Cultivars of glandular-haired alfalfa, Medicago sativa L., such as '54H69', are currently available and marketed as being resistant to potato leafhopper, Empoasca fabae (Harris) (Hemiptera: Cicadellidae). 54H69 and a standard, nonglandular-haired alfalfa 'Choice' were evaluated at two locations in Virginia over a 3-yr period. Dry matter yields and concentrations of crude protein and acid detergent fiber were compared at the first, second, and third harvests. Overall, the two cultivars produced similar dry matter yields of comparable forage quality in the absence of insecticides at both locations in each year. Untreated 54H69 did not produce greater dry matter yields than untreated Choice under either light or heavier potato leafhopper pest pressure. Concentrations of crude protein did not vary between the two cultivars at any harvest. Some differences in concentrations of acid detergent fiber were detected between cultivars, but these differences were not consistent among years, harvests, or between locations. Further comparisons between untreated 54H69 and treated Choice were made, but few significant differences were detected in dry matter yields or forage quality. An economic analysis for the study indicated that a grower planting 54H69 would realize less net revenue than a grower planting Choice, largely because of the seed premium for the glandular-haired cultivar and the evident need to treat 54H69 with insecticide for control of alfalfa weevil, Hypera postica (Gyllenhal) (Coleoptera: Curculionidae), and potato leafhopper.  相似文献

Plant tolerance to herbivory is a key approach for managing pests. In alfalfa, Medicago sativa, the potato leafhopper, Empoasca fabae, is a major pest as a result of the cascade of plant responses to piercing-sucking injury. To identify tolerance to its injury based on alfalfa physiology, experiments were conducted in the field and greenhouse. In our comparison of the response of field-grown alfalfa cultivars to standardized leafhopper densities, net photosynthesis and transpiration rates of 'Geneva' leaves were reduced by 18 and 21%, respectively, by leafhopper presence compared with a rate change of <1% of resistant 'EverGreen' leaves. Under greenhouse conditions, alfalfa clones varied in their level of gas exchange (net photosynthesis and transpiration) and stem elongation responses to leafhopper injury. For example, in the comparison of seven clones, net photosynthesis declined an average of 40.7% with leafhopper injury, although individual clones varied from 26.6 to 74.3% reduction. Internode elongation after 2 d was 60.3% less on injured stems compared with healthy stems, but again, the individual clones varied from 17.3 to 91.9%. In a time-course study of selected clones, clones varied in their level of injury just after and 3 d after insect removal. Gas exchange responses of all clones recovered by 7 d after cessation of injury. In a choice test, leafhoppers spent similar amounts of time on the susceptible clone and the most tolerant clone; however, their precise feeding behaviors were not measured. Thus, the variable response of clones to injury may be either true physiological tolerance or antixenosis from a change in feeding behavior. This study showed putative tolerance to leafhopper injury among alfalfa genotypes, suggesting that tolerance could be the basis for crop protection in alfalfa from potato leafhopper injury.  相似文献

The effects of a 3-m wide uncut alfalfa strip on the within field distribution of Empoasca fabae (Harris) (Homoptera: Cicadellidae) and several species of insect predators were examined for 3 yr (1998-2000). The objectives were to determine whether this uncut strip would serve as a trap crop for E. fabae and a refuge for insect predators at first harvest. Empoasca fabae and insect predators in the families Coccinellidae, Nabidae, Anthocoridae, Chrysopidae, and Hemerobiidae were collected weekly using sweep samples and sticky traps from the uncut strips and up to 40 m into the alfalfa regrowth. For 2 yr, both small- (0.34 ha) and large-scale (approximately 11.3 ha) field trials showed higher numbers of E. fabae in 73% of the uncut strips for 2-3 wk after harvest. Similarly, the number of insect predators found within <50% of the uncut strips was also higher during the first or second week after harvest. In 1999, however, we did not observe higher numbers of E. fabae in the uncut strips. Differences may be because of higher E. fabae population numbers in 1999 compared with 1998 and 2000. This research provides alfalfa growers a potential cultural management technique for E. fabae while conserving predatory insects.  相似文献

