 To examine the potential of a method of double-stranded (ds) RNA infection to Helicobasidium mompa, the transmissibility of dsRNA between strains of this fungus was investigated. Strain V70 was used as a dsRNA donor. The dsRNA recipients were six strains that were mycelially incompatible with V70, plus two monokaryotic strains. Random amplified polymorphic DNA analysis suggested that the mycelially incompatible strains were genetically different mutually; however, the analysis also suggested that V70 was genetically homogeneous with the two monokaryotic strains. When V70 was paired with either of the mycelially incompatible strains, the dsRNAs did not transmit to the recipients at all. When V70 was paired with the two monokaryotic strains, the dsRNAs were transmitted to the monokaryotic strains. The two monokaryotic strains, which had acquired dsRNAs from V70 in the previous experiment, were used as new dsRNA donors in a next experiment so that we could investigate dsRNA transmission from these monokaryotic strains to the six strains used in the previous experiment. One of the monokaryotic strains permitted dsRNA transmission to two of the six recipients. We conclude that we can infect genetically different strains of H. mompa with dsRNA using the monokaryotic strains.  To identify a basidiomycetous fungus isolated from butt rot of Chamaecyparis obtusa, Japanese cypress, its cultural features were examined, and sequences of its nuclear ribosomal 18S and ITS1–5.8S–ITS2 regions were analyzed. In culture, this fungus is characterized by the occurrence of chlamydospores, blastoconidium-like cells, and clavate-to-spathulate hyphal ends at the tips of aerial hyphae, and production of a small basidioma on the mycelial mat after 3 months of incubation. The morphological features of the basidioma are identical to those of Phlebia brevispora. Furthermore, molecular data of the sequences of these strains and P. brevispora showed a high level of similarity. These results appear to justify determining the present fungus as P. brevispora.  Three new species of Agaricales from eastern Honshu, Japan, are described and illustrated. (1) Clitocybe minutella sp. nov. (section Vernae), having white, very small, clitocyboid basidiomata with white, strigose mycelial tomentum at the base of the stipe and trichodermial elements in the pileipellis, occurs on leaf litter in deciduous oak forests. (2) Crepidotus longicomatus sp.nov. (section Echinospori), forms very small (up to 3.5 mm in diameter), reniform basidiomata densely covered overall with white to pale yellow, erect, thick-walled, long hairs. It has melleous, echinulate basidiospores, and was found on fallen dead branches in lowland oak forests. (3) Crepidotus virgineus sp. nov. (section Crepidotus), forming pure white, reniform basidiomata, has smooth, ellipsoid basidiospores, subfusiform-pedicellate to irregularly cylindrical cheilocystidia, and abundant clamp connections. It occurs on fallen dead branches in lowland oak forests.  Two new species and one new combination of Agaricales are described and illustrated from eastern Honshu, Japan, and Ishigaki Island, southwestern Japan: (1) Gymnopus oncospermatis comb. nov. (subsection Vestipedes of section Vestipedes), new to Japan, has pale brown to whitish, collybioid basidiomata with very close lamellae, and forms a distinct white extensive mycelial mat on the dead leaf bases of the palm Satakentia liukiuensis in Ishigaki Island; (2) Hydropus aurarius sp. nov. (subsection floccipedes of section floccipedes), forming clitocyboid basidiomata with a dark brown furfuraceous pileus and stipe and vivid yellow lamellae, found on fallen dead twigs in oak forests; and (3) Tylopilus castanoides sp. nov. (section Oxydabiles), forming medium to large, reddish-brown basidiomata with a minutely scurfy-punctate stipe, found in Carpinio quercion forests. Coprinopsis phlyctidospora (syn: Coprinus phlyctidosporus) from the Netherlands, Japan, New Zealand, and Australia can be segregated into two groups, northern and southern, based on the nucleotide sequences of their ITS regions. The mating type of a C. phlyctidospora isolate was tetrapolar. Mating reactions were compatible between monokaryotic testers derived from basidiospores of a Japanese isolate and dikaryotic isolates obtained from a wide geographic area in Japan. In contrast, mating between the Japanese monokaryotic and dikaryotic isolates from Australia and New Zealand were incompatible. These results indicated that C. phlyctidospora was complex and individuals currently recognized as C. phlyctidospora in the Northern Hemisphere and those in the Southern Hemisphere are distinct taxa. Summary.  