为了探索联合接种甲乙肝疫苗、实验性甲乙肝联合疫苗免疫恒河猴的安全性及免疫原性。实验中挑选了甲肝抗体阴性,乙肝两对半阴性,肝功能指标正常的健康恒河猴24只,随机分为10组。混合或分别接种,进行不同毒株的甲肝灭活疫苗与不同厂家的乙肝疫苗的配对效果比较。并接种了实验性甲乙肝联合疫苗;史克甲乙肝联合疫苗试验组。设甲肝单价灭活疫苗L8株、深圳乙肝单价疫苗作为对照。免疫方案0、4、24w。每只恒河猴接种lml。接种3d内,每天观察动物有无不良反应。接种1针和2针后4w内,每2w采集空腹静脉血,以后每4w采血1次检测抗HAV、抗HBs、ALT、AST直至40w。接种疫苗后4、8、24、28w穿刺肝组织做病理学检查。结果显示接种疫苗后3d内,所有恒河猴均无不良反应。ALT、AST无异常升高。4、8、24、28w肝组织无特殊病理改变。注射2针后4w,除3组外,其余各组抗HAV及抗HBs均阳转。3组抗HAV阳转时间迟至12w。全程免疫后12w(即40w),抗HAVGMT为258.75~37489.50mIU/ml;抗HBsGMT为8263.68~60008.064mIU/ml。甲乙肝疫苗联合免疫及实验性的甲乙肝联合疫苗接种恒河猴安全性及免疫原性均良好。  相似文献

目的采用动物实验及临床观察,探讨益生菌制剂培菲康治疗肝硬化高氨血症的作用机制。方法采用大鼠肝硬化模型,其中治疗组予以培菲康悬液每天灌胃治疗共8周,检测治疗组及对照组血浆内毒素指标;并选用70例肝硬化Child-Pugh分级为B级的患者,其中治疗组口服培菲康胶囊(每次2粒,每日3次)3周,观察治疗前后血浆内毒素变化。结果培菲康治疗组的肝硬化大鼠及患者血浆内毒素均明显低于对照组(P<0.05)。结论培菲康能明显降低肝硬化患者的血氨水平,可作为肝硬化患者的辅助用药。  相似文献

A total of 337 liver abscesses of Awassi sheep that were found in different slaughterhouses in Jordan were examined for their causal agents. Fifteen different bacterial species were isolated from 297 liver abscesses. No bacteria were isolated from the remaining 40 liver abscesses. Fusobacterium necrophorum biovar B were isolated from 195 (58%) abscesses. Arcanobacterium pyogenes, F. necrophorum biovar A, Escherichia coli and Clostridium perfringens were isolated from 41 (12%), 34 (10%), 30 (9%) and 17 (5%), respectively, liver abscesses. Our study suggested that F. necrophorum biovar B is the most prevalent bacterium incriminated for liver abscesses in Awassi sheep.  相似文献

Power-frequency electromagnetic fields (PF-EMFs) at 50 Hz are potential health risk factors. This study aimed to explore the effects of long-term exposure to 50-Hz PF-EMFs on general physiological conditions in Sprague–Dawley (SD) rats. During a 24-week exposure period, the body mass and water and food intake of the animals were recorded regularly. The hematologic parameters were detected every 12 weeks, and blood chemistry analyses were performed every 4 weeks. After sacrifice, morphology was identified by hematoxylin–eosin, Masson, and immunohistochemical staining. Fibrosis-related gene expression and oxidative stress status were also detected. Compared with the control group, exposure to 30, 100, or 500 μT PF-EMF did not exert any effect on body mass, food intake, or water intake. Similarly, no significant differences were found in hematologic parameters or blood chemistry analyses among these groups. Furthermore, morphological assays showed that exposure to PF-EMFs had no influence on the structure of the liver or kidney. Finally, fibrosis-related gene expression and oxidative stress status were unaltered by PF-EMF exposure. The present study indicates that 24 weeks of exposure to PF-EMFs at intensities of 30, 100, or 500 μT might not affect hemograms, blood chemistry, fibrosis, or oxidative stress in the liver or kidney in SD rats. © 2020 Bioelectromagnetics Society  相似文献

Restriction fragment length polymorphism analysis of rRNA genes was employed to genetically compare Fusobacterium necrophorum subsp. necrophorum and F. necrophorum subsp. funduliforme isolates from multiple abscesses of the same liver and isolates from liver abscesses, the ruminal wall, and ruminal contents from the same animal. Four livers with multiple abscesses and samples of ruminal contents, ruminal walls, and liver abscesses were collected from 11 cattle at slaughter. F. necrophorum was isolated from all liver abscesses, nine ruminal walls, and six ruminal content samples. Chromosomal DNA of the isolates was extracted and single or double digested with restriction endonucleases (EcoRI, EcoRV, SalI, and HaeIII); then restriction fragments were hybridized with a digoxigenin-labeled cDNA probe transcribed from a mixture of 16S and 23S rRNAs from Escherichia coli. EcoRI alone or in combination with EcoRV yielded the most discriminating ribopatterns for comparison. Within the subspecies multiple isolates from the same liver were indistinguishable based on the ribopattern obtained with EcoRI. The hybridization patterns of liver abscess isolates were concordant with those of the corresponding isolates from ruminal walls in eight of nine sets of samples. None of the six ruminal content isolates matched either the liver abscess isolates or the ruminal wall isolates. The genetic similarity between the isolates from liver abscesses and ruminal walls supports the hypothesis that F. necrophorum isolates of liver abscesses originate from the rumen.  相似文献

