Extensive experimentation with protargol staining of neurons in celloidin and frozen sections of organs has resulted in the following technic: Fix tissue in 10% aqueous formalin. Cut celloidin sections IS to 25 μ, frozen sections 25 to 40 μ. Place sections for 24 hours in 50% alcohol to which 1% by volume of NH4OH has been added. Transfer the sections directly into a 1% aqueous solution of protargol, containing 0.2 to 0.3 g. of electrolytic copper foil which has been coated with a 0.5% solution of celloidin, and allow to stand for 6 to 8 hours at 37° C. Caution: In this and the succeeding step the sections must not be allowed to come in contact with the copper. From aqueous protargol, place the sections for 24 to 48 hours at 37° C. directly into a pyridinated solution of alcoholic protargol (1.0% aqueous solution protargol, 50 ml.; 95% alcohol, 50 ml.; pyridine, 0.5 to 2.0 ml.), containing 0.2 to 0.3 g. of coated copper. Rinse briefly in 50% alcohol and reduce 10 min. in an alkaline hydroquinone reducer (H3BO3, 1.4 g.; Na2SO3, anhydrous, 2.0 g.; hydroquinone, 0.3 g.; distilled water, 85 cc; acetone, 15 ml.). Wash thoroly in water and tone for 10 min. in 0.2% aqueous gold chloride, acidified with acetic acid. Wash in distilled water and reduce for 1 to 3 min. in 2% aqueous oxalic acid. Quickly rinse in distilled water and treat the sections 3 to 5 min. with 5% aqueous Na2S2O3+5H2O. Wash in water and stain overnight in Einarson's gallocyanin. Wash thoroly in water and place in 5% aqueous phosphotungstic acid for 30 min. From phosphotungstic acid transfer directly to a dilution (stock solution, 20 ml.; distilled water, 30 ml.) of the following stock staining solution: anilin blue, 0.01 g.; fast green FCF, 0.5 g.; orange G, 2.0 g.; distilled water, 92.0 ml.; glacial acetic acid, 8 ml.) and stain for 1 hour. Differentiate with 70% and 95% alcohol; pass the sections thru butyl alcohol and cedar oil; mount.  相似文献

A series of experiments with protargol staining of nerve fibers in mammalian adrenal glands has yielded the following procedure: Fix-1-2 days in a mixture of formamide (Eastman Kodak Company) 10 cc, chloral hydrate 5 g., and 50% ethyl alcohol 90 cc. Wash, dehydrate and embed in paraffin. Cut sections about 15 and mount on slides. Remove the paraffin and run down to distilled water. Mordant 1-2 days in a 1% aqueous solution of thallous (or lead) nitrate at 56-60°C. Wash thru several changes of distilled water and impregnate in 1% aqueous protargol (Winthrop Chemical Company) at 37-40°C. for 1 to 2 days. Rinse quickly in distilled water and differentiate 7-15 seconds in a 0.1% aqueous solution of oxalic acid. Rinse thru several changes of distilled water for a total time of 0.5 to 1.0 rain. Reduce 3-5 rain, in Bodian's reducer: hydroquinone 1 g., sodium sulfite 5 g., distilled water 100 cc. Wash in running water 3-5 min. and tone 5-10 min. in a 0.2% gold chloride solution. Wash 0.5 min. or more and reduce in a 2% oxalic acid solution to which has been added strong formalin, 1 cc. per 100. (Caution. This last reduction is critical and over-reduction can spoil an otherwise good stain; 15-30 seconds usually suffices, and the sections should show only the beginning of darkening to a purplish or gray color.) Wash, fix in hypo, wash, dehydrate and cover.  相似文献

