinylpyrrolidone (PVP) added. The presence of PVP allowed separation of several mitochondrial fractions of a different density. Contrary to mitochondria isolated from plants grown in the presence of nitrate alone, revealing only two fractions, the mitochondria from NH4 +/NO3 -plants were distributed in four fractions. Total amount of mitochondria, as well as specific activities of some nitrogen metabolism enzymes and tricarboxylic acid (TCA) cycle enzymes of all mitochondrial fractions, and respiratory activities of two lower density fractions isolated from plants grown on mixed nitrogen were higher in comparison to mitochondria from nitrate-grown plants.  相似文献

The content of glucose, fructose and saccharose as well as changes in the activities of enzymes involved in their biosynthesis and degradation were studied in tobacco plants infected with potato virus Y (necrotic strain) during the acute-infection period. Over the first part of this period, accumulation of saccharose, glucose and fructose was observed concurrently with decreased activities of the enzymes metabolizing saccharose, glucose and fructose (saccharases, saccharose synthase and hexokinases) and enhancement in the activities of enzymes synthesizing these carbohydrates (saccharosephosphate synthase, glucose-6-phosphate and/or fructose-6-phosphate phosphatases). The subsequent period was characterised by a reduction in both phosphatases that (together with just slightly raised saccharosephosphate synthase) could hardly produce enough sugars for the highly stimulated enzymes such as saccharases, saccharose synthase, and both kinases. Presumably for this reason, the previously increased content of sugars was considerably reduced to the level of control plants. The activities of glucokinase, fructokinase, saccharases and saccharose synthase were strongly increased at the culmination of virus multiplication and negatively correlated with the content of free glucose, fructose and saccharose. Recombinant chalcone synthase (CHS) from Scutellaria baicalensis and stilbene synthase (STS) from Arachis hypogaea accepted CoA esters of long-chain fatty acid (CHS up to the C12 ester, while STS up to the C14 ester) as a starter substrate, and carried out sequential condensations with malonyl-CoA, leading to formation of triketide and tetraketide alpha-pyrones. Interestingly, the C6, C8, and C10 esters were kinetically favored by the enzymes over the physiological starter substrate; the kcat/KM values were 1.2- to 1.9-fold higher than that of p-coumaroyl-CoA. The catalytic diversities of the enzymes provided further mechanistic insights into the type III PKS reactions, and suggested involvement of the CHS-superfamily enzymes in the biosynthesis of long-chain alkyl polyphenols such as urushiol and ginkgolic acid in plants.  相似文献

Glycoglycerolipids are abundant membrane components in the photosynthetic tissues of plants and in cyanobacteria, with highly conserved structures (galactolipids). In non-photosynthetic bacteria, glycoglycerolipids are also widespread but with higher structural diversity. They are synthesized by the action of glycosyltransferases (GT), which transfers a glycosyl unit from a sugar nucleotide donor to diacylglycerol to form monoglycosyldiacylglycerol followed by a second transfer to give diglycosyldiacylglycerol. Both transferase activities are catalysed by different GT enzymes in plants, and many bacteria; however, processive enzymes, in which a single GT transfers the first and second (and eventually more) glycosyl units are also found in some bacteria. In this review, we summarize the diversity of glycosyltransferases involved in glycolipid biosynthesis in bacteria, focussing on mycoplasma enzymes and comparing processive and non-processive glycolipid synthases. Since glycoglycerolipids are key structural components of the plasma membrane in mycoplasmas, the glycolipid synthases involved in their biosynthesis are proposed as targets for the design of new antibiotics against mycoplasma infections.  相似文献

