目的:探讨食管癌组织微小RNA-21(miR-21)、微小RNA-182(miR-182)表达与临床病理特征及预后的关系。方法:选择2010年10月至2014年2月期间在我院治疗的120例食管癌患者为研究对象,采集患者的食管癌组织及癌旁组织,采用荧光定量PCR检测组织中miR-21、miR-182表达量,采用Kaplan-Meier法分析患者生存情况。结果:与癌旁组织相比,食管癌组织中miR-21、miR-182表达量明显升高(P<0.05)。食管癌组织中miR-21、miR-182表达均与TNM分期和淋巴结转移相关(P<0.05)。miR-21低表达患者的5年生存率明显高于高表达患者(P<0.05),miR-182低表达患者的5年生存率明显高于高表达患者(P<0.05)。结论:miR-21、miR-182表达量在食管癌中上调,与食管癌TNM分期和淋巴结转移相关。miR-21高表达以及miR-182高表达患者5年生存率下降,检测miR-21、miR-182表达量在食管癌患者预后预测中具有一定临床意义。  相似文献

BackgroundBreast cancer is an extensively identified malignant tumor and is a prime cause of cancer mortalities in females. It has been shown that alteration of miRNAs expression (up or down regulation) can affect the initiation and progression of many malignancies. We aimed to evaluate the role of circulating miRNA-148a and miRNA-30c in female patients with breast cancer and estimate their usage as potential biomarkers in the diagnosis, prognosis and survival of breast cancer.MethodsThis study included 75 breast cancer female patients.They were compared with 55 apparently healthy female subjects. miRNAs expression analysis was assessed via real-time PCR.ResultsTo discriminate breast cancer patients from controls, miR-30c showed the best performance at a cut off value of ≤20.6 (AUC = 0.998, 97.33% sensitivity, 96.36% specificity, p < 0.001), followed by miR-148a (AUC = 0.995, 94.67% sensitivity, 90.91% specificity, p < 0.001 at a cut off value of ≤0.1), CA 15-3 (AUC = 0.930, 88.0% sensitivity, 81.82% specificity, p < 0.001 at a cut off value of >21.3), and finally CEA (AUC = 0.751, 70.67% sensitivity, 63.64% specificity, p < 0.001 at a cut off value of >2.5).ConclusionmiRNA-148a and miRNA-30c expressions were down regulated in female patients with breast cancer and might be considered as potential blood biomarkers. Both also might have rule in disease treatment and selection of therapeutic targets. Future studies are needed to improve their role in predicting response to treatment and prognosis.  相似文献

目的:探讨窒息新生儿血清mi RNA-21、mi RNA-210及mi RNA-199a的表达及临床意义。方法:选取2015年9月1日到2017年7月31日我院新生儿科收治的窒息新生儿40例作为观察组,另选取同期在我院出生的健康新生儿40例作为对照组,根据阿氏(Apgar)评分将观察组的新生儿分为重度窒息组(12例)和轻度窒息组(28例)。检测所有新生儿血清中mi RNA-21、mi RNA-210及mi RNA-199a的表达水平,并比较观察组与对照组、重度窒息组与轻度窒息组血清中mi RNA-21、mi RNA-210及mi RNA-199a水平,并分析其相关性。结果:观察组的血清mi RNA-21、mi RNA-210水平高于对照组,mi RNA-199a水平低于对照组,差异有统计学意义(P0.05)。重度窒息组的血清mi RNA-21、mi RNA-210水平均高于轻度窒息组,mi RNA-199a水平低于轻度窒息组,差异有统计学意义(P0.05)。观察组血清mi RNA-21与mi RNA-210呈正相关(P0.05),血清mi RNA-21、mi RNA-210与mi RNA-199a水平呈负相关(P0.05)。结论:窒息新生儿血清中mi RNA-21、mi RNA-210呈现高表达,mi RNA-199a呈现低表达,且其表达水平与窒息严重程度相关。  相似文献

Objective

Metastasis is the major cause of death in colorectal cancer patients. Expression of certain miRNAs in the primary tumors has been shown to be associated with progression of colorectal cancer and the initiation of metastasis. In this study, we compared miRNA expression in primary colorectal cancer and corresponding liver metastases in order to get an idea of the oncogenic importance of the miRNAs in established metastases.

