During germination of Citrullus lanatus, pyrophosphate-dependent phosphofructokinase (PFP) activity is induced. The peak of PFP activity coincides with the maximum gluconeogenic flux and high fructose-2,6-bisphosphate (Fru-2,6-P2) concentrations. Determination of cytosolic fructose-1,6 bisphosphatase (FBPase) activity in crude extracts is unreliable because of the high PFP activity. The FBPase activity, after correction for the contaminating PFP, is only one-third of the PFP activity. Purified cytosolic FBPase is inhibited by Fru-2,6-P2. The low cytosolic FBPase activity and high Fru-2,6-P2 most probably result in inadequate in vivo activity to catalyze the observed gluconeogenic flux. The total PFP activity is sufficient to catalyze the required carbon flux.  相似文献

The activity of transglutaminase (TG) was examined in the rat superior cervical ganglion (SCG) during development and after postganglionic nerve crush. During postnatal development the enzyme activity is increased by sevenfold in parallel to protein content of the ganglion and reaches adult levels by day 35 after birth. The endogenous activity (enzyme activity assayed in the absence of the exogenous substrate) during development is transiently elevated with a peak at day 21 postnatal. In the adult ganglion the enzyme specific activity is evenly distributed in all subcellular compartments, but most of it is contained in the cytosol. Within the first hour after axotomy TG activity is rapidly and transiently elevated. The peak value, 80% above control levels, is attained by 30 min postoperative. At this time the activity is increased in all subcellular fractions, but the endogenous activity is selectively increased in the fraction containing nuclei. The enhanced TG activity after axotomy can be prevented by topical treatments with verapamil, an inhibitor of voltage-dependent calcium fluxes across excitable membranes, or with the calcium chelator EGTA. The results show that intracellular TG activity is present in the SCG and that it increases with postnatal growth of the ganglion. After axotomy the enzyme activity is rapidly and transiently increased in the ganglion and this elevation critically depends on calcium fluxes.  相似文献

研究了不同肥力水平的宫川蜜柑根际土壤酶的活性及其与土壤农化特性的关系。结果表明 :高产园的土壤酶活性显著高于低产园的土壤酶活性。经统计分析 ,土壤酶活性与养分含量均呈极显著相关。而且酶的活性在土壤中的分布有一定的规律性。其水平分布是在树冠内半径的 4 /5处至树冠滴水线范围内 ,酶的活性最高 ,由此处向内向外酶的活性逐渐降低 ;其垂直分布是 0~ 2 0 cm土层酶的活性最高 ,随土层的加深而逐渐降低  相似文献

1. Incubation of intact epididymal adipose tissue from fed rats at 37 degrees in an albumin solution at pH7.4 in vitro results in rapid loss of clearing-factor lipase activity until a low activity, stable to prolonged incubation, is attained. The clearing-factor lipase activity of intact tissue from starved rats, which is initially much less than that of tissue from fed rats, is mainly stable to incubation at 37 degrees . 2. Much of the clearing-factor lipase activity of intact epididymal adipose tissue from fed rats is inactivated by collagenase. The enzyme activity of intact tissue from starved rats is not inactivated by collagenase. 3. The clearing-factor lipase activity of fat cells isolated from the epididymal adipose tissue of fed rats is stable to prolonged incubation at 37 degrees . It represents only a small proportion of the total activity of the intact tissue. In starved rats, the isolated fat cells contain a much higher proportion of the activity of the intact tissue. Their activity is also stable at 37 degrees . 4. Incubation of isolated fat cells in a serum-based medium leads to a progressive rise in clearing-factor lipase activity. Actinomycin increases the extent of this rise in activity. No rise in clearing-factor lipase activity occurs when stromal-vascular cells isolated from epididymal adipose tissue are incubated in the medium. 5. The findings indicate that less than 20% of the activity of intact adipose tissue from fed rats is retained when fat cells are isolated from the tissue by collagenase treatment. The activity that is lost could be that which normally functions in the uptake of triglyceride fatty acids by the tissue.  相似文献