Plant secondary metabolism has been a focus of research in recent years due to its significant roles in plant defense and in human medicine and nutrition. A protein engineering strategy was designed to more effectively manipulate plant secondary metabolite (isoflavonoid) biosynthesis. A bifunctional isoflavone synthase/chalcone isomerase (IFS/CHI) enzyme was constructed by in-frame gene fusion, and expressed in yeast and tobacco. The fusion protein was targeted to the endoplasmic reticulum (ER) membrane and the individual enzymatic functions of its component fragments were retained when assayed in yeast. Petals and young leaves of IFS/CHI transgenic tobacco plants produced higher levels of the isoflavone genistein and genistein glycosides as a ratio of total flavonoids produced than did plants transformed with IFS alone. Thus, through a combined molecular modeling, in vitro protein engineering and in planta metabolic engineering approach, it was possible to increase the potential for accumulation of isoflavonoid compounds in non-legume plants. Construction of bifunctional enzymes will simplify the transformation of plants with multiple pathway genes, and such enzymes may find broad uses for enzyme (e.g., cytochrome P450 family) and biochemical pathway engineering.Electronic Supplementary Material Supplementary material is available for this article at and is accessible for authorized users.  相似文献

The methylerythritol 4-phosphate (MEP) pathway synthesizes the precursors of carotenoids and other isoprenoids in bacteria and plant plastids. Despite recent progress in the identification of rate-determining steps, the relative contribution of most pathway enzymes to flux control remains to be established. In this work we investigated whether upregulated levels of hydroxymethylbutenyl diphosphate synthase (HDS) could increase the metabolic flux through this pathway, as judged by endpoint (carotenoid) measurements. Unlike other MEP pathway enzymes, however, increasing the levels of an active HDS protein in carotenoid-producing Escherichia coli cells and transgenic Arabidopsis thaliana plants did not result in an enhanced accumulation of MEP-derived isoprenoids. Our data suggest that enhanced flux through the MEP pathway for peak demand periods in bacteria and plastids does not require increased HDS activity.  相似文献

This review mainly summarizes the recent research progress In elucidating the ecological role of terpenoids and characterization of the enzymes Involved in the terpenold biosynthesis. 大量的研究结果表明,活性氧参与植物乙烯生物合成过程具有明显的普遍性,超氧阴离子自由基是参与乙烯生物合成过程的主要活性氧。近年来研究的焦点主要从乙烯生物合成的关键调控酶ACC合酶及ACC氧化酶的酶活性、酶动力学特性、酶蛋白空间结构、酶基因表达水平等方面来阐明活性氧调控植物乙烯生物合成的机制。最新的研究表明:植物在各种正常或应激的生长条件下首先诱导了活性氧产生水平的变化,活性氧在基因或蛋白质水平上影响ACC合酶和ACC氧化酶的活性水平,从而调节乙烯的生物合成。本文首次综述了活性氧影响植物乙烯生物合成过程的最新研究进展,并对活性氧在植物乙烯生物合成中具有诱导与抑制并存的“双重性”作用进行了探讨。  相似文献

As with all organisms, plants must respond to a plethora of external environmental cues. Individual plant cells must also perceive and respond to a wide range of internal signals. It is now well-accepted that nitric oxide (NO) is a component of the repertoire of signals that a plant uses to both thrive and survive. Recent experimental data have shown, or at least implicated, the involvement of NO in reproductive processes, control of development and in the regulation of physiological responses such as stomatal closure. However, although studies concerning NO synthesis and signalling in animals are well-advanced, in plants there are still fundamental questions concerning how NO is produced and used that need to be answered. For example, there is a range of potential NO-generating enzymes in plants, but no obvious plant nitric oxide synthase (NOS) homolog has yet been identified. Some studies have shown the importance of NOS-like enzymes in mediating NO responses in plants, while other studies suggest that the enzyme nitrate reductase (NR) is more important. Still, more published work suggests the involvement of completely different enzymes in plant NO synthesis. Similarly, it is not always clear how NO mediates its responses. Although it appears that in plants, as in animals, NO can lead to an increase in the signal cGMP which leads to altered ion channel activity and gene expression, it is not understood how this actually occurs.
NO is a relatively reactive compound, and it is not always easy to study. Furthermore, its biological activity needs to be considered in conjunction with that of other compounds such as reactive oxygen species (ROS) which can have a profound effect on both its accumulation and function. In this paper, we will review the present understanding of how NO is produced in plants, how it is removed when its signal is no longer required and how it may be both perceived and acted upon.  相似文献

