AIMS: The aim of the study was to assess the impact of Salmonella present on the slaughter line before processing on broiler carcass contamination during processing. METHODS AND RESULTS: Three Belgian broiler slaughterhouses were each visited twice. Samples were taken from the slaughter line after the cleaning and the disinfection process and before slaughter of the first flock. During the slaughter of the first flock, feathers and neck skins were collected at various points of the slaughter process. Swab samples were also taken from the crates in which the birds were transported. In two slaughterhouses, the slaughter line was contaminated with Salmonella before the onset of slaughter, especially the shackles, conveyer belt and the plucking machine in the dirty zone. During slaughter, the carcasses of the first Salmonella-free flock became contaminated with the same strains as isolated previously from the slaughter line. CONCLUSION: Contamination of the slaughter line with Salmonella leads to carcass contamination. SIGNIFICANCE AND IMPACT OF THE STUDY: Implementation of logistic slaughter is only successful when the cleaning and disinfection process completely eliminates the Salmonella contamination of the slaughter line. Only if this is achieved, will the slaughter of Salmonella-free flocks result in the absence of Salmonella on the carcasses after slaughter.  相似文献

This study aimed to define the origin of Salmonella contamination on swine carcasses and the distribution of Salmonella serotypes in two commercial slaughterhouses during normal activity. Salmonellae were isolated from carcasses, from colons and mesenteric lymph nodes of individual pigs, and from the slaughterhouse environment. All strains were serotyped; Salmonella enterica serotype Typhimurium and Salmonella enterica serotype Derby isolates were additionally typed beyond the serotype level by pulsed-field gel electrophoresis (PFGE) and antibiotic resistance profiling (ARP); and a subset of 31 serotype Typhimurium strains were additionally phage typed. PFGE and ARP had the same discriminative possibility. Phage typing in combination with PFGE could give extra information for some strains. In one slaughterhouse, 21% of the carcasses were contaminated, reflecting a correlation with the delivery of infected pigs. Carcass contamination did not result only from infection of the corresponding pig; only 25% of the positive carcasses were contaminated with the same serotype or genotype found in the corresponding feces or mesenteric lymph nodes. In the other slaughterhouse, 70% of the carcasses were contaminated, and only in 4% was the same genotype or serotype detected as in the feces of the corresponding pigs. The other positive carcasses in both slaughterhouses were contaminated by genotypes present in the feces or lymph nodes of pigs slaughtered earlier that day or from dispersed sources in the environment. In slaughterhouses, complex contamination cycles may be present, resulting in the isolation of many different genotypes circulating in the environment due to the supply of positive animals and in the contamination of carcasses, probably through aerosols.  相似文献

AIMS: A comparison of Enterobacteriaceae, coliform and Escherichia coli counts in chicken carcasses with and without visible faecal contamination was conducted to evaluate the role of contamination as a vehicle for generic E. coli, coliform and other enterobacteria contaminating broiler chicken carcasses when processed under routine commercial operations. METHODS AND RESULTS: Samples were removed from the processing line immediately after evisceration, inside-outside shower and chilling for microbiological analysis. After evisceration, mean counts were significantly different only for E. coli (P < or = 0.05) in chicken carcasses with and without visible faecal contamination. While the spray wash practice was not efficient enough for complete removal of the visible contamination from carcasses, leading to microbiological reduction percentages lower than expected, 25 ppm chlorinated water chilling did reduce the contamination level considerably in all samples. CONCLUSIONS: Carcasses with and without visible faecal contamination harboured E. coli and other potentially hazardous enterobacteria. E. coli was the predominant strain isolated in all samples, Enterobacter cloacae being next most frequent. SIGNIFICANCE AND IMPACT OF THE STUDY: The zero tolerance of visible faecal contamination requirement alone is not sufficient to assure safety and to improve the microbial quality of carcasses.  相似文献

