The critical shear stress of resuspension and rates of erosion for cohesive and loosely structured sediments must be obtained by direct measurements since there is no theoretical calculation. An in situ experiment on sediment resuspension was performed in a shallow lake (Langer See, NE Germany; area = 1.27 km2, zmax = 3.8 m) in summer 2006 using a hydrodynamically calibrated erosion chamber (Ø 20 cm). Shear velocity (u*) was incrementally increased in 11 steps (0–2.19 cm s?1) to initiate resuspension events. Entrainment rates (E) of suspended particulate matter (ESPM), total P (ETP), chlorophyll a (EChl a), and soluble reactive P (ESRP) were determined by mass balance. Two subsequent critical u* (0.53 cm s?1 and 1.48 cm s?1) support the ‘two-layered bed’ model of a fluffy surface aggregate layer (freshly deposited phytodetritus prone to resuspension) and an underlying more consolidated biostabilised layer. Patterns in ESPM (2–106 g m?2 h?1), ETP (11–532 mg m?2 h?1), and EChl a (3–24 μg m?2 h?1) revealed a sediment surface maximum of TP and Chl a and their theoretical vertical logarithmic decrease within 4 mm sediment depth, the maximum thickness of sediment layer entrained. The advective ESRP flux (17 mg m?2 h?1) was 43 times higher than the diffusive SRP flux (0.4 mg m?2 h?1). The TP and Chl a micro-profiles suggest that cohesive sediment bed formation is a function of both settling (fluff) and consolidation (biostabilisation). Thus, sediment microstructure and resuspension behavior depend on each other.  相似文献

在黄土高原水蚀风蚀交错区坡面(40 m×350 m)尺度上进行网格(10 m×10 m)取样,用经典统计学和地统计学相结合研究了180个土壤剖面(0-200 cm)各土层扰动土饱和导水率(Ks) 的空间异质性及分布格局。结果表明: 0-20 cm土层的Ks值(5.36×10-3 cm/s)最大,>20-200 cm各土层的Ks值均小于表层,其值介于4.32×10-3-4.76×10-3 cm/s之间。各土层Ks的变异程度相近,均属于中等变异。>20-200 cm各土层Ks 的Kriging 插值图分布格局也表现出一致性,因此可用>20-40 cm土层的Ks值来代表深层Ks值对土壤水分运动进行模拟。除了0-20 cm 的Ks的基台值(C +C0)为0.154,其它各土层基台值介于0.202-0.276之间,说明0-20 cm的Ks空间异质性小于>20-200 cm各土层。从比值C/(C+C0)来看, 0-20 cm属于中等自相关,>20-200 cm土层属于强的空间自相关性,同样也验证了黄土高原水蚀风蚀交错区土壤剖面饱和导水率具有空间变异特征。  相似文献

In slow mainstream flows (<4–6 cm · s?1), the transport of dissolved nutrients to seaweed blade surfaces is reduced due to the formation of thicker diffusion boundary layers (DBLs). The blade morphology of Macrocystis pyrifera (L.) C. Agardh varies with the hydrodynamic environment in which it grows; wave‐exposed blades are narrow and thick with small surface corrugations (1 mm tall), whereas wave‐sheltered blades are wider and thinner with large (2–5 cm) edge undulations. Within the surface corrugations of wave‐exposed blades, the DBL thickness, measured using an O2 micro‐optode, ranged from 0.67 to 0.80 mm and did not vary with mainstream velocities between 0.8 and 4.5 cm · s?1. At the corrugation apex, DBL thickness decreased with increasing seawater velocity, from 0.4 mm at 0.8 cm · s?1 to being undetectable at 4.5 cm · s?1. Results show how the wave‐exposed blades trap fluid within the corrugations at their surface. For wave‐sheltered blades at 0.8 cm · s?1, a DBL thickness of 0.73 ± 0.31 mm within the edge undulation was 10‐fold greater than at the undulation apex, while at 2.1 cm · s?1, DBL thicknesses were similar at <0.07 mm. Relative turbulence intensity was measured using an acoustic Doppler velocimeter (ADV), and overall, there was little evidence to support our hypothesis that the edge undulations of wave‐sheltered blades increased turbulence intensity compared to wave‐exposed blades. Pulsed nuclear magnetic resonance studies of the longitudinal (T1) and transverse (T2) quadrupolar relaxation times of 7Li, 23Na, 35Cl ions in the absence and presence of human oxy- and carbon monoxyhaemoglobin have been used to investigate the interaction of the ions and the macromolecule.The relaxation data show that Cl? interacts strongly with the haemoglobin but provide no evidence for binding of Na+ up to concentrations of 0.5 m. In the case of Li+, evidence for interaction is obtained at concentrations of about 0.1 m.The dependence of relaxation rate on frequency was followed over a limited frequency range in an attempt to separate the effects of correlation times and exchange rates of the bonded ions on the relaxation. In the case of Cl?, an upper limit for the mean lifetime divided by the number of sites can be set at about 1 × 10?6 second, and a lower limit at about 1 × 10?8 second.  相似文献

