A sterile mineral salts broth was fortified with different additives, inoculated with conidia ofPenicillium rubrum P-13, and incubated quiescently for 14 days or with shaking for 3 to 5 days. Maximal fungal growth and rubratoxin production occurred when the broth contained 20% sucrose. Broth with 10% glucose, 10% fructose, 5% maltose, or 1% asparagine supported formation of substantial amounts of rubratoxin (52.9–78.5 mg/100 ml). When the broth was fortified with glucose plus lysine, arginine aspartic acid, cystine, ammonium citrate, or ammonium phosphate, moderate amounts (27.5–39.5 mg/100 ml) of rubratoxin and mycelium (0.1–1.5 g/100 ml) were produced. Presence in the broth of 5% galactose or starch resulted in accumulation of small amounts (22.2 and 24.6 mg/100 ml, respectively) of rubratoxin and mold tissue (0.70 and 0.5 g/ 100 ml, respectively). Whereas some toxin was recovered from mineral salts broth fortified with lactose or ribose, toxin was not recovered when the mold grew in broth containing mannitol or fumarate. With the exception of gluconate which supported some growth and toxin formation and ethanol which permitted formation of small amounts of toxin, other carbon sources resulted in little or no fungal growth and no toxin formation. Yields of rubratoxin decreased with an increase in amount of agitation or length of incubation ofP. rubrum cultures. Mold growth increased and toxin formation decreased with an increase in volume of culture.  相似文献

The culture yield of a simple method of microtuber production of potato was increased by assessing the interactions of illumination source (Thorn Lighting (Philips) “Colour 84” lamps (TL‐84) or Grolux lamps (Sylvania) in a conventional growth room or natural light in a glasshouse cabinet), type of vessel closure (unventilated or ventilated) and sucrose concentration (1%, 2%, 4% or 8%). Microtuber initiation and growth in unventilated cultures was 100% at 8% sucrose falling to 40–50% at 4% sucrose and was absent at 1% or 2%. With ventilation, rapid tuberisation (90–100%) occurred at initial sucrose concentrations of 2–8%, but only when the medium was allowed to dry before transfer of cultures to short days. Water supplementation of cultures at day 28 prevented tuberisation at 1–4% sucrose up to day 56. The fresh weight and dry weight of microtubers per plant increased significantly with sucrose concentration, with ventilation of cultures and under natural light. In ventilated cultures, the mean number of usable microtubers (± 0.1 g weight) increased from between 1.0–1.4 per plant at 8% sucrose to between 1.6–2.6 per plant at 4% sucrose, with the highest numbers (1.8–2.6 per plant) produced under natural light for the cvs Desirée and King Edward. The mean % dry matter content of microtubers was reduced to 11.3% at 4% sucrose compared with 17.3% at 8% sucrose, but the survival rate of microtubers after 6 months storage was unaffected. Microtuber production under short days was improved at a higher intensity of natural light with culture ventilation in a partially‐shaded glasshouse cabinet, whilst using reduced inputs (lower sucrose supply and no lamps).  相似文献

The amino acid composition of the silage dry matter, silage nitrogen (g/16 gN) and the molar composition of the total measured amino acids (mM/100 mM) of five maize silages was measured and compared with results from the U.S.A.As the dry matter content of the silages increased, the total amino acid content decreased but was generally higher than values reported from America. As the grain content of the silages increased there was a decrease in lysine content which was reflected in an increased concentration of glutamic acid and proline. The lysine content of U.K. silages was higher than those from the U.S.A.  相似文献

大田栽培条件下,在大豆始花期叶面喷施以植物多糖(P1)、植物多糖和5-氨基乙酰丙酸(P2)以及植物多糖、5-氨基乙酰丙酸和缩节胺(P3)为有效成分复配的3种制剂,研究不同植物多糖类复合制剂对大豆叶绿素含量、光合蒸腾特性、干物质积累与分配以及籽粒产量的影响.结果表明:喷施3种制剂35 d内,大豆叶片叶绿素含量与对照相比明显增加,且随生育进程下降的趋势有所延缓;喷施P1和P3使大豆叶片光合速率和水分利用效率分别提高13.2%和10.3%以上.与对照相比,喷施3种制剂促进了大豆地上部于物质积累量的增加、提高了叶片干物质向荚的分配比例,花后干物质同化量对籽粒的贡献率增加17.1%以上;喷施P1和P3后,大豆单株荚数、单株粒数和百粒重显著增加,喷施P2后变化不显著;喷施3种制剂使大豆增产5.9%以上.3种植物多糖类复合制剂可促进大豆叶绿素合成、延缓叶片衰老、改善叶片光合潜能和水分状况,有效调控大豆干物质积累和花后同化物分配,进而实现增产.  相似文献