AIMS: The purpose of this study was to investigate the prevalence of Salmonella in pigs at the moment of slaughter and in the slaughterhouse environment. METHODS AND RESULTS: In total, five different commercial slaughterhouses were sampled during eight slaughterhouse visits. Carcass swabs, colon content and mesenteric lymph nodes were taken to reflect the animal status and from the slaughterhouse environmental samples were taken. Salmonella was isolated from 37% of the carcass samples as a mean value. High variations were noticed between different slaughterhouses (between 0 and 70%) and sampling days in the same abattoir (between 3 and 52%). A correlation was found between the carcass contamination and the status of the delivered animals (P=0.01675). Cross contamination was estimated to account for 29% of the positive carcasses. The slaughterhouse environment was highly contaminated; before starting the slaughtering activities 25% of the samples were positive on average. The most prevalent serotypes isolated at the slaughterhouse environment and from the colon content were S. Typhimurium, S. Livingstone and S. Derby. On carcasses S. Typhimurium was predominately isolated (71%). The biggest variability of serotypes was found in the mesenteric lymph nodes. Serologically 56.3% of the pigs were found positive for Salmonella using a cut-off level of the optical density percentage higher than 10 (O.D.% > or = 10). While on individual pig level the correlation between the bacteriological and serological data was poor, because of recent Salmonella infections, a better correlation was found at the herd level on the moment of slaughtering. CONCLUSION: A high degree of carcass contamination is noticed after slaughtering. This contamination resulted from the delivery of Salmonella-positive pigs and cross-contamination from the slaughterhouse environment. SIGNIFICANCE AND IMPACT OF THE STUDY: In pigs, Salmonella carriage is high, but it is obvious that slaughterhouse hygiene is a determinative factor for managing carcass contamination.  相似文献

Aims:  The present study aimed to assess the Arcobacter contamination on bovine carcasses postevisceration and postcooling in two slaughterhouses and in ready-to-eat minced beef.
Methods and Results:  Carcasses ( n  = 247) were sampled at four sites in two slaughterhouses and 100 minced beef samples were collected at retail. Isolation was performed by a quantitative and qualitative Arcobacter selective method, and the isolates were identified by multiplex PCR, after which a part of them were characterized by enterobacterial repetitive intergenic consensus (ERIC)-PCR. Although arcobacters were isolated from 37% of the bovine carcasses postevisceration with the chest and the foreleg as most contaminated sites, cooling the carcasses for at least 24 h reduced the incidence of Arcobacter (7%) on the carcass surface significantly. Arcobacter butzleri was the species most frequently isolated, although co-contamination with multiple species also occurred. At retail, arcobacters were present in 9% of the minced beef samples, with Arcobacter butzleri as the dominant species.
Conclusions:  Forced air cooling of bovine carcasses for at least 24 h decreased the number of positive carcasses, but did not eliminate all arcobacters.
Significance and Impact of the study:  This study demonstrates that maintaining good hygiene practices throughout the food supply chain is crucial to ensure safe food products at the consumer level.  相似文献

AIMS: A comparison of the prevalence of Salmonella in chicken carcasses with and without visible faecal contamination during commercial slaughter practice was made. The relationship between Enterobacteriaceae, coliform and Escherichia coli counts and Salmonella status was also evaluated to establish the likelihood of using these groups as 'index' organisms to predict the presence of pathogen. METHODS AND RESULTS: Samples were removed immediately after evisceration, after the inside-outside shower and after chilling from the processing line for microbiological analysis. Of the carcasses visibly uncontaminated with faeces after the evisceration step 20% harboured salmonellas and 20.8% of the visibly contaminated carcasses were positive for the pathogen. When E. coli, coliforms and Enterobacteriaceae were used as predictor variables the error rates ranged from 33.3 to 60% for both sample types. CONCLUSIONS: There was no indication that any of the groups of organisms analysed could predict the incidence of salmonellas on the samples studied. Positive results for the pathogen were obtained at every tested step of the slaughtering process regardless of whether or not faecal contamination was present. SIGNIFICANCE AND IMPACT OF THE STUDY: The present study demonstrated that carcasses not visibly contaminated with faeces carried Salmonella as well as the visibly contaminated carcasses.  相似文献