The biodistribution of long-circulating PEG-liposomes in a subcutaneous mouse model of established mixed infection abscesses was investigated to assess their possible role as drug carriers in the treatment of small, undrainable intra-abdominal abscesses. There was a 10-30-fold greater localisation of (67)Ga-labelled PEG-liposomes in abscesses compared to uninfected normal skin samples. Over 3% of the injected dose (ID) of liposomes was present in the abscesses 24 h after liposome administration in contrast to 0.1% in normal skin sections. The percentage ID present in the liver, spleen and kidneys was 17%, 4% and 2% per organ respectively. Five days after liposome injection, 2% ID could still be recovered from the abscesses. Using colloidal gold-labelled PEG-liposomes, it was shown that there was a 4-fold greater density of liposome clusters in the subcutaneous tissue surrounding the capsule than in the core of the abscesses. The clusters within the abscesses were distributed evenly. We conclude that PEG-liposomes localise to a significant degree at the infection focus in our mouse model and may provide a new approach to the antimicrobial treatment of intra-abdominal abscesses.  相似文献

An attempt was made to determine the time of onset of liver abscesses in intensively fed calves. Determinations of seram proteins (total protein, albumin, globulin, albumin/ globulin ratio, agarose-gel electrophoretic separation and formol-gel test) and, in a part of the material, of serum-GOT and -GPT were made in serial samples taken once a month from 125 calves. Liver abscesses were found in 48 (38.4 %) when slaughtered at 6–7 months of age. Total protein and globulin increased, the albumin-globulin ratio decreased with increasing age. Calves with liver abscesses had a significantly (0.05 > P > 0.01) higher globulin concentration at the last sampling. At this sampling the material was of a limited size and the difference has to be interpreted with caution. No other significant differences were found. Nor were any rises observed in S-GOT and S-GPT referable to the formation of liver abscesses. Bacteriological studies were not made but, in analogy with observations on a similar animal material in another part of Sweden, the infectious agent in the liver abscesses was assumed to be Sph. necrophorus. The explanation of the almost absent changes in the serum proteins in calves with liver abscesses may be the low antigenicity of this bacterium. In conclusion it is stated that the present serial investigations of the serum protein pattern in young cattle provide no definite guidance for establishing the time of onset of liver abscesses.  相似文献

Sixteen patients with pyogenic liver abscesses were studied over 10 years to discover the causative organisms of the condition. Pus was subjected to Gram-negative smear or gas-liquid chromatography to detect volatile acids characteristic of anaerobes and then cultured. All isolates were identified by conventional methods and tested for sensitivity to appropriate antimicrobial agents. Bacteria were grown from the liver abscesses in all 16 patients. Streptococcus milleri Lancefield group F was the commonest organism isolated from the pyogenic liver abscesses, being found in 13 patients. If Strep milleri is isolated care should be taken not to mistake it for an anaerobe, and finding the organism in the blood should alert the clinician to the possible presence of a liver abscesses.  相似文献

The effect of aging and development in male Wistar-derived rats on the profile of endogenous metabolites excreted in the urine was investigated using both (1)H NMR spectroscopy and HPLC-TOF MS using electrospray ionisation (ESI). The endogenous metabolites were profiled in samples collected from male rats every two weeks from just after weaning at 4 weeks up to 20 weeks of age. Multivariate data analysis enabled clusters to be visualised within the data according to age, with urine collected at 4 and 6 weeks showing the greatest differences by both analytical techniques. Markers detected by (1)H NMR spectroscopy included creatinine, taurine, hippurate and resonances associated with amino acids/fatty acids, which increased with age, whilst citrate and resonances resulting from glucose/myoinositol declined. A number of ions were detected by HPLC-MS that were only present in urine samples at 4 weeks of age in both positive and negative ESI, with a range of ions, including e.g. carnitine, increasing with age. Age predictions by PLS-regression modelling demonstrated an age-related trend within these data, between 4 and 12 weeks for HPLC-MS and 4-16 weeks for NMR. The possible utility of these techniques for metabonomic investigations of age-related changes in the rat is discussed and the importance of employing suitable control animals in pharmacological and toxicological studies is highlighted.  相似文献