Paraffin sections of nervous tissue, which had been fixed in Hofker's fluid, stained readily with protargol solution without the addition of metallic copper or other activator. Amidolsulfite mixtures reduced the protargol more rapidly and completely than hydroquinone-sulfite. Intensification of the stain could be secured by reducing with 0.5% amidol (or pyrogallol) solution after gold toning. The completeness of staining of unmyelinated fibers of the dorsal roots of cat spinal nerves was checked by estimating the number of fibers in a root and the cells of its associated ganglion. A fiber cell ratio of 1:1 was found hi 4 specimens, indicating within limits of error that all fibers were stained. An improvement of die original Hofker's mixture as a fixative was obtained by using a mixture of formic acid, 5 cc.; trichloracetic acid, 10 g.; n-propyl alcohol, 20 cc.; and n-butyl alcohol, 60 cc. (instead of the acetic, trichloracetic, ethyl alcohol mixture used hi the original formula). The following arbitrary method is suggested. Fix 12 to 24 hours, pass to water thru graded ethyl alcohol, wash several hours, dehydrate and embed in paraffin. Cut, mount, and remove the paraffin, pass to water and impregnate 2 or 3 days at 27 to 30$$C. in a 0.5% aqueous solution of protargol (Winthrop Chemical Co.). Rinse 2 or 3 seconds and reduce with 0.5% amidol (Agfa brand used) in 5% sodium sulfite solution. Wash, tone with 0.1% gold chloride, wash and reduce with 0.5% amidol (no sulfite), wash, dehydrate and cover. The method works well on spinal nerve roots, cerebrum, cerebellum, and spinal cord, and moderately well on nerve trunks including sympathetic nerves. A paraffin section method is described with a yellow-brown-black color range comparable to that of Ranson's pyridine silver block stain. After impregnation with activated protargol and reduction with a fine grain photographic developer, silver nitrate impregnation and reduction are repeated as often as necessary. The procedure is as follows:

Place hydrated sections of tissue fixed in chloral hydrate (25 g. in 100 ml. of 50% alcohol) in 1% aqueous protargol (Winthrop Chemical Co.) containing 5-6 g. metallic copper for 12-24 hours. After rinsing in 2 changes of distilled water, reduce 5 to 10 minutes in: Elon (Eastman Kodak Co.) 0.2 g., Na2SO3, dessicated, 10 g., hydroquinone 0.5 g., sodium borate powder 0.1 g., distilled water 100 ml. Wash thoroly in 4 or 5 changes of distilled water and place in 1% aqueous AgNO3 for 10-20 minutes at 28°-50° C. Rinse in 2 or 3 changes of distilled water and reduce in the elon-hydroquinone solution. After thoroly washing in 4 or 5 changes of distilled water, examine under microscope.

If too pale, treat again in silver nitrate for 10-20 minutes, rinse, reduce 5-10 minutes and wash thoroly until nerve fibers show distinct microscopic differentiation, then dehydrate, clear and mount.  相似文献

Controlled silver staining of connective tissue fibers and sometimes of these fibers and cells simultaneously can be obtained. 1. Fix in 10% formalin. Embed in paraffin and cut sections as usual, but do not mount them on slides. Deparaffinize and hydrate through xylene, alcohols and distilled water and henceforth treat them the same as frozen sections. Real frozen sections can also be used. 2. Treat with a freshly prepared 1% solution of KMnO4, usually 15-60 sec, sometimes up to 10 min. 3. Wash in distilled water, 5-10 sec. 4. Decolorize in 2% potassium metabisulfite, 10-20 sec. 5. Place in distilled water, 1 min. 6. Sensitize with 2% iron alum, 1 min. 7. Place in distilled water, 1 min. 8. Impregnate in Gomori's silver oxide solution, 2 min. 9. Wash in a 1.5% aqueous solution of pyridine, about 15 sec. 10. Reduce in a mixture containing 0.25% gelatin and 2% formalin 1 min. 11. Repeat steps 7 to 10 once or several times until the connective tissue fibers are completely stained. For cell staining (which may fail) proceed as follows: After the first insufficient staining of the connective tissue fibers, rinse in distilled water, dip for 1 sec in Gomori's solution and reduce immediately in gelatin-formalin without previous washing in pyridined water. This step can be repeated. 12. If the staining is too strong, decolorize as needed in 2% iron alum. 13. Toning in 0.2% gold chloride, 5 min or more, followed by fixation in 5% sodium thiosulfate, 1 min, is optional. Counterstain as desired. 14. Wash in tap water, dehydrate, clear in xylene and mount in balsam. The same technique applied to sections attached to slides gives good results but inferior to that obtained in paraffin sections processed in the loose, unmounted condition.  相似文献