To investigate the microbiological conditions of hunted wild boar carcasses and factors that contribute to the microbial carcass contamination, skin and carcass meat swab samples from 210 hunted wild boars were collected from freshly shot animals. The mean aerobic colony counts (ACCs) and Enterobacteriaceae counts on the skin were 5.2 and 3.6 log10 CFU/cm2, with 1.4% of animals’ skin tested positive for Salmonella spp. Slightly higher mean ACC and Enterobacteriaceae counts of 5.4 and 3.8 log10 CFU/cm2 were obtained from carcass meat with Salmonella spp. prevalence of 1.9%. Inadequate hygiene practices in handling and dressing wild boar carcasses, such as evisceration in the laying position on the ground and practice of skin and interior carcass surface washing after evisceration, were found to have the most significant influence on the microbiological conditions of final carcasses. Therefore, these findings indicate the need for the implementation and strict adherence to good hygiene practice in hunting estates and game handling establishments.  相似文献

In this study of the behaviour of the fractions of unretted and frost-retted fibre straws in damp air, a production scale method to separate fibre and shive from fibre plants was introduced and tested on bast fibre plants (Linum usitatissimum L. and Cannabis sativa L.). The method consists of optional drying of stalks, unloading bales, milling the straws with a hammer mill, separating the fractions from air stream with a cyclone and finally separating fibres from shives with a screening drum. Fractions were characterized focusing on technologically indicative properties such as equilibrium moisture content, ash and microbiological quality. Unretted fractions of the bast fibre plant stem reached higher equilibrium moisture contents than the retted fractions, and hemp fibres absorbed more moisture from air than did the Linum fibres. In very humid air, all fractions began to lose weight due to moulding. The weight decrease during the first week was lower in frost-retted than in unretted fractions. The frost-retted fractions appeared to be more resistant to humidity in the short term. The total number of microbes and especially the numbers of yeasts and moulds can be used as a criterion of hygienic level. For green fractions, the mould level was similar in fibres and in shives, but frost-retted shives contained more moulds than the unretted shives. The mould content of a fraction had no direct correlation with the moulding tendency of the fraction. The ash contents of fibres were somewhat higher than those of shives, due to a probable soil contamination. Ash content did not have significant correlation with microbiological quality, although ash is a possible risk factor for hygienic quality. According to the results of this study it is highly important to study the quality of the production chain of bast fibre plants to ensure the quality of industrial products. From the producer's point of view, raw material with defined quality can be directed to the most suitable application. The behaviour of fractions in various ambient atmospheres, and other quality aspects such as hygienic level can be used as criteria for defining the most appropriate product applications.  相似文献

The bacteriological quality of beef and pig carcasses was assessed at 9 Norwegian abattoirs by sampling 10 carcasses at multiple sites on each of several visits. On beef carcasses the following sites consistently carried higher numbers of bacteria: the Brisket, the Fore-ribs, the Flank groin, and the Round medial. There was no evidence that beef slaughter and dressing in the hanging position was superior to methods where the carcass was lying until the hide puller. On pork carcasses the Cheek, and the Abdomen lateral surface (belly) were most heavily contaminated. The hygienic quality of pork carcasses in abattoirs where singeing was a separate step tended to be better than where a combined singeing and dehairing machine was used. This survey suggests that bacteriological monitoring of slaughter at this level of sampling and visiting is able to detect consistently poor hygienic practices. Where direct comparisons with data from other countries could be made, the present investigation indicates that the bacterial counts on Norwegian beef and pork carcasses are of the same order or better.  相似文献

