No significant differences between the serum protein concentrations of the totally starved pigs and control animals given a milk protein supplemented diet were found. Serum lipid levels rose in the totally starved group. During the first 42 days of the experimental period sharp falls in serum protein concentrations were noted in the protein deprived pigs. The losses were greatest in the albumin and two β-globulin fractions, γ-globulin levels rose but at a much slower rate than in the control group. In the pigs which were refed a protein containing diet during the second 42 day period, the concentrations of all the serum protein fractions except for γ-globulin had reached control levels by the end of the investigation. The pigs which continued with the protein free diet showed further losses of albumin, a reduction in some α-globulin fractions and a cessation of net γ-globulin synthesis. The trends observed by measurements of protein bound carbohydrate were similar to those obtained from polypeptide determinations indicating quantitative rather than qualitative changes in the serum proteins. The reduction in the serum lipid concentrations of the pigs undergoing protein starvation was largely accounted for by losses of α- and β-lipoproteins. The protein deprived pigs maintained their initial body weight, while a continuous fall in weight was found in the totally starved group and a continuous increase in the control pigs.  相似文献

Recently, decreased activity levels have been observed in pigs treated postoperatively with transdermal delivery of fentanyl (TD-fentanyl) after isoflurane anaesthesia. Whether the change in behaviour is related to opioid-induced sedation or to insufficient pain relief remains to be investigated. This study was therefore undertaken to evaluate the effect of TD-fentanyl 50 μg h-1 on the activity level with and without isoflurane anaesthesia. Eight pigs (25.4 ± 5.2 kg) were submitted to a cross-over study and given two treatments; 1) fentanyl patch applied after 30 minutes of anaesthesia (treatment A/F) and 2) fentanyl patch without anaesthesia (treatment F). The pigs' behaviour was observed from a video recording instantaneously every 10 minutes for 24 h before treatments and up to 72 h after the patch attachment. Venous blood samples were taken 1, 6, 12, 24, 48 and 72 h after the patch application. The behaviour recordings showed that TD-fentanyl did not produce sedation in any pig. No differences were found between the two treatments in activity level, weight gain or serum fentanyl concentration. This concentration measured after 24 h was 0.27 ± 0.11 ng ml-1 and 0.47 ± 0.40 ng ml-1 in the A/F and F group, respectively.  相似文献

The fermentative production of uridine diphosphate N-acetylglucosamine (UDPAG) from 5′-UMP and glucosamine by dried cells of baker’s yeast was studied. UDPAG was found to accumulate in a reaction system containing 5′-UMP, glucosamine, fructose, inorganic phosphate and magnesium ions with dried baker’s yeast as an enzyme source. UDPAG was separated from the reaction mixture by means of anion exchange column chromatography and was identified by several biochemical methods.

The reaction conditions for the fermentative production of UDPAG were examined. The yield of UDPAG was about 40~66% based on 5′-UMP added when fermentation conditions were optimized. The concentration of glucosamine and potassium phosphate buffer, and pH as well as the water content of dried cells greatly affected the formation of UDPAG.  相似文献

分别采用5 mW·mm-2 He-Ne激光辐照、10.08 kJ·m-2·d-1 增强UV-B辐射及二者组合对"晋麦8号"小麦幼苗进行处理,测定各处理幼苗叶片可溶性糖、还原性糖、蔗糖合成酶(SS)、蔗糖磷酸合成酶(SPS) 以及α,β-淀粉酶的变化.研究分析He-Ne激光对增强UV-B辐射引起小麦损伤的修复效应.结果表明:He-Ne激光辐照可使UV-B辐射后小麦幼苗可溶性糖含量升高,还原性糖含量降低,SS,SPS活力降低,α,β-淀粉酶活力升高.该结果同时表明可溶性糖、还原性糖、SS、SPS、α,β-淀粉酶的变化同小麦幼苗损伤修复的能力相关,一定剂量的He-Ne激光辐照可部分修复增强UV-B对小麦幼苗糖代谢的辐射损伤.  相似文献

The impact of porcine epidemic diarrhea virus (PEDv) infection on the US pork industry has mainly been attributed to the mortality that it causes in suckling piglets, and, consequently, much effort has been invested in the quantification of its effect in sow farms. However, no information on the performance of surviving pigs that were exposed to the PEDv as piglets is available. Here, a retrospective cohort study to evaluate the impact of porcine epidemic diarrhea virus (PEDv) infection on growing pigs’ performance, as indicated by mortality, average daily gain (ADG), average daily feed intake (ADFI), and feed conversion ratio (FCR) was performed using production records from weaned pigs in nursery and wean-to-finish sites from sow farms that became PEDv-infected between May 2013 and June 2014. Production records from the first batch of growing pigs weaned in infected flows after the PEDv outbreak (“infected batches”) were compared with those from pigs weaned within the previous 14 to 120 days (“control batches”). Performance records from infected and control batches, paired by flow, were compared using non-parametric paired tests. Mortality, ADG and FCR were significantly different in PEDv-positive (infected) compared with PEDv-negative (control) batches, with a mean increase of mortality and FCR of 11% and 0.5, respectively, and a decrease of ADG of 0.16 lb/day. Our results demonstrate a poorer performance of growing pigs weaned after a PEDv outbreak compared with those weaned within the previous 14-120 days, suggesting that in addition to the mortality induced by PEDv in suckling pigs, the disease also impairs the performance of surviving pig. These findings help to quantify the impact of PEDv infection in the US and, ultimately, contribute to efforts to quantify the cost-effectiveness of disease prevention and control measures.  相似文献