Freshly collected effluent was treated with 2 ml formalin per litre and used to replace 150 g/kg of the dry matter of an all-meal control diet of bacon pigs. Diets containing effluent from either unwilted or wilted crops were compared with the control diet which was also given wet by adding water. Liveweight gains of 10 pigs per treatment from 65 to 86 kg on diets containing effluents from unwilted and wilted crops and on the control diet were 753, 715 and 719 ± 16.8 g/day, respectively, and in the same order feed conversion ratios (corrected to dry matter content of the control diet) were 2.94, 3.10 and 3.07 ± 0.075. None of these differences were significant. There were no significant differences in killing-out yield, in hardness of subcutaneous back-fat when measured by a penetrometer or in abnormal odour rating of subcutaneous back-fat.Formalin at levels of 1 and 2 ml/l and formic acid at levels of 1, 2 and 6 ml/l were added as potential preservatives to 1000 ml quantities of freshly collected silage effluent, which was then stored at 23°C. The 2 ml/l level of formalin and 6 ml/l level of formic acid prevented mould growth for 26 and 19 days, respectively.Freshly collected effluent was stored for 3.5, 14, 28, 56, 112 and 240 days after treatment, with formalin at the rate of 3 ml/l, and used in a pig-feeding trial. Pigs were fed either on a control all-meal diet or an equal dry matter intake of 150 g/kg effluent DM and 850 g/kg meal DM.A total of 120 animals were individually penned and fed twice daily on the experimental diets from 33 kg to slaughter at 82 kg liveweight. Overall mean liveweight gains for effluent and control diets were 646 and 656 ± 7.5 g/day, respectively, and the corresponding feed conversion ratios (DM basis) were 2.69 and 2.64 ± 0.030, neither of the differences being significant. Animal performance did not deteriorate with increasing period of effluent storage. No significant differences were found in carcase lean, back-fat thickness nor hardness of back-fat.A digestibility study was carried out on freshly collected effluent from another source. The level of dietary inclusion was equal to that in the previous experiments and the digestibility of dry matter was 0.831 and 0.819, and of nitrogen 0.822 and 0.827 for the control and effluent diets, respectively. A value of 11.4 MJ/kg dry matter was calculated by difference for the digestible energy of this effluent. In a further digestibility trial, digestible energy values of 14.6 and 14.2 MJ/kg dry matter were calculated by difference for effluents stored for 3.5 and 300 days, respectively.  相似文献

The present investigation clarified the effect of enzymes or other substances formed during the germination process on the vomitoxin level of contaminated oats. The studies found that oats containing vomitoxin germinated very poorly; the decrease in toxins was also slight. The amount of pure vomitoxin added to toxin-free grain decreased (barley 53 %, oats 40 %, wheat 20 %) during germination (5 d). In homogenized mixture of germinated grain (2.4 and 7 d) and toxic grain no decrease in toxin amount occurred during a 1–7 day period. In contrast, when germinating toxin free grains and toxic oats in a grain mixture the toxin level decreased at first, but later rose considerably. On the basis of these results, the utilization of germination processes for the improvement of grain containing vomitoxin is of questionable value.  相似文献

Summary The effect of substrate (buckwheat seeds) pretreatment on the growth and the sporulation behaviour of Penicillium roqueforti is presented. When a saccharifying enzyme (-amylase) is added to a medium which exhibits a low water content (0.46 g water/g initial dry matter, IDM), a more rapid internal colonization of the seeds occurs, but the final spore production does not increase and remains close to 8.109 spores/g dry matter (DM) at 500 h. No carbon source limitation is then observed. The addition of casein hydrolysate to this medium gives rise to a great increase of the sporulation, since 14.5 109 spores/g DM are obtained after 600 h. This result is attained by a better spore yield from the mycelium, the substrate colonization being unchanged. High water content (0.60 g water/g IDM) of buckwheat seeds induces a shorter cultivation time along with a higher biomass production. However, the spore content of the medium remains close to the low water content one, but 60% total spores are external against 30% to 35% in the other media.  相似文献