Scanning electron microscopy was used in conjunction with normal microbiological procedures to evaluate the stomacher as a means of recovering bacteria from chicken skin. Stomaching removed most bacteria from the skin and viable counts of skin micro-organisms obtained by this method were not significantly different from those obtained by blending. The stomacher represents a useful alternative to blending techniques commonly used to assess contamination of poultry carcasses.  相似文献

Thermotolerant Campylobacter is among the more prevalent bacterial pathogens that cause foodborne diseases. This study aimed at evaluating the occurrence of thermotolerant Campylobacter contamination in chicken carcasses and processing plant stations (chilling water, scalding water, defeathering machinery, evisceration machine, and transport crates) in two of the Chilean main slaughterhouses. AIMS: To determine the prevalence of Salmonella enterica serovars in chicken carcasses in slaughterhouses in Spain and to examine genotypic relations among these serovars. METHODS AND RESULTS: A total of 336 chicken carcasses were collected from six slaughterhouses in Northwestern Spain. Salmonellae were isolated (ISO-6579-1993), serotyped, phage-typed, ribotyped and antibiotyped against 20 antibiotics. Salmonella strains were detected in 60 (17.9%) carcasses. Isolates belonged to nine different serotypes, with Salm. Enteritidis being the most common. Three strains (5%) were resistant to one antibiotic and 24 (40%) were multi-resistant (to more than one antibiotic). The most frequently encountered resistances were to sulphamides, fluoroquinolones and tetracycline. Ribotyping was able to differentiate isolates of the same serotype and phage type. CONCLUSIONS: The Salmonella serotypes and phage types detected are among those most frequently associated with human diseases in Spain. The large percentage of antimicrobial resistant strains is a matter for concern. A high genetic relationship between strains from different slaughterhouses was found. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provides detailed information about Salmonella isolates from poultry in Spain. It emphasizes the importance of controlling this pathogen in poultry products, and suggests the need for more prudent use of antibiotics.  相似文献

A study was conducted to determine the prevalence of Campylobacter with antimicrobial resistance from chickens, pigs, dairy cows, healthy farm workers, and children hospitalized with diarrhea in northern Thailand. Resistance was highest in pig samples and lowest in healthy farm workers. Resistance to fluoroquinolones and tetracycline was high in all study populations. The increased prevalence of resistant isolates from meat samples collected at markets, compared to isolates collected from animals on the farm or the slaughterhouse, suggests that contamination after carcasses leave the slaughterhouse is an important factor in the spread of resistant bacteria into the human food chain.  相似文献

Pseudomonas spp. in Wiltshire bacon cover brines taken from seven bacon factories were enumerated at regular intervals over six months. There were differences between factories as assessed by the geometric mean counts (12–2907/ml) or by the percentage of samples exceeding 103/ml (0–75%). Heavy pseudomonad contamination of brines was associated with large populations of Pseudomonas spp. on fresh pork carcasses and vice versa. The curing of defrosted pork carcasses also led to high Pseudomonas spp. counts in brines. The average time for 90% reduction in a Pseudomonas population in brine was 35 d. These data can aid interpretation of the Pseudomonas count of bines when it is used as a part of microbiological quality control. A set of advisory standards proposed.  相似文献