Laboratory experiments were conducted to characterize the trichome-based defense of glandular-haired alfalfa, Medicago sativa L., against the potato leafhopper, Empoasca fabae (Harris). Within-plant variability in leafhopper resistance was examined by caging adult leafhoppers to either basal or apical stem internodes of the leafhopper-resistant, glandular-haired M. sativa genotype G98A or the susceptible, nonglandular-haired M. sativa 'Ranger'. Young, actively secreting glandular trichomes are located on apical internodes of G98A, whereas senesced gland heads are found on older, basal internodes of G98A. After 96 h, the highest cumulative leafhopper mortality and lowest number of excretory droplets were associated with apical internodes of G98A. No difference was detected in mortality and feeding levels among insects caged to basal internodes of G98A and basal and apical internodes of Ranger. The influence of abiotic factors on leafhopper resistance was evaluated by caging adult leafhoppers to either G98A or Ranger under four combinations of low and high light (250 and 1,000 micromol s(-1) m(-2)) and temperature regimes (17 and 30 degrees C). After 96 h, the highest cumulative mortality was associated with leafhoppers confined to G98A under high light and high temperature conditions. Temperature level and plant type also had an effect on the production of excretory droplets, resulting in the highest number of excretory droplets being associated with Ranger under the high temperature regime. These results indicate that certain regions of M. sativa G98A are better protected against the potato leafhopper than others and that temperature influences resistance levels of glandular-haired alfalfa.  相似文献

Eight proprietary genotypes of glandular-haired alfalfa, Medicago sativa L., supplied by two different companies, were compared for the degree and types of resistance to the potato leafhopper, Ernpoasca fabae (Harris), and hopperburn. A tube cage no-choice bioassay was developed to test leafhopper mortality, feeding, settling preferences, severity of hopperburn symptoms (in this case, defined as both yellowing and stem growth reduction), and trichome density and type on feeding sites. Leafhopper mortality was both strongly and significantly associated with feeding and leaf trichome density; decreased hopperburn symptom severity was weakly, although significantly, associated with increased mortality. To quantify hopperburn in terms of both yellowing and stem growth reduction, we developed a ranking system that reduces overall hopperburn expression to a single number that considers the varying responses to both types of symptoms. Great variability in leafhopper settling, leafhopper mortality, and stem glandular trichome density was detected among alfalfa genotypes, suggesting that genotypic differences may be based on the concentration and/or chemical constituency of the trichome exudates. We postulate that, among variably resistant genotypes of glandular-haired alfalfa, differences among leafhopper responses and hopperburn severity are linked to forced movement from the stems to the leaves as refuge feeding sites. Principal component analysis was performed to reduce the 10 variables down to five biologically significant factors. Scores for these factors were then used to develop resistance indices for potato leafhopper resistance, hopperburn resistance, and an overall glandular-haired alfalfa resistance index.  相似文献

Experiments were performed to elucidate resistance of glandular-haired alfalfa, Medicago sativa L., to the potato leafhopper, Empoasca fabae (Harris). The primary objective was to examine the potential role of the glandular trichomes and stem lignification for imparting resistance to this pest. During free-choice foliar discoloration experiments, the resistant alfalfa clone FGplh13 expressed lower levels of injury than the susceptible P5373 after 13 d. No-choice tests performed using nymphal potato leafhoppers showed higher levels of mortality associated with the resistant glandular-haired clone FGplh13, and no nymphs survived after 48 h. Additional no-choice experiments using FGplh13 and P5373 alfalfa with the glandular and nonglandular trichomes intact or removed show that mortality of nymphs and adults decreased after removal of the glandular trichomes from FGplh13. Nymphal and adult mortality and nymphal development time did not differ on FGplh13 with the trichomes removed and P5373 with the trichomes intact or removed. Firstinstar, and possibly second-instar, potato leafhoppers were entrapped by an exudate produced by the glandular trichomes on the FGplh13 alfalfa. Stem lignification and the number of vascular bundles did not differ between FGplh13 and P5373 alfalfas. The glandular trichomes on FGplh13 alfalfa appeared to provide the major host resistance factor, with resistance to adults being chemically based and resistance to nymphs being chemically and mechanically based. To maintain levels of potato leafhopper resistance, breeders would appear to benefit by continuing to select for the expression of the glandular trichome phenotype.  相似文献