Methods

We analyzed the expression of miRNA-21, miRNA-31 and miRNA-373 in corresponding formalin-fixed paraffin-embedded (FFPE) tissue samples of primary colorectal cancer, liver metastasis and healthy tissues of 29 patients by quantitative real-time PCR.

Results

All three miRNAs were significantly up-regulated in the primary tumor tissues as compared to healthy colon mucosa of the respective patients (p < 0.01). MiRNA-21 and miRNA-31 were also higher expressed in liver metastases as compared to healthy liver tissues (p < 0.01). No significant difference of expression of miRNA-31 and miRNA-373 was observed between primary tumors and metastases. Of note, miRNA-21 expression was significantly reduced in liver metastases as compared to the primary colorectal tumors (p < 0.01).

Conclusion

In the context of previous studies demonstrating increased miRNA-21 expression in metastatic primary tumors, our findings raise the question whether miRNA-21 might be involved in the initiation but not in the perpetuation and growth of metastases.  相似文献

目的:采用荧光定量PCR法检测前列腺癌循环miRNA-141表达,并研究五苓胶囊对其影响。方法:选取我院泌尿外科收治的前列腺癌患者20例,随机分为A、B两组,各10例;A组给予常规治疗,B组在常规治疗基础上给予加用五苓胶囊治疗。选取同期住院前列腺增生患者20例,正常对照组为同时期体检健康者20例。采用荧光定量PCR法检测正常对照组、前列腺增生组以及前列腺癌组血清miRNA-141的表达情况,以及前列腺癌组治疗前TPSA(前列腺特异性抗原)含量。结果:前列腺癌组的血清miRNA-141的表达水平为正常对照组的(11.22±8.19)倍,明显高于前列腺增生组(1.65±1.73)和正常对照组,差异有统计学意义(P0.05);前列腺增生组和正常对照组比较无明显差异(P0.05);前列腺癌组血清miRNA-141的表达水平与TPSA水平呈正相关关系,相关系数r=0.753,有统计学意义(P0.05)。治疗后,前列腺癌A组与B组的血清miRNA-141的表达水平均有所改善,与A组(5.82±4.51)相比较,B组的miRNA-141的表达水平(1.93±0.67)明显降低,差异有统计学意义(P0.05)。结论:前列腺癌患者血清中miRNA-141表达水平明显升高,且与TPSA水平呈正相关关系,五苓胶囊能够降低miRNA-141表达。  相似文献

目的:探讨microRNA-21表达水平对新疆地区食管癌早期诊断及预后评估的临床价值。方法:收集我院收治的100例食管癌患者,其中哈萨克族患者50例,汉族患者50例,采用RT-PCR以及RT-qPCR的方法对患者血清microRNA-21表达水平进行检测并比较。结果:食管癌患者癌组织中microRNA-21表达水平均高于癌旁组织,差异具有统计学意义(P0.05);有淋巴结转移、ⅡB+Ⅲ期、低分化癌组织中的miRNA-21相对表达量高于无淋巴结转移、I+ⅡA期以及高分化癌组织患者,差异具有统计学意义(P0.05);miRNA-21在不同民族、性别、年龄分组中表达无明显差异(P0.05)。结论:microRNA-21表达水平对新疆地区食管癌患者癌组织中表达水平较高,在有淋巴转移、ⅡB+Ⅲ期以及低分化癌组织中表达水平较高,民族、性别以及年龄对microRNA-21水平的影响较小,因此microRNA-21检测对食管癌的早期诊断及预后判断具有指导意义。  相似文献

MicroRNA(miRNA)是一类非编码小分子RNA,参与基因转录后水平调控,与肿瘤的形成、侵袭及转移、治疗及预后过程密切相关。研究发现,miRNA-200家族包括miR-141、miR-200a、miR-200b、miR-200c和miR-429在卵巢癌中的表达改变明显,且参与卵巢癌的发生、侵袭转移、卵巢癌细胞对紫杉醇的敏感性及复发过程,可能是用于卵巢癌的早期诊断、治疗、预后预测的指标。本文主要对miRNA-200家族在卵巢癌中的研究进展进行综述。  相似文献