Carotenoids are Indispensable to plants and play a critical role in human nutrition and health. Significant progress has been made in our understanding of carotenoid metabolism in plants. The biosynthetic pathway has been extensively studied.Nearly all the genes encoding the biosynthetic enzymes have been isolated and characterized from various organisms. In recent years, there is an increasing body of work on the signaling pathways and plastid development, which might provide global control of carotenoid biosynthesis and accumulation. Herein, we will highlight recent progress on the biosynthesis,regulation, and metabolic engineering of carotenoids in plants, as well as the future research towards elucidating the regulatory mechanisms and metabolic network that control carotenoid metabolism.  相似文献

The pathway of nitrogen assimilation in plants   总被引:5,自引:0,他引:5  
The major route of nitrogen assimilation has been considered for many years to occur via the reductive amination of α-oxoglutarate, catalysed by glutamate dehydrogenase. However, recent work has shown that in most bacteria an alternative route via glutamine synthetase and glutamine: 2-oxoglutarate aminotransferase (glutamate synthase) operates under conditions of ammonia limitation. Subsequently the presence of a ferredoxin-dependent glutamate synthase in green leaves and green and blue-green algae, and a NAD(P)H and ferredoxin-dependent enzyme in roots and other non-green plant tissues, has suggested that this route may also function in most members of the plant kingdom. The only exceptions are probably the majority of the fungi, where so far most organisms studied do not appear to contain glutamate synthase. Besides the presence of the necessary enzymes there is other evidence to support the contention that the assimilation of ammonia into amino acids occurs via glutamine synthetase and glutamate synthase, and that it is unlikely that glutamate dehydrogenase plays a major role in nitrogen assimilation in bacteria or higher plants except in circumstances of ammonia excess.  相似文献

Faba bean (Vicia faba L. var. minor cv. Alborea) and pea (Pisum sativum L. cv. Lincoln) plants, inoculated with Rhizobium leguminosarum biovar. viciae strain GRA19, were treated with salt (100 mM NaCl) and/or nitrate (8 mM KNO3) to test whether plants grown with inorganic-nitrogen are more tolerant to salinity than plants entirely reliant upon fixed nitrogen. According to the growth inhibition recorded, pea plants dependent on dinitrogen fixation proved more tolerant to salt stress than those N-fertilized, in contrast to results obtained for faba bean plants. This study therefore confirms that plants dependent on nitrogen fixation are not always more sensitive to salinity than are N-fertilized plants. Nitrate addition did not reduce the specific nitrogenase activity in pea, but did in faba bean. However, nodulation was inhibited in both legumes. The specific nitrogenase activity was more affected by salt treatment in N-fertilized plants for both legumes. The activity of the enzymes mediating ammonium assimilation in nodules (GS, NADH-GOGAT) was inhibited by salt stress both in N-fixing and in N-fertilized pea and faba bean plants.  相似文献

SA has been shown to play an important signaling role in the activation of various plant defense responses following pathogen attack. These responses include the induction of local and systemic disease resistance, the potentiation of host cell death, and the containment of pathogen spread. The mechanisms through which SA mediates these effects are varied and can involve alterations in the activity or synthesis of certain enzymes, increased defense gene expression, potentiation of several defense responses, and/or the generation of free radicals. Through the analysis of mutant plants exhibiting aberrant responses to pathogen infection, many genes encoding products involved in the SA-mediated defense pathway(s) have been isolated. In addition, mounting evidence suggests that certain defense responses can be activated via a SA-independent pathway(s). This review focuses primarily on recent discoveries pertaining to the SA signaling pathway(s) leading to disease resistance; however, a very brief discussion of the SA-independent pathway (s) and its ability to cross-talk with the SA pathway is also presented.  相似文献