Survival of nymphs and adult males and females of potato leafhopper, Empoasca fabae (Harris), on six potato (Solanum spp.) genotypes with varied levels of resistance was evaluated and related to type, density, and droplet/head size of foliar glandular trichomes. Survival was greater on Solanum tuberosum cultivars ('Allegany' and 'Elba') compared with accessions of the wild S. berthaultii (PI 473331 and PI 473334). The barrier provided by glandular trichomes may affect survival of E.fabae by restricting nymph and adult feeding. PI 473331 was the most pubescent host, bearing type A and type B trichomes and the most unsuitable host for E. fabae. Allegany possesses leaflets with low trichome densities and small gland diameter and was the most suitable host for adult E. fabae survival. Females generally had higher survival rates than males. Nymphal and adult leafhoppers differed greatly in their survival rates, particularly on S. tuberosum x S. berthaultii hybrids (Q174-2 and NY123). Interestingly, nymphs had longer (190 d) mean lethal time (LT50) than adults (31 d) when held on Q174-2, and a lower LT50 (19 d) when held on NY123 compared with adults (104 d). Starvation, dehydration, or both were likely associated with mortality of nymphs confined on PI 473331 and PI 473334. Nymphs on these accessions died at a comparable rate to those confined on a starvation treatment and more rapidly than those supplied with water only, sucrose solution or other potato genotypes. Darkened trichome exudates were observed on insects held on PI 473331 and PI 473334, consistent with previous reports that glandular trichomes of S. berthaultii contain phenolic oxidation chemistry. This study aids in the identification of the probable mechanisms of resistance to E. fabae used by S. berthaulti and its hybrids with S. tuberosum.  相似文献

A field study was conducted in 1996 and 1997 to determine survival and spatial patterns of alfalfa weevil, Hypera postica (Gyllenhal), larvae within and between windrows of swathed alfalfa, Medicago sativa L., forage as dry matter concentration increased. After harvest, the percentage of live weevil larvae residing in swathed forage decreased as swath dry matter percentage increased. Conversely, larval populations in stubble between windrows increased with increasing swath dry matter. Larval populations in stubble under windrowed forage increased slightly as windrowed forage dry matter increased. Applications of these results for the development of alfalfa harvest systems for improved cultural control of alfalfa weevil are discussed.  相似文献

Strain distribution for the alfalfa weevil, Hypera postica (Gyllenhal), was determined for the five major alfalfa-growing regions of New Mexico. Mitochondrial DNA from field-collected weevils at each location was digested and sequenced to differentiate among strains. Eastern, Egyptian, and western strains were recovered, along with a population not matching any known strain. The majority of sites examined had populations of two strains intermingled within a field. This is the first confirmation of the presence of all three strains within the same geographic region. Possible implications for alfalfa weevil management are discussed.  相似文献

The potato leafhopper, Empoasca fabae (Harris), is a key pest of alfalfa, Medicago sativa L., in part because of the leafhopper's ability to disrupt upward translocation within phloem tissues. To determine if leafhopper injury also disrupts basal translocation necessary for regrowth and perenniality of alfalfa, we used radiolabeled 14CO2 to measure the basal transport of photoassimilates in injured and healthy plants. In one experiment, less 14C was transported to lower stem tissue of leafhopper-injured plants in comparison to the same tissue of healthy plants in early vegetative and early reproductive stages of alfalfa development. In a second experiment, less 14C was transported to lower stem, crown, and root tissues of injured plants in comparison to the same tissues of healthy, early reproductive plants. The disruption of basal transport caused by potato leafhopper may impact carbon storage and mobilization subsequent to defoliation, winter survival, and nitrogen fixation.  相似文献

It is argued that this variation is due to chromosome fusion and that A. draba is presently evolving to a lower number. Although potato leafhopper, Empoasca fabae (Harris) (Hemiptera: Cicadellidae), is highly polyphagous, classic host studies do not recognize grapevines (Vitis spp.), as suitable hosts. Recently, injury has been reported and reproduction documented within grape vineyards, suggesting a host expansion for the leafhopper. To document this apparent expansion in host use, we determined whether grape plants were suitable hosts for potato leafhopper reproduction, measured the consequence of feeding injury on gas exchange rates of grape leaves, and compared the susceptibility to feeding injury among cultivars. We found that potato leafhopper adults survived equally well on grape (Vitis vinifera L.), alfalfa (Medicago sativa L.), and fava bean (Vicia faba L.). The total number of offspring was greater on fava bean but did not differ between alfalfa and grape. Injury to grapevines was assessed by measuring gas exchange responses of leaves in field cages and in greenhouse tests. We found marginally significant declines in photosynthesis and transpiration rates in the field (9.6 and 13.2%, respectively), and much stronger effects in greenhouse tests (ranging between 22 and 52%). Our results verify that Vitis is a suitable host, and that potato leafhopper is capable of injuring its gas exchange physiology. We discuss possible explanations for the host expansion, and its potential to damage commercial grapevines.  相似文献