The antifreeze glycoproteins (AFGP) of polar fish have the ability to depress the freezing temperature of water approximately 500 times the amount expected based on the number of AFGP molecules in solution; yet AFGP solutions have a purely colligative melting point depression. The difference of solution melting and freezing temperatures is the antifreeze activity of AFGP. One characteristic of AFGP activity that requires further examination is the effect of concentration on antifreeze activity, especially whether the activity saturates at high concentrations or the measured activity increases ad infinitum. This study first surveys the activity of the various antifreeze components from both Pagothenia borchgrevinki and the Arg-containing antifreeze glycoprotein from Eleginus gracilis (EgAF). It was found that all AFGP components examined have a plateau in activity at high concentration, but the actual value of the plateau activity differs between the different length AFGP components and between AFGP and EgAF. While the low molecular weight components of both AFGP and EgAF lose activity at deep supercooling, at high concentration activity is restored. The activity data is then shown to fit a reversible kinetic model of AFGP activity, and the coefficients obtained are used to compare the activity differences between AFGP components and between AFGP and EgAF. The model is also shown to describe the activity of the antifreeze protein of the fish Pseudopleuronectes americanus and the thermal hysteresis protein of the insect, Tenebrio molitor.  相似文献

Diacylglycerol kinase activity is found in both adult and fetal lung. Approximately 27 and 52% of the total activity is found in microsomes and cytosol, respectively. The activity is maximal at pH 7.4. The apparent Km for ATP is 0.11 mM and 0.21 mM for cytosol and microsomes, respectively. The apparent Km for dioleoylglycerol is 0.05 mM for cytosol and 0.14 for microsomes. Maximal activity in cytosol and microsomes is obtained with 2.0 mM dexoycholate. Other detergents cannot substitute for deoxycholate. Phosphatidylglycerol stimulates activity in the absence and in the presence of deoxycholate. Phosphatidylserine also stimulates activity, whereas phosphatidylethanolamine was inactive and phosphatidylcholine inhibited the reaction. Linoleic acid produced inhibition. The general properties of the enzyme were similar for fetal and adult lung. Diacylglycerol kinase from microsomes and cytosol fraction from both fetal and adult lung was most active with dioleoylglycerol and diacylglycerol from egg phosphatidylcholine. Significantly lower activity was obtained with dipalmitoylglycerol. Phosphatidylglycerol did not alter the relative substrate preferences. The activity in microsomes increased with development from 19 days gestation to a maximal activity at 21 days gestation. Maximal activity was about 2-fold higher than the adult. The activity dropped rapidly reaching adult values prior to birth (22 days gestation). The activity in cytosol fractions increased gradually from 19 days gestation, reaching adult values by 22 days gestation.  相似文献

The influence of water activity on microbial and enzymatic processes is reviewed. Methods for measuring and calculating water activity are summarized. The effect of a decreased water activity on product formation with algae, bacteria, yeast and fungi, as well as the role of water activity both in inactivation and preservation of microorganisms, is discussed. The relation between water activity and enzyme stability, enzyme activity and enzyme specificity is discussed. It is concluded that water activity should not be considered to be a sole regulatory parameter in biotechnical processes, but that it deserves greater attention in process design and development than is presently given.  相似文献

The relationship between the early-age activity of Mediterranean fruit flies (medflies) or other fruit flies and their lifespan has not been much studied, in contrast to the connections between lifespan and diet, sexual signaling, and reproduction. The objective of this study is to assess intra-day and day-to-day activity profiles of female Mediterranean fruit flies and their role as biomarker of longevity as well as to explore the relationships between these activity profiles, diet, and age-at-death throughout the lifespan. We use advanced statistical methods from functional data analysis (FDA). Three distinct patterns of activity variations in early-age activity profiles can be distinguished. A low-caloric diet is associated with a delayed activity peak, while a high-caloric diet is linked with an earlier activity peak. We find that age-at-death of individual medflies is connected to their activity profiles in early life. An increased risk of mortality is associated with increased activity in early age, as well as with a higher contrast between daytime and nighttime activity. Conversely, medflies are more likely to have a longer lifespan when they are fed a medium-caloric diet and when their daily activity is more evenly distributed across the early-age span and between daytime and nighttime. The before-death activity profile of medflies displays two characteristic before-death patterns, where one pattern is characterized by slowly declining daily activity and the other by a sudden decline in activity that is followed by death.  相似文献

