We affinity-purified the tobacco plastid-encoded plastid RNA polymerase (PEP) complex by the alpha subunit containing a C-terminal 12 x histidine tag using heparin and Ni(2+) chromatography. The composition of the complex was determined by mass spectrometry after separating the proteins of the >900 kDa complex in blue native and SDS polyacrylamide gels. The purified PEP contained the core alpha, beta, beta', beta" subunits and five major associated proteins of unknown function, but lacked sigma factors required for promoter recognition. The holoenzyme efficiently recognized a plastid psbA promoter when it was reconstituted from the purified PEP and recombinant plastid sigma factors. Reconstitution of a plastid holoenzyme with individual sigma factors will facilitate identification of sigma factor-specific promoter elements.  相似文献

Plastid-encoded plastid RNA polymerase (PEP) is essential for chloroplast development and plastid gene expression in Arabidopsis thaliana. However, PEP is a large complex, and many proteins in this complex remain to be identified. We previously reported that Delayed Greening 238 (DG238) interacts with PEP subunit protein FLN1 and may function as a PEP-associated protein and participate in early chloroplast development and PEP-dependent plastid gene expression. DG238 contains Domain of Unknown Function 143 (DUF143), whose function is currently unknown. Here, we found that a deficiency of the DUF143 domain in DG238 affected its localization, which resulted in abnormal interactions with PEP-associated proteins in the chloroplast. Furthermore, DG238 lacking the DUF143 domain or DG238 with only this domain failed to function. Interestingly, the lack of conserved amino acids 193–217 of the DUF143 domain in DG238 also affected its function. In addition to FLN1, DG238 also interacts with other PEP-associated proteins, including FSD2, FSD3, MRL7-L, and MRL7, to regulate plastid gene expression. These results suggest that the DUF143 domain is necessary for the functioning of the PEP-associated protein DG238 in chloroplasts.  相似文献

RNA polymerases of cyanobacteria contain a novel core subunit, gamma, which is absent from the RNA polymerases of other eubacteria. The genes encoding the three largest subunits of RNA polymerase, including gamma, have been isolated from the cyanobacterium Anabaena sp. strain PCC 7120. The genes are linked in the order rpoB, rpoC1, rpoC2 and encode the beta, gamma, and beta' subunits, respectively. These genes are analogous to the rpoBC operon of Escherichia coli, but the functions of rpoC have been split in Anabaena between two genes, rpoC1 and rpoC2. The DNA sequence of the rpoC1 gene was determined and shows that the gamma subunit corresponds to the amino-terminal half of the E. coli beta' subunit. The gamma protein contains several conserved domains found in the largest subunits of all bacterial and eukaryotic RNA polymerases, including a potential zinc finger motif. The spliced rpoC1 gene from spinach chloroplast DNA was expressed in E. coli and shown to encode a protein immunologically related to Anabaena gamma. The similarities in the RNA polymerase gene products and gene organizations between cyanobacteria and chloroplasts support the cyanobacterial origin of chloroplasts and a divergent evolutionary pathway among eubacteria.  相似文献
