1. Corticotropin-stimulated lipolysis in adipocytes of rats, mice, hamsters, guinea pigs and rabbits. Melanotropins elicited high lipolytic activity only in guinea pig and rabbit adipocytes. Opiate peptides were active only in rabbit adipocytes. Pituitary and chorionic gonadotropins and somatotropin were lipolytic in guinea pig adipocytes. Other hormones tested including prolactin, somatostatin, substance P, neurotensin, angiotensin II, thyrotropin releasing hormone and pancreatic polypeptide were devoid of lipolytic activity in all of the adipocytes studied. 2. In the rabbit adipocytes gamma-melanotropin was lipolytic only at high doses. At these doses the peptide inhibited the lipolytic response to a high dose of corticotropin. 3. Lipolysis stimulated by vasoactive intestinal peptide and epinephrine in rat adipocytes was antagonized by insulin. The lipolytic hormones corticotropin, epinephrine, vasoactive intestinal peptide and secretin suppressed basal and insulin-stimulated lipogenesis.  相似文献

New Zealand White rabbits were injected subcutaneously with 20 mg/kg body weight of diethylnitrosamine (DEN), twice per week, starting when they were 1 week old. The animals were sacrificed 6 to 12 months after the first injection and lung tissues were processed for light microscopy. Using serotonin (5HT) and neuron specific enolase (NSE) as markers for the endocrine cells, tissue sections were stained immunocytochemically by the avidin-biotin complex method. Numerous neuroepithelial bodies (NEBs) positive for 5HT, but negative for NSE, were seen in the alveolar duct regions of DEN-treated rabbits. On the other hand, an increased number of solitary endocrine cells immunoreactive for NSE was found in bronchial or bronchiolar epithelia. The results indicate that DEN induced increases in two distinct types of endocrine cells: the component cells of NEBs are positive for 5HT and solitary cells are positive for NSE.  相似文献

The survival, growth, and egg-laying capacity of young adult Angiostrongylus cantonensis, surgically transferred from intracranial sites into pulmonary arteries, were studied. A variety of experimental animals (rats, guinea pigs, mice, and mastomys) were chosen as donor animals and as recipient hosts (rats, guinea pigs, and rabbits). These species were specifically chosen to span the spectrum of host permissiveness relative to worm development in an attempt to understand the mechanisms which underlie species-dependent resistance. Recipient animals were monitored not only for the development of parasites per se but also for antibody production and histopathologic changes. The results indicated that these procedures were technically feasible, with good worm development following intra-rat transfers, as early as 15 days after initial exposure. Studies were performed to analyze the constraints of development both on initial, i.e., prelung and subsequent i.e., postlung development. When worms were obtained from permissive species such as rat or mastomys, transfer into rats resulted in good growth and development; however, worms which developed initially in exposed mice or guinea pigs developed less well in the rat. Conversely, worms which developed initially in permissive host such as the rat, when transferred into a variety of less permissive hosts such as the guinea pig and rabbit, apparently did not survive and caused significant morbidity and mortality within the nonpermissive host. Histopathologic evaluation revealed a strong eosinophilic perivascular and peribronchiolar infiltration as well as granulomatous reactions surrounding the worms in the lungs of recipient guinea pigs and rabbits, changes not observed in the lungs of permissive rat recipients. As reaginic antibody responses were also more prominent in nonpermissive than in permissive animals, it is possible that IgE responses may be more directly related to the phenomenon of morbidity and/or permissiveness than are other aspects of immune response. In support of this contention was the finding of nearly equivalent hemagglutinating antibody production between permissive rats and nonpermissive guinea pigs and rabbits.  相似文献

Pneumocystis carinii is the most important opportunistic pathogen of humans in the world. Pneumocystis carinii is experimentally detected in the lungs of rats, mice, rabbits, and monkeys, however, the organisms from different mammals are identical in microscopic morphology. The present study tried to find out more mammalian hosts of P. carinii and also to differentiate the organisms from different mammals by karyotyping. Rats, mice, hamsters, rabbits, cats, and dogs were successfully infected by P. carinii, but guinea pigs and pigs were not. Karyotype of P. carinii from rabbits showed similar size range of chromosomes with that of the prototype, but in different pattern. The patterns from cats and dogs were also different from that of rats. The present study confirms that cats and dogs are infected by P. carinii and at least total three karyotype strains of P. carinii are proven in Korea.  相似文献

Endocrine cells (apudocytes) and neuroepithelial bodies (NEBs) were revealed in the lungs of rats of different ages by Grimelius' argyrophilic method. Solitary apudocytes were found among the bronchial epitheliocytes, they had the oval, columnar or triangular shape. NEBs comprise groups of argyrophilic cells, in some cases the penetration of neural fibers into the bodies and their branching in the terminals are observed. Apudocytes and NEBs in 1-, 7-, and 15-day-old rats are more numerous in the epithelium of small than big bronchi. In 21- and 30-day-old and in adult animals apudocytes and NEBs are very scarce. The elements under study are likely to take part in the early postnatal development of the rat lungs.  相似文献