A total of 75 compounds, including antioxidants, preservatives, gallic acid and p-hydroxybenzoic acid esters, hydroquinones, hydroxyquinolines, phenol derivatives, and related compounds, were screened for their antibotulinal activity in prereduced Thiotone-yeast extract-glucose broth. The most effective inhibitors of Clostridium botulinum growth and toxin production were long-chain esters of p-hydroxybenzoic acid and gallic acid, antioxidants, and butylphenol derivatives. The antioxidant nordihydroguaiaretic acid at 100 microgram/ml delayed the growth and toxin production for the entire incubation period (7 days). Other antioxidants, such as butylated hydroxytoluene, butylated hydroxyanisole, and tert-butylhydroquinone were also very effective (at 200 to 400 microgram/ml) for the inhibition of C. botulinum growth and toxin production. Toxin was detected, although no detectable growth was found by daily absorbance measurements, in the prereduced medium containing 50 to 400 microgram of 8-hydroxyquinoline, pentylphenol, tert-pentylphenol, 3,5-ditert-butylphenol, 3,5-ditert-butylcatechol, (2-hydroxydiphenyl)methane, or (4-hydroxydiphenyl)methane per ml.  相似文献

Summary Dry matter increase of Agaricus bisporus mycelium in liquid culture, was found to be directly proportional to quantities of fungal derived chitin and extracellular laccase for up to 28 days following inoculation. Fungal ergosterol showed a similar relationship to mycelial growth which was sustained for 56 days of culture. In cultures grown on autoclaved rye grain a correlation was found between the three biochemical indices and linear extension of the mycelium. Each method gave a similar biomass estimate for cultures grown on cereal grains.  相似文献

Growth on a medium containing succinic acid as the sole carbon source produced 1 g (dry weight) of mycelium per liter of medium by 50 days of incubation, whereas 25 g of mycelium was produced in 10 days when glucose was also present in the medium. Primary shunt metabolism took place during growth on succinic acid in spite of the extremely slow growth. Mycelia grown on succinic acid contained a higher percentage of residual mycelium and phosphate, but a lower percentage of mannitol, carbohydrate, lipid, and water-soluble nitrogen, than mycelia grown on a mixture of glucose and succinic acid. Thus, although primary shunt metabolism is favored by rapid growth on a rich, balanced sugar medium, it can also take place during extremely restricted growth in a medium containing succinic acid as the sole carbon source.  相似文献

为提高灰毡毛忍冬"渝蕾1号"悬浮培养体系中绿原酸的含量,该研究探讨了B_5培养基中不同浓度的无机盐对灰毡毛忍冬"渝蕾1号"悬浮培养细胞生物量及绿原酸含量的影响,通过在悬浮培养体系中添加不同浓度的无机盐,采用重量法测定灰毡毛忍冬"渝蕾1号"悬浮培养细胞的生物量及采用高效液相色谱法测定绿原酸的含量。结果表明:当硝态氮和铵态氮配比与B_5培养基中硝态氮和铵态氮配比一致时,即NO_3~-/NH_4~+摩尔比值为13∶1时,培养体系有利于细胞的生长和绿原酸的积累。当KNO_3浓度为3.5 g·L~(-1)时,细胞生物量达到最大,为19.26 g·L~(-1);当KNO_3在较低浓度(0.5 g·L~(-1)和1.5 g·L~(-1))时,积累较多的绿原酸。NO_3~-的两项研究结果均与对照浓度(2.5g·L~(-1))有一定的差异。另外,对(NH_4)_2SO_4来说,在高于对照浓度0.134 g·L~(-1),即浓度为0.268 g·L~(-1)时,生物量和绿原酸含量都达到了最大。P、Ca、Mg三种矿质元素的研究结果表明,当Na H_2PO_4·2H_2O浓度为0.10 g·L~(-1)、Ca Cl_2的浓度为0.20 g·L~(-1)时,细胞的生长和绿原酸的积累均可达到最大值;而对Mg~(2+)来说,低浓度促进细胞的生长,高浓度促进绿原酸的积累。兼顾细胞生物量和绿原酸含量两个指标,需选择适中的浓度。这些结果均与对照浓度有一定的差异。这说明灰毡毛忍冬"渝蕾1号"悬浮细胞所需无机盐的浓度与B_5培养基无机盐的浓度有一定的差异,选择适宜的浓度可促进其悬浮细胞的生长及次生代谢产物绿原酸的积累。该研究结果为绿原酸的工业化生产打下了基础。  相似文献

