Racemic γ-ionone, partly resolved via its menthydrazone, was used for total synthesis of β,γ-carotene enriched in the 6′R and 6′S enantiomers. By CD correlation with natural β,γ-carotene isolated from Caloscypha fulgens 6′S-chirality is demonstrated for the natural carotene. Biosynthetic implications regarding the cyclization reaction are discussed.  相似文献

In a leaf flavonoid analysis of six Fuchsia species and seven Fuchsia hybrids, flavonols were found to be abundant in all taxa except F. procumbens. Flavone glycosides were found in only three species: luteolin 7-glucoside in F. splendens; and luteolin and apigenin 7-glucuronides and 7-glucuronidesulphates, tricin 7-glucuronidesulphate and diosmetin 7-glucuronide from one or both of the New Zealand species F. procumbens and F. excorticata. Luteolin 7- glucuronidesulphate is reported for the first time. Other less common phenolics identified include the flavanone, eriodictyol 7-glucoside from F. excorticata, a galloylglucose from F. triphylla, and a galloylglucosesulphate present in all taxa. Eight of the flavonoid glycosides proved useful as marker substances for particular Fuchsia species: quercetin 3- rhamnoside, 3-glucuronide and 3-rutinoside for F.fulgens; quercetin and kaempferol 3-galactosides for F. boliviana var. luxurians; diosmetin 7-glucuronide for F. excorticata and apigenin 7-glucuronide and 7-glucuronidesulphate for F. procumbens. The chemical results on the hybrids support the view that the cultivar ‘Mary’ is a hybrid of F. boliviana var. luxurians and F. triphylla and that both F.fulgens and F. triphylla are involved as parents of the cultivars ‘Koralle’ and ‘Traudchen Bondstedt’.  相似文献

The morphology and the anatomy of the 13 species presently included in the genusFulgensiawere surveyed.Caloplaca aureaandC. paulii, two species regarded as closely related toFulgensiaby earlier authors, were also included in the study.Fulgensiawas found to exhibit a great variation in both morphology and anatomy. The differences in size, shape and septation of the spores, in the anatomy of cortex and exciple, and in the gross morphology, will presumably motivate a different taxonomic treatment in the future. A tentative separation of the species into four different groups is proposed here. Group A comprisesF. australisandF. chanousiae, which appear close to species inCaloplacasect.Gasparrinia.Groups B and C, each comprises one species,F. schistidiiandF. canariensisrespectively and group D comprises the remaining nine species, including the type speciesF. fulgens. The last group may be further split by a more detailed investigation. The present separation ofFulgensiainto two subgenera cannot be regarded as taxonomically sound. The genus is probably polyphyletic and the different groups are probably related to different groups within the large genusCaloplaca.  相似文献

Mulberry, belonging to the order Rosales, family Moraceae, and genus Morus, has received attention because of both its economic and medicinal value, as well as for its important ecological function. The genus Morus has a worldwide distribution, however, its taxonomy remains complex and disputed. Many studies have attempted to classify Morus species, resulting in varied numbers of designated Morus spp. To address this issue, we used information from internal transcribed spacer (ITS) genetic sequences to study the taxonomy of all the members of generally accepted genus Morus. We found that intraspecific 5.8S rRNA sequences were identical but that interspecific 5.8S sequences were diverse. M. alba and M. notabilis showed the shortest (215 bp) and the longest (233 bp) ITS1 sequence length, respectively. With the completion of the mulberry genome, we could identify single nucleotide polymorphisms within the ITS locus in the M. notabilis genome. From reconstruction of a phylogenetic tree based on the complete ITS data, we propose that the Morus genus should be classified into eight species, including M. alba, M. nigra, M. notabilis, M. serrata, M. celtidifolia, M. insignis, M. rubra, and M. mesozygia. Furthermore, the classification of the ITS sequences of known interspecific hybrid clones into both paternal and maternal clades indicated that ITS variation was sufficient to distinguish interspecific hybrids in the genus Morus.  相似文献