A silver staining method for paraffin sections of material fixed in HgCl2, sat. aq., with 5% acetic acid is as follows. Process the sections through the usual sequence of reagents, and including I-KI in 70% alcohol, thiosulfate (5% aq.), washing and back to 70% alcohol containing 5% of NH4OH (conc. aq.). After 3 minutes in the ammoniated alcohol, wash through tap water and 2 changes of distilled water and silver 5-10 minutes at 25°C. in 15% AgNO3 aq. to which 0.02 ml. of pyridine per 100 ml. has been added. Blot the slide, but not the section and do not rinse. Reduce at 45°C. in 0.1% pyrogallol in 55% alcohol, then rinse in 55% alcohol and wash in water. The remainder of the process consists of gold toning, intensifying in oxalic acid, fixing in 5% Na2S2O3, washing, dehydrating, clearing and covering. When the specimen contains much smooth muscle, the I-KI solution is acidified before use by adding 2 ml. of 1N nitric acid per 100 ml., and the sections treated for 3 minutes instead of the usual 2 minutes. Formalin should not be added to sublimate-acetic, but specimens that do not contain strongly argyrophilic nonneural tissue may be fixed in formalin or, preferably, Bouin's fluid. Sections of tissue after the latter type of fixation will not require the I-KI and thiosulfate but can go from 95% alcohol to the ammoniated alcohol. The advantages of fixing in HgCl2-acetic acid are suppression of the staining of connective tissue and intensifying the staining of nerve fibers.  相似文献

The following procedure for staining Negri bodies in sections is based on methods previously described by MacNeal, by Haynes, and by Richter:

Fixation:
  1. 1. Zenker's solution 4 hours at 37°C or Dominici's 3 hours.
  2. 2. 70% alcohol, 12 to 18 hours at room temperature.
  3. 3. 80% alcohol, about 5 to 6 hours.
  4. 4. 90% alcohol, about 4 to 6 hours.
  5. 5. Absolute alcohol about 16 hours.
  6. 6. Ether and absolute alcohol aa, about 8 hours.
  7. 7. 16 to 24 hours in the following mixture: celloidin 1 g., methyl salycilate 25 cc., abs. alcohol 25 cc., ether 25 cc.
  8. 8. Chloroform and paraffin, 2 to 3 hours.
  9. 10. Paraffin, 1 to 1 1/2 hours.
  10. 11. Embed.


staining:
  1. 1. Cut sections 4 to 5 μ.
  2. 2. Bring section to water and cover with Lugol's iodine for 10 minutes.
  3. 3. Decolorize with a 2% sodium thiosulfate (hypo).
  4. 4. Wash thoroly with water.
  5. 5. Cover with a mixture of equal parts of 0.5% phloxine and 1% eosin Y (National Aniline brand) and leave for 15 minutes.
  6. 6. Wash with water and stain 2 to 5 minutes in 0.1% azure B (National Aniline).
  7. 7. Wash with 96% alcohol and decolorize in a mixture of 2 parts absolute alcohol with 1 part clove oil, ordinarily for not more than 1/2 to 1 minute.
  8. 8. Dehydrate rapidly, clear, and mount in Yucatan Elemi.

The following method of staining bacterial flagella is ecommended for use on smears made from suspensions of 10 to 16-tour agar slant cultures, incubated 30 minutes at 37°C before spreadng on thoroly cleaned and named slides:
  1. Cover with fixative (100 cc. of 1/4 sat. aqu. solution picric acid, with 5 g. tannic acid and 7.5 g. ferrous sulfate).
  2. Wash with tap water, dry and cover with Fontana spirochaete stain; heat to steaming and allow to act for 1 to 2 minutes. Wash in ap water. The stain is prepared as follows: To 25 cc. 2% AgNO3 add dilute ammonia till the precipitate which forms redissolves; then add more AgNO3 till a faint turbidity results. A clear solution is useess.