2H-Pyran-2-ones 1 were transformed with various hydrazines into (E)- or (Z)-α,β-didehydro-α-amino acid (DDAA) derivatives 4 (and 7) containing a highly substituted pyrazolyl moiety attached at the β-position. With heterocyclic hydrazines, the products 4 were accompanied also by decarboxylated enamines E-6. In order to separate (E/Z)-mixtures of acids, they were transformed to the corresponding methyl esters 9 and 10 by the application of diazomethane. Catalytic hydrogenation under high pressures with Pd/C as a catalyst resulted in the formation of racemic alanine derivatives 11.  The karyotype of Flammulina velutipes (Curt. : Fr.) Sing. was investigated using contour-clamped homogeneous electric fields (CHEF) gel electrophoresis. A parental dikaryotic stock, JA, was resolved into at least eight chromosomal DNA bands ranging from 1.4- to 4.9-megabase (Mb) pairs. Overall, little size variation was found among monokaryotic strains with a few major exceptions. Among 13 monokaryotic progenies examined, 11 strains were resolved into at least eight chromosomal DNA bands in a manner similar to the parent dikaryon, whereas the other 2 were resolved into at least seven chromosomes lacking the 2.1-Mb chromosome possessed in the former. A slightly larger size variation was found in a chromosome carrying ribosomal DNA. An estimated haploid genome size of this stock was 24.0 Mb or more. Summary.  The effect of taurine (Tau) and taurine chloramine (Tau-Cl) on the production of TNF-α, IL-1β, and IL-6 by peripheral blood mononuclear cells of healthy volunteers was examined. Cells were stimulated with bacterial lipopolysaccharide (LPS) in the presence of either Tau or Tau-Cl. After 24 h culture the cytokine concentrations were measured in both culture supernatants (secreted) and cell lysates (cell-associated) using ELISA. In LPS-stimulated cells Tau-Cl inhibited both the secreted and cell-associated IL-1β and IL-6, while exerted dual effect on TNF-α production: raising it slightly at low and reducing at higher concentration. By contrast, Tau had no significant effect on the cytokine production. These results indicate that Tau-Cl modulates synthesis of pro-inflammatory cytokines, and therefore it may play a role in the initiation and propagation of immune response.  Chlamydospores of Entoloma clypeatum f. hybridum were described on the mycorrhizas and rhizomorphs associated with Rosa multiflora. Their developmental pattern seems to be the Nyctalis type. This is the first report on chlamydospore formation on the mycorrhizae in entolomatoid fungi. Summary.  The paper describes two methods of the synthesis of ethyl (3R,4S)- and (3S,4S)-4-[(benzyloxycarbonyl)amino]-5-[(tert-butyloxycarbonyl)amino]-3-hydroxypentanoates, useful for the syntheses of edeine analogs. Differently N-protected (S)-2,3-diaminopropanoic acid was used as a substrate in both procedures. The absolute configuration of newly generated asymmetric carbon atoms C-3 in β-hydroxy-γ,δ-diamino products was assigned by means of 1H NMR spectroscopy after their transformation into corresponding piperidin-2-ones.  Seven species of the genus Lophiostoma were the subject of this study. Among these, Lophiostoma mucosum is described and illustrated as a new species. All other species, L. macrostomum, L. semiliberum, L. arundinis, L. caulium, L. caudatum, and L. winteri, are reported for the first time in Japan. A key to the species of Lophiostoma in Japan is given.  Four new species of Crinipellis and Marasmius (Agaricales, Basidiomycetes) in eastern Honshu, Japan, are described and illustrated: (1) Crinipellis conchata sp. nov. (section Excentricinae), forming a conchate pileus and a strongly excentric, short stipe, was found on a dead twig of Trachelospermum asiaticum in Mt. Takao, Tokyo; (2) Marasmius funalis sp. nov. (section Androsacei), forming a densely white-hispid, dark brown stipe bearing numerous setiform caulocystidia, was found on a dead twig of Cryptomeria japonica or on leaf litter in Tokyo and Kanagawa; (3) Marasmius maculosus sp. nov. (section Sicci), having a relatively large, reddish-brown pileus distinctly mottled with pale colored spots and Siccus-type cheilocystidia and pileipellis cells with relatively long setulae, was found on leaf litter in the lowland forest of Kanagawa and Chiba; and (4) Marasmius sasicola sp. nov. (section Marasmius), having a small, plicate-sulcate pileus, a filiform, wiry, blackish stipe, collariate lamellae, and Siccus-type cheilocystidia and pileipellis elements, was found on fallen dead leaves of grass bamboo in Kanagawa. Summary.  