Understanding the systematics and evolution of clavicipitoid fungi has been greatly aided by the application of molecular phylogenetics. They are now classified in three families, largely driven by reevaluation of the morphologically and ecologically diverse genus Cordyceps. Although reevaluation of morphological features of both sexual and asexual states were often found to reflect the structure of phylogenies based on molecular data, many species remain of uncertain placement due to a lack of reliable data or conflicting morphological characters. A rigid, darkly pigmented stipe and the production of a Hirsutella-like anamorph in culture were taken as evidence for the transfer of the species Cordyceps cuboidea, Cordyceps prolifica, and Cordyceps ryogamiensis to the genus Ophiocordyceps. Data from ribosomal DNA supported these species as a single group, but were unable to infer deeper relationships in Hypocreales. Here, molecular data for ribosomal and protein coding DNA from specimens of Ophiocordyceps cuboidea, Ophiocordyceps ryogamiensis, Ophiocordyceps paracuboidea, Ophiocordyceps prolifica, Cordyceps ramosopulvinata, Cordyceps nipponica, and isolates of Polycephalomyces were combined with a broadly sampled dataset of Hypocreales. Phylogenetic analyses of these data revealed that these species represent a clade distinct from the other clavicipitoid genera. Applying the recently adopted single system of nomenclature, new taxonomic combinations are proposed for these species in the genus Polycephalomyces, which has been historically reserved for asexual or anamorphic taxa.  相似文献

Eight NE Atlantic and Mediterranean species, which were originally assigned to the genus Schizoporella (Family Schizoporellidae) when introduced, are redescribed and stabilized by typification. Seven of these species are transferred to the bitectiporid genus Schizomavella: S. fischeri, S. glebula, S. neptuni, S. obsoleta, S. richardi, S. triaviculata, and S. triaviculata var. paucimandibulata, which is here raised to species rank. The eighth species, Schizoporella fayalensis, is transferred to the lanceoporid genus Stephanotheca. Schizomavella obsoleta and S. glebula are considered junior subjective synonyms of S. fischeri and S. richardi, respectively. Two new species are described: Schizomavella rectangularis n. sp. from the Strait of Gibraltar, and Schizomavella phterocopa n. sp. from the Great Meteor Bank. A new subgenus, Calvetomavella n. subgen. is established as a result of a phylogenetic analysis based on morphological characters; it includes S. neptuni, S. triaviculata, S. paucimandibulata and S. phterocopa n. sp., together with Schizomavella discoidea and Schizomavella noronhai. The rest of the species remain in the nominotypical subgenus Schizomavella.  相似文献

The genus Agrobacterium was created a century ago by Conn who included it in the family Rhizobiaceae together with the genus Rhizobium. Initially, the genus Agrobacterium contained the non-pathogenic species Agrobacterium radiobacter and the plant pathogenic species Agrobacterium tumefaciens and Agrobacterium rhizogenes. At the end of the past century two new pathogenic species, Agrobacterium rubi and Agrobacterium vitis, were added to the genus. Already in the present century these species plus Agrobacterium larrymoorei were reclassified into genus Rhizobium. This reclassification was controversial and for a time both genus names were used when new species were described. Few years ago, after a taxonomic revision based on genomic data, the old species A. rhizogenes was maintained in the genus Rhizobium, the old species A. vitis was transferred to the genus Allorhizobium and several Rhizobium species were transferred to the genus Agrobacterium, which currently contains 14 species including the old species A. radiobacter, A. tumefaciens, A. rubi and A. larrymoorei. Most of these species are able to produce tumours in different plants, nevertheless the genus Agrobacterium also encompasses non-pathogenic species, one species able to nodulate legumes and one human pathogenic species. Taking into account that the species affiliations to five Agrobacterium genomospecies have not been determined yet, an increase in the number of species within this genus is expected in the near future.  相似文献

The generic segregates of the widespread fruticose genus Ramalina (mostly based on empirical data on morphology, cortex anatomy and secondary metabolites) are studied using maximum parsimony, maximum likelihood and Bayesian analyses of nuclear LSU and ITS sequences. The species examined include the three species aggregates within Niebla from the western coasts of North America, all species except one assumed to belong to the same genus from Macaronesia and the Mediterranean basin, the type species of Dievernia and Ramalina, and representatives of the genus Fistulariella. The genus Niebla is strongly supported when restricted to species from the New World, and all species referred to it from Macaronesia and the Mediterranean basin belong to Ramalina (R. bourgeana, R. crispatula, R. cupularis, R. hamulosa, R. portosantana, R. rosacea, R. subwebbiana and R. webbii). No support is found for the genera Dievernia and Fistulariella. The internal topology of the large genus Ramalina is unresolved and needs further studies.  相似文献

Background

Sporotrichosis is a chronic (sub)cutaneous infection caused by thermodimorphic fungi in the order, Ophiostomatales. These fungi are characterized by major differences in routes of transmission, host predilections, species virulence, and susceptibilities to antifungals. Sporothrix species emerge in the form of outbreaks. Large zoonoses and sapronoses are ongoing in Brazil and China, respectively. Current diagnostic methods based on morphology and physiology are inaccurate due to closely related phenotypes with overlapping components between pathogenic and non-pathogenic Sporothrix. There is a critical need for new diagnostic tools that are specific, sensitive, and cost-effective.