Commercially produced haem fraction of abattoir blood with a dry matter content of 35% was treated with formic acid of 80–85% purity at the rate of 3:100 (v/v) which transformed the “haem” from a liquid to a solid state and reduced pH from 7.2 to 3.6. The product was included at approximately 5% of the dietary dry matter for pigs over the live weight range 33–80 kg. In one experiment the diet containing the haem was compared with diets containing either 7 or 10% herring meal on an air dry basis. The pigs on the 10% herring meal diet grew slightly, but non-significantly, faster and more efficiently than pigs on the other two diets. The carcases of the pigs on the haem diet contained less lean and more fat than those on the herring meal diets, apparently because of dietary deficiencies of isoleucine and methionine + cystine. In the second experiment, diets with and without the haem fraction were formulated by linear programme taking ingredient costs into account. The performance and carcase composition of pigs on the haem diet were equal to or better than those on the control diet.The total lysine content of the haem fraction of abattoir blood treated with formic acid was 115 g/kg dry matter with an availability (1-fluoro-2,4-dinitrobenzene-reactive lysine) of 86%. In relation to the requirements of the finishing pig the limiting amino acids appear to be isoleucine and methionine + cystine.  相似文献

The influence of whole, crushed and ground barley and oats on intake, performance and carcass characteristics of lambs was examined. A comparison of diets was conducted with individually-fed male and female lambs in six groups. The study was repeated in three successive years (1995–1997) using the same experimental design. A total of 192 lambs of a mean initial age of 74 days (SD 13.0) and live weight 20.6 kg (SD 4.54) were daily fed their respective concentrates, 72 g dry matter (DM)/kg metabolic live weight (kgW0.75), for 98 days from weaning to slaughter. Hay was provided ad libitum.The lambs adapted more quickly to diets containing barley than to those containing oats. Compared to oats, the total daily DM intake was higher on barley (1162 vs. 990 g DM/lamb, 89 vs. 78 g DM/kgW0.75, P < 0.001). Hay consumption was significantly lower on oats than on barley (182 vs. 344 g DM/day, P < 0.001). On oats the lambs experienced energy and protein deficiencies with their energy and protein intakes being 20% below feeding recommendations. On the barley diets the energy and protein requirements of the lambs were satisfied. When processed cereals were offered, there was no increase in the total DM, energy or protein intakes. Processing did not improve the organic matter digestibility of barley or oats.The growth rate (P < 0.01), carcass weight (P < 0.001) and slaughter percentage (P < 0.001) were higher on a barley diet than on oats. A barley diet resulted in more kidney fat (P < 0.01) and fattier carcasses. Processing had no positive influence on the live weight gain. On the contrary, lambs receiving crushed or ground barley or oats had a lower final live weight than those receiving whole grains (P < 0.001). The utilisation of whole grain was more efficient than that of processed grain. There were no advantages in processing barley and oats for the feeding of lambs.  相似文献

A 10−3 m β-indolebutyric and 2,4-dichlorophenoxyacetic acid concentration stimulated the fresh weight increase and dry matter production in the mycelium of staticClaviceps purpurea cultures to 170–180% of the control value. Higher concentrations severely inhibited growth. α-Naphthylacetic acid was less active. β-Indoleacetic acid differed markedly from the other auxins, as it stimulated the fresh weight increase, but at the same time inhibited dry matter production. In submerse cultivation, the differences between the test auxins disappeared; none of them stimulated growth and only inhibition of growth, by concentrations of over 10−5–10−4 m, was observed. The used strain did not produce alkaloids in saprophytic cultures, even in the presence of auxins.  相似文献

