This study determined the effects of dietary branched-chain amino acids (AA) (BCAA) on growth performance, expression of jejunal AA and peptide transporters, and the colonic microflora of weanling piglets fed a low-protein (LP) diet. One hundred and eight Large White × Landrace × Duroc piglets (weaned at 28 days of age) were fed a normal protein diet (NP, 20.9 % crude protein), an LP diet (LP, 17.1 % crude protein), or an LP diet supplemented with BCAA (LP + BCAA, 17.9 % crude protein) for 14 days. Dietary protein restriction reduced piglet growth performance and small-intestinal villous height, which were restored by BCAA supplementation to the LP diet to values for the NP diet. Serum concentrations of BCAA were reduced in piglets fed the LP diet while those in piglets fed the LP + BCAA diet were similar to values for the NP group. mRNA levels for Na+-neutral AA exchanger-2, cationic AA transporter-1, b0,+ AA transporter, and 4F2 heavy chain were more abundant in piglets fed the LP + BCAA diet than the LP diet. However, mRNA and protein levels for peptide transporter-1 were lower in piglets fed the LP + BCAA diet as compared to the LP diet. The colonic microflora did not differ among the three groups of pigs. In conclusion, growth performance, intestinal development, and intestinal expression of AA transporters in weanling piglets are enhanced by BCAA supplementation to LP diets. Our findings provide a new molecular basis for further understanding of BCAA as functional AA in animal nutrition.  相似文献

Salmon protein hydrolysates (SPH) from two different rest raw materials were evaluated in diets for weaning piglets. Four experimental diets were included in the study: a diet based on plant protein with soy protein as the main protein source (Diet PP), a diet based on fishmeal in exchange for soy protein (Diet FM) and two diets in which different SPH replaced fishmeal in the FM diet. The experimental diets were fed to piglets from the day of weaning until 32 d postweaning. In addition to the record of performance data, an intestinal sampling for mucosal morphometry and microbiota 16S rRNA gene sequencing were performed at day 11 on a subset of the animals. The duodenal villi absorption area was significantly larger in piglets receiving Diets SPH compared with Diet PP (< 0.02). A significant positive correlation between duodenal villi height and average daily gain during the first 11 d postweaning was detected. Only small differences in intestinal microbiota community and no differences in growth performance were detected between the experimental diets. To conclude, SPH seem to be an interesting novel protein source in weanling piglets.  相似文献

本试验将40头21 d断奶的三元杂交猪随机分为4组,用4组不同日粮组成及营养水平的饲料(I:基础日粮+2%喷雾干燥血浆蛋白粉(SDPP);Ⅱ、Ⅲ和Ⅳ组分别在基础日粮中添加1%,2%和3%小肽营养素)饲养21 d后称重、测定血液生化指标并进行统计分析.结果表明:添加小肽营养素对断奶仔猪的生产性能无显著影响;各处理组的血清总蛋白、血糖和血清尿素差异不显著,2%小肽组的球蛋白、免疫球蛋白含量显著高于其它组;小肽产品可以部分等蛋白取代断奶仔猪日粮中的血浆蛋白粉.通过了解小肽营养素替代喷雾干燥血浆蛋白粉对21 d断奶仔猪的生产性能和血液生化指标的影响,为肽类产品在断奶仔猪日粮中的应用提供了依据.  相似文献

Performance, nutrient digestibility, feeding behaviour, diarrhoea frequency and faecal microbial diversity was studied in weaned piglets allotted to dry feed (diet A), liquid feed with water (diet B) or liquid feed with wet wheat-distillers grain (diet C). There were no differences in digestibility of organic matter and energy, and the performance was similar between piglets fed liquid diets. Piglets fed diet A spent more time on eating/drinking compared with piglets fed liquid diets. The frequency of diarrhoea decreased (p?<?0.05) from from diet A (11 out of 16) to diet B (6 out of 16) and further to diet C (1 out 16). The type of diet did not affect the diversity of coliform bacteria or lactobacilli, but the composition of the Lactobacillus flora was changed. The lower diarrhoea frequency when feeding diet C may be due to a prophylactic effect induced by the inclusion of wet wheat-distillers grain in the diet.  相似文献