There are different approaches to define the soil available water (SAW) for plants. The objectives of this study are to evaluate the SAW values of 12 arable soils from Hamadan province (western Iran) calculated by plant available water (PAW), least limiting water range (LLWR) and integral water capacity (IWC) approaches and to explore their relations with Dexter’s index of soil physical quality (i.e., S-value). Soil water retention and mechanical resistance were determined on the intact samples which were taken from the 5–10 cm layer. For calculation of LLWR and IWC, the van Genuchten-Mualem model was fitted to the observed soil water retention data. Two matric suctions (h) of 100 and 330 cm were used for the field capacity (FC). There were significant differences (P?<?0.01) between the SAW values calculated by PAW100, PAW330, LLWR100, LLWR330 and IWC. The highest (i.e., 0.210 cm3 cm?3) and the lowest (i.e., 0.129 cm3 cm?3) means of SAW were calculated for the IWC and LLWR330, respectively. The upper limit of LLWR330 for all of the soils was h of 330 cm, and that of LLWR100 (except for one soil that was air-filled porosity of 0.1 cm3 cm?3) was h of 100 cm. The lower limit of LLWR330 and LLWR100 for five soils was h of 15,000 cm and for seven soils was mechanical resistance of 2 MPa. The IWC values were smaller than those of LLWR100 for two soils, equal to those of LLWR100 for three soils and greater than those of LLWR100 for the rest. There is, therefore, a tendency to predict more SAW using the IWC approach than with the LLWR approach. This is due to the chosen critical soil limits and gradual changes of soil limitations vs. water content in the IWC calculation procedure. Significant relationships of SAW with bulk density or relative bulk density were found but not with the clay and organic matter contents. Linear relations between IWC and LLWR100 or LLWR330 were found as: IWC?=??0.0514 + 1.4438LLWR100, R 2?=?0.83; and IWC?=??0.0405 + 2.0465LLWR330, R 2?=?0.84, respectively (both significant at P?<?0.01). Significant relationships were obtained between the SAW values and S indicating the suitability of the index S to explain the availability of soil water for plants even when complicated approaches like IWC are considered. Overall, the results demonstrate the importance of the choice of the approach to be used and its critical limits in the estimation of the soil available water to plants.  相似文献

We estimated the grazing impact of the heterotrophic flagellate Collodictyon triciliatum on the harmful, bloom-forming cyanobacterium Microcystis aeruginosa in an experimental pond during a Microcystis bloom from summer to winter in 2010. For these experiments, we calculated the grazing rates from the digestion rate of C. triciliatum and its food vacuole contents. During the study period, M. aeruginosa exhibited one bloom event with a maximum density of 1.1 × 105 cells ml?1. The cell density of C. triciliatum fluctuated from below the detection limit to 291 cells ml?1. The number of M. aeruginosa cells ingested by C. triciliatum food vacuoles ranged between 0.4 and 10.8 cells flagellate?1, and the digestion rate of C. triciliatum at 25 °C was 0.73 % cell contents min?1. The grazing rate of C. triciliatum on the M. aeruginosa prey was 0.2–6.9 cells flagellate?1 h?1, and its grazing impact was 0.0–25.3 % standing stock day?1. The functional response of C. triciliatum to the M. aeruginosa prey followed the Michaelis–Menten model of significance (r 2 = 0.873, p < 0.001) in our experimental systems, in which the prey concentration varied from 1.0 × 104 to 2.1 × 106 cells ml?1. The maximum grazing rate was 6.2 prey cells grazer?1 h?1, and the half-saturation constant was 1.2 × 105 cells ml?1. We present evidence that C. triciliatum grazing explained the remarkable decrease in M. aeruginosa cell density in the pond. The present study is the first demonstration of the high potential of protistan grazing on M. aeruginosa to reduce cyanobacterial blooms.  相似文献