To identify the means by which phytohemagglutinin (PHA) exerts its toxicity on the potato leafhopper, four different methods (thick and semi-thin sectioning combined with immunofluorescent staining, in vitro receptor autoradiography, and immunoelectron microscopy) were used to elucidate the PHA target tissue, binding site, and its effects on this tissue. Sixteen 1- or 2-day-old female potato leafhoppers were fed for 36 h on each of three treatments: a control, diet or a diet containing either the PHA-E subunit or the PHA-L subunit. The PHA-E subunit, but not PHA-L, had previously been shown to be lethal. The insects were then prepared for both light and confocal microscopy. Analysis of images showed that PHA bound only to the surface of midgut epithelial cells of the potato leafhopper. PHA-E caused severe disruption, disorganization, and elongation of the brush border microvilli, and swelling of the epithelial cells into the lumen of the gut, leading to complete closure of the lumen. Furthermore, PHA-E stimulated the division of midgut epithelial cell nuclei, leading to two nuclei in each cell. Nuclei later elongated and degraded. In contrast, PHA-L had little effect on the epithelial cells of the midgut. It did not strongly bind to the surface of epithelial cells and caused much less disruption of brush-border microvilli, less disorganization of the cells and less elongation of nuclei. Strong binding of PHA occurred solely on the cell membrane of the brush border microvilli of epithelial cells. In contrast, the controls (i.e., midgut tissue, blocking agent, PHA, and antibodies) showed that midgut tissue was not autofluorescent and showed no fluorescent binding signal. Analysis of both bright- and dark-field images obtained by autoradiography and immunoelectron microscopy confirmed these findings.  相似文献

Sensilla on the antennae of adult alfalfa weevils, Hypera postica (Gyllenhal),were studied with scanning and transmission electron microscopes to determine the sensilla structure and possible chemo- and mechanoreceptive functions for food detection.Five types of sensilla trichodea (s.t.) occur (length 35 – 135 μm); s.t. I (71 – 135-μm long)are thick-walled without radial pores and s.t. II (50 – 75-μm long), the most abundant s.t., are multiporous. Bifurcated and trifurcated types (75 –85 -μm long) also occur. Five types of sensilla basiconica (s.b.) are present and include s.b. I (26 – 50-μm long), a fluted uniporous type with a rugose surface and encircling ledges; s.b. II (15 – 35-gm long), a common multiporous type with many longitudinal grooves; and s.b. IV (4 – 7-μm long), which are 3 apical, short pegs. Sensilla styloconica occur in a small band among the s.b. type II. One or 2 epidermal gland ducts are adjacent to most s.t. and some s.b.  相似文献

The alfalfa weevil Hypera postica is a serious economic pest in most alfalfa grown in many countries worldwide. Digestive α-amylase and pectinase activities of larvae were investigated using general substrates. Midgut extracts from larvae showed an optimum activity for α-amylase against starch at acidic pH (pH 5.0). α-Amylase from larval midgut was more stable at mildly acidic pH (pH 5–6) than highly acidic and alkaline pH. The enzyme showed its maximum activity at 35°C. α-Amylase activity was significantly decreased in the presence of Ca2+, Mg2+ and sodium dodecylsulfate. On the contrary, K+ and Na+ did not significantly affect the enzyme activity. Zymogram analysis revealed the presence of one band of α-amylase activity in in-gel assays. Pectinase activity was assayed using agarose plate and colorimetric assays. Optimal pH for pectinase activity in the larval midgut was determined to be pH 5.0. Pectinase enzyme is more stable at pH 4.0–7.0 than highly acidic and alkaline pH. However, the enzyme was more stable at slightly acidic pH (pH 6.0) when incubation time increased. Maximum activity for the enzyme incubated at different temperatures was observed to be 40°C. Optimum pH activity for α-amylase and pectinase is not completely consistent with the pH prevailing in the larval midgut. This is the first report of the presence of pectinase activity in H. postica.  相似文献

  1. Pissodes castaneus represents an emerging pest species for Pinus spp. production in Latin America. This species attacks all the cultivated pine species in Patagonia Argentina causing wood damage and tree death. The objectives of this work were to describe the host preference behaviour of P. castaneus and the influence of wood damage by conspecifics on its host selection.
  2. In two-choice bioassays, P. castaneus showed a feeding preference for Pinus contorta over P. ponderosa. However, percent weight gain when feeding on either species was similar.
  3. In other bioassays, P. castaneus spent more time in areas with twigs on which other conspecifics had recently fed, although they were able to successfully feed on twigs without previous damage.
  4. These results show that while P. castaneus can successfully colonize different pine species, P. contorta may be more susceptible to attack than P. ponderosa. Silvicultural management should be prioritized considering the weevil's preference for attacking damaged hosts.

