STUDIES ON THE PATHWAY OF SULFIDE PRODUCTION IN A COPPER-ADAPTED YEAST   总被引:1,自引:0,他引:1  
Metabolism of some sulfur-containing substances was studiedin a copper-resistant strain of yeast (R), its parent strain(P) and respiratory-deficient(RD) mutants from them. The resultsobtained are as follows:
  1. Using sulfate, sulfite and thiosulfate as sulfur sources, Rproducedmore H2S than P, and both of these had the activityhigher than their RD mutants. All of them produced a large amountof H2S from cysteine, but only little from methionine, cysteinesulfinic acid and S-sulfocysteine.
  2. From sulfite and thiosulfate,P and R produced more H2S inaerobicthan in anaerobic condition.With sulfate and cysteine, however,H2S production did not differunder those conditions.
  3. In both P and R, the sulfate-to-sulfiteand sulfite-to-sulfidereactions were remarkably lowered byiron and zinc deficiencies.But the cysteine-to-sulfide reactionwas not affected by themetal-deficiencies.
  4. H2S productionfrom sulfate was remarkably depressed by highconcentrationsof pantothenate.
  5. Rates of reaction steps on a plausible pathway from sulfatetosulfide and to organic sulfur compounds areestimated forthe strainsused. R is characterized by its largecapacity ofthe reaction step from sulfate to sulfite, and excessivesulfitethus formed is liberatedas sulfide not by the way ofcysteine.

A cane molasses-based medium for the biomass production of biocontrol agent Rhodosporidium paludigenum was statistically optimized. Molasses concentration (after pretreatment), yeast extract, and initial pH were identified by the Plackett–Burman design to show significant influence on the biomass production. The three factors were further optimized by central composite design and response-surface methodology. The statistical analysis indicated the optimum values of the variables were 89.98 g/L for cane molasses, 2.35 g/L for yeast extract and an initial pH of 8.48. The biomass yield at the optimal culture achieved 15.89 g/L in flask fermentation, which was 2.1 times higher than that at the initial NYDB medium. In a 10-L fermenter, 18.97 g/L of biomass was obtained after 36 hr of cultivation. Moreover, the biocontrol efficacy of the yeast was investigated after culture optimization. The results showed the yeast harvested in the optimal medium maintained its initial biocontrol properties by reducing the percentage of decayed apples to below 20%.  相似文献

为考察将STA基因导入酿酒酵母的可能性和研究糖化酵母在不同核倍性下STA基因的表达效应,从酿酒酵母HU-TY-1A及糖化酵母5101-7C、5206-1B、5301-14D出发,通过原生质体电融合技术,构建了11株核倍性分别为2n、4n、5n及8n的酵母多倍体.所用电融合参数为:交变电场强度200v/cm,频率1MHz;电脉冲强度9kv/cm,宽度40μsec,脉冲数2个,间隔10sec,波形为方波.在糖化酵母单一亲株融合组合中,融合株检出采用菌落形态作为初检指标取得一定效果.其它融合组合则采用遗传标记检出更为可靠.所得融合率约为5×10~(-5)—1×10~(-3).所有融合株经15代以上传代培养,遗传稳定.  相似文献

Activity of hydrogen sulfide production from sulfite was studiedusing a copper-resistant yeast strain (R), its parent strain(P), and the culture of R in the medium without copper addition(R(0)). More hydrogen sulfide was produced under aerobic conditionthan under anaerobic condition. Sulfide producing activity wasin the order of R(0)>P>R under either condition. Stationaryphase cells produced more sulfide than logarithmic phase cellswhen cultured without copper, while the reverse was the casewith R, cultured in copper medium. Sulfide production was inhibitedby high concentrations of sulfite and by salicylaldoxime. 探讨酵母菌杀菌系统(即30株杀菌酵母菌)对38株真菌或细菌的杀菌活性。结果显示杀菌酵母菌不仅对念珠菌、新型隐球菌和烟曲霉菌等真菌有杀菌活性,而且对其不相关的葡萄球菌、肠杆菌科、绿脓杆菌、不动杆菌和副溶血性弧菌也有杀菌活性,提示该系统具有广泛的杀菌活性,特别是K36。  相似文献

1. Acid-inactivated yeast invertase could not be regenerated in the presence of the proteolytic enzymes trypsin, pepsin, and chymotrypsin. 2. Certain foreign proteins of non-enzymatic nature partially inhibited the reactivation of acid-inactivated invertase. 3. Certain proteins as gelatin, lacto-globulin, and carbohydrate-free horse crystalbumin did not prevent the reactivation of invertase at all. 4. Highly purified reactivated invertase was shown to exhibit an effect typical of original native invertase; that is, acceleration of its activity in presence of foreign protein at pH 3.0. 5. Native invertase was not digested by trypsin and chymotrypsin. 6. The addition of trypsin and chymotrypsin to reactivating invertase did not affect the invertase which had already reverted to the active form, but prevented further reactivation of inactive invertase.  相似文献