The quantity of histamine and the number of mast cells in the skin of the pika were measured and compared with rabbits, guinea pigs and rats. The ranking of regional histamine levels in the skin of the pika was: perianal region greater than abdomen greater than interscapular region = back greater than lumbus greater than head greater than auricle, and the average value of the 7 regions was 22.6 micrograms/g. The level of histamine in the 6 regions, except the auricle, was 2-5 times that of rabbits and guinea pigs. In the auricle of each of the 4 kinds of animal (pika, rabbit, guinea pig and rat), the levels were almost identical. With respect to histamine levels, those in the pika resembled those in rats. The number of mast cells in the skin of the pika was less than in rats, and was greater than that in rabbits and guinea pigs. The average value was 9.9/mm2.  相似文献

Ultrastructural changes in certain types of adenocytes of old rats, guinea pigs, rabbits and dogs are characterized by specific peculiarities. However, the direction of these changes mainly coincides. The somatotropic function of rats, guinea pigs and rabbits is suppressed, lactotrophic and corticotrophic cells being activated with aging. The phenomenon of the overstrain of gonadotrophic and thyrotrophic cells is registered. Dystrophic changes in the peripheral glands are observed.  相似文献

Tracheal mucous transport was measured using similar techniques in several species. One- to 10-microliter quantities of 99mTc-macroaggregated albumin (99mTc-MAA) were instilled via oral intubation in the distal trachea of rats, rabbits, and dogs. Tracheostomies were used for the instillation in guinea pigs. All animals were anesthetized with halothane for the instillation and allowed to recover immediately in restrainers. Clearance of the 99mTc-MAA in rats and guinea pigs was measured by a slit-collimated NaI scanner. In rabbits and dogs a series of gamma-camera scintiphotos were taken. Clearance was faster and more efficient in dogs than in the other species. No significant differences existed among the rats, rabbits, and guinea pigs in the percentages of the originally deposited material remaining at the instillation site after 1 h (P greater than 0.2). Mean values and standard deviations were 83 +/- 23%, 81 +/- 22% and 70 +/- 20% for rats, guinea pigs, and rabbits, respectively. However, in the dogs a mean of 14 +/- 12% remained at the original site of deposition after only 25 min indicating much more rapid clearance. Mean leading-edge velocities were 9.8 +/- 2.1 (SD) for dogs, 3.2 +/- 1.1 for rabbits, 2.7 +/- 1.4 for guinea pigs, and 1.9 +/- 0.7 mm/min for rats. Clearance patterns qualitatively among the species. In dogs the material moved as a few discrete boluses, whereas in the other species the activity spread toward the larynx. The relatively slow mucous transport of rats, rabbits, and guinea pigs could have important implications in inhalation toxicological studies.  相似文献

Our present understanding of the morphology of neuroepithelial bodies (NEBs) in mammalian lungs is comprehensive. Several hypotheses have been put forward regarding their function but none has been proven conclusively. Microscopic data on the innervation that appears to affect the reaction of NEBs to stimuli have given rise to conflicting interpretations. The aim of this study has been to check the validity of the hypothesis that pulmonary NEBs receive an extensive vagal sensory innervation. The fluorescent neuronal tracer DiI was injected into the vagal sensory nodose ganglion and NEBs were visualized in toto by using immunocytochemistry and confocal microscopy on 100-μm-thick frozen sections of the lungs of adult rats. The most striking finding was the extensive intraepithelial terminal arborizations of DiI-labelled vagal afferents in intrapulmonary airways, apparently always co-appearing with calcitonin gene-related peptide (CGRP)-immunoreactive NEBs. Not all NEBs received a traced nerve fibre. Intrapulmonary CGRP-containing nerve fibres, including those innervating NEBs, always appeared to belong to a nerve fibre population different from the DiI-traced fibres and hence did not arise from the nodose ganglion. Therefore, at least some of the pulmonary NEBs in adult rats are supplied with sensory nerve fibres that originate from the vagal nodose ganglion and form beaded ramifications between the NEB cells, thus providing support for the hypothesis of a receptor function for NEBs. We report here quantitative data on the Feyrter (single) cells (APUD cells) and neuroepithelial bodies (grouped Feyrter cells), in the lungs of rabbit fetuses at 26, 27.5 and 29 days gestational age, during normoxia and short term chronic hypoxia. The apparent number of these cells declines during this period; we suggest that this might be due to increased hypoxemia. Moreover, the number of cells in the lungs of fetuses from short term chronically hypoxic mothers is lower than in the normoxic animals. These findings are in agreement with our previous studies in short term chronically hypoxic neonatal rabbits, and suggest that the increased hypoxemia in the fetus, caused by the induction of hypoxia in the mother, constitutes a stimulus for secretory activity of the Feyrter cells and neuroepithelial bodies (NEBs). This in turn could be part of the mechanism responsible for maintaining the pulmonary vasoconstriction due to hypoxemia. Our results from fetuses of normoxic does provide base line data on the chronological importance of the Feyrter cells and NEBs.  相似文献