Properties of autolytic breakdown of rat skeletal muscle proteins in the alkaline pH range have been reported. The activity is almost exclusively localized in the myofibrillar fraction, but is not solubilized with Triton X-100. The activity is affected by the KCI concentration in the reaction mixture. In 0.6 M and the more concentrated KCI solutions, the maximum activity is attained. The optimum pH of the activity is in the range of pH 7.5~9.5, and the optimum temperature is between 47~57°C.

This autolytic activity seems to be different from catheptic activity which shows its optimum pH in the acid pH range. Moreover, though more than half of the catheptic activity of rat skeletal muscle is recovered in the myofibrillar fraction, the catheptic activity in the myofibrillar fraction can be removed from the fraction by the extraction with dilute saline solution containing Triton X-100.  相似文献

The relation between the delta-aminolevulinate-synthase and heme-oxygenase activities and the contents of cytochromes b5 and P-450 in rat liver after phenobarbital and CoCl2 injections was studied. Two hours after a single injection of phenobarbital the delta-aminolevulinate-synthase activity is increased, showing a further rise after 24 hrs. The content of cytochrome b5 is not changed, while that of cytochrome P-450 is increased 24 hrs after the injection. The heme-oxygenase activity remains unaffected thereby. The increase in the enzyme activity and cytochrome P-450 content induced by phenobarbital is eliminated by a preliminary administration of actinomycin D. The administration of CoCl2 is accompanied by a decrease in the delta-aminolevulinate-synthase activity after 2 hrs and its further increase after 24 hrs. The heme-oxygenase activity shows a sharp rise 24 hrs after the injection. The rise in the delta-aminolevulinate-synthase activity induced by CoCl2 is removed by actinomycin D. CoCl2 decreases the content of cytochromes b5 and P-450 24 hrs after the injection. It is assumed that the correlation between the delta-aminolevulinate-synthase activity and cytochrome P-450 content is observed only in the case when the heme-oxygenase activity is not increased. The cytochrome b5 content is independent of the changes in the activity of the key enzyme of heme synthesis and depends to a certain extent on the rate of heme degradation by heme-oxygenase.  相似文献

Summary The annual and diurnal activity cycle for four naturally feeding brown trout separately confined in netting cages on the bed of Windermere is described. All the fish showed a similar annual cycle of maximum activity during May and June, one fish showing a second activity peak during the autumn. The fish also showed a similar diurnal activity rhythm of low activity during the night and increased activity during the day with a pronounced increase at dawn.The possible influence of light and temperature on the fishes activity is briefly discussed.  相似文献

This paper shows that the palmitoyl-CoA hydrolase activity of postheparin serum of the rat is mainly derived from the liver. The identity of this activity with the heparin-releasable hepatic triacylglycerol hydrolase activity is established. The consequences of the different substrate specificities of the hepatic and extrahepatic enzymes for the measurement of the overall postheparin serum lipase activity are discussed. Treatment of the rats with either a corticosteroid or with streptozotocin was found to lower the lipolytic activity from the liver and to enhance the extrahepatic activity. Also in human postheparin serum, palmitoyl-CoA hydrolase activity is shown to behave identical with hepatic triacylglycerol hydrolase activity. The possible function of the liver in the serum triacylglycerol metabolism is discussed in connection with the proposed mechanism for the role of extrahepatic lipoprotein lipase in atherogenesis.  相似文献

Summary The activity of succinic dehydrogenase and malic dehydrogenase was observed histochemically in the gland stomach of rats, and also the relationship between the secretory activity of the gastric gland cells and the process of the TCA cycle in the cells was studied.Histochemically, enzyme activity is plainly visible in the gastric parietal cells but in the gastric chief cells and mucous neck cells.The secretory activity of the cells was promoted by the administration of food, the sub-cutaneous injection of histamine, histidine, acetylcholine or eserin.The activity of succinic dehydrogenase appears to be constant regardless of secretory activity except in a few cases. The activity of malic dehydrogenase increases as secretory activity is promoted. It seems very unlikely that one step in the cycle (the transformation of malic acid into oxalacetic acid) would be accelerated while the other step (the transformation of succinic acid into fumaric acid) is not. This inconsistency of activity may be attributed to the histochemical reaction. Thus the increase of malic dehydrogenase activity is seen as an acceleration of the whole TCA cycle. It is our conclusion, therefore, that the source of energy within the cell, i.e. the TCA cycle, is a process which parallels secretory activity.  相似文献