Methodology

We developed a panel of novel markers, based on calmodulin (CAL) gene sequences, for the large-scale diagnosis and epidemiology of clinically relevant members of the Sporothrix genus, and its relative, Ophiostoma. We identified specific PCR-based markers for S. brasiliensis, S. schenckii, S. globosa, S. mexicana, S. pallida, and O. stenoceras. We employed a murine model of disseminated sporotrichosis to optimize a PCR assay for detecting Sporothrix in clinical specimens.

Results

Primer-BLAST searches revealed candidate sequences that were conserved within a single species. Species-specific primers showed no significant homology with human, mouse, or microorganisms outside the Sporothrix genus. The detection limit was 10–100 fg of DNA in a single round of PCR for identifying S. brasiliensis, S. schenckii, S. globosa, S. mexicana, and S. pallida. A simple, direct PCR assay, with conidia as a source of DNA, was effective for rapid, low-cost genotyping. Samples from a murine model of disseminated sporotrichosis confirmed the feasibility of detecting S. brasiliensis and S. schenckii DNA in spleen, liver, lungs, heart, brain, kidney, tail, and feces of infected animals.

Conclusions

This PCR-based method could successfully detect and identify a single species in samples from cultures and from clinical specimens. The method proved to be simple, high throughput, sensitive, and accurate for diagnosing sporotrichosis.  相似文献

Due to severe declines in abundance throughout southern California, the green abalone (Haliotis fulgens Philippi 1845) became protected under a state-sponsored fishery moratorium in 1997 and was declared a NOAA NMFS Species of Concern in 2004. Recently, H. fulgens was chosen for possible stock restoration via translocation of wild adults to depleted habitat and supplementation through releasing cultured individuals. Before a management plan could be developed, however, an understanding of the species’ natural population genetic structure was needed. We used a genomic technique called restriction site associated DNA sequencing (RADSeq) to address the issue. RADSeq enabled discovery of 1,209 single nucleotide polymorphisms theoretically spread genome-wide in H. fulgens. Analyses suggested the species may be panmictic throughout our sampled range, with an effective population size (Ne) of 1,100–3,600. Hence, limitations to management, such as requiring local broodstock and restricting translocation potential, might be unnecessary. Sites with larger populations may be suitable sources for restoration of depleted sites (e.g. the Palos Verdes Peninsula), although the extent of local adaptation remains unknown. Despite this potential for restoration, results gathered on a sample of cultured H. fulgens illustrated how quickly genetic diversity can be lost through captive breeding. To help mitigate a drop in Ne due to hatchery supplementation, we recommend collection and replacement of ≥100 wild abalone per generation for broodstock and close management of the proportion of cultured individuals in the wild. Successful implementation will depend on operational capacity and the resilience of the source populations to broodstock collection.  相似文献

Anaporrhutine gorgoderids (Digenea: Gorgoderidae: Anaporrhutinae) found in the body cavity of six species of elasmobranchs from the orders Carcharhiniformes, Myliobatiformes and Orectolobiformes from Australian waters were found to belong to the genus Staphylorchis. Although these specimens were morphologically variable, sequences of ITS2 and 28S ribosomal DNA from specimens from three host families and two host orders were identical. Based on morphological and molecular data these specimens were identified as the type-species of the genus, Staphylorchis cymatodes. New measurements are provided for S. cymatodes, and for the first time genetic data are presented for this species. In addition to providing new morphological and molecular data for S. cymatodes, the previously described species S. gigas, S. parisi and S. scoliodonii, are here synonymised with S. cymatodes. This implies that S. cymatodes, as conceived here, has remarkably low host-specificity, being recorded from eight elasmobranch species from four families and three orders, has a wide geographical distribution in the Indo-west Pacific from off India, in the Bay of Bengal, to Moreton Bay in the Coral Sea, and is morphologically plastic, with body size, size of specific organs and body shape differing dramatically between specimens from different host species. The genus Staphylorchis now contains only two valid species, S. cymatodes and S. pacifica.  相似文献