Intact, 14- to 21-day fetal rat lung pairs, neonatal lungs, and cultured 15- and 16-day lung explants were examined in 2-micron-thick glycol methacrylate sections stained by PAS-lead hematoxylin. Selected stages were also studied in histochemical preparations for aliesterase and formaldehyde-induced monoamine fluorescence, as well as by scanning and transmission electron microscopy. Neuroepithelial bodies (NEBs) first appear in pseudoglandular lungs at 15 days in vivo as pyramidal groups of basal, diffusely lead-hematoxylin-positive cells in glycogen-depleted epithelium of main and lobar bronchi. By day 16, primitive NEBs occur within three to four generations of the terminal buds, and older, proximal bodies are larger and more distinctive than at 15 days. Aliesterase activity is first detected in basally located, developing NEBs on day 16. During the canalicular and alveolar sac periods, NEBs appear and mature on a proximal-to-distal gradient along the airway, as they do in developing rabbit and human lungs. As earlier-formed airways elongate, additional NEBs appear and supplement the population already present. By days 20-21, NEBs occur at all airway levels down to the bronchiolo-alveolar junctions, and many of the cells have discrete PAS- and lead-hematoxylin-positive, infranuclear granules. Near term some NEBs exhibit serotonin fluorescence after incubation in 5-hydroxytryptophan and have abundant, ca. 100-nm, electron-dense granules. These are concentrated toward the cell base like the stained granules visualized by light microscopy. Similar results were obtained from lungs placed in organ culture. From 2 days in culture to a time equivalent to term, NEB formation parallels that in vivo, indicating that developmental requirements are met in in vitro. Taken altogether, morphologic and cytochemical evidence suggests that NEBs of rats are functional in late fetal life and that their development is relatively independent of extrapulmonary influences and of the intraepithelial ingrowth of sensory nerve endings.  相似文献

In the gastric mucous membrane of 40 rats representing 4 age groups (pubertal, mature, old and very old age) per 10 animals in each, thickness of the mucous membrane has been measured by the argyrophil method of Grimelius and the argentaffin method of Masson - Hamperl and the number of the endocrine cells has been counted. The thickness of the mucous membrane and the length of its glands, as well as the number of argyrophil cells increase from the pubertal towards the mature age. From the mature up to the old age the number of argyrophil cells rises in the antral part, and in the acid-producing zone it significantly decreases. The number of argentaffin cells in the stomach does not change from the pubertal towards the old age. In the very old age, against the background of atrophy of gastric glands, the number of both argyrophil and argentaffin cells decreases in both parts of the stomach studied. The data obtained are discussed taking into consideration functional role of various types of endocrine cells, producing biologically active peptides and monoamines.  相似文献

Ethynyloestradiol was administered to rabbits, guinea pigs and rats, and the concentration of the steroid in blood was measured by radioimmunoassay. In both rabbits and guinea pigs, levels of conjugated steroid were much higher than those of the freely extractable form. Whereas considerable amounts of steroid were present in a congugated form in plasma 24 h after injection, none was present at this time in a freely extractable form. There were significant differences between young and adult rabbits and guinea pigs in the rate at which ethynyloestradiol was metabolized. The amounts present in the freely extractable form in rats were higher than in the other two species but no steroid was detected in the conjugated fraction. The results are compared with previous findings in humans.  相似文献

Rhodanese levels have been measured in liver, kidney and plasma from a number of species. Liver activity was low in marmosets, pigeons and beagle bitches. Levels were high in rats and somewhat lower in hamsters and guinea pigs while levels in two strains of rabbits were intermediate between guinea pigs and marmosets. The relationship between hepatic and plasma rhodanese and cyanide sensitivity is discussed.  相似文献