A moderate threonine deficiency may affect differently tissue protein metabolism. In this study, we compared protein metabolism in the small and large intestines, the liver, and the carcass of piglets (Sus scrofa) pair-fed either a control well-balanced diet (C: 9.3 g threonine/kg diet) or a low threonine diet (LT: 6.5 g threonine/kg diet) for 2 weeks. In the small intestine, the LT diet did not modify protein deposition, fractional protein synthesis rate (KS) and AA protein composition. Ubiquitin mRNA level, a component of the ubiquitin-dependent proteolytic pathway, was significantly decreased in the jejunum of the LT piglets. Protein deposition measured in the carcass and the colon, and KS measured in the semitendinosus muscle and the colon, did not differ between LT and C piglets. Nevertheless, in these compartments, threonine content was reduced indicating deposition of proteins less rich in threonine. In the liver, protein retention was reduced, KS was increased and AA protein composition was modified in the LT compared to the C piglets. In conclusion compared to the other compartments, small intestinal protein metabolism seems to be less sensitive to a moderate dietary threonine deficiency. This indicates that dietary threonine extraction by the small intestine may reduce threonine availability for the other tissues when young piglets were fed a diet marginally deficient in threonine.  相似文献

The influence of feeding, egg load and stage of ovarian development on the sexual receptivity of some insect species has been reported. This work investigated the effects of different brewer's yeast concentrations in the diet offered to females of West Indian fruit fly, Anastrepha obliqua, on their sexual receptivity. Wild females were fed on artificial diets containing 2.0 g brewer's yeast per 100 mL of water (low‐yeast diet, LY diet), 6.5 g yeast per 100 mL of water (high‐yeast diet, HY diet) or only sucrose (Su diet) and then placed with the males. Female sexual receptivity tests were carried out in acrylic boxes in the laboratory under controlled conditions (temperature, humidity and light). Females fed on the HY diet were more sexually receptive to male courtship than the other females. Additionally, when fed on the same diet (HY diet), those females with higher egg load were more sexually receptive. The data suggest that the ingestion of a protein source is essential for increasing sexual receptivity and that egg load is the most important factor affecting sexual receptivity of A. obliqua females.  相似文献

为了探讨大豆寡糖(SBOS)作为抗生素替代物对断奶仔猪生长性能的影响及其作用机制,饲养试验选用21日龄断奶的环江香猪12头,随机分为2组,每组6头,分别饲喂添加0.5%SBOS日粮和抗生素对照日粮,试验期为14 d。记录每天的采食量,分别于试验开始和结束时称取空腹体重并采集血液,肝素抗凝离心分离血浆,测定生化参数和游离氨基酸水平。消化试验选用10头环江香猪,随机分为2组,处理同饲养试验。预试期4 d,正试期3 d,收集粪样,指示剂法测定干物质、粗蛋白和粗脂肪的消化率。结果表明,与抗生素对照组相比,SBOS组仔猪平均日采食量、日增重和料重比均无显著变化;血浆尿素氮和血氨含量显著下降,总蛋白含量显著升高(P<0.05);血浆甘氨酸、组氨酸、赖氨酸、精氨酸和蛋氨酸的含量均显著升高(P<0.05);粗蛋白和粗脂肪消化率有提高趋势。提示SBOS可替代抗生素用于断奶仔猪日粮。  相似文献