Axoplasm is selectively impregnated by the following steps: (1) fixation in 10% formalin or in 10% formalin with added sucrose, 15%, and concentrated NH4OH, 1%, for 1-7 days; (2) frozen sections; (3) extraction of the sections in 95% ethyl alcohol, absolute alcohol, xylene, and 95% ethyl alcohol and absolute alcohol, 1 hr each; (4) distilled water, 3 changes of 10 min each; (5) 20% AgNO3 (aq.) at 25°C, 30 min; (6) distilled water, 3 changes of 1-2 sec each; (7) 6.9% K2CO3, 1 hr; (8) water, 3 changes of about 1 min each; (9) 0.2%AuCl3, 2 min; (10) distilled water; (11) 5% Na2S2O3, 2 min; (12) washing, clearing and mounting. This procedure is proposed as a simplified stain for axoplasm, with other tissue components remaining unstained. The few reagents necessary suit this method for histochemical investigation of the mechanism of silver staining.  相似文献

Technics for free-living forms such as Paramecium and for parasitic forms such as the opalinid ciliates are described.

Paramecium: Fix paramecia in hot Schaudinn's fluid containing 5% of glacial acetic acid for 5-15 minutes. (A hot water bath for maintaining the proper temperature of the fixative is described.) Dehydrate up to 83% alcohol. Mount the specimens on albuminized cover glasses. (A table for mounting animals on cover glasses is described.) Apply a thin layer of collodion to the cover glass to prevent the loss of the specimens during the subsequent handling. Pass through descending grades of alcohol to water. Mordant in 4% iron alum for 24 hours. Stain in 0.5% hematoxylin for 24 hours. Destain in saturated aqueous picric acid. Rinse in tap water, expose to ammonia vapor for a second, and then rinse again in tap water. Wash in running water for 1 hour. Dehydrate. Clear, then mount in damar.

Opalinid Ciliates: Make smears on cover glasses and fix them while wet. If the opalinids are to be subsequently stained in hematoxylin, fix in hot Schaudinn's fluid (containing 5% of glacial acetic acid) for 5-15 minutes. Pass through descending grades of alcohol to water. Mordant in iron alum for 24 hours. Stain in hematoxylin for 24 hours. Destain in saturated aqueous picric acid. For Feulgen reaction, fix in a modified weak Flemming's fluid for 1 hour. Wash in running water for 30 minutes. Hydrolyze. Leave 3 hours in fuchsin decolorized with H2SO3 (Feulgen formula). Wash in H2SO3, then in running water for 15 minutes. Dehydrate up to 95% alcohol. Counterstain with fast green FCF for 2 minutes. Dehydrate in absolute alcohol. Clear, then mount in damar.  相似文献

The following procedure is recommended: Fix ces-todes and trematodes (while held flat between glass slides) 0.5-2.0 hr. in the following mixture: formalin, 15; acetic acid (gl.), 5; glycerol, 10; 95% ethyl alcohol, 24; distilled H2O, 46; all proportions by volume. After freeing them from the slides, wash thoroughly in running water and stain immediately thereafter. Stock staining solution: ferric ammonium alum (violet cryst.), 2 g.; distilled H2O (cold) 100 ml.; after solution, add 2 ml. concentrated H2SO4, bring to a boil; add 1 g. coelestin blue B (Nat. Aniline), boil 3-5 min.; cool and add 10 ml. absolute methyl alcohol and 10 ml. glycerol. Dilute 1 vol. with 3 vol. distilled H20 for use. Stain 5-30 min., depending on size of specimens. Wash with 2 changes 0.5 hr. each of distilled H2O, then 50% isopropyl alcohol 12-16 hr., 50% isopropyl alcohol 2 hr., followed by graded isopropyl alcohol for dehydration. Ether: ethyl alcohol (equal parts), 1 hr., is followed by embedding in celloidin in a sheet just thick enough to cover the specimens. Trim embedded specimens and dehydrate with isopropyl alcohol, 80%, 90% and absolute. Clear in beechwood creosote. Mount in balsam with cover glasses that overlap the edges of the celloidin 1-2 mm. While drying at 37°C, refill edges of mount with fresh balsam as needed. When dry, remove excess balsam and ring the edges with ordinary gloss enamel paint.  相似文献