A comparison was made between different anaesthetics for general anaesthesia in growing pigs, with focus on minor surgery under field conditions and for experiments in clinical research. Healthy crossbreed pigs (HampshirexYorkshirexSwedish Landrace) weighing 20–45 kg were used. The anaesthetics combinations compared were 1) azaperone plus metomidate (AM), 2) Zoletil® (zolazepam + tiletamine) plus xylazine (ZX), and 3) Zoletil® plus xylazine plus ketamine (ZXK). Parameters measured were: heart rate, respiratory rate, blood pressure, body temperature, and depth of analgesia (pin-prick). Minor surgery was performed to test the reliability of the “pin-prick” tests. It was clearly shown that AM produces anaesthesia with good cardiovascular stability and is a drug combination that is suitable for minor surgery. ZX also produces a good anaesthesia characterized by reliable and rapid induction. Good cardiovascular function is maintained, and the laryngeal relaxation makes intubation possible. These characteristics are very useful in a laboratory environment, as easy handling to avoid stress is necessary for research. Although it is difficult to evaluate the quality of analgesia from this study, it is concluded that ZX did not provide a superior anasthesia and analgesia compared to AM in crossbreed pigs. However, these drugs are too expensive for regular use in ambulatory practice. Ferrokinetic investigations have been carried out in pigs which had undergone protein starvation and rehabilitation. After long term protein deprivation haematocrit levels, blood haemoglobin levels, serum iron concentrations, serum total iron binding capacity and blood volume per kg body weight were all reduced in the deficient animals. The values for the rehabilitated animals were in the normal range. The erythrocytes of the protein starved pigs showed an increased rate of uptake of radioactive iron compared with the control pig. However, the turnover of iron as a whole was reduced greatly in the protein deprived group compared to the rehabilitated and control pigs. Accumulation of iron occurred in the livers, spleens and kidneys of the protein starved animals. The rehabilitated pigs had lower liver stores of iron than the control pig. On the basis of the data presented it has been concluded that these effects are mainly due to a shortage of amino acids for protein synthesis.  相似文献

By using e.s.r. spectroscopy coupled with the spin trapping technique we have detected the formation of free radical intermediates by rat liver microsomes incubated with either ethanol, 2-propanol or 2-butanol in the presence of a NADPH regenerating system and 4-pyridyl-l-oxide-t-butyl nitrone (4-POBN) as spin trap. The e.s.r. spectra have been identified as due to the hydroxyalkyl free radical adducts of 4-POBN.

The free radical formation depends upon the activity of the microsomal monoxygenase system and is blocked by omitting NADP+ from the incubation mixture, by anaerobic incubation or by enzyme denaturation. The involvement of hydroxyl radicals (OH) produced through a Fenton-type reaction from endogenously formed hydrogen peroxide is suggested by the opposite effects exerted on the e.s.r. signal intensity by azide and catalase. Consistently, iron chelation by desferrioxamine inhibits the free radical formation, while the supplementation of EDTA-iron increases it by several fold. Inhibitors of cytochrome P450-dependent monoxygenase system reduce to various extents the production of free radical intermediates suggesting that reactive oxygen species might be formed at the active site of cytochrome P450 where they react with alkyl alcohol molecules.

The data presented support the hypothesis that free radical species are generated during the microsomal metabolism of alcohols and suggest the possibility that ethanol-derived radicals might play a role in the pathogenesis of the liver lesions consequent upon alcoholic abuse.  相似文献

The following diuretics have been examined for their influence on renal clearances in pigs: chlorthiazidum (2 mg/kg), hydrochlor-thiazidum (0.2 and 2 mg/kg), furosemidum (0.5 mg/kg) and mer- salylum (2.5 and 10 mg/kg). The investigation comprised determinationof the clearances of inulin, endogenous creatinine, urea, PAH, sodium, potassium and chloride, before and after the administration of the diuretics. Ghlorthiazidum, hydrochlorthiazidum and mersalylum in the low dose did not affect the clearances of inulin, endogenous creatinine, urea and PAH. When furosemidum was administered, there was first a slight increase and then a decrease in those clearances. After administration of 10 mg/kg mersalylum, there was a strong decrease in the inulin, endogenous creatinine, urea and PAH clearances, and simultaneously glucose and protein could be found in the urine. All four diuretics caused a markedly increased excretion of sodium and chloride, while the excretion of potassium was only moderately increased. The effect on the excretion of sodium and chloride was of about the same order of magnitude for chlorthiazidum and hydrochlorthiazidum, though the effect of the latter was slightly more prolonged. In contrast to those two diuretics, furosemidum had a very strong but short effect. The influence of mersalylum on the excretion of sodium and chloride was somewhat stronger and more prolonged than that of the thiazides. Histological examinations of the kidneys of the pigs given 10 mg/kg mersalylum revealed pronounced tubular degeneration, particularly in the proximal tubules.  相似文献