目的:观察血浆miRNA-1、miRNA-21在冠心病(CHD)与无冠脉病变患者中含量的差异,探讨其在CHD合并糖尿病(DM)患者的经皮冠状动脉介入治疗(PCI)后发生支架内再狭窄(ISR)与非支架内再狭窄(NISR)患者的含量差异及其对ISR的预测价值。方法:入选CHD支架置入术后患者(CHD组,n=187)、无冠脉病变患者(对照组,n=195)。根据指南,将对照组分为正常对照组(n=150)、单纯DM组(n=45);将CHD组分为单纯CHD组(n=119)、CHD合并DM组(n=68),又将CHD组分为ISR组(n=48)及NISR组(n=139);再将ISR组分为单纯ISR组(n=26)和ISR合并DM组(n=22)。收集各组患者血浆,提取总miRNA,检测miRNA-1、miRNA-21含量并分析各组的含量差异。结果:与对照组比较,CHD组miRNA-1、miRNA-21含量均升高(P<0.05);与NISR组比较,ISR组miRNA-1、miRNA-21含量均升高(P<0.05)。与单纯CHD组比较,CHD合并DM组ISR发生率明显升高;而且与单纯ISR组比较,ISR合并DM组miRNA-21含量升高(P<0.05),而单纯ISR组与ISR合并DM组miRNA-1含量无明显差异(P<0.05)。Logistics分析表明,与CHD患者相关危险因素DM、miRNA-1、miRNA-21的OR值分别为2.132,3.066,1.924(P<0.05,P<0.01);ISR患者相关危险因素DM、miRNA-21的OR值分别为3.091,5.654(P<0.05,P<0.01);CHD合并DM患者PCI术后发生ISR相关危险因素miRNA-21的OR值为9.148(P<0.01)。ROC曲线评价表明,miRNA-1,miRNA-21与CHD患者发生ISR的AUC分别为0.854,0.857;miRNA 21与CHD合并DM患者发生ISR的AUC为0.783。结论:血浆miRNA-1、miRNA-21对诊断CHD患者支架置入术后ISR发生有重要临床意义,miRNA-21对诊断CHD合并DM患者支架置入术后ISR发生尤其有重要临床意义。  相似文献

目的:探讨老年非小细胞肺癌(non-small cell lung cancer,NSCLC)患者抗癌治疗前血浆纤维蛋白原(fibrinogen,FIB)和D-二聚体(D-dimer)的预后意义。方法:测定97例肺癌患者(肺癌组)及36例健康体检者(对照组)血浆D-dimer、FIB水平并进行比较,并分析其与NSCLC临床病理因素之间关系及预后价值。结果:肺癌组血浆FIB、D-dimer水平高于健康对照组(P0.05)。肺癌组FIB与TNM分期有关,D-dimer与淋巴结转移和TNM分期有关。单因素分析提示FIB、D-dimer、肿瘤大小、淋巴结转移、TNM分期与总体生存时间(overall survival,OS)和无进展生存时间(progression free survival,PFS)相关,而多因素分析仅提示D-dimer、FIB是老年NSCLC患者的独立危险预后因素。结论:检测老年NSCLC患者抗癌治疗前纤维蛋白原和D-二聚体可以指导预后,为肺癌个体化治疗提供一定的参考价值。  相似文献

To investigate the global expression profile of miRNAs in primary breast cancer (BC) and normal adjacent tumor tissues (NATs) and its potential relevance to clinicopathological characteristics and patient survival, the genome-wide expression profiling of miRNAs in BC was investigated using a microarray containing 435 mature human miRNA oligonucleotide probes. Nine miRNAs of hsa-miR-21, hsa-miR-365, hsa-miR-181b, hsa-let-7f, hsa-miR-155, hsa-miR-29b, hsa-miR-181d, hsa-miR-98, and hsa-miR-29c were observed to be up-regulated greater than twofold in BC compared with NAT, whereas seven miRNAs of hsa-miR-497, hsa-miR-31, hsa-miR-355, hsa-miR-320, rno-mir-140, hsa-miR-127 and hsa-miR-30a-3p were observed to be down-regulated greater than twofold. The most significantly up-regulated miRNAs, hsa-mir-21 (miR-21), was quantitatively analyzed by TaqMan real-time PCR in 113 BC tumors. Interestingly, among the 113 BC cases, high level expression of miR-21 was significantly correlated with advanced clinical stage (P = 0.006, Fisher's exact text), lymph node metastasis (P = 0.007, Fisher's exact text), and shortened survival of the patients (hazard ratio [HR]=5.476, P < 0.001). Multivariate Cox regression analysis revealed this prognostic impact (HR=4.133, P = 0.001) to be independent of disease stage (HR=2.226, P = 0.013) and histological grade (HR=3.681, P = 0.033). This study could identify the differentiated miRNAs expression profile in BC and reveal that miR-21 overexpression was correlated with specific breast cancer biopathologic features, such as advanced tumor stage, lymph node metastasis, and poor survival of the patients, indicating that miR-21 may serve as a molecular prognostic marker for BC and disease progression.  相似文献