A weeping pulsed radar reflectometer designed for measuring the spatial electron density distribution in the Globus-M spherical tokamak with a minor plasma radius of a=24 cm, a major radius of R=36 cm, a toroidal field of B T=0.5 T, a plasma current of I p=200 kA, and an average density of n=(3–10)×1013 cm?3 is described. The reflectometer operation is based on the reflection of microwaves with a carrier frequency f from a plasma layer with the critical density n=(0.0111f)2, where n is the electron density in units of 1014 cm?3 and f is the microwave frequency in GHz. By simultaneously probing the plasma at different frequencies, it is possible to recover the electron density profile. Microwave pulses with different frequencies are obtained by frequency sweeping. To increase the range of measured densities, channels with fixed frequencies are also used; as a result, the instrument has eleven frequency channels: a 19.5-GHz channel, eight channels in the 26-to 40-GHz frequency range, a 51.5-GHz channel, and a 60-GHz channel, which corresponds to eleven points in the density profile: 0.47×1013 cm?3, eight points in the (0.8–1.95)×1013-cm?3 range, 3.27×1013 cm?3, and 4.5×1013 cm?3. The reflectometer allows detailed measurements of the density profile with a time resolution of several tens of microseconds, which can be useful, in particular, in studying the processes related to the formation of an internal transport barrier in plasma. The first results obtained using this reflectometer in the Globus-M tokamak under various operating conditions are discussed.  相似文献

Using a liquid chromatography method that separates the two sulfonium diastereoisomers of adenosylmethionine, we have found that immature soybeans, soybean callus culture, radish leaves, yeast and rat liver contain only the (S)-sulfonium form of S-adenosylmethionine. Our findings contradict the suggestion by Stolowitz and Minch that 10–20% of naturally-occurring adenosylmethionine may have the (R)-configuration at the sulfonium pole. Absence of the (R)-sulfonium isomer of adenosylmethionine in biological materials indicates that the (R)-sulfonium form of adenosylmethionine present in commercial adenosylmethionine samples is an artifact of the isolation procedure. Our method of measuring the isomers of adenosylmethionine enabled us to readily determine the rate of racemization and hydrolysis of adenosylmethionine. Our rate constants for racemization (Kr) and hydrolysis (Kh) were 2.4 × 10?6 sec?1 and 12.3 × 10-?6 sec?1, respectively; values which are noticeably different from those of Wu and co-workers which were obtained with a more complicated method (Kr = 8 × 10?1 sec?1; Kh = 6 × 10?6 sec?1). We believe the absence of the (R)-isomer in vivo is best explained by stabilization of the (S)-isomer as suggested by Wu et al. Although the tissues we have analysed contained the (S)-sulfonium form of adenosylmethionine exclusively, when ethionine-resistant soybean cell lines were given ethionine, they accumulated both sulfonium diastereoisomers of adenosylethionine.  相似文献

The heterotrophic planktonic bacteria in two high-arctic lakes were studied by direct microscope count and the enzymatic uptake of 14C labelled glucose which generally conformed to Michaelis-Menten kinetics. Bacterial numbers and activity in oligo-trophic Char Lake ranged from 0.1 to 2.0×10?3 bacteria/l and a maximum uptake velocity (Vmax) of 1.8 × 10?3μg glucose l? h?1. Nearby Meretta Lake received waste water from the Department of Transport Base at Resolute and this eutrophication was reflected in higher bacterial numbers of 2-80 × 108/1 and Kmax of 0.1 × 10?1-7.5 × 10?1 fig glucose l?1 h?1 The Kmax per cell in Char Lake was 3 × 10?11μg glucose l?1 h?1 and changed little between the period of solid ice cover in May and ice-free conditions in August. Bacterial cycles could not be related to phytoplankton cycles in either lake. Comparison of kinetic data from several lakes suggests a relationship between the bacterial uptake rate of glucose and phytoplankton production. Both bacterial numbers and activity in Char Lake may be very close to the minima to be expected in undisturbed freshwater environments.  相似文献