Elucidation of the mechanisms of liver fibrogenesis is important to treat liver fibrosis.In this study,we established rat models of liver fibrosis with stages from 0–1,2,and 3–4 to 4 at 2,4,6,and 8 weeks,respectively,by injection of pig serum.Liver fibrogenesis was detected by Masson’s trichrome staining.Rat non-parenchymal cells(NPCs)were enriched 4-fold by Percoll density gradient centrifugation.Protein extracts from NPCs were prepared at 4 and 8 weeks,separated by two-dimensional electrophoresis,and then stained with Coomassie Blue G-250.At 4 weeks,we identified 18 non-redundant differentially expressed proteins of which protein disulfide-isomerase associated protein 3(PDIA3)and NDUV showed consistent expression at protein and mRNA levels from 4 to 8 weeks.PDIA3 was found to be down-regulated by Western blotting in the rat model and immunohistochemically in human liver.Our results revealed important aspects of the pathogenesis/progression of liver fibrosis and demonstrated important changes in protein expression levels of NPCs at various stages of liver fibrosis.  相似文献

Sphaerophorus necrophorus (bovine liver abscess isolates) antiserum was fractionated and labeled with fluorescein isothiocyanate. The fluorescent-antibody (FA) conjugate proved to be species-specific and facilitated the detection of S. necrophorus cells in liver abscesses, viscera, and ruminal contents of cattle. Brightly fluorescing S. necrophorus cells were observed in experimentally inoculated soil incubated anaerobically at 37 and 4 C for 8 and 10 months, respectively. When incubated under moist conditions (80% water holding capacity) at 37 C, the test organism survived in both sterile and unsterile soil for as long as 8 weeks. Results strongly support the feasibility of using FA techniques concurrent with cultural methods for rapid detection of S. necrophorus infections.  相似文献

Antimicrobial therapy without surgical drainage or therapeutic aspiration was effective in the management of four patients with deep abscesses ranging in diameter from 1.3 to 10.0 cm. Two of the patients had multiple hepatic abscesses, one had hepatic, intra-abdominal and intrapelvic abscesses, and one had an intrapelvic abscess alone. Anaerobic bacteria were isolated from the blood or abscesses in all four patients, and an aerobic-anaerobic infection was present in one patient. The patients were treated with metronidazole, alone or in combination with other antibiotics, for 3 to 6 weeks. Therefore, in selected patients with deep abscesses, a therapeutic trial of antimicrobial agents instead of surgery may be justified.  相似文献

报道了用戊型肝炎(HepatitisE,HE)病人粪便悬液感染恒河猴后的组织病理学、血液生化与免疫学以及病毒学分子生物学检测的结果。三只实验猴在感染后第3~4周均出现ALT异常;粪便以及肝脏与胆囊组织超薄切片中电镜观察到27~34nm大小的病毒样颗粒;病理组织切片观察表明,肝脏组织有典型的急性炎症病灶;粪便与血清经RTnPCR扩增到戊型肝炎病毒(HepatitisEVirus,HEV)特异性片段,粪便排毒从感染后第7天持续至第50天左右,病毒血症迟于粪便排毒,出现于感染后两周左右,维持1~2周;ELISA检测发现,实验猴血清中HEVIgG抗体水平在感染后3~4周阳转,4~5个月后转阴。这些实验结果提示,恒河猴作为HEV感染实验动物模型是理想的,建立系统的恒河猴实验模型对探讨HEV感染发病机理、机体免疫应答以及临床诊断与疫苗研制具有重要意义。  相似文献

目的:检测Toll样受体4(TLR4)在四氯化碳诱导的肝硬化大鼠骨髓血窦内皮细胞的表达,为进一步研究肝硬化时骨髓损伤的发生机制提供实验依据。方法:选择Wistar大鼠给予腹腔注射CCl4,一周两次,建立肝硬化大鼠模型。分别于建模8周和12周检测大鼠血浆内毒素的水平,免疫组化检测大鼠骨髓血窦内皮细胞上TLR4的表达情况,RT-PCR测定骨髓组织中TLR4mRNA的表达,分析TLR4的表达与内毒素血症间的关系。结果:给予CCl4 8周和12周时,对照组大鼠血浆内毒素水平分别为(0.216±0.024)Eu/ml和(0.133±0.022)Eu/ml,模型组大鼠血浆内毒素水平分别为(0.626±0.021)Eu/ml和(0.725±0.031)Eu/ml,分别较对照组显著升高,差异均有统计学意义(P〈0.001);骨髓血窦内皮细胞TLR4蛋白表达及骨髓组织中TLR4mRNA的表达均显著高于对照组,差异均有统计学意义(P〈0.05)。大鼠骨髓TLR4蛋白和mRNA表达与血浆内毒素水平均呈显著正相关(r=0.841,0.803,P均〈0.001)。结论:CCl4诱导的肝硬化大鼠骨髓血窦内皮细胞TLR4表达升高,并伴随大鼠内毒素血症的发生,提示肝硬化时肠源性内毒素血症可能参与了骨髓的造血功能的损害和病变。  相似文献