MicroRNAs (miRNAs) play a critical role in gastric cancer progression and metastasis. This study investigated the role of miRNA-135a in early gastric cancer (EGC) including lymph node (LN) metastasis. We examined the correlation between miRNA-135a expression and clinical outcomes in 59 patients who underwent surgery for EGC. Using gastric cancer cell lines, we performed functional and target gene analyses. miRNA-135a expression was down-regulated in 33.9% of patients. These patients showed a significantly more advanced stage (TNM stage≥IB, 35.0% vs. 12.8%, p = 0.045) and higher rate of LN metastasis (30.0% vs. 5.1%, p = 0.014) than those with up-regulation of miRNA-135a expression. In a multivariate analysis, down-regulation of miRNA-135a was an independent risk factor for LN metastasis (adjusted odds ratio, 8.04; 95% confidence interval, 1.08–59.81; p = 0.042). Functional analyses using gastric cancer cell lines showed that miRNA-135a suppressed cell viability, epithelial-mesenchymal transition, cell invasion, and migration. ROCK1 was a target of miRNA-135a and its expression was inversely correlated to that of miRNA-135a. ROCK1 expression was significantly increased in EGC patients with LN metastasis than in those without LN metastasis. Our results confirm the tumor-suppressive role of miRNA-135a, and demonstrate its role in LN metastasis in EGC. miRNA-135a and its target gene ROCK1 may be novel therapeutic and prognostic targets for EGC.  相似文献

目的:探讨基于CT血管成像的血流储备分数对肺癌淋巴结转移的诊断价值。方法:2018年1月到11月选择在本院进行诊治的肺癌患者60例,所有患者都给予常规CT检查与CT血管成像,记录成像特征与相关血流储备分数-血流量(blood flow,BF)、血容量(blood volume,BV)、平均通过时间(mean transit time,MTT),判断诊断价值。结果:在60例患者中,病理确诊为淋巴结转移20例,非淋巴结转移40例。转移组的毛刺征、分叶征、棘突征、空泡征等CT征象发生率显著高于非转移组(P0.05)。转移组的肺动脉BF、BV值显著低于非转移组(P0.05),MTT值显著高于非转移组(P0.05)。在60例患者中,Spearman相关分析显示淋巴结转移与MTT成显著正相关性(P0.05),与BF、BV值成显著负相关性(P0.05)。结论:CT血管成像在肺癌中的应用能反映患者的血流储备分数状况,有利于判断患者的淋巴结转移情况,有很好的应用价值。  相似文献

【目的】通过高通量测序的方法获得PCV2感染3D4/21细胞的miRNAs表达谱,并探讨miRNA-98在PCV2复制中的作用。【方法】本研究以猪肺泡巨噬细胞系3D4/21细胞为细胞模型,对PCV2感染过程中的3D4/21细胞进行miRNAs差异表达分析,筛选与病毒复制相关的特异性miRNAs,并探讨其在PCV2复制中的作用。【结果】经高通量测序,获得PCV2感染3D4/21细胞的miRNAs表达谱,结合实验室前期研究筛选获得miRNA-98。实验表明,miRNA-98的表达量随PCV2感染时间的延长而持续升高,其变化趋势与Cap蛋白表达变化基本一致,由此推测miRNA-98与PCV2复制正相关。过表达miRNA-98可显著上调Cap蛋白的表达量和PCV2的复制。进一步的研究表明,miRNA-98参与调节宿主免疫相关细胞因子的表达和PCV2的复制。【结论】miRNA-98可通过调节免疫相关细胞因子的表达调控宿主免疫功能,帮助PCV2逃逸宿主免疫,促进PCV2在3D4/21细胞中的复制。这些发现不仅为深入了解PCV2与宿主之间的关系提供了新视角,还有望为猪圆环病毒相关疾病的防控提供新的抗病毒策略。  相似文献