The authenticity of controversial species is a significant challenge for systematic biologists. Moschidae is a small family of musk deer in the Artiodactyla, composing only one genus, Moschus. Historically, the number of species in the Moschidae family has been debated. Presently, most musk deer species were restricted in the Tibetan Plateau and surrounding/adjacent areas, which implied that the evolution of Moschus might have been punctuated by the uplift of the Tibetan Plateau. In this study, we aimed to determine the evolutionary history and delimit the species in Moschus by exploring the complete mitochondrial genome (mtDNA) and other mitochondrial gene. Our study demonstrated that six species, M. leucogaster, M. fuscus, M. moschiferus, M. berezovskii, M. chrysogaster and M. anhuiensis, were authentic species in the genus Moschus. Phylogenetic analysis and molecular dating showed that the ancestor of the present Moschidae originates from Tibetan Plateau which suggested that the evolution of Moschus was prompted by the most intense orogenic movement of the Tibetan Plateau during the Pliocene age, and alternating glacial-interglacial geological eras.  相似文献

The present article offers a detailed review of the taxonomy, distribution and palaeoecology of the genus Semigenetta. The study is based on new craniodental and postcranial remains of the genus from the early late Miocene (Tortonian) locality of Hammerschmiede (Bavaria, Germany). Most of the new specimens are attributed to the medium-sized species Semigenetta sansaniensis, whereas one lower carnassial is assigned to the large-sized Semigenetta grandis, making Hammerschmiede 4 the first known locality with two species of the genus. The variability of the material of S. sansaniensis from Europe allows us to revise the taxonomic weight of some previously used characters, and to identify the smaller-sized late Miocene form Semigenetta ripolli as a junior synonym of the former. Such an evolutionary transition of S. sansaniensis towards smaller forms is explained by niche partitioning with larger carnivorans of similar ecology, such as the herein reported S. grandis. Additionally, the species Semigenetta huaiheensis is here considered as a junior synonym of Semigenetta elegans.  相似文献

Mesotheres (Notoungulata: Typotheria) are among the most common mammals found in upper Miocene to Pliocene deposits of central Argentina, including the classic type Monte Hermoso locality, which defines the Montehermosan South American Land Mammal “Age”. Nevertheless, the correct name for the mesothere species from this site has been shrouded in uncertainty for well over a century due to questions of taxonomic priority, specimen provenance, and ontogenetic changes in dental formula. Since the mesotheres from Monte Hermoso were named, three distinct species have been formally considered as the type species of the genus: (1) Pseudotypotherium bravardi; (2) “Pseudotypotheriummaendrum; and (3) Pseudotypotherium exiguum. However, none of these species is a nominal species of the Pseudotypotherium genus; all three were originally referred to Typotherium. Article 67.2 of the International Code of Zoological Nomenclature (ICZN, 1999) indicates that only species considered as nominal species are eligible to set the type; in the case of Pseudotypotherium, these include: P. pulchrum, P. carlesi, P. hystatum, and P. carhuense. We conclude that Pseudotypotherium pulchrum F. Ameghino, 1904 (holotype MACN A 10299, Museo Argentino de Ciencias Naturales “Bernardino Rivadavia”, Ameghino Collection), is the type species of the mesotheriid notoungulate genus from Monte Hermoso. According to Article 68.2, F. Ameghino fixed the type by original designation in 1904 when he described P. pulchrum and included “n. g., n. sp.”. Two of the other species previously considered species P. (= T.) bravardi and P. (= T.) exiguum are invalid as type species according to Article 70.2, since their designations overlooked the previous type fixation. The third species (M. (= T.) maendrum) represents a different mesothere genus (Mesotherium) that only occurs in younger (Pleistocene) deposits. Our analysis puts an end to a historical debate that has been ongoing for more than a century regarding the identity of this well-represented late Miocene–Pliocene mesotheriine genus (Pseudotypotherium). This study provides a solid taxonomic foundation for future studies on intraspecific and ontogenetic variation of Pseudotypotherium pulchrum.  相似文献

The Lower Jurassic genus Eodasycladus is discussed according to the characters of type species and compared with the other species known in the literature. The architecture of two species attributed to the genus Palaeodasycladus, P. alanensis Soka?, and P. benceki Soka?, is examined and an alternative organization of the thallus is prospected. P. alanensis is considered a valid species, but its characters require to transfer the taxon to the genus Eodasycladus. P. benceki is considered synonymous with P. alanensis.  相似文献