目的 探讨复合微生态制剂对断奶仔猪生长性能、粪便乳酸菌和大肠埃希菌数、血液红胞数以及血红蛋白含量及其对免疫水平的影响.方法 选择25日龄的断奶仔猪共108头,平均分为两组:Ⅰ组和Ⅱ组,每组3个重复,每个重复18只,Ⅰ组饲喂全价料+2.0‰复合微生态制剂;Ⅱ组为对照组,饲喂全价料,实验期为42 d.结果 至实验结束时,复合微生态制剂组的料重比显著低于对照组(P <0.05);14、28和42 d时复合微生态制剂组粪便中的大肠埃希菌数比对照组低7.94%,2.90%和4.89%,与对照组相比差异有统计学意义(P<0.05);14d时,复合微生态制剂组粪便乳酸菌数与对照组相比差异有统计学意义(P<0.05),28和42 d时,两组粪便乳酸菌数差异无统计学意义(P>0.05).14、28和42 d时复合微生态制剂组血液中红细胞数和血红蛋白浓度均显著高于对照组.两组血清尿素氮水平差异无统计学意义(P>0.05),复合微生态制剂组血清总蛋白和IgG的含量与对照组相比有升高的趋势,但差异无统计学意义(P>0.05),复合微生态制剂组血清IgA和IgM的浓度显著高于对照组(P<0.05).结论 复合微生态制剂可以显著降低断奶仔猪的料重比,降低粪便中大肠埃希菌数量,促进血液红细胞和血红蛋白的含量,提高断奶仔猪的免疫水平.  相似文献

The effect of dietary supplementation with Saccharomyces cerevisiae on gilthead seabream (Sparus aurata L.) was studied. Four replicates of fish (n = 6) were fed with a commercial diet containing 0 (control, no yeast added) or 10 mg per kilogram of heat-killed (30 min, 60°C) S. cerevisiae. After 4 weeks, half of the fish (two replicates) were injured and continued with the same diet. At 3 and 7 days post-wounding, samples of blood, skin mucus, skin and liver were obtained from each fish. The results showed that calcium concentrations were significantly higher (with respect to control fish) in the serum from fish sampled at 3 days post-wounding, whereas antioxidant enzymes in the skin mucus were altered after wounding (at both 3 and 7 days). Histological analyses revealed oedema, signs of inflammation and white cell recruitment together with a reduction in the epidermis layer in the wounded regions of fish fed control diet. Yeast supplementation did not change growth performance and helped maintain the normal serum calcium concentrations in wounded fish. Furthermore, a reduction in inflammation around wounds in the animals fed yeast with respect to that fed control diet was evident in the histological study. Furthermore, increased levels of stress-related gene expression in liver and skin from wounded fish were obtained. Overall, yeast supplementation seemed to be a functional and appropriate dietary additive to improve skin recovery reducing the stress resulting from wounds.  相似文献

The mean assimilation efficiency of aquarium acclimatized Oreochromis mossambiaue fed on a diet of Microcystis aeruginosa collected from Hartbeespoort Dam was determined as 50,8% for total organic matter, 63,7% for protein and 75,5% for phosphorus. Transmission electron microscopic examination of faeces of fish fed on M. aeruginosa, revealed that most Microcystis cell walls had become permeable allowing cell contents to leach out. Further digestion resulted in the break down of the cell wall structure. Up to 25% of the cells, however, appeared intact after passing through the fish. Fish fed on a diet of M. aeruginosa lost mass initially, but after 21 days showed a slight gain in mass. The high protein content of M. aeruginosa nay have inhibited efficient metabolism and would have led to reduced growth in fish.  相似文献

Whole cells and cell walls of the mycelial and yeast forms ofBlastomyces dermatitidis grown in four different media were analyzed for differences in lipid, fatty acid, carbohydrate, and protein contents. The bound (saponifiable) fatty acids of yeast and mycelial whole cells (but not the cell walls) vary considerably in response to growth medium. The percentage of readily extractable lipid varied somewhat in whole cells. The percentage of carbohydrate and protein of whole cells and cell walls are little affected by the medium in which the cells are grown.  相似文献