As best characterized for rats, it is clear that pulmonary neuroepithelial bodies (NEBs) are contacted by a plethora of nerve fiber populations, suggesting that they represent an extensive group of multifunctional intraepithelial airway receptors. Because of the importance of genetically modified mice for functional studies, and the current lack of data, the main aim of the present study was to achieve a detailed analysis of the origin and neurochemical properties of nerve terminals associated with NEBs in mouse lungs. Antibodies against known selective markers for sensory and motor nerve terminals in rat lungs were used on lungs from control and vagotomized mice of two different strains, i.e., Swiss and C57-Bl6. NEB cells were visualized by antibodies against either the general neuroendocrine marker protein gene-product 9.5 (PGP9.5) or calcitonin gene-related peptide (CGRP). Thorough immunohistochemical examination of NEB cells showed that some of these NEB cells also exhibit calbindin D-28 k (CB) and vesicular acetylcholine transporter (VAChT) immunoreactivity (IR). Mouse pulmonary NEBs were found to receive intraepithelial nerve terminals of at least two different populations of myelinated vagal afferents: (1) Immunoreactive (ir) for vesicular glutamate transporters (VGLUTs) and CB; (2) expressing P2X2 and P2X3 ATP receptors. CGRP IR was seen in varicose vagal nerve fibers and in delicate non-vagal fibers, both in close proximity to NEBs. VAChT immunostaining showed very weak IR in the NEB-related intraepithelial vagal sensory nerve terminals. nNOS- or VIP-ir nerve terminals could be observed at the base of pulmonary NEBs. While a single NEB can be contacted by multiple nerve fiber populations, it was clear that none of the so far characterized nerve fiber populations contacts all pulmonary NEBs. The present study revealed that mouse lungs harbor several populations of nerve terminals that may selectively contact NEBs. Although at present the physiological significance of the innervation pattern of NEBs remains enigmatic, it is likely that NEBs are receptor–effector end-organs that may host complex and/or multiple functional properties in normal airways. The neurochemical information on the innervation of NEBs in mouse lungs gathered in the present study will be essential for the interpretation of upcoming functional data and for the study of transgenic mice.  相似文献

Beta3-adrenergic agonists are well-recognited to promote lipid mobilisation and adipose tissue remodeling in rodents, leading to multilocular fat cells enriched in mitochondria. However, effects of beta3-adrenergic agonists on glucose transport are still controversial. In this work, we studied in white adipose tissue (WAT) the influence of sustained beta3-adrenergic stimulation on the glucose transport and on the mitochondrial monoamine oxidase (MAO) activity. As one-week administration of CL 316243 (CL, 1 mg/kg/d) induces beta-adrenergic desensitization in rat but not in guinea pig adipocytes, attention was paid to compare these models. When expressing glucose uptake as nmoles of 2-deoxyglucose/100 mg cell lipids, maximally stimulated uptake was increased in adipocytes of WAT from treated rats but not from treated guinea pigs. However, basal hexose uptake was also increased in CL-treated rats and, as a consequence, the dose-dependent curves for insulin stimulation were similar in control and CL-treated rats when expressed as fold increase over basal. Insulin-induced lipogenesis was unchanged in rat or guinea pig adipocytes after CL-treatment. The glucose carriers GLUT4 and corresponding mRNA were increased in subcutaneous WAT or in brown adipose tissue (BAT) but not in visceral WAT or muscles of CL-treated rats. There was an increase of MAO activity in WAT and BAT, but not in liver, of CL-treated rats while no change was detected in guinea pigs. These findings show that only rat adipocytes, which are beta3-adrenergic-responsive, respond to chronic beta3-AR agonist by an increase of GLUT4 content and MAO activity, despite a desensitization of all beta-adrenoceptor subtypes.  相似文献

alpha 1-Adrenergic activation stimulated phosphorylase and phosphoinositide turnover in hepatocytes from guinea pigs, rats and rabbits. Chlorethylclonidine inhibited these effects in rat and rabbit cells but not in guinea pig hepatocytes; low concentrations of 5-methyl urapidil blocked the alpha 1 actions in guinea pig and rabbit liver cells, but not in rat hepatocytes. Binding competition experiments also showed high affinity for 5-methyl urapidil in liver membranes from guinea pigs and rabbits and low affinity in those from rats. The data indicated that guinea pig hepatocytes express alpha 1A-, rat hepatocytes alpha 1B- and rabbit hepatocytes alpha 1C- adrenoceptors. This was confirmed by Northern analysis using receptor subtype-selective probes.  相似文献

Effectiveness of two different samples of PPD tuberculins was studied quantitatively. No differences were found in the effect on skin test in rabbits, extent of skin reaction in guinea pigs, edema of foot-pad in rats and inhibition of spleen cell migration in guinea pigs. Differences were observed in the systemic febrile reaction in rabbits and in examinations of the thickness of infiltrated skin in guinea pig tests. The results may serve as a basis for standardization of systemic reaction in rabbits.  相似文献

Alveolar macrophages (AM) were collected by repeated endobronchial lavage from mice, rats, guinea pigs, and rabbits, and titrated into cultures of mitogen-stimulated syngeneic or autochthonous lymphocytes. Significant species differences were detected in regard to AM activity in the cultures. AM from guinea pigs and mice stimulated PHA-induced lymphoproliferation, while those from rats and rabbits were inhibitory; blood or peritoneal macrophages were not inhibitory in any of the species examined.  相似文献