Celloidin sections from formalin-fixed brain and spinal cord of primates are stored in 70% alcohol after cutting, soaked in 2% pyridine in 50% alcohol for 6-8 hr at 37 C, and transferred to 1% concentrated NH4OH in 50% alcohol 15-18 hr at 20-25 C. After washing and flattening, the sections are transferred to 1% silver protein solution containing 30 ml of 0.2 M H3BO3/100 ml. Impregnation is accomplished in 50 ml screw-top jars, 50 mm in diameter, which are filled to a depth of 35 mm, and have 1 gm of copper foil, 0.002 inch thick added. The foil is folded in loose accordion-fashion, pierced and threaded, cleaned in 5% HNO3, rinsed in distilled water, and suspended in the solution just above the sections by fastening the thread to the jar lid. The sections are impregnated for 24 hr at 37 C, rinsed in distilled water, reduced in a solution of 5% Na2SO3 and 1% hydroquinone for 10 min, washed in distilled water and toned in 0.2% gold chloride for 5 min. After rinsing in distilled water, the sections are transferred to 1% oxalic acid for 45-60 sec, washed in distilled water and placed in 5% Na2S2O3 for 5 min. Sections are then washed, dehydrated to 95% alcohol, cleared in terpineol, followed by 3 changes in xylene, and mounted.  相似文献

Developing and established nerve fibers in the retina and in superficial tracts of the brain can be stained and viewed en bloc. The method was developed on chick embryos of 2 days of incubation to several months post-hatching but could be used on other material provided that the objects of interest were within 35 μ of the surface. Procedure: (1) Place the entire eye or head in 50% pyridine for at least 16 hr. (2) Wash well for 5-7 hr with hourly changes of distilled water or with running tap water for 4-6 hr followed by several changes of distilled water. (3) Transfer to 95% ethanol for 16-48 hr. (4) Impregnate with 1.5% AgNO3 for 2 days at 37 C. (5) Submerge in water and, when staining the retina, remove the vitreous body and apply an aqueous solution of 0.25% pyrogallic acid in 1.25% formalin by directing a narrow stream of this reducer against the retina for 2-5 sec. Wash the eye with distilled water 30-60 sec after applying the reducer. When staining the brain, remove the meninges under water, direct the stream of reducer against the brain for 20-30 sec, and rinse the brain immediately after the nerves have stained. (6) Dissect the specimen and make temporary mounts in glycerol; or, dehydrate and clear for resin mounting. The technique stains both mature and growing axons with their growth cones and sometimes their cell bodies. The fiber patterns show best on the surfaces of the retina and brain. The stain works consistently and is suited to the study of both normal and abnormal development.  相似文献

Lesions produced in the cerebral cortex of rats were studied by Nauta's method for degeneration. The brains were perfused with physiological NaCl solution, followed by 10% neutral (CaCO3) formalin. The brains were removed and stored in the formalin for 2 wk to 1 yr. Experimental modifications of the staining method showed that its sensitivity for fine degenerating fibers could he enhanced by the following changes: (a) omitting 0.05% potassium permanganate; (b) replacing the hydroquinone-oxalic acid mixture with 0.1% pyrogallol. Procedure: (1) frozen sections to water; (2) 0.5% phosphomolybdic acid, 45 min; (3) distilled water, 1 min; (4) 0.1% pyrogallol (aq.), 2 min; (5) distilled water, 3 washes of 1 min each; (6) 1.5% silver nitrate (aq.), 30 min; (7) distilled water, 1 min, (8) Laidlaw's ammoniated silver carbonate, 10110 sec; (9) Nauta's reducer, 1-2 min; (10) distilled water, 1 min; (11) 1.0% Na2S2O3, 2 min; (12) distilled water 3 changes, 1 min each; (13) dehydrate, clear, and cover. This method gave equally good results on degenerating axons in both cortex and thalamus.  相似文献