Twenty Norwegian Landrace pigs were divided into 5 groups and fed a basal diet consisting of a mixture of dried skim milk and whey powder together with ground barley. The diet was supplemented with 0, 0.2, 0.8, 1.2 and 2.2 μg selenium as sodium selenite and was fed for 12 weeks. The muscle selenium level was increased by a factor of about 4 and the liver selenium by a factor of about 12 when the dietary selenium supplement was increased from zero to 2.2 μg/g. There was a significant linear correlation between dietary selenium and selenium concentrations in tissues. Possible benefit for humans consuming meat from animals having received the selenium doses used in this experiment are discussed. dietary selenium; tissue levels; pigs.  相似文献

Brevibacterium flavum mutants defective in the phosphoenolpyruvate (PEP)-dependent glucose phosphotransferase system (PTS) were selected with high frequency by 2-deoxyglucose-resistance. Most of them (DOGr) still had the fructose-PTS and grew not only on fructose but also on glucose like the wild-type strain. A mutant having 1 /8th the fructose-PTS activity of the wild strain but normal glucose-PTS activity was isolated as a xylitol-resistant mutant. It grew on glucose but not on fructose. The glucose-PTS was active on glucose, glucosamine, 2-deoxyglucose and mannose, and slightly on methyl-a-glucoside and N-acetylglucosamine, but not on fructose or xylitol. The fructose-PTS acted on fructose and xylitol, and to some extent on glucose but not on glucosamine or 2-deoxyglucose. Mutants unable to grow on glucose (DOGrGlc-) derived from a DOGr mutant were all defective in the fructose-PTS. All revertants able to grow on glucose derived from a DOGrGlc“ mutant had the fructose-PTS. The glucokinase activity was about 2/3rds the glucose activity of the fructose-PTS. All the DOGrGlc- mutants had normal levels of glucokinase. One of these mutants grew on maltose and sucrose, which were hydrolyzed to glucose. Thus, glucokinase seems to contribute to the phosphorylation of glucose liberated inside the cell. The fructose-PTS was induced by fructose and repressed by glucose. The glucose repression was not observed in a mutant defective in the glucose-PTS.  相似文献

Heat stress is one of the major limiting factors of production efficiency in the swine industry. The aims of the present study were 1) to observe if hemorheological and hematological parameters could be associated to physiological acclimation during the first days of heat stress exposure and 2) to determine if water restriction could modulate the effect of thermal heat stress on physiological, hematological and hemorheological parameters. Twelve Large White male pigs were divided into an ad libitum and a water restricted group. All pigs were submitted to one week at 24°C (D-7 to D-1). Then, at D0, temperature was progressively increased until 32°C and maintained during one week (D1 to D7). We performed daily measurements of water and feed intake. Physiological (i.e., skin temperature, rectal temperature, respiratory rate), hematological and hemorheological parameters were measured on D-6, D-5, D0, D1, D2 and D7. Water restriction had no effect on physiological, hematological and hemorheological parameters. The first days of heat stress caused an increase in the three physiological parameters followed by a reduction of these parameters suggesting a successful acclimation of pigs to heat stress. We showed an increase in hematocrit, red blood cell aggregation and red blood cell aggregation strength during heat stress. Further, we observed an important release of reticulocytes, an increase of red blood cell deformability and a reduction of feed intake and blood viscosity under heat stress. This study suggests that physiological acute adaptation to heat stress is accompanied by large hematological and hemorheological changes.  相似文献

The respiratory responses of substrate-depleted excised roottips to a range of sugars, sugar phosphates, and sugar alcoholshave been determined by measuring oxygen uptake by the directmethod of Warburg. Sucrose, dextrose, and laevulose are the only sugars which promotea high level of oxygen uptake. The effects of azide and DNP on the oxygen uptake promoted bysucrose and by dextrose are described. Mannose is a strong inhibitor of respiration. This inhibitionis reversed by the simultaneous addition of those sugars whichalso reverse the growth inhibition caused by mannose. Mannoseinhibits the respiration of sucrose and of glycolytic intermediates.Galactose is slowly respired and does not, even at high concentration,inhibit the respiration of sucrose. The results are discussed in relation to the growth effectsof the sugars tested.  相似文献

Biological availability of 23 alcohols, 16 aldehydes and 2 ketones was compared by the mini-test with chicks. Chicks can utilize methanol and ethanol, but not the alcohols of carbon chain from 3 to 9. Lauryl and myristyl alcohols were well utilized but those of higher carbon chain than 14 were not, mainly due to low digestibility. Glycerol and D-sorbitol were well utilized but not 4 other poly alcohols tested. Aldehydes and ketones with free carbonyl group showed low availability or even toxicity, but their derivatives with masked group showed no detrimental effect. Acetaldehyde, acetal and aldehydes of carbon chain longer than 9 were partially utilized.  相似文献