Cuticular membranes (CM) were isolated enzymatically from leaves of amphibious and submerged plants, and the oxygen permeability of aerial and aquatic CMs was compared using a specially constructed oxygen electrode. Their thicknesses were estimated from transmission electron micrographs of intact leaves. When dry CMs were moistened, the permeability of typical aerial CMs changed differently from that of typical aquatic CMs during the desiccation, apparently reflecting different internal structures. The cuticle thickness was mostly < 100 nm, and the range of permeance values was 5–143 × 10?6 m s?1, corresponding to resistance values of 7–210 × 103 s m?1. The resistance of an aerial CM was always higher than that of an aquatic one from the same species. Only the aquatic CMs showed a weak proportionality between resistance and thickness. The relation between resistance and thickness varied from aquatic to aerial CMs in a species specific way.  相似文献

Enzymatic acylation of rutin and esculin with aromatic, aliphatic and aryl-aliphatic acids using Candida antarctica lipase in tert amyl alcohol as solvent was investigated under low water content. Whatever the acyl donor used, the conversion yields and initial rates for esculin were higher than for rutin. For a given flavonoid, the performance of these reactions depended on the acyl donor structures. For aliphatic acids, conversion yields and initial rates of both flavonoids were respectively in the ranges of 68–90% and of 9.5×10?2–72×10?2 mmol l?1 h?1. For aromatic acids, the reaction occurred only with the aryl subgroup (cinnamic, hydrocinnamic, 3,4-dihydroxyhydrocinnamic and 4-hydroxyphenyl acetic acids) and was drastically influenced by the presence of side chain and substitution patterns of the aromatic ring. Except for hydrocinnamic acid (75%, 23.4×10?2 mmol l?1 h?1), with these acids the conversion yields and initial rates were lower and in the range of 10–45% and of 0.7×10?2 to 12.1×10?2 mmol l?1 h?1. Unsaturation of the side chain of the hydrocinnamic acid decreased the esculin conversion rate from 75 to 13% and initial rate from 23.4 to 1.76×10?2 mmol l?1 h?1. The presence of hydroxyl or nitro-groups on the aromatic ring of the aryl aliphatic acid also reduced conversion yields and initial rates. Even without a spacer, the non-phenolic ring acid (quinic acid) was reactive and lead to conversion yields of about 20 and 23% respectively for rutin and esculin.  相似文献

Purified, water-soluble polysaccharide of Phellodendron amurense Ruprecht (P-WSPS) was obtained in highly homogeneous state. The partial specific volume and intrinsic viscosity were, respectively, 0.56 and 11.68 dL/g. The S value and molecular weight depended strongly on the P-WSPS concentration, but extrapolation to infinite dilution gave Sw,200 = 10.67 × 10?13 s?1 and M0 = 625 × 103 from velocity-sedimentation and equilibrium-sedimentation experiments, respectively, results that showed the highly branched structure of P-WSPS. The molecular weight calculated from the D0 value (1.035 × 10?7) and S0 value agreed with that from the equilibrium-sedimentation experiment to within ~±9%, showing that the diffusion method is useful for rapid estimation of molecular weight.  相似文献

The ultimate tensile strength (σUT) and the modulus of elasticity (E) of Rhodnius extensible cuticle are σUT = 2.20 × 107 Nm?2, E = 2.43 × 108 Nm?2 (unplasticised); σUT = 1.43 × 107 Nm?2, E = 9.45 × 106 Nm?2 (plasticised with 5HT) and σUT = 9.05 × 106 Nm, E = 2.46 × 106 Nm?2 (plasticised in pH 5 buffer).The mechanical properties of cuticle from insects which have deposited additional layers of cuticle after they have been fed differ from those of cuticle from unfed insects. This is possibly due to the different composition of the additional cuticle: it is suggested that the post-feeding cuticle is providing protection and a template for the next instars cuticle.The maximum strain of extensible cuticle from starved insects is related to the amount of matrix protein present.  相似文献