The epoxy resin was removed from semithin (1 μm) sections by immersing them for 30 sec in sodium methoxide (Mayor et al., J. Biophys. Biochem. Cytol., 9: 909-10, 1961) and then processed as follows: (1) left for 1-3 hr at 60 C in a mixture of formalin, 25 ml; glacial acetic acid, 5 ml; CrO3, 3 gm; and distilled water, 75 ml: (2) oxidized 10 min in a 1:1:6 v/v mixture of 2.5% KMnO4, 5% H2SO4 and distilled water: (3) bleached in 1% oxalic acid, and (4) stained for 15 min in aldehyde fuchsin, 0.125% in 70% alcohol, or in a 1% aqueous solution of toluidine blue. Brains of rat with surgical lesions 3-5 days old are fixed in 10% neutralized formalin (excess of CaCO3), 20 μ serial frozen sections cut therefrom and kept in neutralized formalin for an additional 24-48 hr. The sections are soaked in distilled water 12-24 hr, transferred to 50% alcohol containing 0.75 ml of concentrated NH4OH (sp. gr. 0.91) per 100 ml 12-24 hr, placed in distilled water 2-3 hr and then in silver-pyridine solution (AgNO3 3% aq., 20 ml; pyridine, 1 ml) for 48 hr. Test sections are transferred directly to each one of 3 ammoniated silver-solutions, pH 12.8, 13.0 and 13.2, made as follows: To 200 ml of solution 1 (silver nitrate, 6.4 gm; alcohol 96%, 220 ml; NH4OH (sp. gr. 0.91), 28 ml and distilled water, 440 ml) is added respectively 8-12 ml, 12-16 ml and 16-20 ml of solution 2 (2% NaOH) to give the pH desired. The test sections are studied and the optimal ammoniated silver solution chosen. Two baths of ammoniated silver are used, the section placed with continuous agitation into the first bath for 30 sec and the second bath for 60 sec. The sections are then transferred directly into a reducing bath (formalin 10%, 2ml; alcohol 96%, 5 ml; citric acid 1%, 1.5 ml and distilled water, 4.5 ml) for 2 min and from there to 5% Na2S2O3 for 1 min, rinsed in 3 changes of distilled water, dehydrated and mounted.  相似文献

Sections of potato tuber and epidermis from veins on the dorsal side of the sunflower leaf have proved most satisfactory in demonstrating plasmodesma. Kill fresh sections 5 minutes in a solution of 0.75 g. KI, 0.50 g. iodine in 100 cc. H2O. Swell 5 minutes in 10% H2SO4. Mordant 5 minutes in a solution of 1.25 g. KI, 1.00 g. iodine in 100 cc. of 5% H2SO4. Wash in 5% H2SO4 until iodine starts to fade. Stain in a mixture of 0.5% gentian violet solution in 5% H2SO4 made up to a deep green color. Wash in a solution of 30 cc. glycerin, 2 g. ZnCl2, 0.2 g. iodine, and a bit of KI in 60 cc. of water. Brush with a camel's hair brush, wash and mount in the same solution.  相似文献

Cells from monolayer culture of Chinese hamster line Don were treated by Colcemid (0.1 μg/ml) for 2 hr, trypsinized and spun; resuspended in 0.5% sodium citrate solution for 10 min, respun, and then resuspended in a small volume of the supernatant. Slide preparations were made by smearing, followed by air drying for 1 min at room temperature. They were fixed and stained by the following sequence: 2.5% glutaraldehyde in Millonig's buffer, 30 min; distilled water, 6 min, 5 changes; ammoniacal silver at 18-26 C, 10 sec; distilled water, 30 min, 5 changes; 2.5% formalin, 2 min; and distilled water, 3 changes during 15 min. Staining solution: add 225 ml of 5% Na2CO3 to 75 ml of 10% AgNO3, then add concentrated NH4OH slowly, drop by drop, until the solution is transparent. Finally add 300 ml of dstilled water. Cells treated with cold 0.25 N HCl before fixation were not stained. Sequence modifications show that chromatin does not reduce silver by itself. This method stains the sites of high histone concentrations in mitotic chromosomes of cytogenetic preparations.  相似文献