Alterations in hepatic metabolism of S-amino acids were monitored over one week in male rats treated with a single dose of ethanol (3 g/kg, ip). Methionine and S-adenosylhomocysteine concentrations were increased rapidly, but S-adenosylmethionine, cysteine, and glutathione (GSH) decreased following ethanol administration. Activities of methionine adenosyltransferase, cystathionine γ-lyase and cystathionine β-synthase were all inhibited. γ-Glutamylcysteine synthetase activity was increased from t = 8 hr, but GSH level did not return to control for 24 hr. Hepatic hypotaurine and taurine levels were elevated immediately, but reduced below control in 18 hr. Changes in serum and urinary taurine levels were consistent with results observed in liver. Cysteine dioxygenase activity was increased rapidly, but declined from t = 24 hr. The results show that a single dose of ethanol induces profound changes in hepatic S-amino acid metabolism, some of which persist for several days. Ethanol not only inhibits the cysteine synthesis but suppresses the cysteine availability further by enhancing its irreversible catabolism to taurine, which would play a significant role in the depletion of hepatic GSH.  A Scleroderma species collected on sandy soil under trees of Lithocarpus edulis in Saitama Prefecture, central Japan, is identified as Scleroderma laeve, a new record for Japan.  A sporeless mutant dikaryon, completely defective in sporulation, was isolated from mycelial protoplasts of Pleurotus eryngii mutagenized by UV irradiation. Newly established dikaryons between one component monokaryon from the mutant, and 12 different wild type monokaryons from 3 other wild type dikaryons, all exhibited the sporeless phenotype, whereas those between the other monokaryon and the same wild type monokaryons all produced normal fruiting bodies. These results indicated that the sporeless mutation was induced in one of two nuclei of the mutant and was dominant. In the wild type basidia, the pattern of nuclear behavior during sporulation corresponded to the pattern C nuclear behavior as defined by Duncan and Galbraith. Cytological observation revealed that in the sporeless mutant meiosis was blocked at the meta-anaphase I in most basidia and hence basidiospores and sterigmata were not produced. Although fruiting bodies of the sporeless mutant showed a somewhat leaning growth, their gross morphology and its fruiting body productivity were comparable to that of the original wild type strain. Based on these results, it was considered that the sporeless mutant could serve as a potential material in breeding of sporeless P. eryngii commercial strains. Summary.  The 14-residue peptaibol antibiotic trichovirin I 4A of the structure Ac-Aib-L-Asn-L-Leu-Aib-L-Pro-L-Ala-L-Val-Aib-L-Pro-Aib-L-Leu-Aib-L-Pro-L-Leuol (Aib = α-aminoisobutyric acid, Leuol = leucinol) was synthesized by stepwise conventional solution phase synthesis using the Z/OtBu(OMe) strategy and HOBt/EDC as coupling reagents. Intermediates were fully characterized and the identity of the synthetic peptide with the component 4A of the natural, microheterogeneous peptide mixture was proven by electrospray mass spectrometry, HPLC, and bioassay.  We examine a generalised SIR model for the infection dynamics of four competing disease strains. This model contains four previously-studied models as special cases. The different strains interact indirectly by the mechanism of cross-immunity; individuals in the host population may become immune to infection by a particular strain even if they have only been infected with different but closely related strains. Several different models of cross-immunity are compared in the limit where the death rate is much smaller than the rate of recovery from infection. In this limit an asymptotic analysis of the dynamics of the models is possible, and we are able to compute the location and nature of the Takens–Bogdanov bifurcation associated with the presence of oscillatory dynamics observed by previous authors.  Three aryl alcohol oxidases (AAOs; EC 1.1.3.7) I, II, and III from the culture filtrate of a strain of white-rot fungus Pleurotus ostreatus were purified by multistep chromatography. Each of the purified AAOs I, II, and III had the same molecular masses of 70 kDa and 72 kDa on gel filtration chromatography and sodium dodecyl sulfate-polyacrylamide gel electrophoresis, respectively. Their optimum temperature was 40°C, but their optimum pHs differed slightly. The N-terminal amino acid sequence of AAOs I, II, and III was determined to be Ala-Asp-Lys-Asp-Tyr-Ile-Val-Val-Gly-Ala, which showed significant similarity to those of Pleurotus eryngii (80% identity) and Pleurotus ostreatus Florida (60% identity).