Feeding experiments were carried out on the benthic harpacticoid copepod Tisbe cucumariae Humes, using seven different diets of various dried and ground macroalgae and marsh grass, algal Aufwuchs, diatoms, polychaete meat, and cereal. In short-term experiments (1 h), 14C-labelled foods were used to measure ingestion rate of non-ovigerous adult females (individual dry wt = 5.57 ± 2.49 μg). No significant difference was found among the rates at which all foods, except polychaete meat, were ingested (12.7 to 17.3 × 10?2μg dry wt· ind?1· h?1). Polychaete meat was consumed faster (23.9 × 10?2μg dry wt·ind ?1· h?1). The nutritional value of the foods was estimated in long-term experiments (22 days) by measuring development time and survival of T. cucumariae. Both these variables were significantly correlated with the nitrogen, protein content, and C:N ratio of the foods. No relation was found, however, with the amount of carbon, calories and available calories in the diets. Thus, nitrogen (protein) content of the food was the factor limiting secondary production of the copepods.  相似文献

The migratory history of tapertail anchovy Coilia nasus in the Yangtze River Estuary, China was investigated using otolith Sr:Ca ratios and two-dimensional images of the Sr level from an X-ray electron probe microanalyzer (EPMA). The results showed that 17 of the 22 young-of-the-year (YOY) specimens had low Sr:Ca ratios (1.2–2.4?×?10?3;1.5?±?0.3?×?10?3) at the central otolith area, indicating their riverine origin and initial freshwater residence. In addition, 11 of the 14 adult specimens had low Sr:Ca ratios (1.3–2.2?×?10?3; 1.7?±?0.4?×?10?3) at the central otolith area but showed alternating changes between high (>4.0?×?10?3) and low (<2.5?×?10?3) values outside of this region, reflecting their riverine origin and the migration between freshwater and estuarine habitats. These 28 specimens represented the anadromous population in this region. The other 5 YOY specimens had high Sr:Ca ratios (3.6–5.9?×?10?3; 4.8?±?0.8?×?10?3) throughout the life history. Similarly, the other 3 adult specimens had high Sr:Ca ratios (4.0–5.7?×?10?3; 4.8?±?0.7?×?10?3) at the central otolith area but showed alternating changes between low and high values outside this region, suggesting that estuarine-origin non-anadromous individuals occurred in this region. The average of the otolith Sr:Ca ratios and Sr level mapping along the life-history transects could be used as a scalar for charting the migratory history of the tapertail anchovy in the Yangtze River Estuary: <2.0?×?10?3 for freshwater residence and 3.5–6.0?×?10?3 for estuarine residence.  相似文献

All manifestations of cardiovascular disease (CVD) are substantially more common in patients with type 2 diabetes mellitus (T2DM) than in non-diabetic individuals. The current study evaluated KALRN, a gene previously linked to CVD, as a contributor to CVD in a sample enriched for T2DM. Specifically, the potential modifying effect of cigarette smoking was examined. A total of 28 SNPs in KALRN were genotyped in 1001 European Americans from 369 Diabetes Heart Study (DHS) families, as well as 762 population-based controls. The association between each SNP and both qualitative and quantitative CVD disease phenotypes was determined using generalized estimating equations and variance component models, respectively. Selected KALRN SNPs were found to be associated with both the qualitative (T2DM, CVD, metabolic syndrome) and quantitative traits (C-reactive protein and abdominal aortic calcified plaque). Interaction analysis and stratification were then used to test whether smoking modulates the genetic effects of KALRN. The strongest evidence of a modifying effect of smoking status was observed for rs9289231 and intima-media thickness (p=9.0x10?4) and abdominal aortic calcified plaque (p=3.0×10?4). Overall, following stratification by smoking status, the evidence of association with quantitative traits was more pronounced in smokers compared to non-smokers. The strongest association for smokers was between rs1720960 and abdominal aortic calcified plaque (p=2.6x10?5), while in non-smokers there was no observed association. KALRN variants are associated with measures of CVD and T2DM in the DHS sample with smoking status observed to have a significant modifying effect on these associations.  相似文献