Abstract

We evaluated the differential expression of several microRNAs (miRNAs) among malignant cells in ascites and matched omental metastasis in patients with epithelial ovarian cancer (EOC). Ascites and omental tumors were collected prospectively from five patients who were undergoing primary surgical cytoreduction. Patient samples were processed and treated with carboplatin, paclitaxel and combination chemotherapy. Cell viability was evaluated and miRNA profiling was performed on both tumor cells from ascites fluid and omental cake. Quantitative real-time PCR (RT-q-PCR) and western blots were used to evaluate expressions of miRNA-21 and miRNA -214 and associated proteins. Malignant cells in ascites showed greater cell viability when treated with carboplatin compared to omental metastasis. A significant up-regulation of miRNA-21 and miRNA-214 was observed in malignant cells of ascites compared to omental metastasis; this was confirmed by both cell viability assay and RT-q-PCR. Ours is the first report that demonstrates significant up-regulation of miRNA-21 and miRNA-214 in tumor cells from ascites of patients with EOC compared to omental metastasis. This finding has important implications for intrinsic carboplatin resistance in these patients.  相似文献

摘要 目的:探讨急性心力衰竭患者血清miRNA-21、肌红蛋白(MYO)、心肌肌酸激酶同工酶(CK-MB)与心功能和预后的关系。方法:选择2018年2月至2019年2月于我院接诊的95例急性心力衰竭患者为研究对象(病例组),另选择同期在我院进行检查的80例健康人为对照组,分析两组患者血清miRNA-21、MYO、CK-MB、左心室射血分数(LVEF)、左心房内径(LAD)、左心室舒张末期内径(LVEd)水平变化情况及和预后相关性。结果:病例组患者血清miRNA-21、MYO、CK-MB水平显著高于对照组,差异显著(P<0.05);病例组患者LAD、LVEd水平显著高于对照组,LVEF水平显著低于对照组,差异显著(P<0.05);II级患者血清miRNA-21、MYO、CK-MB水平显著低于III级、IV级患者,III级患者血清miRNA-21、MYO、CK-MB水平显著低于IV级患者,差异显著(P<0.05);预后良好组患者血清miRNA-21、MYO、CK-MB水平显著低于预后不良组,差异显著(P<0.05);Spearman相关分析显示,血清miRNA-21、MYO及CK-MB和LVEF之间均呈负相关,血清miRNA-21、MYO及CK-MB和LAD、LVEd之间均呈正相关,(P<0.05);Logistic 回归分析结果显示,血清miRNA-21、MYO及CK-MB及LVEF、LAD、LVEd为急性心力衰竭患者预后的影响因素(P<0.05)。结论:急性心力衰竭患者血清miRNA-21、MYO、CK-MB的表达升高,其与心功能和预后密切相关。  相似文献

Long noncoding RNAs (lncRNAs) CASC11 is an oncogenic lncRNA in gastric cancer and colorectal cancer. Our study aimed to investigate the role of lncRNA CASC11 in bladder cancer. In this study we showed that plasma lncRNA CASC11 was upregulated, while plasma miRNA-150 was downregulated in patients with early-stage bladder cancer than in healthy controls. Altered expression of plasma lncRNA CASC11 and miRNA-150 separated patients with bladder cancer from healthy controls. lncRNA CASC11 expression was inversely correlated with miRNA-150 expression in patients with bladder cance but not in healthy controls. Overexpression of lncRNA CASC11 mediated the inhibition of miRNA-150 expression in cancer cells, while miRNA-150 overexpression did not significantly alter lncRNA CASC11 expression. lncRNA CASC11 overexpression promoted, while miRNA-150 overexpression inhibited cancer cell proliferation. miRNA-150 also attenuated the enhancing effects of lncRNA CASC11 overexpression on cancer cell proliferation. However, overexpression of lncRNA CASC11 showed no significant effects on cancer cell migration and invasion. Therefore, lncRNA CASC11 may promote cancer cell proliferation in bladder cancer, and the actions of lncRNA CASC11 are likely through miRNA-150.  相似文献
