Clathrin in mitotic spindles   总被引:3,自引:0,他引:3  
Subconfluent cultures ofMadin-Darby canine kidney (MDCK) and CV-1 cells were immunostained withtwo monoclonal antibodies (MAbs), MAb X-22 and MAb 23, against clathrinheavy chain and with polyclonal antiserum against a conserved region ofall mammalian clathrin light chains. In interphase MDCK and CV-1 cells,staining by all three antibodies resulted in the characteristicintracellular punctate vesicular and perinuclear staining pattern. Inmitotic cells, all three anti-clathrin antibodies strongly stained the mitotic spindle. Staining of clathrin in the mitotic spindle was colocalized with anti-tubulin staining of microtubular arrays in thespindle. Staining of the mitotic spindle was evident in mitotic cellsfrom prometaphase to telophase and in spindles in mitotic cellsreleased from a thymidine-nocodazole block. In CV-1 cells, staining ofclathrin in the mitotic spindle was not affected by brefeldin A. OnWestern blots, clathrin was detected, but not enriched, in isolatedspindles. The immunodetection of clathrin in the mitotic spindle maysuggest a novel role for clathrin in mitosis. Alternatively, therecruitment of clathrin to the spindle may suggest a novel regulatorymechanism for localization of clathrin in mitotic cells.

Formation of the complete spindles during the budding process of Saccharomyces uvarum was investigated by fluorescence microscopy of protoplasted cells. Protoplasts were treated with anti-tubulin antibodies and DAPI, a fluorescent dye staining DNA. Thus, both chromatin and spindles could be visualized. Duplication as well as formation of separated spindle pole bodies during the different stages of budding are documented, demonstrating the occurrence and behaviour of microtubules during yeast cell cycle.  相似文献

A new cell type named telocyte (TC) has recently been identified in various stromal tissues, including skeletal muscle interstitium. The aim of this study was to investigate by means of light (conventional and immunohistochemical procedures) and electron microscopy the presence of TCs in adult human neuromuscular spindles (NMSs) and lay the foundations for future research on their behaviour during human foetal development and in skeletal muscle pathology. A large number of TCs were observed in NMSs and were characterized ultrastructurally by very long, initially thin, moniliform prolongations (telopodes – Tps), in which thin segments (podomeres) alternated with dilations (podoms). TCs formed the innermost and (partially) the outermost layers of the external NMS capsule and the entire NMS internal capsule. In the latter, the Tps were organized in a dense network, which surrounded intrafusal striated muscle cells, nerve fibres and vessels, suggesting a passive and active role in controlling NMS activity, including their participation in cell‐to‐cell signalling. Immunohistochemically, TCs expressed vimentin, CD34 and occasionally c‐kit/CD117. In human foetus (22–23 weeks of gestational age), TCs and perineural cells formed a sheath, serving as an interconnection guide for the intrafusal structures. In pathological conditions, the number of CD34‐positive TCs increased in residual NMSs between infiltrative musculoaponeurotic fibromatosis and varied in NMSs surrounded by lymphocytic infiltrate in inflammatory myopathy. We conclude that TCs are numerous in NMSs (where striated muscle cells, nerves and vessels converge), which provide an ideal microanatomic structure for TC study.  相似文献

Proprioceptive innervation of moufflon extrinsic ocular musculature and m. levator palpebrae superioris was studied. Muscle spindles and Golgi tendon organs were found. The first ones are usually between 1st and 2nd order muscle fascicles. The muscle spindles are highly represented in the extrinsic ocular muscles, but less numerous in m. levator palpebrae superioris. Their number varies according to muscles and individuals. In the same subject, also the ratio between the number of the muscle spindles found in m. rectus dorsalis and that of m. levator palpebrae superioris was examined. Besides, the histological structure of the intrafusal fascicles was investigated. Particular attention was devoted to the nerve supply of the muscle spindle. By means of impregnating methods, sensory and motor endings were identified. Primary and secondary sensory endings only in a few cases showed their usual pattern: motor fibres can end in form of plates or trails. Golgi tendon organs were observed between the tendon and the muscular tissue and are always less numerous.  相似文献

In a previous paper (Inbar, 1972/III) a new adaptive model was proposed for muscular control. According to that scheme muscle spindles (MS) supply continuous information about the system dynamics. In the present study integral pulse frequency modulation (IPFM) is used to correlate MS afferent signals, recorded from frog sartorius muscle, to the same muscle dynamics, in order to establish the feasibility of the proposed MS role in the adaptive muscle model.  相似文献