The Aminobacter sp. strain MSH1 has potential for pesticide bioremediation because it degrades the herbicide metabolite 2,6-dichlorobenzamide (BAM). Production of the BAM-degrading bacterium using aerobic bioreactor fermentation was investigated. A mineral salt medium limited for carbon and with an element composition similar to the strain was generated. The optimal pH and temperature for strain growth were determined using shaker flasks and verified in bioreactors. Glucose, fructose, and glycerol were suitable carbon sources for MSH1 (μ?=?0.1 h?1); slower growth was observed on succinate and acetic acid (μ?=?0.01 h?1). Standard conditions for growth of the MSH1 strain were defined at pH 7 and 25 °C, with glucose as the carbon source. In bioreactors (1 and 5 L), the specific growth rate of MSH1 increased from μ?=?0.1 h?1 on traditional mineral salt medium to μ?=?0.18 h?1 on the optimized mineral salt medium. The biomass yield under standard conditions was 0.47 g dry weight biomass/g glucose consumed. An investigation of the catabolic capacity of MSH1 cells harvested in exponential and stationary growth phases showed a degradation activity per cell of about 3?×?10?9 μg BAM h?1. Thus, fast, efficient, large-scale production of herbicide-degrading Aminobacter was possible, bringing the use of this bacterium in bioaugmentation field remediation closer to reality.  相似文献

Using existing heat transfer data, a relatively simple expression was developed for estimating the effective thickness of the boundary layer of air surrounding cylinders. For wind velocities from 10 to 1000 cm/second, the calculated boundary-layer thickness agreed with that determined for water vapor diffusion from a moistened cylindrical surface 2 cm in diameter. It correctly predicted the resistance for water vapor movement across the boundary layers adjacent to the (cylindrical) inflorescence stems of Xanthorrhoea australis R. Br. and Scirpus validus Vahl and the leaves of Allium cepa L. The boundary-layer thickness decreased as the turbulence intensity increased. For a turbulence intensity representative of field conditions (0.5) and for νwindd between 200 and 30,000 cm2/second (where νwind is the mean wind velocity and d is the cylinder diameter), the effective boundary-layer thickness in centimeters was equal to [Formula: see text].  相似文献

1. The microbial metabolism of organic matter in rivers has received little study compared with that of small streams. Therefore, we investigated the rate and location of bacterial production in a sixth‐order lowland river (Spree, Germany). To estimate the contribution of various habitats (sediments, epiphyton, and the pelagic zone) to total bacterial production, we quantified the contribution of these habitats to areal production by bacteria. 2. Large areas of the river bottom were characterized by loose and shifting sands of relatively homogenous particle size distribution. Aquatic macrophytes grew on 40% of the river bottom. Leaf areas of 2.8 m2 m?2 river bottom were found in a 6.6 km river stretch. 3. The epiphyton supported a bacterial production of 5–58 ng C cm?2 h?1. Bacterial production in the pelagic zone was 0.9–3.9 μg C L?1 h?1, and abundance was 4.0–7.8 × 109 cells L?1. Bacterial production in the uppermost 2 cm of sediments ranged from 1 to 8 μg C cm?3 h?1, and abundance from 0.84 to 6.7 × 109 cells cm?3. Bacteria were larger and more active in sediments than in the pelagic zone. 4. In spite of relatively low macrophyte abundance, areal production by bacteria in the pelagic zone was only slightly higher than in the epiphyton. Bacterial biomass in the uppermost 2 cm of sediments exceeded pelagic biomass by factors of 6–22, and sedimentary bacterial production was 17–35 times higher than in the overlying water column. 5. On a square meter basis, total bacterial production in the Spree was clearly higher than primary productivity. Thus, the lowland river Spree is a heterotrophic system with benthic processes dominating.