The Asian Tree Toad genus Pedostibes, as currently understood, exhibits a conspicuously disjunct distribution, posing several immediate questions relating to the biogeography and taxonomy of this poorly known group. The type species, P. tuberculosus and P. kempi, are known only from India, whereas P. hosii, P. rugosus, and P. everetti are restricted to Southeast Asia. Several studies have shown that these allopatric groups are polyphyletic, with the Indian Pedostibes embedded within a primarily South Asian clade of toads, containing the genera Adenomus, Xanthophryne, and Duttaphrynus. Southeast Asian Pedostibes on the other hand, are nested within a Southeast Asian clade, which is the sister lineage to the Southeast Asian river toad genus Phrynoidis. We demonstrate that Indian and Southeast Asian Pedostibes are not only allopatric and polyphyletic, but also exhibit significant differences in morphology and reproductive mode, indicating that the Southeast Asian species’ are not congeneric with the true Pedostibes of India. As a taxonomic solution, we describe a new genus, Rentapia gen. nov. to accommodate the Southeast Asian species.  相似文献

Phloeocharis Mannerheim is the largest genus within the problematic rove beetle subfamily Phloeocharinae, with a single extinct and 44 recent species recorded from the Holarctic Region. Until now, the oldest fossil record of Phloeocharis was known from Late Cretaceous (Turonian) amber from New Jersey, USA. Here we describe 2Phloeocharis burmana n. sp. from mid-Cretaceous (Albian–Cenomanian) Kachin amber from northern Myanmar, as the earliest extinct species of this genus. Our finding also sheds light on the biogeography of Phloeocharis, since no recent or extinct species have so far been recorded from the Oriental Region. Furthermore, the discovery of 2P. burmana n. sp. extends the Mesozoic diversity of the phloeocharine rove beetles both taxonomically and morphologically, particularly from Kachin amber.  相似文献

Taxonomy of the genus Leveillula has long been considered as a challenge in powdery mildew systematics. The rDNA diversity has recently been used for phylogenetic analysis of several specimens of the genus Leveillula. In the present study, additional rDNA ITS sequences are provided and a new phylogenetic analysis is carried out aiming at a better understanding of the genetic diversity in the genus Leveillula. New analyses confirmed that L. taurica is unique in the genus, as it exhibits an intraspecific gene sequence diversity considerably higher than in other species. In several cases L. taurica s. lat. on a certain host plant species has a sequence different from L. taurica on other host plants. Moreover, DNA data indicated different lineages among L. taurica specimens which were hardly distinguishable by morphology. More than one genotype occurring on a single host is sometimes possible. According to these results, several races such as Leveillula on Artemisia, Acroptilon, Onobrychis, are molecularly well characterized. While there is enough molecular evidence to delimit such races as independent taxa, clear morphological delimitations between these new and already published taxa are very difficult or even impossible. However, ecological features, and above all, host specificity for biotrophic fungi such as powdery mildew, would be a good criterion to discriminate cryptic taxa along with rDNA sequences. In fact, many collections of Leveillula strains on different hosts show their own type of conidial morphology, which is usually consistent for a strain on a single host species. Hence, we have proposed to describe new species for Leveillula on some host plants such as Artemisia, Acroptilon, Echinops and Onobrychis.  相似文献

Artemisia L. is a complex genus of medicinal importance. Publicly available chloroplast genomes of few Artemisia species are insufficient to resolve taxonomic discrepancies at species level. We report chloroplast genome sequences of two further Artemisia species: A. maritima (151,061 bp) and A. absinthium (151,193 bp). Both genomes possess typical quadripartite structure comprising of a large single copy, a small single copy and a pair of long inverted repeats. The two genomes exhibited high similarities in genome sizes, gene synteny, GC content, synonymous and non-synonymous substitutions, codon usage, amino acids frequencies, RNA editing sites, microsatellites, and oligonucleotide repeats. Transition to transversion ratio was <1. Maximum likelihood tree showed Artemisia a monophyletic genus, sister to genus Chrysanthemum. We also identified 20 highly polymorphic regions including rpoC2-rps2, trnR-UCU-trnG-UCC, rps18-rpl20, and trnL-UAG-rpl32 that could be used to develop authentic and cost-effective markers to resolve taxonomic discrepancies and infer phylogenetic relationships among Artemisia species.  相似文献

We describe a new nematode species, Baylisascaris potosis n. sp., isolated from captive kinkajou, Potos flavus, from the Cooperative Republic of Guyana. The nematode was found in fecal specimens, identified morphologically, and confirmed genetically. The new species is similar to Baylisascaris procyonis, Baylisascaris columnaris, and other Baylisascaris species, but is distinguished by the position of the male phasmidial pole. Nuclear and mitochondrial DNA sequence analyses confirmed that the new species is phylogenetically distinct from all the members of the genus Baylisascaris, and groups with B. procyonis and B. columnaris. This nematode is the 10th species assigned to the genus Baylisascaris.  相似文献