Feeding high-protein diets in animals can lead to a decrease of nitrogen utilization efficiency, and then promote the environmental pollution. Recently, more reports have demonstrated that lowering protein level in diets supplemented with specific amino acids can address these problems. However, the whole proteome alteration in the skeletal muscle of weaned piglets fed low-protein diets is poorly understood. Here, we applied the isobaric tags for relative and absolute quantification approach to investigate this alteration. We fed weaned piglets with normal protein diet (20% crude protein) and low-protein diet supplemented with lysine, methionine, threonine, and tryptophan (17% crude protein) for 25 days. Then proteomic profiling of skeletal muscles was performed. In total, 1354 proteins were quantified in the porcine skeletal muscle proteome. 132 proteins were identified as differentially expressed proteins between the two groups. Differentially expressed proteins were significantly enriched in various nutrient metabolism including lipid, carbohydrate, and amino acid metabolism. Interestingly, a total of 20 differentially expressed proteins, which are involved in the oxidative phosphorylation pathway, were all down-regulated by the low-protein diet feeding. Further immunoblotting confirmed the down-regulations of MT-ATP8, COX2, NDUFA6, and SDHB, four selected proteins among these 20 proteins. Meanwhile, the ATP level in the low-protein diet group was also reduced. These findings for the first time reveal that oxidative phosphorylation pathway is suppressed in longissimus dorsi muscle of weaned piglets fed low-protein diet supplemented with limiting amino acids, which may provide new insights into further formula design and the choice of limiting amino acids in diets.  相似文献

The cell walls of the yeast and mycelial forms of Yarrowia lipolytica were isolated and purified. Electron microscopy studies showed no differences between both types of cell walls. Chemical analysis revealed that the yeast cell wall contained 70% neutral carbohydrate, 7% amino sugars, 15% protein, 5% lipids and 0.8% phosphorus. Mycelial cell walls contained 70% carbohydrate, 14% aminosugars, 6% protein, 5% lipids and 0.6% phosphorus. Three polysaccharides: -glucan, mannan and chitin were detected. Proteins were solubilized from both cell wall fractions and separated by polyacrylamide gel electrophoresis. About 50 protein bands were detected, four of them corresponding to glycoproteins. A 21-day study was conducted to determine whether isoleucine might limit the performance of piglets fed low-crude protein (CP), amino acid (AA)-supplemented diets and to investigate the potential benefits of low-CP diets on gastrointestinal health in weaned pigs. Ninety-six piglets (initial BW = 6.44 ± 0.14 kg), housed four per pen, were randomly assigned to one of four diets, resulting in six replicate pens per diet. Dietary treatments were as follows: (1) 210 g/kg CP diet, (2) 190 g/kg CP diet deficient in isoleucine, (3) 190 g/kg CP diet supplemented with crystalline isoleucine up to the level in the 210 g/kg CP diet and (4) 170 g/kg CP diet supplemented with isoleucine and valine on the ideal protein ratio basis (60% and 70% relative to lysine, respectively). Pigs were allowed to adapt to the new environment for 4 days before the experiment commenced. Overall, pigs fed the 210 g/kg CP diet had higher (P < 0.05) average daily gain and lower (P < 0.05) feed : gain ratio compared with those fed the other diets. The faecal consistency score of pigs fed the 210 g/kg CP diet was higher (P < 0.05) than those fed the other diets. Pigs fed the 170 g/kg diet had lower (P=0.02) small intestine weight than those fed the 210 g/kg CP diet. Pigs fed the 210 g/kg CP diet had deeper (P < 0.05) crypt in the duodenum and ileum and higher (P < 0.05) ammonia N concentration in caecal digesta than those fed the other diets. There were no effects of diet on microbial population and volatile fatty acid concentration in the caecal digesta except for propionic acid whose concentration was higher (P < 0.05) for pigs fed the 170 g/kg diet than those fed the 190+isoleucine and the 210 g/kg CP diets. The results indicate that the low-CP, AA-supplemented diet reduced crypt hypertrophy, ammonia N concentration in the caecal digesta, small intestine weight and the performance of piglets. Also, the results of the current study were inconclusive with respect to whether isoleucine may limit the performance of pigs fed a low-CP, AA-supplemented diet.  相似文献