AIMS: Bovine meat is the principal source of human contamination of attaching and effacing Escherichia coli, including enterohaemorrhagic E. coli O157. The aim was to study the prevalence of these strains on bovine carcasses in Algeria. METHODS AND RESULTS: Two-hundred and thirty carcasses were swabbed and analysed by classical microbiological methods for total E. coli counts and for the presence of pathogenic E. coli. The E. coli counts were high, with a 75th percentile of 444.75 CFUs cm(-2). For pathogenic E. coli, more than 7% of the tested carcasses were positive for E. coli O157. Eighteen E. coli O157 strains were isolated and typed by multiplex PCR. The main isolated pathotype (78%) was eae+ stx2+ ehxA+. In addition to E. coli O157, other attaching and effacing E. coli (AEEC) were also detected from carcasses by colony hybridization after pre-enrichment and plating on sorbitol MacConkey agar using eae, stx1 and stx2 probes. Thirty carcasses (13%) on the 230 analysed harboured at least one colony positive for one of the tested probes. These positive carcasses were different from those positive for E. coli O157. Sixty-six colonies (2.9%) positive by colony hybridization were isolated. The majority (60.6%) of the positive strains harboured an enteropathogenic E. coli-like pathotype (eae+ stx-). Only three enterohaemorrhagic E. coli (EHEC)-like (eae+ stx1+) colonies were isolated from the same carcass. These strains did not belong to classical EHEC serotypes. CONCLUSIONS: In this study, the global hygiene of the slaughterhouse was low, as indicated by the high level of E. coli count. The prevalence of both E. coli O157 and other AEEC was also high, representing a real hazard for consumers. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first study of this type in Algeria, which indicates that the general hygiene of the slaughterhouse must be improved.  相似文献

Aerobic plate counts (APC 37°C and APC 25°C) and Escherichia coli enumerations (Petrifilm) were used to determine sources of bacterial contamination during sheep dressing, determine the hygienic efficacy of hand wash and knife 'sterilization'procedures and compare the hygiene efficiency of conventional and inverted sheep dressing systems. The major slaughterline sources of microbial contamination were: fleece > workers' hands > faecal pellets > knife blades. Aerobic plate counts (APC 37°C) exceeding log 4.4 cfu cm-2 were considered indicative of direct fleece contact, whereas E. coli numbers exceeding log 3.3 cfu cm-2 were considered indicative of direct faecal contact. A 44°C water hand rinse removed 90% of the microbial contamination from workers' hands, but rinsed hands, particularly those contacting the fleece, still carried a microbial population exceeding log 4.0 cfu cm-2. A 44°C rinse followed by an 82°C water dip reduced the contamination on knife blades to less than log 3.0 cfu cm-2. Inverted dressing systems produced carcasses with a lower contamination level than conventional systems. With both systems little increase in contamination occurred after pelt removal. The areas of highest contamination were the forequarter region with inverted dressing and the hindquarter with conventional dressing. In both cases these regions are the sites where cuts are made through the skin. With both systems contamination around these cuts was entirely consistent with direct fleece contact resulting from 'rollback'.  相似文献

AIMS: To identify sources of Escherichia coli on beef by characterizing strains of the organism on animals, equipment and product at beef-packing plant. METHODS AND RESULTS: Generic E. coli were recovered from hides, carcasses, beef trimmings, conveyers and ground beef during the summer of 2001 (750 isolates) and winter of 2002 (500 isolates). The isolates were characterized by Random Amplification of Polymorphic DNA (RAPD). The numbers of E. coli recovered from dressed carcasses were less than the numbers recovered from hides. The numbers recovered from chilled carcasses were too few for meaningful analysis of the strains present on them but the numbers recovered from trimmings and ground beef were larger. The RAPD patterns showed that the majority of isolates from hides, carcasses, beef trimmings, conveyers and ground beef were of similar RAPD types, but a few unique RAPD types were recovered from only one of those sources. The E. coli populations present on the hides of incoming animals and in the beef-processing environment were highly diverse. Randomly selected E. coli isolates from each of the five sources were further characterized by pulsed-field gel electrophoresis (PFGE). Most genotypes of E. coli defined by PFGE corresponded to the E. coli types defined by RAPD. CONCLUSIONS: The hides of the incoming animals appeared to be only one of the sources of the E. coli on trimmings and in ground beef, as additional sources were apparently present in equipment used for carcass breaking. SIGNIFICANCE AND IMPACT OF THE STUDY: This study indicates that hazardous microbiological contamination of meat may occur after the dressing of carcasses at commercial beef-packing plants, which suggests that attention should be given to the control of the contamination of meat during carcass breaking as well as during the dressing of carcasses.  相似文献