After herpes simplex virus infection of hamster kidney cells there is an induction of nucleoside phosphotransferase activity which can utilize AMP as phosphate donor. The activity is immunologically specific for the infected cell and is induced concomitantly with the virus-coded pyrimidine deoxynucleoside kinase activity. It was discovered that there is sphingomyelinase activity in the rat liver nuclei. The maximum of enzyme activity is at pH 7.1. The data obtained demonstrated that the main part of sphingomyelinase is located in the nuclear membrane. Comparison of sphingomyelinase activity in cell nuclei, liver and brain homogenates shows high level of enzyme activity in the nuclei. The properties of cytoplasmic aminoacyl-tRNA synthetase and aminoacyl-transferring enzymes in the myocardium were examined and methods for the assay of the activity of these enzyme systems were developed. Aminoacyl-tRNA synthetase activity was measured from the rate of incorporation of 14C-labelled amino acid into aminoacyl-tRNA. Transferase activity was measured from the rate of incorporation of amino[14C]acyl-tRNA into protein in the presence of a standard preparation of hepatic ribosomes. Aminoacyl-tRNA synthetase activity is labile once the heart has been homogenized, whereas transferase activity is stable. The source of energy for synthetase activity is ATP; that for transferase is GTP. Transferase activity was inhibited by puromycin and stimulated by dithiothreitol, whereas synthetase activity was unaffected.  相似文献

The filamentous fungus Fusarium lateritium produces cyanide hydratase when grown in the presence of cyanide. The cyanide hydratase protein produced at a high level in Escherichia coli shows a low but significant nitrilase activity with acetonitrile, propionitrile and benzonitrile. The nitrilase activity is sufficient for growth of the recombinant strain on acetonitrile, propionitrile or benzonitrile as the sole source of nitrogen. The recombinant enzyme shows highest nitrilase activity with benzonitrile. Site-directed mutagenesis of the F. lateritium cyanide hydratase gene indicates that mutations leading to a loss of cyanide hydratase activity also lead to a loss of nitrilase activity. This suggests that the active site for cyanide hydratase and nitrilase activity in the protein is the same. This is the first evidence of cyanide hydratase having nitrilase activity.  相似文献

In the previous work we have found two peaks of catalase activity at acid and neutral pH in partially destroyed bacteria E. coli K12 KS400. The present study indicates that catalase activity with two pH-optimums is sensitive to pH of cultivation medium. The relative catalase activity of frozen-thawed bacteria preparations measured at pH 3.5 increased two-fold and activity measured at pH 7.0 didn't change by shift of medium pH from value 5.5 to 7.0. In analogical preparations of bacteria grown in slightly alkaline media activity with acid maximum was not observed, but activity with neutral maximum rose to 130% in comparison with the intact cells was revealed. Two peaks of activity differed in their sensitivity to bacteria destruction, heating, inhibition by NaN3 and AMT, oxidative stress. The analysis of recent literature information and experimental data leads us to conclude that the activity with neutral pH-optimum consists of two known catalase forms HPI and HPII in E. coli. The ratio of HPI and HPII is 70 and 30%, respectively what was concluded from inhibition of catalase activity with neutral pH-optimum by AMT. Properties of catalase activity with acid pH-optimum didn't corresponding to any known enzyme forms. It is suggested the activity measured at pH 3.5 is results of some unstable activator which acts in acid pH range. It is possible that the described activity with acid pH-optimum is specific for the used E. coli strain. Investigation of another strain of E. coli K12 AB1157 confirmed this idea where the activity peak with acid pH-optimum was not detected.  相似文献