The aim of the experiment on 180 weaned piglets (8.9 kg body weight) was to investigate the influence of high fibre diets formulated with different fibrous ingredients on performance, nutrient digestibility, diarrhoea incidence and numbers of faecal microbiota. The dietary treatments included a Control diet and five high fibre diets formulated with different fibre sources including wheat bran, soybean hulls, naked oat hulls, palm kernel expeller and bamboo fibre. The high fibre diets averaged 14.6% neutral detergent fibre with different non-starch polysaccharides (NSP) components and were fed ad libitum for 28 d. Faecal samples were collected during the last 3 d of the experiment and the apparent total tract digestibility of nutrients and fibre components were determined. Pigs fed the Control and wheat bran diets had a higher (≤ 0.05) average daily gain (ADG) than pigs fed the palm kernel expeller and bamboo meal diets. The reduced ADG for pigs appeared to be related to reductions in the digestibility of gross energy and dry matter, respectively. The feed-to-gain ratio was significantly higher (≤ 0.05) for pigs fed the fibre diets. The digestibility of NSP components was different among the treatments. The diarrhoea incidence was not affected by treatments. The abundance of faecal bifidobacteria was significantly higher (≤ 0.05) for pigs fed the wheat bran diet than for pigs fed the bamboo meal diet. It was concluded that the diets formulated with different fibre sources when fed to weaned piglets have different effects on pig performance, nutrient digestibility and numbers of faecal microbiota. The wheat bran diet rich in arabinoxylans enabled a better performance than the other tested diets with fibre addition.  相似文献

The objective of this study was to investigate the effects of free amino acids supplementation to protein restricted diet on the intestinal morphology and proteome composition in weaning pigs. Weanling piglets were randomly fed one of the three diets including a corn-soybean based control diet and two lower protein diets with or without free amino acids supplementation for 2 weeks. The jejunum samples of piglets were collected for morphology and proteome analysis. Compared with the control diet, the protein restricted diet had a significant lower average daily gain and higher feed conversion rate. Free amino acids supplementation to the protein restricted diet significantly improved average daily gain and higher feed conversion rate, compared with the protein restricted diet. The villous height in pigs fed the protein restricted diet was lower than that of the control and free amino acids diet. Using two-dimensional gel electrophoresis and mass spectrometry, we identified 16 differentially expressed protein spots in the jejunum of the weaning piglet. These proteins were related to stress and immune response, the metabolism of carbohydrates and lipids, and tissue structure. Based on the proteome and ELISA analysis, free amino acids diet significantly down-regulated the jejunal expression of stress protein heat shock 60 kDa protein. Our results indicated that amino acids supplementation to the protein restricted diet could enhance weight gain and feed efficiency in weanling pigs through improving intestinal nutrient absorption and transportation, gut health, and mucosal immunity.  相似文献

Dietary supplementation with a high level of zinc oxide (ZnO) has been shown to reduce the incidence of diarrhea in weanling pigs, but the underlying mechanisms remain largely unknown. Intestinal–mucosal mast cells, whose maturation and proliferation is under the control of the stem cell factor (SCF), play an important role in the etiology of diarrhea by releasing histamine. The present study was conducted to test the novel hypothesis that supplementing ZnO to the diet for weanling piglets may inhibit SCF expression in the small intestine, thereby reducing the number of mast cells, histamine release, and diarrhea. In Experiment 1, 32 piglets (28 days of age) were weaned and fed diets containing 100 or 3000 mg zinc/kg (as ZnO) for 10 days (16 piglets per group). In Experiment 2, two groups of 28-day-old piglets (8 piglets per group) were fed the 100- or 3000-mg zinc/kg diet as in Experiment 1, except that they were pair-fed the same amounts of feed. Supplementation with a high level of ZnO reduced the incidence of diarrhea in weanling piglets. Dietary Zn supplementation reduced expression of the SCF gene at both mRNA and protein levels, the number of mast cells in the mucosa and submucosa of the small intestine and histamine release from mucosal mast cells. Collectively, our results indicate that dietary supplementation with ZnO inhibits SCF expression in the small intestine, leading to reductions in the number of mast cells and histamine release. These findings may have important implications for the prevention of weaning-associated diarrhea in piglets.  相似文献