Previous studies suggest that the positive response of transplanted rice (Oryza sativa L.) to nursery fertiliser application was due to increased seedling vigour or possibly to increased nutrient content. This paper presents results of two glasshouse experiments designed to test the hypothesis that seedling vigour was responsible for the response of transplanted seedlings to nursery treatments. The aim of the present study was to explore the concept of seedling vigour of transplanted rice and to determine what plant attributes conferred vigour on the seedlings. Seedling vigour treatments were established by subjecting seedlings to short-term submergence (0, 1 and 2 days/week) in one experiment and to leaf clipping or root pruning and water stress in another to determine their effect on plant growth after transplanting. Submerging seedlings increased plant height but depressed shoot and root dry matter and root:shoot ratio of the seedling at 28 days after sowing. After transplanting these seedlings, prior submergence depressed shoot dry matter at 40 days. Nursery nutrient application increased plant height, increased root and shoot dry matter, but generally decreased root:shoot ratio. Pruning up to 60% of the roots at transplanting decreased shoot and root dry matter, P concentration in leaves at panicle initiation (PI) and straw dry matter and grain yield at maturity. By contrast, pruning 30% of leaves depressed shoot and root dry matter by 30% at PI, and root dry matter and straw and grain yield by 20% at maturity. The combined effects of leaf clipping and root pruning on shoot, root and straw dry matter were largely additive. It is concluded that the response of rice yield to nursery treatments is largely due to increased seedling vigour and can be effected by a range of nutritional as well as non-nutritional treatments of seedlings that increase seedling dry matter, nutrient content, and nutrient concentration. Impairment of leaf growth and to a lesser extent root growth in the nursery depressed seedling vigour after transplanting. However, rather than increasing stress tolerance, seedling vigour was more beneficial when post transplant growth was not limited by nutrient or water stresses.  相似文献

Fusarium moniliforme was grown on a carob aqueous extract in a chemostat for fungal protein production. The substrate was adjusted to provide 0.5% carob sugars supplemented with inorganic salts. The dilution rate varied from 0.086 to 0.227 hr?1 under constant conditions of temperature (30°C), pH (4.5), and oxygen saturation (60–80%). A yield of 0.709 g dry mycelium/g consumed carob sugar and a productivity value of 0.687 g dry mycelium/liter hr?1 were obtained at μ = 0.205 hr?1. The maintenance coefficient was 0.077 g carob sugar/g dry mycelium hr?1. While the carbohydrate and purine content of dry mycelium increased at μ values from 0.114 to 0.205 hr?1 both true (Lowry) and crude (N × 6.25) protein contents decreased at the same μ range. Maximum values of 36.3% true and 47.9% crude protein of dry mycelium were obtained at μ = 0.114 hr?1, whereas a minimum purine content of 99.8 μmol/g corresponding to 6.42% nucleic acids was recorded at μ = 0.086 hr?1. It was concluded that a continuous fermentation of carob aqueous extract using F. moniliforme should be operated at growth rates of approximately 0.205 hr?1 in order to maximize protein production.  相似文献

在大田栽培条件下,采用生物质多糖(P1)、生物质多糖和5 氨基乙酰丙酸复配(P2),以及生物质多糖、5-氨基乙酰丙酸和缩节胺为有效成分复配(P3)的3种不同制剂,研究在冬小麦始花期叶面喷施制剂对其产量构成、蔗糖、可溶性总糖、干物质贮运以及氮磷养分累积与转移的影响.结果表明: 喷施3种制剂使冬小麦穗粒数和千粒重增加,增产8.5%以上;喷施20 d内,小麦旗叶蔗糖含量较对照明显增加;喷施P1和P3使小麦籽粒可溶性糖含量分别比对照增加4.5%和11.0%.P3增加了小麦花后干物质及氮磷养分累积量,分别较对照增加48.5%、116.9%和18.1%,P3还显著提高了小麦花后干物质及养分累积对产量的贡献,但花前养分向籽粒转移对产量的贡献小于其他处理.小麦增产与植物多糖类复合制剂有效调控营养器官光合产物输出、籽粒可溶性糖积累,以及促进花后干物质和氮磷养分累积有关.  相似文献