Field and laboratory-choice experiments were conducted to understand aspects of host plant orientation by the Colorado potato beetle, Leptinotarsa decemlineata Say (Coleoptera: Chrysomelidae), in Virginia. In laboratory bioassays, L. decemlineata oriented to volatiles emitted by potato, Solanum tuberosum L., foliage over both tomato, Lycopersicon esculentum L., and eggplant, Solanum melongena L., foliage, and eggplant over tomato foliage, all of which had been mechanically damaged. Field choice tests revealed more L. decemlineata adults, larvae, and egg masses on eggplant than on tomato. In other experiments, counts of live L. decemlineata on untreated paired plants and counts of dead beetles on imidacloprid-treated plants did not differ between potato and eggplant. L. decemlineata was significantly attracted to eggplant over both tomato and pepper. To determine whether feeding adults affected orientation to host plants, an imidacloprid-treated eggplant or potato plant was paired with an untreated eggplant or potato plant covered in a mesh bag containing two adult male beetles. Significantly more adults were attracted to eggplant with feeding male beetles paired with another eggplant than any other treatment combination. These results indicate that the presence of male L. decemlineata on plants affects host plant orientation and suggests that the male-produced aggregation pheromone may be involved.  相似文献

Abstract 1 The dose–response of azadirachtin on vine weevil, Otiorhynchus sulcatus (Fabricius), reproduction is investigated by confining adults to feed on treated Taxus × media leaves, and by counting and evaluating development in the resulting eggs. 2 A dosage‐dependent reduction in oviposition is discovered for foliar surface residues of azadirachtin, with an EC50 of 25–50 parts per million (p.p.m) and 99.2% inhibition of viable egg production with 100 p.p.m. 3 Switching weevils from treated to untreated foliage allows reproductive capability to be restored for weevils that cease egg laying after azadirachtin exposure of 50 p.p.m. Weevils that had already started laying eggs in untreated groups soon cease oviposition once switched to azadirachtin‐treated foliage. 4 A transovarial effect results in a decrease in the percentage of viable eggs as the azadirachtin concentration increases. 5 The amount of feeding on foliage does not appreciably decrease at these hormonally effective concentrations, and adult weevil mortality is only slightly greater in the azadirachtin‐treated groups. Therefore, the overall effect of azadirachtin on weevil populations in the field is difficult to assess, except by collecting weevils to determine whether they are able to lay viable eggs.  相似文献

Since the erection of the weevil subfamily Baridinae by Schönherr in 1836, no phylogenetic hypothesis using cladistic methods has been proposed for this extraordinarily diverse group. This study provides the first hypothesis for the evolution of Baridinae using phylogenetic methods, including 301 taxa and 113 morphological characters. Despite fairly well‐resolved results, indicating paraphyly of nearly all of the currently recognized intrasubfamilial divisions, no change to the current classification is made. Even though groupings are proposed based on the final results, it is believed that more rigorous analyses need to be made prior to a re‐evaluation and subsequent alteration of the current classification. © 2010 The Linnean Society of London, Zoological Journal of the Linnean Society, 2010.  相似文献

Constant precision sampling plans for the white apple leafhopper, Typhlocyba pomaria McAtee, were developed so that it could be used as an indicator species for system stability as new integrated pest management programs without broad-spectrum pesticides are developed. Taylor's power law was used to model the relationship between the mean and the variance, and Green's constant precision sequential sample equation was used to develop sampling plans. Bootstrap simulations of the sampling plans showed greater precision (D = 0.25) than the desired precision (Do = 0.3), particularly at low mean population densities. We found that by adjusting the Do value in Green's equation to 0.4, we were able to reduce the average sample number by 25% and provided an average D = 0.31. The sampling plan described allows T. pomaria to be used as reasonable indicator species of agroecosystem stability in Washington apple orchards.  相似文献