The following procedure stains the atrioventricular conduction system selectively. (1) Wash the fresh heart with physiological saline solution to free it of blood; (2) fix it in 10% formalin containing 0.5% HIO4 for 1 hr; (3) wash in 3 changes of distilled water for 20 min; (4) keep in 80% alcohol for 12 hr to 2 wk; (5) wash with distilled water; (6) treat with a dilute Schiff's reagent containing 0.1 gm of basic fuchsin per 100 ml for 0.5-2 min; (7) rinse in three changes of 2% Na2SO3 in 0.2 N HCI for 3-5 min; (8) wash and examine in 80% alcohol; store in 80% alcohol.  相似文献

Tissues were fixed at 20° C for 1 hr in 1% OsO4, buffered at pH 7.4 with veronal-acetate (Palade's fixative), soaked 5 min in the same buffer without OsO4, then dehydrated in buffer-acetone mixtures of 30, 50, 75 and 90% acetone content, and finally in anhydrous acetone. Infiltration was accomplished through Vestopal-W-acetone mixtures of 1:3, 1:1, 3:1 to undiluted Vestopal. After polymerisation at 60° C for 24 hr, 1-2 μ sections were cut, dried on slides without adhesive, and stained by any of the following methods. (1) Mayer's acid hemalum: Flood the slides with the staining solution and allow to stand at 20°C for 2-3 hr while the water of the solution evaporates; wash in distilled water, 2 min; differentiate in 1% HCl; rinse 1-2 sec in 10% NH,OH. (2) Iron-trioxyhematein (of Hansen): Apply the staining solution as in method 1; wash 3-5 min in 5% acetic acid; restain for 1-12 hr by flooding with a mixture consisting of staining solution, 2 parts, and 1 part of a 1:1 mixture of 2% acetic acid and 2% H2SO4 (observe under microscope for staining intensity); wash 2 min in distilled water and 1 hr in tap water. (3) Iron-hematoxylin (Heidenhain): Mordant 6 hr in 2.5% iron-alum solution; wash 1 min in distilled water; stain in 1% or 0.5% ripened hematoxylin for 3-12 br; differentiate 8 min in 2.5%, and 15 min in 1% iron-alum solution; wash 1 hr in tap water. (4) Aceto-carmine (Schneider): Stain 12-24 hr; wash 0.5-1.0 min in distilled water. (5) Picrofuchsin: Stain 24-48 hr in 1% acid fuchsin dissolved in saturated aqueous picric acid; differentiate for only 1-2 sec in 96% ethanol. (6) Modified Giemsa: Mix 640 ml of a solution of 9.08 gm KH2PO4 in 1000 ml of distilled water and 360 ml of a solution of 11.88 gm Na2HPO4-2H2O in 1000 ml of distilled water. Soak sections in this buffer, 12 hr. Dissolve 1.0 gm of azur I in 125 ml of boiling distilled water; add 0.5 gm of methylene blue; filter and add hot distilled water until a volume of 250 ml is reached (solution “AM”). Dissolve 1.5 gm of eosin, yellowish, in 250 ml of hot distilled water; filter (solution “E”). Mix 1.5 ml of “AM” in 100 ml of buffer with 3 ml of “E” in 100 ml of buffer. Stain 12-24 hr. Differentiate 3 sec in 25 ml methyl benzoate in 75 ml dioxane; 3 sec in 35 ml methyl benzoate in 65 ml acetone; 3 sec in 30 ml acetone in 70 ml methyl benzoate; and 3 sec in 5 ml acetone in 95 ml methyl benzoate. Dehydrated sections may be covered in a neutral synthetic resin (Caedax was used).  相似文献