Acid pretreatment of cellulosic wastes improved their susceptibility to Fusarium acuminatum enzymes. The effectiveness of acid pretreatment was demonstrated with an increase in both fungal growth and enzyme activities. A growth yield of 0.15 g/100 ml was achieved on medium containing 5% acid pretreated pods of bean for 60 minutes. Avicelase (C1), carboxymethylcellulase (Cx) and B-glucosidase (C2) reached their maximal biosynthesis on acid pretreated wheat bran, sugar-cane bagasse and sawdust-containing media, respectively. Xylanase and pectinase attained their highest accumulation on pretreated pods of bean media. A mixture of free sugars has been released by acid pretreatment. O.199 g dry mycelium was obtained when the fungus was grown on 100 ml of medium containing hydrolysate of 10% H2SO4 pretreated pods of bean for 30 min. No cellulase enzymes could be detected on hydrolysate medium at the time that low contents of both xylanase and pectinase were accumulated.  相似文献

This study provides observations on the effects of lead and cadmium ions on the growth of two species of marine fungi, Corollospora lacera and Monodictys pelagica. On solid media lead appeared to have no effect on the radial rate of growth of fungi. Exposure to increasing cadmium concentrations on solid media resulted in significant reduction (p < 0.05) in the radial mycelial growth rates of both fungi, especially in M. pelagica. These results reveal significant difference in species sensitivity toward cadmium and, essentially, insensitivity toward lead exposure. In liquid cultures, the metal content of mycelia (metal mass found in mycelium, in mg), and the concentration of metal in dry mycelium (metal mass in 1g of mycelium, in mg g(-1)) were both found to increase (p < 0.05) with the increase in the metal cation concentration, while mycelium dry mass decreased. As it was observed on solid media, cadmium cation affected more severely (p < 0.05) the growth of M. pelagica in liquid cultures. Ergosterol content of mycelia of C. lacera exposed to increasing cadmium cation concentration decreased, similarly to the trend observed for dry mycelial mass. It was found that ca. 93% of all lead sequestered by C. lacera is located extracellularly. M. pelagica was found to bioaccumulate over 60 mg of cadmium and over 6 mg of lead per 1 g of mycelium, while C. lacera bioaccumulated over 7 mg of cadmium and up to 250 mg of lead per 1 g of mycelium. Overall, the results indicate that both metal ions affect the growth of marine fungi with lead being accumulated extracellularly in the mycelia. Both metals accumulated by fungi may then enter the marine ecosystem food web, of which marine fungi are integral members.  相似文献

Ergosterol is a membrane component specific to fungi that can be used to estimate fungal biomass using appropriate factors of conversion. Our objectives were to determine the limits of use of ergosterol content as a measure of biomass for aquatic hyphomycetes, and to evaluate a previously established ergosterol-to-biomass conversion factor. We varied inoculum quality, growth medium, and degree of shaking of four aquatic hyphomycete species. In cultures inoculated with homogenized mycelium, we found a significant effect of shaking condition and culture age on ergosterol content. In liquid cultures with defined medium, ergosterol content reached 10 to 11 microg/mg of mycelium (dry mass) and varied by factors of 2.2 during exponential growth and 1.3 during stationary phase. The increase in ergosterol content during exponential phase could be attributed, at least in part, to rapid depletion of glucose. Oxygen availability to internal hyphae within the mycelial mass is also responsible for the differences found between culture conditions. Ergosterol concentration ranged from 0.8 to 1.6 microg/mg in static cultures inoculated with agar plugs. Ergosterol content varied by a factor of 4 in two media of different richnesses. For different combinations of these parameters, strong (r(2) = 0.83 to 0.98) and highly significant (P < 0.001) linear relationships between ergosterol and mycelial dry mass (up to 110 mg) were observed. Overall, the ergosterol content varied by a factor of 14 (0.8 to 11 mg/g). These results suggest that care must be taken when the ergosterol content is used to compare data generated in different field environments.  相似文献
